Hemoglobin A1c (HbA1c) is a major component of glycated hemoglobin in human red blood cells. It has been proven to be a significant biomarker for the diagnosis of diabetes; its content in fresh red cells in diabetes blood reflects the average level of blood glucose over the previous three months. Thus, HbA1c level has been used for the assessment of long-term glycemic control in diabetes; the level of 6.5% HbA1c has been certified as a critical cut-off for the diabetes diagnosis. The current commonly used method for HbA1c quantification is based on cation-exchange high performance liquid chromatography (CX-HPLC). The method has advantages such as high stability, rapidity, and automation, but there are still some unidentified peaks of Hb species in CX-HPLC (VARIANT Ⅱ system); in particular, the presence of HbA3 (a glutathiolated Hb) affects the accurate determination of HbA1c. HbA3 is usually present in healthy adult blood samples at 2%-4%, but the concentration of HbA3 increases due to the protection of erythrocytes from oxidation, resulting in decreased HbA1c. However, the relative location of the HbA3 peak in the CX-HPLC clinical chromatogram has not been established. To address this issue, we extracted Hb species from fresh blood samples obtained from a hospital in an anaerobic environment to avoid possible redox reactions of Hb and glutathione. After the extraction, the Hb samples were analyzed using two methods: a low-resolution CX-HPLC (5/50 mm column) currently used for diabetes diagnosis and a high-resolution cationic exchange HPLC (Mono-S 5/50 mm column), to identify the peak corresponding to HbA3. The CX-HPLC analysis of fresh blood samples indicated that the unknown peak P3 located between HbA1c and HbA0 peaks corresponded to the HbA3 peak between HbA1c and HbA0 in the Mono-S-HPLC. Microarray isoelectric focusing (IEF) was used for the micro-preparation of HbA3, HbA1c, and HbA0 in healthy blood samples; then, the micro-prepared species of HbA3, HbA1c, and HbA0 were individually identified via Mono-S-HPLC. The results of the CX-HPLC, Mono-S-HPLC, and microarray IEF experiments indicated that the P3 peak might correspond to HbA3. To confirm this, glutathiolated Hb samples were synthesized via acetylphenylhydrazine and analyzed using both the Mono-S- and CX-HPLC systems. The results showed that the content of both glutaminated hemoglobin of HbA3 in Mono-S-HPLC and P3 in CX-HPLC increased, implying the peak of P3 with the retention time of 1.50 min in CX-HPLC was the peak corresponding to HbA3 in Mono-S-HPLC and microarray IEF. Based on the above experiments and our previous results, the influence of HbA3 on both the analysis of HbA1c in blood samples and the diabetes diagnosis needs to be considered and discussed. The study results are significant for the tentative assignment of peak P3 and for offering more information on diabetes diagnosis using CX-HPLC in the clinical setting.

Herein, the quench model of the moving exchange boundary (MEB) was first created via a ligand of 5,5'-dithiobis(2-nitro-benzoic acid) (DTNB) and group of 3-mercaptopropionic acid (MPA) capped on QDs, and then the recovery model was formed via MPA and 2-nitro-5-thiobenzoic acid (TNB) capped on QDs. The theory on MEB dynamics and width was developed based on the two reversible models, the simulation was conducted for the illumination of MEB, and the protocol was described for the MEB runs. The experiments revealed that (i) the quench model could be created via DTNB and MPA capped on QDs and the recovery one could be in situ formed via MPA and TNB capped on QDs, showing the feasibility of MEB models; (ii) the simulations on MEB dynamics and width were in coincidence with the theoretic predictions, showing the validity of two models; and (iii) the experiments demonstrated the validity of models, predictions, and simulations. The models and theory have potential for development of a biosensor, nanoparticle characterization, separation science, and an affinity assay of ligand-QDs.

