Encouraging and astonishing developments have recently been achieved in image-based diagnostic technology. Modern medical care and imaging technology are becoming increasingly inseparable. However, the current diagnosis pattern of signal to image to knowledge inevitably leads to information distortion and noise introduction in the procedure of image reconstruction (from signal to image). Artificial intelligence (AI) technologies that can mine knowledge from vast amounts of data offer opportunities to disrupt established workflows. In this prospective study, for the first time, we develop an AI-based signal-to-knowledge diagnostic scheme for lung nodule classification directly from the computed tomography (CT) raw data (the signal). We find that the raw data achieves almost comparable performance with CT, indicating that it is possible to diagnose diseases without reconstructing images. Moreover, the incorporation of raw data through three common convolutional network structures greatly improves the performance of the CT models in all cohorts (with a gain ranging from 0.01 to 0.12), demonstrating that raw data contains diagnostic information that CT does not possess. Our results break new ground and demonstrate the potential for direct signal-to-knowledge domain analysis.

This study investigated the impact of Human Papillomavirus (HPV) on inflammation and growth in oral epithelial cells, with a focus on the role of Interleukin-37 (IL37). Oral epithelial cells, including HOEC and HSC-3 cells, were employed in the research. The results revealed that HPV significantly induced inflammation in both types of oral epithelial cells, concurrently promoting cell growth and inhibiting apoptosis. IL37, a cytokine, was found to mitigate HPV-induced inflammation in oral epithelial cells. Moreover, IL37 counteracted HPV's effects on apoptosis and cell viability in oral epithelial cells. The study also identified a reduction in autophagy in HPV-infected oral epithelial cells, a phenomenon alleviated by IL37. Furthermore, chemical inhibition of autophagy was observed to attenuate HPV-induced inflammation and growth in oral epithelial cells. These findings contribute valuable insights into the pathogenesis of inflammation in oral epithelial cells associated with HPV and oral cancers, offering potential avenues for novel therapeutic strategies.

The tumor-associated macrophages (TAM) play a crucial role in lung adenocarcinoma (LUAD), which can cause the proliferation, migration and invasion of tumor cells. In particular, TAMs mainly regulate changes in the tumor microenvironment thereby contributing to tumorigenesis and progression. Recently, an increasing number of studies are using single-cell RNA (Sc-RNA) sequencing to investigate changes in the composition and transcriptomics of the tumor microenvironment. We obtained Sc-RNA sequencing data of LUAD from GEO database and transcriptome data with clinical information of LUAD patients from TCGA database. A group of important genes in the state transition of TAMs was identified by analyzing TAMs at the single-cell level, while 5 TAM-related prognostic genes were obtained by omics data integration, and a prognostic model was constructed. GOBP analysis revealed that TAM-related genes were mainly enriched in tumor-promoting and immunosuppression-related pathways. After ROC analysis, it was found that the AUC of the prognosis model reached 0.751, with well predictive effectiveness. The 5 unique genes, HLA-DMB, HMGN3, ID3, PEBP1, and TUBA1B, was finally identified through synthesized analysis. The transcriptional characteristics of 5 genes were determined through GEPIA2 database and RT-qPCR. The increased expression of TUBA1B in advanced LUAD may serve as a prognostic indicator, while low expression of PEBP1 in LUAD may have the potential to become a therapeutic target.

Glycyrrhiza (Fabaceae) species are rich in metabolites and widely used in medicine. Research on the chloroplast genome of Glycyrrhiza is important for understanding its phylogenetics, biogeography, genetic diversity, species identification, and medicinal properties. In this study, comparative genomics and phylogenomics of Glycyrrhiza were analyzed based on the chloroplast genome. The chloroplast genomes of six Glycyrrhiza species were obtained using various assembly and annotation tools. The final assembled chloroplast genome sizes for the six Glycyrrhiza species ranged from 126,380 bp to 129,115 bp, with a total of 109–110 genes annotated. Comparative genomics results showed that the chloroplast genomes of Glycyrrhiza showed typically lacking inverted repeat regions, and the genome length, structure, GC content, codon usage, and gene distribution were highly similar. Bioinformatics analysis revealed the presence of 69–96 simple sequence repeats and 61–138 long repeats in the chloroplast genomes. Combining the results of mVISTA and nucleotide diversity, four highly variable regions were screened for species identification and relationship studies. Selection pressure analysis indicated overall purifying selection in the chloroplast genomes of Glycyrrhiza, with a few positively selected genes potentially linked to environmental adaptation. Phylogenetic analyses involving all tribes of Fabaceae with published chloroplast genomes elucidated the evolutionary relationships, and divergence time estimation estimated the chronological order of species differentiations within the Fabaceae family. The results of phylogenetic analysis indicated that species from the six subfamilies formed distinct clusters, consistent with the classification scheme of the six subfamilies. In addition, the inverted repeat-lacking clade in the subfamily Papilionoideae clustered together, and it was the last to differentiate. Co-linear analysis confirmed the conserved nature of Glycyrrhiza chloroplast genomes, and instances of gene rearrangements and inversions were observed in the subfamily Papilionoideae.

The exploration of unleaded free-cutting Cu40Zn brass with excellent mechanical and tribological properties has always drawn the attention of researchers. Due to its attractive properties combining metals and ceramics, Ti3AlC2 was added to Cu40Zn brass using high-energy milling and hot-pressing sintering. The effects of Ti3AlC2 on the microstructure, mechanical and tribological properties of Cu40Zn-Ti3AlC2 composites were studied. The results showed that Ti3AlC2 could suppress the formation of ZnO by adsorbing oxygen impurity and promote the formation of the β phase by releasing the β-forming element Al to the substrate. The hardness and wear resistance of Cu40Zn-Ti3AlC2 composites increased with increasing Ti3AlC2 content from 0 to 5 wt.%. The proper Ti3AlC2 additive was beneficial to both the strength and plasticity of the composites. The underlying mechanisms were discussed.

Interfacial solar water evaporation has emerged as an effective technique for converting solar energy to thermal energy, which can then be applied to clean water production and sewage treatment. This review discusses the primary characterization techniques used to evaluate the key aspects of interfacial solar water evaporators: the intrinsic and performance evaluation. Each technique is discussed with a brief explanation of its foundations, followed by carefully selected examples. This review is aimed at assisting materials chemists and physicists, particularly students, in choosing the most appropriate techniques for characterizing photothermal materials.

