Abstract Based on the ultrasonic welding technology, this study uses three different welding widths to weld copper cables with different specifications. The influence of welding width on the mechanical properties and microstructure of each group of welded joints was systematically studied for the first time. The thermal shock test was carried out for each group of welded joints under optimum welding width to simulate the influence of severe temperature change environment on joint performance. It is found that the cross-sectional area of joint is 20 mm2 and optimal welding width of joint composed of two and three cables is 7 mm. The optimal welding temperature of the joint composed of four cables is 5 mm. Under the optimal welding width, the average shear strength of two-cable joint reaches 309.4 N. The four-cable joint is only 232.2 N. Moreover, the welding strength weakens significantly as the number of cables and the peak temperature decreases. The high temperature of bonding interface is the key factor to form a good weld. The peak temperature during welding is negatively correlated with the porosity of joint and positively correlated with peeling strength of joint. In addition, the morphology of ultrasonically welded joints has changed obviously after thermal shock test. With the participation of oxygen, the surface of welded joint is gray and bright brass, while the interior of joint is purple due to lack of oxygen. Moreover, the phenomenon of atomic diffusion and thermal expansion generates joints which were initially in a mechanically interlocked form and welding interface of the metallurgical bond under the action of high temperature. So the maximum joint peel strength is slightly improved.

The status of coinfection during the national outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Omicron BA.5.2 or BF.7 in China in the winter of 2022, which is suspected to contribute substantially to the overloaded severe cases, needs to be investigated. We analyzed the coinfection status of 385 severe patients infected with the Omicron variant in Guangzhou using metagenomic sequencing. We found that 317 (82.3 %) patients were coinfected with at least one additional pathogen(s), including bacteria (58.7 %), fungi (27.1 %) and viruses (73.5 %). Pseudomonas aeruginosa (P. aeruginosa) (24.2 %), Staphylococcus aureus (S. aureus) (14.0 %), and Klebsiella pneumoniae (K. pneumonia) (13.4 %) ranked as the top three coinfected bacteria. Aspergillus fumigatus (A. fumigatus) (39.5 %), Pneumocystis jirovecii (P. jirovecii) (24.4 %) and Canidia albicans (C. albicans) (22.1 %) were the top three coinfected fungi. Epstein-Barr virus (EBV) (63.1 %), Human herpesvirus 7 (HHV-7) (34.8 %), and Herpes simplex virus 1 (HSV-1) (32.6 %) were the top three coinfected viruses. Of note, the detection of multiple coinfections of potential pathogenic bacteria, fungi, and viruses, despite lacking consistent patterns, highlighted a complicated synergistic contribution to disease severity. Our study presents the most comprehensive spectrum of bacterial, fungal, and viral coinfections in Omicron-associated severe coronavirus disease 2019 (COVID-19), implying that the coinfection of conditional pathogens might synergistically deteriorate the Omicron infection outcomes.

Abstract Objective To investigate whether the ultrasound microcystic pattern (MCP) can accurately predict borderline ovarian tumors (BOTs). Methods A retrospective collection of 393 patients who met the inclusion criteria was used as the study population. Indicators that could well identify BOT in different pathological types of tumors were derived by multivariate unordered logistic regression analysis. Finally, the correlation between ultrasound MCP and pathological features was analyzed. Results (1) MCP was present in 55 of 393 ovarian tumors, including 34 BOTs (34/68, 50.0%), 16 malignant tumors (16/88, 18.2%), and 5 benign tumors (5/237, 2.1%). (2) Univariate screening showed significant differences (P 10 cyst locules, a solid component, blood flow, and MCP among BOTs, benign ovarian tumors, and malignant ovarian tumors. With the intelligent development of vehicles, the number of vehicles equipped with millimeter-wave (mmWave) radars is increasing, and the possibility of interference between radars is rising dramatically. In automatic driving, it will be common for target detection to be affected by multiple interfering radars. Addressing the mutual interference challenges, an adaptive interference detection method based on support vector machines (SVMs) is proposed. First, a window selection is performed on the received signal and features describing the difference between the normal signal and the interference are extracted. Then, we use a nonlinear SVM to distinguish between the interference and the normal signal. After completing the localization of the interference, we use an autoregressive (AR) prediction model to reconstruct the target echo signal. Results from both multiple interference simulation scenarios and real experimental scenarios show that the accuracy of interference localization and the effect of interference mitigation of the proposed method outperforms the mainstream methods.

Abstract High strength aluminum alloys are widely used but their strength is reduced as nano-precipitates coarsen rapidly in medium and high temperatures, which greatly limits their application. Single solute segregation layers at precipitate/matrix interfaces are not satisfactory in stabilizing precipitates. Here we obtain multiple interface structures in an Al-Cu-Mg-Ag-Si-Sc alloy including Sc segregation layers, C and L phases as well as a newly discovered χ-AgMg phase, which partially cover the θ′ precipitates. By atomic resolution characterizations and ab initio calculations, such interface structures have been confirmed to synergistically retard coarsening of precipitates. Therefore, the designed alloy shows the good combination of heat resistance and strength among all series of Al alloys, with 97% yield strength retained after thermal exposure, which is as high as 400 MPa. This concept of covering precipitates with multiple interface phases and segregation layers provides an effective strategy for designing other heat resistant materials.

Abstract Background Tibetans are genetically adapted to high-altitude environments. Though many studies have been conducted, the genetic basis of the adaptation remains elusive due to the poor reproducibility for detecting selective signatures in the Tibetan genomes. Results Here, we present whole-genome sequencing (WGS) data of 1001 indigenous Tibetans, covering the major populated areas of the Qinghai–Tibetan Plateau in China. We identify 35 million variants, and more than one-third of them are novel variants. Utilizing the large-scale WGS data, we construct a comprehensive map of allele frequency and linkage disequilibrium and provide a population-specific genome reference panel, referred to as 1KTGP. Moreover, with the use of a combined approach, we redefine the signatures of Darwinian-positive selection in the Tibetan genomes, and we characterize a high-confidence list of 4320 variants and 192 genes that have undergone selection in Tibetans. In particular, we discover four new genes, TMEM132C, ATP13A3, SANBR, and KHDRBS2, with strong signals of selection, and they may account for the adaptation of cardio-pulmonary functions in Tibetans. Functional annotation and enrichment analysis indicate that the 192 genes with selective signatures are likely involved in multiple organs and physiological systems, suggesting polygenic and pleiotropic effects. Conclusions Overall, the large-scale Tibetan WGS data and the identified adaptive variants/genes can serve as a valuable resource for future genetic and medical studies of high-altitude populations.

