However, several other diseases are often associated with thrombosis although their frequency is not easily estimated. Eosinophilia is one of these conditions. From a clinical viewpoint it is very difficult to understand which conditions might lead to a thrombotic event because the underlying pathophysiological mechanisms are different. Here, we report a case of idiopathic hypereosinophilia associated to venous thromboembolism without any other associated prothrombotic condition.Keywords: venous thromboembolism, deep vein thrombosis, hypereosinophilia, allergy

To investigate the modification of the protein C anticoagulant pathway after vigorous exercise, we measured ProC® Global assay, a protein C activity dependent clotting time, in 20 healthy subjects before and immediately after maximal treadmill exercise, and at 5, 15, 30 and 60 min in the recovery phase. The most evident change was a shortening of ProC® Global clotting time from the average basal value of 123 sec to 84 sec at 30 min in post-exercise. Our study shows that the coagulation unbalance observed after strenuous exercise and with no consequence in healthy individuals with normal PC level, could increase the thrombophilic risk in silent carriers of significant defects of the protein C system and occasionally trigger an episode of deep vein thrombosis.Keywords: ProC® Global, protein C unbalance after physical exercise, physical exercise; thrombophilic risk

Abstract

In the -omics era, analysis of genome and transcriptome have become extremely relevant for the elucidation of the genetic cause of a number of diseases, previously undiagnosed. In addition, microbiome analysis is becoming relevant in many pathological conditions.Identification of genetic variants is very efficient with techniques such as comparative genome hybridization (CGH)-array and with whole genome sequence (WGS) or whole exome sequence (WES) performed with next-generation sequence (NGS) methodologies. Most importantly, correct classification of variants and elucidation of their clinical significance are tasks of extreme relevance for the correct diagnosis and, often, also to indicate the most efficient therapeutic choices. However, over the years our understanding of significance of genetic variants has dramatically improved, therefore many cases would require reevaluation and, on occasions, retesting. In this article, we reviewed the major advances in the genomic diagnostics field focusing, in particular, at addressing the relevance of periodic reevaluation of results and retesting patients when significantly novel technologies are developed, focusing also on economical, legal and ethical points.

Abstract

DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limit

Abstract

DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I (p) steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at similar to 8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I (p) beam injection was coupled to a radiative divertor. Abstract

DIII-D physics research addresses critical challenges for the operation of ITER and the next generation of fusion energy devices. This is done through a focus on innovations to provide solutions for high performance long pulse operation, coupled with fundamental plasma physics understanding and model validation, to drive scenario development by integrating high performance core and boundary plasmas. Substantial increases in off-axis current drive efficiency from an innovative top launch system for EC power, and in pressure broadening for Alfven eigenmode control from a co-/counter-I p steerable off-axis neutral beam, all improve the prospects for optimization of future long pulse/steady state high performance tokamak operation. Fundamental studies into the modes that drive the evolution of the pedestal pressure profile and electron vs ion heat flux validate predictive models of pedestal recovery after ELMs. Understanding the physics mechanisms of ELM control and density pumpout by 3D magnetic perturbation fields leads to confident predictions for ITER and future devices. Validated modeling of high-Z shattered pellet injection for disruption mitigation, runaway electron dissipation, and techniques for disruption prediction and avoidance including machine learning, give confidence in handling disruptivity for future devices. For the non-nuclear phase of ITER, two actuators are identified to lower the L-H threshold power in hydrogen plasmas. With this physics understanding and suite of capabilities, a high poloidal beta optimized-core scenario with an internal transport barrier that projects nearly to Q = 10 in ITER at ∼8 MA was coupled to a detached divertor, and a near super H-mode optimized-pedestal scenario with co-I p beam injection was coupled to a radiative divertor. The hybrid core scenario was achieved directly, without the need for anomalous current diffusion, using off-axis current drive actuators. Also, a controller to assess proximity to stability limit

Abstract

Introduction: oncololytic adenoviruses (OAds), viruses constructed to replicate in tumor cells, improve the outcome of cancer therapy in some cases, such as sarcomas. However, the molecular heterogeneity of tumors requires specific and personalized cancer treatments in order to set up more adequate and effective therapies. Methods: by using the array Comparative Genomic Hybridization (array-CGH), a molecular approach method, we aimed to identify chromosomal aberrations or Copy Number Variants (CNVs) in three different tumor cell lines: HCT116, SW872 and A2058 selected for their Coxsackievirus and Adenovirus receptor (CAR) expression profile. Results: the cells showed several duplications of genes involved in replicative Adenovirus cycle (binding, internalization, escape) in the core transport, and in the escape of the viral DNA from the capsid. Conclusion: in this study, our aim was to identify chromosomal alterations in genes involved in the OAd replication cycle process. Array-CGH method could be useful to design a platform for a screening analysis in order to identify mutations that can contribute to oncolytic virotherapy approach generating a personalized strategy for tumor suppression.