Abstract Background C-X-C motif chemokine ligand 9 (CXCL9), which is involved in the pathological processes of various human cancers, has become a hot topic in recent years. We developed a radiomic model to identify CXCL9 status in ovarian cancer (OC) and evaluated its prognostic significance. Methods We analyzed enhanced CT scans, transcriptome sequencing data, and corresponding clinical characteristics of CXCL9 in OC using the TCIA and TCGA databases. We used the repeat least absolute shrinkage (LASSO) and recursive feature elimination(RFE) methods to determine radiomic features after extraction and normalization. We constructed a radiomic model for CXCL9 prediction based on logistic regression and internal tenfold cross-validation. Finally, a 60-month overall survival (OS) nomogram was established to analyze survival data based on Cox regression. Results CXCL9 mRNA levels and several other genes involving in T-cell infiltration were significantly relevant to OS in OC patients. The radiomic score (rad_score) of our radiomic model was calculated based on the five features for CXCL9 prediction. The areas under receiver operating characteristic (ROC) curves (AUC-ROC) for the training cohort was 0.781, while that for the validation cohort was 0.743. Stimulus-responsive materials are fundamental to the broad and ever-growing field of intelligence research, which bridge intelligent systems with the Internet of Things (IoT) in future lifestyles. Among these materials, writable materials have received great interest; however, carbonization and irreversible writing processes are generally inevitable for extensively investigated organic compounds. Photochromism is a potential mode of composing information. Nevertheless, inorganic materials usually exhibit weak photochromic effects. Here, a novel strategy of designing high-entropy perovskite (HEP) oxides is put forward to develop a new inorganic photochromic system with satisfying performance. A series of HEP oxides are synthesized for the first time. Benefiting from excellent photochromic features, real-time information encoding was achieved. The mechanism-related photochromism is also discussed. Distinct from the previous works, it is believed that the present photochromic-based HEP oxides provide a new and manyfold research space for the future development of conventional writable materials and the disclosing of unprecedented properties and phenomena.

Abstract With the widespread use of highly active antiretroviral therapy (HARRT), the survival time of AIDS patients has been greatly extended. However, the incidence of lung cancer in HIV‐infected patients is increasing and has become a major problem threatening the survival of AIDS patients. The aim of this study is to use Weighted Gene Co‐expression Network Analysis (WGCNA) and differential gene analysis to find possible key genes involved in HIV‐infected lung cancer. In this study, using lung tissue samples from five pairs of HIV‐infected lung cancer patients, second‐generation sequencing was performed and transcriptomic data were obtained. A total of 132 HIV‐infected lung cancer‐related genes were screened out by WGCNA and differential gene expression analysis methods. Based on gene annotation analysis, these genes were mainly enriched in mitosis‐related functions and pathways. In addition, in protein–protein interaction (PPI) analysis, a total of 39 hub genes were identified. Among them, five genes (ASPM, CDCA8, CENPF, CEP55, and PLK1) were present in both three hub gene lists (intersection gene, DEGs, and WCGNA module) suggesting that these five genes may become key genes involved in HIV‐infected lung cancer.

Abstract Background Sorafenib resistance is a key impediment to successful treatment of patients with advanced hepatocellular carcinoma (HCC) and recent studies have reported reversal of drug resistance by targeting ferroptosis. The present study aimed to explore the association of fatty acid synthase (FASN) with sorafenib resistance via regulation of ferroptosis and provide a novel treatment strategy to overcome the sorafenib resistance of HCC patients. Methods Intracellular levels of lipid peroxides, glutathione, malondialdehyde, and Fe2+ were measured as indicators of ferroptosis status. Biological information analyses, immunofluorescence assays, western blot assays, and co-immunoprecipitation analyses were conducted to elucidate the functions of FASN in HCC. Both in vitro and in vivo studies were conducted to examine the antitumor effects of the combination of orlistat and sorafenib and CalcuSyn software was used to calculate the combination index. Results Solute carrier family 7 member 11 (SLC7A11) was found to play an important role in mediating sorafenib resistance. The up-regulation of FASN antagonize of SLC7A11-mediated ferroptosis and thereby promoted sorafenib resistance. Mechanistically, FASN enhanced sorafenib-induced ferroptosis resistance by binding to hypoxia-inducible factor 1-alpha (HIF1α), promoting HIF1α nuclear translocation, inhibiting ubiquitination and proteasomal degradation of HIF1α, and subsequently enhancing transcription of SLC7A11. Orlistat, an inhibitor of FASN, with sorafenib had significant synergistic antitumor effects and reversed sorafenib resistance both in vitro and in vivo. Conclusion Targeting the FASN/HIF1α/SLC7A11 pathway resensitized HCC cells to sorafenib. The combination of orlistat and sorafenib had superior synergistic antitumor effects in sorafenib-resistant HCC cells.

Composite helical springs (CHSs) are mainly used in transportation and aerospace fields, such as automobile suspension, railway bogie and aircraft engine systems. It has become a trend to replace the traditional metal helical springs with CHSs with the advantage of energy conservation during service and emission reduction during manufacturing. The advantages of CHSs such as low weight, high specific strength, high specific modulus, corrosion resistance, fatigue resistance and high strain energy storage capacity mean that it has great development potential. The static and dynamic performances of CHSs together determine whether they can be used on a large scale in the engineering field. The static performance of the CHS determines their service-load range and the dynamic performance determines the service life and safety performance of CHSs under dynamic load environmental conditions. Therefore, it is an important task to optimize the static and dynamic performance of CHSs. To provide a reliable reference for the development of CHSs, this review analyzes the important factors affecting the static and dynamic performance optimization of CHSs from the perspectives of theory, finite element method (FEM), and experiment. In addition, the outlook in the research of CHSs are discussed.

4-Methylcatechol (4-MC) is an agonist of various neurotrophic factors, which can upregulate the expression of Heme oxygenase 1 (HO-1) protein by activating nuclear factor erythroid 2-related factor 2 (Nrf2), thereby inhibiting oxidative stress-induced neural stem cell death. During RANKL-stimulated osteoclast differentiation, intracellular reactive oxygen species (ROS) levels were increased. Nonetheless, the effect of 4-MC on osteoclast formation and bone resorption function has not been researched. In this study, we investigated the effect of HO-1 upregulation by 4-MC on RANKL-induced osteoclastogenesis and explored the molecular mechanism of HO-1 upregulation by 4-MC. We found that the small molecule compound 4-MC could bind to Keap1 amino acid residue of glycine GLY 367, isoleucine ILE 559 and valine VAL 606, with a predicted binding energy of −4.99 kcal/mol. 4-MC was found to inhibit osteoclast differentiation in vitro by activating Nrf2 to scavenge ROS, inhibiting NF-κB phosphorylation, and alleviating osteoporosis in ovariectomized (OVX) mice. Taken together, 4-MC reduces ROS by inhibiting Keap1, thereby preventing OVX-induced bone loss.