In capillary electrophoresis, determination of the basic physical and chemical properties of compounds, such as absolute mobility (m0) and dissociation constant (pKa), is of great practical significance. This is because the aforementioned properties are often used for the qualitative or quantitative analyses of the relevant compounds toward their application as potential drugs. Lovastatin is a potential drug candidate that can reduce the levels of cholesterol and low-density lipoprotein cholesterol in the blood, as well as prevent atherosclerosis and coronary heart disease. For a more convenient and rapid investigation of the properties and applications of lovastatin, it is necessary to determine its m0 and pKa values. However, existing research on capillary electrophoresis for lovastatin and other related drugs focus on their quantitative determination, and their action mechanism and functions. Unfortunately, there are very few studies aimed at the determination of the m0 and pKa values of lovastatin. Based on related studies, this paper herein proposed a novel method to determine m0 and pKa of lovastatin. The present study mainly included a calculation method and experimental verification. The calculation method was based on capillary zone electrophoresis (CZE) and the empirical formula of ion mobility. First, on the basis of the empirical formula, the calculation formula for m0 was derived from the relationship between the actual mobility (mact), effective mobility (meff) and m0. Second, for a monovalent acid (HA), according to the calculation formula for m0 part, considering the hydrogen ion concentration as the independent variable and the reciprocal of meff as the dependent variable, a straight line was obtained on the coordinate axis. From the slope of this straight line, the dissociation equilibrium constant Ka was obtained directly, and pKa was calculated easily. After the derivation of m0 and pKa in the theoretical part, the feasibility and reliability of this method were verified by using it to determine the m0 and pKa values of several organic acids and bases (barbituric acid, benzoic acid, benzylamine, phenol, and m-cresol) in the experimental part. The existing miniature capillary electrophoresis (CE) devices use a tablet or a computer for data processing and analysis, which hinders their portability. In order to solve this problem, a smartphone-based CE device was proposed, which allowed for real portable quantitative analysis. The device integrated the functions of capacitively coupled contactless conductivity detection (C4D) and Bluetooth communication. Furthermore, a Kotlin language-based application with a user-friendly interface was developed. The application could not only control the electrophoresis run in the CE device but also receive the data from the C4D detector in real time, display the electrophoretogram, and process the data. The peak areas could be calculated automatically on the smartphone, and the migration time could be obtained. The size of the developed device was 20 cm×20 cm×15 cm, and its weight was 2 kg. Quaternary ammonium salts (QAs) in disinfectors (dodecyl dimethyl benzyl ammonium bromide (DDBAB) and dodecyl trimethyl ammonium bromide (DTAB)) were used as the analytes to verify the performance of the developed device. The experimental data showed that the linear ranges of DDBAB and DTAB were from 20 to 1000 and from 30 to 1000 μmol/L, respectively. The correlation coefficients (R2) of DDBAB and DTAB were 0.9995 and 0.9989, respectively, indicating good linearity between the peak area and concentration. The limits of detection (LODs) of DDBAB and DTAB were 10 and 13 μmol/L, respectively. The intra-day relative standard deviations (RSDs, n=3) of DDBAB and DTAB were 1.9% and 2.7% respectively, revealing good repeatability. In addition, a mixture of DDBAB and DTAB was tested. Two QAs were separated within 8 min, showing good selectivity. Finally, QAs in a bromo geramine disinfector used in the field were tested to further validate the performance of the designed device. The recoveries of DDBAB and DTAB were 100.5%-101.5% and 96.2%-99.3%, respectively, indicating good accuracy. The developed device has the advantages of good linearity, low LOD, good repeatability, high accuracy, and real portability, and it can be used for the quantitative detection of QAs in disinfectors.

Assays of uric acid (UA) in human urine and blood are of great significance to clinic diagnoses of many diseases, such as gout, renal failure, and hyperuricemia. Portable and visual quantification method, however, has been rarely developed for POCT detection of UA in urine and blood samples. In this work, we developed the electrophoresis titration (ET) model, the relevant sensing method and the portable chip device for portable and visual detection of UA in both urine and blood based on moving reaction boundary (MRB). In the model, uricase catalyzes UA in the anode well to allantoin, producing hydrogen peroxide, which oxidizes leucocrystal violet dye without charge to crystal violet (CV+) with one positive charge. Under an electric field, a MRB was created between the violet CV+ moving from the anode well into the channel and the alkaline sodium acetate in channel, resulting in an ET sensing. The model indicated that the distance of MRB under given conditions was as a function of UA content, implying an extraordinary simple sensing for UA. Based on the MRB-ET model, a series of experiments were conducted. The results evidently validated the model and method of ET-MRB. The experiments not only demonstrated the high facility and portability of MRB-ET model, but also showed the visuality, selectivity and rapidity. Herein, a facile thermometer-like model of electrophoresis titration (ET) biosensor was proposed as an alternative tool for miRNA assay via moving reaction boundary (MRB) chip. For proof-of-concept demonstration, miRNA-122 and catalyzed hairpin assembly (CHA) were chosen as the model analyte and amplification, respectively. In the developed ET system, miRNA triggered the CHA with two hairpin probes (H1, H2) to yield H1–H2 duplexes with negative charges. Under an electric field, the duplexes moved into ET channel, and neutralized the acidic TAE buffer creating an MRB indicated by SYBR Green I (SGI). The model revealed that the MRB distance was as a function of logarithmic miRNA-122 content, indicating a facile sensing model. The relevant experiments were conducted and systemically validated the model of miRNA ET. Under the optimized conditions, the linear range of ET sensor was from 20 fM to 1 nM and the limit of detection (LOD) was 10 fM, showing a more than 100-fold sensitive increase in contrast to the one with a single CHA amplification. The mechanism of sensitive increase was well unveiled by the designed experiments. In addition, the ET biosensor had good selectivity, stability (less than 5% for intra-day and inter-day) and recovery (96%–110%), and was successfully applied for the assay of miRNA-122 and miRNA let-7a in real bio-fluids of serum and cancer cell lysate. Evidently, the proposed biosensor might be used as an alternative assay tool after nucleic acid amplification due to its high simplicity, sensitivity, specificity, linearity, stability and recovery.