Abstract

Antimicrobial peptides can protect the gastric mucosa from bacteria, but Helicobacter pylori (H. pylori) can equally colonize the gastric apparatus. To understand beta-defensin function in H. pylori-associated chronic gastritis, we investigated susceptibility, human beta-defensin mRNA expression, and DNA methylation changes to promoters in the gastric mucosa with or without H. pylori infection. We studied the expression of HBD2 (gene name DEFB4A), HBD3 (DEFB103A), and HBD4 (DEFB104) using real-time PCR in 15 control and 10 H. pylori infection patient gastric specimens. This study demonstrates that H. pylori infection is related to gastric enhancement of inducible HBD2, but inducible HBD3 and HBD4 expression levels remained unchanged. HBD2 gene methylation levels were overall higher in H. pylori-negative samples than in H. pylori-positive samples. We also assessed antimicrobial susceptibility using growth on blood agar. The H. pylori strain Tox+ was susceptible to all defensins tested and their analogs (3N, 3NI). These results show that HBD2 is involved in gastritis development driven by H. pylori, which facilitates the creation of an epigenetic field during H. pylori-associated gastric tumorigenesis.

Abstract

Diagnosing a complex genetic syndrome and correctly assigning the concomitant phenotypic traits to a well-defined clinical form is often a medical challenge. In this work, we report the analysis of a family with complex phenotypes, including microcephaly, intellectual disability, dysmorphic features, and polydactyly in the proband, with the aim of adding new aspects for obtaining a clear diagnosis. We performed array-comparative genomic hybridization and quantitative reverse transcriptase PCR (qRT-PCR) analyses. We identified a deletion of chromosome 20p12.1 involving the macrodomain containing 2/mono-ADP ribosylhydrolase 2 gene (MACROD2) in several members of the family. This gene is actually not associated with a specific syndrome but with congenital anomalies of multiple organs. qRT-PCR showed higher levels of a MACROD2 mRNA isoform in the individuals carrying the deletion. Our results, together with other data reported in the literature, support the hypothesis that the deletion in MACROD2 can affect correct embryonic development and that the presence of another associated event, such as epigenetic modifications at the MACROD2 locus, can influence the level of severity of the pathology.

Abstract

Diagnosing a complex genetic syndrome and correctly assigning the concomitant phenotypic traits to a well-defined clinical form is often a medical challenge. In this work, we report the analysis of a family with complex phenotypes, including microcephaly, intellectual disability, dysmorphic features, and polydactyly in the proband, with the aim of adding new aspects for obtaining a clear diagnosis. We performed array-comparative genomic hybridization and quantitative reverse transcriptase PCR (qRT-PCR) analyses. We identified a deletion of chromosome 20p12.1 involving the macrodomain containing 2/mono-ADP ribosylhydrolase 2 gene (MACROD2) in several members of the family. Abstract

As a tribute to Bruno Lombardo, our senior co-worker who sadly passed away in 2014, the geologic setting and metamorphic evolution of the first eclogites discovered in the Eastern Himalaya are summarised in the context of U-Pb analytical results from the original granulitized eclogite samples. These eclogites occur at the top of the Main Central Thrust (MCT) zone in the Ama Drime Range (Arun - Phung Chu Valley), southeastern Tibet. The eclogites record five superposed metamorphic stages: a first eclogite-facies stage ( P > 1.5 GPa, T > 580°C), two granulite-facies stages at decreasing pressure ( P = 0.8–1.0 GPa, T > 750°C; P ~ 0.4 GPa, T ~ 750°C), an amphibolite-facies stage ( T = 700°C, P = 0.4–0.6 GPa) and a late retrogression stage ( T < 690°C, P