A novel composite floor consisting of an upper chord of a grouted square steel tube truss and a lower chord of a reinforced concrete slab is proposed to address existing problems with precast composite floors, such as a complex construction process, high cost, significant resource consumption, and severe environmental pollution. Sixteen-point loading tests were carried out on five simply supported one-way slabs to simulate the stress state of the floor under a uniform load and to investigate the stiffness, cracking moment, load-carrying capacity, and failure mode of the composite floor system. The results showed that the composite floor system exhibited ductile failure, with cracks uniformly distributed at the bottom of the slab; additionally, the grouted steel tube did not experience uplift or out-of-plane buckling, and the truss welds did not fracture, indicating that the composite floor system still had further deformation capacity and load-carrying capacity with satisfactory stress conditions. After unloading, the residual deformation of the composite floor accounted for 28% to 36% of the maximum deflection, demonstrating good deformation recovery ability. The bottom slab thickness and truss height are key parameters that influence the new composite floor, and increasing the two parameters enhances the cracking load, deformation resistance, and flexural capacity of the composite floor, with a significant improvement achieved by increasing the truss height. The floor slab should have a minimum thickness of 60 mm, as required by China’s Code for Design of Concrete Structures, and the truss height-to-span ratio should be 1:30, which meets the deformation and load-carrying requirements of the floor and conforms to the design concept of green buildings.

Introduction: Colorectal cancer (CRC) is the fourth most common cancer worldwide, with high morbidity and mortality rates. In recent years, high-fat diet has been shown to increase CRC morbidity, highlighting the possibility of the application of hypolipidemic drugs for CRC treatment. In this study, we preliminarily evaluated the effects and mechnisms of ezetimibe against CRC through the blockage of lipid absorption in small intesine.Methods: In this study, CRC cell proliferation, invasion, apoptosis, and autophagy were evaluated using cellular and molecular assays. Fluorescent microscopy, and a flow cytometric assay were used to assess mitochondrial activity in vitro. A subcutaneous xenograft mouse model was used to evaluate the effects of ezetimibe in vivo.Results: We found that ezetimibe inhibited CRC cell proliferation, and migration, and facilitated autophage-associated apoptosis in HCT116 and Caco2 cells. Ezetimibe-induced mitochondrial dysfunction in CRC cells was found to be correlated with mTOR signaling activity.Discussion: Ezetimibe exhibits effects against CRC through the promotion of cancer cell death via mTOR signaling-dependent mitochondrial dysfunction, highlighting its potential value in CRC therapy.

A radar absorbing material named waffle structured composite (WSC) was proposed based on the periodic structure element formed by one-step double-sided knitting process with the advantage of continuous manufacture. The unique waffle structured pattern shaped by the knitted loops achieved the integration of structure and function. By realizing the collaborative mechanisms of absorption and scattering, the radar cross section (RCS) below −10 dB was achieved in the range of 6–8 GHz and 12–18 GHz towards the wale and course directions, respectively, and RCS reduction peak reaches to −30 dB. Through the S-parameter analysis and electromagnetic simulation, it can be revealed that the low-frequency RCS reduction in the wale direction was mainly dominated by the absorption mechanism, and the high-frequency RCS reduction in the course direction was led by the scattering mechanism. Furthermore, to further explore the scattering and absorption mechanism, phase difference, dielectric loss and magnetic loss were investigated in details. It was verified that the absorption of WSC is mainly originated from dielectric loss with slight magnetic loss, and Debye relaxation plays the important role in dielectric loss. The WSC proposed in this paper is equipped with characteristics of good RCS reduction and continuous large-scale manufacture, which is promising in practical engineering application of radar stealth.

Abstract Primary liver cancer (PLC) is a common malignancy with high morbidity and mortality. Poor prognosis and easy recurrence on PLC patients calls for optimizations of the current conventional treatments and the exploration of novel therapeutic strategies. For most malignancies, including PLC, immune cells play crucial roles in regulating tumor microenvironments and specifically recognizing tumor cells. Therefore, cellular based immunotherapy has its instinctive advantages in PLC therapy as a novel therapeutic strategy. From the active and passive immune perspectives, we introduced the cellular based immunotherapies for PLC in this review, covering both the lymphoid and myeloid cells. Then we briefly review the combined cellular immunotherapeutic approaches and the existing obstacles for PLC treatment.

Abstract Plants belonging to family Paeoniaceae are not only economically important ornamental plants but also medicinal plants used as an important source of traditional Chinese medicine. Owing to the complex network evolution and polyploidy evolution of this family, its systematics and taxonomy are controversial and require a detailed investigation. In this study, three complete chloroplast genomes of sect. Paeonia, one of the sections of Paeonia, were sequenced and then analysed together with 16 other published chloroplast genomes of Paeoniaceae species. The total lengths of the chloroplast genomes of these species were 152,153–154,405 bp. A total of 82–87 protein-coding genes, 31–40 tRNA genes and 8 rRNA genes were annotated. Bioinformatics analysis revealed 61–74 simple sequence repeats (SSRs) in the chloroplast genomes, most of which have A/T base preference. Codon usage analysis showed that A/U-ending codons were more positive than C/G-ending codons, and a slight bias in codon usage was observed in these species. A comparative analysis of these 19 species of Paeoniaceae was then conducted. Fourteen highly variable regions were selected for species relationship study. Phylogenetic analysis revealed that the species of sect. Paeonia gathered in one branch and then divided into different small branches. P. lactiflora, P. anomala, P. anomala subsp. veitchii and P. mairei clustered together. P. intermedia was related to P. obovata and P. obovata subsp. willmottiae. P. emodi was the sister to all other species in the sect. Paeonia.

Osteoporosis affects around 200 million people globally, with menopausal women accounting for the bulk of cases. In the occurrence and development of osteoporosis, a key role is played by osteoclasts. Excessive osteoclast-mediated bone resorption activity reduces bone mass and increases bone fragility, resulting in osteoporosis. Thus, considerable demand exists for designing effective osteoporosis treatments based on targeting osteoclasts. Eltanexor (Elt; KPT-8602) is a selective nuclear-export inhibitor that covalently binds to and blocks the function of the nuclear-export protein exportin-1 (XPO1), which controls the nucleus-to-cytoplasm transfer of certain critical proteins related to growth regulation and tumor suppression, such as p53, IκBα [nuclear factor-κB (NF-κB) inhibitor α] and FOXO1; among these proteins, IκBα, a critical component of the NF-κB signaling pathway that primarily governs NF-κB activation and transcription. How Elt treatment affects osteoclasts remains poorly elucidated. Elt inhibited the growth and activity of RANKL-induced osteoclasts in vitro in a dose-dependent manner, and Elt exerted no cell-killing effect within the effective inhibitory concentration. Mechanistically, Elt was found to trap IκBα in the nucleus and thus protect IκBα from proteasome degradation, which resulted in the blocking of the translocation of IκBα and NF-κB p65 and the consequent inhibition of NF-κB activity. The suppression of NF-κB activity, in turn, inhibited the activity of two transcription factors (NFATc1 and c-Fos) essential for osteoclast formation and led to the downregulation of genes and proteins related to bone resorption. Our study thus provides a newly identified mechanism for targeting in the treatment of osteoporosis.

