Stressful events can alter organism physiology at several levels triggering allostatic responses. Telomeres are well-conserved repetitive DNA sequences mainly localised at chromosome's ends, playing a crucial role in DNA stability. Analyses of telomere dynamics are new tools to assess consequences of environmental stress in non-model organisms like fish. In this study, the relationship between freshwater tolerance and telomere dynamics was investigated in the gills of the European sea bass Dicentrarchus labrax . Fluorescent in situ hybridisation of telomeric sequences revealed distal telomeres as well as intrachromosomal telomeres known as interstitial telomere sequences. In order to better understand telomere dynamics in the gills of D. labrax , we used quantitative PCR to measure telomere length and mRNA expression of the catalytic subunit of telomerase reverse transcriptase tert. For the calculation of the relative telomere length, two reference genes were tested: the single copy gene mc2r , encoding melanocortin 2 receptor and the multicopy gene 18S , encoding the 18S ribosomal RNA . We proposed a novel normalisation method to calculate the relative telomere length using both, single and multiple copy genes as references. Cell dynamics was also investigated by measuring mRNA expression of genes involved in apoptosis ( caspase 8 and 9 ), cell proliferation ( proliferation cell nuclear antigen ), aerobic mitochondrial metabolism (ATP citrate-synthase), anaerobic metabolism ( lactate dehydrogenase a ) and antioxidant enzymatic defences ( superoxide dismutase 1 and 2, catalase ). Following a 15-days fresh water exposure, telomere dynamics was not significantly modified in the gills of freshwater tolerant fish. But freshwater intolerant fish exhibited telomere attrition relative to saltwater controls, and lower expression of tert in gills relative to freshwater tolerant fish. This modification of telomere dynamics in intolerant individuals was found to be correlated with lower antioxidant enzymatic defences, a higher aerobic metabolic marker and a lower cellular turnover. These data bring new perspectives for the use of telomere dynamics as an integrative marker to study environmental stress in fish, while considering individual phenotypic plasticity in response to freshwater exposure.

WOS:000475657500006; International audience; In teleosts, the regulation of hydromineral balance has a direct impact on several physiological functions, biochemical processes, and can influence behaviour, distribution and survival. As European sea bass Dicentrarchus labrax undertake seasonal migrations from seawater (SW) to brackish, estuarine and fresh water (FW) in their habitat, this study investigates their capacity to tolerate fresh water and explores intraspecific variations in physiological responses. Juvenile D. labrax were transferred from SW to FW at various ages. Freshwater-tolerant and non-tolerant phenotypes were discriminated according to behavioural and morphological characteristics. About 30% of the fish exposed to FW were identified as freshwater intolerant following FW challenges performed at different ages. Interestingly, intolerant fish exhibited the same phenotypic traits: erratic swimming, lower speed, isolation from the shoal and darker colour. Freshwater-intolerant fish were also characterised by a significant lower blood osmolality compared to tolerant fish, and significantly lower Na+/K+-ATPase α1a expression in the posterior kidney. An imbalance in ion regulatory mechanisms was further confirmed by a blood Na+/Cl− ratio imbalance observed in some freshwater-intolerant fish. The analysis of glucocorticoid and mineralocorticoid receptor expression levels in gills and kidney revealed significant differences between freshwater-intolerant and -tolerant fish in both organs, suggesting differential stress-related responses. This study clearly shows an intraspecific difference in the responses following FW transfer with a decreased renal ion uptake capacity as a major cause for freshwater intolerance.

