Abstract Climate change is currently one of humanity’s greatest threats. To help scholars understand the psychology of climate change, we conducted an online quasi-experimental survey on 59,508 participants from 63 countries (collected between July 2022 and July 2023). In a between-subjects design, we tested 11 interventions designed to promote climate change mitigation across four outcomes: climate change belief, support for climate policies, willingness to share information on social media, and performance on an effortful pro-environmental behavioural task. Participants also reported their demographic information (e.g., age, gender) and several other independent variables (e.g., political orientation, perceptions about the scientific consensus). In the no-intervention control group, we also measured important additional variables, such as environmentalist identity and trust in climate science. We report the collaboration procedure, study design, raw and cleaned data, all survey materials, relevant analysis scripts, and data visualisations. This dataset can be used to further the understanding of psychological, demographic, and national-level factors related to individual-level climate action and how these differ across countries.

Congenital diaphragmatic hernia (CDH) is a relatively common and genetically heterogeneous structural birth defect associated with high mortality and morbidity. We describe eight unrelated families with an X-linked condition characterized by diaphragm defects, variable anterior body-wall anomalies, and/or facial dysmorphism. Using linkage analysis and exome or genome sequencing, we found that missense variants in plastin 3 (PLS3), a gene encoding an actin bundling protein, co-segregate with disease in all families. Loss-of-function variants in PLS3 have been previously associated with X-linked osteoporosis (MIM: 300910), so we used in silico protein modeling and a mouse model to address these seemingly disparate clinical phenotypes. The missense variants in individuals with CDH are located within the actin-binding domains of the protein but are not predicted to affect protein structure, whereas the variants in individuals with osteoporosis are predicted to result in loss of function. A mouse knockin model of a variant identified in one of the CDH-affected families, c.1497G>C (p.Trp499Cys), shows partial perinatal lethality and recapitulates the key findings of the human phenotype, including diaphragm and abdominal-wall defects. Both the mouse model and one adult human male with a CDH-associated PLS3 variant were observed to have increased rather than decreased bone mineral density. Together, these clinical and functional data in humans and mice reveal that specific missense variants affecting the actin-binding domains of PLS3 might have a gain-of-function effect and cause a Mendelian congenital disorder.

IntroductionThe Arabidopsis abc1k1/pgr6 (Activity of BC1 complex/proton regulation 6) mutant is characterized by photosynthetic and conditional developmental phenotypes triggered by stressful red as well as high light. The Arabidopsis ABC1-like kinases belong to the atypical kinase family and contain conserved ATP-binding and hydrolysis motifs, but their physiological requirement has never been investigated.MethodsBy mutation to asparagine, we demonstrate that the highly conserved active site aspartate residue within ATP-binding motif VIIb is required for the physiological functions of ABC1K1.ResultsComplementation of the abc1k1 knock out mutant with ABC1K1 D400N, failed to restore the wildtype phenotype.DiscussionThese results provide in vivo evidence for a critical role of the active site aspartate residue (D400) of ABC1K1.

Plants and algae are faced with a conundrum: harvesting sufficient light to drive their metabolic needs while dissipating light in excess to prevent photodamage, a process known as nonphotochemical quenching. A slowly relaxing form of energy dissipation, termed qH, is critical for plants' survival under abiotic stress; however, qH location in the photosynthetic membrane is unresolved. Here, we tested whether we could isolate subcomplexes from plants in which qH was induced that would remain in an energy-dissipative state. Interestingly, we found that chlorophyll (Chl) fluorescence lifetimes were decreased by qH in isolated major trimeric antenna complexes, indicating that they serve as a site for qH-energy dissipation and providing a natively quenched complex with physiological relevance to natural conditions. Next, we monitored the changes in thylakoid pigment, protein, and lipid contents of antenna with active or inactive qH but did not detect any evident differences. Finally, we investigated whether specific subunits of the major antenna complexes were required for qH but found that qH was insensitive to trimer composition. Because we previously observed that qH can occur in the absence of specific xanthophylls, and no evident changes in pigments, proteins, or lipids were detected, we tentatively propose that the energy-dissipative state reported here may stem from Chl-Chl excitonic interaction.

