Staphylococcus aureus is an important pathogen associated with various infections, including foodborne outbreaks worldwide. Due to possible contamination during food preparation by food handlers, the presence of this species in kitchens environmental is a risk to global health, mainly S. aureus biofilm-producing. Moreover, there are few reports of the evaluation of biofilm production of S. aureus obtained from food handlers in Brazil. The study aimed to characterize phenotypically and genotypically biofilm production associated with ica-dependent and ica-independent genes in S. aureus isolated from food handlers in this country. A total of 74 Staphylococcus sp. strains were isolated and 37 (50%) were identified as S. aureus through the sau gene detection. All S. aureus strains exhibited a high ability to form biofilm and detected several genes involved in this process, spotlighting the icaA (86.5%), icaD (86.5%), sasG (78.7%), atlA (78.7%) and aap (78.7%). Furthermore, the predominance of biofilm-associated genes in the strong biofilm-producing isolates by TB and TCP was found, presenting a significant association between methods and genes detected (p = S. aureus biofilm-producing, triggering the transmission of this genus isolated worldwide.

O objetivo foi avaliar a qualidade microbiológica da carne moída e quibe cru comercializados em Presidente Prudente-SP. Foram coletados 30 amostras de carne moída e quibe crupara a quantificação de coliformes totais e termotolerantes, aeróbios mesófilos, bolores e leveduras. Foi aplicado um formulário para cada estabelecimento onde foram observadas condições higiênicas sanitárias e adequações do ambiente para o manuseio da carne. Foi observado condições higiênicas insatisfatórias devidoa ausência de paramentação dos funcionários e a presença de animais ou insetos no ambiente. Foi notado altas contagens para aeróbios mesofilos, bolores e leveduras e coliformes totais. Coliformes termotolerantesestava presente em várias amostras de carne moída e quibe cru, entretanto, todos os valores estavam abaixo do parâmetro estabelecido pela legislação. Os dados despertam a atenção em relação a Saúde Pública uma vez que as falhas das condições higiênico-sanitárias podem representar um risco potencial à saúde dos consumidores. .

Hospital environment has factors that promote increased infections caused by microorganisms allowing them to remain viable for long periods on surfaces and fomites. The aim of our study was to identify the viability of Candida albicans in hospital environment common surfaces with biological fluids presence and disinfectants. Four surfaces (cotton fabric, synthetic fabric, egg crate foam mattress and, ceramic floor) were contaminated with a strain of Candida albicans ATCC 26790 and with an association of distilled water, blood, saliva and urine. Also, combinations of surfaces and fluids with addition of 70% alcohol and 2% sodium hypochlorite were tested. Viability was identified at 1, 3, 7, 14 and 21 days by Colony Forming Units counting. C. albicans presented 14 days viability in hospitals fabrics with absence of liquids and maintained this persistence under influence of distilled water and blood. It also showed low growth with urine addition and there was growth inhibition with 2% sodium hypochlorite while with 70% alcohol and blood there was persistence for up to 7 days in synthetic fabric and mattress. The understanding of these yeasts persistence in hospital surfaces and fomites is essential for a better delineation of protocols control the spread and these infections prevention in healthcare environments. El entorno hospitalario cuenta con factores que favorecen el aumento de infecciones provocadas por microorganismos, lo que les permite permanecer viables durante largos periodos en superficies y fómites. El objetivo de nuestro estudio fue identificar la viabilidad de Candida albicans en superficies comunes en un ambiente hospitalario con presencia de fluidos biológicos y desinfectantes. Cuatro superficies (tela de algodón, tela sintética, colchón de espuma tipo caja de huevos y piso de cerámica) fueron contaminadas con una cepa de Candida albicans ATCC 26790 y con una asociación de agua destilada, sangre, saliva y orina. Además, se probaron combinaciones de superficies y fluidos con la adición de 70% de alcohol y 2% de hipoclorito de sodio. La viabilidad se identificó a los 1, 3, 7, 14 y 21 días contando las Unidades Formadoras de Colonias. C. albicans mostró viabilidad durante 14 días en tejidos hospitalarios sin líquidos y mantuvo esta persistencia bajo la influencia de agua destilada y sangre. También mostró bajo crecimiento con la adición de orina e inhibición del crecimiento con hipoclorito de sodio al 2% mientras que con alcohol al 70% y persistencia de sangre hasta por 7 días en tela sintética y colchón. Comprender la persistencia de estas levaduras en las superficies y fómites hospitalarios es fundamental para un mejor diseño de protocolos de control de la