Abstract

Relacionar el aprendizaje activo y la responsabilidad social universitaria, resaltando la importancia de actividades académicas con valores para sentar las bases de atención primaria de salud. Diseño observacional, descriptivo y multidisciplinario con participación de profesores y estudiantes universitarios de las carreras de Medicina, Nutrición y Odontología, de 2o-4o semestres para brindar atención primaria de salud. Mediante consentimiento informado se atendió a más de 300 pobladores de diferentes comunidades incluyendo, niños, adolescentes y adultos. Los estudiantes educaron en salud con talleres de concientización y dinámicas grupales en temas relacionados al bienestar social como cuidado de mascotas, lavado de manos, toxoplasmosis, COVID-19, parasitosis alimentaria, plato del buen comer y salud bucal; se tomaron muestras de sangre para determinación de glicemia, colesterol, triglicéridos, grupo sanguíneo y antígeno prostático. Con este trabajo comunitario de atención a la salud, en escenarios reales, se logró el aprendizaje significativo en las áreas de bioquímica, histología, fisiología, nutrición, patología especial, patología y medicina bucal con trato digno al paciente.

17. Peonidin-3- O -glucoside and Resveratrol Increase the Viability of Cultured Human hFOB Osteoblasts and Alter the Expression of Genes Associated with Apoptosis, Osteoblast Differentiation and Osteoclastogenesis.

Abstract

High-throughput RNA-sequencing can determine the impact of nutrients and their combinations on gene transcription levels in osteocytes, and clarify the biological pathways associated with their impact on bone tissues. Previously, we reported that resveratrol (RES) and peonidin-3-O-glucoside (POG) increased osteoblastogenesis, as well as reduced osteoclastogenesis in transgenic teleost fish models. Here, we perform whole-genome transcriptomic profiling of osteoblasts treated with POG or RES to provide a comprehensive understanding of alterations in gene expression and the molecular mechanisms involved. Cultured human fetal osteoblastic hFOB 1.19 cells were treated with the test compounds, and then RNA was used to prepare RNA-seq libraries, that were sequenced using a NovaSeq 6000. Treatment with POG or RES increased osteoblast proliferation and reduced apoptosis. Transcriptomic profiling showed that of the 29,762 genes investigated, 3177 were differentially expressed (1481 upregulated, 1696 downregulated, FDR ≤ 0.05) in POG-treated osteoblasts. In the RES-treated osteoblasts, 2288 genes were differentially expressed (DGEs, 1068 upregulated, 1220 downregulated, FDR ≤ 0.05). Ingenuity® Pathway Analysis (IPA) of DGEs from RES or POG-treated osteoblasts revealed significant downregulation of the apoptosis, osteoarthritis and HIF1α canonical pathways, and a significant reduction in Rankl mRNA expression. Abstract

Background To support occupational physicians (OPs) in the implementation of workers' health surveillance (WHS), a training programme was developed. Aims (i) To evaluate the effects of a WHS training programme for OPs on knowledge, self-efficacy and skills to implement WHS. (ii) To evaluate to what extent a WHS training programme is acceptable and feasible for implementation in practice. Methods A single-blinded randomized controlled trial with waiting-list control group was used. The WHS training programme consisted of an e-learning and a 4.5-h online training session. OPs completed a knowledge test (0–8), self-efficacy questionnaires on knowledge and skills (6–60), and vignette assignments (0–16) to measure skills. OPs completed the questionnaires, either before and after the WHS training programme (intervention group), or before the training programme (control group) while receiving the training programme after the waiting period. All OPs completed questionnaires about the training's acceptability, and feasibility for implementation in practice. ANCOVA and Poisson regression analyses were conducted. Results The self-efficacy score (M = 44.1 versus M = 37.2) (P < 0.001) and skills score (M = 9.6 versus M = 8.3) (P < 0.05) of OPs in the training group (N = 16) were higher than the control group (N = 23). No effect was found on knowledge. Evaluation of acceptability and feasibility showed that 21 (58%) OPs were very satisfied with the training part on initiating WHS, and 29 (85%) would recommend the WHS training programme to colleagues. Conclusions This WHS training programme has a positive effect on self-efficacy and skills of OPs to implement WHS, and may be acceptable and feasible to implement in practice. [ABSTRACT FROM AUTHOR]

Abstract

El objetivo de la investigación fue aislar y caracterizar cepas de Vibrio obtenidas de agua marina en la línea de playa y la zona adyacente a la plataforma de extracción petrolera CX-11 del Lote Z1 ubicado a 17 km de la ciudad de La Cruz (Tumbes, Perú). Las cepas fueron aisladas en caldo mineral Bushnell Haas suplementado con petróleo crudo, luego fueron subcultivadas en caldo de tripticasa soya (TSB) hasta obtener colonias puras, las cuales fueron caracterizadas mediante tinción de Gram y las pruebas de oxidasa y catalasa; además fueron identificadas mediante secuenciamiento de un fragmento de sus genes 16S ARNr. Se verificó que las cepas portaran genes de benceno di oxigenasas. Se pudo aislar seis cepas correspondientes a Vibrio fluvialis y Vibrio spp., que portaban los genes indicados. La investigación demostró que en aguas de la línea de playa y zonas adyacentes a la plataforma de extracción CX-11 del Lote Z1, existen cepas de Vibrio fluvialis y Vibrio spp. con capacidad para degradar hidrocarburos.

