However, the significance of the TRIM family of E3 ubiquitin ligases in head and neck squamous cell carcinoma (HNSCC) has not been determined. In this study, the roles of TRIM E3 ubiquitin ligases in lymphovascular invasion in head and neck squamous cell carcinoma (HNSCC) were evaluated.Materials and Methods: TRIM expression and related parameters were obtained from UbiBrowser2.0, UALCAN, TIMER, TISIDB, LinkedOmics, STRING, and GeneMANIA databases. Immunohistochemistry was used to confirm their expression.Results: TRIM2, TRIM11, TRIM28, and TRIM56 were upregulated in HNSCC with lymphovascular invasion. TRIM expression was strongly associated with immune infiltration, including key treatment targets, like PD-1 and CTL4. Co-expressed genes and possible ubiquitination substrates included tumor-related factors. The TRIMs had predicted roles in ubiquitination-related pathways and vital signaling pathways, eg, MAPK, PI3K-Akt, and JAK-STAT signaling pathways.Conclusion: Ubiquitination mediated by four TRIMs might be involved in the regulation of tumor immunity, laying the foundation for future studies of the roles of the TRIM family on the prediction and personalized medicine in HNSCC. In addition to standard medication treatment, participants in the experimental group with a low-lectin diet, while those in the control group received standard medication treatment alone. Clinical outcomes were assessed through evaluations conducted by physicians and teachers, implementation of the Conners Parent Rating Scales, and analysis of gut microbiota composition.Results: The results revealed significant improvements in symptom reduction and attention allocation rate within the experimental group, surpassing those observed in the control group. Specifically, the experimental group exhibited lower physician ratings, teacher ratings, and attention allocation rate compared to the control group. Moreover, analysis of gut microbiota composition identified notable distinctions between the two groups.Conclusion: These findings provide compelling evidence and valuable guidance supporting the integration of a low-lectin diet as an adjunctive intervention for managing ADHD.Keywords: ADHD, gut microbiota, low-lectin diet, integrative intervention, children

We present 97 new high-quality strong lensing candidates found in the final $\sim 350\,\rm deg^2$, that completed the full $\sim 1350\,\rm deg^2$ area of the Kilo-Degree Survey (KiDS). Together with our previous findings, the final list of high-quality candidates from KiDS sums up to 268 systems. The new sample is assembled using a new Convolutional Neural Network (CNN) classifier applied to $r$-band (best seeing) and $g,~r,~i$ color-composited images separately. This optimizes the complementarity of the morphology and color information on the identification of strong lensing candidates. We apply the new classifiers to a sample of luminous red galaxies (LRGs) and a sample of bright galaxies (BGs) and select candidates that received a high probability to be a lens from the CNN ($P_{\rm CNN}$). In particular, setting $P_{\rm CNN}>0.8$ for the LRGs, the $1$-band CNN predicts 1213 candidates, while the $3$-band classifier yields 1299 candidates, with only $\sim$30\% overlap. For the BGs, in order to minimize the false positives, we adopt a more conservative threshold, $P_{\rm CNN} >0.9$, for both CNN classifiers. This results in 3740 newly selected objects. The candidates from the two samples are visually inspected by 7 co-authors to finally select 97 "high-quality" lens candidates which received mean scores larger than 6 (on a scale from 0 to 10). We finally discuss the effect of the seeing on the accuracy of CNN classification and possible avenues to increase the efficiency of multi-band classifiers, in preparation of next-generation surveys from ground and space., Comment: Published by APJ