International audience; Chaque année 900 à 1000 étudiants sont inscrits dans le portail MONOD de l’UE Biologie Intégrative des organismes. Le département Biologie-Ecologie a fait le choix de favoriser l’enseignement pratique pour favoriser l’apprentissage. Cette UE de 5 ECTS compte donc 30h de TP, 10.5h de TD pour 9h de CM. L’enjeu de cette UE est de sensibiliser les étudiants à toutes les facettes de la biologie tout en leur faisant acquérir, par la pratique, un certain nombre d’apprentissage basiques spécifiques à la biologie (reconnaitre et classer les organismes avec les méthodes dédiées…) et des compétences plus transversales : rédiger un compte-rendu d’observation, faire un diagramme de manipulation tout en respectant un horaire. Le premier verrou est donc de faire acquérir de nombreuses compétences à différents niveaux et dans le même temps. À cela s’ajoute le nombre et la diversité des profils étudiants mais aussi le nombre et la diversité des profils enseignants.Savoir rédiger un compte-rendu d’observation et d’expérimentation est une compétence essentielle pour un scientifique et suit la logique de rédaction d’une publication. L’objectif de notre étude a donc été d’optimiser cet apprentissage et de pallier le manque d’uniformité à la fois sur la forme et le délai de rendu des corrections des comptes rendus. De plus, les corrections des enseignants sont souvent incomprises et mal vécues. L’autoévaluation et l’évaluation par les pairs semblent des méthodes qui permettent aux étudiants de mieux comprendre les attendus. Dans l’autoévaluation, l’étudiant est face à lui-même et n’a pas à craindre de l’image qu’il renvoie. L’évaluation par les pairs permet une relation apprenant-apprenant. On n’est pas dans un rapport hiérarchique. Par contre, la crainte du regard de l’autre peut-être un biais. L’anonymat est une solution pour pallier ce biais. Utiliser cette méthode via une interface informatique de type Moodle permet la mise en place d’une uniformisation plus contrôlée, et un paramétrage facilité de la répartition, de l’anonymat et de la notation. Elle nécessite en préalable une bonne pratique des outils de bureautique par les étudiants. Notre objectif a donc été d’utiliser la plateforme Moodle pour utiliser l’outil atelier à la fois pour l’autoévaluation des 2 premiers TP et l’évaluation par les pairs du TP3, suivi d’une évaluation classique par l’enseignant pour le TP4. Nous avons bénéficié pour cela d’un soutien de la Faculté des Sciences de l’université de Montpellier via l’appel à projet TICE 2016-2017.Nos résultats concernent à la fois l’appréciation des étudiants pour cet outil mais aussi le bilan sur l’évolution des notes au cours de l’année et en comparaison avec les promotions des années précédentes n’ayant pas eu ce dispositif. Cet outil ne peut que perdurer s’il a l’adhésion à la fois des étudiants et des enseignants. Nos résultats montrent que les étudiants sont majoritairement favorables à cet outil s’il est utilisé tôt dans l’apprentissage. Le retour des enseignants est plus hétérogène notamment sur le fait d’intégrer cette évaluation dans la moyenne de l’UE. Nous commenterons aussi l’outil, l’avantage et l’inconvénient des différents paramètres testés mais aussi le temps de gestion en regard de l’utilisation de cet outil. Enfin, nous proposerons des perspectives d’amélioration afin d’optimiser son utilisation. En effet, la mise en place et la maintenance est assez lourde durant l’opération. Une sensibilisation des enseignants et une formation à l’outil permettrait de tirer tous les avantages de cet outil.

International audience; The copepod Eurytemora affinis has an unusually broad salinity range, as some populations have recently invaded freshwater habitats independently from their ancestral saline habitats. Prior studies have shown evolutionary shifts in ion transporter activity during freshwater invasions and localization of ion transporters in newly discovered “Crusalis organs” in the swimming legs. The goals of this study were to localize and quantify expression of ion transport enzymes V-type H+-ATPase (VHA) and Na+/K+-ATPase (NKA) in the swimming legs of E. affinis and determine the degree of involvement of each leg in ionic regulation. We confirmed the presence of two distinct types of ionocytes in the Crusalis organs. Both cell types expressed VHA and NKA, and in the freshwater population the location of VHA and NKA in ionocytes was, respectively, apical and basal. Quantification of in situ expression of NKA and VHA established the predominance of swimming leg pairs 3 and 4 in ion transport in both saline and freshwater populations. Increases in VHA expression in swimming legs 3 and 4 of the freshwater population (in fresh water) relative to the saline population (at 15 PSU) arose from an increase in the abundance of VHA per cell rather than an increase in the number of ionocytes. This result suggests a simple mechanism for increasing ion uptake in fresh water. In contrast, the decline in NKA expression in the freshwater population arose from a decrease in ionocyte area in legs 4, likely resulting from decreases in number or size of ionocytes containing NKA. Such results provide insights into mechanisms of ionic regulation for this species, with added insights into evolutionary mechanisms underlying physiological adaptation during habitat invasions.
