Abstract Background Hepatocellular carcinoma (HCC) is a common gastrointestinal malignancy with a high incidence and poor prognosis. The subunits of the integrator complex (INTS1-14) play a crucial role in regulating genes dependent on RNA Polymerase II, which may be associated with cancer. However, the role of INTSs in HCC remains unclear. This study aims to comprehensively analyze the clinical value and potential role of INTS family genes in HCC through systematic bioinformatics analysis. Methods We employed various public databases, including UALCAN, HPA, Kaplan–Meier Plotter, GEPIA2, TNMplot, STRING, TIMER, and TISIDB, to investigate the expression levels, clinicopathological correlations, diagnostic and prognostic value, genetic alterations, co-expression network, molecular targets, and immune infiltration of INTSs in HCC. Additionally, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were utilized to investigate the biological functions of genes associated with INTSs. Furthermore, Western blot, real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-qPCR), and immunohistochemistry techniques were employed to assess the expression of relevant proteins and genes. The proliferation of HCC cells was evaluated using the CCK8 assay. Results We found that in HCC, there was a significant upregulation of INTSs at the transcriptional level, particularly INTS1, INTS4, INTS7, and INTS8. Additionally, the protein levels of INTS1 and INTS8 were notably elevated. The overexpression of these INTSs was strongly correlated with tumor stages in HCC patients. INTS1, INTS4, INTS7, and INTS8 exhibited significant diagnostic and prognostic value in HCC. Moreover, their expression was associated with immune infiltrations and activated status, including B cells, CD8 + T cells, CD4 + T cells, NK cells, macrophages, and dendritic cells. Functional predictions indicated that INTS1, INTS4, INTS7, and INTS8 were involved in various cancer-related signaling pathways, such as TRAIL, IFN-gamma, mTOR, CDC42, Apoptosis, and the p53 pathway. Furthermore, we observed a significant upregulation of INTS1, INTS4, INTS7, and INTS8 expression in HCC cell lines compared to normal liver cell lines. The level of INTS1 protein was higher in cancerous tissues compared to adjacent non-cancerous tissues (n = 16), and the suppression of INTS1 resulted in a significant decrease in the proliferation of Huh7 cells. Conclusion These findings indicate the potential of INTS family genes as diagnostic biomarkers and therapeutic targets in HCC. Further research is needed to understand the underlying mechanisms and explore clinical applications.

This study undertakes a systematic analysis of the hydrological changes before and after the implementation of the Comprehensive Remediation Project in the lower reaches of the Ganjiang River. It focuses on changes in downstream inflow, ratios of flow distribution, and water levels, as well as water velocity near the gates. The results indicate a significant improvement in the spatial distribution of water resources in the lower reaches of the Ganjiang River. The project enhances the inflow from the northern and southern branches, positively influencing downstream water usage and the ecological environment. Building upon these findings, the study proposes operational recommendations tailored to different hydrological years, such as timely adjustments to the southern branch's water inflow and optimizing flow distribution ratios. This research provides a scientific basis for the implementation and dispatch of comprehensive remediation projects and offers insights into water resource management in similar regions. In order to meet the complex driving and braking conditions of automobiles, a highly efficient dual-rotor in-wheel motor (DRIWM) structure suitable for various driving and braking conditions of automobiles is proposed. To improve the energy recovery efficiency of variable voltage charging regenerative braking of DRIWM, a combination of two sets of power battery packs and switching modules is proposed. The regenerative braking control strategy of DRIWM based on rule base and maximum power is designed. Simulation results show that the control of variable torque coefficient based on rule base is better than the control of fixed torque coefficient on target regenerative braking torque. At the same time, the simulation results demonstrate that the energy recovered according to the respective ideal voltages of the internal and external motors is 5978 J. Compared with the results according to the ideal voltage of the internal and external motors, the recovered energy is improved around 3.67% and 27%, respectively. For the recovered energy, it is 5978 J. It is verified that the proposed variable voltage charging scheme based on dual power battery pack has advantages in energy recovery.

Purposes With the continuous promotion of the dual carbon goal, new energy has ushered in explosive development, and the capacity and space for new energy consumption have become a concern for all sectors of society. In the research on existing new energy consumption capacity and space, the consumption measures are relatively single and focus more on theoretical research, which is not applicable, propose specific methods and processes. Methods In this article the time series production simulation method was adopted to build a new energy consumption capacity and consumption space method and process for large-scale new energy grid connection on the Panoramic Power System Operation Simulation Analysis Platform. Findings The new energy consumption capacity of Shanxi Province during the 14th and 15th Five Year Plans was estimated. On the basis. Based on this, comprehensive consumption measures were taken to increase its new energy utilization rate to over 95%, improving the new energy consumption capacity. On the basis of a new energy utilization rate of 95%, the new energy consumption space under various scenarios was calculated for different proportion of newly added wind and solar power, different production timing, different new energy utilization rates, and different years, guiding the new energy planning of Shanxi Province and assisting in the implementation of the dual carbon target.

Dendritic fibrous nanosilica (DFNS) was functionalized using microcrystalline chitosan, derived from shrimp exoskeletons, to act as a robust anchor, resulting in DFNS@Chitosan. In order to prevent the restacking of chitosan sheets, the supramolecular polymerized chitosan not only served as a spacer but was also incorporated into cement-based composites. The physical-chemical characteristics of DFNS@Chitosan were assessed through various analytical techniques such as TEM, SEM, TGA, FTIR, AFM, XPS, and EDX. The potency and auto-induced contraction of Cement-based composite materials fortified with DFNS@Chitosan were probed. The incorporation of DFNS@Chitosan resulted in an increase in both compressive and interfacial stretching potency of the cement-based composites. Furthermore, the presence of DFNS@Chitosan effectively inhibited the occurrence of auto-induced contraction in the cement-based paste. This research endeavor is anticipated to promote an alternative utilization of DFNS and shrimp waste shells in the development of sustainable building materials.

ABSTRACTIdentification and the time since deposition (TsD) estimation of body fluid stains from a crime scene could provide valuable information for solving the cases and are always difficult for forensics. Microbial characteristics were considered as a promising biomarker to address the issues. However, changes in the microbiota may damage the specific characteristics of body fluids. Correspondingly, incorrect body fluid identification may result in inaccurate TsD estimation. The mutual influence is not well understood and limited the codetection. In the current study, saliva, semen, vaginal secretion, and menstrual blood samples were exposed to indoor conditions and collected at eight time points (from fresh to 30 days). High-throughput sequencing based on the 16S rRNA gene was performed to characterize the microbial communities. The results showed that a longer TsD could decrease the discrimination of different body fluid stains. However, the accuracies of identification still reached a quite high value even without knowing the TsD. Correspondingly, the mean absolute error (MAE) of TsD estimation significantly increased without distinguishing the types of body fluids. The predictive TsD of menstrual blood reached a quite low MAE (1.54 ± 0.39 d). In comparison, those of saliva (6.57 ± 1.17 d), semen (6.48 ± 1.33 d), and vaginal secretion (5.35 ± 1.11 d) needed to be further improved. The great effect of individual differences on these stains limited the TsD estimation accuracy. Overall, microbial characteristics allow for codetection of body fluid identification and TsD estimation, and body fluids should be identified before estimating TsD in microbiome-based stain analyses.IMPORTANCEEmerged evidences suggest microbial characteristics could be considered a promising tool for identification and time since deposition (TsD) estimation of body fluid stains. However, the two issues should be studied together due to a potential mutual influence. The current study provides the first evidence to understand the mutual influence and determines an optimal process for codetection of identification and TsD estimation for unknown stains for forensics. In addition, we involved aged stains into our study for identification of body fluid stains, rather than only using fresh stains like previous studies. This increased the predictive accuracy. We have preliminary verified that individual differences in microbiotas limited the predictive accuracy of TsD estimation for saliva, semen, and vaginal secretion. Microbial characteristics could provide an accurate TsD estimation for menstrual blood. Our study benefits the comprehensive understanding of microbiome-based stain analyses as an essential addition to previous studies.