Synthesizing realistic medical images provides a feasible solution to the shortage of training data in deep learning based medical image recognition systems. However, the quality control of synthetic images for data augmentation purposes is under-investigated, and some of the generated images are not realistic and may contain misleading features that distort data distribution when mixed with real images. Thus, the effectiveness of those synthetic images in medical image recognition systems cannot be guaranteed when they are being added randomly without quality assurance. In this work, we propose a reinforcement learning (RL) based synthetic sample selection method that learns to choose synthetic images containing reliable and informative features. A transformer based controller is trained via proximal policy optimization (PPO) using the validation classification accuracy as the reward. The selected images are mixed with the original training data for improved training of image recognition systems. To validate our method, we take the pathology image recognition as an example and conduct extensive experiments on two histopathology image datasets. In experiments on a cervical dataset and a lymph node dataset, the image classification performance is improved by 8.1% and 2.3%, respectively, when utilizing high-quality synthetic images selected by our RL framework. Cervical intraepithelial neoplasia (CIN) grade of histopathology images is a crucial indicator in cervical biopsy results. Accurate CIN grading of epithelium regions helps pathologists with precancerous lesion diagnosis and treatment planning. Although an automated CIN grading system has been desired, supervised training of such a system would require a large amount of expert annotations, which are expensive and time-consuming to collect. In this paper, we investigate the CIN grade classification problem on segmented epithelium patches. We propose to use conditional Generative Adversarial Networks (cGANs) to expand the limited training dataset, by synthesizing realistic cervical histopathology images. While the synthetic images are visually appealing, they are not guaranteed to contain meaningful features for data augmentation. To tackle this issue, we propose a synthetic-image filtering mechanism based on the divergence in feature space between generated images and class centroids in order to control the feature quality of selected synthetic images for data augmentation. Our models are evaluated on a cervical histopathology image dataset with a limited number of patch-level CIN grade annotations. Extensive experimental results show a significant improvement of classification accuracy from 66.3% to 71.7% using the same ResNet18 baseline classifier after leveraging our cGAN generated images with feature-based filtering, which demonstrates the effectiveness of our models., Comment: MICCAI 2019

However, the concentrations of MC-LR in drinking water and the synergistic effect of MC-LR and HBV on hepatocellular carcinogenesis through their disturbance of redox balance have not been fully elucidated.Methods: We measured the MC-LR concentrations in 168 drinking water samples of areas with a high incidence of HCC. The relationships between MC-LR and both redox status and liver diseases in 177 local residents were analyzed. The hepatoma cell line HepG2 transfected with C-terminal truncated hepatitis B virus X gene (Ct-HBX) were treated with MC-LR. Reactive oxygen species (ROS), superoxide dismutase (SOD), glutathione (GSH) and malondialdehyde (MDA) were measured. Cell proliferation, migration, invasion, and apoptosis were assessed with cell activity assays, scratch and transwell assays, and flow cytometry, respectively. The mRNA and protein expression-related redox status genes were analyzed with qPCR and Western blotting.Results: The average concentration of MC-LR in well water, river water and reservoir water were 57.55 ng/L, 76.74 ng/L and 132.86 ng/L respectively, and the differences were statistically significant (P < 0.05). The MC-LR levels in drinking water were correlated with liver health status, including hepatitis, clonorchiasis, glutamic pyruvic transaminase abnormalities and hepatitis B surface antigen carriage (all P values < 0.05). The serum MDA increased in subjects who drank reservoir water and were infected with HBV (P < 0.05). In the cell experiment, ROS increased when Ct-HBX-transfected HepG2 cells were treated with MC-LR, followed by a decrease in SOD and GSH and an increase in MDA. MC-LR combined with Ct-HBX promoted the proliferation, migration and invasion of HepG2 cells, upregulated the mRNA and protein expression of MAOA gene, and downregulated UCP2 and GPX1 genes.Conclusion: MC-LR and HBV may synergistically affect redox status and play an important role in hepatocarcinoma genesis.Keywords: hepatocellular carcinoma, microcystins-LR, hepatitis B virus X gene, redox balance

Collective dynamics often play an important role in determining the stability of ground states for both naturally occurring materials and metamaterials. We studied the temperature dependent dynamics of antiferromagnetically ordered superdomains in a square artificial spin lattice using soft x-ray photon correlation spectroscopy. We observed an exponential slowing down of superdomain wall motion below the AF onset temperature, similar to the behavior of typical bulk antiferromagnets. Electrical control of structural and physical properties is a long-sought, but elusive goal of contemporary science and technology. We demonstrate that a combination of strong spin-orbit interactions (SOI) and a canted antiferromagnetic (AFM) Mott state is sufficient to attain that goal. The AFM insulator Sr2IrO4 provides a model system in which strong SOI lock canted Ir magnetic moments to IrO6-octahedra, causing them to rigidly rotate together. A novel coupling between an applied electrical current and the canting angle reduces the N\'eel temperature and drives a large, non-linear lattice expansion that closely tracks the magnetization, increases the electron mobility, and precipitates a unique resistive switching effect. Our observations open new avenues for understanding fundamental physics driven by strong SOI in condensed matter, and provide a new paradigm for functional materials and devices., Comment: 5 figures; to be published in Physical Review Letters

We report evidence of spontaneous formation of a heterogeneous network of superdomains in two-dimensional square artificial spin ice nanostructures in externally applied magnetic fields. The magnetic heterogeneity is locally disordered but has a zig-zag texture at longer length scales. Resonant coherent soft-x-ray scattering off such textures give rise to unique internal structure in Bragg peaks. 40. Emergence of a Balanced Core through Dynamical Competition in Heterogeneous Neuronal Networks

