2. Research Image Management Practices Reported by Scientific Literature: An Analysis by Research Domain

Research data management is essential for safeguarding and prospecting data generated in a scientific context. Specific issues arise regarding data in image format, as this data typology poses particular challenges and opportunities; however, not much attention has been given to data as images. We reviewed 109 articles from several research domains where images were used either as data or metadata to understand how researchers specifically deal with this data format, and what are your habits and behaviors. We use the Web of Science (WoS), considering its five main areas of research. We included in the initial corpus the most relevant articles by research domain, selecting the ten most cited articles in WoS, by year, between 2010 and 2021. The selected articles should be in English and in open access. The results found that images have been used in scientific works numerous times, but, unfortunately, few are those in which they are the central element of the study. Photography is the type of image most used in most domains. In terms of the instruments used, the Technology and Life Sciences and Biomedicine domains use the microscope more, while the Arts and Humanities and Physical Sciences domains use the camera more. We found that the images are mostly produced in the context of the project, rather than reused by third parties. As for their collection scenario, these are mostly produced/used in a laboratory context. The overwhelming majority of the images present in the articles are digital, and only a small part is analog. We verify that Arts and Humanities are more likely to perform qualitative types of analyses, while Life Sciences and Biomedicine overwhelmingly use quantitative analyses. As for the issues of sharing and depositing, Life Sciences and Biomedicine is the domain that stands out the most in the tasks of depositing and sharing images. It was found that the licenses of a project are intrinsically related to the motivations for sharing results with third parties. Description, a fundamental step in the data management process, is neglected by a large number of researchers. The images are mostly not described or annotated and when this happens, researchers don’t provide much detail about this.

Face às prováveis mudanças climáticas afetando as áreas agricultáveis do planeta, foi feita uma revisão bibliográfica sobre os efeitos do aumento da temperatura no metabolismo da planta de batata, enfatizando suas consequências no desenvolvimento vegetativo e na produtividade da cultura. Embora a ênfase maior desse artigo se prenda ao efeito do clima na redução de alimentos, alerta-se ainda para o risco de erosão genética pelo desaparecimento de espécies silvestres, de grande importância para programas de melhoramento genético. A partir dessas informações e de dados climáticos das regiões produtoras publicados em literatura nacional, foi realizada uma análise prospectiva da produção de batata no Brasil. Para tal, tomou-se como base o aumento da temperatura de até 5ºC até o final deste século e uma faixa ótima de produtividade da batata de 10ºC a 25ºC, conforme indicado em literatura especializada. Foram selecionados seis municípios representando as regiões geográficas onde se cultiva a batata: São Joaquim-SC; Guarapuava-PR; Cristalina-GO; Mucugê-BA; Vargem Grande do Sul-SP e Araxá-MG. Para municípios com prevalência de temperaturas mais baixas, como São Joaquim, o aumento de temperatura preconizado não deverá restringir a produção, porém serão necessárias adequações de épocas de plantio. Por outro lado, a situação é preocupante em regiões e épocas de cultivo sujeitas a temperaturas altas, como Mucugê e Cristalina, onde o cultivo deverá se restringir a poucos meses do ano, contrastando com o plantio contínuo, durante o ano todo, ora sendo realizado. O intuito deste artigo é alertar, e não alarmar, a cadeia produtiva de batata no sentido de se preparar para os ajustes fitotécnicos visando a preservar a bataticultura nacional face a um eventual aquecimento global. Considerações sobre o melhoramento genético para atenuar, pelo menos em parte, esta situação é discutida. The future of the potato production in Brazil is discussed upon prediction of temperature rise due to global warming. A literature review was carried out on the effects of high temperatures on the potato plant metabolism, and their consequences upon vegetative development and yield. Although higher emphasis was devoted to the climate effect on food production, the risk of genetic erosion by disappearance of wild species is stressed, with consequences to the future of plant breeding. Based on this information and on climate data from the main growing regions published on Brazilian documents, we carried out a prospective analysis of the potato production in Brazil. For that, a temperature rise of up to 5ºC was considered to the end of the century, and the range of 10ºC to 25ºC as the optimum for potato growth, according to specialized literature. For the study, six counties, representative of the main growing regions, were selected: São Joaquim, Santa Catarina State; Guarapuava, Paraná State; Cristalina, Goiás State; Mucugê, Bahia State; Vargem Grande do Sul, São Paulo State and Araxá, Minas Gerais State. For cooler counties, such as São Joaquim, major drawbacks on potato production are not expected, even though adjustments in planting season should be required. However, cropping on those counties subject to high temperatures which today allow potato production all year round, like Mucugê, and Cristalina, is expected to be restricted to few months of the year. The aim of this article was to alert, not alarm, the Brazilian potato chain in order to promote the management changes to preserve the crop in case the expected temperature rise comes true. In addition, the role of plant breeding to counteract the negative effects of high temperature is discussed.

The δ15N composition of bottom-feeding fish (iliophagous = Apareiodon affinis, Cyphocharax nagelii, Prochilodus lineatus, Steindachnerina brevipinna and S. insculpta; detritivorous = Loricariichthys platymetopon and Liposarcus anisitsi; benthophagous = Satanoperca pappaterra and Hoplosternum littorale) and their primary food sources were investigated in the upper Paraná River floodplain during rainy seasons in different environments (lotic and lentic). Two hypotheses were tested: i) that the trophic position and isotopic values of the investigated organisms (fish and food resources) vary spatially; and ii) that trophic position and isotopic compositions differ among iliophagous, detritivorous and benthophagous fish. C4 macrophytes, periphyton and phytoplankton were isotopically different in sites analyzed. Significant isotopic differences occurred in the species of each trophic category. Spatial differences were observed in the isotopic composition of P. lineatus and L. platymetopon, whose values were more enriched in the Paraná River and Pau Véio Lake. Significant spatial differences in trophic position were observed for L. platymetopon and H. littorale, which presented the highest values in the Paraná and Baía rivers, respectively. Trophic positions were significantly different among the species that composed each trophic category. These findings demonstrate that in energy-flow studies in detrital food chains generalizations concerning the grouping of fish into trophic categories and/or habitats should only be carried out after careful investigations of the local/specific trophic dynamics of the organisms. A composição de δ15N de peixes exploradores de fundo (iliófagos = Apareiodon affinis, Cyphocharax nagelii, Prochilodus lineatus, Steindachnerina brevipinna e S. insculpta; detritívoros = Loricariichthys platymetopon e Liposarcus anisitsi; e bentófagos = Satanoperca pappaterra e Hoplosternum littorale) e suas fontes alimentares foram investigadas na planície de inundação do alto rio Paraná durante a estação de chuvas em ambientes lóticos e lênticos. Duas hipóteses foram testadas: i) que a posição trófica e os valores isotópicos dos organismos investigados (peixes e fontes alimentares) variam espacialmente; e ii) que a posição trófica e a composição isotópica diferem entre os peixes iliófagos, detritívoros e bentófagos. Macrofitas C4, perifíton e fitoplâncton foram isotopicamente diferentes entre os locais analisados. Diferenças isotópicas ocorreram entre as espécies de cada categoria trófica. Diferenças espaciais foram observadas na composição isotópica de P. lineatus e L. platymetopon, as quais tiveram valores mais enriquecidos no rio Paraná e ressaco do Pau Véio. Diferenças espaciais significativas nas posições tróficas foram verificadas para L. platymetopon e H. littorale, as quais apresentaram os maiores valores nos rios Paraná e Baía, respectivamente. As posições tróficas foram significativamente diferentes entre as espécies que compuseram cada categoria trófica. Estes resultados demonstram que em estudos de fluxo de energia em cadeias alimentares detritais generalizações a respeito do agrupamento de peixes em categorias tróficas e/ ou habitats devem ser conduzidos somente após investigações criteriosas da dinâmica trófica local/ específica dos organismos.