Bombesin-like receptor 3 (BRS3), an orphan G protein-coupled receptor (GPCR), plays important roles in our biological system while the exact mechanisms behind it are less known. To get insights of the biological effects upon BRS3 activation, we utilized quantitative proteomics approach to explore the dynamic protein profiling during the stimulation by its ligand. At different time points after stimulation with BRS3 surrogate agonist, the protein profiling in BRS3 overexpressed HEK 293 cells BRS3 (HEK 293-BRS3) was analyzed by nano-LC-MS/MS. In total, 1593 cellular proteins were confidently identified and quantified, including 146 proteins dysregulated at multiple time points and 319 proteins only altered at one time point. Data analysis indicated that BRS3 activation could regulate cell death, survival, and protein synthesis, particularly mRNA translation. Key signaling pathways were revealed for BRS3 signal transduction. In particular, 21 of our identified proteins are involved in the rapamycin (mTOR) signaling pathway. The promotion of mTOR was further confirmed through monitoring its indicative targets upon BRS3 activation. Upon the inhibition of mTOR by rapamycin, cell proliferation was dramatically reversed. Our proteomics data collectively demonstrate that BRS3 activation will lead to cascades of signal transduction and promote cell proliferation. The developed strategy might be utilized to discover the roles of other GPCRs and improve our understanding of their unknown functions.

Nucleic acid binding proteins (NABPs) mediate a broad range of essential cellular functions. However, it is very challenging to comprehensively extract whole cellular NABPs due to the lack of approaches with high efficiency. To this end, carbon nanomaterials, including graphene oxide (GO), carboxylated graphene (cG), and carboxylated carbon nanotube (cCNT), were utilized to extract cellular NABPs in this study through a new strategy. Our data demonstrated that GO, cG, and cCNT could extract nearly 100% cellular DNA in vitro. Conversely, their RNA extraction efficiencies were 60, 50, and 29%, respectively, partially explaining why GO has the highest NABPs yield compared to cG and cCNT. We further found that ionic bond mediated by cations between RNA and functional groups of nanomaterials facilitated RNA absorption on nanomaterials. About 2400 proteins were successfully identified from GO-enriched NABPs sample, and 88% of annotated NABPs were enriched at least 2 times compared to cell lysate, indicating the high selectivity of our strategy. The developed method was further applied to compare the NABPs in two lung cancer cell lines with different tumor progression abilities. According to label-free quantification results, 118 differentially expressed NABPs were discovered and 6 candidate NABPs, including ACAA2, GTF2I, VIM, SAMHD1, LYAR, and IGF2BP1, were successfully validated by immunoassay. The level of SAMHD1 in the serum of lung cancer patients was measured, which significantly increased upon cancer progression. Our results collectively demonstrated that GO is an ideal nanomaterial for NABPs selective extraction, which could be broadly used in varied physiological and pathophysiological settings.

As a vital enzyme, alkaline phosphatase (ALP) has great clinical significance in diagnoses of bone or liver cancer, bone metastases, rickets, and extrahepatic biliary obstruction. However, there is still no really portable chip for the ALP assay in blood. Herein, a simple electrophoresis titration (ET) model was developed for ALP detection via a moving reaction boundary (MRB). In the model, ALP catalyzed the dephosphorylation of a 4-methylumbelliferyl phosphate disodium salt (4-MUP) substrate in the cathode well to 4-methylumbelliferone ([4-MU]-) with a negative charge and blue fluorescence under UV excitation. After the catalysis, an electric field was used between the cathode and the anode. Under the electric field, [4-MU]- moved into the channel and neutralized the acidic Tris-HCl buffer, resulting in the quenching of [4-MU]- and creating a MRB. The ET system just had an ET chip, a lithium cell, a UV LED and an iPhone used as a recorder, having no traditional expensive power supply and fluorescence detector. The relevant method was developed, and a series of experiments were conducted via the ET chip. The experiments showed: (i) a MRB could be formed between the [4-MU]- base and the acidic buffer, and the MRB motion had a linear relationship with the ALP activity, validating the ET model; (ii) the ET run was not impacted by many interferences, implying good selectivity; and (iii) the ET chip could be used for portable detection within 10 min, implying an on-site and rapid analysis. In addition, the ET method had a relatively good sensitivity (0.1 U L-1), linearity (V = 0.033A + 3.87, R2 = 0.9980), stability (RSD 2.4-6.8%) and recoveries (101-105%). Finally, the ET method was successfully used for ALP assays in real serum samples. All the results implied that the developed method was simple, rapid and low-cost, and had potential for POCT clinical ALP assays.

