Abstract Identifying and characterizing sedimentary evolution patterns are crucial for assessing the distributions of source and reservoir rocks, which are fundamental to hydrocarbon exploration. This study analyzed the stratigraphic sequence, lithological characteristics, sedimentary lithofacies, individual well sedimentary sequences, and seismic reflection properties. The analysis revealed six fourth-order sequences, including progradational and regressive sequences, indicative of water level changes. The sediment sources for the second and third sub-members of the Eocene Shahejie Formation's third member (Es3 2+3) in the Nanpu Sag were identified as the Baigezhuang and Xinanzhuang Uplifts. Predominantly, the sandstones are lithic arkose and feldspathic litharenite, both of which exhibit low compositional and structural maturity. Notably, 22 lithofacies and 8 lithofacies associations suggest fan delta processes. This study identified three fundamental seismic reflection package reflection types. These lithofacies associations, sedimentary sequences, and seismic reflections serve as critical indicators for determining sedimentary environments. The results from the sedimentary facies analysis indicate that the Es3 2+3 Formation developed fan delta deposits, controlled by the sequence of the sedimentary evolution pattern. The potential of these fan delta sediments to form oil and gas reservoirs is significant. Therefore, precise characterization of the sedimentary evolution pattern is essential for a comprehensive understanding of basin dynamics and hydrocarbon potential.

Yellowfin tuna (Thunnus albacares) is popular with consumers due to its delicious meat and high nutritional value. It is a globally recognized high-end marine economic fish. With the advancement of fishing equipment and the increase in fishing efforts, the amount of tuna resources has decreased significantly. To make up for the insufficient supply of tuna in the market and protect wild populations, it is essential to carry out research on artificial aquaculture technology for tuna and to gradually establish full-cycle cultures of tuna. In order to study the differences in the nutritional components and quality of yellowfin tuna muscles of different sizes, three different-sized yellowfin tuna, J1 (4.2±1.2) kg, J2 (22.5± 2.5) kg, and J3 (50.8±3.9) kg, were used as the research subjects, and conventional biochemical analysis methods were used to compare and analyze the proximate compositions, amino acids, fatty acids, and mineral elements of tuna muscle. The results showed that (1) the moisture level of the J1 group was significantly higher than that of the J2 and J3 groups (PJ2>J1. The content of nonessential amino acids was J3>J2>J1 (P0.05). The fatty acids in yellowfin tuna muscle were mainly PUFAs, and the content of DHA accounted for 37.46%~39.18% of the total fatty acid content. Abstract The rigid hull encasing Tartary buckwheat seeds necessitates a laborious dehulling process before flour milling, resulting in considerable nutrient loss. Investigation of lignin composition is pivotal in understanding the structural properties of tartary buckwheat seeds hulls, as lignin is key determinant of rigidity in plant cell walls, thus directly impacting the dehulling process. Here, the lignin composition of seed hulls from 274 Tartary buckwheat accessions is analyzed, unveiling a unique lignin chemotype primarily consisting of G lignin, a common feature in gymnosperms. Furthermore, the hardness of the seed hull showed a strong negative correlation with the S lignin content. Genome‐wide detection of selective sweeps uncovered that genes governing the biosynthesis of S lignin, specifically two caffeic acid O‐methyltransferases (COMTs) and one ferulate 5‐hydroxylases, are selected during domestication. This likely contributed to the increased S lignin content and decreased hardness of seed hulls from more domesticated varieties. Genome‐wide association studies identified robust associations between FtCOMT1 and the accumulation of S lignin in seed hull. Transgenic Arabidopsis comt1 plants expressing FtCOMT1 successfully reinstated S lignin content, confirming its conserved function across plant species. These findings provide valuable metabolic and genetic insights for the potential redesign of Tartary buckwheat seed hulls.

Abstract A single type of signal processing means that it is difficult to analyze vibration signals comprehensively and effectively. By comprehensively using wavelet analysis techniques, a comprehensive and in-depth study of aero-engine vibration conditions is realized as a way to carry out health management. By introducing various types of wavelet analysis techniques and using Labview2022 programming, corresponding signal processing tools are developed for the analysis of the collected vibration signals. The comprehensive analysis of aero-engine vibration signals based on the wavelet transform method is realized, and the corresponding products are successfully applied in engineering practice.

Abstract Background In this study, by analyzing the correlation between various components of health-related physical fitness (HPF) and liver function indicators, the indicators of physical fitness that were highly correlated with liver function and could be monitored at home were screened to prevent more serious liver disease in the future, and to provide experimental basis for prescribing personalized exercise. Methods A total of 330 faculties (female = 198) of a university were recruited. The indicators of HPF and liver function were measured. Spearman correlation analysis, multivariate linear regression, and cross-lagged panel model was used to data statistics. Results In males, body fat (BF) was positively correlated with alanine aminotransferase (ALT); vital capacity and the vital capacity index were positively correlated with albumin; and vertical jump was positively correlated with globulin and negatively correlated with the albumin-globulin ratio (P

In this study, we conduct a reliability characterisation study of cold standby systems. Utilising synthetic rectangular formulas and cold preparedness equivalent models for cold standby systems, we analyse the lifetimes of several typical configurations, including series, parallel, and k/n:m voting systems. This study proposes system equivalent models for various types of cold standby systems, all composed of components that follow the same exponential distribution. We use the equivalent model to determine the optimal timing for the use of cold spares and derive the reliability function and residual lifetime function for each type of system. To demonstrate the validity of our model, the Monte Carlo simulation is strategically designed based on the system failure rate function. The experimental results are then compared with those obtained from the numerical model, highlighting that the numerical method incurs a lower time cost.