BackgroundTalaromycosis is one of the most common opportunistic infections in human immunodeficiency virus (HIV) infected patients. However, few researches have explored the prevalence in Southern China and fully assessed the value of the Mp1p antigen screening for the diagnosis of talaromycosis.Methodology/principal findingsWe performed a cross-sectional study of HIV-infected antiretroviral therapy (ART)-naïve adult patients who were seen in 2018 at Guangzhou Eighth People's Hospital, Guangzhou Medical University. Serum samples collected from all the 784 enrolled patients were tested for Mp1p antigen using double-antibody sandwich enzyme-linked immunosorbent assay. A culture of pathogen was conducted in 350 clinically suspected patients to confirm talaromycosis. The overall prevalence of talaromycosis based on the Mp1p antigen detection was 11.4% (89/784) and peaked at 32.2% (75/233) in patients with CD4+ ≤50 Nr/μl. Logistic regression analysis found Mp1p antigen positive rate decreased with the increase in CD4+ counts (OR 0.982, 95% CI 0.977-0.987, PConclusions/significanceMp1p antigen screening can be an effective tool for more efficient diagnosis of Talaromycosis, especially in HIV/AIDS patients with low CD4+ counts. Future validation studies are needed.

Abstract Objectives Idiopathic inflammatory myopathies (IIM) are a class of autoimmune diseases with high heterogeneity that can be divided into different subtypes based on clinical manifestations and myositis‐specific autoantibodies (MSAs). However, even in each IIM subtype, the clinical symptoms and prognoses of patients are very different. Thus, the identification of more potential biomarkers associated with IIM classification, clinical symptoms, and prognosis is urgently needed. Methods Plasma and urine samples from 79 newly diagnosed IIM patients (mean disease duration 4 months) and 52 normal control (NC) samples were analysed by high‐performance liquid chromatography of quadrupole time‐of‐flight mass spectrometry (HPLC‐Q‐TOF‐MS)/MS‐based untargeted metabolomics. Orthogonal partial least‐squares discriminate analysis (OPLS‐DA) were performed to measure the significance of metabolites. Pathway enrichment analysis was conducted based on the KEGG human metabolic pathways. Ten machine learning (ML) algorithms [linear support vector machine (SVM), radial basis function SVM, random forest, nearest neighbour, Gaussian processes, decision trees, neural networks, adaptive boosting (AdaBoost), Gaussian naive Bayes and quadratic discriminant analysis] were used to classify each IIM subtype and select the most important metabolites as potential biomarkers. Results OPLS‐DA showed a clear separation between NC and IIM subtypes in plasma and urine metabolic profiles. KEGG pathway enrichment analysis revealed multiple unique and shared disturbed metabolic pathways in IIM main [dermatomyositis (DM), anti‐synthetase syndrome (ASS), and immune‐mediated necrotizing myopathy (IMNM)] and MSA‐defined subtypes (anti‐Mi2+, anti‐MDA5+, anti‐TIF1γ+, anti‐Jo1+, anti‐PL7+, anti‐PL12+, anti‐EJ+, and anti‐SRP+), such that fatty acid biosynthesis was significantly altered in both plasma and urine in all main IIM subtypes (enrichment ratio > 1). Random forest and AdaBoost performed best in classifying each IIM subtype among the 10 ML models. Using the feature selection methods in ML models, we identified 9 plasma and 10 urine metabolites that contributed most to separate IIM main subtypes and MSA‐defined subtypes, such as plasma creatine (fold change = 3.344, P = 0.024) in IMNM subtype and urine tiglylcarnitine (fold change = 0.351, P = 0.037) in anti‐EJ+ ASS subtype. Sixteen common metabolites were found in both the plasma and urine samples of IIM subtypes. Among them, some were correlated with clinical features, such as plasma hypogeic acid (r = −0.416, P = 0.005) and urine malonyl carnitine (r = −0.374, P = 0.042), which were negatively correlated with the prevalence of interstitial lung disease. Conclusions In both plasma and urine samples, IIM main and MSA‐defined subtypes have specific metabolic signatures and pathways. This study provides useful clues for understanding the molecular mechanisms, searching potential diagnosis biomarkers and therapeutic targets for IIM.

Dengue is prevalent in tropical and subtropical regions. As an arbovirus disease, it is mainly transmitted by Aedes aegypti and Aedes albopictus. According to the previous studies, temperature is closely related to the survival of Aedes mosquitoes, the proliferation of dengue virus (DENV) and the vector competence of Aedes to transmit DENV. This review describes the correlations between temperature and dengue epidemics, and explores the potential reasons including the distribution and development of Aedes mosquitoes, the structure of DENV, and the vector competence of Aedes mosquitoes. In addition, the immune and metabolic mechanism are discussed on how temperature affects the vector competence of Aedes mosquitoes to transmit DENV.

The problem of low transformer load rate (less than 60%) in the actual operation of large stadiums and gymnasiums has affected the waste of power energy and the improvement of power cost in the industry. The transformer failure of large stadiums and gymnasiums may further cause serious power accidents in the future. In order to solve the above problems, firstly, this paper analyzes the transformer load rate data of the two large sports venues(Nanchang International Sports and Tennis Stadium in Jiangxi Province)and probes into the design data and calculation coefficient of the transformer of the large sports venues; Secondly, this paper puts forward some suggestions on how to improve the operation load rate of transformers in stadiums from the aspects of transformer design and selection of demand coefficient; Thirdly, the paper establishes the system hardware environment by using firewall, special switch and information database, and then realizes the design of software deduction program through correlation analysis, fault diagnosis and cluster deduction, and completes the design of transformer fault deduction system in large sports venues. Finally, the results clearly show that the transformer load rate is relatively low in large sports stadiums(the highest value is only about 55% and well below the 60% standard). On other hand, compared with the traditional system and based on big data association, the average relative errors of fault points 1, 2 and 3 decreased by 67.8%, 56.4% and 51.6% respectively and the diagnosis time has also been greatly shorten by the transformer fault deduction system. In short, this paper uses association rules to mine and obtain transformer fault characteristics, combined with the advantages of virtual reality technology, designs an effective and reliable transformer fault deduction visualization system for large sports venues.

SARS-CoV-2 breakthrough infections in vaccinated individuals are a public health concern. Here, the authors analyse the clinical characteristics and profile immune alterations among vaccinated and non-vaccinated residents with Delta SARS-CoV-2 infection in Guangzhou.

Background and aimsIt is necessary to identify simple biomarkers that can efficiently predict the efficacy of long-term antiretroviral therapy (ART) against human immunodeficiency virus (HIV), especially in underdeveloped countries. We characterized the dynamic changes in plasma interleukin-18 (IL-18) and assessed its performance as a predictor of long-term virological response.MethodsThis was a retrospective cohort study of HIV-1-infected patients enrolled in a randomized controlled trial with a follow-up of 144 weeks of ART. Enzyme-linked immunosorbent assay was performed to evaluate plasma IL-18. Long-term virological response was defined as HIV-1 RNA 64 pg./mL, a OR 19.10, 95% CI: 2.36–154.80) was the only independent predictor of long-term virological response.ConclusionEarly on-treatment plasma IL-18 could act as a promising indicator for long-term virological response in patients with HIV-1 infection. Chronic immune activation and inflammation may represent a potential mechanism; further validation is necessary.