Mitochondria play essential roles in both energy metabolism and cell signaling, which are critical for cell survival. Although significant efforts have been invested in understanding mitochondrial biology, methods for intact mitochondria preparation are technically challenging and remain to be improved. New methods for heterogeneous mitochondria purification will therefore boost our understanding on their physiological and biophysical properties. Herein, we developed a novel recycling free-flow isoelectric focusing (RFFIEF) with post-pH gradient sample injection (post-PGSI) for preparative separation of mitochondria. Crude mitochondria of rabbit liver obtained from differential centrifugation were purified by the developed method according to their pI values as six fractions. Transmission electron microscope images revealed that intact mitochondria existed in two fractions of pH 6.24 and 6.61, degenerative mitochondria were in two fractions of pH 5.46 and 5.72, and inner membrane vesicles (IMVs) appeared in the fractions of pH 4.70 and 5.04. Membrane potential measurement proved a dramatic difference between intact mitochondria and IMVs, which reflected the bioactivity of obtained populations. Particularly, proteomics analyses revealed that more number of proteins were identified in the intact fractions than that of IMVs or crude mitochondria, which demonstrated that RFFIEF could be powerful tool for the preparation of intact organelle as well as their proteomic and in-depth biological analysis.

Circulating tumor exosomes harbor plenty of cancer biological information, which have emerged as promising targets for cancer early detection and diagnosis. Human serum and saliva are unique diagnostic body fluids, which contain numerous circulating exosomes. It is necessary to establish standardized isolation method and compare their proteome profiling for translational medicine. High abundant proteins in these body fluids were removed before exosomes isolation and obtained exosomes were further confirmed by morphology analysis and surface biomarker test. Label free quantification was applied to systematically compare the protein profiling in saliva and serum exosomes. 319 and 994 exosomal proteins were identified from saliva and serum by LC-MS/MS, respectively. To explore their utility for cancer proteomics, we systematically compared the proteome of saliva and serum exosomes from healthy subjects and lung cancer patients. In particular, 11 potential candidates were coincidently discovered in both body fluids of lung cancer patients. Our finding enforced the hypothesis that cancer related proteins were presented in saliva and serum exosomes, which promoted the unique features of exosomes in our body fluids. A circulating exosomes based body fluid test could be easily established for monitoring cancer once these candidates were validated.

Poor control of electroosmotic flow (EOF) often occurs in chip electrophoretic titration (ET) of moving reaction boundary due to both absence of PMMA channel coating and coexistence of acid-base. Also, chip ET still suffers from low reproducibility, instability, disposability and high cost as well. To address these issues, a reusable ET chip (60 mm × 60 mm × 5.0 mm) was designed via an upper PMMA sheet, a middle silicon membrane and a lower sheet. In the upper sheet, enlarged channel (500 μm × 200 μm × 12 mm) was fabricated and linked to the anodic and cathodic wells. The upper sheet, middle membrane and lower sheet could be easily assembled together in 20 s as an ET chip of supporting 1.4 kg tension force, indicating good mechanical stability. Regeneration and reassembling of chip could be finished in 50 s. EOF could be simply suppressed by mass chemical cross-linked polyacrylamide gel or physical agarose gel in the enlarged channel. The EOF-free chip could be well used for: (i) acid-base ET; (ii) EOF monitoring; (iii) pKa determination; (iv) protein content analysis. The EOF-free chip showed the following merits: (i) 98% success rate in contrast to less than 30% in normal channels (80 μm × 20 μm × 5.0 mm); (ii) more than 30 times reuse, indicating reusability and low cost; (iii) less than 10% of relative standard deviation, suggesting fair stability; and (iv) good recovery and accuracy. The developed chip makes great advancement towards real use of chip in acid- base titration, EOF monitoring, pKa determination and protein content analysis.

An ionic coordination hybrid hydrogel is formed with ionic and covalent crosslinked networks via one-step copolymation. It is demonstrated that the hybrid crosslinker gel is used as a separation matrix for bioseparation, showing high separation efficiency, in contrast to traditional polyacrylamide gels. The hydrogel might serve as a novel matrix for bioseparation.