Recently, composite materials consisting of ionic liquids (ILs) and metal–organic frameworks (MOFs) have attracted a great deal of attention due to their fantastic properties. Many theoretical studies have been performed on their special structures and gas separation applications. Yet, the mechanism for the diffusion of ILs inside MOF channels still remains unclear. Here, the DFT calculations (e.g., rigid and relaxed potential energy surface, PES, scan) together with frontier orbital analysis, natural charge analysis, and energy decomposition analysis were performed to investigate the diffusion behavior of a typical IL, [C4mim][PF6], into the ZIF-8 SOD cage. The PES profiles indicate that it is quite difficult for the cation [C4min]+ to diffuse into the cage of ZIF-8 through the pristine pores because of the large imidazole steric hindrance, which results in a large energy barrier of ca. 40 kcal·mol−1 at the least. Interestingly, the PES reveals that a successful diffusion could be obtained by thermal contributions, which enlarge the pore size through swing effects at higher temperatures. For example, both [C4mim]+ and [PF6]− could easily diffuse through the channel of the ZIF-8 SOD cage when the pore size was increased to 6.9 Å. Subsequently, electronic structure analyses reveal that the main interactions between [PF6]− or [C4mim]+ and ZIF-8 are the steric repulsion interactions. Finally, the effects of the amounts of [C4mim][PF6] on the ZIF-8 structures were investigated, and the results show that two pairs of [C4mim][PF6] per SOD cage are the most stable in terms of the interaction between energies and structural changes. With these findings, we propose that the high-temperature technique could be employed during the synthesis of IL@MOF membranes, to enrich their family members and their industrial applications.

Abstract Background Tartary buckwheat (Fagopyrum tataricum) is an important food and medicine crop plant, which has been cultivated for 4000 years. A nuclear genome has been generated for this species, while an intraspecific pan-plastome has yet to be produced. As such a detailed understanding of the maternal genealogy of Tartary buckwheat has not been thoroughly investigated. Results In this study, we de novo assembled 513 complete plastomes of Fagopyrum and compared with 8 complete plastomes of Fagopyrum downloaded from the NCBI database to construct a pan-plastome for F. tartaricum and resolve genomic variation. The complete plastomes of the 513 newly assembled Fagopyrum plastome sizes ranged from 159,253 bp to 159,576 bp with total GC contents ranged from 37.76 to 37.97%. These plastomes all maintained the typical quadripartite structure, consisting of a pair of inverted repeat regions (IRA and IRB) separated by a large single copy region (LSC) and a small single copy region (SSC). Although the structure and gene content of the Fagopyrum plastomes are conserved, numerous nucleotide variations were detected from which population structure could be resolved. The nucleotide variants were most abundant in the non-coding regions of the genome and of those the intergenic regions had the most. Mutational hotspots were primarily found in the LSC regions. The complete 521 Fagopyrum plastomes were divided into five genetic clusters, among which 509 Tartary buckwheat plastomes were divided into three genetic clusters (Ft-I/Ft-II/Ft-III). The genetic diversity in the Tartary buckwheat genetic clusters was the greatest in Ft-III, and the genetic distance between Ft-I and Ft-II was the largest. Based on the results of population structure and genetic diversity analysis, Ft-III was further subdivided into three subgroups Ft-IIIa, Ft-IIIb, and Ft-IIIc. Divergence time estimation indicated that the genera Fagopyrum and Rheum (rhubarb) shared a common ancestor about 48 million years ago (mya) and that intraspecies divergence in Tartary buckwheat began around 0.42 mya. Conclusions The resolution of pan-plastome diversity in Tartary buckwheat provides an important resource for future projects such as marker-assisted breeding and germplasm preservation.

As a nutrient-rich multigrain crop, buckwheat is a typical “medicinal food homology” crop that is rich in flavonoids, including rutin and various vitamins. It has positive anti-oxidant and anti-tumour properties and lowers blood pressure. However, due to strict self-crossing characteristics, slow progress has been made in Tartary buckwheat (TB) cross-breeding, resulting in the slow breeding of new varieties of new TB varieties, which has limited the improvement of yield and quality. Therefore, mutant breeding is a rapid and effective technique for broadening and innovating TB breeding. In recent years, improving qualities related to yield, lodging resistance, and stability have become key points in TB breeding. Based on the above findings, excellent, potentially valuable TB lines with rich phenotypes were obtained for the TB mutation library via ethyl methanesulfonate (EMS), laying a foundation for creating new TB germplasms. In this study, we systematically investigated more than 10 agronomic traits of JQ2 and JQ4 mutants, including plant type, leaf colour, grain type, grain colour, grain number per plant, grain length, grain width, grain weight per plant, and 1000-grain weight. The results show that the maximum number of grains per plant was 1956, the weight was 32.84 g, and the 1000-grain weight was 30.89 g. The maximum number of grains per JQ4 plant was 2308, and the weight was 44.82 g. The maximum 1000-grain weight was 24.7 g. Among the 295 JQ2 mutants and 153 JQ4 mutants, 10 flavonoids (orientin, morin, quercetin, kaempferol, luteolin, naringin, hesperetin, myricetin, hesperidin, and rutin) were detected with near infrared spectroscopy (NIR). The mutants were divided into five groups according to the flavonoid content of the JQ2 mutants, of which the first group included 31 individual lines. and the second to fifth groups included 70, 69, 72, and 53 lines, respectively. The JQ4 mutants were divided into four classes, of which 41, 50, 32, and 30 were individual lines, respectively, with the highest rutin content being 82.06 mg/g. In summary, through systematic analysis and screening of the agronomic traits and flavonoid contents of JQ2 and JQ4 mutant seeds, we obtained three lines with a high 1000-grain weight, including two JQ2 mutant lines (30.89 g) and one JQ4 mutant line, which reached 24.70 g and ten lines with high grain weight per plant. This included 8 JQ2 mutants and 2 JQ4 mutants, as well as 72 high-rutin mutants (including 31 lines from JQ2 and 41 lines from JQ4 mutants). These elite lines provide the material basis for creating TB germplasms with excellent qualities and cultivation characteristics.

