Abstract Metabolic abnormalities represent one of the pathological features of chronic obstructive pulmonary disease (COPD). Glutamic pyruvate transaminase 2 (GPT2) is involved in glutamate metabolism and lipid synthesis pathways, whilst the exact roles of GPT2 in the occurrence and development of COPD remains uncertain. This study aims at investigating how GPT2 and the associated genes modulate smoking‐induced airway epithelial metabolism and damage by reprogramming lipid synthesis. The circulating or human airway epithelial metabolomic and lipidomic profiles of COPD patients or cell‐lines explored with smoking were assessed to elucidate the pivotal roles of GPT2 in reprogramming processes. We found that GPT2 regulate the reprogramming of lipid metabolisms caused by smoking, especially phosphatidylcholine (PC) and triacylglycerol (TAG), along with changes in the expression of lipid metabolism‐associated genes. GPT2 modulated cell sensitivities and survival in response to smoking by enhancing mitochondrial functions and maintaining lipid and energy homeostasis. Our findings provide evidence for the involvement of GPT2 in the reprogramming of airway epithelial lipids following smoking, as well as the molecular mechanisms underlying GPT2‐mediated regulation, which may offer an alternative of therapeutic strategies for chronic lung diseases.

Abstract Background Peripheral blood carries a reservoir of mRNAs that regulate cardiac structure and function potential. Although it is well recognized that the typical symptoms of Myxomatous Mitral Valve Disease (MMVD) stage B2 are long-standing hemodynamic disorder and cardiac structure remodeling caused by mitral regurgitation, the transcriptomic alterations in blood from such dogs are not understood. Results In the present study, comparative high-throughput transcriptomic profiling of blood was performed from normal control (NC) and naturally-occurring MMVD stage B2 (MMVD) dogs. Using Weighted Gene Co-expression Network Analyses (WGCNA), Gene Ontology (GO), and Kyoto Encyclopedia of Gene and Genomes (KEGG), we identified that the turquoise module was the most highly correlated with echocardiographic features and found 64 differentially expressed genes (DEGs) that were significantly enriched in platelet activation related pathways. Therefore, from the turquoise module, we selected five DEGs (MDM2, ROCK1, RIPK1, SNAP23, and ARHGAP35) that, according to real-time qPCR, exhibited significant enrichment in platelet activation related pathways for validation. Mesoproterozoic (ca. 1.3 Ga) magmatism in the North China Craton (NCC) was dominated by mafic intrusions (dolerite sills) with lesser amounts of granitic magmatism, but our lack of knowledge of this magmatism hinders our understanding of the evolution of the NCC during this period. This study investigated porphyritic granites from the Huade–Kangbao area on the northern margin of the NCC. Zircon dating indicates the porphyritic granites were intruded during the Mesoproterozoic between 1285.4 ± 2.6 and 1278.6 ± 6.1 Ma. The granites have high silica contents (SiO2 = 63.10–73.73 wt.%), exhibit alkali enrichment (total alkalis = 7.71–8.79 wt.%), are peraluminous, and can be classified as weakly peraluminous A2-type granites. The granites have negative Eu anomalies (δEu = 0.14–0.44), enrichments in large-ion lithophile elements (LILEs; e.g., K, Rb, Th, and U), and depletions in high-field-strength elements (HFSEs; e.g., Nb, Ta, and Ti). εHf(t) values range from –6.43 to +2.41, with tDM2 ages of 1905–2462 Ma, suggesting the magmas were derived by partial melting of ancient crustal material. The geochronological and geochemical data, and regional geological features, indicate the Mesoproterozoic porphyritic granites from the northern margin of the NCC formed in an intraplate tectonic setting during continental extension and rifting, which represents the response of the NCC to the break-up of the Columbia supercontinent.

In this letter, we propose the design and manufacture of an all-dielectric metasurface using high-permittivity ceramics. The unit structure consists of a rectangular ceramic, which is asymmetric in the x and y direction. Electric and magnetic resonances are generated to form cross-polarization reflection. As an example, cross-polarization reflection is demonstrated both numerically and experimentally. This work not only gives the designing method but also discusses how to manufacture a large sample and the difference between ideal simulation and the practical sample. After a large number of simulations and experiments, the feasible design methods and problems needing attention in design are given. The experiment result is according to the adjusted simulated result. The result confirms the feasibility of the design and processing of an all-dielectric metasurface in the microwave band. If we can find a suitable material preparation process or optimize the unit structure that is easy to process, the all-dielectric polarization conversion metasurface can realize a maximum conversion efficiency of nearly 100% and form a broadband working band. Since such metasurfaces are made of low-loss high-permittivity ceramics, they are of important value especially in high temperature or high-power applications. This method can also be used to design all-dielectric metasurfaces in other frequencies by changing the geometry parameters.

Parent’s nutrition knowledge, attitudes, and dietary practices (KAP) play imperative roles in preventing malnutrition for themselves and their children. Our study aimed to determine the status and contributing factors of nutrition KAP among parents of children and adolescents. A total of 1746 parents (mean age 39.67 ± 5.38 years, females accounting for 69.82%) of primary and junior high school students in Weifang, China, completed a self-reported KAP questionnaire in August 2021. An analysis of Pearson product-moment correlation was conducted to determine the relationship between knowledge, attitudes, and practices. Chi-square test, followed by a multivariable robust Poisson regression analysis, was performed to identify the contributing factors to parents’ KAP. A 65.94% awareness rate of nutritional knowledge was observed. The Northeast China Block is a major component of the Central Asian Orogenic Belt, and its tectonic evolution has attracted much research attention. Ordovician strata are important in reconstructing the tectonic evolution of the Northeast China Block. This paper presents the results of sedimentological, zircon U–Pb, and geochemical analyses of sandstones of the Luohe Formation in the Wunuer area, Northern Great Xing’an Range, Northeast China. Lithological data, sedimentary structures, and grain-size analysis indicate that the Luohe Formation was deposited in a shallow marine environment. Detrital zircon U–Pb dating yields age peaks of 463, 504, 783, 826, 973, and 1882 Ma for sandstones from the Luohe Formation. The youngest zircon grain age of 451 ± 6 Ma represents the maximum depositional age of the Luohe Formation. The peak age at 463 Ma is consistent with the timing of post-collisional magmatism and the formation of the Duobaoshan island arc, while the peak at 504 Ma is consistent with the timing of magmatic activity related to the collision between the Erguna and Xing’an blocks. The peaks at 788, 826, 973, and 1882 Ma correspond to magmatism in the Erguna block, these ages indicate that the sandstones of the Luohe Formation were derived mainly from the Erguna block. Sandstone modal compositional analysis indicates that the provenance of the Luohe Formation was mainly a magmatic arc. The geochemical compositions of the sandstones suggest that the source rocks have continental island arc signatures. Based on the depositional age, sedimentary environment, provenance, and regional geology, it is concluded that the Luohe Formation was deposited in a back-arc basin setting during the formation of the Duobaoshan island arc–basin system in response to subduction of the Paleo-Asian oceanic plate.