An aptamer-linked assay of a target biomarker (e.g., thrombin) is facing the challenges of long-term run, complex performance, and expensive instrument, unfitting clinical diagnosis in resource-limited areas. Herein, a facile chip electrophoresis titration (ET) model was proposed for rapid, portable, and low-cost assay of thrombin via aptamer-linked magnetic nanoparticles (MNPs), redox boundary (RB), and horseradish peroxidase (HRP). In the electrophoresis titration-redox boundary (ET-RB) model, thrombin was chosen as a model biomarker, which could be captured within 15 min by MNP-aptamer 1 and HRP-aptamer 2, forming a sandwich complex of (MNP-aptamer 1)-thrombin-(HRP-aptamer 2). After MNP separation and chromogenic reaction of 3,3',5,5'-tetramethylbenzidine (TMB) within 10 min, an ET-RB run could be completed within 5 min based on the reaction between a 3,3',5,5'-tetramethylbenzidine radical cation (TMB•+) and l-ascorbic acid in the ET channel. The systemic experiments based on the ET-RB method revealed that the sandwich complex could be formed and the thrombin content could be assayed via an ET-RB chip, demonstrating the developed model and method. In particular, the ET-RB method had the evident merits of simplicity, rapidity (less than 30 min), and low cost as well as portability and visuality, in contrast to the currently used thrombin assay. In addition, the developed method had high selectivity, sensitivity (limit of detection of 0.04 nM), and stability (intraday: 3.26%, interday: 6.07%) as well as good recovery (urine: 97-102%, serum: 94-103%). The developed model and method have potential to the development of a point-of-care testing assay in resource-constrained conditions.

A traditional immobilized pH gradient (IPG) has a high stability for isoelectric focusing (IEF) but suffers from time-consuming rehydration, focusing and staining-imaging as well as complex performance. To address these issues, an IEF system with an array of 24 IPG columns (10 mm × 600 μm × 50 μm) and dynamic scanning imaging (DSI) was firstly designed for protein focusing. Moreover, two IPG columns (pH 4–9 and pH 6.7–7.7 of 10 mm in length) were firstly synthesized for IEF. A series of experiments were carried out based on the IEF array. In contrast to a traditional IPG IEF with more than 10 h rehydration, 5–14 h IEF and ca 10 h stain-imaging, the IEF array had the following merits: 25 min rehydration for sample loading, 4 min IEF, and 2 min dynamics scanning of 24 columns, well addressing the issues of traditional IEF. Furthermore, the IEF array had fair sensitivity (LOD of 60 ng), good recovery (95%), and high stability (1.02% RSD for intra-day and 2.16% for inter-day). Finally, the developed array was successfully used for separation and determination of HbA1c (a key biomarker for diabetes diagnosis) in blood samples. All these results indicated the applicability of the developed IEF array to diabetes diagnosis.

In this work, charge-to-mass ratio (C/M) and band broadening analyses were combined to provide better guidance for the design of free-flow zone electrophoresis carrier buffer (CB). First, the C/M analyses of hemoglobin and C-phycocyanin (C-PC) under different pH were performed by CLC Protein Workbench software. Second, band dispersion due to the initial bandwidth, diffusion, and hydrodynamic broadening were discussed, respectively. Based on the analyses of the C/M and band broadening, a better guidance for preparation of free-flow zone electrophoresis CB was obtained. Series of experiments were performed to validate the proposed method. The experimental data showed high accordance with our prediction allowing the CB to be prepared easily with our proposed method. To further evaluate this method, C-PC was purified from crude extracts of Spirulina platensis with the selected separation condition. Results showed that C-PC was well separated from other phycobiliproteins that have similar physicochemical properties, and analytical grade product with purity up to 4.5 (A620/A280) was obtained.

In this work, we developed the concept of enzyme catalysis–electrophoresis titration (EC-ET) under ideal conditions, the theory of EC-ET for multiplex enzymatic assay (MEA), and a related method based on a moving reaction boundary (MRB) chip with a collateral channel and cell phone imaging. As a proof of principle, the model enzymes horseradish peroxidase (HRP), laccase and myeloperoxidase (MPO) were chosen for the tests of the EC-ET model. The experiments revealed that the EC-ET model could be achieved via coupling EC with ET within a MRB chip; particularly the MEA analyses of catalysis rate, maximum rate, activity, Km and Kcat could be conducted via a single run of the EC-ET chip, systemically demonstrating the validity of the EC-ET theory. Moreover, the developed method had these merits: (i) two orders of magnitude higher sensitivity than a fluorescence microplate reader, (ii) simplicity and low cost, and (iii) fairly rapid (30 min incubation, 20 s imaging) analysis, fair stability (

A multiple covalent crosslinked gel of poly(acrylamide-acrylic acid) was synthesized by using acrylic acid and acrylamide both as a comonomer and as a covalent crosslinker. The resultant gel showed a high crosslinking degree, tight micropores, but similar softness and marked separation capacity, in contrast to traditional polyacrylamide gels. The gel might serve as a novel matrix in bioseparation.

