Abstract Wildlife tagging provides critical insights into animal movement ecology, physiology, and behavior amid global ecosystem changes. However, the stress induced by capture, handling, and tagging can impact post-release locomotion and activity and, consequently, the interpretation of study results. Here, we analyze post-tagging effects on 1585 individuals of 42 terrestrial mammal species using collar-collected GPS and accelerometer data. Species-specific displacements and overall dynamic body acceleration, as a proxy for activity, were assessed over 20 days post-release to quantify disturbance intensity, recovery duration, and speed. Differences were evaluated, considering species-specific traits and the human footprint of the study region. Over 70% of the analyzed species exhibited significant behavioral changes following collaring events. Herbivores traveled farther with variable activity reactions, while omnivores and carnivores were initially less active and mobile. Recovery duration proved brief, with alterations diminishing within 4–7 tracking days for most species. Herbivores, particularly males, showed quicker displacement recovery (4 days) but slower activity recovery (7 days). Individuals in high human footprint areas displayed faster recovery, indicating adaptation to human disturbance. Our findings emphasize the necessity of extending tracking periods beyond 1 week and particular caution in remote study areas or herbivore-focused research, specifically in smaller mammals.

This paper explores the possibility of using recycled seashells from marine coasts of the Mali Ston Bay in Dubrovnik-Neretva County, Croatia, to create a novel bio material derived from waste seashells. In this research, two types of waste seashells, mussel (Mytilus galloprovincialis) and oyster (Ostrea edulis), combined with natural, non-toxic binders (bone glue) were investigated experimentally. The goal is to develop a sustainable material, suitable for the production of furniture and decorative objects. The parameters studied, included physical and mechanical properties of this material. The results showed that this bio composite material, derived from recycled seashells, is hygroscopic and has low compression strength. It should be used for making furniture components that don't bear heavy loads and it is suitable only for interior applications. This study presents an eco-friendly and sustainable material option, while optimizing the recycling of food waste materials.

Abstract The Tibellus oblongus spider is an active hunter that does not spin webs and remains highly underinvestigated in terms of the venom composition. Here, we describe venom glands transcriptome and venom proteome analysis for unveiling the polypeptide composition of Tibellus oblongus spider venom. The resulting EST database includes 1733 records, including 1263 nucleotide sequences with ORFs, of these 942 have been identified as toxin-coding. The database of peptide sequences was built based on of the transcriptomics results. It contains 217 new toxins, 212 of them were detected in the T. oblongus venom by the proteomics.

In the social network Facebook from April 2020 to October 2020 an online survey using the "Google Form" was conducted to study the perception of the COVID-19 pandemic by the residents of Ukraine. The study involved 550 participants residing in the Ukraine-controlled territory and the city of Kyiv. The data are as follows: the coronavirus is considered a serious danger by 33.3±2.0% of respondents and 11.6±1.4% agree with certain doubts (in total – Σ44.9±2.1%); 21.1±1.7% believe that the coronavirus is not serious and 17.3±1.6% believe that it is probably not serious (Σ38,4±2.1%). Adenosine 5' triphosphate (ATP) is a universal intracellular energy source and an evolutionarily ancient, ubiquitous extracellular signal in diverse species. Here, we report the generation and characterization of single-wavelength genetically encoded fluorescent sensors (iATPSnFRs) for imaging extracellular and cytosolic ATP from insertion of circularly permuted superfolder GFP into the epsilon subunit of F0F1-ATPase from Bacillus PS3. On the cell surface and within the cytosol, iATPSnFR1.0 responds to relevant ATP concentrations (30 μM to 3 mM) with fast increases in fluorescence. iATPSnFRs can be genetically targeted to specific cell types and sub-cellular compartments, imaged with standard light microscopes, do not respond to other nucleotides and nucleosides, and when fused with a red fluorescent protein function as ratiometric indicators. After careful consideration of their modest pH sensitivity, iATPSnFRs represent promising reagents for imaging ATP in the extracellular space and within cells during a variety of settings, and for further application-specific refinements.