The balance between excitation and inhibition is crucial for neuronal computation. It is observed that the balanced state of neuronal networks exists in many experiments, yet its underlying mechanism remains to be fully clarified. Theoretical studies of the balanced state mainly focus on the analysis of the homogeneous Erd$\ddot{\text{o}}$s-R\'enyi network. However, neuronal networks have been found to be inhomogeneous in many cortical areas. In particular, the connectivity of neuronal networks can be of the type of scale-free, small-world, or even with specific motifs. In this work, we examine the questions of whether the balanced state is universal with respect to network topology and what characteristics the balanced state possesses in inhomogeneous networks such as scale-free and small-world networks. We discover that, for a sparsely but strongly connected inhomogeneous network, despite that the whole network receives external inputs, there is a small active subnetwork (active core) inherently embedded within it. The neurons in this active core have relatively high firing rates while the neurons in the rest of the network are quiescent. Surprisingly, the active core possesses a balanced state and this state is independent of the model of single-neuron dynamics. The dynamics of the active core can be well predicted using the Fokker-Planck equation with the mean-field assumption. Our results suggest that, in the presence of inhomogeneous network connectivity, the balanced state may be ubiquitous in the brain, and the network connectivity in the active core is essentially close to the Erd$\ddot{\text{o}}$s-R\'enyi structure. The existence of the small active core embedded in a large network may provide a potential dynamical scenario underlying sparse coding in neuronal networks.

41. Modeling of novel resist technologies

In response to the difficulties posed by the resolution, line edge roughness, sensitivity (RLS) trade-off to traditional chemically amplified resist (CAR) systems used for extreme ultraviolet lithography, a number of novel resist technologies have been proposed. In this paper, the effect of quencher loading on three resist technologies is analyzed via an error propagation-based resist simulator. In order of increasing novelty as well as complexity, they are: Conventional CAR with quencher, CAR with photodecomposable base, and PSCAR 2.0, a CAR system with photodecomposable base as well as an EUV-activated UV-sensitive resist component. Simulation finds the more complicated resist systems trade in an increase in resist stochastics for improved deprotection slopes, yielding a net benefit in terms of line width roughness.

Procedure details included recording delivery sites and those in BT. Asthma Control Questionnaire (ACQ) scores and spirometric measurements were recorded 1-day before treatment and 6 months post-treatment to explore the effects of BT activation number and site on clinical response.Results: ACQ score (3.19± 1.14 vs 1.26± 0.63), forced expiratory volume in 1 sec (FEV1)% predicted (55.53± 21.66 vs 66.19± 22.50), FEV1 (1.53± 0.74 vs 1.93± 0.82), and forced vital capacity (FVC) (2.49± 0.86 vs 2.92± 0.94) significantly increased after three BT sessions compared with pre-session. Major bronchial ablation did not significantly improve BT response in asthma patients. Multivariate logistic regression identified baseline ACQ score and baseline FEV1% predicted as independent factors affecting the clinical response to BT. Correlation and regression analysis revealed a significant linear relationship between baseline ACQ and ACQ improvement, as well as a linear relationship between the third session activation number and ACQ improvement. Based on subgroup analysis of activation number, cohort C (activations ≥ 200) had better lung function, lower non-responding rate, and better long-term effectiveness than the other two cohorts. The activation number in the third BT session showed the strongest predictive ability compared with the first two sessions.Conclusion: Main bronchial ablation did not markedly affect clinical response to BT. Baseline ACQ and baseline FEV1% predicted were independent factors affecting clinical response to BT. Increasing the activation number might promote the therapeutic efficacy of BT, and the activation number in the third BT session correlated with and predicted the BT response.Keywords: refractory asthma, bronchial thermoplasty, activation, response, correlation