Background:SERPINE1, a serine protease inhibitor involved in the regulation of the plasminogen activation system, was recently identified as a cancer-related gene. However, its clinical significance and potential mechanisms in pan-cancer remain obscure.Methods: In pan-cancer multi-omics data from public datasets, including The Cancer Genome Atlas (TCGA) and Genotype-Tissue Expression (GTEx), and online web tools were used to analyze the expression of SERPINE1 in different cancers and its correlation with prognosis, genetic alteration, DNA promoter methylation, biological processes, immunoregulator expression levels, immune cell infiltration into tumor, tumor mutation burden (TMB), microsatellite instability (MSI), immunotherapy response and drug sensitivity. Further, two single-cell databases, Tumor Immune Single-cell Hub 2 (TISCH2) and CancerSEA, were used to explore the expression and potential roles of SERPINE1 at a single-cell level. The aberrant expression of SERPINE1 was further verified in clear cell renal cell carcinoma (ccRCC) through qRT-PCR of clinical patient samples, validation in independent cohorts using The Gene Expression Omnibus (GEO) database, and proteomic validation using the Clinical Proteomic Tumor Analysis Consortium (CPTAC) database.Results: The expression of SERPINE1 was dysregulated in cancers and enriched in endothelial cells and fibroblasts. Copy number amplification and low DNA promoter methylation could be partly responsible for high SERPINE1 expression. High SERPINE1 expression was associated with poor prognosis in 21 cancers. The results of gene set enrichment analysis (GSEA) indicated SERPINE1 involvement in the immune response and tumor malignancy. SERPINE1 expression was also associated with the expression of several immunoregulators and immune cell infiltration and could play an immunosuppression role. Besides, SERPINE1 was found to be related with TMB, MSI, immunotherapy response and sensitivity to several drugs in cancers. Finally, the high expression of SERPINE1 in ccRCC was verified using qRT-PCR performed on patient samples, six independent GEO cohorts, and proteomic data from the CPTAC database.Conclusion: The findings of the present study revealed that SERPINE1 exhibits aberrant expression in various types of cancers and is associated with cancer immunity and tumor malignancy, providing novel insights for individualized cancer treatment.

Abstract Background Bovine parainfluenza virus type 3 (BPIV3) infection often causes respiratory tissue damage and immunosuppression and further results in bovine respiratory disease complex (BRDC), one of the major diseases in dairy cattle, caused huge economical losses every year. However, the pathogenetic and immunoregulatory mechanisms involved in the process of BPIV3 infection remain unknown. Proteomics is a powerful tool for high-throughput identification of proteins, which has been widely used to understand how viruses interact with host cells. Methods In the present study, we report a proteomic analysis to investigate the whole cellular protein alterations of MDBK cells infected with BPIV3. To investigate the infection process of BPIV3 and the immune response mechanism of MDBK cells, isobaric tags for relative and absolute quantitation analysis (iTRAQ) and Q-Exactive mass spectrometry-based proteomics were performed. The differentially expressed proteins (DEPs) involved in the BPIV3 invasion process in MDBK cells were identified, annotated, and quantitated. Results A total of 116 proteins, which included 74 upregulated proteins and 42 downregulated proteins, were identified as DEPs between the BPIV3-infected and the mock-infected groups. These DEPs included corresponding proteins related to inflammatory response, immune response, and lipid metabolism. These results might provide some insights for understanding the pathogenesis of BPIV3. Fluorescent quantitative PCR and western blotting analysis showed results consistent with those of iTRAQ identification. Interestingly, the upregulated protein MKK3 was associated with the p38 MAPK signaling pathway. Conclusions The results of proteomics analysis indicated BPIV3 infection could activate the p38 MAPK pathway to promote virus replication.

Abstract Background The fundamental initial step in the metabolic process of fatty acids (FA) is facilitated by Acyl‐coenzyme A synthetase (ACS). ACS enables FAs to engage in both anabolic and catabolic pathways by creating thioester bonds with CoA, thus triggering their activation. This paper demonstrated the effect of in vitro cells on the ACS family of key enzymes in FA metabolism in response to inflammatory or lipid stimulating factors and provided insight into the relevant metabolic mechanisms. Methods The expression levels of 27 ACS family members were assessed across various lung epithelial cell lines, such as HBE, SPC‐1A, or NCI‐H460, in response to external challenges including lipopolysaccharide (LPS), lysophosphatidylcholine (lysoPC), cigarette smoking extract (CSE), cholesterol, and interference with the ACSL5 gene in response to FASN, CPT1A inhibitors. Results Our data showed that ACSF2 was highly expressed in SPC‐A1 and NCI‐H460 cells compared with HBE cell, while ACSL4 and ACSVL4 were lower expressed. The 27 ACS family genes associated with FA metabolism varied under several different exogenous stimuli.