Abstract

Introduction: Numerous studies over the past 30 years have shown that bioactive compounds present in functional foods, including vitamins, minerals and phytochemicals reduce cancer risk. For example, vitamins A and D derivatives found in fruits, vegetables, and dairy products, reduce the growth of breast, bladder, head, neck, lung, prostate, and skin cancers. However, the effects of these combined vitamins have not been previously reported for breast cancer. Aims: To investigate the activities of vitamin A (all-trans-retinoic acid; ATRA), as well as vitamins D2 and D3 in combination in the breast epithelial cancer cell lines T47D:A18, MCF-7, and SK-BR-3 and perform whole genome analysis in MCF-7 cells using RNA-seq. Methods: Breast cancer cells were cultured in appropriate media and treated with ATRA, D2 or D3 alone in concentrations from 1-10 μg/ml, or in combination at 1, 5, and 10 μg/ml. The CellTiter-Glo® 2.0 assay, Caspase-Glo®3/7, Caspase®8, and ApoTox-Glo™ Triplex assays measured cell viability and apoptosis. The effect of treatment on autophagy in MCF-7 cells was measured with a CYTO-ID® Autophagy Detection Kit 2.0. The whole transcriptome analysis was assessed using mRNA-seq and qPCR. Results: Separately, ATRA, D2, and D3 all reduced the viability of all breast cancer cell lines tested, with median inhibitory concentrations (IC50) between 2.1 to 31.7 μg/ml. However, when breast cancer cells were treated with combinations of ATRA+D2+D3, the IC50 was reduced indicating synergism. In MCF-7 cells, 5-flurouracil (5-FLU) had an IC50 of 1.37 µg/ml, while the vitamin A and D combination had an IC50 of 1.5 µg/ml, indicating the combination was ~90% as effective as 5- FLU. Treatment of MCF-7 cells with ATRA+D2+D3 enhanced caspase 3/7 activity, as well as the expression of Bax, BAD, PTEN and p53 (apoptosis canonical pathway), as well as induced autophagy. Whole genome analysis of treated MCF-7 cells showed a significant upregulation in gene expression in the autophagy, endoplasmic reticulum stress and the unfolded protein response apoptosis canonical pathways. Furthermore, MCF-7 cells treated with ATRA+D2+D3 demonstrated significant downregulation of gene expression in estrogen-mediated S phase entry and estrogen signaling canonical pathways suggesting antiestrogenic effects. Conclusions: Vitamins A and D combinations had synergistic effects in breast cancer cells and induced both apoptosis and autophagy. Transcriptional profiling showed significant alterations in gene expression patterns and upregulation of multiple cancer signaling pathways supporting the hypothesis that combining vitamins A and D is a more effective treatment than either vitamin alone. Abstract

Background. Chronic kidney disease (CKD) is associated with an elevated risk of all-cause mortality, with cardiovascular death being extensively investigated. However, non-cardiovascular mortality represents the biggest percentage, showing an evident increase in recent years. Klotho is a gene highly expressed in the kidney, with a clear influence on lifespan. Low levels of Klotho have been linked to CKD progression and adverse outcomes. Single nucleotide polymorphisms (SNPs) of the Klotho gene have been associated with several diseases, but studies investigating the association of Klotho SNPs with noncardiovascular death in CKD populations are lacking. Methods. The main aim of this study was to assess whether 11 Klotho SNPs were associated with non-cardiovascular death in a subpopulation of the National Observatory of Atherosclerosis in Nephrology (NEFRONA) study (n ¼ 2185 CKD patients). Results. After 48 months of follow-up, 62 cardiovascular deaths and 108 non-cardiovascular deaths were recorded. We identified a high non-cardiovascular death risk combination of SNPs corresponding to individuals carrying the most frequent allele (G) at rs562020, the rare allele (C) at rs2283368 and homozygotes for the rare allele (G) at rs2320762 (rs562020 GG/AG þ rs2283368 CC/CT þ rs2320762 GG). Among the patients with the three SNPs genotyped (n ¼ 1016), 75 (7.4%) showed this combination. Furthermore, 95 (9.3%) patients showed a low-risk combination carrying all the opposite genotypes (rs562020 AA þ rs2283368 TT þ rs2320762 GT/TT). All the other combinations [n ¼ 846 (83.3%)] were considered as normal risk. Using competing risk regression analysis, we confirmed that the proposed combinations are independently associated with a higher fhazard ratio [HR] 3.28 [confidence interval (CI) 1.51-7.12]g and lower [HR 6 × 10-6 (95% CI 3.3 × 10-7-1.1 × 10-5)] risk of suffering a non-cardiovascular death in the CKD population of the NEFRONA cohort compared with patients with the normal-risk combination. Conclusions. Determination of three SNPs of the Klotho gene could help in the prediction of non-cardiovascular death in CKD. © The Author(s) 2020. Abstract