To obtain lily polysaccharides efficiently and characterize the structure of polysaccharides, the polysaccharides from lily was extracted by using microwave-assisted aqueous two-phase extraction in this paper. The best extraction process was determined by single factor experiment and orthogonal experiment. The crude extract of lily polysaccharides was purified via DEAE-52 and Sephadex G-100 column chromatography in turn to obtain a single fraction (LP2-SG). The molecular weight and monosaccharide composition of LP2-SG were determined by high performance gel permeation chromatography and gas chromatography, respectively, and then its structure was preliminarily characterized. The results showed that the optimum extracting parameters to achieve the highest yield of polysaccharides 10.15%±0.11% from lily by microwave-assisted aqueous two-phase extraction was obtained under the microwave power of 400 W, ammonium sulfate mass fraction of 20%, ethanol volume fraction of 50%, and solid-to-liquid ratio of 1:30 g/mL. LP2-SG with a molecular weight of 530.51 kDa was composed of mannose, glucose, and galactose with a molar ratio of 11.63:29.85:8.46. LP2-SG has no characteristic absorption at 260 nm and 280 nm, and it had typical infrared absorption characteristics of polysaccharides. The results of this study provide an important reference for the efficient extraction and deep development of lily polysaccharides.

Abstract Background Effective pain management is closely related to nurses’ knowledge, attitudes and empathy regarding pain. Nursing educators and managers should understand the relationship between nurses’ pain management knowledge, attitudes and empathy level, and take targeted measures accordingly. Currently, there is limited study exploring the relationship between pain empathy and pain knowledge and attitudes among nurses in North China. Objectives The purpose of this study was to investigate the level of nurses’ pain management knowledge and attitudes and pain empathy, to analyze the factors influencing pain empathy, and to explore the relationship between these two variables. Design This study was a quantitative, descriptive-correlation design. Setting and participants The study population was registered nurses in North China, the sample included 177 registered nurses in North China. Methods Data were collected with the “General data questionnaire”, “Knowledge and attitudes survey regarding pain” (KASRP) and the “Empathy for pain scale” (EPS) via Wechat mini program “Questionnaire Star”. Results The 177 registered nurses completed the survey. The averege correct rate for KASRP was (51.94 ± 9.44)%, and none of the respondents achieved a percentage score of >80%. The mean score for pain empathy was (2.78 ± 0.78), the empathy reactions dimension was (2.99 ± 0.77), and the body and mind discomfort dimension was (2.71 ± 0.80). The results of multiple stepwise linear regression showed that whether they had received empathy training, whether they had greater trauma or severe pain and whether they had negative emotions were independent influencing factors for EPS scores. Abstract Aims Left ventricular (LV) remodelling after acute myocardial infarction (AMI) is associated with heart failure and increased mortality. There was no consensus on the definition of LV remodelling, and the prognostic value of LV remodelling with different definitions has not been compared. We aimed to find the optimal definition and develop a prediction nomogram as well as online calculator that can identify patients at risk of LV remodelling. Methods and results This prospective, observational study included 829 AMI patients undergoing percutaneous coronary intervention from January 2015 to January 2020. Echocardiography was performed within the 48 h of admission and at 6 months after infarction to evaluate LV remodelling, defined as a 20% increase in LV end‐diastolic volume (LVEDV), a 15% increase in LV end‐systolic volume (LVESV), or LV ejection fraction (LVEF)

Abstract Background Limited data exist regarding preoperative serum sodium (Na) and 30-day mortality in adult patients with tumor craniotomy. Therefore, this study investigates their relationship. Methods A secondary retrospective analysis was performed using data from the ACS NSQIP database (2012–2015). The principal exposure was preoperative Na. The outcome measure was 30-day postoperative mortality. Binary logistic regression modeling was conducted to explore the link between them, and a generalized additive model and smooth curve fitting were applied to evaluate the potential association and its explicit curve shape. We also conducted sensitivity analyses and subgroup analyses. Results A total of 17,844 patients (47.59% male) were included in our analysis. The mean preoperative Na was 138.63 ± 3.23 mmol/L. The 30-day mortality was 2.54% (455/17,844). After adjusting for covariates, we found that preoperative Na was negative associated with 30-day mortality. (OR = 0.967, 95% CI:0.941, 0.994). For patients with Na ≤ 140, each increase Na was related to a 7.1% decreased 30-day mortality (OR = 0.929, 95% CI:0.898, 0.961); for cases with Na > 140, each increased Na unit was related to a 8.8% increase 30-day mortality (OR = 1.088, 95% CI:1.019, 1.162). The sensitivity analysis and subgroup analysis indicated that the results were robust. Conclusions This study shows a positive and nonlinear association between preoperative Na and postoperative 30-day mortality in adult patients with tumor craniotomy. Appropriate preoperative Na management and maintenance of serum Na near the inflection point (140) may reduce 30-day mortality.

Passive vibration isolation techniques with low-frequency characteristics have been a hot topic in the aerospace field. A hydraulic inertial vibration isolator is a highly effective type of isolator for controlling low-frequency vibrations. It typically consists of a main spring, a minor spring, an inertial mass, and a fluid domain. Due to its multi-domain nature, analyzing the isolation mechanism of this type of isolator is challenging. The bond graph method is employed to establish the dynamic model of the isolator. Subsequently, the state equations of the isolator are derived, and the energy equations of both the mechanical and the fluid parts of the isolator are obtained. Based on this, the energy transfer characteristics between the mechanical and fluid domains inside the isolator under external excitation are discussed. The time-domain response of the forces transmitted to the foundation is analyzed. It is shown that the anti-resonance frequency occurs when the forces transmitted to the foundation generated by the main spring and the fluid pressure are equal to that of the minor spring. To verify the proposed method’s correctness, a prototype of the isolator is designed and a carefully designed experiment is conducted. The acceleration transmissibility of the isolator is used to conduct a comparative study. The results show that the theoretical results are in good agreement with the experimental results. To depict the dynamic characteristics of the isolator under large amplitude vibration, the nonlinear dynamic model of the isolator is developed, and the corresponding force transmissibility of the isolator is formulated. The energy flow between the mechanical and the fluid domains under this condition is also analyzed. The results indicate that the energy flow responses exhibit a similar change tendency to the force transmissibility. However, the peak of the energy ratio between the mechanical subsystem and the fluid is the same as the linear condition, suggesting that this value is determined by the amplification ratio of the isolator. This research provides enhanced physical insight to understand the dynamic characteristics of this type of isolator and will help to shorten the design cycle of the isolator.

The burning of Ethylene–Propylene–Diene Monomer (EPDM) rubber generates substantial smoke, posing a severe threat to the environment and personal safety. Considering the growing emphasis on safety and environmental protection, conventional non-smoke-suppressing flame retardants no longer satisfy the present application requirements. Consequently, there is an urgent need to develop a novel flame retardant capable of suppressing smoke formation while providing flame retardancy. Sepiolite (SEP), a porous silicate clay mineral abundant in silica and magnesium, exhibits notable advantages in the realm of flame retardancy and smoke suppression. This research focuses on the synthesis of two highly efficient flame-retardant smoke suppression systems, namely AEGS and PEGS, using Enteromorpha (EN), graphene (GE), sepiolite (SEP), ammonium polyphosphate (APP), and/or piperazine pyrophosphate (PPAP). The studied flame-retardant systems were then applied to EPDM rubber and the flame-retardant and smoke suppression abilities of EPDM/AEGS and EPDM/PEGS composites were compared. The findings indicate that the porous structure of sepiolite plays a significant role in reducing smoke emissions for EPDM composites during combustion.

Background Adenocarcinoma, the most prevalent histological subtype of non-small cell lung cancer, is associated with a significantly higher likelihood of bone metastasis compared to other subtypes. The presence of bone metastasis has a profound adverse impact on patient prognosis. However, to date, there is a lack of accurate bone metastasis prediction models. As a result, this study aims to employ machine learning algorithms for predicting the risk of bone metastasis in patients. Method We collected a dataset comprising 19,454 cases of solitary, primary lung adenocarcinoma with pulmonary nodules measuring less than 3 cm. These cases were diagnosed between 2010 and 2015 and were sourced from the Surveillance, Epidemiology, and End Results (SEER) database. Utilizing clinical feature indicators, we developed predictive models using seven machine learning algorithms, namely extreme gradient boosting (XGBoost), logistic regression (LR), light gradient boosting machine (LightGBM), Adaptive Boosting (AdaBoost), Gaussian Naive Bayes (GNB), multilayer perceptron (MLP) and support vector machine (SVM). Results The results demonstrated that XGBoost exhibited superior performance among the four algorithms (training set: AUC: 0.913; test set: AUC: 0.853). Furthermore, for convenient application, we created an online scoring system accessible at the following URL: https://www.xsmartanalysis.com/model/predict/?mid=731symbol=7Fr16wX56AR9Mk233917, which is based on the highest performing model. Conclusion XGBoost proves to be an effective algorithm for predicting the occurrence of bone metastasis in patients with solitary, primary lung adenocarcinoma featuring pulmonary nodules below 3 cm in size. Moreover, its robust clinical applicability enhances its potential utility.