Abstract The conventional method used to obtain a tumor biopsy for hepatocellular carcinoma (HCC) is invasive and does not evaluate dynamic cancer progression or assess tumor heterogeneity. It is thus imperative to create a novel non-invasive diagnostic technique for improvement in cancer screening, diagnosis, treatment selection, response assessment, and predicting prognosis for HCC. Circulating tumor DNA (ctDNA) is a non-invasive liquid biopsy method that reveals cancer-specific genetic and epigenetic aberrations. Owing to the development of technology in next-generation sequencing and PCR-based assays, the detection and quantification of ctDNA have greatly improved. In this publication, we provide an overview of current technologies used to detect ctDNA, the ctDNA markers utilized, and recent advances regarding the multiple clinical applications in the field of precision medicine for HCC.

The protein phosphatase 2C (PP2C) gene family, which is known to participate in cellular processes, is one of the most conserved plant-specific gene families that regulate signal transduction in eukaryotic organisms. However, the PP2C gene family of Glycyrrhiza uralensis Fisch. has not yet been systematically characterized. In this study, 76 PP2C genes of G. uralensis (GuPP2Cs) were identified. Phylogenetic analysis divided these GuPP2Cs into 12 subfamilies. The gene structure and protein motifs of the GuPP2Cs were then analysed. The analysis of the transcriptomic data of the GuPP2Cs revealed that nine genes were highly sensitive to abscisic acid (ABA) stress and might respond to ABA treatment. These nine genes were validated via quantitative real-time polymerase chain reaction. This study provides a foundation for future studies on the functional PP2C genes of G. uralensis and the improvement of the cultivation and quality of G. uralensis.

ObjectiveThis study aimed to compare limited open reduction and transepiphyseal intramedullary fixation with Kirschner wire (LOR-TIKW) versus open reduction and internal fixation with plate and screw (ORIF-PS) for treatment of irreducible distal radius diaphyseal–metaphyseal junction (DMJ) fracture in older children.MethodsData of children (aged 10–14 years) treated in our hospital for distal radius DMJ fractures with LOR-TIKW or ORIF-PS from January 2018 to December 2019 were retrospectively analyzed. Follow-up was until radiographic union. Demographic, clinical, and radiographic data; treatment cost; healing time; functional outcome (by Price criteria); complications; and postoperative angulation and displacement were compared between children treated by the two methods. Statistical analysis was performed with alpha set at P < 0.05.ResultsA total of 26 children were included: 14 treated with LOR-TIKW and 12 with ORIF-PS. Operation time was less (22.1 min vs. 46.7 min, P < 0.0001), surgical incision smaller (2.43 cm vs. 5.00 cm, P < 0.0001), cost of internal fixation lower (US$, 40.6 vs. 2020, P < 0.0001), and healing time shorter (4.79 weeks vs. 5.64 weeks, P = 0.03) with LOR-TIKW; however, postoperative fracture angulation was slightly larger (1.07° vs. 0.83°, P = 0.85) and displacement slightly more (0.86 mm vs. 0.58 mm, P = 0.44) in the LOR-TIKW group. Rate of union, functional outcome, and complications were not significantly different between the groups.ConclusionFor irreducible DMJ fracture of distal radius in older children, LOR-TIKW appears to be a promising method with several advantages over ORIF-PS.

Purpose: The purpose of this study is to use the data in the GEO database to analyze, screen biomarkers that can diagnose tuberculosis, and verification of candidate biomarkers.Materials and methods: GSE158767 dataset were used to process WGCNA analysis, differential gene analysis, Gene ontology and KEGG analysis, protein-protein network analysis and hub genes analysis. Based on our previous study, the intersect between WGCNA and differential gene analysis could be used as candidate biomarkers. Then, the enzyme-linked immunosorbent assay was used to validate candidate biomarkers, and receiver operating characteristic was used to assess diagnose ability of candidate biomarkers.Results: A total of 412 differential genes were screened. And we obtained 105 overlapping genes between DEGs and WGCNA. GO and KEGG analysis showed that most of the differential genes were significantly enriched in innate immunity. A total of 15 hub genes were screened, and four of them were verified by Enzyme-linked immunosorbent assay. CCL5 performed well in distinguishing the healthy group from the TB group (AUC = 0.723). And CCL19 performed well in distinguishing the TB group from the ORD groups (AUC = 0.811).Conclusion: CCL19, C1Qb, CCL5 and HLA-DMB may play important role in tuberculosis, which indicated four genes may become effective biomarkers and could be conveniently used to facilitate the individual tuberculosis diagnosis in Chinese people.

Osteoclasts are of hematopoietic lineage and have the ability to degrade mineralized bone tissues. Abnormalities in osteoclastic activity under certain pathological conditions are common in bone diseases such as osteoporosis, osteosclerosis, and arthritis. Although many kinds of drugs are currently used to treat osteoporosis, they have obvious adverse reactions and limitations. CYT387 is a new small-molecule Janus kinase (JAK) inhibitor involved in hematopoiesis, immune modulation, fertility, lactation, and embryonic development. However, it has remained unclear whether CYT387 functionally impacts osteoclast formation. Our study demonstrated through osteoclast formation assay in vitro, that the use of CYT387 is a potential drug candidate for treating osteoclast-associated bone disease. The effects of CYT387 on osteoclast formation, bone resorption, NFATc1 activation, and especially intracellular ROS levels were investigated in vitro. Further, we examined the preclinical prospects of CYT387 using an oophorectomy (OVX) mouse model of osteoporosis with its anti-osteoclast activity in vivo. On the whole, this study shows that CYT387 holds promise for treating osteoclast-related bone illnesses including osteoporosis.

Abstract Liver cancer has become the sixth most diagnosed cancer and the fourth leading cause of cancer death worldwide. Hepatocellular carcinoma (HCC) is responsible for up to 75–85% of primary liver cancers, and sorafenib is the first targeted drug for advanced HCC treatment. However, sorafenib resistance is common because of the resultant enhancement of aerobic glycolysis and other molecular mechanisms. Aerobic glycolysis was firstly found in HCC, acts as a hallmark of liver cancer and is responsible for the regulation of proliferation, immune evasion, invasion, metastasis, angiogenesis, and drug resistance in HCC. The three rate-limiting enzymes in the glycolytic pathway, including hexokinase 2 (HK2), phosphofructokinase 1 (PFK1), and pyruvate kinases type M2 (PKM2) play an important role in the regulation of aerobic glycolysis in HCC and can be regulated by many mechanisms, such as the AMPK, PI3K/Akt pathway, HIF-1α, c-Myc and noncoding RNAs. Because of the importance of aerobic glycolysis in the progression of HCC, targeting key factors in its pathway such as the inhibition of HK2, PFK or PKM2, represent potential new therapeutic approaches for the treatment of HCC.