Although secretory IgA (SIgA) is the dominant antibody in mucosal secretions, the capacity of the SIgA–antigen complex to prime the activation of dendritic cells (DCs) and T cells in the intestinal epithelium is not well understood. To this end, the SIgA–ETEC F5 immune complexes (ICs) were prepared via Ni-NTA pull-down. After injecting the ICs into the intestines of SPF BALB/c mice, most ICs were observed in the Peyer’s patch (PP). We established a microfold (M) cell culture model in vitro for transport experiments and the inhibition test. To evaluate the priming effect of mucosal immunity, we employed the DC2.4 stimulation test, T lymphocyte proliferation assays, and cytokine detection assays. We found that the ICs were taken up via clathrin-dependent endocytosis through M cells. The high expression of costimulatory molecules CD86, CD80, and CD40 indicated that the ICs promoted the differentiation and maturation of DC2.4 cells. The stimulation index (SI) in the complex group was significantly higher than in the control group, suggesting that the ICs stimulated the proliferation of primed T cells. The secretion of some cytokines, namely TNF-α, IFN-γ, IL-2, IL-4, IL-5, and IL-6, in spleen cells from the immunized mice was upregulated. These results indicate that ETEC F5 delivery mediated by SIgA in PPs initiates mucosal immune responses.

Slightly off-axis digital holography is proposed using transmission grating to obtain quantitative phase distribution. The experimental device is based on an improved 4f optical system in which a two-window input plane is used to form the object beam and reference beam. Then, the two beams are diffracted into multiple orders by the transmission grating placed at the Fourier plane. By applying a modified Michelson configuration, the interference patterns can be generated by the object and reference beams from different diffraction orders. After translating the grating, a random phase shift can be introduced to the hologram. To demonstrate the feasibility of our method, both thick and thin phase specimens are retrieved using two carrier phase-shifting holograms. Furthermore, we use the phase reconstruction algorithm based on the NVIDIA CUDA programming model to reduce the retrieval time. Meanwhile, we optimize the discrete cosine transform (DCT)-based least-squares unwrapping algorithm to unwrap the phase. By porting the entire phase reconstruction process to the graphics processing unit (GPU), the phase retrieval acceleration and execution efficiency significantly improve. To demonstrate the feasibility of our method, it is found that our method can measure the surface profiles of standard elements, such as a plano-convex cylinder lens and a microlens array, with a relative error of about 0.5%. For holograms with a different phase shift, the root-mean-square (RMS) value of the phase difference for the main imaging region is about 0.2 rad. By accelerating the phase reconstruction with GPU implementation, a speedup ratio of about 20× for the thick phase specimen and a speedup ratio of about 15× for the thin-phase specimen can be obtained for holograms with a pixel size of 1024 × 1024.

Northeast China composes the main part of the Central Asian Orogenic Belt. Traditionally, Northeast China has been considered a collage of several microcontinental blocks. However, the tectonic evolution of these blocks remains uncertain. Igneous rocks can be used to infer the magmatic histories of the blocks and thus help reconstruct their evolution. In this study, we present new zircon U–Pb and whole-rock geochemical data for Carboniferous igneous rocks from the Wunuer area, northern Great Xing’an Range, Northeast China, to constrain the Carboniferous amalgamation of the united Xing’an–Erguna and Songnen–Zhangguangcai Range massifs. On the basis of zircon U–Pb dating results, we identify two main stages of magmatism, i.e., early Carboniferous (332–329 Ma) and late Carboniferous (312–310 Ma). The early Carboniferous igneous rocks include diorites and granodiorites, with the former being classified as calc-alkaline to tholeiitic and the latter as tholeiitic. Both rock types are enriched in Th and U and depleted in Nb and Ti. The rocks display slightly fractionated rare earth element (REE) patterns, with an enrichment in light REEs and a depletion in heavy (H)REEs. The geochemical characteristics of the early Carboniferous rocks indicate that they formed in a subduction-related continental-arc setting. The late Carboniferous igneous rocks include monzogranites and syenogranites, both of which are classified as high-K calc-alkaline rocks and show enrichment in Th, U, and Rb and depletion in Nb and Ti. The rocks display strongly fractionated REE patterns, with an enrichment in light REEs and a depletion in HREEs. The geochemical characteristics of the late Carboniferous rocks indicate that they formed in a syn-collisional tectonic setting. Combining the new geochronological and geochemical results and inferred tectonic settings with regional magmatic data, we propose a new three-stage model to interpret the late Paleozoic tectonic evolution of the united Xing’an–Erguna and Songnen–Zhangguangcai Range massifs of Northeast China: (1) early Carboniferous (360–340 Ma) subduction of the Paleo-Asian oceanic plate beneath the united Xing’an–Erguna Massif and formation of the Wunuer oceanic basin in the Yakeshi area; (2) early to late Carboniferous (340–310 Ma) sustained subduction of the Paleo-Asian oceanic plate beneath the united Xing’an–Erguna Massif and initiation of subduction of the Wunuer oceanic basin; and (3) late Carboniferous–early Permian (310–275 Ma) syn-collisional to post-collisional tectonic transition between the united Xing’an–Erguna Massif and the Songnen–Zhangguangcai Range Massif.

BackgroundBacterial vaginosis (BV) is the most common microbiological syndrome in women of childbearing age, causing numerous adverse health issues in pregnant women. Several observational studies have discussed the association between vitamin D deficiency and the risk of BV during pregnancy, but the results were inconclusive. Therefore, this meta-analysis aimed to explore the association between vitamin D deficiency and BV risk in pregnant women.Materials and methodsWe searched four databases, including PubMed, Embase, Cochrane Library, and Web of Science, from their inception to July 2022. Pooled odds ratios (OR) with corresponding 95% confidence intervals (CI) were estimated using random effects models. Additionally, we conducted subgroup analyses to identify the potential sources of between-study heterogeneity. Sensitivity analysis was performed using the method of exclusion, one study at a time. Publication bias was examined using Egger’s test and funnel plot.ResultsA total of 14 studies from 13 articles including 4,793 participants were eligible for this meta-analysis. The outcome showed that vitamin D deficiency may increase the risk of BV during pregnancy by 54% (OR, 1.54; 95% CI, 1.25–1.91; P < 0.001). In subgroup analyses, positive associations were also found in studies that were: conducted in black women (OR, 1.56; 95% CI, 0.98–2.48; P = 0.060), focused on the first trimester of pregnancy (OR, 2.22; 95% CI, 1.35–3.64; P = 0.002), of high quality (OR, 3.05; 95% CI, 1.26–7.41; P = 0.014), and adjusted for confounders (OR, 1.28; 95% CI, 1.06–1.55; P = 0.012). Sensitivity analysis reported that BV risk during pregnancy resulting from vitamin D deficiency increased by 157% (OR, 2.57; 95% CI, 1.50–4.42; P = 0.001) when removing the first two high-weight studies. Publication bias was observed using Egger’s test (t = 3.43, P = 0.005) and a visual funnel plot.ConclusionThis meta-analysis showed that vitamin D deficiency is positively associated with the risk of BV during pregnancy. Further high-quality prospective cohort studies are needed to determine whether vitamin D intake reduces the prevalence of BV in pregnant women.