11. The role of grapevine leaf morphoanatomical traits in determining capacity for coping with abiotic stresses: a review

Worldwide, there are thousands of Vitis vinifera grape cultivars used for wine production, creating a large morphological, anatomical, physiological and molecular diversity that needs to be further characterised and explored, with a focus on their capacity to withstand biotic and abiotic stresses. This knowledge can then be used to select better adapted genotypes in order to help face the challenges of the expected climate changes in the near future. It will also assist grape growers in choosing the most suitable cultivar(s) for each terroir; with adaptation to drought and heat stresses being a fundamental characteristic. The leaf blade of grapevines is the most exposed organ to abiotic stresses, therefore its study regarding the tolerance to water and heat stress is becoming particularly important, mainly in Mediterranean viticulture. This review focuses on grapevine leaf morphoanatomy - leaf blade form, leaf epidermis characteristics (cuticle, indumentum, pavement cells and stomata) and anatomy of mesophyll - and their adaptation to abiotic stresses. V. vinifera xylem architecture and its adaptation capacity when the grapevine is subjected to water stress is also highlighted since grapevines have been observed to exhibit a large variability in responses to water availability. The hydraulic properties of the petiole, shoot and trunk are also reviewed. Summarising, this paper reviews recent advances related to the adaptation of grapevine leaf morphoanatomical features and hydraulic architecture to abiotic stresses, mainly water and heat stress, induced primarily by an ever-changing global climate.

Assessing sustainability of the wine industry requires improved characterization of its environmental impacts, namely in terms of water use. Therefore, quantification of water inputs and wastewater (WW) outputs is needed to highlight inefficiencies in wine production and related consequences for the environment. Water use and WW generation in irrigated viticulture and oenology remains insufficiently quantified for dry Mediterranean regions (e.g. South Portugal). This paper is focused on wine production under warm and dry climate conditions in the winegrowing region of Alentejo (South Portugal). This region experiences increasingly dry conditions, while the irrigated area keeps expanding, which puts exacerbates the pressure on existing local and regional water resources. Additionally, more erratic variation in climate conditions and the tendency for increasingly extreme climate events (e.g. heat waves) pose more challenges to Alentejo’s wine sector. We conclude that quantitative information on water use and management is not always easy to obtain or access, which hinders improved strategies and/or policies for water use at farm, winery and region-level. Up-to-date statistics and robust metrics can help to better characterize water use and WW flows for Alentejo’s wine region, while optimizing management in vineyards and wineries, in companies and region-wide. 16. Physiological and agronomical responses to environmental fluctuations of two Portuguese grapevine varieties during three field seasons

Extensive agricultural losses are attributed to heat, often combined with drought. These abiotic stresses occur in the field simultaneously, namely in areas with Mediterranean climate, where grapevine traditionally grows. The available scenarios for climate change suggest an increase in the frequency of heat waves and severe drought events in summer, also affecting the South of Portugal. In this work we monitored several production-related parameters and evaluated the state of the oxidative stress response apparatus of two grapevine varieties, Touriga Nacional (TN) and Trincadeira (TR), with and without irrigation, during three field seasons (2010 to 2012). Overall, results point to a high correlation of most yield and stress-associated parameters with the specific characteristics of each variety and to each season rather than the irrigation treatments. In the season with the driest winter, 2012, the lack of irrigation significantly affected yield in TR, while in the two other seasons the impact of the irrigation regime was much lower. In 2012, the yield of TN was affected by environmental conditions of the previous season. The irrigation treatments significantly affected berry size rather than quality.

17. Leaf morphoanatomy traits in white grapevine cultivars with distinct geographical origin

This study aims to compare the leaf morphoanatomical characteristics of seven Vitis vinifera subsp. vinifera white cultivars with different geographical origin, grown side by side at the same “terroir”. The leaf morphoanatomical traits analyzed under light and scanning electron microscopy (SEM) showed large variability among genotypes. Significant differences between cultivars were observed in single leaf area, specific leaf area, leaf density and in the thickness of cuticle, upper and lower epidermal cells and mesophyll. Leaf dry mass per area presented a significant positive correlation with leaf density but showed no correlation with leaf thickness, results that can be explained by the quite different mesophyll structure, epidermal cells and cuticle thickness. ‘Alvarinho’, ‘Encruzado’, ‘Macabeu’ and ‘Viosinho’ were the cultivars with the highest thickness of cuticle and mesophyll tissues. Under SEM magnification three types of stomata were identified: sunken, at the same level and raised above, with the same level type presenting the higher percentage in all cultivars. Stomata density presented significant differences between cultivars, with ‘Macabeu’ showing the highest value and ‘Alvarinho’ and ‘Arinto’ the lowest ones. The hairs on the lower surface presented a similar woolly aspect in all cultivars. Calcium oxalate crystals, raphids and druses were visible and widely distributed in the parenchyma tissues. The observed differences in leaf traits among genotypes suggest a major role of leaf anatomy in determining grapevine capacity for coping with different environmental conditions.

The association of maghemite–rhodium citrate (MRC) nanoparticles (NPs) has the potential to increase specificity of the cytotoxic action. However, the interaction of these NPs with cells, their uptake mechanism, and subcellular localization need to be elucidated. This work evaluates the uptake mechanism of MRC NPs in metastatic and nonmetastatic breast cancer-cell models, comparing them to a nontumor cell line. MRC NPs uptake in breast cancer cells was more effective than in normal cells, with regard to both the amount of internalized material and the achievement of more strategic intracellular distribution. Moreover, this process occurred through a clathrin-dependent endocytosis pathway with different basal expression levels of this protein in the cell lines tested. This paper presents results of mechanical tests on a prototype of an innovative structural strengthening in form of self-monitoring fabric. Smart textile employs carbon fibers conductivity for measuring strains while monitoring changes of electric resistance under increasing load. A general solution was tested in a series of calibrating tests on strengthening of small size concrete slabs. Promising results of simple specimen, has encouraged the research team to perform the next tests using mastered carbon fibre reinforced fabric. Main tests were performed on natural scale RC beam. Smart textile proved its efficiency in both: strengthening and monitoring of strains during load increase. New strengthening proposal was given 10% increase of loading capacity and the readings of strain changes were similar to those obtained in classical methods. In order to calibrate the prototype and to define range limits of solution usability, textile sensor was tested in areas of large deformations (timber beam) and aswell as very small strains (bridge bearing block). In both cases, the prototype demonstrated excellent performance in the range of importance for structural engineering. 20. Leaf morphoanatomy of four red grapevine cultivars grown under the same terroir