This work presents the new model called CRHyME (Climatic Rainfall Hydrogeological Modelling Experiment), a tool for geo-hydrological hazard evaluation. CRHyME is a physically based and spatially distributed model written in the Python language that represents an extension of the classic hydrological models working at the basin scale. CRHyME's main focus consists of simulating rainfall-induced geo-hydrological instabilities such as shallow landslides, debris flows, catchment erosion and sediment transport into a river. These phenomena are conventionally decoupled from a hydrological routine, while in CRHyME they are simultaneously and quantitatively evaluated within the same code through a multi-hazard approach. CRHyME is applied within some case studies across northern Italy. Among these, the Caldone catchment, a well-monitored basin of 27 km2 located near the city of Lecco (Lombardy), was considered for the calibration of solid-transport routine testing, as well as the spatial-scale dependence related to digital terrain resolution. CRHyME was applied across larger basins of the Valtellina (Alps) and Emilia (Apennines) areas (∼2600 km2) which have experienced severe geo-hydrological episodes triggered by heavy precipitation in the recent past. CRHyME's validation has been assessed through NSE (Nash–Sutcliffe efficiency) and RMSE (root mean square error) hydrological-error metrics, while for landslides the ROC (receiver operating characteristic) methodology was applied. CRHyME has been able to reconstruct the river discharge at the reference hydrometric stations located at the outlets of the basins to estimate the sediment yield at some hydropower reservoirs chosen as a reference and to individuate the location and the triggering conditions of shallow landslides and debris flows. The good performance of CRHyME was reached, assuring the stability of the code and a rather fast computation and maintaining the numerical conservativity of water and sediment balances. CRHyME has shown itself to be a suitable tool for the quantification of the geo-hydrological process and thus useful for civil-protection multi-hazard assessment.

Surface-enhanced Raman spectroscopy is a powerful technique for the ultra-sensitive detection of organic analytes. In this paper, the preparation of SERS substrates based on silver nanorods (AgNRs) is proposed, exploiting a simple protocol which does not require complex procedures and/or sophisticated and expensive instrumentation. For this purpose, various syntheses of AgNRs were tested, and the best one for preparing the SERS active substrate proved to be the one which does not involve surfactants as nanoparticle stabilizers. The plasmonic properties of the selected substrate can be modified based on the concentration of the deposited nanoparticles, allowing for the experimentation of different excitation wavelengths. Positive results were obtained on reference solutions of three natural dyes of historical interest using both green exciting radiation (532 nm) and two near-infrared ones (785 and 850 nm; the latter is combined with the SSE™ technology for further fluorescence quenching). Furthermore, the substrates of AgNRs were found to be suitable for SERS measurements even in dry-state conditions, i.e., only exploiting the electromagnetic interaction between the nanostructured substrate and the dye molecules absorbed onto a wool fibre.

Indoor radon is one of the most significant contributors to lung cancer after smoking. Mitigation strategies based on protecting buildings with radon barrier materials, combined with home ventilation or room pressurization, are regularly used. A scale model room made from a porous ignimbrite rich in radon precursors was used as an analogue to test the efficiency of fifteen airtight membranes to reduce radon levels, also in combination with room pressurization. The results of these experiments were considered together with previous ones to propose the scale model room approach as a tool for rapidly evaluating the performance of specially designed radon barrier materials, and for radiation exposure assessment. Relative reduction of indoor radon (RIR) ranges from −20 to −94%. The most effective materials were FPO membrane, single-component silane-terminated polymer membranes and synthetic resins. The presence of additives likely modified the composition and structure of some products, improving their radon barrier capacity. The introduction of room pressurization further reduced radon levels in the model room where the membranes were applied. The overpressure necessary to reach RIRs of the order of 85–90% is very low for materials that powerfully stop radon even without ventilation, but necessarily higher for poorer membranes.