This study investigated roles of nuclear factor E2-related factor 2 (Nrf2) in regulating pyroptosis and PD development.Methods: Cellular and rat PD models established by 6-OHDA exposure were subjected to Nrf2 overexpression. Neurobehavioral functions were assessed by the traction test, Morris Water Maze, and open field test. Cell proliferation was analyzed by MTS assay, while flow cytometry was applied to quantify levels of reactive oxygen species (ROS) and apoptosis. Nissl bodies in rat brains were detected by Nissl staining, and cell apoptosis in brain tissues was assessed by terminal deoxynucleotidyl transferase dUTP nick-end labeling. Differential expression of lncRNA and mRNA was characterized by deep sequencing.Results: A cellular PD model was successfully established by inducing PC12 cell differentiation with nerve growth factor-β and exposing differentiated cells to 6-OHDA. Cells exhibited significantly increased ROS levels, enhanced pyroptosis, and inhibited Nrf2 phosphorylation. The rat PD model exhibited impaired muscle strength, increased pyroptosis, and repressed Nrf2 phosphorylation. Nrf2 overexpression effectively repressed pyroptosis in both cellular and rat PD models. Marked alterations of lncRNA and mRNA profiles were induced by Nrf2 overexpression in the cellular PD model, which involved multiple signaling pathways. Silencing of the lncRNA AABR07032261.5 significantly promoted pyroptosis in the cellular PD model.Conclusion: Nrf2 suppressed PD pathogenesis in cellular and animal models by promoting AABR07032261.5, which repressed pyroptosis.Keywords: Nrf2, lncRNAs, AABR07032261.5, oxidative stress, pyroptosis, Parkinson’s disease

- Abstract
We develop and compare e-variables for testing whether k samples of data are drawn from the same distribution, the alternative being that they come from different elements of an exponential family. We consider the GRO (growth-rate optimal) e-variables for (1) a ‘small’ null inside the same exponential family, and (2) a ‘large’ nonparametric null, as well as (3) an e-variable arrived at by conditioning on the sum of the sufficient statistics. (2) and (3) are efficiently computable, and extend ideas from Turner et al. (2021) and Wald (1947) respectively from Bernoulli to general exponential families. We provide theoretical and simulation-based comparisons of these e-variables in terms of their logarithmic growth rate, and find that for small effects all four e-variables behave surprisingly similarly; for the Gaussian location and Poisson families, e-variables (1) and (3) coincide; for Bernoulli, (1) and (2) coincide; but in general, whether (2) or (3) grows faster under the alternative is family-dependent. We furthermore discuss algorithms for numerically approximating (1).

The gut microbiota is a complex ecosystem composed of many bacteria and their metabolites.It plays an irreplaceable role in human digestion, nutrient absorption, energy supply, fat metabolism, immune regulation, and many other aspects. Exploring the structure and function of the gut microbiota, as well as their key genes and metabolites, will enable the early diagnosis and auxiliary diagnosis of diseases, new treatment methods, better effects of drug treatments, and better guidance in the use of antibiotics. The identification of gut microbiota plays an important role in clinical diagnosis and treatment, as well as in drug research and development. Therefore, it is necessary to conduct a comprehensive review of this rapidly evolving topic. Traditional identification methods cannot comprehensively capture the diversity of gut microbiota. Currently, with the rapid development of molecular biology, the classification and identification methods for gut microbiota have evolved from the initial phenotypic and chemical identification to identification at the molecular level. This review integrates the main methods of gut microbiota identification and evaluates their application. We pay special attention to the research progress on molecular biological methods and focus on the application of high-throughput sequencing technology in the identification of gut microbiota. Both inflammatory processes and glutamatergic systems have been implicated in the pathophysiology of mood-related disorders. However, the role of caspase-1, a classic inflammatory caspase, in behavioral responses to chronic stress remains largely unknown. To address this issue, we examined the effects and underlying mechanisms of caspase-1 on preclinical murine models of depression. We found that loss of caspase-1 expression in Caspase-1-/- knockout mice alleviated chronic stress-induced depression-like behaviors, whereas overexpression of caspase-1 in the hippocampus of wild-type (WT) mice was sufficient to induce depression- and anxiety-like behaviors. Furthermore, chronic stress reduced glutamatergic neurotransmission and decreased surface expression of glutamate receptors in hippocampal pyramidal neurons of WT mice, but not Caspase-1-/- mice. Importantly, pharmacological inhibition of caspase-1-interleukin-1β (IL-1β) signaling pathway prevented the depression-like behaviors and the decrease in surface expression of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs) in stressed WT mice. Finally, the effects of chronic stress on both depression- and anxiety-like behaviors can be mimicked by exogenous intracerebroventricular (i.c.v.) administration of IL-1β in both WT and Caspase-1-/- mice. Taken together, our findings demonstrate that an increase in the caspase-1/IL-1β axis facilitates AMPAR internalization in the hippocampus, which dysregulates glutamatergic synaptic transmission, eventually resulting in depression-like behaviors. These results may represent an endophenotype for chronic stress-induced depression.