GFI1 is a transcriptional repressor and plays a pivotal role in regulating the differentiation of hematopoietic stem cells (HSCs) towards myeloid and lymphoid cells. Serial transplantation of Gfi1 deficient HSCs repopulated whole hematopoietic system but in a competitive setting involving wild-type HSCs, they lose this ability. The underlying mechanisms to this end are poorly understood. To better understand this, we used different mouse strains that express either loss of both Gfi1 alleles (Gfi1-KO), with reduced expression of GFI1 (GFI1-KD) or wild-type Gfi1/GFI1 (Gfi1-/GFI1-WT; corresponding to the mouse and human alleles). We observed that loss of Gfi1 or reduced expression of GFI1 led to a two to four fold lower number of HSCs (defined as Lin−Sca1+c-Kit+CD150+CD48−) compared to GFI1-WT mice. To study the functional influence of different levels of GFI1 expression on HSCs function, HSCs from Gfi1-WT (expressing CD45.1 + surface antigens) and HSCs from GFI1-KD or -KO (expressing CD45.2 + surface antigens) mice were sorted and co-transplanted into lethally irradiated host mice. Every 4 weeks, CD45.1+ and CD45.2 + on different lineage mature cells were analyzed by flow cytometry. At least 16 weeks later, mice were sacrificed, and the percentage of HSCs and progenitors including GMPs, CMPs and MEPs in the total bone marrow cells was calculated as well as their CD45.1 and CD45.2 expression. In the case of co-transplantation of GFI1-KD with Gfi1-WT HSCs, the majority of HSCs (81% ± 6%) as well as the majority of mature cells (88% ± 10%) originated from CD45.2 + GFI1-KD HSCs. In the case of co-transplantation of Gfi1-KO HSCs with Gfi1-WT HSCs, the majority of HSCs originated from CD45.2+ and therefore from Gfi1-KO (61% ± 20%); however, only a small fraction of progenitors and mature cells originated from Gfi1-KO HSCs (

Abstract The Huizhou Depression in the Pearl River Mouth basin has prospective hydrocarbon potential, with Miocene sandstones as its main oil and gas-bearing reservoir. The sandstones in Miocene formation of the Z21 offshore oil–gas field composed of medium-grained, moderately sorted subarkose and lithic arkose. In this study, a total of six depositional lithofacies, namely Massive fine- to medium-grained sandstone (Sm), ripple cross-laminated fine-grained sandstone (Sr), parallel-laminated siltstone and claystone (Fl), lenticular siltstone (Sl), parallel-bedded fine-grained sandstone (Sp), wavy laminated siltstone (Sw), and two depositional systems, namely nearshore sand bar (SB) and sand sheet (SS) were identified based on core observations and seismic study. Distributions of the porosity (13.9%) and permeability (35.8 mD) reveal that the Miocene sandstones have characteristics of low porosity and low permeability, with high heterogeneity. The sedimentary system, primary texture and diagenesis jointly control the reservoir quality. Sandstones with sand bars as well coarse-grained tend to exhibit a higher quality. Mechanical compaction and calcite (average 6.81%) cementation are the major determinants to reductions in porosity and permeability. The total clay minerals (average 5.27%) generally lead to reduction of porosity, whereas chlorite coatings and illite within a certain content range may enhance the preservation of porosity in eodiagenesis. Dissolution of feldspar and debris contribute significantly to improving the reservoir quality.

The aim of this study was to investigate the circadian rhythm of muscle-related gene expression in mackerel tuna under different weather conditions. The experiment was carried out under two weather conditions at four sampling times (6:00, 12:00, 18:00, and 24:00) to determine the expression of growth, function, and rhythm genes: white muscle rhythm genes were rhythmic on sunny and cloudy days, except for PER3 and RORA; all functional genes had daily rhythmicity. Red muscle had daily rhythmicity on both sunny and cloudy days; functional genes had daily rhythmicity except for MBNL. The expression levels of the rhythm gene PER1 were determined to be significantly different by independent t-test samples in white muscle at 6:00, 12:00, 18:00, and 24:00 under different weather conditions; the expression levels of the functional genes MBNL and MSTN were both significantly different. In the red muscle, the expression of the rhythm genes PER3, REVERBA, and BMAL1 was determined by independent t-test samples at 6:00, 12:00, 18:00, and 24:00 on cloudy and sunny days; the functional gene MBNL was significantly different. The present study showed that mackerel tuna muscle rhythm genes and functional genes varied significantly in expression levels depending on weather, time of day, and light intensity and that the expression levels of myogenic genes were closely related to clock gene expression. The fish were also able to adapt to changes in light intensity in different weather conditions through positive physiological regulation.

Rhabdomyosarcoma (RMS) is a soft tissue tumor, which is the most common in the head, neck, limbs, and trunk. RMS originating from the epididymis is extremely rare. Herein, we reported a 34-year-old patient with RMS on the right epididymis. For this case, right epididymal mass resection was performed and intraoperative freezing suggested a malignant tumor. Right testicular radical resection was subsequently adopted, with right epididymal alveolar RMS being pathologically diagnosed. Alternating VAC/VI chemotherapy was given after surgery, and tumor recurrence has not been found so far.