The traditional recycling free-flow isoelectric focusing (RFFIEF) suffered from complex structure, tedious operations and poor extensibility as well as high cost. To address these issues, a novel reciprocating free-flow isoelectric focusing device (ReFFIEF) was developed for proteins or peptides pre-fractionation. In the new device, a reciprocating background flow was for the first time introduced into free flow electrophoresis (FFE) system. The gas cushion injector (GCI) used in the previous continuous free-flow electrophoresis (CFFE) was redesigned for the reciprocating background flow. With the GCI, the reciprocating background flow could be achieved between the GCI, separation chamber and transient self-balance collector (tSBC). In a run, process fluid flowed to and from, forming a stable reciprocating fluid flow in the separation chamber. A pH gradient was created within the separation chamber, and at the same time proteins were focused repeatedly when passing through the chamber under perpendicular electric field. The ReFFIEF procedure was optimized for fractionations of three model proteins, and the optimized method was further used for pre-fractionation of model human serum samples. As compared with the traditional RFFIEF devices developed about 25 years ago, the new ReFFIEF system showed several merits, such as simple design and structure, user-friendly operation and easy to extend as well as low cost.

In this work, we proposed the concept of fluorescent speed sensing (FSS) of glycoprotein in chip supramolecular electrophoresis (CSE). To achieve a stable FSS run, we firstly synthesized a novel boronic acid-based diben-zoxanthene receptor (Rp3) by a simple one-pot reaction and the complex probe of F1@Rp3 containing Rp3 and fluorescein (F1), and performed a series of experiments to assess the complex probe's ability for recognition of saccharides and glycoproteins. Secondly, we revealed the mechanism of 'balance electromigration of supramolecular complex probe' in FSS and developed the relevant theory of FSS via a series of theoretical derivation. The FSS theory showed that the moving speed of F1 was a function of glycoprotein content, indicating a novel model of glycoprotein sensing. To demonstrate the validity and utility of developed model and theory, IgG and ovalbumin were selected as model glycoproteins for the relevant experiments. The results verified the validity of FSS concept to glycoprotein assay with good accuracy (less than 6.33 % difference from standard HPLC method), stability (less than 6.04 % RSD) and linearity (higher than R2 = 0.99) as well as fair sensitivity (8.48 ng). Finally, the FSS was successfully used for the analysis of HbA1c (a key biomarker in diabetes blood). The developed FSS concept and complex probe have great potential for glycoprotein sensing, particularly for the diagnosis of diabetes.

Melamine was sometimes adulterated to dairy products for false protein content increase in developing countries. However, a portable sensor has not been developed for on-spot determination of melamine in dairy products yet. Herein, a distance-based sensor was advanced for the quantification of melamine in dairy products based on chip electrophoretic titration (ET) of moving neutralization boundary (NB) and EDTA photocatalysis. In the chip sensor, EDTA, H2O2, and leucomalachite green (LMG) were added in the anode well. Under UV light, EDTA photocatalyzes H2O2 and colorless LMG as H2O and color malachite green (MG) with one positive charge. When applying an electric field, the MG in the anode well migrated into the channel and was neutralized with the base in the channel, resulting in colorless MG-OH and NB. Extracellular vesicles (EVs) are cell-derived microparticles present in most body fluids, mainly including microvesicles and exosomes. EV-harbored proteins have emerged as novel biomarkers for the diagnosis and prediction of different cancers. We successfully isolated microvesicles and exosomes from human saliva, which were further characterized comprehensively. Salivary EV protein profiling in normal subjects and lung cancer patients was systematically compared through utilizing LC-MS/MS-based label-free quantification. 785 and 910 proteins were identified from salivary exosomes and microvesicles, respectively. According to statistical analysis, 150 and 243 proteins were revealed as dysregulated candidates in exosomes and microvesicles for lung cancer. Among them, 25 and 40 proteins originally from distal organ cells were found in the salivary exosomes and microvesicles of lung cancer patients. In particular, 5 out of 25 and 9 out of 40 are lung-related proteins. Six potential candidates were selected for verification by Western blot, and four of them, namely, BPIFA1, CRNN, MUC5B, and IQGAP, were confirmed either in salivary microvesicles or in exosomes. Our data collectively demonstrate that salivary EVs harbor informative proteins that might be used for the detection of lung cancer through a noninvasive way.