diseminación y prevención de estas infecciones en entornos sanitarios. O ambiente hospitalar possui fatores que promovem o aumento das infecções causadas por microrganismos, permitindo que permaneçam viáveis por longos períodos em superfícies e fômites. O objetivo do nosso estudo foi identificar a viabilidade de Candida albicans em superfícies comuns de ambiente hospitalar com presença de fluidos biológicos e desinfetantes. Quatro superfícies (tecido de algodão, tecido sintético, colchão de espuma de engradado de ovo e piso de cerâmica) foram contaminadas com uma cepa de Candida albicans ATCC 26790 e com uma associação de água destilada, sangue, saliva e urina. Além disso, foram testadas combinações de superfícies e fluidos com adição de álcool 70% e hipoclorito de sódio 2%. A viabilidade foi identificada em 1, 3, 7, 14 e 21 dias por contagem de Unidades Formadoras de Colônia. C. albicans apresentou viabilidade de 14 dias em tecidos hospitalares com ausência de líquidos e manteve essa persistência sob influência de água destilada e sangue. Também apresentou baixo crescimento com adição de urina e inibição do crescimento com hipoclorito de sódio 2% enquanto com álcool 70% e sangue persistência por até 7 dias em tecido sintético e colchão. O entendimento da persistência dessas leveduras em superfícies hospitalares e fômites é essencial para um melhor delineamento de protocolos de controle de disseminação e prevenção dessas infecções em ambientes de saúde.

Staphylococcus aureus biofilms have been recognized as a leading cause of multiple infections, including implant-associated infections and chronic wounds. We evaluated the colonization capacity of two distinct textured prostheses by different bacterial strains. Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Proteus mirabilis and Enterococcus faecalis were evaluated. Initially, the hydrophobicity and biofilm formation capacity were determined. Subsequently, 20 fragments of vascular prosthesis and 20 silicone prostheses were embedded in suspensions with the microorganisms and incubated. The prostheses were then sown in culture medium and incubated for 48 hours. Petri dishes were photographed and analyzed by fractal dimension. The Kruskal-Wallis test and the Dunn test were applied for the analysis of biofilm formation. To compare the mean intensity for the type of bacteria and the type of prosthesis, a general linear model was applied. Staphylococcus aureus was the bacterium with the highest colonization density in both prostheses (p = 0.0001). E. coli showed strong adherence in the biofilm formation capacity test (p = 0.0001), however, it did not colonize either prosthesis. We demonstrated that Staphylococcus aureus has a greater affinity for vascular and silicone prostheses than other bacteria. Se ha reconocido que las biopelículas de Staphylococcus aureus son una de las principales causas de infecciones múltiples, incluidas las infecciones asociadas a implantes y las heridas crónicas. Evaluamos la capacidad de colonización de dos prótesis texturizadas distintas por diferentes cepas bacterianas. Se evaluaron Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Proteus mirabilis y Enterococcus faecalis. Inicialmente se determinó la hidrofobicidad y la capacidad de formación de biopelículas. Posteriormente, se embebieron 20 fragmentos de prótesis vasculares y 20 prótesis de silicona en suspensiones con los microorganismos y se incubaron. A continuación, las prótesis se sembraron en medio de cultivo y se incubaron durante 48 horas. Las placas de Petri se fotografiaron y analizaron por dimensión fractal. Se aplicaron la prueba de Kruskal-Wallis y la prueba de Dunn para el análisis de la formación de biopelículas. Para comparar la intensidad media para el tipo de bacteria y el tipo de prótesis, se aplicó un modelo lineal general. Demonstramos que o Staphylococcus aureus tem maior afinidade por próteses vasculares e de silicone do que outras bactérias.

Outbreaks and deaths related to Foodborne Diseases (FBD) occur constantly in the world, as a result of the consumption of contaminated foodstuffs with pathogens such as Listeria monocytogenes, Escherichia coli, Staphylococcus aureus, Salmonella spp, Clostridium spp. and Campylobacter spp. The purpose of this review is to discuss the main omic techniques applied in foodborne pathogen and to demonstrate their functionalities through the food chain and to guarantee the food safety. The main techniques presented are genomic, transcriptomic, secretomic, proteomic, and metabolomic, which together, in the field of food and nutrition, are known as "Foodomics." This review had highlighted the potential of omics to integrate variables that contribute to food safety and to enable us to understand their application on foodborne diseases. The appropriate use of these techniques had driven the definition of critical parameters to achieve successful results in the improvement of consumers health, costs and to obtain safe and high-quality products.