The zinc finger protein Growth Factor Independence 1 (GFI1) acts as a transcriptional repressor regulating differentiation of myeloid and lymphoid cells. A single nucleotide polymorphism of GFI1, GFI1-36N, has a prevalence of 7% in healthy Caucasians and 15% in acute myeloid leukemia (AML) patients, hence most probably predisposing to AML. One reason for this is that GFI1-36N differs from the wildtype form GFI1-36S regarding its ability to induce epigenetic changes resulting in a derepression of oncogenes. Using proteomics, immunofluorescence, and immunoblotting we have now gained evidence that murine GFI1-36N leukemic cells exhibit a higher protein level of the pro-proliferative protein arginine N-methyltransferase 5 (PRMT5) as well as increased levels of the cell cycle propagating cyclin-dependent kinases 4 (CDK4) and 6 (CDK6) leading to a faster proliferation of GFI1-36N leukemic cells in vitro. As a therapeutic approach, we subsequently treated leukemic GFI1-36S and GFI1-36N cells with the CDK4/6 inhibitor palbociclib and observed that GFI1-36N leukemic cells were more susceptible to this treatment. The findings suggest that presence of the GFI1-36N variant increases proliferation of leukemic cells and could possibly be a marker for a specific subset of AML patients sensitive to CDK4/6 inhibitors such as palbociclib.

Abstract Background Tartary buckwheat is an important minor crop species with high nutritional and medicinal value and is widely planted worldwide. Cultivated Tartary buckwheat plants are tall and have hollow stems that lodge easily, which severely affects their yield and hinders the development of the Tartary buckwheat industry. Methods Heifeng No. 1 seeds were treated with ethylmethanesulfonate (EMS) to generate a mutant library. The dwarf mutant ftdm was selected from the mutagenized population, and the agronomic characteristics giving rise to the dwarf phenotype were evaluated. Ultra-fast liquid chromatography-electrospray ionization tandem mass spectrometry (UFLC-ESI–MS/MS) was performed to determine the factors underlying the different phenotypes between the wild-type (WT) and ftdm plants. In addition, RNA sequencing (RNA-seq) was performed via the HiSeq 2000 platform, and the resulting transcriptomic data were analysed to identify differentially expressed genes (DEGs). Single-nucleotide polymorphism (SNP) variant analysis revealed possible sites associated with dwarfism. The expression levels of the potential DEGs between the WT and ftdm mutant were then measured via qRT-PCR and fragments per kilobase of transcript per million mapped reads (FPKM). Result The plant height (PH) of the ftdm mutant decreased to 42% of that of the WT, and compared with the WT, the mutant and had a higher breaking force (BF) and lower lodging index (LI). Lower GA4 and GA7 contents and higher contents of jasmonic acid (JA), salicylic acid (SA) and brassinolactone (BR) were detected in the stems of the ftdm mutant compared with the WT. Exogenous application of GAs could not revert the dwarfism of the ftdm mutant. On the basis of the transcriptomic analysis, 146 homozygous SNP loci were identified. In total, 12 DEGs with nonsynonymous mutations were ultimately identified, which were considered potential candidate genes related to the dwarf trait. When the sequences of eight genes whose expression was downregulated and four genes whose expression was upregulated were compared, SKIP14, an F-box protein whose sequence is 85% homologous to that of SLY1 in Arabidopsis, presented an amino acid change (from Ser to Asn) and was expressed at a lower level in the stems of the ftdm mutant compared with the WT. Hence, we speculated that this amino acid change in SKIP14 resulted in a disruption in GA signal transduction, indirectly decreasing the GA content and downregulating the expression of genes involved in GA biosynthesis or the GA response. Further studies are needed to determine the molecular basis underlying the dwarf phenotype of the ftdm mutant. Conclusion We report a Tartary buckwheat EMS dwarf mutant, ftdm, suitable for high-density planting and commercial farming. A significant decrease in GA4 and GA7 levels was detected in the ftdm mutant, and 12 DEGs expressed in the stems of the ftdm mutant were selected as candidates of the dwarfing gene. One nonsynonymous mutation was detected in the SKIP14 gene in the ftdm mutant, and this gene had a lower transcript level compared with that in the WT.

Abstract Background Acute pancreatitis is a sudden inflammation of the pancreas. Although interleukin‐23 (IL‐23) is associated with the severity of acute pancreatitis, the underlying mechanism remains largely unknown. Herein, its regulatory mechanisms were explored in this study. Methods RNA‐sequencing analysis selected the differently expressed genes in cerulean‐induced acute pancreatitis mice. Polymerase chain reaction analysis determined IL‐23 expression in cyclin‐dependent kinase 2 (Cdk2) short hairpin RNA (shRNA)‐pretreated or DDB1–cullin‐4‐associated factor‐2 (DCAF2)‐overexpressed RAW264.7 cells or CDKs inhibitor AT7519/cullin ring‐finger ubiquitin ligase inhibitor MLN4924‐treated bone marrow‐derived macrophages in the presence of lipopolysaccharides (LPS). Pancreatic damages were evaluated in AT7519‐treated pancreatitis mice. Results Pancreatitis mice displayed an increased expression on IL‐23 and a decreased expression of Cdk2. Inhibiting Cdk2 by shRNA or AT7519 significantly induced IL‐23 expression in LPS‐treated RAW cells. Moreover, AT7519 treatment significantly aggravated the severity of acute pancreatitis in mice. Furthermore, AT7519 remarkably increased DCAF2 expression, which was also induced by MLN4924 no matter with or without AT7519 in vitro. On the contrary, overexpressing DCAF2 blocked the stimulatory effect of AT7519 on IL‐23 expression. Conclusion Cdk2 negatively regulates IL‐23 expression by inhibiting DCAF2 in acute pancreatitis, indicating that Cdk2 might serve as a promising therapeutic target for acute pancreatitis.

Due to the existence of system noise and unknown state variables, it is difficult to realize unbiased estimation with minimum variance for the parameter estimation of canonical state space model. This paper presents a new least squares estimator based on bias compensation principle to solve this problem, transforms canonical state space into the form suitable for the least square algorithm, introduces an augmented parameter vector and an auxiliary variable, derives parameter estimation formula based on noise compensation, realizes the unbiased estimation, and gives the specific algorithm. A simulation example is provided to verify the effectiveness of the estimator.