The prevalence and mortality of cardiovascular disease are relatively high. Currently, depression has been proven to be an independent risk factor for the occurrence and poor prognosis of cardiovascular disease. Psycho-cardiovascular comorbidity, as a reciprocal cause and effect, affects each other, leading to the deterioration of clinical prognosis and forming a vicious circle. Coronary artery disease comorbidity with depression is a common disease in psycho-cardiology medicine. This paper expounds on the exploration of the treatment model of psycho-cardiology from the aspects of epidemiological characteristics, comorbidity mechanism, screening, diagnosis, and treatment.

Abstract Terrestrial ecosystems can exhibit various behaviors in response to climate change and human activities. Nonlinear and abrupt shifts in ecosystems are particularly important as they indicate substantial modifications in ecosystem structure and function, posing a threat to the provision of ecosystem services. Here we distinguish between linear, curvilinear, and abrupt shifts in ecosystem productivity from 2000 to 2020 in China's Loess Plateau. We utilize spatial Random Forest models to analyze the driving factors behind these change patterns. Our findings indicate that 84.1% of the ecosystems experienced a positive change in plant productivity, while a small proportion (2.5%) exhibited a negative change. Abrupt shifts are prevalent in both positive and negative changes in ecosystem productivity, with negative changes often manifesting as abrupt shifts (79.3%). Negative changes in plant productivity, particularly the negative abrupt shifts, are primarily associated with human activities characterized by increased nighttime light and urbanization. Land conversion to forest is linked to a curvilinear trajectory in plant productivity, characterized by nonlinear changes with acceleration. Higher water availability and a wetter environment are more likely to promote positive changes in plant productivity. Moderate warming trends contribute to abrupt positive changes in plant productivity, while high warming trends are associated with increased negative abrupt and curvilinear changes. We highlight the importance of accounting for diverse change behaviors in ecosystems for the development of targeted conservation and restoration measures.

Ecological restoration is the guarantee of regional ecological security. Accurate identification of restoration areas is crucial for planning ecological programs. However, the question of how to recognize the range of restoration areas at the pixel scale in order to improve restoration efficiency still requires further discussion. In this study, a framework that integrates ecological security and feasibility of restoration through multi-criteria decision analysis was developed to identify the ecological restoration priority areas (ERPAs) in the Qinghai-Tibet Plateau (QTP). The results showed that areas with low ecological security accounted for 58.33 % of the QTP, primarily concentrated in the northwest. Areas that were feasible to be restored, represented 26.11 % of the QTP. The relatively high feasibility areas (F > 0.6) were mainly located in the northern and southwestern of the QTP, covering 23.88 % of the total feasible areas. This study ultimately identified 6.69 × 105 km2 of ERPAs, encompassing 26.05 % of the QTP. The high-priority areas were concentrated in the southwest and scattered in the northern part of the study area, accounting for 21.47 % of the ERPAs. The low-priority areas with the largest proportion (57.77 % of ERPAs) were widely distributed in the eastern part of the study area. Therefore, more attention should be paid to the southwestern region of the QTP in the subsequent ecological restoration efforts. The framework, which takes into consideration ecological security and restoration feasibility, can significantly enhance the spatial accuracy of ecological restoration area identification, providing a practical reference for further environmental optimization.

Objective The study focuses on the development of critical elements in coal as a new area of mineral resource exploration. Methods Various analytical methods including X-ray diffraction, X-ray fluorescence spectrum, and inductively coupled plasma-mass spectrometry were used to investigate the mineralogy and geochemical characteristics of Early Permian middling coal in the southern margin of the North China Plate. Results The results indicate that the Early Permian coal in this region is mainly composed of minerals such as kaolinite, illite, tobelite, chlorite, and calcite, with SiO2, Al2O3, and CaO as the main elements. The coal seam samples also show enrichment of Li, with an average content of 76.83×10-6, as well as slight enrichment of Zr and Th. Other elements, such as Co, Zn, Rb, Cd, and Cs, are significantly depleted compared to the world average for hard coal. Conclusion Through correlation analysis and geochemical interpretation, it was concluded that the Li element in the Early Permian coal is mainly associated with clay minerals like kaolinite and is controlled by terrigenous debris. The uplift, weathering, and denudation of Mesoproterozoic K-feldspar granite and Carboniferous Benxi Formation palaeoweathering crust bauxite in the northern Yinshan ancient land, influenced by Variscan tectonic movement, became the primary source area for the Early Permian coal in the southern margin of the North China Plate. The salty and anoxic reducing sedimentary environment of the peat water medium favors the adsorption and deposition of Li by clay minerals, particularly kaolinite.

Abstract Background Glioblastoma (GBM) is a highly aggressive primary brain tumor with a poor prognosis. This study investigates the therapeutic potential of human Vγ9Vδ2 T cells in GBM treatment. The sensitivity of different glioma specimens to Vγ9Vδ2 T cell-mediated cytotoxicity is assessed using a patient-derived tumor cell clusters (PTCs) model. Methods The study evaluates the anti-tumor effect of Vγ9Vδ2 T cells in 26 glioma cases through the PTCs model. Protein expression of BTN2A1 and BTN3A1, along with gene expression related to lipid metabolism and glioma inflammatory response pathways, is analyzed in matched tumor tissue samples. Additionally, the study explores two strategies to re-sensitize tumors in the weak anti-tumor effect (WAT) group: utilizing a BTN3A1 agonistic antibody or employing bisphosphonates to inhibit farnesyl diphosphate synthase (FPPS). Furthermore, the study investigates the efficacy of genetically engineered Vγ9Vδ2 T cells expressing Car-B7H3 in targeting diverse GBM specimens. Results The results demonstrate that Vγ9Vδ2 T cells display a stronger anti-tumor effect (SAT) in six glioma cases, while showing a weaker effect (WAT) in twenty cases. The SAT group exhibits elevated protein expression of BTN2A1 and BTN3A1, accompanied by differential gene expression related to lipid metabolism and glioma inflammatory response pathways. Importantly, the study reveals that the WAT group GBM can enhance Vγ9Vδ2 T cell-mediated killing sensitivity by incorporating either a BTN3A1 agonistic antibody or bisphosphonates. Both approaches support TCR-BTN mediated tumor recognition, which is distinct from the conventional MHC-peptide recognition by αβ T cells. Furthermore, the study explores an alternative strategy by genetically engineering Vγ9Vδ2 T cells with Car-B7H3, and both non-engineered and Car-B7H3 Vγ9Vδ2 T cells demonstrate promising efficacy in vivo, underscoring the versatile potential of Vγ9Vδ2 T cells for GBM treatment. Conclusions Vγ9Vδ2 T cells demonstrate a robust anti-tumor effect in some glioma cases, while weaker in others. Elevated BTN2A1 and BTN3A1 expression correlates with improved response. WAT group tumors can be sensitized using a BTN3A1 agonistic antibody or bisphosphonates. Genetically engineered Vγ9Vδ2 T cells, i.e., Car-B7H3, show promising efficacy. These results together highlight the versatility of Vγ9Vδ2 T cells for GBM treatment.

Abstract Recent concurrent processes of vegetation greening and reduced resilience (the capacity to recover from disturbances) worldwide have brought many uncertainties into sustainable ecosystems in the future. However, little is known about the conditions and extent to which greening affects resilience changes. Here we assess both vegetation dynamics and resilience in China’s Loess Plateau from 2000 to 2020 using satellite-based vegetation data and an early warning indicator. Our results reveal an overall greening trend in vegetated areas, while resilience shifted from gains to losses at a breakpoint in 2010. Vegetation greening generally contributed to resilience gains, whereas increased temperature and precipitation variability contributed to the resilience loss observed in 2011–2020. These findings provide empirical evidence that vegetation greening does not necessarily correspond to an increase in resilience. We therefore recommend integrating resilience indicators into ecological restoration and conservation efforts to gain a more comprehensive understanding of vegetation states and support effective ecosystem stewardship.