Routine native immobilized pH gradient isoelectric focusing (IPG-IEF) and two-dimensional gel electrophoresis (2DE) are still suffering from unfortunate reproducibility, poor resolution (caused by protein precipitation) and instability in characterization of intact protein isoforms and posttranslational modifications. Based on the concept of moving reaction boundary (MRB), we firstly proposed a tunable non-IPG-IEF system to address these issues. By choosing proper pairs of catholyte and anolyte, we could achieve desired cathodic and anodic migrating pH gradients in non-IPG-IEF system, effectively eliminating protein precipitation and uncertainty of quantitation existing in routine IEF and 2DE, and enhancing the resolution and sensitivity of IEF. Then, an adjustable 2DE system was developed by combining non-IPG-IEF with polyacrylamide gel electrophoresis (PAGE). The improved 2DE was evaluated by testing model proteins and colon cancer cell lysates. The experiments revealed that (i) a tunable pH gradient could be designed via MRB; (ii) up to 1.65 fold improvement of resolution was achieved via non-IPG-IEF; (iii) the sensitivity of developed techniques was increased up to 2.7 folds; and (iv) up to about 16.4% more protein spots could be observed via the adjustable 2DE as compared with routine one. The developed techniques might contribute to complex proteome research, especially for screening of biological marker and analysis of extreme acidic/alkaline proteins.

Antimicrobial peptides (AMPs) are usually small and cationic biomolecules with broad-spectrum antimicrobial activities against pathogens. Purifying them from complex samples is essential to study their physiochemical properties. In this work, free-flow zone electrophoresis (FFZE) was utilized to purify AMPs from yeast fermentation broth. Meanwhile, gel filtration chromatography (GFC) was conducted for comparison. The separation efficiency was evaluated by SDS-PAGE analysis of the fractions from both methods. Our results demonstrated as follows: (i) FFZE had more than 30-fold higher processing capacity as compared with GFC; (ii) FFZE could achieve 87% purity and 89% recovery rate while in GFC these parameters were about 93 and 82%, respectively; (iii) the former had ∼2-fold dilution but the latter had ∼13-fold dilution. Furthermore, Tricine-SDS-PAGE, Native-PAGE, and gel IEF were carried out to characterize the purified AMPs. We found that two peptides existed as a pair with the molecular mass of ∼5.5 and 7.0 kDa, while the same pI 7.8. These two peptides were proved to have the antimicrobial activity through the standardized agar diffusion method. Therefore, FFZE could be used to continuously purify AMPs with high bioactivity, which will lead to its wide application in the clinical and pharmaceutical fields.

In this work, we developed the model and theory of free-flow moving reaction boundary electrophoresis (FFMRB) for continuous protein concentration for the first time. The theoretical results indicated that (i) the moving reaction boundary (MRB) can be quantitatively designed in free-flow electrophoresis (FFE) system; (ii) charge-to-mass ratio (Z/M) analysis could provide guidance for protein concentration optimization; and (iii) the maximum processing capacity could be predicted. To demonstrate the model and theory, three model proteins of hemoglobin (Hb), cytochrome C (Cyt C) and C-phycocyanin (C-PC) were chosen for the experiments. The experimental results verified that (i) stable MRBs with different velocities could be established in FFE apparatus with weak acid/weak base neutralization reaction system; (ii) proteins of Hb, Cyt C and C-PC were well concentrated with FFMRB; and (iii) a maximum processing capacity and recovery ratio of Cyt C enrichment were 126. mL/h and 95.5% respectively, and a maximum enrichment factor was achieved 12.6 times for Hb. All of the experiments demonstrated the protein concentration model and theory. In contrast to other methods, the continuous processing ability enables FFMRB to efficiently enrich diluted protein or peptide in large volume solution.

Uneven flow in free-flow electrophoresis (FFE) with a gravity-induced fraction collector caused by air bubbles in outlets and/or imbalance of the surface tension of collecting tubes would result in a poor separation. To solve these issues, this work describes a novel collector for FFE. The collector is composed of a self-balance unit, multisoft pipe flow controller, fraction collector, and vacuum pump. A negative pressure induced continuous air flow rapidly flowed through the self-balance unit, taking the background electrolyte and samples into the fraction collector. The developed collector has the following advantages: (i) supplying a stable and harmonious hydrodynamic environment in the separation chamber for FFE separation, (ii) effectively preventing background electrolyte and sample flow-back at the outlet of the chamber and improving the resolution, (iii) increasing the preparative scale of the separation, and (iv) simplifying the operation. In addition, the cost of the FFE device was reduced without using a multichannel peristaltic pump for sample collection. Finally, comparative FFE experiments on dyes, proteins, and cells were carried out. It is evident that the new developed collector could overcome the problems inherent in the previous gravity-induced self-balance collector.