Background Mental health complaints occur frequently among healthcare workers. A workers' health surveillance mental health module (WHS-MH) was found to be effective in improving work functioning of nurses, although not implemented regularly. Therefore, training for occupational physicians and nurses to facilitate the implementation of WHS-MH was developed and evaluated. Aims This study was aimed to evaluate the effect of training on knowledge, self-efficacy and motivation to implement WHS-MH, and to evaluate participants' satisfaction with the training. Methods Three-hour training was held among 49 occupational physicians and nurses in the Netherlands. Effect on knowledge, self-efficacy and motivation was assessed using knowledge tests and questionnaires before and immediately after the training. Satisfaction with the training was measured using questionnaires after the training. Results A mean knowledge test score of 5.3 (SD = 1.6) was found before training, which did not significantly increase after training (M = 5.6, SD = 1.8). In total, 43% agreed or strongly agreed to have sufficient skills to implement WHS-MH, which significantly increased to 78% after the training. Furthermore, 87% agreed or strongly agreed to be motivated to initiate WHS, which significantly increased to 94% after the training. The majority of participants were satisfied with the training. Conclusions Training may enhance the implementation of the WHS-MH through increasing self-efficacy and motivation. However, no effect on level of knowledge to implement WHS-MH was found. Abstract

Background: Lateral epicondylitis (LE) is a highly prevalent musculoskeletal disorder in workers, often associated with physically demanding work. Knowledge of work‐relatedness of LE is crucial to develop appropriate preventive measures. This study investigates the prospective association between work‐related physical risk factors and LE. Methods: A systematic literature review was conducted in MedLine using PubMed from January 1, 2010 until February 16, 2021. Published reports were included if: (1) LE was clinically assessed, (2) exposure to work‐related physical risk factors was assessed, and (3) associations between LE and work‐related physical risk factors were reported in prospective studies. Quality of evidence was assessed using the Grading of Recommendations, Assessment, Development and Evaluation. Results: In total, 318 workers with LE from a population of 5036 workers in five studies were included. Meta‐analyses revealed high‐quality evidence for associations between LE and a Strain Index (SI) score >5.1 (odds ratio [OR]: 1.75, 95% confidence interval (CI): 1.11–2.78) and moderate‐quality evidence for forearm rotation >4 h/day or forearm rotation ≥45° for ≥45% time (OR: 1.85, 95% CI: 1.10–3.10). Gripping, flexion and extension of the wrist, and repetitive movements showed no significant associations with LE. Conclusion: High‐quality evidence was found indicating that a higher SI increased the risk of LE. Moderate‐quality evidence was found for an association between forearm rotation and LE. No associations were found between other physical risk factors and LE. Primary preventive interventions should focus on a reduction of the SI and of high forearm rotation in work. Abstract

Los peces venenosos y tóxicos forman parte de la riqueza en biodiversidad del Perú, sus venenos y toxinas contienen metabolitos que pueden emplearse para producir sustancias farmacológicas. La investigación tuvo como objetivo identificar, mediante ADN barcoding, las especies de peces venenosos y tóxicos en el manglar de Tumbes. Se recolectaron 46 ejemplares de estos peces que fueron identificados morfológicamente. Para el análisis de ADN barcoding, se obtuvo sus secuencias de un fragmento del gen de la subunidad I de la citocromo oxidasa (COI), con las cuales se realizó su identificación en Blast y Bold. Se realizó el análisis filogenético mediante Neighbor joining (NJ), automatic barcode gap discovery (ABGD) y modelo generalizado mixto de Yule y coalescencia (GMYC), que confirmaron la asignación de especies. Mediante ADN barcoding se identificó las especies: Daector dowi, Diodon holocanthus, Sphoeroides andersonianus, S. annulatus, S. kendalli, S. sechurae, Gymnura marmorata, Urotrygon asterias y U. peruanus. En algunos ejemplares la identificación mediante ADN barcoding, caracteres morfológicos y análisis filogenético fueron disimiles como en los géneros Sphoeroides y Urotrygon, por lo que deberían realizarse más estudios para confirmar la presencia de complejos de especies altamente emparentadas en ellos.