Background: The aim of this study was to develop and internally validate a treatment failure risk nomogram in a Chinese population of patients with Drug-Resistant Tuberculosis with surgical therapy. Methods: We developed a prediction model based on a dataset of 132 drug-resistant tuberculosis (DR-TB) patients. The least absolute shrinkage and selection operator regression model was used to optimize feature selection for the treatment failure risk model. Multivariable logistic regression analysis was applied to build a predicting model incorporating the feature selected in the least absolute shrinkage and selection operator regression model. Discrimination, calibration, and clinical usefulness of the predicting model were assessed using the C-index, calibration plot, and decision curve analysis. Internal validation was assessed using the bootstrapping validation. Findings: Predictors contained in the prediction nomogram included Lesion, Treatment history, Recurrent chest infection (RCI) and Multidrug-resistant tuberculosis (MDR-TB) or Extensively drug-resistant tuberculosis (XDR-TB). The model displayed good discrimination with a C-index of 0.905 and good calibration. A high C-index value of 0.876 could still be reached in the interval validation. Decision curve analysis showed that the nomogram was clinically useful when an intervention was decided at the treatment failure possibility threshold of 1%. Interpretation: This study developed a novel nomogram with relatively good accuracy to help clinicians access the risk of treatment failure in MDR/XDR-TB patients when starting surgery. With an estimate of individual risk, clinicians and patients can make more suitable decisions regarding surgery. This nomogram requires external validation, and further research is needed to determine whether the nomogram is appropriate for predicting surgery risk in MDR/XDR-TB patients.

The helical spring is one of the fundamental mechanical elements in a wide range of industrial applications. In this study, a novel helical spring made of multistrand carbon fiber/epoxy composite were designed and fabricated by an integrated formation process to achieve flexible adjustment of the spring constant without changing geometric size. Five types of composite helical springs were fabricated by controlling different reinforcement twist and fiber volume content (Vf), forming three twistless composite multistrand helical springs (CMHSs) with Vf of 45%, 50%, and 55% and two multistrand twisted CMHSs with twist coefficient of 4 and 6r/10 cm and Vf of 55%. Quasi-static compression tests showed that the spring constant could be adjusted effectively by changing the Vf and reinforcement twist. By applying finite element analysis, the general correlation of the effects of Vf and reinforcement twist on the spring constant of the CMHSs were further analyzed and discussed. It was proved that the increase of reinforcement twist and Vf can both improve the spring constant, among which the effect of reinforcement twist dominated due to the stress homogenization of the twisting structure. Furthermore, a prediction of spring constant as a function of twist ranging from 0 to 40r/10 cm were conducted based on experimental and simulation results, which was of engineering significance for the application of composite helical springs.

Metamaterials possess attractive electromagnetically left-handed properties yet their nature as an artificial structure rather than a material restricts their further applications. In this article we propose to use Co-based ferromagnetic microwires as periodical metamaterial units interweaved into a fabric to realize a “metacomposite” for steering wave propagation. By introducing ferromagnetic microwires in quartz fabric at three curvatures and periodic lengths, the electromagnetic response and microwave behaviors on both horizontally- and vertically-arranged directions were presented and investigated. Tensile behaviors of metacomposites with different ferromagnetic microwire morphologies were monitored by non-contact full field strain measurement system. At X band (8.2–12.4 GHz), metacomposites containing vertically-arranged ferromagnetic microwires show negative permittivity due to the dielectric resonance of microwire arrays. The dielectric response of metacomposites conformed to Lorentz-oscillator mode, behaving similarly to diluted plasma. When ferromagnetic microwires reached the lowest periodic curved length 2.5 mm with the highest curvature was obtained, the metacomposites presented the maximum microwave absorption of 34% and tensile stress of 273 MPa. The incorporation of ferromagnetic microwires exerts trivial influence on the mechanical properties yet homogenizes the strain and reduces the stress concentration on metacomposites. The metacomposite with a periodic curved length of 7.5 mm has less interweaving points and the highest surface float length, therefore achieving the maximum tensile strength of 458 MPa. The single negative metacomposites containing ferromagnetic microwires are featured with excellent structural properties and amenable microwave left-handed properties in controlling wave-propagating behavior, which is of practical engineering significance in structural health monitoring, sensing and stealth applications.

Abstract Background Hepatocellular carcinoma (HCC) is a common primary malignant tumor which usually progresses to an advanced stage because of late diagnosis. Sorafenib (Sora) is a first line medicine for advanced stage HCC; however, it has been faced with enormous resistance. Simvastatin (Sim) is a cholesterol-lowering drug and has been reported to inhibit tumor growth. The present study aims to determine whether Sora and Sim co-treatment can improve Sora resistance in HCC. Methods The HCC cell line LM3 and an established Sora-resistant LM3 cell line (LM3-SR) were used to study the relationship between Sora resistance and aerobic glycolysis. Cell proliferation, apoptosis and glycolysis levels were analyzed by western blotting, flow cytometry analysis and biomedical tests. A xenograft model was also used to examine the effect of Sim in vivo. Detailed mechanistic studies were also undertaken by the use of activators and inhibitors, and lentivirus transfections. Results Our results demonstrated that the resistance to Sora was associated with enhanced aerobic glycolysis levels. Furthermore, LM3-SR cells were more sensitive to Sim than LM3 cells, suggesting that combined treatment with both Sora and Sim could enhance the sensitivity of LM3-SR cells to Sora. This finding may be due to the suppression of the HIF-1α/PPAR-γ/PKM2 axis. Conclusions Simvastatin can inhibit the HIF-1α/PPAR-γ/PKM2 axis, by suppressing PKM2-mediated glycolysis, resulting in decreased proliferation and increased apoptosis in HCC cells, and re-sensitizing HCC cells to Sora.

Bidens plants are annuals or perennials of Asteraceae and usually used as medicinal materials in China. They are difficult to identify by using traditional identification methods because they have similar morphologies and chemical components. Universal DNA barcodes also cannot identify Bidens species effectively. This situation seriously hinders the development of medicinal Bidens plants. Therefore, developing an accurate and effective method for identifying medicinal Bidens plants is urgently needed. The present study aims to use phylogenomic approaches based on organelle genomes to address the confusing relationships of medicinal Bidens plants. Illumina sequencing was used to sequence 12 chloroplast and eight mitochondrial genomes of five species and one variety of Bidens. The complete organelle genomes were assembled, annotated and analysed. Phylogenetic trees were constructed on the basis of the organelle genomes and highly variable regions. The organelle genomes of these Bidens species had a conserved gene content and codon usage. The 12 chloroplast genomes of the Bidens species were 150,489 bp to 151,635 bp in length. The lengths of the eight mitochondrial genomes varied from each other. Bioinformatics analysis revealed the presence of 50–71 simple sequence repeats and 46–181 long repeats in the organelle genomes. By combining the results of mVISTA and nucleotide diversity analyses, seven candidate highly variable regions in the chloroplast genomes were screened for species identification and relationship studies. Comparison with the complete mitochondrial genomes and common protein-coding genes shared by each organelle genome revealed that the complete chloroplast genomes had the highest discriminatory power for Bidens species and thus could be used as a super barcode to authenticate Bidens species accurately. In addition, the screened highly variable region trnS-GGA-rps4 could be also used as a potential specific barcode to identify Bidens species.