This study compare leaf morphoanatomical characteristics of four red cultivars - ‘Touriga Nacional’, ‘Trindadeira’, ‘Cabernet Sauvignon’ and ‘Syrah’ -, grown side by side at the same terroir. The analyzed leaf traits, under light and scanning electron microscopy, showed large variability among genotypes. ‘Trincadeira’ has the biggest single leaf area and ‘Cabernet Sauvignon’ the smallest one. ‘Touriga Nacional’ showed the lowest leaf dry weight and ‘Trincadeira’ the highest one, nonetheless there was no significantly differences in leaf dry mass per area and in leaf density. Leaf dry mass per area was positively correlated with leaf density but showed no correlation with leaf thickness. The French genotypes presented higher thickness of the leaf anatomical traits than the two Portuguese ones. ‘Trincadeira’ showed significantly highest stomata density while the other cultivars showed no significant differences among them. The analyses of the three types of stomata revealed that ‘Trincadeira’ has the lower percentage of raised above and the highest percentage of sunken stomata while ‘Cabernet Sauvignon’ showed the opposite behaviour. The hairs on the lower surface presented a similar woolly aspect in all cultivars. The possible role of leaf morphoanatomical characteristics in determining the cultivars adaptation to abiotic stresses is suggested and discussed.

Climate change introduced new challenges to vinegrowers in the Mediterranean areas such as the hot and dry winegrowing region of Alentejo, south Portugal. Warmer and drier conditions are harmful for grape yield and berry quality attributes and for vine’s longevity, mainly when optimal thresholds are exceeded. Therefore winegrowers are forced to move from rainfed to irrigated production systems making Alentejo’ wine production strongly based on available water resources for irrigation. This work aims to review and discuss ecophysiological and agronomical data obtained in irrigation trials set up at different terroirs of Alentejo. In the last four decades, classical bioclimatic temperature-based indices showed a significantly positive trend, while the dryness index present a negative trend over time. Furthermore, ecophysiological data collected in deficit irrigation experiments are reviewed and discussed, focusing on the effects of drought and heat stress on vigour, yield and berry composition. Emphasis is given on the indirect effects of leaf senescence on cluster exposure and consequences on berry temperature and composition. In order to promote the sustainability and quality of wine production in these hot and dry terroirs short- to long-term adaptation measures are suggested. The limitations and risks of using deficit irrigation during heat waves are also underlined.

The ecology of anopheline species (Diptera, Culicidae) was studied in the vicinity of the Serra da Mesa Reservoir, State of Goiás, Brazil. Climatic factors and frequency of anopheline populations were analyzed. Bimonthly human-bait and Shannon trap captures were conducted for 36 consecutive months (January 1997 through December 1999). A total of 5,205 adult anophelines belonging to five species were collected. Anopheles darlingi was the most frequently collected anopheline (61.4%), followed by An. albitarsis s.l. (35.4%), An. triannulatus. (2.5%), An. oswaldoi (0.4%), and An. evansae (0.2%). The water level and vegetation along the banks of the reservoir were crucial to the frequency of the various anopheline species. Climatic factors had a secondary influence. The reservoir's water-level stability, increased frequency of An. darlingi, and the arrival of gold prospectors were responsible for the increase in malaria cases.

OBJETIVO: Observar detalhes morfológicos de ovos de Haemagogus leucocelaenus visualizados pela primeira vez por microscopia eletrônica de varredura (MEV) e realizar morfometria das principais estruturas. MÉTODOS: Foram utilizados ovos de Hg. leucocelaenus provenientes de fêmeas capturadas na Reserva Biológica do Tinguá, RJ, sendo parte destinada à eclosão e outra ao processamento de MEV, dos quais três foram submetidos à análise morfométrica. O material foi fixado em glutaraldeído 2,5% e pós-fixado em tetróxido de ósmio 1%, ambos em tampão cacodilato de sódio 0.1M, pH 7.2, processado e observado ao MEV Jeol 5310. Medições foram realizadas com o auxílio do software de análise Semafore. RESULTADOS: Os ovos apresentaram contorno elíptico com aproximadamente 574 µm de comprimento e 169 µm de largura, sendo o índice do ovo (l/wratio) 3,39 µm. O exocório é extremamente regular, possuindo ornamentação hexagonal e algumas vezes pentagonal. Nas células coriônicas, observaram-se tubérculos simetricamente dispostos com relação ao eixo longitudinal, e, no interior delas, tubérculos menores, individualizados, dispostos na periferia, e poucos agrupados no centro. A superfície do retículo coriônico não apresentou rugosidades. O aparelho micropilar apresenta colar proeminente, contínuo, com disco micropilar bem evidente. CONCLUSÕES: A ornamentação do exocório apresenta diferenças em relação aos tubérculos das células coriônicas e ao retículo coriônico externo entre os ovos de Hg. leucocelaenus comparados aos ovos de Hg. janthinomys e Hg. equinus, bem como com relação aos de Aedes aegypti, Ae. albopictus e Ae bahamensis.

Estabelecemos a influência exercida pela cobertura vegetal de quatro diferentes biótopos em áreas do Parque Nacional do Iguaçu (PNI) sobre a fauna local de mosquitos. Realizaram-se capturas bimestrais, em isca humana e armadilha Shannon, em três diferentes períodos do dia, em ambiente silvestre e domiciliar, durante 24 meses consecutivos. Dentre os 20.273 espécimes de fêmeas adultas capturadas, pertencentes a 44 espécies, Ochlerotatus serratus (10,3%), Haemagogus leucocelaenus (9,7%), Mansonia titillans (9,6%) e Chagasia fajardoi (8,8%) foram predominantes. Anopheles cruzii, Runchomyia theobaldi, Wyeomyia aporonoma e Wy. confusa ocorreram exclusivamente em áreas com vegetação bem preservada e densa configuração. Culex nigripalpus, Oc. pennai, Oc. serratus, Sabethes purpureus e Sa. albiprivus foram capturados nos três biótopos essencialmente silvestres. Na mata no entorno da represa foram capturadas principalmente An. albitarsis s.l., An. galvaoi, An. evansae, An. fluminensis, Coquillettidia venezuelensis, Cq. juxtamansonia, Wy. quasilongirostris e Onirion personatum. As espécies que apresentaram maiores incidências na área sob ação antrópica foram Ch. fajardoi, Cq. fasciolata, Cq. nitens e Ma. titillans.