Two novel Gram-positive bacteria, designated strains REN8T and REN14T, were isolated from baijiu pit mud in Sichuan Province, China. REN8T achieved the best growth at 37°C, a pH of 8.0, and a NaCl concentration of 2%, while REN14T displayed optimal growth at 37°C, a pH of 6.0, and a NaCl concentration of 1%. 16S rRNA and genomic phylogenetic analysis showed that REN8T and REN14T were clustered with the genus Planococcus. The genomic DNA G + C contents of REN8T and REN14T were 46.7 and 45.1 mol%, respectively. The dDDH and ANI values were 24.5 and 80.43% between REN8T and P. salinarum (the most closely related type strain) and 25.1 and 82.42% between REN14T and P. soli (the most closely related type strain). Genomic analysis showed that several carbohydrate-active enzymes and secondary metabolite gene clusters existed in REN8T and REN14T. Chemotaxonomic characteristics of REN8T and REN14T included major fatty acids, predominant menaquinones, and polar lipids, all of which were consistent with the genus Planococcus. Based on the polyphasic taxonomic method, these two strains represent two novel species of the genus Planococcus; the name Planococcus beigongshangi sp. nov. is proposed for the type strain REN8T (=JCM 33964T = GDMCC 1.2213T), and the name Planococcus beijingensis sp. nov. is proposed for the type strain REN14T (=JCM 34410T = GDMCC 1.2209T). The addition of REN8T and REN14T might improve the quality of huangjiu by considerably increasing the amino acid nitrogen content and acidity and decreasing the bioamine content, with no significant change in alcohol content.

Abstract Background Clinical translation of therapeutic nuclear acid, particularly those targeting tumor progression, has been hampered by the intrinsic weaknesses of nuclear acid therapeutic including poor systemic stability, rapid clearance, low membrane permeability and lack of targeting ability. Small nuclear acid engineered into carrier-free nanodrugs with structural stability and disease targeting may be viable to overcome pharmaceutical obstacles of nuclear acid. Methods A general method through a mild and simple chemistry was established to convert therapeutic miRNA into an infinite Auric-sulfhydryl coordination supramolecular miRNA termed IacsRNA with near-spherical nanostructure, high colloid as well as anti-hydrolysis stability and low macrophage uptakes. Results IacsRNA presented the increased half-life period in circulation and accumulation at tumor sites in comparison to normal miRNA. Moreover, Iacs-miR-30c showed no toxicity of viscera and sanguis system in the 5-time injection dosage of the treatment. More importantly, Iacs-miR-30c potently suppressed the Wnt signaling pathway in vitro and in vivo, and effectively sensitized both potency of 5-Fu in PDX model of colon cancer and Anti-PD1 in B16F10 homograft model of melanoma. Conclusion Collectively, this work amply confirmed the design of IacsRNA as a general and viable strategy of nano-pharmaceutic to concert flimsy therapeutic miRNA into potential drugs. Considering from a broader perspective, the miRNA-initiated infinite coordination self-assembly strategy has distinct advantages in resurrecting nuclear acid therapeutics, probably bringing new inspiration to RNA-derived therapeutics of a great variety of human diseases including cancer. Objective To investigate the effect of continuous positive airway pressure (CPAP) treatment on cognitive function in stroke patients with obstructive sleep apnoea (OSA) by exploring randomised controlled trials (RCTs).Methods Published RCTs that assessed the therapeutic effects of CPAP on cognition in stroke patients with OSA, compared with controls or sham CPAP, were included. Electronic databases, including MEDLINE, Embase and Cochrane library, were searched in October 2020 and October 2021. Risk of bias was assessed using the Cochrane collaboration tools. A random effects or fixed effects model was used according to heterogeneity. The outcomes were global cognitive gain, improvement in cognitive domain and subjective sleepiness.Results 7 RCTs, including 327 participants, comparing CPAP with control or sham CPAP treatment were included. 6 RCTs with 270 participants reported results related to global cognition, and CPAP treatment had no significant effects on global cognitive gain in stroke patients with OSA (standardised mean difference (SMD), 0.18; 95% CI, –0.07 to 0.42; p=0.153). Abstract Background Stroke is the major cause of adult neurocognitive disorders (NCDs), and presents a significant burden on both of the families and society. To improve the cerebral injury, we generated a blood–brain barrier penetrating peptide TAT‐LBD‐Ngn2, in which Ngn2 (Neurogenin2) is a classical preneural gene that enhances neurogenesis, and neural precursor cells survival and differentiation. We previously demonstrated that it has a short‐term protective effect against cerebral ischemia‐reperfusion injury. However, it is uncertain if TAT‐LBD‐Ngn2 could promote neurogenesis to exhibit long‐term therapeutic impact. Methods and results In present study, TAT‐LBD‐Ngn2 was administered for 14 or 28 days following bilateral common carotid arteries occlusion (BCCAO). After confirming that TAT‐LBD‐Ngn2 could cross the brain blood barrier and aggregate in the hippocampus, we conducted open field test, Morris water maze and contextual fear conditioning to examine the long‐term effect of TAT‐LBD‐Ngn2 on cognition. We discovered that TAT‐LBD‐Ngn2 significantly improved the spatial and contextual learning and memory on both days 14 and 28 after BCCAO, while TAT‐LBD‐Ngn2 exhibited anxiolytic effect only on day 14, but had no effect on locomotion. Using western blot and immunofluorescence, TAT‐LBD‐Ngn2 was also shown to promote neurogenesis, as evidenced by increased BrdU+ and DCX+ neurons in dentate gyrus. Meanwhile, TAT‐LBD‐Ngn2 elevated the expression of brain derived neurotrophic factor rather than nerve growth factor compared to the control group. Conclusions Our findings revealed that TAT‐LBD‐Ngn2 could dramatically promote learning and memory in long term by facilitating neurogenesis in the hippocampus after global cerebral ischemia, indicating that TAT‐LBD‐Ngn2 may be an appealing candidate for treating poststroke NCD.

