1. Bonding-Directed Crystallization of Ultra-Long One-Dimensional NbS3 van der Waals Nanowires.

Abstract

The rediscovery of one-dimensional (1D) and quasi-1D (q-1D) van der Waals (vdW) crystals ushered the realization of nascent physical properties in 1D that are suitable for applications in photonics, electronics, and sensing. However, despite renewed interest in the creation and understanding of the physical properties of 1D and q-1D vdW crystals, the lack of accessible synthetic pathways for growing well-defined nanostructures that extend across several length scales remains. Using the highly anisotropic 1D vdW NbS3-I crystal as a model phase, we present a catalyst-free and bottom-up synthetic approach to access ultralong nanowires, with lengths reaching up to 7.9 mm and with uniform thicknesses ranging from 13 to 160 nm between individual nanowires. Control over the synthetic parameters enabled the modulation of intra- and interchain growth modalities to selectively yield only 1D nanowires or quasi-2D nanoribbons. Comparative synthetic and density functional theory (DFT) studies with a closely related nondimerized phase, ZrS3, show that the unusual preferential growth along 1D can be correlated to the strongly anisotropic bonding and dimeric nature of NbS3-I.

Abstract

BACKGROUND: The Latinx population experiences some of the highest rates of chronic disease, including obesity and type II diabetes. Such conditions may be especially burdensome in rural Latinx communities that often face barriers to accessing disease prevention resources and public health programs. METHODS: Diverse stakeholders (i.e., patients, community members, system of healthcare clinics, community food bank) tailored an existing cookbook, based on the U.S. Department of Agriculture MyPlate healthy eating and dietary guidelines, for local ingredients, health literacy, and language for rural Latinx and Indigenous Latin Americans. The cookbook recipes were disseminated widely via virtual cooking demonstrations, food distribution events, and social media. Pre- and posttest surveys were used to assess changes in diabetes knowledge measured by the 24-item American Diabetes Association Diabetic Knowledge Questionnaire and confidence in dietary behavior change over time measured by 4 questions of the 17-item Mediterranean Diet Index. A mixed effects, repeated measures analysis was conducted with gender ID, age range and educational attainment included as covariates and assessment interval as the predictor (pretest vs posttest) and change in confidence about adhering to four specific components of the Mediterranean diet. Focus groups elicited information on participants motivation and ability to use the recipes and eat healthy foods following the virtual cooking demonstration participation. RESULTS: A total of 20 virtual cooking demonstrations were conducted and 60 participants completed a pretest survey and 54 a posttest survey, a subsample (n = 19) participated in one of three focus groups. Most participants were female, identified as Latinx/Hispanic, were between the ages of 40-49, and spoke Spanish. 17% identified as Indigenous Latin American specifically as Purépecha, an indigenous group from Michoacán, Mexico. Survey and focus group findings indicated at posttest an increase in diabetes knowledge among participants with no prior diagnosis of chronic health conditions and more confidence in limiting sugary beverages and refined wheat pasta/white rice among indigenous participants. Focus group discussions explicated the quantitative findings. CONCLUSION: This study brought together patients and key stakeholders committed to addressing the social determinants of health and it mobilized the community to develop culturally vetted health education materials. The findings indicate the need for increased access to evidence-based nutrition education and to culturally appropriate food products that can be easily incorporated into daily food preparation.

7. Hope, Optimism, Gratitude, and Wellbeing among Health Professional Minority College Students

Abstract

Objectives: To (1) describe the level of hope, optimism, and gratitude in a sample of minority health professional college students. (2) To examine the association between hope, optimism, and gratitude with wellbeing domains. Participants: One hundred and thirty-two (n = 132) college students from the Nursing, Medicine, and Allied Health programs. Methods: Cross-sectional survey study assessed wellbeing across the five domains of positive emotion, engagement, relationships, meaning, and accomplishment. Results: Mean and Standard Deviation on hope, optimism, and gratitude were 50.2 ± 5.6; 21.7 ± 4.34; and 36.3 ± 5.35, respectively. Hope, optimism, and gratitude were positively associated with domains of wellbeing, controlling for gender and age. Conclusion: Students maintained a positive outlook in life. Hopeful, optimistic, and grateful students experienced positive emotion, were more engaged in their daily activities, had more supportive relationships, had a better sense of direction in life, and more often accomplished their goals. Interpretations and future directions are discussed.