This paper lays out a theoretical and analytical framework for researching and reflecting on the peacebuilding role of education in conflict-affected contexts. The 4Rs framework recognizes that working toward "positive peace" (Galtung 1976, 1990) requires working toward peace with social justice and reconciliation, challenging dominant "security-first" and "liberal peace" models, and gaining a better understanding of how education might support these processes in building sustainable and peaceful postconflict societies. The 4Rs framework combines dimensions of recognition, redistribution, representation, and reconciliation to explore what sustainable peacebuilding might look like through a social justice lens. The paper addresses the cultural translation of these concepts, highlighting the need for locally embedded interpretations. Rather than a fixed theoretical model, the 4Rs approach is designed as a heuristic device that promotes a dialogue among key stakeholders on the dilemmas and challenges in the field of education in emergencies. We highlight the application of a 4Rs framework through a recent case study of Myanmar, which demonstrates both the interrelated connections and the tensions between the different "Rs." Finally, we reflect on the challenges and limitations of the approach, and the tasks ahead.

OBJETIVO: Estabelecer a influência exercida pelos fatores climáticos na freqüência mensal da fauna de mosquitos em áreas do Parque Nacional da Serra da Bocaina, Vale do Paraíba, SP. MÉTODOS: Foram realizadas capturas mensais por meio de isca humana, em três diferentes períodos do dia, durante 24 meses consecutivos (janeiro de 1991 a dezembro de 1992). RESULTADOS: Foram capturados 11.808 espécimes adultos pertencentes a 28 espécies. Anopheles cruzii, Runchomyia reversa e Ru. frontosa foram as espécies mais presentes em todos os meses do ano; An. cruzii ocorreu preferencialmente de outubro a fevereiro; Ru. reversa e Ru. frontosa, em setembro; An. lutzi, Chagasia fajardoi, Coquillettidia chrysonotum, Aedes serratus, Trichoprosopon simile, Wyeomyia theobaldi, Ru. humboldti e Ru. theobaldi, pertencentes ao segundo grupo de espécies mais abundantes, ocorreram preferencialmente nos meses mais quentes, úmidos e chuvosos. A temperatura e as precipitações pluviométricas influenciaram positivamente na incidência da maioria das espécies. CONCLUSÕES: A cobertura vegetal de Mata Atlântica bem preservada, as precipitações pluviométricas e a temperatura foram determinantes para a incidência da fauna de mosquitos no parque. Os meses mais chuvosos, úmidos e quentes contribuíram para o aumento da diversidade e da densidade. O período de setembro a março foi o mais favorável. Cerca de 70% das espécies chegaram a desaparecer no período de abril a agosto, os meses mais frios e secos do ano.OBJECTIVE: To establish the influence of climatic factors on the monthly frequency of mosquito fauna in areas of the National Park of Serra da Bocaina, Brazil. METHODS: Human bait collections were conducted once a month, in diurnal and nocturnal hours, throughout 24 months, from January 1991 to December 1992. RESULTS: A total of 11,808 adult mosquitoes belonging to 28 different species were collected. Anopheles cruzii, Runchomyia reversa, and Ru. frontosa were the most commonly found species each month. An. cruzii was found mostly from October to February; Ru. reversa and Ru. frontosa predominated in September. An. lutzi, Chagasia fajardoi, Coquillettidia chrysonotum, Aedes serratus, Trichoprosopon simile, Wyeomyia theobaldi, Ru. humboldti, and Ru. theobaldi are the second most prevalent species and they were found mostly throughout the hottest, rainiest and most humid months. Temperature and rainfall have positively influenced the incidence of the majority of the species. CONCLUSIONS: The exuberant and well-preserved tropical forest of Mata Atlântica, rainfalls, and temperatures had a direct and significant contribution to the mosquito fauna in the park. In the months with greater rainfalls, temperatures and humidity, there was an increase in the density and diversity of mosquito species. The period from September to March was the most favorable for their proliferation. In the colder and drier period, from April to August, about 70% of the species showed a significant decline in number, and they were hardly found during these months.

27. Hepatitis C prevalence and risk factors in hemodialysis patients in Central Brazil: a survey by polymerase chain reaction and serological methods

An hemodialysis population in Central Brazil was screened by polymerase chain reaction (PCR) and serological methods to assess the prevalence of hepatitis C virus (HCV) infection and to investigate associated risk factors. All hemodialysis patients (n=428) were interviewed in eight dialysis units in Goiânia city. Blood samples were collected and serum samples screened for anti-HCV antibodies by an enzyme-linked immunosorbent assay (ELISA). Positive samples were retested for confirmation with a line immunoassay (LIA). All samples were also tested for HCV RNA by the PCR. An overall prevalence of 46.7% (CI 95%: 42-51.5) was found, ranging from 20.7% (CI 95%: 8.8-38.1) to 90.4% (CI 95%: 79.9-96.4) depending on the dialysis unit. Of the 428 patients, 185 were found to be seropositive by ELISA, and 167 were confirmed positive by LIA, resulting in an anti-HCV prevalence of 39%. A total of 131 patients were HCV RNA-positive. HCV viremia was present in 63.5% of the anti-HCV-positive patients and in 10.3% of the anti-HCV-negative patients. Univariate analysis of risk factors showed that the number of previous blood transfusions, transfusion of blood before mandatory screening for anti-HCV, length of time on hemodialysis, and treatment in multiple units were associated with HCV positivity. However, multivariate analysis revealed that blood transfusion before screening for anti-HCV and length of time on hemodialysis were significantly associated with HCV infection in this population. These data suggest that nosocomial transmission may play a role in the spread of HCV in the dialysis units studied. In addition to anti-HCV screening, HCV RNA detection is necessary for the diagnosis of HCV infection in hemodialysis patients.

Os processos expansivos situados ao nível da região selar constituem um variável número de patologias que, apesar da localização semelhante, têm características clínicas, radiológicas, patológicas e origens diferentes. Assim sendo, diferentes tipos de abordagens são utilizados no tratamento cirúrgico destas lesões. Dentre os acessos transcranianos convencionais e os mais sofisticados acessos à base do crânio, o acesso sub-frontal proporciona visão direta das estruturas neurovasculares da região. Este estudo descreve 19 tumores operados por esta via, no período compreendido de fevereiro de 1997 a março de 1998. Onze destas lesões eram adenomas pituitários, 3 eram craniofaringiomas, 2 eram meningiomas, 1 germinoma e 1 lesão inflamatória do nervo óptico. A remoção total dos tumores foi conseguida em 17 casos. Não houve mortalidade operatória e as complicações mais comuns foram: anosmia unilateral, que ocorreu em 12 casos; diabetes insipidus transitório, que acometeu 4 pacientes; e déficit isquêmico progressivo, que ocorreu em 1 paciente. Estes resultados demonstram que a via sub-frontal é uma opção segura para a cirurgia dos tumores desta região.

