Abstract

Sweet basil (Ocimum basilicum) leaves are rich in bioactive compounds that present therapeutic benefits for human health. Ultrasonic-assisted extraction (UAE) is frequently used to obtain phenolic compounds from plants/herbal sources. However, few works have developed multi-variable studies to find the optimal conditions to extract the maximum amount of compounds, especially when applied to UAE via a sonotrode. The purpose of this work was to perform a multi-variable study by employing a Box–Behnken design to collect the highest active compound content from Ocimum basilicum leaves. The efficacy of the design was endorsed by ANOVA. The studied parameters for UAE via a sonotrode were the ethanol/water ratio, amplitude, and time. The analyzed responses were the rosmarinic acid, the sum of phenolic acids, and the sum of phenolic compounds content. The optimal conditions were found to be 50% ethanol/water, 50% amplitude, and 5 min. Twenty bioactive compounds were identified by HPLC-ESI-TOF-MS when the extract was collected by applying the optimal conditions. Ocimum basilicum may be appreciated as a valuable source of important bioactive substances for pharmaceutical use. [ABSTRACT FROM AUTHOR]

43. Exploring Efavirenz supramolecular complexes to reduce Staphylococcus aureusvirulence and resistance factors

Abstract

Antimicrobial resistance represents a significant global challenge that demands the development of innovative pharmaceutical strategies. Efavirenz, an antiviral drug, also exhibits antivirulence and antibiofilm effects on Staphylococcus aureus. However, efavirenz oral bioavailability is low and variable, limited by its poor aqueous solubility. In this study, the complexation of efavirenz with α-cyclodextrin and meglumine were investigated to identify an optimized delivery system. The characterization of the solids by thermal analysis, scanning electron microscopy, electron probe microanalysis and X-ray diffraction demonstrated the efficacy of the kneading method for the preparation of a multicomponent complex. Dissolution of efavirenz from the supramolecular powders into water containing 1 % sodium lauryl sulfate was measured by UV/Vis absorption spectroscopy. Results show that the complexation leads to the formation of a solid with a significantly better dissolution profile and facilitates the attainment of efavirenz concentration in aqueous solution that exceeds that achieved by dissolving the pure compound. Furthermore, it was observed that the complex reduces the biofilm formation and hemolysis caused by S. aureus. In conclusion, the complexation of efavirenz with α-cyclodextrin and meglumine might represent an effective strategy for the development of a formulation with an enhanced dissolution profile and promising reduction in virulence and biofilm formation by S. aureus.