Background: Excessive osteoclast formation disrupts bone homeostasis, thereby significantly contributing to pathological bone loss associated with a variety of diseases. Protein S-palmitoylation is a reversible post-translational lipid modification catalyzed by ZDHHC family of palmitoyl acyltransferases, which plays an important role in various physiological and pathological processes. However, the role of palmitoylation in osteoclastogenesis has never been explored. Consequently, it is unclear whether this process can be targeted to treat osteolytic bone diseases that are mainly caused by excessive osteoclast formation. Materials and methods: In this study, we employed acyl-biotin exchange (ABE) assay to reveal protein S-palmitoylation in differentiating osteoclasts (OCs). We utilized 2-bromopalmitic acid (2-BP), a pharmacological inhibitor of protein S-palmitoylation, to inhibit protein palmitoylation in mouse bone marrow-derived macrophages (BMMs), and tested its effect on receptor activator of nuclear factor κβ ligand (RANKL)-induced osteoclast differentiation and activity by TRAP staining, phalloidin staining, qPCR analyses, and pit formation assays. We also evaluated the protective effect of 2-BP against estrogen deficiency-induced bone loss and bone resorption in ovariectomized (OVX) mice using μCT, H&E staining, TRAP staining, and ELISA assay. Furthermore, we performed western blot analyses to explore the molecular mechanism underlying the inhibitory effect of 2-BP on osteoclastogenesis. Results: We found that many proteins were palmitoylated in differentiating OCs and that pharmacological inhibition of palmitoylation impeded RANKL-induced osteoclastogenesis, osteoclast-specific gene expression, F-actin ring formation and osteoclastic bone resorption in vitro, and to a lesser extent, osteoblast formation from MC3T3-E1 cells. Furthermore, we demonstrated that administration of 2-BP protected mice from ovariectomy-induced osteoporosis and bone resorption in vivo. Mechanistically, we showed that 2-BP treatment inhibited osteoclastogenesis partly by downregulating the expression of c-Fos and NFATc1 without overtly affecting RANKL-induced activation of osteoclastogenic AKT, MAPK, and NF-κB pathways. Conclusion: Pharmacological inhibition of palmitoylation potently suppresses RANKL-mediated osteoclast differentiation in vitro and protects mice against OVX-induced osteoporosis in vivo. Mechanistically, palmitoylation regulates osteoclast differentiation partly by promoting the expression of c-Fos and NFATc1. Thus, palmitoylation plays a key role in promoting osteoclast differentiation and activity, and could serve as a potential therapeutic target for the treatment of osteoporosis and other osteoclast-related diseases. The translational potential of this article: The translation potential of this article is that we first revealed palmitoylation as a key mechanism regulating osteoclast differentiation, and therefore provided a potential therapeutic target for treating osteolytic bone diseases.

Abstract Food safety risk management is an important cross-boundary issue around the world from both theoretical and practical perspectives. Public–public collaboration is the most basic form of cross-boundary governance. This study investigates the main factors affecting the governance effectiveness in public–public collaboration for food safety risk management through a questionnaire survey in Jiangsu Province and Guangxi Zhuang Autonomous Region, China, using a multivariable linear regression model based on principal component analysis. The results show that although the governance effectiveness in public–public collaboration is affected by many factors, the most important of which are the behavior and capabilities of legislative, administrative regulation and enforcement, and environmental improvement government (public) agencies, professionalism of government agencies, and laws and regulations as the basis for government agencies to perform their responsibilities. This research provides a case study for the academic community to better understand the main problems facing public–public collaboration for food safety risk management in China. It also provides insights into promoting public–public collaboration in developing countries.

Exploring the relationship and driving forces between supply–demand of ecosystem services (ESs) and urbanization can help solve the environmental problems and promote regional sustainable development. This study analyzed the spatio-temporal distribution characteristics of supply–demand of ESs and comprehensive urbanization level (CUL) in Shaanxi Province from 2010 to 2019 and assessed the coupling relationship between ecosystem service supply–demand ratio (ESSDR) and CUL using the coupling coordination degree (CCD) model. Random forests and geographically weighted regression methods were utilized to characterize the contribution and spatial distribution of the drivers of CCD. The results showed that: (1) except for habitat quality, the ESSDR of the other three types of services as well as the comprehensive services showed a decreasing trend, CUL exhibited increasing trend; (2) Although CCD was generally increasing, a significant portion (78.51%) of regions still remained uncoordinated, with relatively better coordination shown around the Guanzhong urban agglomeration, which has a higher urbanization level; (3) The CCD in Shaanxi Province was primarily influenced by local financial income, the secondary industry, and temperature forces. In regions with high and increasing CCD, the tertiary industry was the decisive force. In other areas, there were significant spatial variations in the driving forces. These findings provide a coupled and coordinated perspective for urban ecological management, which can provide scientific reference and practical guidance for cities with different development modes.

In order to address the issues of poor stability in vehicle speed and deteriorated spraying quality caused by changes in road slope and the decrease in overall mass due to liquid spraying, this study focuses on analyzing the structure and longitudinal dynamic characteristics of a 4WID high ground clearance self-propelled electric sprayer. By utilizing MATLAB/Simulink software, three subsystems, namely, the inverse longitudinal dynamics model, torque distribution model, and motor model, are established. The model takes into account the effects of longitudinal driving resistance, slope, and vehicle roll angle on the distribution of loads among the four wheels during slope driving. A seven-degrees-of-freedom dynamic model is developed. A hierarchical control structure is designed, incorporating an upper-level PID controller and a lower-level fuzzy PID controller, to control the overall system. The control algorithms are tailored to the specific characteristics of the sprayer’s operation, and simulation experiments are conducted under the corresponding operating conditions. Building upon this, a sensor-equipped experimental platform is set up in the self-propelled sprayer manufactured by the team in the preliminary stage. Real vehicle tests are conducted in two scenarios: transition transportation and field operations, with the evaluation of the overall vehicle speed serving as the performance metric to validate the correctness of the model and the control theory.

Chemotherapy resistance in cancer is an essential factor leading to high mortality rates. Tumor multidrug resistance arises as a result of the autophagy process. Our previous study found that compound 1-nitro-2 acyl anthraquinone-leucine (C2) exhibited excellent anti-colorectal cancer (CRC) activity involving autophagy and apoptosis-related proteins, whereas its underlying mechanism remains unclear. A notable aspect of this study is how C2 overcomes the multidrug susceptibility of HCT116/L-OHP, a colon cancer cell line that is resistant to both in vitro and in vivo oxaliplatin (trans-/-diaminocyclohexane oxalatoplatinum; L-OHP). In a xenograft tumor mouse model, we discovered that the mixture of C2 and L-OHP reversed the resistance of HCT116/L-OHP cells to L-OHP and inhibited tumor growth; furthermore, C2 down-regulated the gene expression levels of P-gp and BCRP and decreased P-gp’s drug efflux activity. It is important to note that while C2 re-sensitized the HCT116/L-OHP cells to L-OHP for apoptosis, it also triggered a protective autophagic pathway. The expression levels of cleaved caspase-3 and Beclin 1 steadily rose. Expression of PI3K, phosphorylated AKT, and mTOR were decreased, while p53 increased. We demonstrated that the anthraquinone derivative C2 acts as an L-OHP sensitizer and reverses resistance to L-OHP in HCT116/L-OHP cells. It suggests that C2 can induce autophagy in HCT116/L-OHP cells by mediating p53 and the PI3K/AKT/mTOR signaling pathway.

Insect development is intricately governed by hormonal signaling pathways, yet the pivotal upstream regulator that potentiates hormone activation remains largely elusive. The migratory locust, Locusta migratoria, exhibits population density-dependent phenotypic plasticity, encompassing traits such as flight capability, body coloration, and behavior. In this study, we elucidated a negative correlation between population density and ontogenetic development during the nymphal stage of locusts. We found that the level of density influences the developmental trajectory by modulating transcript abundance within the ecdysone signaling pathway, with knockdown of the prothoracicotropic hormone (PTTH) resulting in developmental delay. Transcriptomic analysis of locust brains across solitary and gregarious phases revealed significant differential expression of genes involved in various pathways, including protein synthesis, energy metabolism, hormonal regulation, and immunity. Notably, knockdown experiments targeting two energy regulators, adipokinetic hormone (AKH) and insulin-like polypeptide 1 (ilp1), failed to elicit changes in the developmental process in solitary locusts. However, knockdown of immunoglobulin (IG) significantly shortened the developmental time in higher-density populations. Collectively, our findings underscore the regulatory role of population density in determining developmental duration and suggest that an immune-related gene contributes to the observed differences in developmental patterns.