With the development of new power systems, the construction concept imposes new requirements on the internal environment of the equipment in terms of moisture protection. The study addresses the requirements of dehumidification and temperature control in the internal environment of prefabricated substations and builds the test platform for prefabricated substations to investigate the effect of coordinated operation of distributed thermoelectric dehumidifier (DTD) and centralized thermoelectric dehumidifier (CTD) on the prefabricated bay environment. It is found that: distributed dehumidifiers are effective in dehumidifying environments with humidification rates less than 2.848 ml/min; CTD should be used when the humidification rate is greater than 2.848 ml/min. DTD its relative humidity regulation can be strong but cause the ambient temperature to rise, CTD power consumption is low and the ambient temperature change is small. When the coordinated operation of the distributed and centralized dehumidifier is used to reduce the relative humidity from 95% to 69.5%, its total power consumption is only 0.260 kWh and the ambient temperature increases by 3.2 °C. Therefore, the comprehensive performance advantage is obvious, which is conducive to improving the ability to resist sudden changes in the environment and ensuring stable equipment operation.

The emerging tissue-engineered bio-assemblies are revolutionizing the regenerative medicine, and provide a potential program to guarantee predictive performance of stem-cell-derived treatments in vivo and hence support their clinical translation. Mesenchymal stem cell (MSC) showed the attractive potential for the therapy of nervous system injuries, especially spinal cord injury (SCI), and yet failed to make an impact on clinical outcomes. Herein, under the guidance of the embryonic development theory that appropriate cellular coarctations or clustering are pivotal initiators for the formation of geometric and functional tissue structures, a developmentally engineered strategy was established to assemble DPMSCs into a bio-assembly termed Spinor through a three-level sequential induction programme including reductant, energy and mechanical force stimulation. Spinor exhibited similar geometric construction with spinal cord tissue and attain autonomy to released exosome with the optimized quantity and quality for suppressing cicatrization and inflammation and promoting axonal regeneration. As a spinal cord fascia and exosome mothership, Spinor guided the in-situ neuroplasticity of spinal cord in vivo, and caused the significant motor improvement, sensory recovery, and faster urinary reflex restoration in rats following SCI, while maintaining a highly favorable biosafety profile. Collectively, Spinor not only is a potentially clinical therapeutic paradigm as a living “exosome mothership” for revisiting Prometheus' Myth in SCI, but can be viewed allowing developmentally engineered manufacturing of biomimetic bio-assemblies with complex topology features and inbuilt biofunction attributes towards the regeneration of complex tissues including nervous system.

Aluminum appears as a promising structural material as it shows many benefits such as a great strength to weight ratio, low maintenance costs, resistance to corrosion, recyclability, etc. Accordingly, characterizing the behavior and resistance of different aluminum sections under various loading conditions is essential. The current paper presents an experimental investigation on aluminum sections with simple and complex geometries to study the effect of cross-section shape on the buckling response of the aluminum sections. Firstly, eight tensile coupon tests were conducted on coupons of aluminum alloy 6061-T6 to accurately determine its material properties. Geometrical imperfections on the surface of each specimen were then measured using a 3D scanner. Further, eight stub column tests were also performed to study the behavior of I, H and complex cross-sections under pure compression. In addition to this, 12 cross-section tests under combined compression and bending are currently under way to study the buckling behavior of these specimens under eccentric compression.

This paper investigates the ultimate resistance of aluminum circular hollow sections (C.H.S.) under simple load cases as affected by local buckling and material strain hardening. Following the development of advanced non-linear shell finite element models and their validation against existing test data, extensive non-linear numerical analyses were carried out, varying several key parameters such as alloy types, load case (compression or pure bending) and various section dimensions. The numerical results are used as references to analyze the structural performance of aluminum C.H.S., and to assess the accuracy of design equations based on the Overall Interaction Concept (O.I.C.). The O.I.C.-based proposal is shown to yield more accurate, consistent, and continuous resistance predictions than Eurocode 9 recommendations.

Aluminum has a bright future as a structural material due to its excellent corrosion resistance, durability, lightweight, and complete recyclability. However, it is necessary to fully understand its mechanical behaviour under various loading conditions to make it competitive with other materials, such as concrete or steel, for civil engineering applications, especially as primary load-bearing structural members. To develop an in-depth knowledge on the behaviour of extruded aluminum sections of various shapes, an extensive experimental study was undertaken with specific emphasis on analyzing the buckling response of rectangular and square hollow sections (RHS and SHS) with 6061-T6 aluminum alloy under compressive loads. In this regard, six stub column tests were performed under axial compression, while eleven short beam-column tests under eccentric compression are currently in progress. Additionally, six tensile coupon tests were performed to obtain the full material stress–strain curve, and initial geometrical imperfections were measured mechanically and using a 3D scanner. Finally, the results of the stub column tests were compared to the resistances calculated using the Canadian standard CSA S157, which were generally conservative compared to the experimental observations.

Aluminium alloys exhibit a non-linear stress–strain material response with significant strain-hardening effects. Whereas the latter shall be considered in the design of aluminium elements, they are largely ignored in most design codes, such as Eurocode 9 and the Canadian Standards. Preliminary studies on typical aluminium I-shapes indeed indicated that as much as 40% extra resistance could be reached through more accurate and appropriate design recommendations. Accordingly, an original design approach based on the Overall Interaction Concept (O.I.C.) is developed to better account for the actual behaviour of aluminium structural shapes. In this respect, non-linear finite element models are developed within ABAQUS and further validated against experimental data. Subsequently, comprehensive parametric studies are performed to collect numerical reference results, and comparisons are performed with resistance predictions from well-known design standards. The performance of the newly developed O.I.C.-based design approach is shown to significantly improve accuracy, safety, consistency, and reliability levels.

Abstract Background Transmitted drug resistance (TDR) that affects the effectiveness of the first-line antiretroviral therapy (ART) regimen is becoming prevalent worldwide. However, its prevalence and transmission among HIV-1 treatment-naïve patients in Guangdong, China are rarely reported. We aimed to comprehensively analyze the prevalence of TDR and the transmission clusters of HIV-1 infected persons before ART in Guangdong. Methods The HIV-1 treatment-naïve patients were recruited between January 2018 and December 2018. The HIV-1 pol region was amplified by reverse transcriptional PCR and sequenced by sanger sequencing. Genotypes, surveillance drug resistance mutations (SDRMs) and TDR were analyzed. Genetic transmission clusters among patients were identified by pairwise Tamura-Nei 93 genetic distance, with a threshold of 0.015. Results A total of 2368 (97.17%) HIV-1 pol sequences were successfully amplified and sequenced from the enrolled 2437 patients. CRF07_BC (35.90%, 850/2368), CRF01_AE (35.56%, 842/2368) and CRF55_01B (10.30%, 244/2368) were the main HIV-1 genotypes circulating in Guangdong. Twenty-one SDRMs were identified among fifty-two drug-resistant sequences. The overall prevalence of TDR was 2.20% (52/2368). Among the 2368 patients who underwent sequencing, 8 (0.34%) had TDR to protease inhibitors (PIs), 22 (0.93%) to nucleoside reverse transcriptase inhibitors (NRTIs), and 23 (0.97%) to non-nucleoside reverse transcriptase inhibitors (NNRTIs). Two (0.08%) sequences showed dual-class resistance to both NRTIs and NNRTIs, and no sequences showed triple-class resistance. A total of 1066 (45.02%) sequences were segregated into 194 clusters, ranging from 2 to 414 sequences. In total, 15 (28.85%) of patients with TDR were included in 9 clusters; one cluster contained two TDR sequences with the K103N mutation was observed. Conclusions There is high HIV-1 genetic heterogeneity among patients in Guangdong. Although the overall prevalence of TDR is low, it is still necessary to remain vigilant regarding some important SDRMs.