Abstract With the development of lipidomics, the significance of lipid metabolism in various lung diseases has been noticed recently. Among them, cholesterol metabolism plays a crucial role in the pathological process of chronic obstructive pulmonary diseases (COPD). Cholesterol accumulation could lead to mitochondrial dysfunction, such as impaired respiration, imbalanced dynamics and decreased adenosine triphosphate production, while mitochondrial dysfunction, especially altered mitochondrial dynamics, could also disrupt lipid homeostasis in cells. This review focuses on cholesterol metabolism and mitochondrial function in COPD and discusses the potential mechanisms of the steroidogenic acute regulatory‐related lipid transfer domain family (STARD), especially STARD3. The review provides new insights for understanding the pathological process between STARD and mitochondria and new therapeutic targets in COPD.

Abstract Background Respiratory fungal exposure is known to be associated with various allergic pulmonary disorders. Eosinophils have been implicated in tissue homeostasis of allergic inflammation as both destructive effector cells and immune regulators. What contributions eosinophils have in Aspergillus fumigatus (Af)-induced allergic lung inflammation is worthy of investigating. Methods We established the Af-exposed animal asthmatic model using eosinophil-deficient mice, ∆dblGATA1 mice. Airway inflammation was assessed by histopathological examination and total cell count of bronchoalveolar lavage fluid (BALF). The protein level in BALF and lung mRNA level of type 2 cytokines IL-4, IL-5, and IL-13 were detected by ELISA and qRT-PCR. We further studied the involvement of endoplasmic reticulum (ER) stress, apoptosis, and autophagy by western blots, qRT-PCR, immunofluorescence, TUNEL, or immunohistochemistry. RNA-Seq analysis was utilized to analyze the whole transcriptome of Af-exposed ∆dblGATA1 mice. Results Hematoxylin and eosin (HE) staining and periodic acid–Schiff staining (PAS) showed that airway inflammation and mucus production were alleviated in Af-challenged ∆dblGATA1 mice compared with wild-type controls. The protein and mRNA expressions of IL-4, IL-5, and IL-13 were reduced in the BALF and lung tissues in Af-exposed ∆dblGATA1 mice. The results demonstrated that the significantly increased ER stress markers (GRP78 and CHOP) and apoptosis executioner caspase proteases (cleaved caspase-3 and cleaved caspase-7) in Af-exposed wild-type mice were all downregulated remarkably in the lungs of ∆dblGATA1 mice with Af challenge. In addition, the lung autophagy in Af-exposed ∆dblGATA1 mice was found elevated partially, manifesting as higher expression of LC3-II/LC3-I and beclin1, lower p62, and downregulated Akt/mTOR pathway compared with Af-exposed wild-type mice. Additionally, lung RNA-seq analysis of Af-exposed ∆dblGATA1 mice showed that biological processes about chemotaxis of lymphocytes, neutrophils, or eosinophils were enriched but without statistical significance. Conclusions In summary, eosinophils play an essential role in the pathogenesis of Af-exposed allergic lung inflammation, whose deficiency may have relation to the attenuation of type 2 immune response, alleviation of ER stress and apoptosis, and increase of autophagy. These findings suggest that anti-eosinophils therapy may provide a promising direction for fungal-induced allergic pulmonary diseases.

Abstract Background Hypercholesterolemia is found in patients with chronic lung inflammation, during which airway epithelial cells play important roles in maintenance of inflammatory responses to pathogens. The present study aims at molecular mechanisms by which cholesterol changes airway epithelial sensitivity in response to smoking. Methods Human bronchial epithelial cells (HBEs) were stimulated with cigarette smoke extract (CSE) and mice were exposed to CS/lipopolysaccharide (LPS) as models in vitro and in vivo. Severe COPD patients and healthy volunteers were also enrolled and the level of cholesterol in plasma was detected by metabolomics. Filipin III and elisa kits were used to stain free cholesterol. Mitochondrial function was detected by mitotracker green, mitotracker green, and Seahorse. Mitochondrial morphology was detected by high content screening and electron microscopy. The mRNA and protein levels of mitochondrial dynamics‐related proteins were detected by RT‐qPCR and Western blot,respectively. BODIPY 493/503 was used to stain lipid droplets. Lipidomics was used to detect intracellular lipid components. The mRNA level of interleukin (IL)‐6 and IL‐8 were detected by RT‐qPCR. Results We found that the cholesterol overload was associated with chronic obstructive pulmonary disease (COPD) and airway epithelia‐driven inflammation, evidenced by hypercholesterolemia in patients with COPD and preclinical models, alteration of lipid metabolism‐associated genes in CSE‐induced airway epithelia and production of ILs. External cholesterol altered airway epithelial sensitivity of inflammation in response to CSE, through the regulation of STARD3‐MFN2 pathway, cholesterol re‐distribution, altered transport and accumulation of cholesterol, activities of lipid transport regulators and disorder of mitochondrial function and dynamics. MFN2 down‐regulation increased airway epithelial sensitivity and production of ILs after smoking, at least partially by injuring fatty acid oxidation and activating mTOR phosphorylation. Conclusions Our data provide new insights for understanding molecular mechanisms of cholesterol‐altered airway epithelial inflammation and for developing diagnostic biomarkers and therapeutic targets to improve patient outcomes.