Abstract

Il Ghiacciaio Superiore di Coolidge, localizzato in alta Valle Po nel territorio comunale di Crissolo (CN), è l'ultimo sopravvissuto di un gruppo di apparati glaciali originariamente ospitati sul versante nord-orientale del Monviso. L'area, dominata dalla maestosa piramide del "Re di Pietra", oltre a comprendere la riserva naturale speciale del Pian del Re è a sua volta parte integrante del Sito di Interesse Comunitario (SIC) denominato "Gruppo del Monviso e Bosco dell'Allevè" (Codice IT1160058) nonché parte del Parco Naturale del Fiume Po. Nel 2013 il Massiccio del Monviso è stato formalmente riconosciuto come nuova Riserva della Biosfera nazionale e transfrontaliera nell'ambito del programma UNESCO Man and Biosphere. La fitta rete di sentieri e punti di appoggio, che consente un'agevole esplorazione a 360° del massiccio, rende inoltre questo settore delle Alpi Cozie uno dei luoghi più frequentati della catena alpina occidentale. L'itinerario descritto si sviluppa a partire dalle sorgenti del F. Po, situate al Pian del Re (2020 m), per giungere fino al Rifugio Quintino Sella (2640 m), localizzato alla base dell'imponente parete NE del Monviso. Il percorso si snoda attraverso un paesaggio suggestivo, profondamente segnato dal modellamento delle successioni meta-ofiolitiche da parte dei ghiacciai pleistocenici e, nell'area più prossima alla piramide del Monviso, dalla riavanzata delle fronti glaciali che ha contraddistinto la Piccola Età Glaciale. Le evidenze lasciate dal crollo di buona parte del Ghiacciaio pensile di Coolidge, avvenuto nell'estate del 1989, consentono inoltre di effettuare interessanti osservazioni sugli effetti indotti dal ritiro (generalmente graduale, talvolta parossistico) delle masse glaciali.

Abstract

In order to promote geosite conservation in the project entitled 'PROactive management of GEOlogical heritage in the PIEMONTE Region', we propose a comprehensive study involving the Monviso Massif (MM) geothematic area, one of the most outstanding symbols of the Alps and particularly of the Cottian Alps. Specifically, at the MM, the inventory of a number of different geosites whose conservation and development require different geologic and some additional non-geological expertise is considered: (1) some of the best preserved ophiolites in the Alps and the associated Cu-Fe mineralizations; (2) the lithostructural units in the Germanasca Valley; (3) the first primary source of jade in the Alps at the MM and its importance in terms of Neolithic to Bronze Age-polished stone implements; (4) the world-famous minerals such as coesite and giant pyrope, as well as type localities for new minerals (including carlosturanite); (5) the area, now buried under a debris flow, where Hannibal is thought to have regrouped his army while crossing the Alps; and (6) the biodiversity of lichens, microfungi and cyanobacteria colonizing the ophiolites, which can give additional value for the environmental assessment and evaluation of the MM outcrops. Following geodiversity identification, further stages will be devoted to develop appropriate plans for geodiversity conservation in this area.

Abstract

Osteoporosis is a chronic, systemic, metabolic disease characterized by loss of bone mass, disruption of the micro architectural structure of bone tissue and changes in the physical properties of bone matrix that leads to an increased risk of fracture. The rise in average age in western society increases the need for novel therapeutic strategies for this disease; therefore, in order to identify novel drugs, systems for in vitro screening for molecules capable of stimulating osteoblast differentiation are of increasing relevance. Indeed, detailed analysis and characterization in cellular and animal models are necessary preclinical steps towards the development of novel therapeutic options. Here we present a review of cellular models for osteoblast differentiation and animal models for osteoporosis as well as available methodologies to evaluate bone parameters in vivo. These models are useful to identify novel targets and to develop safer and more effective therapeutic strategies with new drugs able to reduce fractures risk in human.

AbstractData on the biology of Torpedo torpedofrom the Ionian Sea are absent, and in the Mediterranean Sea, most of the published works focus on the reproductive biology of the species and are not recent. Several biological and ecological aspects of Torpedo torpedowere investigated between March and May 2019 on the coast of Sicily (Ionian Sea) between Avola and Marzamemi. Morphological (disc width–weight relationships, total length-disc width relationships), population (size-frequency distribution, sex ratio, fecundity), and ecological (habitat, diet) characteristics were studied. Stomach content analysis revealed that T. torpedois a generalist piscivore, feeding on a wide variety of benthic and benthopelagic fish: Levin’s index value (Bi) was 0.82. The analysis of the sex ratio showed a higher proportion of males vs. females. Females were, on average, larger than males. Fecundity showed a positive correlation with body size, with the smallest mature female being 14.9 cm in disc width and 24.8 cm in total length. The ovarian fecundity ranged from 3 to 20 eggs. In the study area, both adults and juveniles of the species were caught in considerable numbers with trammel nets. We also provide data from some specimens of Torpedo marmoratacaught during the study period.