An aging society is coming in China with more and more serious problems such as osteoporosis and osteoarthritis. Glucosamine is one of important components from proteoglycan in human articular cartilage matrix, which can improve articular cartilage metabolism and promote bone and joint health. Commercially available glucosamine usually exists in the form of glucosamine hydrochloride, glucosamine sulfate and their double salts, and produced through acid and enzymatic hydrolysis and fermentation. Clinical studies have shown that it has high safety with no serious adverse reactions found and exhibits the health functions of promoting bone and joint health, antioxidation, regulating immunity, resisting bacteria and inflammation, etc. In health food, research in glucosamine is often focused on promoting bone health. In this article, the health functions, preparation techniques, safety and use of glucosamine are reviewed, while suggestions are given to improve the quality standard and production techniques due to no specific quality standard and standardized production process of glucosamine currently available, aiming at providing a reference for the development, supervision and regulation of glucosamine-based health foods.

CO2 geological utilization and storage (CGUS) is an important technology to achieve a deep cut of global CO2 emissions. CO2 leakage from the subsurface may impair the performance of CGUS projects, and the CO2 leakage through wellbores is the most common leakage pathway. This paper proposes a workflow for wellbore CO2 leakage risk management, and the workflow consists of the following steps: i) leakage risk identification; ii) leakage risk evaluation; iii) leakage risk monitoring; iv) leakage handling. Representative approaches in each step of the workflow are systematically reviewed. Key challenges of wellbore CO2 leakage risk management include: lack of effective detection and evaluation approaches to tackle the CO2 leakage risk induced by cement failure; lack of low-cost acid resistance alloys and CO2-resistant cement; lack of automated monitoring systems that could enable automated shutdowns of the wellbore whenever certain warning criteria are met.

Defects with varying geometric distribution in the rock mass play a crucial role in determining the stability of engineered rock masses. Previous studies have primarily investigated the initiation, propagation, and coalescence behaviors of flaws within rock masses. Nevertheless, there has been limited analysis of flawed rock masses under different loading rates. In this study, we present the deformation localization and cracking process of three types of inclined flawed sandstone specimens through conducting uniaxial compression and acoustic emission tests at four different levels of loading rate. The results supported the following findings: 1) Sandstone specimens with different flaw inclination angles exhibit the loading rate strengthening effect, and the strengthening effect gradually decreases with the increase of loading rate. 2) As the loading rate increases, the type of crack emergence changes from wing cracks to anti-tensile cracks, and the time of flaw initiation is shortened. 3) The cumulative acoustic emission counts were higher for the low-loading rate specimens than for the high-loading rate specimens. 4) Tensile cracks typically occur as the initial cracks. Anti-tensile cracks often coexist with wing cracks in rock specimens that have undergone tensile damage. Coplanar secondary cracks are the primary indication of shear damage formation in rock specimens. 5) The increase in loading rate promotes the transition of rock specimens from the mixed tensile-shear damage mode to the shear damage mode. These research results are of great theoretical significance and engineering value for understanding the failure mechanism of rock mass containing flaws and proposing effective measures to prevent cracking and ensure the safety of brittle solid structures.

Background High mortality rates are prevalent among patients with non-small-cell lung cancer (NSCLC), and effective therapeutic targets are key prognostic factors. Fascin actin-bundling protein 1 (FSCN1) promotes NSCLC; however, its role as an RNA-binding protein in NSCLC remains unexplored. Therefore, we aimed to explore FSCN1 expression and function in A549 cells. Method We screened for alternative-splicing events and differentially expressed genes (DEGs) after FSCN1 silence via RNA-sequencing (RNA-seq). FSCN1 immunoprecipitation followed by RNA-seq were used to identify target genes whose mRNA expression and pre-mRNA alternative-splicing levels might be influenced by FSCN1. Results Silencing FSCN1 in A549 cells affected malignant phenotypes; it inhibited proliferation, migration, and invasion, and promoted apoptosis. RNA-seq analysis revealed 2,851 DEGs and 3,057 alternatively spliced genes. Gene ontology-based functional enrichment analysis showed that downregulated DEGs and alternatively splicing genes were enriched for the cell-cycle. FSCN1 promoted the alternative splicing of cell-cycle-related mRNAs involved in tumorigenesis (i.e., BCCIP, DLGAP5, PRC1, RECQL5, WTAP, and SGO1). Combined analysis of FSCN1 RNA-binding targets and RNA-seq data suggested that FSCN1 might affect ACTG1, KRT7, and PDE3A expression by modulating the pre-mRNA alternative-splicing levels of NME4, NCOR2, and EEF1D, that were bound to long non-coding RNA transcripts (RNASNHG20, NEAT1, NSD2, and FTH1), which were highly abundant. Overall, extensive transcriptome analysis of gene alternative splicing and expression levels was performed in cells transfected with FSCN1 short-interfering RNA. Our data provide global insights into the regulatory mechanisms associated with the roles of FSCN1 and its target genes in lung cancer.

Background: The present investigation planned to study the cardiovascular worries in Rose Bengal (RB) induced thrombotic rats by analysing biochemical profile of plasma, platelet membrane and cardiac tissue. Methods: Male Wistar rats were categorized into two groups. The group I was Control (C) rats and received glucose, whereas group II was Rose Bengal (50 mg/kg) induced thrombotic rats. The animals sacrificed and biological samples collected were analysed for various biochemical parameters. Results: In rats RB induced heart damage/thrombosis showed significantly increased levels of lipid peroxidation (LPO), nitrate and nitrite. In RB administered group the antioxidant activities were lowered significantly i.e., reduced glutathione, glutathione S-transferase, glutathione peroxidase, superoxide dismutase and catalase when compared with control rats. Plasma enzymes SGOT, SGPT were decreased and Na+-K+ ATPase was significantly increased in group II in comparison with group I. There is no significant change in plasma hexokinase enzyme in RB administered rats when compared with control rats. The non-enzymatic and enzymatic antioxidants levels were decreased significantly in experimental groups II to compare with control group I rats. A significant increase in platelet membrane fluidity observed in group II rats when compared with group I rats in DPH and pyrene employed method. A significant raise in LPO of platelet membrane was observed in the experimental rats. Plasma nitrite and nitrate levels were increased in group II when compared to group I. Furthermore, the haematological parameters were also increased in group II when compared to group I rats. Conclusion: This investigation pointed out that the nitric oxide scavenging levels increased and might be protected the oxidative stress and free radicals generated by RB induced cardiac thrombosis in rats.

ObjectivePrimary adult choroid plexus carcinomas (PACPCs) are extremely rare brain tumors. The existing literature primarily comprises case reports, which limits our understanding of this uncommon disease. This study aims to describe the clinical characteristics and prognosis of PACPCs, as well as to identify optimal treatment strategies.MethodsWe conducted a comprehensive analysis of clinical data from 7 patients with PACPCs who underwent surgical treatment at the Department of Neurosurgery, Beijing Tiantan Hospital, between March 2011 and March 2023. Additionally, a thorough search of the PubMed database was performed using the keywords “choroid plexus carcinoma” or “choroid plexus carcinomas” within the time frame of August 1975 to April 2023, which yielded a total of 28 identified cases. Subsequently, we evaluated risk factors for progression-free survival (PFS) and overall survival (OS) based on the pooled cases.ResultsThe pooled cohort, consisting of 7 cases from our institution and 28 cases from the literature, included 20 males and 15 females with a mean age of 44.3 ± 14.7 years (range: 21-73 years). Gross-total resection (GTR) and non-GTR were achieved in 22 (62.9%) and 13 (37.1%) patients, respectively. Radiotherapy and chemotherapy were administered to 29 (90.6%) and 13 (40.6%) patients, respectively. After a mean follow-up of 21.0 ± 26.7 months (range: 2-132 months), 18 patients were alive, and 11 patients had died. The multivariate Cox regression model demonstrated that non-GTR (HR 5.262, 95% CI 1.350-20.516, p=0.017) was a negative prognostic factor for OS. However, we did not find any risk factors for PFS.ConclusionComplete surgical resection should be considered as the primary treatment approach for this rare disease. Chemotherapy and radiotherapy appear to have limited effectiveness in treating this condition. Further research with large cohorts is needed to validate our conclusions.