This paper presents the bifurcation approach to analyze divergent loss of stability of the symmetric solution of a nonlinear dynamic model in Lyapunov's critical case of a single zero root. Under such a condition, material birth-annihilation bifurcations of multiple stationary states take place. Moreover, the equilibrium surface of stationary states in a small neighborhood of the symmetric solution is a generalized Whitney fold. In the simplest case of a fold peculiarity, the corresponding bifurcation manifold locally coincides with the discriminant manifold of a third-degree polynomial that determines the manifold of stationary states in a small neighborhood of the symmetric solution. An algorithm to construct the corresponding polynomial is introduced. Through the algorithm, the bifurcation manifold is determined, and the conditions for safe/unsafe loss of stability of the symmetric solution are derived analytically. The proposed approach to analyze divergent loss of stability is implemented for a nonlinear bicycle model of a two-axle wheeled vehicle. It represents a further development of Pevzner–Pacejka's well-known graph-analytical method. The paper determines the critical values of constructive parameters that are responsible for safe/unsafe loss of stability of the vehicle's straight-line motion, and it discusses strategies for the bifurcation approach to analyze divergent loss of stability.

Background: Treatment of isolated and combined open soft tissue wounds remains relevant due to the growth of severe injuries, comorbid pathologies, and immunosuppressive conditions, along with a decrease in antibiotic sensitivity of microorganisms. The aim of this study was to compare the effectiveness of the jet oxygen-sorption treatment (JOST) and flow sorption debridement techniques (FSDT) in the therapy of experimental soft tissue wounds. Methods and Results: The effectiveness of the JOST and FSDT was compared in 2 series of experiments on 288 laboratory Wistar rats with simulated soft tissue wounds. Series 1 (S1) involved 144 animals divided into 2 control groups (CG) and 2 experimental groups (EG); the effectiveness of the developed techniques for the aseptic wound treatment was studied in these groups of animals. Series 2 (S2) involved 144 animals divided into 2 CGs and 2 EGs; in S2, purulent soft tissue wounds were studied. The effectiveness of the developed techniques in the complex treatment of experimental wounds was assessed immediately, on Days 1, 3, 5, 7 and 10 after simulating the pathological process. The assessment included the animals’ condition, the dynamics of the course of reparative processes (local symptoms of inflammation; granulation; epithelialization of wounds; and size and dynamics of the area of the defect), and histological research methods. The use of the JOST has practically no benefits in aseptic wound treatment, compared to FSDT. The use of the JOST and FSDT contributed to a significant acceleration of healing process in aseptic wounds that was expressed in the decreased local inflammatory reactions, higher activity of metabolic processes based on the dynamics of RNA and SH-groups. The most pronounced positive dynamics in the treatment of purulent wounds was observed when applying FSDT. Compared with the findings obtained in CG1-S2, FSDT contributed to a reduction in necrolysis termination by 25.7%, fibrinolysis - by 25.5%, granulation - by 20.0%, epithelization - by 18.9%, wound discharge - by 27.8%. Conclusion: The developed technique of FSDT in the complex treatment of soft tissue wounds provides the most pronounced positive dynamics, accelerates reparative tissue processes, and reduces the duration of wound cleansing and healing for both aseptic and purulent wounds.

The dynamics of modern life in its cultural and social diversity indicates a special interest in modern yoga and its active implementation. The authors of the article try to identify those determinants that contribute to the development of yoga in modern conditions. They emphasize that interest in yoga is being formed under the influence of special factors characteristic of a post-industrial society with new sensations of time. The spread of new intellectual sensitivity also poses certain challenges for humans. Thus, this complex of sociocultural determinants activates interest in the cultural representations of the East. The desire to create personal and social reality sets up certain receptions and practices, and yoga is called upon to fulfill its special mission: it encourages us to study our basic outlooks on life, to become aware of which ones are fruitless nowadays, to search for more suitable ones nowadays in order to substitute the first ones.

