Abstract

The creation and maintenance of a stable and long-lasting relationship are crucial for adult individual well-being, with single people reporting poorer mental health when compared to those engaged in romantic relationships. Among the factors that may predict the quality of romantic dyads, scholars have addressed dyadic perfectionism. However, few is known about which of the perfectionism dimensions better predicts the status of being single or engaged in a romantic relationship. The present study aims to verify whether perfectionism is associated with the relationship status, and which perfectionism dimension is involved. More specifically, the interpersonal aspects of dyadic perfectionism were expected to be positively associated with the probability of belonging to the 'single status'. Contributions of demographic variables as well as effects of mental health symptoms were controlled as they are relevant factors influencing romantic relationships. A hierarchical logistic regression analysis was conducted to evaluate the predictive utility of age and gender (controlled for at Step 1), depression and anxiety (entered at Step 2) and intra-and interpersonal perfectionism dimensions (controlled at Step 3) to predict the participants' relationship status (0 = single, 1 = romantic partner). 25. A Brief Online Intervention Based on Dialectical Behavior Therapy for a Reduction in Binge-Eating Symptoms and Eating Pathology.

Abstract

Dysregulated eating behaviors, comprising subthreshold and clinical binge-eating disorder (BED) and bulimia nervosa (BN), are increasing among the general population, with a consequent negative impact on one's health and well-being. Despite the severity of these outcomes, people with BED and BN often face a delay in receiving a diagnosis or treatment, often due to difficulties in accessing care. Hence, evidence-based and sustainable interventions for eating symptomatology are needed. The present study aims to assess the effectiveness of a web-based 10-session multidisciplinary group intervention based on Dialectical Behavior Therapy (DBT) for BED and BN, aimed at reducing psychological distress and binge-eating-related symptomatology in a sample of patients with dysregulated eating behaviors and including one session of nutritional therapeutic education. A total of 65 participants (84.6% F; age M = 38.5 ± 13.2; experimental group, N = 43; treatment-as-usual group, TAU, N = 22) took part in the study. The results show, after the 9 weekly sessions, a significant reduction in binge-eating-related symptomatology and general psychopathology and an increased self-esteem and eating self-efficacy in social contexts in the experimental group compared to the treatment-as-usual group (T0 vs. T1). Improvements in the experimental group were significantly maintained after one month from the end of the intervention (T2) in terms of binge-eating symptoms, general psychopathology, and eating self-efficacy in social contexts. This study supports the effectiveness of a brief web-based multidisciplinary group intervention in reducing eating symptomatology and psychological distress and enhancing self-esteem and eating self-efficacy in a group of people with dysregulated eating behaviors. Abstract

High variability of influenza vaccine efficacy requires the identification of modulators of immunisation that may be targeted as adjuvants in health psychology interventions. Psychosocial and behavioural variables such as psychological stress, greater negative and lower positive affectivity, poor sleep, loneliness, and lack of social support, have been associated with abnormal immune and inflammatory responses and negative health outcomes, yet their effects in modulating vaccine efficacy are yet to be fully understood. We conducted an updated systematic review of longitudinal and experimental studies examining the effects of such variables in predicting immune response to influenza vaccine. PubMed, Medline, PsycINFO, CINAHL and Scopus were searched up to November 2022. Twenty-five studies met the inclusion criteria for qualitative synthesis and 16 provided data for meta-analysis. Low positive and high negative affect were associated with low antibodies and weak cell-mediated immunity following vaccination in qualitative synthesis. Literature on sleep disturbance, loneliness and social support was limited and yielded inconsistent results. Psychological stress was associated with poorer antibody response in meta-analysis. In conclusion, findings from this review suggest a need for further longitudinal and experimental studies on these factors to support their inclusion as target variables in vaccine adjuvant interventions. Abstract