Background Women are underrepresented in stroke thrombectomy trials, and the impact of sex differences in outcomes after stroke thrombectomy is unclear. We performed a multicenter propensity matching study to define sex‐related differences in outcome after endovascular thrombectomy and integrated results in a meta‐analysis. Methods We included patients with anterior circulation large vessel occlusion consecutively treated with thrombectomy at 2 Comprehensive Stroke Centres (2016–2023). Selection criteria reflected international guidelines. Through systematic review we selected all studies reporting endovascular thrombectomy outcomes in anterior circulation large vessel occlusion stroke, applying propensity score matching. MEDLINE, EMBASE, and Cochrane CENTRAL were searched up to August 15, 2023 according to predefined protocol (OSF.io/je3da). Data were extracted by 2 independent raters, pooled estimates calculated according to random‐effect modeling meta‐analysis and reported as odds ratio (OR) and standard 95% CI. Outcomes were good functional outcome, defined as modified Rankin Scale score 0–2 at 90 days after stroke, and symptomatic intracranial hemorrhage, adjudicated according to European Cooperative Acute Stroke Study II criteria. Results After matching, 698 patients (349 women versus 349 men) had similar cardiovascular risk factors, baseline features, and treatment approach. No significant differences were found for good functional outcome (OR = 0.89, 95% CI = 0.66–1.2) and symptomatic intracranial hemorrhage (OR = 1.00, 95% CI = 0.44–2.26) in the cohort study by sex. Systematic review identified 3 studies (n = 3706), all of high quality. No differences emerged in rates of good functional outcome (OR = 1.00, 95% CI = 0.79–1.21) or symptomatic intracranial hemorrhage (OR = 0.85, 95% CI = 0.60–1.19) depending on sex. Conclusion Women receiving endovascular thrombectomy for anterior circulation large vessel occlusion related stroke have similar rates of good functional outcome and symptomatic intracranial hemorrhage compared to men.

Giacomo Torno was born in 1539 (or 1541) in Naples. At the age of 18 he joined the Clerics Regular Theatines in San Paolo Maggiore and was welcomed on 30 October 1558. He suffered a stroke on 4 December 1608 and died 45 days later. Contemporary sources report that he appeared to be tormented by the devil during his illness, tormented by constant spasms in his arm, which caused him great discomfort. During the analysis of his mummified body, a discontinuity of the skin surface at the level of the sacrum was discovered. All morphological features indicate a wound that developed during the subject’s life. Based on historical sources, the fracture of the first coccygeal vertebra, the appearance of the lesion near death, and the shape of the lesion, it appears that this is the first recorded instance of the Kennedy terminal ulcer, identified through both direct and indirect sources

Landslides are natural hazards that can cause severe damage and loss of life. Optical cameras are a low-cost and high-resolution alternative among many monitoring systems, as their size and capabilities can vary, allowing for flexible implementation and location. Computer vision is a branch of artificial intelligence that can analyze and understand optical images, using techniques such as optical flow, image correlation and machine learning. The application of such techniques can estimate the motion vectors, displacement fields, providing valuable information for landslide detection, monitoring and prediction. However, computer vision also faces some challenges such as illumination changes, occlusions, image quality, and computational complexity. In this work, a computer vision approach based on Lucas-Kanade optical dense flow was applied to estimate the motion vectors between consecutive images obtained during landslide simulations in a laboratory environment. The approach is applied to two experiments that vary in their illumination and setup parameters to test its applicability. We also discuss the application of this methodology to images from Sentinel-2 satellite optical sensors for landslide monitoring in real-world scenarios.

BackgroundMicroRNAs (miRNAs) are small non-coding RNAs capable of regulating gene expression post-transcriptionally. MiRNAs are recognized as key regulators of diverse biological and developmental processes. During the pregnancy–puerperal cycle, numerous changes occur in the female body for the formation, growth, and development of the baby. After birth, there is a critical period in child development, as rapid gains in the physical, cognitive, and socio-emotional domains constitute the “building blocks” of children’s later growth.ObjectiveThe aim of this study was to investigate the association between maternal expression of hsa-miR-423-5p during the first and second trimesters of pregnancy and neurocognitive development at 90 days of life in infants. Methods: This is a longitudinal study included in a population-based cohort study, carried out in a city in southern Brazil. The Bayley III was used to assess the babies’ cognitive development. Blood samples from mothers were obtained for RNA extraction from serum and analysis of miRNA expression by qRT-PCR.ResultsIn total, 87 dyads (mother–baby) were included. The average gestational age was 15.86 weeks (SD ± 5.55). An association of maternal miRNA with infant cognitive development was found; as maternal miR-423-5p increases, infants’ cognitive development increases by 2.40 (95% CI 0.37; 4.43, p = 0.021) points at 3 months of age.ConclusionIn this context, it is suggested to use this miRNA as a biomarker of child neurocognitive development detectable in the prenatal period, thus allowing the planning of early interventions.