However, studies regarding the epidemiology and dissemination of S. aureus and Methicillin-resistant S. aureus (MRSA) within the Primary Health Care in Brazil are scarce. The current study aimed to detect and characterize S. aureus and MRSA strains from the nasal cavities of 63 healthcare working in primary health care units in order to determine the prevalence of S. aureus and MRSA, biofilm formation and resistance profile of these isolates. PCR reactions were performed for detecting mecA, icaA and ical) genes. The phenotypic antimicrobial susceptibility was assessed by the disk diffusion method and biofilm formation by the Congo Red Agar (CRA) method. The MRSA isolates were typed for the Staphylococcal Cassette Chromosome mec (SCCmec). The prevalence of nasal carriage of S. aureus was 74.6%. of which 72.3% were MRSA carrying SCCmec type I (24.4%), III (34.1%), IV (36.6%). Two (4.9%) isolates presented a non-typeable cassette by the performed technique. The antimicrobial susceptibility evaluation evidenced penicillin resistance in 66.1% of S. aureus, erythromycin resistance in 49.2%. while 37.3% were resistant to oxacillin, 28.8% to cefoxitin, 5.1% to levofloxacin and 5.1% to clindamycin. All isolates were biofilm producers and 96.6% of the strains contained the ica biofilm-forming genes (icaA and/or ical)). We have demonstrated a high prevalence of S. aureus and MRSA carriage among health care working in Primary health Care units, the presence of SCCmec types I, III and IV, in addition to their high ability to form biofilm, factors that possibly contribute to the dissemination and persistence of these pathogens within the primary care services. These observations highlight the importance of broadening the perspective of Health Care-Associated Infections prevention, including all health care levels, which are currently little explored. In addition, the dynamics and resistance mechanisms of S. aureus transmission still need to be further clarified to enable the implementation of more effective prevention measures. The gold standard for quantifying bacteria both in routine diagnostics and in research is plating followed by count of colony-forming units (CFU). But, manual CFU counting on plates is time-consuming and subjective. We evaluated fractal dimension as a new methodology for evaluating CFU. Twenty fragments of expanded polytetrafluoroethylene (ePTFE) synthetic vascular prosthesis and 20 silicone prostheses were embedded in bacterial suspensions and incubated. The prostheses were then sown in solid culture medium and incubated for 48 h. Petri dishes were photographed and analyzed by fractal dimension. There was correlation between the number of CFU in manual counting and the fractal dimension analysis (p = 0.0001). We demonstrated that fractal dimension is a useful method for microbiological analyses in researches. Escherichia coliisone of the main microorganisms that cause urinary tract infection (UTI). The objective of this work was to evaluate the presence of virulence genes, the biofilm formation and the potential inhibition of biofilms by essential oil of ginger and star anise in Escherichia coliisolated from a clinical laboratory at Presidente Prudente-SP. In the biofilm formation, the titration microplate technique was used and the polymerase chain reaction was used to evaluate the presence of the virulence genes. The results showed that 100% of the isolates formed biofilms. Only six isolates presented reduction of biofilms in presence of essential oil. The most frequent virulence gene was fimH(100%) followed by kpsMTII (99.9%) papC(98.9%) and fliC (84.8%). We emphasize that the microorganisms may be in a phase of progressive accumulation of virulence factors which facilitates the increase of the severity of the host infection

O açaí é considerado um alimento muito nutritivo e atualmente, tem sido bastante consumido na forma de “açaí na tigela”. Este trabalho teve como finalidade avaliar a qualidade microbiológica da preparação “açaí na tigela” fornecida em uma cidade do Oeste Paulista. Foram coletadas 20 amostras de açaí na tigela de diversos estabelecimentos e em seguida foi realizada a análise microbiológica. Os resultados deste trabalho demonstraram que cerca de 35% das amostras foram consideradas inadequadas para a quantidade bolores e leveduras. Cerca de sete amostras apresentaram valores mínimos para presença de coliformes totais e nenhumas das amostras foram positivas para presença de Salmonella sp., coliformes termotolerantes e Escherichia coli. Destacamos a importância de se investigar e avaliar as condições encontradas nos alimentos prontos para o consumo, pois falhas nas condições higiênico-sanitárias podem representar um risco potencial à saúde dos consumidores.