The biology major has developed rapidly in recent years. Biology is a science that penetrates every aspect of human life and is one of the core majors in most agricultural colleges and universities. However, many teachers lack practical experience in the subject. To overcome this problem, in recent years, we have been trying to introduce new reforms into our teaching. This article provides some insight into the way that biology majors have been reformed, which will help educators in agricultural colleges and universities. At present, teachers implement the “Industrial Innovation and Entrepreneurship Talent Cultivation” (IIETC) model, but it is not clear whether this helps biology majors to master the course and improve their practical skills. In this study, the IIETC model is outlined, and the academic achievement and satisfaction of students taught under the IIETC model are assessed. A T-test is used to examine potential differences between IIETC and traditional teaching models. In-depth interviews and questionnaires were given to two groups of students who followed different teaching models as part of an exploratory study. The aim was to explore how effective IIETC is at helping biology majors master the course and improve students’ wellbeing. Our results show that compared with traditional teaching methods, the IIETC model has a significant positive impact on the academic performance and happiness of biology students. Students trained under the IIETC model were more active and scored more highly in their final exams. They were more likely to feel that they had achieved success and happiness through the course (P = 0.03). The outcomes of this research reveal a novel teaching reform that improved students’ enthusiasm for innovation and entrepreneurship during the ongoing COVID-19 pandemic. The effects are very encouraging and deserve further exploration and expansion in future work.

In the last 20 years, accumulating evidence indicates that the gut microbiota contribute to the development, maturation, and regulation of the host immune system and mediate host anti-pathogen defenses. Lactobacillus casei (L.casei) is a normal flora of the gastrointestinal tract in mammals and, as a great mucosal delivery vehicle, has wide use in bioengineering. However, the diarrhea prevention role of commensal intestinal microbiota interfered by the recombinant L.casei (rL.casei) in newborn piglets is not well understood. In our study, newborn piglets orally fed with the rL.casei surface displayed the fimbrial protein K88 of enterotoxigenic Escherichia coli (ETEC) and their feces were collected for a period of time after feeding. The next-generation sequencing of these fecal samples showed that the relative abundance of L.casei was significantly increased. The oral administration of rL.casei altered the intestinal microbial community as evidenced by altered microbial diversity and microbial taxonomic composition. Remarkably, the functional enhancing of the intestinal bacterial community by rL.casei was positively correlated with membrane transport, replication, and repair (p < 0.05). The specific antibody detection indicates that high levels of anti-K88 secretory immunoglobulin A (sIgA) were induced in fecal samples and systemic immunoglobulin G was produced in serum. The diarrhea rate in piglets caused by ETEC K88 was decreased by about 24%. Thus, the oral administration of rL.casei not only activated the mucosal and humoral immune responses in vivo but also contributed to shape the intestinal probiotics in newborn piglets and to significantly reduce the diarrhea rates of newborn piglets.

Abstract Background Chronic obstructive pulmonary disease (COPD) is characterized by inflammation, airflow obstruction, and lung damage resulting from multiple causes, in particular the exposure to cigarette smoke. Growing evidence supports an important role for lipid metabolism in the development of COPD, where ferroptosis plays a critical role in lipid metabolism and cholesterol homeostasis. The aim of the present study was to investigate the role of cigarette smoke extract (CSE) in the induction and formation of ferroptosis and its potential relation with intracellular cholesterol accumulation. Methods Filipin III staining and cholesterol content assay kit were used to detect intracellular cholesterol level. BODIPY C11 581/591 and malondialdehyde (MDA) assay kit were used to detect lipid perioxidation. Quantitative RT‐PCR was used to detect the mRNA level of ferroptosis‐related and cholesterol‐related genes. The protein expression of phosphatidylinositol 3‐kinase (PI3K) signaling pathway and related proteins were analyzed by Western blot. Results We found that CSE and exogenous cholesterol induced ferroptosis, while the ferroptosis agonist increased intracellular cholesterol levels. The inhibition of ferroptosis activated the cholesterol efflux and synthesis. We also found regulatory roles of PI3K/AKT pathway, especially the role of p85α, in the development of ferroptosis. Conclusions CSE‐induced ferroptosis in human bronchial epithelia changed intracellular cholesterol homeostasis. The interaction between cholesterol metabolism and ferroptosis may play an important role in the development of COPD.

Abstract During the 2020 Spring Festival in China, the outbreak of a novel coronavirus, named COVID‐19 by WHO, brought on a worldwide panic. According to the clinical data of infected patients, radiologic evidence of lung edema is common and deserves clinical attention. Lung edema is a manifestation of acute lung injury (ALI) and may progress to hypoxemia and potentially acute respiratory distress syndrome (ARDS). Patients diagnosed with ARDS have poorer prognosis and potentially higher mortality. Although no effective treatment is formally approved for COVID‐19 infection, support of ventilation with oxygen therapy and sometimes mechanical ventilation is often required. Treatment with systemic and/or local glucocorticoids might be helpful to alleviate the pulmonary inflammation and edema, which may decrease the development and/or consequences of ARDS. In this article, we focus on the lung edema and ALI of patients with this widely transmitted COVID‐19 infection in order to provide clinical indications and potential therapeutic targets for clinicians and researchers.

Abstract Methodologies of genome editing are rapidly developing with the improvement of gene science and technology, mechanism‐based understanding, and urgent needs. In addition to the specificity and efficiency of on‐target sites, one of the most important issues is to find and avoid off‐targets before clinical application of gene editing as a therapy. Various algorithms, modified nucleases, and delivery vectors are developed to localize and minimize off‐target sites. The present review aimed to clarify off‐targets of various genome editing and explore potentials of clinical application by understanding structures, mechanisms, clinical applications, and off‐target activities of genome editing systems, including CRISPR/Cas9, CRISPR/Cas12a, zinc finger nucleases, transcription activator‐like effector nucleases, meganucleases, and recent developments. Current genome editing in cancer therapy mainly targeted immune systems in tumor microenvironment by ex vivo modification of the immune cells in phases I/II of clinical trials. We believe that genome editing will be the critical part of clinical precision medicine strategy and multidisciplinary therapy strategy by integrating gene sequencing, clinical transomics, and single cell biomedicine. There is an urgent need to develop on/off‐target‐specific biomarkers to monitor the efficacy and side‐effects of gene therapy. Thus, the genome editing will be an alternative of clinical therapies for cancer with the rapid development of methodology and an important part of clinical precision medicine strategy.