Alternative splicing is one of the main regulation pathways in living cells beyond simple changes in the level of protein expression. Most of the approaches proposed in proteomics for the identification of specific splicing isoforms require a preliminary deep transcriptomic analysis of the sample under study, which is not always available, especially in the case of the re-analysis of previously acquired data. Herein, we developed new algorithms for the identification and validation of protein splice isoforms in proteomic data in the absence of RNA sequencing of the samples under study. The bioinformatic approaches were tested on the results of proteome analysis of human melanoma cell lines, obtained earlier by high-resolution liquid chromatography and mass spectrometry (LC-MS). A search for alternative splicing events for each of the cell lines studied was performed against the database generated from all known transcripts (RefSeq) and the one composed of peptide sequences, which included all biologically possible combinations of exons. The identifications were filtered using the prediction of both retention times and relative intensities of fragment ions in the corresponding mass spectra. The fragmentation mass spectra corresponding to the discovered alternative splicing events were additionally examined for artifacts. Selected splicing events were further validated at the mRNA level by quantitative PCR.

This paper analyses a general qualitative depiction of changes in the configuration of the non-linear model of a tractor-semitrailer with excessive turnability under stationary circular motions within a broad range of control parameters, such as the longitudinal velocity of motion and angle of rotation of the wheels of the controlled unit. It allows us to foresee the jackknifing effect and to estimate the conditions necessary for safe travel. The proposed analysis combines the transparent geometric approach to analyse the configuration of a tractor-semitrailer, which was developed earlier by J.P. Pauwelussen, and the ability to investigate real bifurcations of the non-linear equation of handling and the non-linear equation of folding angle tractor-semitrailer analytically. Based on this approach, we obtain new analytical dependencies that determine the boundaries of divergent loss of stability of the entire manifold of stationary states of a tractor-semitrailer model and estimates of the maximum values of the angle of folding under different values of the vehicle's longitudinal velocity. High-performance liquid chromatography (HPLC) is widely used for separation of complex peptide mixtures before mass spectrometry-based proteome analysis. In this analysis, reversed phase HPLC (RPHPLC) using non-polar stationary phases such as surface-modified silica containing alkyl groups (e.g., C18) is typically employed. Because of the high heterogeneity of proteomic samples, multidimensional separation approaches gained increasing attention recently to tackle this complexity and extremely high range of concentrations. In two-dimensional liquid chromatography, hydrophilic interaction chromatography (HILIC) is often a method of choice for combination with RP-HPLC because it uses reversed-phase type eluents and allows efficient separation of polar peptides. Due to the high degree of orthogonality in this two-dimensional separation space, it is tempting to develop approaches for predicting peptide retention times for HILIC-based separations similar to the ones for RP-HPLC. Recent successful efforts in this area were focused on developing retention coefficient (RC)-based approaches. Herein, we explored the feasibility of using a statistical thermodynamic model for prediction of peptide retention times in HILIC separations and determined the phenomenological parameters of the model for a bare silica column. The performance of the developed model was tested using HPLC-MS analysis of a set of synthetic peptides, as well as a tryptic peptide mixture. The Tibellus oblongus spider is an active predator that does not spin webs and remains poorly investigated in terms of venom composition. Here, we present a new toxin, named Tbo-IT2, predicted by cDNA analysis of venom glands transcriptome. The presence of Tbo-IT2 in the venom was confirmed by proteomic analyses using the LC-MS and MS/MS techniques. The distinctive features of Tbo-IT2 are the low similarity of primary structure with known animal toxins and the unusual motif of 10 cysteine residues distribution. Recombinant Tbo-IT2 (rTbo-IT2), produced in E. coli using the thioredoxin fusion protein strategy, was structurally and functionally studied. rTbo-IT2 showed insecticidal activity on larvae of the housefly Musca domestica (LD100 200 μg/g) and no activity on the panel of expressed neuronal receptors and ion channels. The spatial structure of the peptide was determined in a water solution by NMR spectroscopy. The Tbo-IT2 structure is a new example of evolutionary adaptation of a well-known inhibitor cystine knot (ICK) fold to 5 disulfide bonds configuration, which determines additional conformational stability and gives opportunities for insectotoxicity and probably some other interesting features.