Abstract Background Male sterility (MS) has important applications in hybrid seed production, and the abortion of anthers has been observed in many plant species. While most studies have focused on the genetic factors affecting male sterility, the dynamic gene expression patterns of pollen abortion in male sterile lines have not been fully elucidated. In addition, there is still no hybrid oat that is commercially planted due to the lack of a suitable system of male sterility for hybrid breeding. Results In this study, we cultivated a male sterile oat line and a near-isogenic line by crossbreeding to elucidate the expression patterns of genes that may be involved in sterility. The first reported CA male sterile (CAMS) oat line was used for cross-testing and hybridization experiments and was confirmed to exhibit a type of nuclear sterility controlled by recessive genes. Oat stamens of two lines were sampled at four different developmental stages separately. Paired-end RNA sequencing was performed for each sample and generated 252.84 Gb sequences. There were 295,462 unigenes annotated in public databases in all samples, and we compared the histological characteristics and transcriptomes of oat stamens from the two oat lines at different developmental stages. Our results demonstrate that the sterility of the male sterile oat line occurs in the early stage of stamen development and is primarily attributable to abnormal meiosis and the excessive accumulation of superoxide. Conclusions To the best of our knowledge, this study is the first to decipher the dynamic expression profiles of pollen abortion CAMS and CA male fertile (CAMF) oat lines, which may represent a valuable resource for further studies attempting to understand pollen abortion and anther development in oats.

Abstract Spikelet rot disease (SRD) is an emerging disease of the grain surface of naked oat in China that affects both grain yield and quality. The typical symptom is discoloration from the black structures of the causal fungi. Here, we investigated the fungal communities on the grain surfaces of cultivar Bayou 13 grown in ten ecological oat-producing regions of China, to identify the main pathogens of naked oat SRD. Our results showed that the growth of Alternaria spp. and Davidiella spp. exhibited a competitive relationship and was mainly affected by the elevations of all 10 ecological regions. The dominant pathogens were Davidiella spp. in Shannan Prefecture in Tibet and Haidong Prefecture in Qinghai Province and Alternaria spp. in the other eight regions. The ratios of black pathogens of interest to all pathogens in Shannan Prefecture and Haidong Prefecture were significantly lower than those of the other eight regions, thus indicating that SRD mainly occurred in regions below 2000 m (elevation). We isolated black fungal pathogens from grain surfaces and deduced that they were Alternaria spp. by sequence comparison. The blackened appearance of the grain surfaces was more evident under spray inoculation with a spore suspension of Alternaria than under the control in greenhouse experiments. The recovered pathogen was the same as the pathogen used for inoculation. We thus concluded that Alternaria alone causes naked oat SRD and mainly infects naked oat in regions below 2000 m, which provides a basis for the recognition and management of SRD of naked oat.

Silymarin and milk thistle oil have unique biological benefits; however, applying silymarin to milk thistle oil remains a challenge. In this research, the content of silymarin in milk thistle oil conditions using enzyme-mediated solvent extraction was investigated and optimized by response surface methodology. The optimal extraction conditions using enzyme-mediated solvent extraction were as follows: the enzyme-added content was 3.06 mg/mL, the enzymatic hydrolysis temperature was 55.09 °C, and the enzymatic hydrolysis time was 66.28 min. Oil extracted by the enzyme-mediated assisted solvent was further compared with those extracted with n-hexane and cold pressing. Results indicated that the oil extraction using the enzyme-mediated assisted solvent had a lower acid value (2.20 ± 0.01 mg/g) and the highest α-tocopherol content (0.62 ± 0.00 mg/g), total phenols (7.67 ± 0.01 mg/g), and flavonoids (1.06 ± 0.13 mg/g). Furthermore, the antioxidant capacity of milk thistle oils was further investigated. The results showed that the enzyme-mediated assisted solvent-extracted oil had the strongest antioxidant capacity with lower lipid oxide content. Therefore, enzyme-mediated solvent extraction is an excellent method for extracting milk thistle oil.

Exploring high-efficiency, low-cost, and long-life bifunctional self-supporting electrocatalysts is of great significance for the practical application of advanced rechargeable Zn-air batteries (ZABs), especially flexible solid-state ZABs. Herein, ultrathin CoFe-layered double hydroxide (CoFe-LDH) nanosheets are strongly coupled on the surface of leaf-like bimetallic metal-organic frameworks (MOFs)-derived hybrid carbon (Co@NC) nanoflake nanoarrays supported by carbon cloth (CC) via a facile and scalable method for rechargeable and flexible ZABs. This interfacial engineering for CoFe-LDHs on Co@NC improves the electronic conductivity of CoFe-LDH nanosheets as well as achieves the balance of oxygen evolution reduction (OER) and oxygen reduction reaction (ORR) activity. The unique three-dimensional (3D) open interconnected hierarchical structure facilitates the transport of substances during the electrochemical process while ensuring adequate exposure of OER/ORR active centers. When applied as an additive-free air cathode in rechargeable liquid ZABs, CC/Co@NC/CoFe-LDH-700 demonstrates high open-circuit potential of 1.47 V, maximum power density of 129.3 mW cm−2, and satisfactory specific capacity of 710.7 mAh g−1Zn. Further, the flexible all-solid-state ZAB assembled by CC/Co@NC/CoFe-LDH-700 displays gratifying mechanical flexibility and stable cycling performance over 40 h. More significantly, the series-connected flexible ZAB is further verified as a chain power supply for LED strips and performs well throughout the bending process, showing great application prospects in portable and wearable electronics. This work sheds new light on the design of high-performance self-supporting non-precious metal bifunctional electrocatalysts for OER/ORR and air cathodes for rechargeable ZABs.