Since prefabricated cabin substations became a comprehensive application achievement of the new infrastructure intelligent integrated system, its electrical equipment failure, and power system safety due to humidity condensation is of great concern. In this paper, thermoelectric cooling superconducting thermoelectric dehumidifiers (TCSTD) are developed and the optimal operating conditions and the dehumidification performance of the TCSTD under actual operating conditions are investigated through tests. The results show that the optimal dehumidification rate of the TCSTD is 188.4 ml/h when relative humidity is 95% and the outlet air velocity is 2.3 m/s. The addition of TCSTD on top of thermal assistance can further reduce the relative humidity to 53.05%, a reduction of up to 32.36%. TCSTD is used in high temperature, high humidity environments to reduce not only the ambient relative humidity but also the moisture content of the air. The proposal is that attention should be paid to both dehumidification and relative humidity per unit of power consumption to meet the requirements for moisture protection and energy saving in prefabricated cabin substations. The results of the study will provide a reference for the application of the new prefabricated cabin substation.

Background: Major depressive disorder (MDD) is a serious mental illness characterized by mood changes and high suicide rates. However, no studies are available to support a blood test method for MDD diagnosis. The objective of this research was to identify potential peripheral blood biomarkers for MDD and characterize the novel pathophysiology.Methods: We accessed whole blood microarray sequencing data for MDD and control samples from public databases. Biological functions were analysed by GO and KEGG pathway enrichment analyses using the clusterprofile R package. Infiltrated immune cell (IIC) proportions were identified using the CIBERSORT algorithm. Clustering was performed using the ConsensusClusterPlus R package. Protein–protein interactions (PPI) were assessed by constructing a PPI network using STRING and visualized using Cytoscape software. Rats were exposed to chronic unpredictable mild stress (CUMS) for 6 weeks to induce stress behaviour. Stress behaviour was evaluated by open field experiments and forced swimming tests. Flow cytometry was used to analyse the proportion of CD8+ T cells. The expression of the corresponding key genes was detected by qRT–PCR.Results: We divided MDD patients into CD8H and CD8L clusters. The functional enrichment of marker genes in the CD8H cluster indicated that autophagy-related terms and pathways were significantly enriched. Furthermore, we obtained 110 autophagy-related marker genes (ARMGs) in the CD8H cluster through intersection analysis. GO and KEGG analyses further showed that these ARMGs may regulate a variety of autophagy processes and be involved in the onset and advancement of MDD. Finally, 10 key ARMGs were identified through PPI analysis: RAB1A, GNAI3, VAMP7, RAB33B, MYC, LAMP2, RAB11A, HIF1A, KIF5B, and PTEN. In the CUMS model, flow cytometric analysis confirmed the above findings. qRT–PCR revealed significant decreases in the mRNA levels of Gnai3, Rab33b, Lamp2, and Kif5b in the CUMS groups.Conclusion: In this study, MDD was divided into two subtypes. We combined immune infiltrating CD8+ T cells with autophagy-related genes and screened a total of 10 ARMG genes. In particular, RAB1A, GNAI3, RAB33B, LAMP2, and KIF5B were first reported in MDD. These genes may offer new hope for the clinical diagnosis of MDD.

Abstract Background Bone mass loss in aging is linked with imbalanced lineage differentiation of bone marrow mesenchymal stem cells (BMMSCs). Recent studies have proved that histone deacetylases (HDACs) are regarded as key regulators of bone remodeling. However, HDACs involve in regulating BMMSC bio-behaviors remain elusive. Here, we investigated the ability of HDAC9 on modulation of autophagy and its significance in lineage differentiation of BMMSCs. Methods The effects of HDAC9 on lineage differentiation of BMMSCs and autophagic signaling were assessed by various biochemical (western blot and ChIP assay), morphological (TEM and confocal microscopy), and micro-CT assays. Results Sixteen-month mice manifested obvious bone mass loss and marrow fat increase, accompanied with decreased osteogenic differentiation and increased adipogenic differentiation of BMMSCs. Further, the expression of HDAC9 elevated in bone and BMMSCs. Importantly, HDAC9 inhibitors recovered the lineage differentiation abnormality of 16-month BMMSCs and reduced p53 expression. Mechanistically, we revealed that HDAC9 regulated the autophagy of BMMSCs by controlling H3K9 acetylation in the promoters of the autophagic genes, ATG7, BECN1, and LC3a/b, which subsequently affected their lineage differentiation. Finally, HDAC9 inhibition improved endogenous BMMSC properties and promoted the bone mass recovery of 16-month mice. Conclusions Our data demonstrate that HDAC9 is a key regulator in a variety of bone mass by regulating autophagic activity in BMMSCs and thus a potential target of age-related bone loss treatment.

ABSTRACTThe novel coronavirus (2019-nCoV) infection caused pneumonia. we retrospectively analyzed the virus presence in the pharyngeal swab, blood, and the anal swab detected by real-time PCR in the clinical lab. Unexpectedly, the 2109-nCoV RNA was readily detected in the blood (6 of 57 patients) and the anal swabs (11 of 28 patients). Importantly, all of the 6 patients with detectable viral RNA in the blood cohort progressed to severe symptom stage, indicating a strong correlation of serum viral RNA with the disease severity (p-value = 0.0001). Meanwhile, 8 of the 11 patients with annal swab virus-positive was in severe clinical stage. However, the concentration of viral RNA in the anal swab (Ct value = 24 + 39) was higher than in the blood (Ct value = 34 + 39) from patient 2, suggesting that the virus might replicate in the digestive tract. Altogether, our results confirmed the presence of virus RNA in extra-pulmonary sites.