MÉTODOS: Foram realizadas capturas mensais em ambiente silvestre e domiciliar, em isca humana, durante três diferentes períodos do dia, pelo período de 24 meses consecutivos, de janeiro de 1991 a dezembro de 1992. RESULTADOS: Foram capturados 11.808 espécimes adultos, pertencentes a 28 espécies. Ru. reversa e An. cruzii foram predominantes, respectivamente 52,5% e 17,9% do total de mosquitos. Ru. reversa representou 59,4% do total de espécimes no ambiente de mata fechada, seguida por Ru. frontosa com 10,5% e An. cruzii com 9,9%. No ambiente formado por campos de altitude e matas de galeria, o An. cruzii predominou com 48,1%, seguido por Ru. reversa com 28,1%. No ambiente modificado pelo homem, o An. cruzii predominou com 73,7% dos espécimes. Coquillettidia chrysonotum foi a única que se apresentou preferencialmente nesse biótopo: 14,9% no intra, 19,4% no peri e 65,7% no extradomicílio. An. cruzii e Ru. reversa foram constantes em todos os ambientes ao longo do ano. CONCLUSÕES: Com exceção de Cq. chrysonotum, com preferência pelo ambiente modificado pelo homem, os mosquitos apresentam hábitos assinantrópicos no PNSB. An. cruzii, embora assinantrópico, se aproxima e adentra o domicílio para realizar a hematofagia. A presença do Ae. serratus no extra e peridomicílio reforça a importância epidemiológica como vetora potencial de arboviroses. Os Sabethini apresentaram-se exclusivamente silvestres.

The ecology of mosquitoes were studied (Diptera: Culicidae) in areas of Serra do Mar State Park, State of São Paulo, Brazil. Systematized monthly human bait collections were made three times a day, for periods of 2 or 3 h each, in sylvatic and rural areas for 24 consecutive months (January 1991 to December 1992). A total of 24,943 specimens of adult mosquitoes belonging to 57 species were collected during 622 collective periods. Coquillettidia chrysonotum was the most frequent collected mosquito (45.8%) followed by Aedes serratus (6.8%), Cq. venezuelensis (6.5%), Psorophora ferox (5.2) and Ps. albipes (3.1%). The monthly averages of temperature and relative humidity were inserted in the ten-year average limits of maximum and minimum of the previous ten-years. Rainfall accompanied the curve of the ten-year averages. Those climatic factors were influential in the incidence of some species; temperature: Anopheles cruzii, An. mediopunctatus, Ae. scapularis, Ae. fulvus, Cq. chrysonotum, Cq. venezuelensis, Runchomyia reversa, Wyeomyia dyari, Wy. confusa, Wy. shannoni, Wy. theobaldi and Limatus flavisetosus; relative humidity: Ae. serratus, Ae. scapularis, Cq. venezuelensis and Ru. reversa; rainfall: An. cruzii, Ae. scapularis, Ae. fulvus, Cq. venezuelensis Ru. reversa, Wy. theobaldi and Li. flavisetosus.

31. Isoenzymatic variation in the germplasm of Brazilian races of maize (Zea mays L.)

There are more than 200 races of maize (Zea mays L.) divided into three groups (ancient commercial races, the recent commercial races, and indigenous races). Although the indigenous races have no commercial value, they have many important characteristics which can be incorporated into maize breeding programs. Most Brazilian indigenous germplasm race stocks were collected at least 40 years ago, and nothing is known of the genetic variability present in this germplasm. The genetic variability was assayed in 15 populations from four indigenous races of maize (Caingang, Entrelaçado, Lenha and Moroti) and five indigenous cultivars, using five isoenzymatic systems encoded by 14 loci. The analysis revealed a low level of variability among the samples studied. Overall, the mean number of alleles/polymorphic locus was three, 64.3% of the loci analyzed being polymorphic and the estimated heterozygosity was 0.352. The mean number of alleles/polymorphic locus per population was 1.6. A mean of 47.5% of the loci were polymorphic. The mean expected heterozygosity was 0.195, the mean genetic identity was 0.821 and the proportion of total genetic diversity partitioned among populations (Gst) was 0.156. A founder effect could explain the low variability detected.

Large-scale trials of a trapping system designed to collect silvatic Triatominae are reported. Live-baited adhesive traps were tested in various ecosystems and different triatomine habitats (arboreal and terrestrial). The trials were always successful, with a rate of positive habitats generally over 20% and reaching 48.4% for palm trees of the Amazon basin. Eleven species of Triatominae belonging to the three genera of public health importance (Triatoma, Rhodnius and Panstrongylus) were captured. This trapping system provides an effective way to detect the presence of triatomines in terrestrial and arboreal silvatic habitats and represents a promising tool for ecological studies. Various lines of research are contemplated to improve the performance of this trapping system.

The objective of the present study was to investigate the prevalence of Cryptosporidium (Apicomplexa, Cryptosporidiidae) in the snake Crotalus durissus terrificus (Serpentes, Viperidae). Fifty animals were evaluated for the presence of oocysts of Cryptosporidium sp. at the time of arrival and 30 and 60 days later. Intestinal washings with saline solution (1% body weight), fecal samples, and organ scrapings were collected during the study. Oocysts were concentrated by an ether-phosphate-buffered saline sedimentation technique and then separated by a density gradient centrifugation technique. Smears were made with the sediment and submitted to modified acid-fast and auramine-rhodamine staining. Cryptosporidium-positive smears were used as controls for the experimental findings. The overall prevalence of Cryptosporidium sp. oocysts was 14%. Among the positive snakes, oocysts were detected only in the intestinal washing in two specimens, only in the feces in four specimens, and in both materials at least once in one specimen. The positive snakes were predominantly from Santa Maria da Serra city State of São Paulo (57.1%). We also observed that all of the examinations that presented positive results were obtained at least 27 days after the capture of the animals.

Control of Neoclassical Tearing Modes (NTMs) requires an accurate and low latency detection of the mode position. For a burning H-mode ITER plasma, simulations are conducted for both ECE detected via the equatorial port plug and along the line-of-sight of the ECCD launchers. Simulated ECE is detected using synthetic radiometers, with settings chosen to meet the required accuracy. A video bandwidth of 2 kHz is used which allows for an intermediate frequency bandwidth of BIF = 400 MHz for ECE detected via the equatorial port plug. For ECE detected via the ECCD line-of-sight, an intermediate frequency bandwidth of 1.5 GHz and 1 GHz for the 2/1 and 3/2 NTM respectively suffices for accurate location detection. For both ECE systems, the latency requirements for NTM suppression are fulfilled.

Abstract Background There is scarce evidence regarding the association between diet and metabolic syndrome (MetS) in Portuguese population. We aim to evaluate the association between a posteriori dietary patterns (DPs) and MetS and its features. Methods Using random digit dialing, a sample of 2167 adults was selected between 1999 and 2003, in Porto. During a face-to-face interview, a questionnaire was applied, anthropometric measures were taken, blood pressure measured and a fasting blood sample collected. Diet was assessed using a validated food frequency questionnaire, and four DPs were identified in each sex by multivariate finite mixture models. Results After adjustment for age and daily energy intake, comparing to the “healthy” DP, women with the “low fruit and vegetables” DP had a higher odds of high waist circumference (OR = 1.88 95% CI 1.17-3.01) and low HDL-cholesterol (OR = 1.78 95% IC 1.12-2.82) and women in the “red meat and alcohol” DP had higher odds of high waist circumference (OR = 1.45 95% CI 1.01-2.07) and of MetS (OR = 1.57 95% CI 1.07-2.29); men with the “fish” DP had a higher odds of high triglycerides (OR = 1.57 95% CI 1.05-2.35). After further adjustments (education, physical activity, smoking, alcohol drinking, BMI, and menopausal status) no significant associations remained. Conclusions Four distinct DPs were identified in a community sample of Portuguese adults and there was no association with the prevalence of MetS.

