Background The development of liver cirrhosis is usually an asymptomatic process until late stages when complications occur. The potential reversibility of the disease is dependent on early diagnosis of liver fibrosis and timely targeted treatment. Recently, the use of non-invasive tools has been suggested for screening of liver fibrosis, especially in subjects with risk factors for chronic liver disease. Nevertheless, large population-based studies with cost-effectiveness analyses are still lacking to support the widespread use of such tools. The aim of this study is to investigate whether non-invasive liver stiffness measurement in the general population is useful to identify subjects with asymptomatic, advanced chronic liver disease. Methods This study aims to include 30,000 subjects from eight European countries. Subjects from the general population aged ≥ 40 years without known liver disease will be invited to participate in the study either through phone calls/letters or through their primary care center. In the first study visit, subjects will undergo bloodwork as well as hepatic fat quantification and liver stiffness measurement (LSM) by vibration-controlled transient elastography. If LSM is ≥ 8 kPa and/or if ALT levels are ≥1.5 x upper limit of normal, subjects will be referred to hospital for further evaluation and consideration of liver biopsy. The primary outcome is the percentage of subjects with LSM ≥ 8kPa. In addition, a health economic evaluation will be performed to assess the cost-effectiveness and budget impact of such an intervention. The project is funded by the European Commission H2020 program. Discussion This study comes at an especially important time, as the burden of chronic liver diseases is expected to increase in the coming years. There is consequently an urgent need to change our current approach, from diagnosing the disease late when the impact of interventions may be limited to diagnosing the disease earlier, when the patient is asymptomatic and free of complications, and the disease potentially reversible. Ultimately, the LiverScreen study will serve as a basis from which diagnostic pathways can be developed and adapted to the specific socio-economic and healthcare conditions in each country. RESUMEN: Los sistemas de liberación controlada son una prometedora alternativa para el tratamiento de infecciones intracelulares por su capacidad para modificar el perfil de liberación de un principio activo e incluso vectorizar su distribución a nivel celular. En esta investigación se desarrollaron dos sistemas de liberación de doxiciclina utilizando micropartículas y liposomas, elaborados por secado por aspersión y evaporación en fase reversa, respectivamente. Las micropartículas fueron preparadas con dos diferentes mezclas de agentes encapsulantes: gelatina:sacarosa 70:30 y gelatina:sacarosa:caroboxitemilcelulosa 13.64:85.86:0.50, y se caracterizaron en términos de humedad, actividad acuosa, propiedades de fluidez, tamaño, potencial zeta, rendimiento, eficiencia y liberación in vitro. Se diseñaron dos tipos de liposomas: neutros DMPC y aniónicos DMPC:DMPS 1:1, utilizando una concentración de fosfolípido 6 mg/mL. Se estudió el efecto de adición de DMPS en el tamaño y carga superficial de liposomas de DMPC, la capacidad de incorporación de doxiciclina en los dos sistemas y la internalización de los liposomas en células DH82, evaluada por microscopía confocal. El sistema microparticulado compuesto por gelatina:sacarosa:caroboxitemilcelulosa presentó mejores características en términos de Índice de Carr (23.33 %), Relación Hausner (1.30), morfología suave y superficie externa lisa, eficiencia de encapsulación (57.91 %) y menor cantidad de principio activo liberado a pH ácido. En cuanto a los liposomas, aquellos compuestos por DMPC:DMPS 1:1 mostraron ser mayormente internalizados de acuerdo con el perfil de intensidad de fluorescencia. xviii Los resultados indicaron que las dos formulaciones, microparticulada y liposomal, tienen potencial de ser implementadas en el tratamiento de enfermedades intracelulares en las que la doxiciclina tenga una acción terapéutica. ABSTRACT: Drug delivery systems are a promising alternative for intracellular infections treatments due to their capability to modify drug release profiles and even vectorize in a cellular level. Two drug delivery systems of doxycycline were developed in this research with microparticles and liposomes, the first one elaborated by spray drying and the second with reverse-phase evaporation vesicle. Microparticles were prepared with two different wall material mixtures: gelatin:sucrose 70:30 and gelatin:sucrose:carboxymethyl cellulose 13.64:85.86:0.50, and were evaluated in terms of moisture, water activity, flow properties, size, zeta potential, yield, efficiency and in vitro release. Two different types of liposomes were designed: neutral DMPC and anionic DMPC:DMPS 1:1, each one with 6 mg/mL of phospholipid. The DMPS addition effect on DMPC liposomes was studied by its size and surface charge, as well as the doxycycline incorporation capacity on the two systems and the liposomes internalization in DH82 cells by confocal microscopy. The microparticulate system, composed by gelatin:sucrose:carboxymethyl cellulose showed better characteristics in terms of Carr’s Index (23.33 %), Hausner Ratio (1.30), soft morphology and smooth external surface, encapsulation efficiency (57.91 %) and lower quantity of active ingredient released at acid pH. Regarding liposomes, those composed by DMPC:DMPS 1:1 showed to be mainly uptake according to the data provided by the fluorescence intensity profile. Results suggested that both formulations, microparticles and liposomes, have potential to be used in the intracellular diseases treatments in which doxycycline has therapeutical effect.

En el presente anteproyecto que lleva por título ESTUDIO DESCRIPTIVO DE CLIMA ORGANIZACIONAL DE LA EMPRESA INDUFRIAL S.A., se realizará una descripción detallada de los fundamentos teóricos de la Investigación o Marco Conceptual de la misma, de igual forma se realiza una definición conceptual y/o peracional de la variable objeto de estudio, así como de sus características; También se realiza una descripción de los procedimientos metodológicos y del tratamiento estadístico que recibirán los datos de esta investigación. Para la realización de este anteproyecto se hizo la revisión de la literatura existente y a disposición, estableciendo los temas más importantes y relacionados con el título de la investigación, de esta forma se plantean las diferentes definiciones de Clima Organizacional, las Dimensiones, Características, Elementos, Cultura Organizacional, Relaciones Interpersonales, Comunicación, Motivación y Desempeño Laboral en las Organizaciones, etc. Posteriormente se plantea la pregunta de Investigación y los Objetivos que serán la guía de la Investigación, se realizó la definición conceptual y operacional de la variable objeto de estudio, que es Clima Organizacional; también se estableció, el diseño a utilizar, los participantes, el instrumento y el procedimiento a seguir.