Objectives: To evaluate the efficacy and safety of first-line treatment with a dendritic cell vaccination for lung cancer (DCVAC/LuCa), standard of care chemotherapy and Shenqi Fuzheng injection in patients with advanced (stage IIIB/IV) non-small cell lung cancer. Patients and Methods: Patients with histologically or cytologically confirmed recurrent metastatic or advanced NSCLC (stage IIIB/IV) with wild-type epidermal growth factor receptor (EGFR) or EGFR mutation which does not confer increased tumor susceptibility to EGFR-interacting drugs were recruited. For the treatment period, the first cycle of standard of care therapy (SoC) started 2 to 14 days after the leukapheresis procedure. SoC continued 4 to 6 cycles. DCVAC/LuCa was administered from the second cycle of SoC. DCVAC/LuCa was administered in a 3-week cycle schedule (5 doses) and then in a 6-week cycle schedule. Shenqi Fuzheng injection was administered 3 days before each DCVAC/LuCa administration for a total of 14 daily doses. Patients would undergo disease evaluation by computed tomography (CT) scan every 3 months. The primary and secondary endpoint was efficacy with regard to objective response rate (ORR) and progression free survival (PFS). The safety profile was measured by: incidence, type, and severity of all adverse events (AEs), laboratory abnormalities (blood routine test, urine test, and chemical test), physical status, and vital signs. Qi insufficiency was evaluated by tongue diagnosis and questionnaire survey with “Classification and Determination of constitution in TCM.” Results: Twenty-three patients from 3 hospitals who received combination therapy were included. ORR was 34.8% (95% CI:16.4%-57.3%). Median duration of response was 5.51 m (95% CI:2.70-8.32). Median PFS was 10.72 m (95% CI:4.52-16.93), 1-year survival was 77.8%. mOS was 21.97 m (95% CI:13.68-30.25). BackgroundAnti-TIF1γ antibodies are a class of myositis-specific antibodies (MSAs) and are closely associated with adult cancer-associated myositis (CAM). The heterogeneity in anti-TIF1γ+ myositis is poorly explored, and whether anti-TIF1γ+ patients will develop cancer or not is unknown at their first diagnosis. Here, we aimed to explore the subtypes of anti-TIF1γ+ myositis and construct machine learning classifiers to predict cancer in anti-TIF1γ+ patients based on clinical features.MethodsA cohort of 87 anti-TIF1γ+ patients were enrolled and followed up in Xiangya Hospital from June 2017 to June 2021. Sankey diagrams indicating temporal relationships between anti-TIF1γ+ myositis and cancer were plotted. Elastic net and random forest were used to select and rank the most important variables. Multidimensional scaling (MDS) plot and hierarchical cluster analysis were performed to identify subtypes of anti-TIF1γ+ myositis. The clinical characteristics were compared among subtypes of anti-TIF1γ+ patients. Machine learning classifiers were constructed to predict cancer in anti-TIF1γ+ myositis, the accuracy of which was evaluated by receiver operating characteristic (ROC) curves.ResultsForty-seven (54.0%) anti-TIF1γ+ patients had cancer, 78.7% of which were diagnosed within 0.5 years of the myositis diagnosis. Fourteen variables contributing most to distinguishing cancer and non-cancer were selected and used for the calculation of the similarities (proximities) of samples and the construction of machine learning classifiers. The top 10 were disease duration, percentage of lymphocytes (L%), percentage of neutrophils (N%), neutrophil-to-lymphocyte ratio (NLR), sex, C-reactive protein (CRP), shawl sign, arthritis/arthralgia, V-neck sign, and anti-PM-Scl75 antibodies. Anti-TIF1γ+ myositis patients can be clearly separated into three clinical subtypes, which correspond to patients with low, intermediate, and high cancer risk, respectively. Machine learning classifiers [random forest, support vector machines (SVM), extreme gradient boosting (XGBoost), elastic net, and decision tree] had good predictions for cancer in anti-TIF1γ+ myositis patients. In particular, the prediction accuracy of random forest was >90%, and decision tree highlighted disease duration, NLR, and CRP as critical clinical parameters for recognizing cancer patients.ConclusionAnti-TIF1γ+ myositis can be separated into three distinct subtypes with low, intermediate, and high risk of cancer. Machine learning classifiers constructed with clinical characteristics have favorable performance in predicting cancer in anti-TIF1γ+ myositis, which can help physicians in choosing appropriate cancer screening programs.

The prevalence of diabetes-associated cognitive dysfunction (DACD) has increased to 13.5%. Dementia, as the most severe DACD, is the second leading cause of death in patients with diabetes mellitus. Hence, the potential mechanisms of DACD for slowing or halting its progression need to be urgently explored. Given that the sigma-1 receptor (Sig-1R), a chaperone protein located in the endoplasmic reticulum (ER)-mitochondrion contact membranes to regulate ER stress (ERS), is associated with cognitive outcomes in neurodegenerative diseases, this study aimed to investigate the role of astrocytic Sig-1R in DACD and its underlying mechanism. Here, we examined the levels of ERS and complement component 3/3a (C3/C3a) from primary astrocytes with different concentrations of glucose and treatment. Subsequently, HT22 neurons were cultured in different astrocyte-conditioned medium, and the expression of synaptic proteins was detected. We constructed type 1 diabetes mellitus (T1DM) model to evaluate the astrocytic Sig-1R mechanism on synapse and cognitive function changes. In vitro, high glucose concentration downregulated Sig-1R and aggravated ERS in astrocytes, resulting in synapse deficits. PRE-084, a high-affinity and selective Sig-1R agonist, inhibited astrocytic ERS and complement cascades and restored synaptic damage, while the Sig-1R antagonist displayed the opposite results. Moreover, C3a receptor antagonist (C3aRA) could mimic the effect of PRE-084 and exerted neuroprotective effects. In vivo, PRE-084 substantially reduced ER-mitochondrion contact, activation of ERS, and C3/C3a secretion in mice with T1DM. Additionally, the synaptic loss and neurobehavioral dysfunction of mice with T1DM were less pronounced in both the PRE-084 and C3aRA treatment groups. These findings demonstrated that Sig-1R activation reduced the astrocytic ER-mitochondrion contact, ERS activation, and complement-mediated synaptic damage in T1DM. This study suggested the mechanisms and potential therapeutic approaches for treating DACD.

To explore the effects of Chinese raspberry (Fu-Pen-Zi) on diabetes, a polyphenol-enriched extract (FPZ) was prepared. Streptozotocin (STZ)-induced diabetic mice were daily administered FPZ (30 mg/kg body weight) for 18 days, the anti-diabetic effects were evaluated. Compared to STZ alone mice, FPZ-treatment ameliorated STZ-induced type 1 diabetes, reduced body weight loss (by 71.61%), hyperglycemia (fasting blood glucose reduced by 19.23%), hyperlipidemia (hepatic triglycerides reduced by 15.50%) and skeletal muscle atrophy (quadriceps mass increased by 21.46%). FPZ protected pancreatic β-cells from STZ-induced damage and promoted insulin secretion, preserved insulin signaling in liver tissues and skeletal muscles. FPZ improved glucose tolerance, modulated liver glycogen metabolism, and inhibited hepatic gluconeogenesis via activating insulin/Akt/Gsk3β and LKB1/AMPK signaling. Moreover, FPZ reduced liver oxidative stress and steatosis, reduced Atrogin-1 and Trim63 expression, contributes to muscle atrophy inhibition. This study highlights the potential usage of FPZ as nutritional interventions for type 1 diabetes and muscle atrophy management.