Abstract The main factor affecting the gas content in a coal seam is the geological structure, and different types of geological structure have different effects on the gas content in a coal seam. To explore the main controlling factors of gas content in coal seam No. 3 in the Changping minefield, and the effect of different geological structures on gas control in a coal seam, this study analyzes the types and quantities of geological structures in the Changping minefield. Further, geological boreholes are used to sample and analyze coal seams near different geological structures and at different burial depths. The results show that the main controlling factor of gas content in a coal seam is the burial depth of the coal seam, and the gas content tends to increase with the increasing depth. A geological structure has an effect on the occurrence of coal seam gas in a local area, and the gas content in the coal seam near a fault is 1.6 times that in the normal areas at the same depth. The gas content in the upper wall of a normal fault is 21.6% higher than that in the lower wall. The syncline and anticline axes are prone to form gas accumulation areas, where the gas content is 30%‐60% higher than the normal level. Gas accumulation is prone to occur around a collapse column, and the gas content there is 2.47 times higher than that in a normal area at the same burial depth. Combining the formation mechanism and structural characteristics of different geological structures, this study analyzes the effect of geological structures on coal seam gas quantitatively and provides a theoretical basis for the prevention and control of gas disasters in the Changping minefield.

Abstract The communication between endoplasmic reticulum (ER) and mitochondria (Mt) plays important roles in maintenance of intra‐ and extra‐cellular microenvironment, metabolisms, signaling activities and cell‐cell communication. The present review aims to overview the advanced understanding about roles of ER‐Mt structural contacts, molecular interactions and chemical exchanges, signal transmissions and inter‐organelle regulations in ER‐Mt communication. We address how the ER‐Mt communication contributes to the regulation of lipid, amino acid and glucose metabolisms by enzymes, transporters and regulators in the process of biosynthesis. We specially emphasize the importance of deep understanding about molecular mechanisms of ER‐Mt communication for identification and development of biology‐specific, disease‐specific and metabolism‐specific biomarkers and therapeutic targets for human diseases. The inhibitors and modulators of the ER‐Mt communication are categorized according to therapeutic targets. Rapid development of biotechnologies will provide new insights for spatiotemporally understanding the molecular mechanisms of ER‐Mt communication.

Abstract Increasing evidence supports a central role of the immune system in lung diseases. Understanding how immunological alterations between lung diseases provide opportunities for immunotherapy. Exhausted T cells play a key role of immune suppression in lung cancer and chronic obstructive pulmonary disease was proved in our previous study. The present study aims to furthermore define molecular landscapes and heterogeneity of systemic immune cell target proteomic and transcriptomic profiles and interactions between circulating immune cells and lung residential cells in various lung diseases. We firstly measured target proteomic profiles of circulating immune cells from healthy volunteers and patients with stable pneumonia, stable asthma, acute asthma, acute exacerbation of chronic obstructive pulmonary disease, chronic obstructive pulmonary disease and lung cancer, using single‐cell analysis by cytometry by time‐of‐flight with 42 antibodies. The nine immune cells landscape was mapped among those respiratory system diseases, including CD4+ T cells, CD8+ T cells, dendritic cells, B cells, eosinophil, γδT cells, monocytes, neutrophil and natural killer cells. The double‐negative T cells and exhausted CD4+ central memory T cells subset were identified in patients with acute pneumonia. This T subset expressed higher levels of T‐cell immunoglobulin and mucin domain‐containing protein 3 (Tim3) and T‐cell immunoreceptor with Ig and ITIM domains (TIGIT) in patients with acute pneumonia and stable pneumonia. Biological processes and pathways of immune cells including immune response activation, regulation of cell cycle and pathways in cancer in peripheral blood immune cells were defined by bulk RNA sequencing (RNA‐seq). The heterogeneity among immune cells including CD4+, CD8+ T cells and NK T cells by single immune cell RNA‐seq with significant difference was found by single‐cell sequencing. The effect of interstitial telocytes on the immune cell types and immune function was finally studied and the expressions of CD8a and chemokine C–C motif receptor 7 (CCR7) were increased significantly in co‐cultured groups. Our data indicate that proteomic and transcriptomic profiles and heterogeneity of circulating immune cells provides new insights for understanding new molecular mechanisms of immune cell function, interaction and modulation as a source to identify and develop biomarkers and targets for lung diseases.

Orchids have attracted much attention from researchers, because of their richness of species and their great ornamental and medicinal value. Jiangxi Province, which is located in southeastern China and ringed on three sides by mountains, contains many nature reserves and harbors large number of orchids. Here, we conducted field surveys of orchids in 35 nature reserves in Jiangxi, using sampling lines and plots. We also analyzed the relationship between orchid richness and density with environmental variables and studied the relationship among these nature reserves. We found that the mountainous areas of southwestern, southern, and northeastern Jiangxi have a high richness and density of orchids, while the mountainous areas of central and northwestern Jiangxi have low richness and density. Jiulianshan and Jinggangshan are the two most rich-species reserves, with 58 and 55 orchids, respectively. Eight reserves (22% of those surveyed) had fewer than 10 orchids. Compared with soil, climate, and vegetation, topography was more closely related to the richness and density of orchids. Topographical variables explained 19% and 20% of the total variation in SR and SD, respectively. The result of hierarchical clustering analysis showed that the 35 nature reserves of Jiangxi obviously fall into two main clusters, which are separated by the Ganjiang River–Poyang Lake water system. In conclusion, the geographical patterns of richness and the density of orchids in Jiangxi are uneven and are affected by topography and vegetation, while their distribution is affected by the terrain of Jiangxi. Our work explains the richness and density patterns and the assembly mechanism of the orchids in Jiangxi and also provides new ideas for the protection of orchids in this region.