Tartary buckwheat (TB; Fagopyrum tataricum Gaertn.) is an important multigrain crop and medicinal plant, but functional genomics and molecular breeding research in this species have been lacking for quite some time. Here, genome-wide screening was performed to develop simple sequence repeat (SSR) markers associated with six major agronomic traits and the rutin contents of 97 core germplasm resources. A total of 40,901 SSR loci were identified; they were uniformly distributed throughout the TB genome, with a mean distance of 11 kb between loci. Based on these loci, 8,089 pairs of SSR primers were designed, and 101 primer pairs for polymorphic SSR loci were used to genotype the 97 core germplasm resources. The polymorphic SSR loci showed high genetic variation in these core germplasm resources, with an average polymorphic information content (PIC) value of 0.48. In addition, multiple SSR markers, such as SXAU8002 [100-grain weight (HGW)] and SXAU8006 [stem diameter (SD)], were found to be associated with agronomic traits in the two environments. Finally, based on gene functional annotation and homology analysis, a candidate gene, FtPinG0007685500, that may affect the node number and SD of the main stem by participating in lignin synthesis was identified. This study reports the mining of genome-wide SSR loci and the development of markers in TB, which can be used for molecular characterization of the germplasm in its gene pool. In addition, the detected markers and candidate genes could be used for marker-assisted breeding and functional gene cloning in TB.

This article introduces some experimental data from the compression test, the tension test, and the temperature stress test machine (TSTM) test for low-heat Portland (LHP) cement concrete with different fly ash (FA) content. With the proposed equations, the thermal cracks can be predicted with the TSTM test along with the mechanical properties. The higher FA content exhibits a lower hydration degree and a smaller adiabatic temperature rise. Although the FA content decreases the hydration heat and the strength at an early age, there are few influences on the relational expression between compressive strength and tensile strength. The proposed equations can also be applied to estimate the thermal cracks risk in LHP cement concrete. The cracking stress of LHP cement concrete in the temperature stress test is not only related to the tensile stress history but also connected to the compressive stress history. Considering the strength history, an analytical method for evaluating the cracking temperature difference was proposed. With the proposed method, only compressive strength can be used to estimate the thermal cracks risk for LHP cement concrete at different ages. It greatly simplifies the evaluation method of thermal cracks in low-heat Portland cement concrete. Furthermore, this evaluation method can also be applied to other types of concrete.

Leaf nutrient content and its stoichiometric relationships (N/P ratio) are essential for photosynthesis and plant growth and development. Previous studies on leaf nutrient-related functional traits have mainly focused on the species level and regional scale, but fewer studies have investigated the distribution patterns of the leaf N and P contents (LN, LP) and N/P ratios (N/P) in communities and their controlling factors at a large scale; therefore, we used LN, LP, and N/P data at 69 sites from 818 forests in China. The results showed significant differences (p < 0.05) in the LN, LP, and N/P at different life forms (tree, shrub, and herb). Neither LN, LP, nor N/P ratios showed significant patterns of latitudinal variation. With the increase in temperature and rainfall, the LN, LP, and leaf nutrient contents increased significantly (p < 0.001). Across life forms, LN at different life forms varied significantly and was positively correlated with soil P content (p < 0.001). The explanatory degree of climatic factors in shaping the spatial variation patterns of LN and N/P was higher than that of the soil nutrient factors, and the spatial variation patterns of the leaf nutrient traits of different life forms were shaped by the synergistic effects of climatic factors and soil nutrient factors.

Iron-manganese co-oxide film (MeOx) has a high removal efficiency for ammonium (NH4+) and manganese (Mn2+) in our previous studies, but it cannot effectively remove CODMn from water. In this study, the catalytic oxidation ability of MeOx was enhanced by dosage with potassium ferrate (K2FeO4) to achieve the simultaneous removal of CODMn and NH4+ from water in a pilot-scale experimental system. By adding 1.0 mg/L K2FeO4 to enhance the activity of MeOx, the removal efficiencies of CODMn (20.0 mg/L) and NH4+ (1.1 mg/L) were 92.5 ± 1.5% and 60.9 ± 1.4%, respectively, and the pollutants were consistently and efficiently removed for more than 90 days. The effects of the filtration rate, temperature and pH on the removal of CODMn were also explored, and excessive filtration rate (over 11 m/h), lower temperature (below 9.2 °C) and pH (below 6.20) caused a significant decrease in the removal efficiency of CODMn. The removal of CODMn was analyzed at different temperatures, which proved that the kinetics of CODMn oxidation was pseudo-first order. The mature sands (MeOx) from column IV were taken at different times for microscopic characterization. Scanning electron microscope (SEM) showed that some substances were formed on the surface of MeOx and the ratio of C and O elements increased significantly, and the ratio of Mn and Fe elements decreased significantly on the surface of MeOx by electron energy dispersive spectrometer (EDS). However, the elemental composition of MeOx would gradually recover to the initial state after the dosage of Mn2+. According to X-ray photoelectron spectroscopy (XPS) analysis, the substance attached to the surface of MeOx was [(-(CH2)4O-)n], which fell off the surface of MeOx after adding Mn2+. Finally, the mechanism of K2FeO4-enhanced MeOx for CODMn removal was proposed by the analysis of the oxidation process.