Valve-in-ring procedures represent a feasible solution for high-risk patients with surgical repair failure. The risk of left ventricular outflow tract obstruction increases the challenge, and transcatheter approaches to prevent it are technically demanding and often do not resolve it. We demonstrate the feasibility and safety of a transseptal balloon-assisted translocation of the anterior mitral leaflet for valve-in-ring implantation.

Gaspare Beretta (1620-1703) was one of the most important engineers in the Seventeenth century Lombardy. He was involved in many sieges and improved fortresses in the Spanish domains and in other Italian States. After a long career he left a remarkable quantity of maps, works and projects. Beretta was also the author of a journal written in 1656 during the siege laid by Francis Duke of Modena and Louis de Bourbon-Vendôme againts the stronghold of Valenza. This source is kept in the Lombard Historical Society’s Archive (Milan) and nowadays remains unknown. In this essay I will examine the journal focusing on the customs of Early Modern Age siege warfare, before the marquis de Vauban’s revolution. In addition, I will compare this report with Italian and French ones, also using the despatches between Milan and the Spanish court. The main purpose is to highlight the uniqueness of the source, which can be considered the voice of a society forced to withstand the siege efforts.

Abstract The COVID-19 pandemic has affected all domains of human life, including the economic and social fabric of societies. One of the central strategies for managing public health throughout the pandemic has been through persuasive messaging and collective behaviour change. To help scholars better understand the social and moral psychology behind public health behaviour, we present a dataset comprising of 51,404 individuals from 69 countries. This dataset was collected for the International Collaboration on Social & Moral Psychology of COVID-19 project (ICSMP COVID-19). This social science survey invited participants around the world to complete a series of moral and psychological measures and public health attitudes about COVID-19 during an early phase of the COVID-19 pandemic (between April and June 2020). The survey included seven broad categories of questions: COVID-19 beliefs and compliance behaviours; identity and social attitudes; ideology; health and well-being; moral beliefs and motivation; personality traits; and demographic variables. We report both raw and cleaned data, along with all survey materials, data visualisations, and psychometric evaluations of key variables.

Abstract Background Stroke is a leading cause of death and disability worldwide. A major factor in brain damage following ischemia is excitotoxicity caused by elevated levels of the neurotransmitter glutamate. In the brain, glutamate homeostasis is a primary function of astrocytes. Amburana cearensis has long been used in folk medicine and seed extract obtained with dichloromethane (EDAC) have previously been shown to exhibit cytoprotective activity in vitro. The aim of the present study was to analyse the activity of EDAC in hippocampal brain slices. Methods We prepared a dichloromethane extract (EDAC) from A. cearensis seeds and characterized the chemical constituents by 1H and 13C-NMR. Hippocampal slices from P6-8 or P90 Wistar rats were used for cell viability assay or glutamate uptake test. Hippocampal slices from P10-12 transgenic mice SOX10-EGFP and GFAP-EGFP and immunofluorescence for GS, GLAST and GLT1 were used to study oligodendrocytes and astrocytes. Results Astrocytes play a critical role in glutamate homeostasis and we provide immunohistochemical evidence that in excitotoxicity EDAC increased expression of glutamate transporters and glutamine synthetase, which is essential for detoxifying glutamate. Next, we directly examined astrocytes using transgenic mice in which glial fibrillary acidic protein (GFAP) drives expression of enhanced green fluorescence protein (EGFP) and show that glutamate excitotoxicity caused a decrease in GFAP-EGFP and that EDAC protected against this loss. This was examined further in the oxygen–glucose deprivation (OGD) model of ischemia, where EDAC caused an increase in astrocytic process branching, resulting in an increase in GFAP-EGFP. Using SOX10-EGFP reporter mice, we show that the acute response of oligodendrocytes to OGD in hippocampal slices is a marked loss of their processes and EDAC protected oligodendrocytes against this damage. Conclusion This study provides evidence that EDAC is cytoprotective against ischemia and glutamate excitotoxicity by modulating astrocyte responses and stimulating their glutamate homeostatic mechanisms.