Abstract

DNA nanostructures are a promising tool to deliver molecular payloads to cells. DNA origami structures, where long single-stranded DNA is folded into a compact nanostructure, present an attractive approach to package genes; however, effective delivery of genetic material into cell nuclei has remained a critical challenge. Here, we describe the use of DNA nanostructures encoding an intact human gene and a fluorescent protein encoding gene as compact templates for gene integration by CRISPR-mediated homology-directed repair (HDR). Our design includes CRISPR-Cas9 ribonucleoprotein binding sites on DNA nanostructures to increase shuttling into the nucleus. We demonstrate efficient shuttling and genomic integration of DNA nanostructures using transfection and electroporation. These nanostructured templates display lower toxicity and higher insertion efficiency compared to unstructured double-stranded DNA templates in human primary cells. Furthermore, our study validates virus-like particles as an efficient method of DNA nanostructure delivery, opening the possibility of delivering nanostructures in vivo to specific cell types. Together, these results provide new approaches to gene delivery with DNA nanostructures and establish their use as HDR templates, exploiting both their design features and their ability to encode genetic information. This work also opens a door to translate other DNA nanodevice functions, such as biosensing, into cell nuclei.

Abstract

BackgroundIndividual diet components and specific dietary regimens have been shown to impact the gut microbiome.ObjectivesHere, we explored the contribution of long-term diet by searching for dietary patterns that would best associate with the gut microbiome in a population-based cohort.MethodsUsing a priori and a posteriori approaches, we constructed dietary patterns from an FFQ completed by 1800 adults in the American Gut Project. Dietary patterns were defined as groups of participants or combinations of food variables (factors) driven by criteria ranging from individual nutrients to overall diet. We associated these patterns with 16S ribosomal RNA-based gut microbiome data for a subset of 744 participants.ResultsCompared to individual features (e.g., fiber and protein), or to factors representing a reduced number of dietary features, 5 a posteriori dietary patterns based on food groups were best associated with gut microbiome beta diversity (P ≤ 0.0002). Two patterns followed Prudent-like diets-Plant-Based and Flexitarian-and exhibited the highest Healthy Eating Index 2010 (HEI-2010) scores. Two other patterns presented Western-like diets with a gradient in HEI-2010 scores. A fifth pattern consisted mostly of participants following an Exclusion diet (e.g., low carbohydrate). Notably, gut microbiome alpha diversity was significantly lower in the most Western pattern compared to the Flexitarian pattern (P ≤ 0.009), and the Exclusion diet pattern was associated with low relative abundance of Bifidobacterium (P ≤ 1.2 × 10-7), which was better explained by diet than health status.ConclusionsWe demonstrated that global-diet a posteriori patterns were more associated with gut microbiome variations than individual dietary features among adults in the United States. These results confirm that evaluating diet as a whole is important when studying the gut microbiome. It will also facilitate the design of more personalized dietary strategies in general populations.

Abstract

While residents of Los Angeles County appear to have access to seemingly endless postsecondary opportunities within their proximity, the geography of opportunity for accessible options of higher education leading to bachelor’s degrees in the area requires further exploration. This case study explores the spatial contexts and 3-year transfer successes of students attending Sunshine Community College (SCC). Spatial quantitative methods and Geographic Information Systems (GIS) software are used to combine Census American Community Survey (ACS) data on educational attainment with student-level data from the institution and the characteristics of surrounding institutions of higher education from IPEDS to explore the relationship between transfer completion and the geography of college opportunity for community college students. An index was calculated to represent the geography of college opportunity (GCO) surrounding each student within their 5-mile spatial contexts and descriptive and inferential analyses run to inform our understanding of the concept of geography of opportunity and its relation to community college transfer. Several variables calculated in the creation of the GCO Index show a statistically significant negative weak relationship with transfer. For instance, students who are surrounded by fewer institutions of higher education have higher 3-year transfer rates and students who have more institutions of higher education in their vicinity have lower transfer rates. The inclusion of for-profit institutions help account for such findings and, as such, recommendations for future research considerations include exercising caution when including for-profit institutions in the analysis of geography of college opportunity. The results of this study will inform ways to better support students in their persistence toward overall educational goals of transfer and degree attainment and to enhance support structures for students from a range of spatial contexts seeking timely and cost-effective completion outcomes.