Abstract Background Periprostatic (PP) adipose tissue surrounds the prostate, an organ with a high predisposition to become malignant. Frequently, growing prostatic tumor cells extend beyond the prostatic organ towards this fat depot. This study aimed to determine the genome-wide expression of genes in PP adipose tissue in obesity/overweight (OB/OW) and prostate cancer patients. Methods Differentially expressed genes in human PP adipose tissue were identified using microarrays. Analyses were conducted according to the donors' body mass index characteristics (OB/OW versus lean) and prostate disease (extra prostatic cancer versus organ confined prostate cancer versus benign prostatic hyperplasia). Selected genes with altered expression were validated by real-time PCR. Ingenuity Pathway Analysis (IPA) was used to investigate gene ontology, canonical pathways and functional networks. Results In the PP adipose tissue of OB/OW subjects, we found altered expression of genes encoding molecules involved in adipogenic/anti-lipolytic, proliferative/anti-apoptotic, and mild immunoinflammatory processes (for example, FADS1, down-regulated, and LEP and ANGPT1, both up-regulated). Conversely, in the PP adipose tissue of subjects with prostate cancer, altered genes were related to adipose tissue cellular activity (increased cell proliferation/differentiation, cell cycle activation and anti-apoptosis), whereas a downward impact on immunity and inflammation was also observed, mostly related to the complement (down-regulation of CFH). Interestingly, we found that the microRNA MIRLET7A2 was overexpressed in the PP adipose tissue of prostate cancer patients. Conclusions Obesity and excess adiposity modified the expression of PP adipose tissue genes to ultimately foster fat mass growth. In patients with prostate cancer the expression profile of PP adipose tissue accounted for hypercellularity and reduced immunosurveillance. Both findings may be liable to promote a favorable environment for prostate cancer progression.

Abstract Background Obesity is associated with prostate cancer aggressiveness and mortality. The contribution of periprostatic adipose tissue, which is often infiltrated by malignant cells, to cancer progression is largely unknown. Thus, this study aimed to determine if periprostatic adipose tissue is linked with aggressive tumor biology in prostate cancer. Methods Supernatants of whole adipose tissue (explants) or stromal vascular fraction (SVF) from paired fat samples of periprostatic (PP) and pre-peritoneal visceral (VIS) anatomic origin from different donors were prepared and analyzed for matrix metalloproteinases (MMPs) 2 and 9 activity. The effects of those conditioned media (CM) on growth and migration of hormone-refractory (PC-3) and hormone-sensitive (LNCaP) prostate cancer cells were measured. Results We show here that PP adipose tissue of overweight men has higher MMP9 activity in comparison with normal subjects. The observed increased activities of both MMP2 and MMP9 in PP whole adipose tissue explants, likely reveal the contribution of adipocytes plus stromal-vascular fraction (SVF) as opposed to SVF alone. MMP2 activity was higher for PP when compared to VIS adipose tissue. When PC-3 cells were stimulated with CM from PP adipose tissue explants, increased proliferative and migratory capacities were observed, but not in the presence of SVF. Conversely, when LNCaP cells were stimulated with PP explants CM, we found enhanced motility despite the inhibition of proliferation, whereas CM derived from SVF increased both cell proliferation and motility. Explants culture and using adipose tissue of PP origin are most effective in promoting proliferation and migration of PC-3 cells, as respectively compared with SVF culture and using adipose tissue of VIS origin. In LNCaP cells, while explants CM cause increased migration compared to SVF, the use of PP adipose tissue to generate CM result in the increase of both cellular proliferation and migration. Conclusions Our findings suggest that the PP depot has the potential to modulate extra-prostatic tumor cells' microenvironment through increased MMPs activity and to promote prostate cancer cell survival and migration. Adipocyte-derived factors likely have a relevant proliferative and motile role.

Abstract Background Monocarboxylate transporters (MCTs) are transmembrane proteins involved in the transport of monocarboxylates across the plasma membrane, which appear to play an important role in solid tumours, however the role of MCTs in prostate cancer is largely unknown. The aim of the present work was to evaluate the clinico-pathological value of monocarboxylate transporters (MCTs) expression, namely MCT1, MCT2 and MCT4, together with CD147 and gp70 as MCT1/4 and MCT2 chaperones, respectively, in prostate carcinoma. Methods Prostate tissues were obtained from 171 patients, who performed radical prostatectomy and 14 patients who performed cystoprostatectomy. Samples and clinico-pathological data were retrieved and organized into tissue microarray (TMAs) blocks. Protein expression was evaluated by immunohistochemistry in neoplastic (n = 171), adjacent non-neoplastic tissues (n = 135), PIN lesions (n = 40) and normal prostatic tissue (n = 14). Protein expression was correlated with patients' clinicopathologic characteristics. Results In the present study, a significant increase of MCT2 and MCT4 expression in the cytoplasm of tumour cells and a significant decrease in both MCT1 and CD147 expression in prostate tumour cells was observed when compared to normal tissue. All MCT isoforms and CD147 were expressed in PIN lesions. Importantly, for MCT2 and MCT4 the expression levels in PIN lesions were between normal and tumour tissue, which might indicate a role for these MCTs in the malignant transformation. Associations were found between MCT1, MCT4 and CD147 expressions and poor prognosis markers; importantly MCT4 and CD147 overexpression correlated with higher PSA levels, Gleason score and pT stage, as well as with perineural invasion and biochemical recurrence. Conclusions Our data provides novel evidence for the involvement of MCTs in prostate cancer. According to our results, we consider that MCT2 should be further explored as tumour marker and both MCT4 and CD147 as markers of poor prognosis in prostate cancer.

Abstract Background Breast-conserving surgery for breast cancer was developed as a method to preserve healthy breast tissue, thereby improving cosmetic outcomes. Thus far, the primary aim of breast-conserving surgery has been the achievement of tumour-free resection margins and prevention of local recurrence, whereas the cosmetic outcome has been considered less important. Large studies have reported poor cosmetic outcomes in 20-40% of patients after breast-conserving surgery, with the volume of the resected breast tissue being the major determinant. There is clear evidence for the efficacy of ultrasonography in the resection of nonpalpable tumours. Surgical resection of palpable breast cancer is performed with guidance by intra-operative palpation. These palpation-guided excisions often result in an unnecessarily wide resection of adjacent healthy breast tissue, while the rate of tumour-involved resection margins is still high. It is hypothesised that the use of intra-operative ultrasonography in the excision of palpable breast cancer will improve the ability to spare healthy breast tissue while maintaining or even improving the oncological margin status. The aim of this study is to compare ultrasound-guided surgery for palpable tumours with the standard palpation-guided surgery in terms of the extent of healthy breast tissue resection, the percentage of tumour-free margins, cosmetic outcomes and quality of life. Methods/design In this prospective multicentre randomised controlled clinical trial, 120 women who have been diagnosed with palpable early-stage (T1-2N0-1) primary invasive breast cancer and deemed suitable for breast-conserving surgery will be randomised between ultrasound-guided surgery and palpation-guided surgery. With this sample size, an expected 20% reduction of resected breast tissue and an 18% difference in tumour-free margins can be detected with a power of 80%. Secondary endpoints include cosmetic outcomes and quality of life. The rationale, study design and planned analyses are described. Conclusion The COBALT trial is a prospective, multicentre, randomised controlled study to assess the efficacy of ultrasound-guided breast-conserving surgery in patients with palpable early-stage primary invasive breast cancer in terms of the sparing of breast tissue, oncological margin status, cosmetic outcomes and quality of life. 40. Phylogenetic relationships in genus Arachis based on ITS and 5.8S rDNA sequences