No. of bitstreams: 0 Previous issue date: 2020-01-01 Studies reported the harmful effects of 2,4-D on body tissues, provoking changes in the anatomy and physiology of the kidneys, liver, and testicles. Thus, the objective was to evaluate if there were alterations in the bone quality of the tibia of rats submitted to feed consumption that were exposed to three different 2,4-D doses. Male Wistar rats were divided into four groups: oral control group (C: feed consumption without 2,4-D contamination); low oral concentration group (G3: contaminated feed with low concentration of 2,4-D); medium concentration group (G6: contaminated feed with medium concentration of 2,4-D); and high concentration group (G9: contaminated feed with high concentration of 2,4-D). The results demonstrated alterations of the mechanical properties and Raman ratios of the tibias of the contaminated groups. The maximum load, maximum stress, elastic modulus, and the cortical area were lower in the G6 and G9 compared to C group. The mineral-to-matrix ratio (relative mineral to organic content) was lower in the G6 and G9 groups compared to C group, but carbonate-to-matrix ratio (indicator of bone turnover) was higher in both groups. Thus, it is possible to suggest that the 2,4-D herbicide performed deleterious effects on the bone quality of male Wistar rats. The aim of this study was to evaluate whether pre- and postcontamination washing of 2 prostheses with different textures can decrease bacterial contamination.The following microorganisms were evaluated: Staphylococcus aureus, Staphylococcus epidermidis, Proteus mirabilis and Enterococcus faecalis. Silicone and expanded polytetrafluoroethylene vascular prostheses were used and divided into 3 groups: prostheses contaminated; prostheses contaminated and treated before contamination; and prostheses contaminated and treated after contamination. Treatments were performed with antibiotic solution, chlorhexidine and lidocaine. After one week of incubation, the prostheses were sown in culture medium, which was incubated for 48 hours. The area of colony formation was evaluated by fractal dimension, an image analysis tool.The antibiotic solution inhibited the growth of S epidermidis and chlorhexidine decrease in 53% the colonization density for S aureus in for both prostheses in the pre-washing. In postcontamination washing, the antibiotic solution inhibited the growth of all bacteria evaluated; there was a 60% decrease in the colonization density of S aureus and absence of colonization for E faecalis with chlorhexidine; and lidocaine inhibited the growth of S aureus in both prostheses.Antibiotic solution showed the highest efficiency in inhibiting bacterial growth, especially for S epidermidis, in both washings. Lidocaine was able to reduce colonization by S aureus in post-contamination washing, showing that it can be used as an alternative adjuvant treatment in these cases. Background The hospital environment is susceptible to bacterial contamination along with survival in fomites and surfaces, allowing dissemination of potential pathogenic strains. The present research aimed to evaluate the influence of biological fluids in bacterial viability on fomites and surfaces commonly present in nosocomial environment. Methods Four different fomites and surfaces (ceramic floor, cotton fabric fragments and synthetic fibers, and eggcrate foam mattress) were contaminated with potential pathogens ( Staphylococcus aureus, Escherichia coli, Enterococcus faecalis, Pseudomonas aeruginosa , and Klebsiella pneumoniae ), then submitted to influence of biological fluids (blood, urine, artificial saliva). The viability of strains was evaluated at 24 hours after contamination and then in intervals of 7 days, by the colony-forming unit count technique. Results S aureus presented viability (>70 days) in all conditions tested, E faecalis and K pneumoniae had decreased viability over time, and E coli did not exhibit a growth relationship with surfaces or fluids. Persistence and adaptability capacity of potential pathogens in fomites and surfaces exposed to the patient are important for guidance, planning, and outlining of protocols for microorganism dissemination control and prevention in the health care environment.

