Abstract

E-TEST (Einstein Telescope Euregio-Meuse-Rhin Site and Technology) is a project recently funded by the European program Ineterreg Euregio Meuse-Rhine. This program is dedicated to innovative cross boarder activities between Belgium, The Netherlands and Germany. With a total budget of15MC and a consortium of 11 partners from the three countries, the objective of the project is twofold. Firstly, to develop an eco-friendly and non-invasive imaging of the geological conditions as well as the development of an observatory of the underground in the EMR region. Secondly, to develop technologies necessary for 3rd generation gravitational wave detectors. In particular, it is proposed to develop a prototype of large suspended cryogenic silicon mirror, isolated from seismic vibrations at low frequency. The total budget of the project is equally spread over the two activities. The first activity is not discussed at all in this report. The E-TEST prototype will have some key unique features: a silicon mirror of 100 kg, a radiative cooling strategy (non contact), a low-frequency hybrid isolation stage, cryogenic sensors and electronics, a laser and optics at 2 microns, a low thermal noise coating.

Abstract

Background and aimsNucleotide-binding oligomerization domain-like receptor-family pyrin domain-containing 3 (NLRP3) inflammasome activation has been shown to result in liver fibrosis. Mechanisms and downstream signaling remain incompletely understood. Here, we studied the role of IL-18 in hepatic stellate cells (HSCs), and its impact on liver fibrosis.Approach and resultsWe observed significantly increased serum levels of IL-18 (128.4 pg/ml vs. 74.9 pg/ml) and IL-18 binding protein (BP; 46.50 ng/ml vs. 15.35 ng/ml) in patients with liver cirrhosis compared with healthy controls. Single cell RNA sequencing data showed that an immunoregulatory subset of murine HSCs highly expresses Il18 and Il18r1 . Treatment of cultured primary murine HSC with recombinant mouse IL-18 accelerated their transdifferentiation into myofibroblasts. In vivo , IL-18 receptor-deficient mice had reduced liver fibrosis in a model of fibrosis induced by HSC-specific NLRP3 overactivation. Whole liver RNA sequencing analysis from a murine model of severe NASH-induced fibrosis by feeding a choline-deficient, L-amino acid-defined, high fat diet showed that genes related to IL-18 and its downstream signaling were significantly upregulated, and Il18-/- mice receiving this diet for 10 weeks showed protection from fibrotic changes with decreased number of alpha smooth muscle actin-positive cells and collagen deposition. HSC activation triggered by NLRP3 inflammasome activation was abrogated when IL-18 signaling was blocked by its naturally occurring antagonist IL-18BP. Accordingly, we observed that the severe inflammatory phenotype associated with myeloid cell-specific NLRP3 gain-of-function was rescued by IL-18BP.ConclusionsOur study highlights the role of IL-18 in the development of liver fibrosis by its direct effect on HSC activation identifying IL-18 as a target to treat liver fibrosis.

Abstract

The third-generation of gravitational wave observatories, such as the Einstein Telescope (ET) and Cosmic Explorer (CE), aim for an improvement in sensitivity of at least a factor of ten over a wide frequency range compared to the current advanced detectors. In order to inform the design of the third-generation detectors and to develop and qualify their subsystems, dedicated test facilities are required. ETpathfinder prototype uses full interferometer configurations and aims to provide a high sensitivity facility in a similar environment as ET. Along with the interferometry at 1550 nm and silicon test masses, ETpathfinder will focus on cryogenic technologies, lasers and optics at 2090 nm and advanced quantum-noise reduction schemes. This paper analyses the underpinning noise contributions and combines them into full noise budgets of the two initially targeted configurations: 1) operating with 1550 nm laser light and at a temperature of 18 K and 2) operating at 2090 nm wavelength and a temperature of 123 K.

Abstract

News diversity in the media has for a long time been a foundational and uncontested basis for ensuring that the communicative needs of individuals and society at large are met. Today, people increasingly rely on online content and recommender systems to consume information challenging the traditional concept of news diversity. In addition, the very concept of diversity, which differs between disciplines, will need to be re-evaluated requiring a interdisciplinary investigation, which requires a new level of mutual cooperation between computer scientists, social scientists, and legal scholars. Based on the outcome of a multidisciplinary workshop, we have the following recommendations, directed at researchers, funders, legislators, regulators, and the media industry: 1. Do more research on news recommenders and diversity. 2. Create a safe harbor for academic research with industry data. 3. Optimize the role of public values in news recommenders. 4. Create a meaningful governance framework. 5. Abstract

User comments have become an essential part of online journalism. However, newsrooms are often overwhelmed by the vast number of diverse comments, for which a manual analysis is barely feasible. Identifying meta-comments that address or mention newsrooms, individual journalists, or moderators and that may call for reactions is particularly critical. In this paper, we present an automated approach to identify and classify meta-comments. We compare comment classification based on manually extracted features with an end-to-end learning approach. We develop, optimize, and evaluate multiple classifiers on a comment dataset of the large German online newsroom SPIEGEL Online and the 'One Million Posts' corpus of DER STANDARD, an Austrian newspaper. Both optimized classification approaches achieved encouraging $F_{0.5}$ values between 76% and 91%. 50. How Would I Treat My Own Chronic Thromboembolic Pulmonary Hypertension in the Perioperative Period?

Abstract

Chronic thromboembolic pulmonary hypertension (CTEPH) results from an incomplete resolution of acute pulmonary embolism, leading to occlusive organized thrombi, vascular remodeling, and associated microvasculopathy with pulmonary hypertension (PH). A definitive CTEPH diagnosis requires PH confirmation by right-heart catheterization and evidence of chronic thromboembolic pulmonary disease on imaging studies. Surgical removal of the organized fibrotic material by pulmonary endarterectomy (PEA) under deep hypothermic circulatory arrest represents the treatment of choice. One-third of patients with CTEPH are not deemed suitable for surgical treatment, and medical therapy or interventional balloon pulmonary angioplasty presents alternative treatment options. Pulmonary endarterectomy in patients with technically operable disease significantly improves symptoms, functional capacity, hemodynamics, and quality of life. Perioperative mortality is <2.5% in expert centers where a CTEPH multidisciplinary team optimizes patient selection and ensures the best preoperative optimization according to individualized risk assessment. Despite adequate pulmonary artery clearance, patients might be prone to perioperative complications, such as right ventricular maladaptation, airway bleeding, or pulmonary reperfusion injury. These complications can be treated conventionally, but extracorporeal membrane oxygenation has been included in their management recently. Patients with residual PH post-PEA should be considered for medical or percutaneous interventional therapy. [ABSTRACT FROM AUTHOR]