Hepatic ischemia/reperfusion injury (HIRI) is a common complication of liver surgery requiring hepatic disconnection, such as hepatectomy and liver transplantation. The aim of this study was to investigate the effects of cordycepin on HIRI and to elucidate the underlying mechanisms. Balb/c mice were randomly divided into six groups: a normal control group, sham group, H-cordycepin group, HIRI group, L-cordycepin (25 mg/kg) + HIRI group, and H-cordycepin (50 mg/kg) + HIRI group. Mice were subjected to I/R, and cordycepin was intragastrically administered for seven consecutive days before surgery. Orbital blood and liver specimens were collected at 6 and 24 h after HIRI. Serum levels of ALT and AST were decreased in the cordycepin pretreatment groups. Notably, cordycepin attenuated the inflammatory response and the production of proapoptosis proteins, while increasing expression of antiapoptosis proteins and decreasing expression of autophagy-linked proteins. Furthermore, cordycepin inhibited activation of the MAPK/NF-κB signaling pathway. Collectively, these results indicate that cordycepin pretreatment ameliorated hepatocyte injury caused by HIRI. As compared with the HIRI group, cordycepin pretreatment mitigated the inflammatory response and inhibited apoptosis and autophagy via regulation of the MAPK/NF-κB signaling pathway.

The shear thickening fluid (STF) with unique rheology is generally combined with polyurethane (PU) skeleton to manufacture flexible sandwich composites which is widely applied in impact resistence field. In this work, experiments and numerical simulations were comparatively analyzed to investigate the non-Newtonian fluid–solid interaction (NFSI) between STF and PU skeleton during low-velocity impact. Optimization of materials selection is proposed to achieve a higher impact resistance performance. It has been found that the synchronous gradient thickening and flow of STF facilitate the load-bearing and stress diffusion performance. In contrast to Newtonian fluids, the internal STF displays both shear resistence of high-viscosity fluid and easy flow characteristic of low-viscosity fluid, creating areas of low stress in composites and improve the impact resistance performance. The fluid-optimized STF-2 sandwich composite exhibits an up to 154.7% lower peak load and 118.82% higher energy absorption. In terms of the skeleton, a lower modulus of PU foam is conductive to a stronger NFSI, which facilitates the energy exchange between STF and skeleton and further improve the impact resistance performance. As a result, the skeleton-optimized composite containing PU-1 exhibits a 20.10% lower peak load and 17.41% higher energy absorption. The NFSI mechanism and material selection optimization between non-Newtonian fluid and large deformation materials proposed in this work provide an analysis idea and design method for flexible buffering composites.

Hepatic ischemia-reperfusion (IR) injury is a clinically significant process that frequently occurs in liver transplantation, partial hepatectomy, and hemorrhagic shock. The aim of this study was to explore the effectiveness of luteolin in hepatic IR injury and the underlying mechanism. BALB/c mice were randomly divided into six groups, including normal controls (NC), luteolin (50 mg/kg), sham procedure, IR+25 mg/kg luteolin, and IR+50 mg/kg luteolin group. Serum and tissue samples were collected at 6 and 24 h after reperfusion to assay liver enzymes, inflammatory factors, expression of proteins associated with apoptosis and autophagy, and factors associated with the extracellular signal-regulated kinase/peroxisome proliferator-activated receptor alpha (ERK/PPARα) pathway. Luteolin preconditioning decreased hepatocyte injury caused by ischemia-reperfusion, downregulated inflammatory factors, and inhibited apoptosis and autophagy. Luteolin also inhibited ERK phosphorylation and activated PPARα.

Helical springs are indispensable mechanical parts widely used in industry. Lightweight is one of the development trends of helical springs. In this study, three kinds of lightweight polymer matrix composite helical springs (PMCHSs) with unidirectional, multistrand, and wrapped textile structural reinforcement (PMCHS-U, PMCHS-M, and PMCHS-W) were designed, manufactured, and evaluated. The performance of these PMCHSs and the relationship between their performance and their corresponding polymer matrix composite spring wire rods (PMCRs) were studied through the torsion test of the PMCRs and the compression and resilience tests of the PMCHSs. The results showed that the performance of the PMCHSs could be effectively improved by using the wrapped structure as the reinforcement. The compression capacity of PMCHS-W was 72.6% and 137.5% higher than that of PMCHS-M and PMCHS-U, respectively. The resilience performance of the PMCHSs decreased with the increase in the spring constant. The performances of the PMCHSs and a steel spring were compared. The results showed that the spring constant of the steel spring could be achieved when the masses of PMCHS-U, PMCHS-M, and PMCHS-W were only 75%, 63%, and 49% of the mass of the steel spring, respectively. This research is of great significance to the improvement in lightweight spring performance.

Background: Surgery is increasingly accepted as an adjunctive approach to treat multidrug-resistant tuberculosis (MDR-TB) or extensively drug-resistant tuberculosis (XDR-TB). However, a model that includes all factors to predict the risk of postoperative complications is lacking.Methods: We developed a prediction model based on 138 patients who had undergone surgery as treatment for drug-resistant tuberculosis (DR-TB) after 24 months. Clinical features on the lesion type (L), treatment history (T), physiologic status of the body (B), and surgical approach (S) were evaluated. Multivariable logistic regression analysis was conducted by clinical features selected in the least absolute shrinkage and selection operator (LASSO) to build a nomogram. The discrimination, calibration, and clinical usefulness of the nomogram were assessed using the C-Index, calibration plots, and decision curves. Internal validation was assessed using bootstrapping.Results: The nomogram contained the features L, B, T, cavitary, recurrent chest infection (RCI) and MDR-TB/XDR-TB. The model displayed good discrimination with a C-Index of 0.879 (95% CI: 0.799–0.967). A high C-Index of 0.824 was achieved in the interval validation. Decision-curve analysis showed that the nomogram was clinically useful if intervention was decided at the non-adherence possibility threshold of 4%.Conclusion: Our novel nomogram could be used conveniently to predict postoperative complication risk in DR-TB patients.

Flexible, lightweight and practical luminous fibers have aroused great interest in the field of smart textiles. In this study, we report flexible aggregation induced emission luminogen (AIEgen) fibers which can be produced in a continuous and large-scale way. Fluorescent nanofibers prepared by conjugate electrospinning are wrapped on the surface of flexible core fiber to form a skin-core AIEgen fiber. The electrospinning parameters including spinning concentration, traction rate, spinning rate and tetraphenylethylene (TPE) concentration were designed and optimized via the non-interactive orthogonal experimental design. The optimized flexible AIEgen fiber retains the excellent mechanical properties of core fiber and is equipped with the fluorescence properties of the wrapped fluorescent nanofibers. In addition, the fabricated AIEgen fibers can still exhibit good fluorescence characteristic after being stretched, bent, washed and rubbed, displaying excellent durability. Different from reported luminescent fibers which required an external power supply or complicated preparation process, the flexible AIEgen fibers fabricated in this study can be prepared continuously in a large-scale way and exhibits effectiveness in clothing display, anti-counterfeiting, information transmission and hazardous gas detection, which shows great application potential for wearable smart textiles.