Psychiatric disorders often require pharmacological interventions to alleviate symptoms and improve quality of life. However, achieving an optimal therapeutic outcome is challenging due to several factors, including variability in the individual response, inter-individual differences in drug metabolism, and drug interactions in polytherapy. Therapeutic drug monitoring (TDM), by measuring drug concentrations in biological samples, represents a valuable tool to address these challenges, by tailoring medication regimens to each individual. This review analyzes the current landscape of TDM in psychiatric practice, highlighting its significance in optimizing drug dosages, minimizing adverse effects, and improving therapeutic efficacy. The metabolism of psychiatric medications (i.e., mood stabilizers, antipsychotics, antidepressants) often exhibits significant inter-patient variability. TDM can help address this variability by enhancing treatment personalization, facilitating early suboptimal- or toxic-level detection, and allowing for timely interventions to prevent treatment failure or adverse effects. Furthermore, this review briefly discusses technological advancements and analytical methods supporting the implementation of TDM in psychiatric settings. These innovations enable quick and cost-effective drug concentration measurements, fostering the widespread adoption of TDM as a routine practice in psychiatric care. In conclusion, the integration of TDM in psychiatry can improve treatment outcomes by individualizing medication regimens within the so-called precision medicine.

Vision starts in retinal photoreceptors when specialized proteins (opsins) sense photons via their covalently bonded vitamin A derivative 11cis retinaldehyde (11cis-RAL). The reaction of non-enzymatic aldehydes with amino groups lacks specificity, and the reaction products may trigger cell damage. However, the reduced synthesis of 11cis-RAL results in photoreceptor demise and suggests the need for careful control over 11cis-RAL handling by retinal cells. This perspective focuses on retinoid(s) synthesis, their control in the adult retina, and their role during retina development. It also explores the potential importance of 9cis vitamin A derivatives in regulating retinoid synthesis and their impact on photoreceptor development and survival. Additionally, recent advancements suggesting the pivotal nature of retinoid synthesis regulation for cone cell viability are discussed.

Con la finalización de la segunda guerra mundial se propagó en Latinoamérica la lógica de la planificación territorial, estimulada desde Estados Unidos a través de centros y entidades hemisféricas que difundieron programas de asistencia técnica. A este fenómeno se sumó la crisis de los llamados “modelos nacionales”, acompañada por un vertiginoso crecimiento demográfico y el desarrollo de las conurbaciones producto de las migraciones campo-ciudad, que pusieron de relieve el creciente problema de la vivienda. El déficit habitacional desbordó las soluciones habitacionales del tipo “llave en mano”, característicos de los decenios precedentes, y en ese contexto, el modelo de autoconstrucción se propuso como solución alternativa. En el marco de los debates continentales alrededor del problema de la vivienda masiva a fines de la década de 1950, el artículo brinda una aproximación a los programas sobre autoconstrucción y asistencia técnica introducidos en el recientemente creado Instituto de la Vivienda de la Provincia de Buenos Aires (IVBA), presidido por Hilario Zalba a partir de 1958. El flamante organismo se insertó en la discusión disciplinar contemporánea llevando adelante políticas que significarían un desplazamiento del rol “dirigista” del Estado argentino hacia los procesos participativos y “descentralizados”, el fomento de la iniciativa privada y la industrialización de la construcción.

Summary: Background: Oral tauroursodeoxycholic acid (TUDCA) is a commercial drug currently tested in patients with amyotrophic lateral sclerosis (ALS) both singly and combined with sodium phenylbutyrate. This retrospective study aimed to investigate, in a real-world setting, whether TUDCA had an impact on the overall survival of patients with ALS who were treated with this drug compared to those patients who received standard care only. Methods: This propensity score–matched study was conducted in the Emilia Romagna Region (Italy), which has had an ALS regional registry since 2009. Out of 627 patients with ALS diagnosed from January 1st, 2015 to June 30th, 2021 and recorded in the registry with available information on death/tracheostomy, 86 patients took TUDCA and were matched in a 1:2 ratio with patients who received only usual care according to age at onset, sex, phenotype, diagnostic latency, ALS Functional Rating Scale-Revised (ALSFRS-R) at first visit, disease progression rate at first visit, and BMI at diagnosis. The primary outcome was survival difference (time from onset of symptoms to tracheostomy/death) between TUDCA exposed and unexposed patients. Findings: A total of 86 patients treated with TUDCA were matched to 172 patients who did not receive treatment. TUDCA-exposed patients were stratified based on dosage (less than or equal to 1000 mg/day or greater) and duration (less than or equal to 12 months or longer) of treatment. The median overall survival was 49.6 months (95% CI 41.7–93.5) among those treated with TUDCA and 36.2 months (95% CI 32.7–41.6) in the control group, with a reduced risk of death observed in patients exposed to a higher dosage (defined as ≥ 1000 mg/day) of TUDCA (HR 0.56; 95% CI 0.38–0.83; p = 0.0042) compared to both the control group and those with lower TUDCA dosages (defined as