Background: Integrase strand-transfer inhibitor (INSTI)-containing regimens have gradually been administered in Guangdong Province, China beginning in 2016, and INSTI-related drug resistance (DR) may occur and should be monitored among HIV-1-infected patients. Objective: To investigate the prevalence of INSTI-related resistance among HIV-1-infected individuals in Guangdong and provide evidence for the optimal administration of INSTIs. Methods: This study recruited 1208 HIV-1-infected patients (including 404 ART-naive and 804 ART-experienced patients) between June 2021 and April 2022. The entire integrase gene was amplified from blood plasma. Demographic and epidemiological information were collected. INSTI mutations and susceptibility were interpreted using the Stanford HIV Drug Resistance Database HIVdb program. Results: Of the 1208 enrolled individuals, 2.65% (32/1208) carried at least one INSTI major or accessory drug resistance mutation (DRM), with 1.49% (6/404) being from ART-naive individuals and 3.23% (26/804) from ART-experienced individuals. Among them, seven polymorphic major mutations were detected. Although no INSTI drug resistance was found among treatment-naive patients, seven ART-experienced patients (0.87%, 7/804) carried mutations conferring resistance to INSTIs. Conclusion: The overall prevalence of INSTI DRMs and DR was comparatively low among ART-naive and ART-treated populations in Guangdong; however, INSTI-related polymorphic mutations were observed. Surveillance should be reinforced before transfer to INSTI-containing regimens.

Dermatomyositis (DM) is an idiopathic inflammatory myopathy characterized by cutaneous manifestations. We first identified the profiles of noncoding RNAs (lncRNAs and miRNAs) in peripheral neutrophil exosomes (EXOs) of DM patients and explored their potential functional roles. Bioinformatics analyses were performed with R packages. Real-time quantitative PCR was used to validate the altered RNAs in DM neutrophil EXO-stimulated human dermal microvascular endothelial cells (HDMECs) and human skeletal muscle myoblasts (HSkMCs). In DM neutrophil EXOs, 124 upregulated lncRNAs (with 1,392 target genes), 255 downregulated lncRNAs (with 1867 target genes), 17 upregulated miRNAs (with 2,908 target genes), and 15 downregulated miRNAs (with 2,176 target genes) were identified. GO analysis showed that the differentially expressed (DE) lncRNAs and DE miRNAs participated in interleukin-6 and interferon-beta production, skeletal muscle cell proliferation and development, and endothelial cell development and differentiation. KEGG analysis suggested that DE lncRNAs and DE miRNAs were enriched in the PI3K–Akt, MAPK, AMPK and FoxO signalling pathways. Many novel and valuable DE lncRNAs and DE miRNAs interacted and cotargeted in the PI3K–Akt, MAPK, AMPK and FoxO signalling pathways. Our study suggests that neutrophil EXOs participate in DM pathogenesis through lncRNAs and miRNAs in the PI3K–Akt, MAPK, AMPK and FoxO signalling pathways.

Abstract Background Radiation exposure negatively affects the regenerative ability and makes reconstruction of bone defects after tumor section difficult. miR-34a is involved in radiation biology and bone metabolism. The aim of this study was to investigate whether miR-34a could contribute to bone regeneration in irradiated bone defects. Methods The expression of miR-34a was analyzed during the osteoblastic differentiation of irradiated BMSCs and bone formation in irradiated bone defects. miR-34a mimics and miR-34a inhibitor were used to upregulate or suppress the expression of miR-34a in BMSCs irradiated with 2 or 4 Gy X-ray radiation. In vitro osteogenesis and subcutaneous osteogenesis were used to assess the effects of miR-34a on the osteogenic ability of radiation-impaired BMSCs. Collagen-based hydrogel containing agomiR-34a or antagomiR-34a were placed into the 3-mm defects of irradiated rat tibias to test the effect of miR-34a on bone defect healing after irradiation. Results miR-34a was upregulated in the process of bone formation after irradiation. Transfecting radiation-impaired BMSCs with miR-34a mimics enhanced their osteoblastic differentiation in vitro by targeting NOTCH1. Overexpression of miR-34a enhanced the ectopic bone formation of irradiated BMSCs. In situ delivery of miR-34a promoted bone regeneration in irradiated bone defects. Conclusions miR-34a promoted the osteoblastic differentiation of BMSCs and enhanced the ectopic bone formation after irradiation. miR-34a promoted bone defect healing in irradiated rat tibias. miR-34a-targeted therapy might be a promising strategy for promoting the reconstruction of bone defects after radiotherapy.

Glucitol-core containing gallotannins (GCGs) are characteristic constituents of the red maple (Acer rubrum) species. To pursue the development of red maple for nutraceutical applications, GCGs-enriched red maple leaves extract (MLE) was evaluated for its effects on obesity, gut dysbiosis and short chain fatty acids (SCFAs) production. Our results demonstrated that MLE alleviated high-fat diet-induced obesity, reduced body weight gain and fat mass, improved liver steatosis and insulin resistance, and mitigated adipose hypertrophy and inflammation. Additionally, MLE increased total SCFAs, acetic acid and n-butyric acid content, but exerted no impact on propionic acid production. Moreover, MLE modulated gut microbiota community structure and certain bacteria relative abundance, including Prevotella and Eubacterium. Our work firstly reports a potential association between colon-derived SCFAs production and metabolic improvement due to GCGs-enriched red maple leaves extract administration, and highlights the utilization of red maple gallotannins as a dietary ingredient for preventing obesity and related metabolic diseases.

Interleukin-33 (IL-33), a member of the IL-1 superfamily, functions as a traditional cytokine and nuclear factor. It is proposed to have an “alarmin” role. IL-33 mediates biological effects by interacting with the ST2 receptor and IL-1 receptor accessory protein, particularly in innate immune cells and T helper 2 cells. Recent articles have described IL-33 as an emerging pro-fibrotic cytokine in the immune system as well as a novel potential target for systemic sclerosis. Here, we review the available information and focus on the pleiotropic expression and pathogenesis of IL-33 in systemic sclerosis, as well as the feasibility of using IL-33 in clinical applications.