Abstract Objective Hepatocellular carcinima is one of the most common tumors in clinic and also one of the leading causes of death from cancer worldwide. Quercetin shows significant effects on blocking the development of various cancers. Methods We used the human hepatocellular carcinoma LM3 and nude mice tumor model to assess the effects of quercetin in hepatocellular carcinoma and clarify its mechanism of action. We collected LM3 cell line treated with different doses of quercetin at different time periods and determined the vital indexes. The liver tissues of mice were collected and used for western boltting (WB), Hematoxylin and Eosin (H&E) and TUNEL staining. Results Results indicated that quercetin suppressed the Hepatocellular carcinoma (HCC) growth both in vivo and in vitro. Quercetin could disturb LM3 cells proliferation and cell cycle distribution, thus inducing apoptosis. At the same time, quercetin inhibited LM3 cells migration and invasion and promoted HCC autophagy. These effects at least partly depended on the down‐regulation of the activation of JAK2 and STAT3 by quercetin. Conclusion Quercetin inhibited hepatocellular carcinoma progression by modulating cell apoptosis, migration, invasion, and autophagy; and its effects were at least partly related with the JAK2/STAT3 signaling pathway.

Abstract Background The treatment for advanced primary hepatocellular carcinoma (HCC) is sorafenib (SORA), while HCC has become increasingly drug resistant with enhanced aerobic glycolysis. The present study aimed to examine the chemotherapeutic effects of a flavonoid proanthocyanidin B2 (PB2) on HCC. Methods Five kinds of HCC cell lines and LO2 were used to test the effect of PB2 on aerobic glycolysis. The proliferation, cell cycle, apoptosis and a xenograft mouse model were analyzed. Lentivirus overexpressed pyruvate kinase M2 (PKM2) or sh-PKM2 was used to verify the target of PB2. The detailed mechanism was investigated by immunofluorescence, co-immunoprecipitation, and western blotting. Results PB2 inhibited the proliferation, induced cell cycle arrest, and triggered apoptosis of HCC cells in vivo and in vitro. PB2 also suppressed glucose uptake and lactate levels via the direct inhibition of the key glycolytic enzyme, PKM2. In addition, PKM2 inhibited the nuclear translocation of PKM2 and co-localization of PKM2/HIF-1α in the nucleus, leading to the inhibition of aerobic glycolysis. Co-treatment with PB2 was also effective in enhancing the chemosensitivity of SORA. Conclusions PB2 inhibited the expression and nuclear translocation of PKM2, therefore disrupting the interaction between PKM2/HSP90/HIF-1α, to suppress aerobic glycolysis and proliferation, and trigger apoptosis in HCC via HIF-1α-mediated transcription suppression.

Nonalcoholic fatty liver disease (NAFLD) has become the most common liver disorder in both China and worldwide. It ranges from simple steatosis and progresses over time to nonalcoholic steatohepatitis (NASH), advanced liver fibrosis, cirrhosis, or hepatocellular carcinoma(HCC). Furthermore, NAFLD and its complications impose a huge health burden to society. The microbiota is widely connected and plays an active role in human physiology and pathology, and it is a hidden ‘organ’ in determining the state of the host, in terms of homeostasis, or disease. Peroxisome proliferator-activated receptors (PPARs) are members of the nuclear receptorsuperfamily and can regulate multiple pathways involved in metabolism, and serve as effective targets forthe treatment of many types of metabolic syndromes, including NAFLD. The purpose of this review is to integrate related articles on gut microbiota, PPARs and NAFLD, and present a balanced overview on how the microbiota can possibly influence the development of NAFLD through PPARs.

Hepatocellular carcinoma (HCC) is the most common primary liver malignancy and is a leading cause of cancer-related deaths globally, with few effective therapeutic options. Bile acids (BAs) are synthesized from cholesterol in the liver and can be modulated by farnesoid X receptor (FXR) and G-protein coupled BA receptor 1 (GPBAR1/TGR5). Alterations in BAs can affect hepatic metabolic homeostasis and contribute to the pathogenesis of liver cancer. Increasing evidence points to the key role of bacterial microbiota in the promotion and development of liver cancer. They are also involved in the regulation of BA synthesis and metabolism. The purpose of this review is to integrate related articles involving gut microbiota, BAs and HCC, and review how the gut microbiota-BA signaling axis can possibly influence the development of HCC.

Hepatic ischemia and reperfusion injury is characterized by hepatocyte apoptosis, impaired autophagy, and oxidative stress. Fenofibrate, a commonly used antilipidemic drug, has been verified to exert hepatic protective effects in other cells and animal models. The purpose of this study was to identify the function of fenofibrate on mouse hepatic IR injury and discuss the possible mechanisms. A segmental (70%) hepatic warm ischemia model was established in Balb/c mice. Serum and liver tissue samples were collected for detecting pathological changes at 2, 8, and 24 h after reperfusion, while fenofibrate (50 mg/kg, 100 mg/kg) was injected intraperitoneally 1 hour prior to surgery. Compared to the IR group, pretreatment of FF could reduce the inflammatory response and inhibit apoptosis and autophagy. Furthermore, fenofibrate can activate PPAR-α, which is associated with the phosphorylation of AMPK.

Objective. The aim of this study is to confirm the hepatocellular protective functions of apigenin and the molecular mechanism on liver fibrosis in mice. Methods. Carbon tetrachloride (CCl4) and bile duct ligature (BDL) mouse fibrosis models were used to investigate the effects of apigenin on liver fibrosis. Sixty-six male C57 mice were randomly divided into eight groups, including the vehicle group, CCl4 group, CCl4+L-apigenin (20 mg/kg) group, CCl4+H-apigenin (40 mg/kg) group, sham group, BDL group, BDL+L-apigenin(20 mg/kg) group, and BDL+H-apigenin(40 mg/kg) group. Serum liver enzymes (ALT and AST), proteins associated with autophagy, and indicators linked with the TGF-β1/Smad3 and p38/PPARα pathways were detected using qRT-PCR, immunohistochemical staining, and western blotting. Results. Our findings confirmed that apigenin could decrease the levels of ALT and AST, suppress the generation of ECM, inhibit the activation of HSCs, regulate the balance of MMP2 and TIMP1, reduce the expression of autophagy-linked protein, and restrain the TGF-β1/Smad3 and p38/PPARα pathways. Conclusion. Apigenin could alleviate liver fibrosis by inhibiting hepatic stellate cell activation and autophagy via TGF-β1/Smad3 and p38/PPARα pathways.