Abstract

El Perú es un país megadiverso que destaca por su biodiversidad de peces, entre ellos los del manglar de Tumbes. Este ecosistema se encuentra amenazado por diversas causas, una de ellas es la introducción de especies invasoras. La investigación se realizó entre junio de 2016 y agosto de 2018, tuvo como objetivo identificar, mediante la técnica de ADN barcode, los peces invasores en el manglar de Tumbes. Se recolectaron peces en 18 puntos de muestreo, estos fueron identificados por sus características morfológicas. Para la identificación mediante ADN barcode, se obtuvo sus secuencias de un fragmento del gen de la subunidad I de la citocromo oxidasa (COI), con las cuales se realizó su identificación en las bases de datos de Blast y Bold. También se realizó análisis filogenético, construyendo árboles basados en los métodos de Neighbor joining (NJ), descubrimiento automático de brecha de barcode (ABDG) y modelo generalizado mixto de Yule y coalescencia (GMYC), con los cuales se confirmó la asignación de especies. De los análisis realizados se concluyó la presencia de cuatro especies invasoras: Poecilia reticulata, Oreochromis mossambicus, O. niloticus y O. urolepis.

47. Desarrollo de una conserva de langostino en aceite vegetal: Tratamiento térmico, contenido nutricional e inocuidad microbiológica

Abstract

La investigación tuvo como objetivo determinar el contenido nutricional y la inocuidad microbiológica de conservas de langostino en aceite vegetal preparadas de forma experimental en la Escuela de Ingeniería Industrial Pesquera (EIIP) de la Universidad Nacional de Tumbes. Las conservas fueron preparadas con langostino que fue descabezado, pelado, desvenado y pre-cocido antes de ser envasado en latas con aceite de girasol. El tratamiento térmico fue de 116 °C por 75 min. El contenido nutricional y análisis microbiológico fueron realizados en laboratorio acreditado. Como resultado se obtuvo que el contenido nutricional fue de 26,47% de proteína, 5,36% de grasa, 66,81% de humedad, 1,32% de ceniza y 0,04% de carbohidratos, con un valor energético de 154,28 kcal/100 g. Las conservas presentaron un pH de 5,04; respecto a su inocuidad microbiológica, resultaron negativas a la presencia de microorganismos mesófilos o termófilos tanto aerobios como anaerobios; de igual manera no mostraron crecimiento bacteriano luego de incubación a 30 a 35 °C por 14 días, así como de 52 a 55 °C por 7 días. La investigación demostró que es posible preparar conservas de langostino en aceite vegetal con buena composición nutricional y que sean inocuas microbiológicamente.

The article conducts a study of the international and national experience of introducing the presumption of guilt principle for officials who are suspected of corruption crimes into legislation of certain states, determining the effectiveness of such implementations, fixing the possibility of introducing the principle of presumption of guilt in international law, assesses the decisions of the competent authorities of Ukraine regarding Ukrainian anti-corruption legislation which enshrined in the legal system of Ukraine legal presumption of guilt principle. The author conducts a study to determine whether the restriction of the legal principle of the presumption of innocence, that is, the introduction of the presumption of guilt, is applied to facilitate and enhance the effectiveness of the fight against corruption, in particular, to easily prove the crime and find evidence of illegal enrichment. Is there a definition of the possibility of introducing a presumption of guilt in relation to crimes for officials concerning corruption by UN international conventions and international conferences for member states, the content and resolution of which they have ratified. The author determines whether the limitation of the presumption of innocence can be limited to a narrower sphere, namely, to act in relation to measures to confiscate property for criminal acts. The author also conducts a study on whether the introduction of the presumption of guilt for officials can take place against the background of the complete abolition of the presumption of innocence at the legislative level, whether there is evidence of the advisability of introducing such fundamental legislative changes

Abstract

Measurements of the adiabatic temperature change (ΔT) and the specific heat transfer (ΔQ) of Ni2.18Mn0.82Ga Heusler alloy were taken in order to quantify the direct giant magnetocaloric effect of the alloy when it is in the vicinity of magneto-structural phase transition (PT) from paramagnetic austenite to ferromagnetic martensite, and their results are presented. A new vacuum calorimeter was used to simultaneously measure ΔT and ΔQ of magnetocaloric materials with a Bitter coil magnet in fields of up to H=140 kOe. Other thermomagnetic properties of this alloy were investigated using standard differential scanning calorimetry and PPMS equipment. The maximal values of magnetocaloric effect in H=140 kOe were found to be ΔT=8.4K at initial temperature 340K and ΔQ=4900 J/kg at 343 K. Using this direct method, we show that the alloy indeed demonstrates the largest value of ΔQ as compared with previously published results for direct measurements of magnetocaloric materials, even though at 140 kOe the magnetic field-induced magnetostructural PT is still not complete.