The tunnel collapse occurred during the construction of the Qingdao Metro Line Ⅰ, resulting in a pit measuring approximately 10 m in diameter and 7 m in depth on the ground surface. The collapse inflicted substantial economic losses, casualties, and subsequent disruptions to ongoing construction activities. To clarify the causes of this collapse and investigate the underlying factors, a comprehensive approach combining field surveys and numerical simulations was undertaken. The results show that the collapse was primarily attributed to inadequate support when facing the challenging geological conditions, poor surrounding rocks (Grade VI) and a high volume of groundwater. The combination of abundant water and sand seeped into the tunnel, culminating in a catastrophic ground collapse. Notably, the arch roof experienced extensive deformation and damage due to the significant seepage pressure, which was exacerbated by the absence of timely support. Then, ground grouting was promptly conducted to repair the affected area and reinforce the tunnel structure. Subsequent monitoring of surface deformations following the resumption of tunneling operations indicated the effectiveness of the disaster treatment approach. These achievements constitute a reliable theoretical foundation for similar projects, particularly in terms of environmental protection and ensuring safe construction practices.

Background: Periodontits (PD) and Alzheimer’s disease (AD) are both associated with ageing and clinical studies increasingly evidence their association. However, specific mechanisms underlying this association remain undeciphered, and immune-related processes are purported to play a signifcant role. The accrual of publicly available transcriptomic datasets permits secondary analysis and the application of data-mining and bioinformatic tools for biological discovery.Aim: The present study aimed to leverage publicly available transcriptomic datasets and databases, and apply a series of bioinformatic analysis to identify a robust signature of immune-related signature of PD and AD linkage.Methods: We downloaded gene-expresssion data pertaining PD and AD and identified crosstalk genes. We constructed a protein-protein network analysis, applied immune cell enrichment analysis, and predicted crosstalk immune-related genes and infiltrating immune cells. Next, we applied consisent cluster analysis and performed immune cell bias analysis, followed by LASSO regression to select biomarker immune-related genes.Results: The results showed a 3 gene set comprising of DUSP14, F13A1 and SELE as a robust immune-related signature. Macrophages M2 and NKT, B-cells, CD4+ memory T-cells and CD8+ naive T-cells emerged as key immune cells linking PD with AD.Conclusion: Candidate immune-related biomarker genes and immune cells central to the assocation of PD with AD were identified, and merit investigation in experimental and clinical research.

Conductive polymer polypyrrole (PPy)-coated lithium-rich manganese-based Li1.2Mn0.54Ni0.13Co0.13O2 (LMNCO) nanotube cathode materials were synthesized by electrospinning and subsequently subjected to low-temperature vapor-phase polymerization. X-ray diffraction (XRD), scanning electron microscopy (SEM) and high-resolution transmission electron microscopy (HRTEM) results confirm that the successful coating of the PPy layer (~2 nm) on the surface of LMNCO nanotubes did not destroy their morphologies or structures. Electrochemical tests indicate that the electrochemical performance of PPy-coated LMNCO nanotubes has been significantly enhanced. At a rate of 1 C, the discharge capacity of the PPy-coated LMNCO cell is 200.1 mAh g−1, and the capacity retention is 99% after 120 cycles. This excellent stability is attributed to the inhibition of side reactions and the protective function of the tubular structure due to the PPy coating layer. Additionally, the rate capability is also improved at a high current density due to the higher electronic and ionic conductivity.

Simultaneous implementation of myoelectric pattern recognition and muscle force estimation is highly demanded in building natural gestural interfaces but a challenging task due to the gesture classification accuracy degradation under varying muscle strengths. To address this problem, a novel method using transformer-based multi-task learning (MTL-Transformer) for the prediction of both myoelectric patterns and corresponding muscle strengths was proposed to describe the inherent characteristics of an individual gesture pattern under different force conditions, thereby improving the accuracy of myoelectric pattern recognition. In addition, the transformer model enabled the characterization of long-term temporal correlations to ensure precise and smooth estimation of the muscle force. The performance of the proposed MTL-Transformer framework was evaluated via experiments of classifying eleven hand gestures and estimating the corresponding muscle force simultaneously, using high-density surface electromyogram (HD-sEMG) recordings from forearm flexor muscles of eleven intact-limbed subjects. The MTL-Transformer framework yielded high classification accuracy (98.70±1.21%) and low root mean square deviation (12.59±2.76%), and outperformed other two common temporally modelling methods significantly ( ${p} < 0.05$ ) in terms of both improved gesture recognition accuracies and reduced muscle force estimation errors. The MTL-Transformer framework is demonstrated as an effective solution for simultaneous implementation of myoelectric pattern recognition and muscle force estimation. This study promotes the development of robust and smooth myoelectric control systems, with wide applications in gestural interfaces, prosthetic and orthotic control.

The energy-driven expansion of artificial surfaces has resulted in severe ecological problems. Scientific evaluation of regional ecological resilience under different scenarios is crucial for promoting ecological restoration. This study chose the Shanxi–Shaanxi–Inner Mongolia Energy Zone (SEZ) and modeled an ecological resilience evaluation based on resistance, adaptability, and recovery. Land-use change and ecological resilience from 1980 to 2020 were then analyzed. Moreover, the SEZ land-use patterns and ecological resilience in 2030 were simulated under business as usual (BAU), energy and mineral development (EMD), and ecological conservation and restoration (ECR) scenarios. The results showed that (1) the SEZ was dominated by cultivated land, grassland, and unused land. (2) Ecological resilience showed a changing trend of decreasing and then increasing, with high ecological resilience areas mainly located in the Yellow River Basin, whereas low ecological resilience areas spread outward from the central urban areas. (3) The ecological resilience level was the lowest under the EMD scenario and the highest under the ECR scenario. This study not only expands the analysis framework of ecological resilience research but also provides scientific support for ecological conservation in ecologically fragile areas with intensive human activity worldwide.

The performance of detonation engines depends on propellant injectors. This study investigates a fluidic-valve injector mounted to a detonation tube. The injector is equipped with a recessed cavity connecting to the fuel plenum. After verifying the theoretical and numerical framework, three cases (I, II, and III) are analyzed, each representing different combinations of initial injector conditions and fuel supply setups. In all cases, a detonation wave is initiated near the headend of the detonation tube. It propagates through the initial section of the tube and undergoes diffraction and deformation at the flush-wall orifice. Among the considered cases, Case III, featuring a pre-pressurized initial injector flowfield and a total-pressure-inlet boundary, demonstrates the best agreement with the experimental results. It reveals a strong interaction between the longitudinally traveling detonation wave and the transverse propellant plume expanding from the orifice, causing the detonation wave to split. One part continues within the tube, while the other diffracts into the injector, creating a recirculation zone. Shock waves propagate within the injector and reflect at the base of the cavity, generating pressure spikes similar to the experimental observations. However, the contact surface separating the burnt products and fresh propellant reaches only a limited distance into the injector, suggesting a short interruption time and rapid recovery of the propellant supply.

As a new concept for systematically evaluating ecosystem services, Gross Ecosystem Product (GEP) provides an effective means to comprehensively reveal the overall status of the ecosystem, the impact of economic activities on the ecological environment, and the effectiveness of ecological protection efforts. GEP accounting has been conducted in various regions; however, GEP’s application in natural reserves still requires further exploration. Taking the Qinling Mountains as the research area, this paper aims to assess the relationship between GEP and economic development on the basis of the GEP accounting system. The results indicated that: (1) From 2010 to 2020, GEP tended to increase continuously and exhibited a distribution pattern with high value regions in the east and west, and low value regions in the north and south. (2) Over the years, the coupling coordination degree between GEP and GDP was in a consistent upward trend. In 2020, a good coupling coordination state between GEP and GDP was achieved in most districts and counties. (3) With the relative development between GEP and GDP, the social economy of most districts and counties lagged behind GEP in 2010. The number of districts and counties lagging in GEP in 2020 increased, while the number of regions with a balanced development of GEP and GDP was still relatively discouraging. (4) In general, elevation, contagion, temperature, population density, and precipitation were the main drivers of coupling coordination degree between GEP and GDP. If the relationship between economic development and ecological environmental protection can be reasonably balanced, it will further promote the sustainable development of nature reserves, and provide a scientific basis for sustainable policy-making in other similar areas.