Background. Hepatic ischemia-reperfusion (IR) injury is one of the severe complications associated with liver surgery and leads to liver dysfunction. PPARγ is always linked with various physiologic pathways, and it can alleviate liver damage in IR injury. Aim. In this study, we explored the potential mechanism of PPARγ in the pathogenesis of hepatic IR injury by mice model. Methods. After treated with si-PPARγ or rosiglitazone, mice were subjected to hepatic ischemia-reperfusion. Liver tissue and blood samples were collected to evaluate liver injury and detected relative mRNA and protein expressions. Results. The expression of PPARγ was increased after reperfusion. And the alleviation of PPARγ aggravated the liver damage in IR; at the same time, upregulation of the expression of PPARγ released the liver damage. And these effects of PPARγ in IR were related to the AMPK/mTOR/autophagy signaling pathway. Conclusion. PPARγ plays an important role in hepatic IR injury at least partly via the AMPK/mTOR/autophagy pathway.

In order to solve defects such as poor integrity, delamination failure, and narrow absorption bandwidth, three-dimensional (3D) gradient honeycomb woven composites (GHWCs) with triangular sections were designed and prepared. Three-dimensional gradient honeycomb woven fabric was crafted with carbon fiber (CF) filaments and basalt fiber (BF) filaments as raw materials on an ordinary loom. Then, the 3D honeycomb woven fabric filled with rigid polyurethane foam was used as the reinforcement, and epoxy resin (EP) doped with carbon black (CB) and carbonyl iron powder (CIP) was conducted as the matrix. The 3D GHWC with triangular sections, which had both EM-absorbing and load-bearing functions, was prepared by the VARTM process. Through the macro test and micro characterization of 3D GHWCs with triangular sections, the overall absorbing properties and mechanical properties of the materials were analyzed. Moreover, the EM-absorbing mechanism and failure mode of the materials were clarified in this work. The results indicated that the CF filament reflective layer effectively improved the EM-absorbing and mechanical properties. Adding a CB/CIP-absorbing agent enhanced the overall EM-absorbing property but reduced the mechanical properties. The increasing number of gradient layers increased the maximum bending load, but the EM-absorbing performance first increased and then decreased. When the thickness was 15 mm, the maximum bending load was 3530 N, and the minimum reflection loss (RLmin) was −21.6 dB. The synergistic effects of EM-absorbing and mechanical properties were the best right now. In addition, this work provided a feasible strategy that adjusting the type of absorber and gradient aperture size ratio could meet the unique requirements of absorbing frequency and intensity, which has excellent application prospects in civil and military fields.

Analysis of the relationships among wild species of section Moutan in the plant genus Paeonia has traditionally been problematic. Interspecies relationships cannot be effectively determined using phenotypic traits alone or through analysis of nuclear or chloroplast DNA fragments. Elucidation of complete chloroplast genome sequences will aid the identification and phylogeny of these species. In this study, the complete chloroplast genomes of three sect. Moutan plants were sequenced and analyzed. Comparative and phylogenetic analyses of the complete chloroplast genomes of all eight species of sect. Moutan were then conducted. The three complete chloroplast genomes gained in this study showed four-part annular structures, and the genome length, structure, GC content, codon usage, and gene distribution were highly similar. There was greater variation in the noncoding regions of the sequences than in the conserved protein-coding regions. Sequence variations in the small single copy (SSC) regions and large single copy (LSC) regions were considerably greater than those in the inverted repeat (IR) regions. Phylogenetic analysis revealed that the species of sect. Moutan clustered in one branch and then subdivided into smaller branches. As for the three complete chloroplast genome sequences obtained in this study, Paeonia jishanensis clustered with another P. jishanensis sequence from the GenBank database, Paeonia qiui clustered with Paeonia rockii, and Paeonia delavayi var. lutea clustered with Paeonia ludlowii. It was also found that the complete chloroplast genomes, LSC regions, and SSC regions all showed great abilities in identification and phylogenetic analysis of the species of sect. Moutan, while IRs regions and highly variable regions were not suitable for the species of sect. Moutan.

Osteoclasts with elevated bone resorption are commonly present in postmenopausal osteoporosis, and other osteolytic pathologies. Therefore, suppressing osteoclast generation and function has been the main focus of osteoporosis treatment. Betulinic acid (BA) represents a triterpenoid mainly purified from the bark of Betulaceae. BA shows multiple biological activities, including antitumor and anti-HIV properties, but its effect on osteolytic conditions is unknown. Here, BA suppressed receptor activator of nuclear factor‐κB ligand (RANKL)‐associated osteoclastogenesis and bone resorptive function, as assessed by tartrate‐resistant acid phosphatase (TRAP) staining, fibrous actin ring generation, and hydroxyapatite resorption assays. Mechanistically, BA downregulated the expression of osteoclastic-specific genes. Western blot analysis revealed that BA significantly interrupted ERK, JNK and p38 MAPK activation as well as intracellular reactive oxygen species (ROS) production, thus altering c-Fos and NFATc1 activation. Corroborating the above findings in cell-based assays, BA prevented ovariectomy-associated bone loss in an animal model. In conclusion, these findings suggest that BA can inhibit osteoclast generation and function as well as the RANKL signaling pathway, and might be used for treating osteoclast-related osteoporosis.

The metacomposites with tailorable negative permittivity including periodic interweaving ferromagnetic mircowires were fabricated by weaving and resin transfer molding methods. By manipulating the alignment direction, periodic curved length, spacing between ferromagnetic microwires for adjusting complex permittivity and permeability, the electromagnetic wave propagation of as-prepared metacomposites under X and Ku bands were evaluated and investigated by experiment and simulation. For metacomposites composed of vertically arranged microwires, for both 1 mm and 0.5 mm spacing, the permittivity turned to negative in certain frequency bands. The simultaneous existence of short periodic curved length and spacing benefits the broadening of frequency band under which metacomposites exhibits single-negative behavior. Significant transmission windows are observed due to negative permittivity occurring at plasma frequency. The simulation results show major loss for the textile structured metacomposites under X and Ku bands derives from electric loss, and small periodic curved length tends to form uniform power loss distribution. Melicope pteleifolia, an important medicinal and horticultural plant, has antipyretic, anti-inflammatory, and analgesic effects. Here, the complete chloroplast genome of M. pteleifolia was sequenced and its phylogenetic relationship was investigated. The complete chloroplast genome of M. pteleifolia was 159,014 bp in size, including a pair of inverted repeat regions (IR, 27,640 bp), a large single copy region (LSC, 85,124 bp), and a small single copy region (SSC, 18,610 bp). The GC content of the chloroplast genome was 38.3%. A total of 133 genes were annotated, including 88 protein-coding genes, 8 rRNA genes, and 37 tRNA genes. Phylogenetic analysis revealed that M. pteleifolia clusters together with species of Toddalia, Zanthoxylum, Tetradium, Phellodendron, and Casimiroa.