Background/Aims: Disseminated tumors, known as metastases, are responsible for ninety-percent of mortality due to cancer. Epithelial to mesenchymal transition, a phenomenon required for morphological conversion of non-motile discoid shaped epithelial cells to highly motile spindle-shaped mesenchymal cells, is thought to be a pre-requisite for metastatic progression. Metastasis-associated 1 (MTA1) protein is a prime inducer of EMT and metastatic progression in all solid tumors including hepatocellular carcinoma (HCC). However, the molecular mechanisms that regulate the expression and function of MTA1 in HCC have not been elucidated. Methods: In silico prediction algorithms were used to find microRNAs (miRNAs) that may target MTA1. We examined the relationship between the expression of MTA1 and miR-183 using quantitative real time PCR. We also determined the levels of the MTA1 protein using immunohistochemistry. Reporter assays, in the presence and absence of the miR-183 mimic, were used to confirm MTA1 as a bona fide target of miR183. The effect of miR-183 on HCC pathogenesis was determined using a combination of in vitro migration and invasion assay, together with in vivo xenograft experiments. The correlation between miR-183 and MTA1 expression was also studied in samples from HCC patients, and in The Cancer Genome Atlas dataset. Results: Analysis of the sequence database revealed that MTA1 is a putative target of miR-183. MTA1 protein and RNA expression showed opposite trends to miR-183 expression in breast, renal, prostate, and testicular tissue samples from cancer patients, and in the metastatic HCC cell line HepG2. An inverse correlation was also observed between MTA1 (high) and miR-183 (low) expression within samples from HHC patients and in the TCGA dataset. Reporter assays in HepG2 cells showed that miR-183 could inhibit translation of a reporter harboring the wild-type, but not the mutant miR-183 3’-untranslated region (UTR). In addition, miR-183 significantly inhibited in vitro migration and invasion in HepG2 cells, and in vivo hepatic metastasis. Conclusion: Our results reveal a novel post-transcriptional regulatory mechanism for MTA1 expression via miR-183, which is suppressed during HCC pathogenesis.

Acute myeloid leukemia (AML) is a group of hematological cancers with metabolic heterogeneity. Oxidative phosphorylation (OXPHOS) has been reported to play an important role in the function of leukemic stem cells and chemotherapy-resistant cells and are associated with inferior prognosis in AML patients. However, the relationship between metabolic phenotype and genetic mutations are yet to be explored. In the present study, we demonstrate that AML cell lines have high metabolic heterogeneity, and AML cells with MLL/AF9 have upregulated mitochondrial activity and mainly depend on OXPHOS for energy production. Furthermore, we show that metformin repressed the proliferation of MLL/AF9 AML cells by inhibiting mitochondrial respiration. Together, this study demonstrates that AML cells with an MLL/AF9 genotype have a high dependency on OXPHOS and could be therapeutically targeted by metformin.

Growth Factor Independence 1 (GFI1) is a transcription factor with an important role in the regulation of development of myeloid and lymphoid cell lineages and was implicated in the development of myelodysplastic syndrome (MDS) and acute myeloid leukaemia (AML). Reduced expression of GFI1 or presence of the GFI1-36N (serine replaced with asparagine) variant leads to epigenetic changes in human and murine AML blasts and accelerated the development of leukaemia in a murine model of human MDS and AML. We and other groups previously showed that the GFI1-36N allele or reduced expression of GFI1 in human AML blasts is associated with an inferior prognosis. Using GFI1-36S, -36N -KD, NUP98-HOXD13-tg mice and curcumin (a natural histone acetyltransferase inhibitor (HATi)), we now demonstrate that expansion of GFI1-36N or –KD, NUP98-HODXD13 leukaemic cells can be delayed. Curcumin treatment significantly reduced AML progression in GFI1-36N or -KD mice and prolonged AML-free survival. Of note, curcumin treatment had no effect in GFI1-36S, NUP98-HODXD13 expressing mice. On a molecular level, curcumin treatment negatively affected open chromatin structure in the GFI1-36N or -KD haematopoietic cells but not GFI1-36S cells. Taken together, our study thus identified a therapeutic role for curcumin treatment in the treatment of AML patients (homo or heterozygous for GFI1-36N or reduced GFI1 expression) and possibly improved therapy outcome.

The gasification experiments of coal chars with CO2 were carried out isothermally, with K, Ca, Ni, and Zn chloride catalysts, adopting a thermal gravimetric analyzer (TGA) from 800 to 1100 °C. The kinetic characteristic of the samples were described using the volumetric model (VM), the grain model (GM), and the random pore model (PRM). The morphology patterns of the samples were tested applying X-ray diffraction (XRD) and the catalytic mechanisms concerning the phase changes were proposed. The results confirm that the gasification rate and char reactivities are enhanced by K, Ca and Ni chlorides, while ZnCl2 inhibited the process. The catalysis ability shows the following cation order: Ca > K > Ni > Zn. Among the models described above, PRM was proven to give the best fitting value and hence adopted to kinetics parameters calculation. The activation energies in promoting conditions were lower than that of the uncatalyzed cases. In view of the catalytic mechanism, the K metals tend to form intermediate complexes and repeatedly connect with coal char, while the Ca species may follow the oxidation-reduction mechanism and the Ni metals catalyze the gasification process.

As a highly nutritious crop, Tartary buckwheat (Fagopyrum tartaricum) strongly adapts and grows in adverse environments and is widely grown in Asia. However, its flour contains a large proportion of the hull that adheres to the testa layer of the groats and is difficult to be removed in industrial processing. Fortunately, rice-Tartary, with the loose and non-adhering hull, provides potentiality of improving Tartary buckwheat that can dehull easily. Here, we performed high-throughput sequencing for two parents (Tartary buckwheat and rice-Tartary) and two pools (samples from the F2 population) and obtained 101 Gb raw sequencing data for further analysis. Sequencing reads were mapped to the reference genome of Tartary buckwheat, and a total of 633,256 unique SNPs and 270,181 unique indels were found in these four samples. Then, based on the Bulked Segregant Analysis (BSA), we identified a candidate genetic region, containing 45 impact SNPs/indels and 36 genes, that might underly non-adhering hull of rice-Tartary and should have value for breeding easy dehulling Tartary buckwheat.

