International audience; Vitamin B12 (cobalamin) is a key determinant of S-adenosyl methionine (SAM)-dependent epigenomic cellular regulations related to methylation/acetylation and its deficiency produces neurodegenerative disorders by elusive mechanisms. Sirtuin 1 deacetylase (SIRT1) triggers cell response to nutritional stress through endoplasmic reticulum (ER) stress. Recently, we have established a N1E115 dopaminergic cell model by stable expression of a transcobalamin-oleosin chimera (TO), which impairs cellular availability of vitamin B12, decreases methionine synthase activity and SAM level, and reduces cell proliferation. In contrast, oleosin-transcobalamin chimera (OT) does not modify the phenotype of transfected cells. Presently, the impaired cellular availability of vitamin B12 in TO cells activated irreversible ER stress pathways, with increased P-eIF-2α, P-PERK, P-IRE1α, ATF6, ATF4, decreased chaperon proteins and increased pro-apoptotic markers, CHOP and cleaved caspase 3, through reduced SIRT1 expression and consequently greater acetylation of heat-shock factor protein 1 (HSF1). Adding either B12, SIRT1, or HSF1 activators as well as overexpressing SIRT1 or HSF1 dramatically reduced the activation of ER stress pathways in TO cells. Conversely, impairing SIRT1 and HSF1 by siRNA, expressing a dominant negative form of HSF1, or adding a SIRT1 inhibitor led to B12-dependent ER stress in OT cells. Addition of B12 abolished the activation of stress transducers and apoptosis, and increased the expression of protein chaperons in OT cells subjected to thapsigargin, a strong ER stress stimulator. AdoX, an inhibitor of methyltransferase activities, produced similar effects than decreased B12 availability on SIRT1 and ER stress by a mechanism related to increased expression of hypermethylated in cancer 1 (HIC1). Taken together, these data show that cellular vitamin B12 has a strong modulating influence on ER stress in N1E115 dopaminergic cells. The impaired cellular availability in vitamin B12 induces irreversible ER stress by greater acetylation of HSF1 through decreased SIRT1 expression, whereas adding vitamin B12 produces protective effects in cells subjected to ER stress stimulation.

Improved knowledge on overland flow (OF) generation and its dynamics (i.e. spatial and temporal variations) is essential to understand catchment hydrology, a prerequisite for better water resources and soil management. In this study, our main objective was to quantify the dynamics of OF during rainfall events and to assess its main factors of control. The research study was undertaken in an agricultural 23-ha catchment of a communal pasture in KwaZulu-Natal (South Africa) experiencing Mediterranean climate and with variations of soil, topography and vegetation conditions. The dynamics of OF was evaluated during three rainfall seasons (2007 to 2010) by using 1x1-m(2) microplots (n=15) located at five landscape positions. At each location, a microplot was equipped with an automatic tipping bucket linked to a logger to estimate the delay between the start of the rain and the start of OF [i.e. the time to runoff initiation (TRI)]. Multivariate analysis was applied to the total OF and TRI data and the information on selected environmental factors (rainfall characteristics; soil type; soil clay content, Clay; proportion of the soil surface covered by vegetation, Cov; proportion of the soil surface covered by crusting, Crust; mean slope gradient, S; soil bulk density, (b); soil water tension at different depths, SWT). The average OF rate over the 3-year study period varied 2.3-fold across the catchment (from 15% footslope to 35% backslope), whereas the average TRI varied by a 10.6-fold factor (between 0.6min at bottomland and 6.4min at footslope). TRI temporal variations correlated the most with event duration (r=0.8) and cumulative amount of rainfall since the onset of the rainy season (r=-0.47), whereas TRI spatial variations were controlled the most by Crust (-0.97