Abstract Background The genus Arachis comprises 80 species and it is subdivided into nine taxonomic sections (Arachis, Caulorrhizae, Erectoides, Extranervosae, Heteranthae, Procumbentes, Rhizomatosae, Trierectoides, and Triseminatae). This genus is naturally confined to South America and most of its species are native to Brazil. In order to provide a better understanding of the evolution of the genus, we reconstructed the phylogeny of 45 species using the variation observed on nucleotide sequences in internal transcribed spacer regions (ITS1 and ITS2) and 5.8 S of nuclear ribosomal DNA. Results Intraspecific variation was detected, but in general it was not enough to place accessions of the same species in different clades. Our data support the view that Arachis is a monophyletic group and suggested Heteranthae as the most primitive section of genus Arachis. The results confirmed the circumscriptions of some sections (Caulorrhizae, Extranervosae), but raised questions about others. Sections Erectoides, Trierectoides and Procumbentes were not well defined, while sections Arachis and Rhizomatosae seem to include species that could be moved to different sections. The division of section Arachis into A and B genome species was also observed in the phylogenetic tree and these two groups of species may not have a monophyletic origin. The 2n = 2x = 18 species of section Arachis (A. praecox, A. palustris and A. decora) were all placed in the same clade, indicating they are closely related to each other, and their genomes are more related to B genome than to the A genome. Data also allowed insights on the origin of tetraploid A. glabrata, suggesting rhizome appeared twice within the genus and raising questions about the placement of that species in section Rhizomatosae. Conclusion The main clades established in this study in general agreed with many other studies that have used other types of evidences and sets of species, being some of them included in our study and some not. Thus, the relationships established can be a useful framework for future systematic reviews of genus Arachis and for the selection of species to pre-breeding programs.

Abstract Background Only a few studies have addressed the molecular pathways specifically involved in carcinogenesis of the distal colon and rectum. We aimed to identify potential differences among genetic alterations in distal colon and rectal carcinomas as compared to cancers arising elsewhere in the large bowel. Methods Constitutional and tumor DNA from a test series of 37 patients with rectal and 25 patients with sigmoid carcinomas, previously analyzed for microsatellite instability (MSI), was studied for BAX, IGF2R, TGFBR2, MSH3, and MSH6 microsatellite sequence alterations, BRAF and KRAS mutations, and MLH1 promoter methylation. The findings were then compared with those of an independent validation series consisting of 36 MSI-H carcinomas with origin from each of the large bowel regions. Immunohistochemical and germline mutation analyses of the mismatch repair system were performed when appropriate. Results In the test series, IGFR2 and BAX mutations were present in one and two out of the six distal MSI-H carcinomas, respectively, and no mutations were detected in TGFBR2, MSH3, and MSH6. We confirmed these findings in the validation series, with TGFBR2 and MSH3 microsatellite mutations occurring less frequently in MSI-H rectal and sigmoid carcinomas than in MSI-H colon carcinomas elsewhere (P = 0.00005 and P = 0.0000005, respectively, when considering all MSI-carcinomas of both series). No MLH1 promoter methylation was observed in the MSI-H rectal and sigmoid carcinomas of both series, as compared to 53% found in MSI-H carcinomas from other locations (P = 0.004). KRAS and BRAF mutational frequencies were 19% and 43% in proximal carcinomas and 25% and 17% in rectal/sigmoid carcinomas, respectively. Conclusion The mechanism and the pattern of genetic changes driving MSI-H carcinogenesis in distal colon and rectum appears to differ from that occurring elsewhere in the colon and further investigation is warranted both in patients with sporadic or hereditary disease.

Abstract Hepatitis C virus (HCV) infection is a global public health problem. Long-distance truck drivers live apart from their family for long periods of time, a lifestyle that favors at-risk behaviors such as unprotected sex with multiple partners and illicit drug use. As data concerning HCV infection in this population are still rare, this paper aims to investigate the prevalence, genotypes/subtypes, and the factors associated with HCV infection in long-distance truck drivers in Brazil. A cross-sectional survey was carried out with 641 Brazilian long-truck drivers who were recruited at a major truck stop located at kilometer 1,296 of the BR-153 highway, which is considered to be one of the longest roads in Brazil. All individuals were interviewed, and their serum samples were tested for the presence of antibodies to HCV (anti-HCV) by ELISA and immunoblot. Anti-HCV positive samples were tested for HCV RNA by PCR amplification of the 5' NC and NS5B regions and were genotyped using the LiPA assay and nucleotide sequencing, respectively. Factors associated with HCV infection were identified with logistic regression. The prevalence of HCV infection was 1.4% (95% CI: 0.7-2.8). History of blood transfusion, sharing of personal hygiene tools, illicit drug use and HBV status were factors independently associated with HCV infection in the study population. HCV RNA was detected in 8/9 anti-HCV positive samples, in which genotypes 1 (n = 3), 2 (n = 2), and 3 (n = 3) were determined by LiPA. Using phylogenetic tree analysis of the NS5B region, subtypes 1a (n = 1), 1b (n = 2), 2b (n = 2) and 3a (n = 3) were identified. These data show that the prevalence of HCV infection among Brazilian truck drivers was similar to that observed for the general population. History of blood transfusion, sharing of personal hygiene tools, illicit drug use and HBV status were predictors of HCV infection. The HCV genotypes/subtypes identified in the study population are consistent with those circulating in Brazil.