BackgroundCardiovascular disease continues to be the leading cause of death globally. Clinical practice guidelines aimed at improving disease management and positively impacting major cardiac adverse events recommend genetic testing for inherited cardiovascular conditions such as dilated cardiomyopathy (DCM), hypertrophic cardiomyopathy (HCM), long QT syndrome (LQTS), hereditary amyloidosis, and familial hypercholesterolemia (FH); however, little is known about how consistently practitioners order genetic testing for these conditions in routine clinical practice. This study aimed to assess the adoption of guideline-directed genetic testing for patients diagnosed with DCM, HCM, LQTS, hereditary amyloidosis, or FH.MethodsThis retrospective cohort study captured real-world evidence of genetic testing from ICD-9-CM and ICD-10-CM codes, procedure codes, and structured text fields of de-identified patient records in the Veradigm Health Insights Ambulatory EHR Research Database linked with insurance claims data. Data analysis was conducted using an automated electronic health record analysis engine. Patient records in the Veradigm database were sourced from more than 250,000 clinicians serving over 170 million patients in outpatient primary care and specialty practice settings in the United States and linked insurance claims data from public and private insurance providers. The primary outcome measure was evidence of genetic testing within six months of condition diagnosis.ResultsBetween January 1, 2017, and December 31, 2021, 224,641 patients were newly diagnosed with DCM, HCM, LQTS, hereditary amyloidosis, or FH and included in this study. Substantial genetic testing care gaps were identified. Only a small percentage of patients newly diagnosed with DCM (827/101,919; 0.8%), HCM (253/15,507; 1.6%), LQTS (650/56,539; 1.2%), hereditary amyloidosis (62/1,026; 6.0%), or FH (718/49,650; 1.5%) received genetic testing.ConclusionsGenetic testing is underutilized across multiple inherited cardiovascular conditions. This real-world data analysis provides insights into the delivery of genomic healthcare in the United States and suggests genetic testing guidelines are rarely followed in practice.

Bucchero is a well-known class of Etruscan ceramics, characterized by being entirely black both on the surface and in the body. In the present investigation, a representative set of bucchero sherds from the excavations of the Etruscan town of Tarquinia (Viterbo, Italy) was analyzed through flame atomic absorption spectroscopy (FAAS), Fourier-transform infrared spectroscopy (FTIR) and near-infrared (NIR) reflection spectroscopy. Both elemental and mineralogical composition data were treated with methods of multivariate analysis. It was thus possible to discriminate between local and imported production, to estimate the firing temperature and to verify the origin of the grey-black color in this peculiar ceramic class. Furthermore, a virtual reconstruction of a bucchero vase starting from fragments was also attempted on the basis of chemical analysis data.

In the present work, a method is proposed that combines spectrofluorimetry with multivariate analysis (cluster and principal components) to non-invasively investigate natural dyes used, mainly in the form of lakes, in illuminations. First, a consistent number of mock-up samples of red and purple lakes (from brazilwood, madder, kermes, cochineal, lac dye, folium and orcein) were prepared following the recipes reported in ancient treatises, then the experimental conditions for their non-invasive investigation were optimized by benchtop instrumentation and transferred to a portable microprobe suitable for in situ analyses. For all the coloring materials examined, it was thus possible to obtain a satisfactory grouping of samples containing the same dye. The analytical protocol was finally extended to the characterization of dyes in the Crescenzago choir books, illuminated manuscripts dating back to the end of the 15th century.