The chemical composition and anti-hyperglycaemic effects of polyphenol-enriched Longan pericarp extracts (LPE) were investigated in the present study. The phenolic profile of LPE was determined by HPLC-DAD/MS analysis. Nine phenolics were identified, and of these, three major components were quantified by LC-MS/MS. The in vitro anti-inflammatory effects of a phenolic-enriched Canadian maple syrup ethyl acetate extract (MS-EtOAc) and 15 purified phenolic constituents were evaluated in a LPS-stimulated RAW 264.7 murine macrophage cell model. MS-EtOAc decreased nitric oxide (NO) and prostaglandin-E2 (PGE2) production at 10–100 μg/mL concentrations. The observed NO inhibition was a direct result of reduced nitric oxide synthase (iNOS) protein and gene expression through suppression of NF-κB transcriptional activation. In addition, MS-EtOAc upregulated cyclooxygenase-2 (COX-2) mRNA and protein expression. Among the 15 pure isolates, (E)-3,3′-dimethoxy-4,4′-dihydroxystilbene was most effective in decreasing both NO and PGE2 levels. However, 4-acetylcatechol, tyrosol, and protocatechuic acid only reduced PGE2 levels. Thus, the potential anti-inflammatory activity of MS-EtOAc can be attributed to its unique combination of compounds and not as a result of a single purified phenolic constituent alone. Future research on the purified phenolic compounds will be useful in understanding the overall in vitro anti-inflammatory effects of maple syrup.

Maple sap has been consumed for centuries as a tonic by the indigenous peoples of eastern North America but is primarily utilized in this region to produce maple syrup. The natural watery form of maple sap makes its application as a functional beverage appealing but due to microbial growth, sterilization or pasteurization would be necessary before sap could be consumed. This study was designed to investigate the chemical composition (sugars, amino acids, organic acids, minerals, and phenolics) and antioxidant effects of maple sap after undergoing pasteurization and sterilization. After both processes, sugars, amino acids, organic acids, and minerals were preserved in the sap samples and they had similar phenolic contents (0.25–0.27 mg/100 g gallic acid equivalents) and antioxidant activities (IC50 ca. 550 μg/mL by DPPH assay). HPLC-DAD analyses revealed over 25 constituents in the sap samples of which 15 were identified using phenolic standards. In addition, one compound, 3′,5′-dimethoxy-4′-hydroxy-(2-hydroxy)acetophenone, not previously reported from maple syrup, was isolated and identified (by NMR) for the first time from maple sap. Therefore, the preservation of chemical constituents and antioxidant activity in maple sap after pasteurization and sterilization warrants its application as a functional beverage beyond its primary use for maple syrup production alone.

The antiproliferative effects of Canadian maple syrup (grades C and D) extracts and fifty-one purified phenolic constituents were evaluated against human tumourigenic (HT-29, HCT-116, and CaCo-2) and non-tumourigenic (CCD-18Co) colon cells. Overall, maple syrup ethyl acetate (MS-EtOAc), butanol (MS-BuOH), and methanol (MS-MeOH) extracts were more active against the tumourigenic versus non-tumourigenic colon cells. At equivalent phenolic levels, the antiproliferative activities of grade D > C maple syrup, and MS-BuOH > MS-MeOH > MS-EtOAc. Among the isolates, gallic acid, catechaldehyde, syringaldehyde, and catechol were most active and their higher levels in grade D MS-BuOH extract could account for the highest observed anticancer effects of that extract. Moreover, the maple syrup extracts did not induce apoptosis of the colon cancer cells but induced cell cycle arrest which was also associated with a decrease in cyclins A and D1 levels. These results suggest that phenolics may impart potential biological effects to maple syrup.

The province of Quebec in Canada leads the world’s production of maple syrup, a natural sweetener obtained by thermal evaporation of sap collected from maple (Acer) species. As part of our laboratory’s detailed chemical investigation of Canadian maple syrup, a novel phenolic compound, 2,3,3-tri-(3-methoxy-4-hydroxyphenyl)-1-propanol, assigned the common name of quebecol, was obtained. Quebecol was isolated using a combination of chromatographic methods and identified by detailed 1D and 2D nuclear magnetic resonance (NMR) and mass spectral (MS) analyses. Liquid chromatography mass spectral (LC-MS) analyses revealed that quebecol is not originally present in maple sap. This observation, as well as the lack of a feasible biosynthetic pathway to explain its origin, suggests that quebecol is formed during the processing and/or extraction of maple syrup. Thus, the identification and biological evaluation of non-natural, process-derived compounds in maple syrup are warranted since such molecules may contribute towards the biological activities reported for this natural sweetener.

Phenolic-enriched extracts of Canadian maple syrup, namely ethyl acetate (MS-EtOAc) and butanol (MS-BuOH), were evaluated for their ability to inhibit carbohydrate hydrolyzing enzymes relevant to type 2 diabetes management. Extracts were standardized to phenolic contents by the Folin–Ciocalteau method and assayed for yeast α-glucosidase inhibitory activities. On normalization to phenolic content, MS-BuOH exhibited higher inhibitory activity than MS-EtOAc (IC50 = 68.38 and 107.9 μg phenolics, respectively). The extracts were further assayed for inhibition of porcine α-amylase and rat α-glucosidase enzymes. MS-BuOH exhibited higher rat α-glucosidase and porcine α-amylase inhibitory activities (IC50 = 135 and 103 μg phenolics, respectively) than MS-EtOAC extract (IC50 > 187 μg phenolics in both assays). These results suggest that maple syrup extracts may have potential for phenolic-mediated type 2 diabetes management, with the MS-BuOH phenolic-enriched fraction having highest bioactivity.

In this paper, we present a volume rendering framework for visualizing 3D flow fields. We introduce the concept of coherence field which evaluates the representativeness of a given streamline set for the underlying 3D vector field. Visualization of the coherence field can provide effective visual feedback to the user for incremental insertion of more streamline seeds. Given an initial set of streamlines, a coherence volume is constructed from a distance field to measure the similarity between the existing streamlines and those in their nearby regions based on the difference between the approximate and the actual vector directions. With the visual feedback obtained from rendering the coherence volume, new streamline seeds can be selected by the user or by a heuristic seed selection algorithm to adaptively improve the coherence volume. An improved volume rendering technique that can render user-defined appearance textures is proposed to facilitate macro-visualization of 3D vector fields.

Fulvestrant (ICI 182,780; ICI) is approved for the treatment of advanced metastatic breast cancer that is unresponsive to other endocrine therapies. Berries are frequently consumed for their antioxidant, anti-inflammatory, and anticancer potential. In this study, we tested the efficacy of two berry extracts (Jamun-EJAE and red raspberry-RRE) and their bioactive compounds (Delphinidin-Del and Ellagic acid-EA) to inhibit cell proliferation with or without a sublethal dose of ICI in various breast cancer cell lines.
