Der Schlaganfall ist definiert als ein akut auftretendes neurologisches Defizit und wird durch Verminderung des intrazerebralen Blutflusses und der Sauerstoffversorgung oder durch Hä-morrhagie verursacht. In der westlichen Welt gilt er nach wie vor als eine der führenden Todesursachen und Gründe für erworbene Behinderung. Neben der großen sozioökonomischen Belastung, die der Schlaganfall darstellt, mangelt es noch immer an suffizienten Behandlungsstrategien, um die Langzeitfolgen zu verbessern oder endogene Resilienzstrategien des Gehirns zu unterstützen. Aus diesem Grund wurde viel Forschung betrieben, um die Pathophysiologie und die molekularen Mechanismen des Schlaganfalls besser zu verstehen, und um potentielle Kandidaten zu identifizieren, die positiven Einfluss auf die Stresstoleranz von Gehirnzellen und speziell von Neuronen nehmen. Eine Strategie verfolgt das Ziel, evolutionär konservierte Selbstschutz- und -heilungskapazitäten des Gehirns zu identifizieren. Das Zinkfingerprotein 580 (Zfp580) ist ein Transkriptionsfaktor, der vielversprechende Charakteristika aufweist, um im Zusammenhang mit dieser sog. endogenen Neuroprotektion (Resilienzsteigerung) diskutiert zu werden. Als Teil der C2H2-Familie von Zinkfingerproteinen (Zfp) enthält das 172 Aminosäuren (AS) lange Protein eine C-terminale DNA-Bindungsstelle und einen prolinreichen N-Terminus, der Protein-Protein-Interaktion ermöglicht. In Endothelzellen ist das Protein bereits gut untersucht. Dort werden ihm angiogenesefördernde, antiinflammatorische und anti-apoptotische Eigenschaften zugeschrieben. Vor allem aber ist es seine zuträgliche Rolle in der Hypoxietoleranz von Myokardzellen, die es zu einem vielversprechenden Kandidaten für die Schlaganfallforschung im Mausmodell macht. Dieses Projekt ist das erste uns bekannte, das sich zum Ziel gesetzt hat, die Expression und Regulation von Zfp580 in den Modellen der MCAo und OGD in Neuronen der Maus zu untersuchen. Wir konnten zeigen, dass das Protein in Neuronen des gesamten murinen Gehirns exprimiert wird, und dass Hypoxie zu einer raschen Herunterregulation des Proteins auf Protein- nicht aber auf mRNA-ebene führte. Wir konnten zudem zeigen, dass sich eine neuronenspezifische Reduktion der mRNA von Zfp580 durch lentivirale Transduktion von kleinen Zfp580-spezifischen Haarnadel-RNAs (shRNA) auf das Zellüberleben nach hypoxischem Stress auswirkte, also neuroprotektiv wirkt. Trotz dieser Ergebnisse erbrachte ein konstitutiver und zelltypübergreifender Knockout von Zfp580 keine Überlebensvorteile im kurzen Erholungsintervall nach MCAo bzw. OGD. Diese Ergebnisse legen nahe, dass die schnelle und effektive Reduktion des Transkriptionsfaktors Teil einer endogenen Antwort der Neuronen auf ischämie-ähnlichen Stress ist. Ebenfalls wird deutlich, dass die Rolle von Zfp580 im Zusammenspiel intra- und interzellulärer Signalwege bei Hypoxie noch nicht endgültig verstanden ist. Trotz dessen ist Zfp580 ein Kandidat, der für eine medikamentöse Akutintervention im Falle des Schlaganfalls zu diskutieren bleibt., Stroke as a sudden neurological deficit caused by an intracerebral reduction of blood flow and oxygen supply or hemorrhage is still one of the major causes of death and acquired disa-bility in the western world. Despite its impact as a socioeconomic burden, therapeutic strategies to improve long- term outcome or minimize lesion sizes are missing. Therefore, research has put great effort into the examination of the pathophysiological and molecular processes in stroke in order to find potential candidates that can improve stroke outcome or foster endogenous relilience strategies against ischemic-like stress. One of these strategies is to foster evo-lutionary conserved self-healing capacities of the brain, so-called endogenous neuroprotection. Zinc finger protein 580 (Zfp580) serves as potential candidate to achieve these goals. Being part of the broad C2H2-familiy of zinc finger proteins, the transcription factor of 172 amino acids contains a C-terminal DNA-binding motif and a proline-rich N-terminus for protein-protein interaction. It has been well examined in endothelial cells where it displays pro-angiogenetic, anti-inflammatory and anti-apoptotic characteristics. However, it is its beneficial role in hypoxic tolerance in myocardial cells which makes it promising to be examined in mouse models of cerebral ischemia. To our knowledge, this project is the first to elucidate the expression and regulation of Zfp580 in neurons in the context of middle cerebral artery occlusion (MCAo) and combined oxygen and glucose deprivation (OGD). We showed that Zfp580 is expressed in neurons throughout the brain and Zfp580 expression levels were quickly decreased after induction of hypoxia on protein, however not on mRNA level. In a model of lentivirus-mediated miR-shRNA transduction in order to efficiently silence Zfp580 in a neuron-specific manner, we provided evidence that the reduction of Zfp580 is neuroprotective after OGD. Despite these findings, a constitutive and cell type unspecific knockout of Zfp580 could not provide beneficial effects on the tolerance against ischemic-like stress in short-term outcome after MCAo. These findings suggest that Zfp580 is subject to a fast and effective endogenous mechanism in the stress response of neurons against ischemic-like stress. Despite this, its role in the complexity of orchestrating signals in and in between brain cells undergoing hypoxia is not conclusively understood. However, since Zfp580 is a protein that seems to play a role in endogenous neuroprotection, it might serve as a druggable candidate in the treatment of acute ischemic stroke.

Experimental butters with a high content of trans-18 : 1 fatty acids and/or cis-9,trans-11-18 : 2 (rumenic acid; RA) were fed to thirty-six New Zealand White rabbits to investigate their effects on adipose tissue (AT) and liver lipogenic activities. Animals received one of three atherogenic (0.2 % cholesterol) diets containing 12 % butter with either a standard fatty acid composition (rich in saturated fatty acids), rich in trans-10-18 : 1 (T10 diet) or in trans-11-18 : 1 plus RA (T11+ RA diet) for 6 or 12 weeks. The ingestion of butters rich in trans fatty acids and/or RA for 6 weeks had little or no effect on liver and AT lipogenesis. The ingestion for 12 weeks of butter rich in T11+ RA decreased perirenal AT weight, lipogenic enzyme and lipoprotein lipase activities, without affecting liver lipid concentration or lipogenic activities except for a decrease in glycerol-3-phosphate dehydrogenase activity. Similar trends, but of a lower magnitude, were observed in rabbits fed the T10 diet for 12 weeks. Ingestion of the T10 or T11+ RA diets for 6 or 12 weeks had no significant effect on plasma metabolites and hormones except for glucose which increased at 6 weeks in the T10 group. Plasma leptin concentration was positively correlated with AT weight but did not differ between the three diets. In conclusion, the supply of butters rich in either T10 or T11+ RA in an atherogenic diet for 12 weeks decreased rabbit AT lipogenesis, with a more marked effect of the T11+RA diet, but had no effect on liver lipogenesis.