Abstract Background Oncogenic point mutations in KIT or PDGFRA are recognized as the primary events responsible for the pathogenesis of most gastrointestinal stromal tumors (GIST), but additional genomic alterations are frequent and presumably required for tumor progression. The relative contribution of such alterations for the biology and clinical behavior of GIST, however, remains elusive. Methods In the present study, somatic mutations in KIT and PDGFRA were evaluated by direct sequencing analysis in a consecutive series of 80 GIST patients. For a subset of 29 tumors, comparative genomic hybridization was additionally used to screen for chromosome copy number aberrations. Genotype and genomic findings were cross-tabulated and compared with available clinical and follow-up data. Results We report an overall mutation frequency of 87.5%, with 76.25% of the tumors showing alterations in KIT and 11.25% in PDGFRA. Secondary KIT mutations were additionally found in two of four samples obtained after imatinib treatment. Chromosomal imbalances were detected in 25 out of 29 tumors (86%), namely losses at 14q (88% of abnormal cases), 22q (44%), 1p (44%), and 15q (36%), and gains at 1q (16%) and 12q (20%). In addition to clinico-pathological high-risk groups, patients with KIT mutations, genomic complexity, genomic gains and deletions at either 1p or 22q showed a significantly shorter disease-free survival. Furthermore, genomic complexity was the best predictor of disease progression in multivariate analysis. Conclusions In addition to KIT/PDGFRA mutational status, our findings indicate that secondary chromosomal changes contribute significantly to tumor development and progression of GIST and that genomic complexity carries independent prognostic value that complements clinico-pathological and genotype information.

Abstract Background Arachis hypogaea (peanut) is an important crop worldwide, being mostly used for edible oil production, direct consumption and animal feed. Cultivated peanut is an allotetraploid species with two different genome components, A and B. Genetic linkage maps can greatly assist molecular breeding and genomic studies. However, the development of linkage maps for A. hypogaea is difficult because it has very low levels of polymorphism. This can be overcome by the utilization of wild species of Arachis, which present the A- and B-genomes in the diploid state, and show high levels of genetic variability. Results In this work, we constructed a B-genome linkage map, which will complement the previously published map for the A-genome of Arachis, and produced an entire framework for the tetraploid genome. This map is based on an F2 population of 93 individuals obtained from the cross between the diploid A. ipaënsis (K30076) and the closely related A. magna (K30097), the former species being the most probable B genome donor to cultivated peanut. In spite of being classified as different species, the parents showed high crossability and relatively low polymorphism (22.3%), compared to other interspecific crosses. The map has 10 linkage groups, with 149 loci spanning a total map distance of 1,294 cM. The microsatellite markers utilized, developed for other Arachis species, showed high transferability (81.7%). Segregation distortion was 21.5%. This B-genome map was compared to the A-genome map using 51 common markers, revealing a high degree of synteny between both genomes. Conclusion The development of genetic maps for Arachis diploid wild species with A- and B-genomes effectively provides a genetic map for the tetraploid cultivated peanut in two separate diploid components and is a significant advance towards the construction of a transferable reference map for Arachis. Additionally, we were able to identify affinities of some Arachis linkage groups with Medicago truncatula, which will allow the transfer of information from the nearly-complete genome sequences of this model legume to the peanut crop.

45. Characterization and transferability of microsatellite markers of the cultivated peanut (Arachis hypogaea)

Abstract Background The genus Arachis includes Arachis hypogaea (cultivated peanut) and wild species that are used in peanut breeding or as forage. Molecular markers have been employed in several studies of this genus, but microsatellite markers have only been used in few investigations. Microsatellites are very informative and are useful to assess genetic variability, analyze mating systems and in genetic mapping. The objectives of this study were to develop A. hypogaea microsatellite loci and to evaluate the transferability of these markers to other Arachis species. Results Thirteen loci were isolated and characterized using 16 accessions of A. hypogaea. The level of variation found in A. hypogaea using microsatellites was higher than with other markers. Cross-transferability of the markers was also high. Sequencing of the fragments amplified using the primer pair Ah11 from 17 wild Arachis species showed that almost all wild species had similar repeated sequence to the one observed in A. hypogaea. Sequence data suggested that there is no correlation between taxonomic relationship of a wild species to A. hypogaea and the number of repeats found in its microsatellite loci. Conclusion These results show that microsatellite primer pairs from A. hypogaea have multiple uses. A higher level of variation among A. hypogaea accessions can be detected using microsatellite markers in comparison to other markers, such as RFLP, RAPD and AFLP. The microsatellite primers of A. hypogaea showed a very high rate of transferability to other species of the genus. These primer pairs provide important tools to evaluate the genetic variability and to assess the mating system in Arachis species.

Centrioles are microtubule-based, barrel-shaped structures that initiate the assembly of centrosomes and cilia(1,2). How centriole length is precisely set remains elusive. The microcephaly protein CPAP (also known as MCPH6) promotes procentriole growth(3-5), whereas the oral-facial-digital (OFD) syndrome protein OFD1 represses centriole elongation(6,7). Here we uncover a new subtype of OFD with severe microcephaly and cerebral malformations and identify distinct mutations in two affected families in the evolutionarily conserved C2CD3 gene. Concordant with the clinical overlap, C2CD3 colocalizes with OFD1 at the distal end of centrioles, and C2CD3 physically associates with OFD1. However, whereas OFD1 deletion leads to centriole hyperelongation, loss of C2CD3 results in short centrioles without subdistal and distal appendages. Because C2CD3 overexpression triggers centriole hyperelongation and OFD1 antagonizes this activity, we propose that C2CD3 directly promotes centriole elongation and that OFD1 acts as a negative regulator of C2CD3. Our results identify regulation of centriole length as an emerging pathogenic mechanism in ciliopathies.

Despite significant advancements in ichthyoplankton collection and data processing, challenges persist in the taxonomic identification of these organisms, particularly their eggs. To overcome these challenges, a novel technique has been developed to facilitate the identification of live eggs collected directly in wild. This user-friendly technique includes the collection, processing of the material, and field incubation. Sampling must be conducted using a pelagic net towed at low speed, preferably during early evening. The material processing involves pre-sorting and sorting to remove eggs and larvae. The separated eggs, kept in an aerated bowl, can be identified based on their morphological and meristic characteristics. Unidentified eggs can be placed in plastic bags with oxygen and incubated directly in the aquatic environment for 48-72 h. After this incubation period, the hatched larvae at the yolk-sac or preflexion stage, are identified to the lowest possible taxonomic level. Depending on the study's purpose, hatched larvae and field-collected larvae can be transported to research centers for further development, released back into the natural environment, or fixed to complete the collection. The application of this technique supports management and monitoring programs by identifying spawning areas through egg identification, forming broodstock, and replenishing threatened species, thereby enhancing scientific collections of ichthyoplankton. Additionally, it reduces mortality in ichthyoplankton techniques, including endangered species. Therefore, we believe that this novel taxonomic technique for identifying live ichthyoplankton represents a paradigm shift in the monitoring, management, and conservation of fish, as well as in ecological stewardship and advances in this area of research., (© 2024. - Abstract
This article discusses questions concerning the future of humanity in the face of threats to the health of populations, whose impact has been exacerbated in the course of inequalities in all parts of the world, pari passu with global development in the hegemonized model since last century. The COVID-19 pandemic is a good example that illustrates this dissonance between development and inequalities. Questions were formulated to be debated about the construction of the future of world society, based on the understanding of the evolutionary character of life on the planet vis-à-vis the evils that affect large contingents of the population and represent powerful risks for this evolutionary process. These questions call attention to the discussion around social participation in the definition and control of public policies, as opposed to the hegemony of private interests in the formulation and execution of these policies, both in the scenarios of each country and in the international context.