Background: High body mass and adiposity during pregnancy can contribute to psychological distress, and body dissatisfaction may be a potential underlying mechanism of this association. Objective. This study aimed to evaluate the mediational role of body dissatisfaction in the relationship between body mass index (BMI) and depressive and anxious symptoms, respectively. Methods: Given the cross-sectional design of this study, two alternative models were investigated, positing that BMI was related to depressive (Model 1a) and anxious symptoms (Model 2a), which, in turn, predicted body dissatisfaction. Seventy-two pregnant women in the third trimester of pregnancy completed the Body Image Disturbance Questionnaire, the Beck Depression Inventory-II and the State-Trait Anxiety Inventory, as well as a demographic form assessing their BMI. Results: As hypothesized, body dissatisfaction mediated the relationship between BMI and psychopathological symptoms. Moreover, the alternative models of reverse mediation were also significant, suggesting that psychopathological symptoms mediated the relationship between BMI and body dissatisfaction. Findings from both the hypothesized and alternative models suggested that, on the one hand, higher distress symptoms associated with body dissatisfaction would result from high BMI and, on the other hand, that body dissatisfaction may result from the effect of BMI on distress symptoms. Conclusions: The present study suggests that body image theory and practice should be implemented by the inclusion of evidence-based clinical interventions for promoting psychological well-being during the antenatal period. 49. The development of new G-quadruplex stabilising ligands using click chemistry

Abstract

G-quadruplexes are highly specific nucleic acid organisations characteristic of G-rich sequences. They are polymorphic quadruple helices with a hydrophobic core of sets of G-quartets each formed by four guanines interacting via Hoogsteen hydrogen bonds, and four negatively charged grooves formed by the phosphate backbone. A considerable number of genes containing G-rich genomic sequences are believed to play a key role in tumour development, e.g. the proto-oncogenes Ckit-1 and Ckit-2, the hypoxia inducible factor 1α promoter HIF-1α and telomeres, therefore these putative G-quadruplexes are an appealing target for cancer therapy. Many different G-quadruplex ligands have been synthesized to date, but the specificity and the low in vitro/in vivo activity of the majority have not been sufficient for them to progress further. The approach described here towards the discovery of G-quadruplex targeting drugs focuses on the synthesis of libraries derived from combinations of different building blocks. "Click-chemistry" Cu(I) catalysed Huisgen 1,3-dipolar cycloaddition has been an essential tool to achieve this objective. Two classes of building blocks have been synthesized, azides and alkynes, which include an aromatic core and basic side chains that is positively charged under physiological conditions. With these fragments, three libraries of G-quadruplex ligands have been generated, extending the aromatic surface by the click reaction, which results in 1,4-triazole ring formation. The aromatic cores and triazole rings in the final compounds are appropriate for interactions with the G-quartets in a quadruplex, and the positively charged side chains interact with the grooves. The diversity introduced in each library enabled me to investigate the binding influence of the core structure and of the number of side chains. The affinity and selectivity of these compounds for G-quadruplexes has been evaluated by binding experiments using FRET, circular dichroism and ESI-MS. Short and long term cytotoxicity studies have been performed on a panel of cancer cells either expressing telomerase, or genes which could be regulated by G-quadruplex sequences; telomerase inhibition has been studied with the cell free TRAP assay. Two lead ligands have been identified N,N'-((4,4'-(Naphthalene-2,7-diyl)bis(1H-l,2,3-triazole- 4,l-diyl))bis(3,1-phenylene))bis(2-(diethylamino)acetamide) (compound 33) and N,N',N''-((4,4',4"-(Benzene-1,3,5-triyl)tris(1H-1,2,3-triazole-4,1-diyl))tris(benzene-3,1-diyl))tris(3-(pyrrolidin-1-yl)propanamide) (compound 46). Compound 46 showed telomerase inhibitory activity in vitro, probably due to stabilisation of the parallel G-quadruplex structure on the telomeres, as suggested by biophysical studies. Furthermore, compound 33 displayed selective cytotoxicity for renal cancer cells RCC4 in vitro, which could be ascribed to G-quadruplex formation in the promoter region of HIF-1α gene, which is up-regulated in renal cancers.