The acceleration of soil erosion by water in most regions of the world in response to the anthropogenic modification of landscapes is a serious threat to natural ecosystem functionalities because of the loss of invaluable constituents such as soil particles and organic carbon (OC). While soil OC erosion is likely to be a major component of the global C cycle, water erosion-induced CO2 emissions remain uncertain. In this study, our main objective was to compare the release of CO2 from eroded topsoils and from the sediments exported by diffuse erosion during an entire rainy season. Conventional tillage (CT) and no-tillage (NT) maize treatments were considered in an attempt to set up best management practices to mitigate gaseous OC losses from agricultural soils. The study was conducted in the KwaZulu-Natal province in South Africa, whereas in many other areas of the developing world, erosion is severe and crop residue scarcity is the main challenge. CO2 emissions from undisturbed 0-0.02 m soil samples collected within 2.25 m x 10 m runoff plots and from exported sediments by water erosion, were evaluated continuously at the laboratory over a 140-day period and compared to soil OC stocks. NT significantly reduced CO2 emissions from both soils and sediments. Overall NT, which exhibited a greater carbon density than CT (17.70 vs 13.19 kg C m(-3)), reduced soil gaseous emissions by 4.4% (10.40 vs 10.88 gCO(2)-C m(-2), P < 0.05) but had a much greater impact on the release of CO2 from eroded sediments (0.185 vs 0.778 gCO(2)-C m(-2)), which corresponded to a 76.3% decrease. For CT, cumulative 141-day emissions were, 19% greater in sediments (0.048 g CO2-C g C-1) compared to soils (0.040 gCO(2)-C g C-1), while for NT, emissions were 33% lower in sediments (0.024 g CO2-C g C-1) compared to soils (0.032 g CO2-C g C-1), these differences being significant at P < 0.05. The lower erosion-induced CO2 emissions under NT could be explained by a high soil aggregate stability (mean weight diameter of 2.29 +/- 0.05 mm for NT vs 1.59 +/- 0.07 mm for CT, P < 0.05) and the associated enhanced protection of SOC from the decomposers. These results on a land management control of water erosion-induced CO2 emissions, might allow improving the impact of terrestrial ecosystems on greenhouse gases concentration in the atmosphere and associated climate change.

The current increase in the global demand for food and fresh water and the associated land use changes or misuses exacerbate water erosion which has become a major threat to the sustainability of the soil and water resources. Soil erosion by rainfall and runoff is a natural and geologic phenomenon, and one of the most important components of the global geochemical cycle. Despite numerous studies on crop lands, there is still a need to quantify soil sheet erosion (an erosion form that uniformly removes fertile upper soil horizons) under grasslands and to assess the factors of the environment that control its spatial variation. For that purpose, fifteen 1 m(2) micro-plots installed within a 23 ha catchment under pasture in the sloping lands of KwaZulu-Natal (South Africa) were monitored during the 2007-2008 rainy season to evaluate runoff (R) and sediment losses (SL). Soil losses computed from the 37 rainfall events with soil erosion averaged 6.45 ton ha(-1) year(-1) with values from 3 to 13 ton ha(-1) year(-1). SL were significantly correlated with the proportion of soil surface coverage by the vegetation (P < 0.01) whereas the slope gradient, and soil characteristics such as bulk density or clay content were not correlated. R and SL increased as the proportion of soil surface coverage decreased and this trend was used to predict the spatial variations of sheet erosion over the 23 ha catchment. Greater sheet erosion occurred at the catchment plateau and at the vicinity of gully head cuts probably in relation to regressive erosion. Mitigating sheet erosion would require an appropriate management of the soil cover through appropriate management of cattle grazing, especially at places where "natural" erosion is likely to occur.

The establishment of a catchment monitoring network is a process, from the inception of the idea to its implementation, the latter being the construction of relevant gauging structures and installation of the various instruments. It is useful that the local communities and other stakeholders are involved and participate in such a process as was realised during the establishment of the hydrological monitoring network in the Potshini catchment in the Bergville district in the KwaZulu-Natal Province in South Africa. The paper illustrates the participatory application of various methods and techniques for establishing a hydrological monitoring network, in a small rural inhabited catchment, to monitor hydrological processes at both field and catchment scale for research purposes in water resources management. The authors conclude that the participation of the local community and other stakeholders in catchment monitoring and instilling the sense of ownership and management of natural resources to the local communities needs to be encouraged at all times. Success stories in water resources management by local communities can be realized if such a process is integrated with other development plans in the catchment at all forums with due recognition of the social dynamics of the communities living in the catchment.

The knowledge of water flow pathways and residence times in a catchment are essential for predicting the hydrological response to a rain storm event. Different experimental techniques are available to study these processes, which are briefly reviewed in this paper. To illustrate this, recent findings from the Weatherley catchment a 1.5 km2 semiarid headwater in South-Africa, are reported in this paper. Beside classical hydrometric measurements of precipitation and runoff different experimental techniques were applied to explore flow paths (i.e. soil moisture and groundwater measurements, natural tracers, and 2-D electrical resistivity tomographies (ERT)).