Abstract Mitochondrial dysfunction contributes to the imbalance of cellular homeostasis and the development of diseases, which is regulated by mitochondria‐associated factors. The present review aims to explore the process of the mitochondrial quality control system as a new source of the potential diagnostic biomarkers and/or therapeutic targets for diseases, including mitophagy, mitochondrial dynamics, interactions between mitochondria and other organelles (lipid droplets, endoplasmic reticulum, endosomes, and lysosomes), as well as the regulation and posttranscriptional modifications of mitochondrial DNA/RNA (mtDNA/mtRNA). The direct and indirect influencing factors were especially illustrated in understanding the interactions among regulators of mitochondrial dynamics. In addition, mtDNA/mtRNAs and proteomic profiles of mitochondria in various lung diseases were also discussed as an example. Thus, alternations of mitochondria‐associated regulators can be a new category of biomarkers and targets for disease diagnosis and therapy.

Abstract The changes associated with the cutting roof and release pressure mining method in the three zones that are susceptible to spontaneous combustion in the goaf with “Y” type ventilation system were studied. For the investigated underground coal mine working face, its three zones of spontaneous combustion have a “U” type ventilation system for the traditional mining method and a “Y” type ventilation system for the cutting roof and release pressure mining method. These systems were monitored using a beam tube method, and the evolution characteristics of spontaneous combustion of the three zones was analysed. The monitoring data showed that when using a traditional mining method with a “U” type ventilation system, the scattered tropical zone converts into the oxidation heating zone after 68.5 m in the goaf, and the suffocative zone presents after 85.5 m. For the cutting roof and release pressure mining method together with a “Y” type ventilation system, the scattered tropical zone converts into the oxidation heating zone after 84 m in the goaf, and the suffocative zone appears after 198.8 m. Compared to traditional mining, the width of the oxidation heating zone increases when using the cutting roof and release pressure method. To prevent spontaneous combustion of the goaf, the comprehensive techniques of the pressure balance for fire control, ground fissure sealing, working face sealing, retained roadways guniting and inert gas fire extinguishing have been used in the studied coal mine. The CO concentration of the goaf is less than 300 ppm during the mining process. Safe mining of the tested working face shows that the cutting roof and release pressure technique can be applied to control the coal seam spontaneous combustion.

Abstract Background Pulmonary cryptococcosis is a common fungal infection frequently seen in immunocompromised patients. Owing to its nonspecific clinical and radiographic features, the differential diagnosis with secondary tuberculosis, malignant tumor, and bacterial pneumonia is sometimes difficult. Many case reports have focused on misdiagnosis of pulmonary cryptococcosis as a malignant tumor. But to the best of our knowledge, the coexistence of pulmonary cryptococcosis and malignant tumor is rarely presented. Case presentation A 52-year-old immunocompetent Han Chinese woman was presented to our emergency department complaining of headache and vomiting accompanied by postural changes. She was diagnosed with pulmonary cryptococcosis according to results of laboratory tests, computed tomography, and percutaneous lung biopsy. Owing to the poor therapeutic effects of 6-month fluconazole treatment, she underwent a second percutaneous lung biopsy and was diagnosed with pulmonary cryptococcosis coexisting with adenocarcinoma. Delayed treatment of malignant tumor resulted in lymph node metastasis, higher pathologic stage, and probably poorer prognosis. Conclusions Our patient’s case serves as a reminder not to misdiagnose pulmonary cryptococcosis coexisting with adenocarcinoma.

Abstract Background SOCS3 (suppressor of cytokine signaling 3) is a negative regulator of JAK/STAT3 signaling pathway and participates in the regulation of lung inflammation in a mouse model with acute lung injury (ALI). However, it is not well understood how SOCS3 regulates lung inflammation in the ALI mouse model. Method In the present study, we investigated the effects of SOCS3 on modulation of Ly6C(+) monocyte phenotypes in a mouse model with lipopolysaccharide (LPS)-induced ALI. Conditional SOCS3(Lyz2cre) mice with myeloid cell-restricted depletion of SOCS3 gene were created by breeding transgenic Lyz2Cre mice with SOCS3(fl/fl) mice. Wilde-type (WT) and SOCS3(Lyz2cre) mice were intratracheal instilled with 5 mg/kg LPS for 2 days. Lung, bronchoalveolar lavage (BAL) and blood were collected for analysis by flow cytometry, ELISA, qRT-PCR and Western blot analysis. Results The studies in the ALI mouse model revealed that myeloid cell-restricted SOCS3 deficiency exacerbated the severity of ALI as compared to the WT mice. The increased severity of ALI in SOCS3-deficient mice was associated with higher populations of neutrophils, T lymphocytes and Ly6C(+) monocytes in the inflamed lung tissues. In addition, CCR2 and CXCL15 were elevated, and accompanied by greater expression and activation of STAT3 in the lung of SOCS3-deficient mice. SOCS3-deficient bone marrow-derived macrophages (BMDMs) expressed a higher amount of TNF-alpha, and adoptive transfer of the SOCS3-deficient Ly6C(+) BMDMs into WT mice enhanced the severity of ALI than adoptive transfer of WT control BMDMs. However, depletion of Ly6C(+) circulating monocytes by anti-Ly6C(+) neutralizing antibody moderately attenuated neutrophil infiltration and resulted in lower prevalence of Ly6C(+) cells in the lung of treated mice. Conclusion Myeloid cell-restricted lack of SOCS3 induced more severe ALI through modulation of Ly6C(+) subtype macrophages. The results provide insight into a new role of SOCS3 in modulation of Ly6C(+) monocyte phenotypes and provide a novel therapeutic strategy for ALI by molecular intervention of macrophages subtypes.