In this paper, a gradient descent algorithm is proposed for the parameter estimation of multi-input and multi-output (MIMO) total non-linear dynamic models. Firstly, the MIMO total non-linear model is mapped to a non-completely connected feedforward neural network, that is, the parameters of the total non-linear model are mapped to the connection weights of the neural network. Then, based on the minimization of network error, a weight-updating algorithm, that is, an estimation algorithm of model parameters, is proposed with the convergence conditions of a non-completely connected feedforward network. In further determining the variables of the model set, a method of model structure detection is proposed for selecting a group of important items from the whole variable candidate set. In order to verify the usefulness of the parameter identification process, we provide a virtual bench test example for the numerical analysis and user-friendly instructions for potential applications.

This paper presented an experimental study on the flexural properties of engineered cementitious composites (ECCs). The bending fatigue damage, residual deformation, and damage characteristics were investigated after a certain number of low stress levels in fatigue load. The composite fracture energy and fiber-bridging fracture energy were calculated by the J integral. It is observed that the number of cracks increased with the increment of stress levels, and most of the cracks were formed during the earlier stage of the dynamic test. The deformation capability decreased with the increment of stress levels while the reduction of the ultimate load was minor after the dynamic load. Furthermore, the strain-hardening phenomenon of the specimen enhanced initially and then weakened with the increment of stress levels. The residual equivalent yield strength became smaller with the increase of stress levels. Meanwhile, the trend was mild at low stress levels and then became steep at high stress levels.

The complete mitochondrial genome of Buffon’s river garfish Zenarchopterus buffonis from South China Sea has been obtained for the first time. It is 16,462 bp long, and contains 21 tRNA genes, 2 rRNA genes, 13 protein-coding genes (PCGs), and 1 control region. The overall base composition is significantly biased (A, G, T, and C was 33%, 12.94%, 29.78%, and 24.27%, respectively) with A + T contents of 62.79%. In total, most PCGs use the initiation codon ATG except COX1 uses GTG. Seven PCGs have a complete codon TAA or TAG, whereas the other six genes end with the incomplete stop codon T–– or TA–. Phylogenetic analysis showed that Z. buffonis forms a monophyletic cluster with the species of Belonidae, Zenarchopteridae, and Sconberesocidae.

Aggressive natural killer cell leukemia (ANKL) is a fatal hematological neoplasm characterized by a fulminating clinical course and extremely high mortality. Current diagnosis of this disease is not effective during the early stages and it is easily misdiagnosed as other NK cell disorders. We retrospectively analyzed the clinical characteristics and flow cytometric immunophenotype of 47 patients with ANKL. Patients with extranodal NK/T cell lymphoma, nasal type (ENKTL) and chronic lymphoproliferative disorder of NK cell (CLPD-NK), who were diagnosed during the same time period were used for comparisons. A new temperature stress test machine (TSTM) was developed to improve temperature control accuracy and efficiency. As there is no uniform standard for the threshold value of TSTM, quite different threshold values are used in the test, which results in the influences of the threshold value on the evaluation of thermal stresses and crack risk of concrete not being determined. To illustrate the importance of an appropriate threshold value, different threshold values were used to evaluate the influences on the thermal stresses and cracking resistance of concrete specimens under a complete restraint test with semi-adiabatic temperature development. The results show that the maximum compressive stress of a concrete specimen with a threshold value 3 με was slightly larger than that of 1.5 με when the growth rate of tensile stress of the concrete specimen with the threshold value 3 με was slightly greater than that of concrete specimen with the threshold value 1.5 με. Based on the combination of crack risk coefficient and restraint degree, a new system for evaluating the crack resistance of concrete was proposed, in which different threshold values were used to estimate their influences on the crack risk of concrete. Thus, an appropriate range of threshold values could be determined.

In this study, three-dimensional (3-D) geostatistical models were constructed to quantify distributions of sandstone and mudstone. We propose a new method that employs weight coefficients to balance the sandstone and mudstone data from irregular well patterns during stochastic modeling. This new method begins with classifying well groups according to well distribution patterns; areas with similar well distribution patterns are classified within the same zone. Then, the distributions of sandstone and mudstone for each zone are simulated separately using the sequential indicator simulation (SIS) method, and the relevant variogram parameters for each zone are computed. In this paper, we used block S6 of the Sulige Gas Field in Ordos Basin in China as a case study. We evaluated the quality of each set of parameters through the vacuation checking method; certain wells were removed to generate equiprobable realizations using different seed numbers. Subsequently, the variogram parameters for the entire S6 area were obtained by assigning different weight coefficients to the parameters of each zone. Finally, a quality assessment of the sandstone and mudstone models of the S6 area was conducted using the horizontal wells, which were not involved in the stochastic modeling process. The results show that these variogram parameters, which were calculated using weight coefficients, are reliable.

Coral reefs occupy a relatively small portion of sea area, yet serve as a crucial source of biodiversity by establishing harmonious ecosystems with marine plants and animals. Previous researches mainly focused on screening several key genes induced by stress. Here we proposed a novel method--correlation analysis after wavelet transform of complex network model, to explore the effect of light on gene expression in the coral Acropora millepora based on microarray data. In this method, wavelet transform and the conception of complex network were adopted, and 50 key genes with large differences were finally captured, including both annotated genes and novel genes without accurate annotation. These results shed light on our understanding of coral's response toward light changes and the genome-wide interaction among genes under the control of biorhythm, and hence help us to better protect the coral reef ecosystems. Further studies are needed to explore how functional connections are related to structural connections, and how connectivity arises from the interactions within and between different systems. The method introduced in this study for analyzing microarray data will allow researchers to explore genome-wide interaction network with their own dataset and understand the relevant biological processes.