Efficiently mitigating the severe air pollution resulting from rapid progress is crucial for the sustainable development of the socio-ecological system. Recently, concerns about nature-based solutions have emerged in the research on the treatment of air pollution. Studies on the purification of PM2.5 using vegetation currently concentrate on the individual scale of tree species or urban vegetation, ignoring the regional scale, which could better assist ecological governance. Therefore, taking the Fenwei Plain of China as the study area, an assessment framework of the air purification service’s spatial distribution reflecting regional vegetation was constructed. The dry deposition model and GeoDetector were used to quantify the spatial-temporal pattern and explore natural driving factors on the removal of PM2.5. The results showed that (1) the PM2.5 purification services offered by various types of vegetation exhibit notable variations. The average removal rates of PM2.5 by vegetation were 0.186%, 0.243%, and 0.435% in 2000, 2010, and 2021, respectively. (2) Meanwhile, a wide range of spatial mismatch exists between the PM2.5 concentration and PM2.5 removal. Insufficient supply regions of PM2.5 purification services account for 50% of the Fenwei Plain. (3) PM2.5 removal was strongly influenced by the types of vegetation and the Normalized Vegetation Index (NDVI), followed by the Digital Elevation Model (DEM), and less affected by meteorological factors; a strong joint effect was shown among the factors. The findings in this research provide a new perspective on regional air pollution management at the regional scale.

The objective of this study was to investigate IL13Ra2 expression in brainstem glioma (BSG) and its correlation with key markers, functions, and prognostic implications, evaluating its therapeutic potential. A total of 80 tumor samples from BSG patients were analyzed. Multiplex immunofluorescence was used to examine six markers—IL13Ra2, H3.3K27M, CD133, Ki67, HLA-1, and CD4—establishing relationships between IL13Ra2 and these markers. Survival analysis, employing Kaplan–Meier and Cox proportional hazard regression models, encompassed 66 patients with complete follow-up. RNA-Seq data from a previously published study involving 98 patients were analyzed using the DESeq2 library to determine differential gene expression between groups. Gene Ontology (GO) enrichment and single-sample gene set enrichment analysis (ssGSEA) via the clusterProfiler library were used to delineate the gene functions of differentially expressed genes (DEGs). Nearly all the BSG patients displayed varying IL13Ra2 expression, with 45.0% (36/80) exhibiting over a 20% increase. Elevated IL13Ra2 levels were notably observed in pontine gliomas, diffuse intrinsic pontine gliomas (DIPGs), H3F3A-mutant gliomas, and WHO IV gliomas. IL13Ra2 expression was strongly correlated with H3.3K27M mutant protein, Ki67, and CD133. Patients with IL13Ra2 expression >20% showed shorter overall survival compared to those with ≤20% IL13Ra2 expression. The Cox proportional hazard regression model identified H3F3A mutations, rather than IL13Ra2 expression, as an independent prognostic factor. Analysis of RNA-Seq data from our prior cohort confirmed IL13Ra2’s correlation with H3.3, CD133, and Ki67 levels. Widespread IL13Ra2 expression in BSG, particularly elevated in the H3F3A mutant group, was strongly correlated with H3F3A mutations, increased proliferation, and heightened tumor stemness. IL13Ra2 represents a promising therapeutic target for BSGs, potentially benefiting patients with H3K27M mutations, DIPGs, WHO Grade IV, and pontine location-specific BSGs, particularly those with H3K27M mutations.

In the present study, an immersion experiment was carried out to examine how N80 steel corrodes when exposed to formation water containing dissolved CO2 and supercritical CO2 (Sc-CO2) along with water vapor. We employed electrochemical and surface analysis methods to examine the influence of various factors, including the temperature and duration of immersion, on the extent of corrosion. The results show that the corrosion patterns of N80 steel in a supercritical CO2 environment and CO2-saturated formation water differed significantly. The presence of similar corrosion features was suggested by the constant structure of the corrosion products identified in the formation water. However, the morphology of the corrosion product was complex in the supercritical CO2 environment, exhibiting features of pitting and localized corrosion. Furthermore, a non-linear trend in the corrosion rate was observed between 40 °C and 120 °C. Specifically, the rate of corrosion declined from 40 °C to 80 °C, but it then resumed its growth from 80 °C to 120 °C. These findings suggest that very high temperatures could lead to the destruction of corrosion products and subsequently enhance the corrosion process.

Background: Long-term ambient particulate matter (PM) exposure exerts detrimental effects on cardiovascular health. Evidence on the relation of chronically exposed ambient PM10 and PM2.5 with coronary stenosis remains lacking. Our aim was to investigate the association of PM10 and PM2.5 with coronary stenosis in patients undergoing coronary angiography. Methods: We performed a retrospective cohort study consisting of 7513 individuals who underwent coronary angiography in Fujian Province, China, from January 2019 to December 2021. We calculated a modified Gensini score (GS) to represent the degree of stenosis in coronary arteries by selective coronary angiography. We fitted linear regressions and logistic models to assess the association of PM10 and PM2.5 with coronary stenosis. We employed restricted cubic splines to describe the exposure-response curves. We performed mediation analyses to assess the potential mediators. Results: Long-term ambient PM10 and PM2.5 (prior three years average) exposure was significantly associated with the GS, with a breakpoint concentration of 47.5 μg/m3 and 25.8 μg/m3 for PM10 and PM2.5, respectively, above which we found a linear positive exposure-response relationship of ambient PM with GS. Each 10 µg /m3 increase in PM10 exposure (β: 4.81, 95 % CI: 0.44–9.19) and PM2.5 exposure [β: 10.50, 95 % CI: 3.14–17.86] were positively related to the GS. The adjusted odds ratio (OR) for each 10 µg/m3 increment in PM10 exposure on severe coronary stenosis was 1.33 (95 % CI: 1.04–1.76). Correspondingly, the adjusted OR for PM2.5 was 1.87 (95 % CI: 1.24–2.99). The mediation analysis indicated that the effect of PM10 on coronary stenosis may be partially mediated through total cholesterol, low-density lipoprotein cholesterol, apolipoprotein B, serum creatinine and blood urea nitrogen, and the effect of PM2.5 may be mediated in part by hemoglobin A1c. Conclusion: Our study provides the first evidence that chronic ambient PM10 and PM2.5 exposure was associated with coronary stenosis assessed by GS in patients with suspected coronary artery disease and reveals its potential mediators.

Neointimal hyperplasia is a complex process after vascular interventions, acute platelet deposition and smooth muscle cell proliferation both contributed to this process. There are still no perfect solutions to solve this problem. Rivaroxaban is a novel anticoagulant that has been widely used in clinic, it has a good pharmacological effects both in vivo and in vitro. Chitosan microparticle rapamycin (MP-rapa) was fabricated, interspaces of polyglycolic acid (PGA) scaffold were used as a reservoir of MP-rapa, and the scaffold was coated with hyaluronic acid rivaroxaban (MP-rapa-riva). Scanning electronic microscopy (SEM) photographs were taken and water contact angles were measured, rat inferior vena cava (IVC) patch venoplasty model was used; patches were harvested at day 14 and examined by immunohistochemistry and immunofluorescence. SEM photographs showed the microparticles rapamycin were inside the interspace of the scaffold, hyaluronic acid rivaroxaban was also successfully coated onto the surface of the scaffold. There was a thinner neointima, fewer proliferating cell nuclear antigen (PCNA) positive cells, fewer macrophages in the MP-rapa and MP-rapa-riva grafts compared to the control PGA graft. The result showed that this scaffold with dual anticoagulation and antiproliferation functions can effectively inhibit venous neointimal hyperplasia, although this is an animal experiment, it showed promising potential clinical application in the future.

Abstract Background Leber’s congenital amaurosis (LCA) is a severe hereditary retinopathy disease that is characterized by early and severe reduction of vision, nystagmus, and sluggish or absent pupillary responses. To date, the pathogenesis of LCA remains unclear, and the majority of cases are caused by autosomal recessive inheritance. In this study, we explored the variant in the Crumbs homologue 1 (CRB1) gene in a Chinese family with LCA. Methods We conducted comprehensive ocular examinations and collected 5 ml of blood samples from members of a Chinese family with LCA. A pathogenic variant was identified by capturing (the panel in NGS) and Sanger sequencing validation. Results A nonsense variant (c.1499C>G) in the 6th exon of CRB1 gene in a Chinese family with LCA was identified, which predicted a change in the protein p. S500*, may lead to loss of gene function. We summarized the 76 variants reported thus far in CRB1 that caused LCA8. Conclusions This study reported a novel variant c.1499C>G (p. S500*) of the CRB1 gene occurred in a Chinese family with LCA, thus expanding the spectrum of CRB1 variants causing LCA.