As bactérias utilizam um sistema de comunicação intercelular denominado quorum sensing (QS) mediado por sinais químicos chamados de autoindutores. Entre os fenótipos regulados pelo QS destacam-se a produção de fatores de virulência, formação de biofilmes, compostos antimicrobianos e a motilidade. Diante da relevância desses fenótipos, há um grande interesse em encontrar substâncias que interfiram nesta comunicação. A inibição do QS é conhecida como quorum quenching (QQ). Dessa forma, este estudo objetivou avaliar o potencial de inibição do QS por compostos fenólicos em bactérias biossensoras do QS e de relevância em alimentos. Inicialmente, foi realizado um estudo in silico de ancoragem molecular de 79 compostos fenólicos pré-selecionados com seis variantes estruturais do ativador transcricional do QS CviR, de Chromobacterium violaceum, a fim de selecionar os compostos de maior potencial de inibição do QS. A atividade antimicrobiana dos compostos selecionados foi investigada pela concentração inibitória mínima (MIC) e pela determinação do crescimento microbiano em caldo de cultura. O efeito inibitório sobre o QS e fatores de virulência foi testado sobre a produção de violaceína em C. violaceum ATCC 12472 e 026, formação de biofilme e motilidade tipo swarming e swimming em Aeromonas hydrophila, Salmonella enterica serovar Montevideo e Serratia marcescens, além de avaliar a sensibilidade térmica e ao antibiótico canamicina dessas estirpes junto com os compostos fenólicos. A partir dos resultados da ancoragem molecular, cinco compostos foram selecionados para estudos in vitro: curcumina, capsaicina, resveratrol, ácido gálico e floridzina. Estes compostos se ligaram com boa afinidade, menor energia potencial, a pelo menos quatro variantes estruturais de CviR. As curvas de crescimento demonstraram que o ácido gálico apresentou MIC de 9,4 mM para C. violaceum ATCC 12472 e 026. Porém, os demais compostos apresentaram MIC superior à maior concentração testada. Os resultados de inibição da produção de violaceína mostraram que curcumina a 25 µM, capsaicina a 70 µM e resveratrol a 6 µM inibiram até 50% da produção do pigmento para a estirpe ATCC 12472. Adicionalmente, curcumina a 5 µM inibiu a produção de violaceína na estirpe 026. Com relação à inibição da formação de biofilme, resveratrol inibiu até 80% em A. hydrophila; capsaicina e curcumina até 40% em S. Montevideo; resveratrol e capsaicina inibiram até 60% em S. marcescens. No teste de motilidade, curcumina inibiu completamente a formação de swarming em S. marcescens, e não foi observada inibição pelos demais compostos nos outros micro-organismos avaliados. Curcumina e resveratrol foram capazes de aumentar a sensibilidade à canamicina nas estirpes avaliadas, em diferentes graus de inibição. Entretanto, não houve inibição no teste de sensibilidade térmica para as estirpes em estudo. Os resultados obtidos para os compostos curcumina e resveratrol podem impulsionar novos estudos de sinergismo com antibióticos, atividade anti-QS e possíveis aplicações nas indústrias alimentícia e farmacêutica. Bacteria use an intercellular communication system termed quorum sensing (QS) mediated by chemical signals called autoinducers. Among the phenotypes regulated by QS, the production of virulence factors, biofilms, antimicrobial compounds, and motility stand out. Given the relevance of these phenotypes, there is a great interest in finding substances that interfere in this communication. The inhibition of quorum sensing is known as quorum quenching (QQ). This study evaluated the potential for QS inhibition by phenolic compounds in QS biosensor bacteria and foodborne microorganisms. Initially, an in silico molecular docking study of 79 pre-selected phenolic compounds with six structural variants of the Chromobacterium violaceum QS transcriptional factor CviR was carried out to select the compounds with the greatest potential for QS interference. The antimicrobial activity of the selected compounds was investigated by the minimum inhibitory concentration (MIC) and by the determination of microbial growth in broth. The inhibitory effect on QS regulated phenotypes and virulence factors was tested on violacein production in C. violaceum ATCC 12472 and 026, biofilm formation and swarming and swimming motility in Aeromonas hydrophila, Salmonella enterica serovar Montevideo and Serratia marcescens, in addition to evaluating thermal sensitivity and microbial inhibition by kanamycin of these strains combined with phenolic compounds. Five compounds were selected in molecular docking studies for in vitro tests: curcumin, capsaicin, resveratrol, gallic acid and phloridizin. These compounds bound with good in silico affinity to at least four CviR structural variants. The growth curves showed that gallic acid had a MIC of 9.4 mM on C. violaceum ATCC 12472 and 026. However, the other compounds had a MIC higher than the highest tested concentration. The results for violacein inhibition showed that curcumin at 25 µM, capsaicin at 70 µM and resveratrol at 6 µM inhibited up to 50% of pigment production in strain ATCC 12472. Additionally, curcumin at 5 µM inhibited the production of violacein in strain 026. Regarding the inhibition of biofilm formation, resveratrol inhibited up to 80% in A. hydrophila; capsaicin and curcumin up to 40% in S. Montevideo; resveratrol and capsaicin inhibited up to 60% in S. marcescens. In the motility test, curcumin completely inhibited swarming in S. marcescens, and no inhibition of the other compounds was observed for the other microorganisms. Curcumin and resveratrol were able to increase the sensitivity to kanamycin in the evaluated strains, with different degrees of inhibition. However, there was no inhibition in the thermal sensitivity test. The results obtained for the compounds curcumin and resveratrol can stimulate further studies of synergisms with anti-QS activity and applications in the food and pharmaceutical industries.