Frequency selective surface (FSS) has been extensively studied due to its potential applications in radomes, antenna reflectors, high-impedance surfaces and absorbers. Recently, a new principle of designing FSS has been proposed and mainly studied in two levels. In the level of materials, dielectric materials instead of metallic patterns are capable of achieving more functional performance in FSS design. Moreover, FSSs made of dielectric materials can be used in different extreme environments, depending on their electrical, thermal or mechanical properties. In the level of design principle, the theory of metamaterial can be used to design FSS in a convenient and concise way. In this review paper, we provide a brief summary about the recent progress in all-dielectric metamaterial frequency selective surface (ADM-FSS). The basic principle of designing ADM-FSS is summarized. As significant tools, Mie theory and dielectric resonator (DR) theory are given which illustrate clearly how they are used in the FSS design. Then, several design cases including dielectric particle-based ADM-FSS and dielectric network-based ADM-FSS are introduced and reviewed. After a discussion of these two types of ADM-FSSs, we reviewed the existing fabrication techniques that are used in building the experiment samples. Finally, issues and challenges regarding the rapid fabrication techniques and further development aspects are discussed.

In this paper, we demonstrate a method of designing all-dielectric metamaterial frequency selective surface (FSS) with ceramic resonators in spatial arrangement. Compared with the traditional way, spatial arrangement provides a flexible way to handle the permutation and combination of different ceramic resonators. With this method, the resonance response can be adjusted easily to achieve pass/stop band effects. As an example, a stop band spatial arrangement all-dielectric metamaterial FSS is designed. Its working band is in 11.65–12.23GHz. By adjusting permittivity and geometrical parameters of ceramic resonators, we can easily modulate the resonances, band pass or band stop characteristic, as well as the working band.

In this paper, we designed a metamaterial absorber performed in microwave frequency band. This absorber is composed of E-shaped dielectrics which are arranged along different directions. The E-shaped all-dielectric structure is made of microwave ceramics with high permittivity and low loss. Within about 1 GHz frequency band, more than 86% absorption efficiency was observed for this metamaterial absorber. This absorber is polarization insensitive and is stable for incident angles. It is figured out that the polarization insensitive absorption is caused by the nearly located varied resonant modes which are excited by the E-shaped all-dielectric resonators with the same size but in the different direction. The E-shaped dielectric absorber contains intensive resonant points. Our research work paves a way for designing all-dielectric absorber.

In this paper, we propose a band enhanced metamaterial absorber in microwave band, which is composed of high-permittivity E-shaped dielectric resonators and metallic ground plate. The E-shaped all-dielectric structure is made of high-temperature microwave ceramics with high permittivity and low loss. An absorption band with 1 GHz bandwidth for both TE and TM polarizations are observed. Moreover, the absorption property is stable under different incident angles. The band enhanced absorption is caused by different resonant modes which lie closely in the absorption band. Due to the enhanced localized electric/magnetic fields at the resonant frequencies, strong absorptions are produced. Our work provides a new method of designing high-temperature and high-power microwave absorbers with band enhanced absorption.

Abstract Magnons and phonons are engineered in artificial lattices with tunable modes and band dispersions. Recent advance in magnon-phonon coupling shined a light on combining magnonic and phononic crystals as hybrid magnon-phonon crystals, benefit from the tunable magnon-phonon coupling, the time-reversal symmetry breaking of magnons, and the long lifetime of phonons. This perspective summarizes lattice-based mutual control of magnons and phonons, and proposes the opportunities provided by the hybrid magnon-phonon crystals.

Abstract Antiferromagnets are promising for nano-scale oscillator in a wide frequency range from gigahertz up to terahertz. Experimentally realizing antiferromagnetic moment oscillation via spin-orbit torque, however, remains elusive. Here, we demonstrate that the optical spin-orbit torque induced by circularly polarized laser can be used to drive free decaying oscillations with a frequency of 2 THz in metallic antiferromagnetic Mn2Au thin films. Due to the local inversion symmetry breaking of Mn2Au, ultrafast a.c. current is generated via spin-to-charge conversion, which can be detected through free-space terahertz emission. Both antiferromagnetic moments switching experiments and dynamics analyses unravel the antiferromagnetic moments, driven by optical spin-orbit torque, deviate from its equilibrium position, and oscillate back in 5 ps once optical spin-orbit torque is removed. Besides the fundamental significance, our finding opens a new route towards low-dissipation and controllable antiferromagnet-based spin-torque oscillators.

Abstract Although computerized cognitive training (CCT) is an effective digital intervention for cognitive impairment, its dose-response relationship is understudied. This retrospective cohort study explores the association between training dose and cognitive improvement to find the optimal CCT dose. From 2017 to 2022, 8,709 participants with subjective cognitive decline, mild cognitive impairment, and mild dementia were analyzed. CCT exposure varied in daily dose and frequency, with cognitive improvement measured weekly using Cognitive Index. A mixed-effects model revealed significant Cognitive Index increases across most dose groups before reaching the optimal dose. For participants under 60 years, the optimal dose was 25 to

Abstract Colonial cities in Asia and Africa, developed under the dual influence of native and sovereign cultures during the colonial era, exhibit unique urban morphological characteristics and reflect the planning ideologies of diverse cultures. Thus, the morphological study of colonial cities in Asia and Africa is crucial for understanding and identifying the historical and cultural value of these cities, which in turn enables the formulation of precise conservation strategies. In light of this, the study develops an urban morphological analysis methodology comprising "Colonial Background Analysis—Morphological Characteristics Analysis—Driving Factor Extraction" based on the Conzenian Approach, which is used to examine the morphology of 21 typical colonial cities in Asia and Africa across various colonial periods. As a result, "Dual-City" emerges as the core spatial characteristic of colonial cities in Asia and Africa, reflecting the morphological differences in road layouts, urban fabric, and urban cores between native and European zones. Additionally, as native settlement sizes, social structures, and cultural identities differ between cities, three major spatial patterns of the "Dual-City" model can be identified: separated, mediated, and integrated spatial patterns, resulting from the varied planning approaches applied by colonizers. Furthermore, suggestions for the conservation of colonial heritage are proposed based on the driving factors analysis from the morphological study.