Abstract Background Patients with critical limb ischemia (CLI) are at great risk of major amputation and cardiovascular events. Adipose-derived mesenchymal stem cell (ADSC) therapy is a promising therapeutic strategy for CLI, but the poor engraftment and insufficient angiogenic ability of ADSCs limit their regenerative potential. Herein, we explored the potential of human umbilical vein endothelial cells (HUVECs)-derived small extracellular vesicles (sEVs) for enhancing the therapeutic efficacy of ADSCs in CLI. Results sEVs derived from hypoxic HUVECs enhanced the resistance of ADSCs to reactive oxygen species (ROS) and further improved the proangiogenic ability of ADSCs in vitro. We found that the hypoxic environment altered the composition of sEVs from HUVECs and that hypoxia increased the level of miR-486-5p in sEVs. Compared to normoxic sEVs (nsEVs), hypoxic sEVs (hsEVs) of HUVECs significantly downregulated the phosphatase and tensin homolog (PTEN) via direct targeting of miR-486-5p, therefore activating the AKT/MTOR/HIF-1α pathway and influencing the survival and pro-angiogenesis ability of ADSCs. In a hindlimb ischemia model, we discovered that hsEVs-primed ADSCs exhibited superior cell engraftment, and resulted in better angiogenesis and tissue repair. Conclusion hsEVs could be used as a therapeutic booster to improve the curative potential of ADSCs in a limb ischemia model. This finding offers new insight for CLI treatment. Graphical Abstract

The Brodmann area (BA)-based map is one of the most widely used cortical maps for studies of human brain functions and in clinical practice; however, the molecular architecture of BAs remains unknown. The present study provided a global multiregional proteomic map of the human cerebral cortex by analyzing 29 BAs. These 29 BAs were grouped into 6 clusters based on similarities in proteomic patterns: the motor and sensory cluster, vision cluster, auditory and Broca’s area cluster, Wernicke’s area cluster, cingulate cortex cluster, and heterogeneous function cluster. We identified 474 cluster-specific and 134 BA-specific signature proteins whose functions are closely associated with specialized functions and disease vulnerability of the corresponding cluster or BA. The findings of the present study could provide explanations for the functional connections between the anterior cingulate cortex and sensorimotor cortex and for anxiety-related function in the sensorimotor cortex. The brain transcriptome and proteome comparison indicates that they both could reflect the function of cerebral cortex, but show different characteristics. These proteomic data are publicly available at the Human Brain Proteome Atlas (www.brain-omics.com). Our results may enhance our understanding of the molecular basis of brain functions and provide an important resource to support human brain research.

Abstract We propose a scheme to construct predictive models for Hamiltonian matrices in atomic orbital representation from ab initio data as a function of atomic and bond environments. The scheme goes beyond conventional tight binding descriptions as it represents the ab initio model to full order, rather than in two-centre or three-centre approximations. We achieve this by introducing an extension to the atomic cluster expansion (ACE) descriptor that represents Hamiltonian matrix blocks that transform equivariantly with respect to the full rotation group. The approach produces analytical linear models for the Hamiltonian and overlap matrices. Through an application to aluminium, we demonstrate that it is possible to train models from a handful of structures computed with density functional theory, and apply them to produce accurate predictions for the electronic structure. The model generalises well and is able to predict defects accurately from only bulk training data.

PurposeHigh grade pleomorphic xanthoastrocytomas (HGPXAs) are very rare and their management and prognostic outcomes remain unclear. To better understand the disease, we aimed to evaluate the risk factors for progression-free survival (PFS) and overall survival (OS), and propose a treatment protocol based on cases from our institute and cases from the literature.MethodsThe authors reviewed the clinical data of 26 patients with HGPXAs who underwent surgical treatment in Department of Neurosurgery of Beijing Tiantan Hospital between August 2014 and September 2021. We also searched the PubMed database using the keywords “anaplastic” combined with “pleomorphic xanthoastrocytoma(s)” between January 1997 and October 2022. Risk factors for PFS and OS were evaluated in the pooled cases.ResultsThe authors’ cohort included 11 males and 15 females with a mean age of 36.7 ± 20.3 years (range: 5.5-71 years). Gross-total resection (GTR) and non-GTR were achieved in 17 (65.4%) and 9 (34.6%) patients, respectively. Radiotherapy and chemotherapy were administered to 22 and 20 patients, respectively. After a mean follow-up of 20.5 ± 21.2 months (range: 0.5-78.1 months), 7 patients suffered tumor recurrence and 6 patients died with a mean OS time of 19.4 ± 10.8 months (range: 8-36 months). In the literature between January 1997 and October 2022, 56 cases of HGPXAs were identified in 29 males and 27 females with a mean age of 29.6 ± 19.6 years (range; 4-74 years). Among them, 24 (44.4%) patients achieved GTR. Radiotherapy and chemotherapy was administered to 31 (62%) patients and 23 (46%) patients, respectively. After a median follow-up of 31.4 ± 35.3 months (range: 0.75-144 months), the mortality and recurrence rates were 32.5% (13/40) and 70% (28/40), respectively. Multivariate Cox regression model demonstrated that non-GTR (HR 0.380, 95% CI 0.174-0.831, p=0.015), age≥30 (HR 2.620, 95% CI 1.183-5.804, p=0.018), no RT (HR 0.334,95% CI 0.150-0.744, p=0.007) and no CT (HR 0.422, 95% CI 0.184-0.967, p=0.042) were negative prognostic factors for PFS. Non-GTR (HR 0.126, 95% CI 0.037-0.422, p=0.001), secondary HGPXAs (HR 7.567, 95% CI 2.221-25.781, p=0.001), age≥30 (HR 3.568, 95% CI 1.190-10.694, p=0.023) and no RT (HR 0.223,95% CI 0.073-0.681, p=0.008) were risk factors for OS.Conclusion:High grade pleomorphic xanthoastrocytomas are very rare brain tumors. Children and younger adults have better clinical outcome than elderly patients. Secondary HGPXAs had worse OS than primary HGPXAs. Complete surgical excision plus RT and CT is recommended for this entity. The frequency of BRAF mutations in HGPXAs is 47.5% (19/40) in this study, however, we do not find the connections between BRAF mutations and clinical outcomes. Future studies with larger cohorts are necessary to verify our findings.

Abstract Maize is a crop that is highly susceptible to the negative effects of low temperature. Low temperature can delay seed germination and cause a decrease in seed vigor, which seriously affects seedling emergence and yield. In this study, 190 maize accessions (inbred lines) with strong germination potential at normal temperature (25°C) were selected from more than 500 accessions to construct a new association mapping panel to further investigate germination under chilling stress (5°C). We re‐sequenced the genomes of the 190 diverse accessions and obtained 4,886,919 high‐quality SNPs. We then used this data to analyze population structure, perform principal components analysis, and construct a phylogenetic tree of the new maize panel. The relative germination rate (RGR) and relative germination index (RGI) are two traits that are significantly related to chilling‐tolerant germination. Genome‐wide association analysis showed that RGR and RGI shared a major QTL, and they also shared the top SNP. There were a total of 26 significant SNPs in common. These SNPs hit directly or indirectly within 37 candidate genes. Among these 37 gene candidates, eight are homologs of genes previously reported to be related to both germination and low‐temperature stress, and another 12 genes related to low‐temperature stress or other abiotic stresses such as drought, salinity, oxidative, and high light stress. In addition, RGR and RGI had another 15 and 26 significant SNPs, respectively, which were associated with 17 and 92 candidate genes, respectively. Further qRT‐PCR analysis using 26 chilling‐tolerant and 22 chilling‐sensitive accessions implied that Zm00001eb272370, Zm00001eb272390, and Zm00001eb272400 associated with the top SNP, may play different roles during cold‐germination. Thus, our study not only established a new association mapping panel suitable for investigation of germination at low temperature but also provided valuable genetic resources for future studies to improve chilling‐tolerant maize varieties.