Hemoglobin A1c (HbA1c) has been proven to be a key biomarker for diabetes screening, and glutathiolation of HbA1c (viz., GSS-HbA1c) has been identified. However, the impact of GSS-HbA1c on the measurement of HbA1c for diabetes screening has not been quantitatively assessed yet. To address the issue, the micropreparative capillary isoelectric focusing (cIEF) developed in our previous work was used for the high resolution separation and purification of hemoglobin (Hb) species. The main fractions of HbA0, HbA3 and HbA1c extracted from the developed cIEF were identified by validated Mono S method. The proposed GSS-HbA1c fractions in the cIEF were pooled and identified by electrospray ionization mass spectrometry (ESI-MS). The HbA1c enzyme-linked immunosorbent assay (ELISA) kit was employed for further quantitative analysis of GSS-HbA1c. A total of 34 blood samples with HbA1c levels from 4.2% to 13.4% were assessed via the above comprehensive strategy of IEF-HPLC-MS-ELISA. It was demonstrated that the HbA1c levels detected by cation exchange LC were considerably influenced by the glutathiolation of Hb and the range of detected GSS-HbA1c values was between 0.23% and 0.74%. The results and developed cIEF methods have considerable significances for investigation of diabetes and clinical diagnosis.

A moving neutralization boundary (MNB) was developed as a novel model of visual offline sample stacking for the trace analysis of heavy metal ions (HMIs) by capillary zone electrophoresis (CZE). In the stacking system, the motion direction of MNB to cathode was used with 2.1 mmol/L HCl-98 mmol/L KCl-trace metal ions in the anodic solution and 4.0 mmol/L NaOH-96 mmol/L KCl in the cathodic solution. The voltage was constant at 180 V and the flow rate of the anolyte and catholyte was 1 mL/min. The metal ions in the gel after stacking were detected by capillary electrophoresis. The calibration curves showed good linear relationship (r > or = 0.998 5) in the concentration range used in the experiments. The pre-concentration factors were up to 80 - 150 and the limits of detection (LODs) were 0.163, 0.256, 0.077, 0.153, 0.203, 0.062 and 0.142 mg/L for Cu(II), Zn(II), Ni(II), Mg(II), Ca(II), Cr(III) and Fe(III), respectively, obviously lower than the national standards. The intra-day and inter-day assay precisions were good (the relative standard deviations (RSDs) less than 7.42%). Finally, the developed method has been successfully used for the stacking and the detection of heavy metal ions in electroplate waste water.

We developed a novel polyacrylamide gel electrophoresis (PAGE) method to stack and separate human hemoglobins (Hbs) based on the concept of moving reaction boundary (MRB). This differs from the classic isotachophoresis (ITP)-based stacking PAGE in the aspect of buffer composition, including the electrode buffer (pH 8.62 Tris-Gly), sample buffer (pH 6.78 Tris-Gly), and separation buffer (pH 8.52 Tris-Gly). In the MRB-PAGE system, a transient MRB was formed between alkaline electrode buffer and acidic sample buffer, being designed to move toward the anode. Hbs carried partial positive charges in the sample buffer due to its pH below pI values of Hbs, resulting in electromigrating to the cathode. Hbs would carry negative charges quickly when migrated into the alkaline electrode buffer and be transported to the anode until meeting the sample buffer again. Thus, Hbs were stacked within a MRB until the transient MRB reached the separation buffer and then separated by zone electrophoresis with molecular sieve effect of the gel. The experimental results demonstrated that there were three clear and sharp protein zones of Hbs (HbA1c, HbA0, and HbA2) in MRB-PAGE, in contrast to only one protein zone (HbA0) in ITP-PAGE for large-volume loading (≥15 μl), indicating high stacking efficiency, separation resolution, and good sensitivity of MRB-PAGE. In addition, MRB-PAGE was performed in a conventional slab PAGE device, requiring no special device. Thus, it could be widely used in separation and analysis of diluted protein in a standard laboratory.

Here, a simple micro free-flow electrophoresis (μFFE) was developed for fluorescence sensing of monosaccharide via supermolecule interaction of synthesized boronic acid functionalized benzyl viologen (ο-BBV) and fluorescent dye. The μFFE contained two open electrode cavities and an ion-exchange membrane was sandwiched between two polymethylmethacrylate plates. The experiments demonstrated the following merits of developed μFFE: (i) up to 90.5% of voltage efficiency due to high conductivity of ion-exchange membrane; (ii) a strong ability against influence of bubble produced in two electrodes due to open design of electrode cavities; and (iii) reusable and washable separation chamber (45 mm × 17 mm × 100 μm, 77 μL) avoiding the discard of μFFE due to blockage of solute precipitation in chamber. Remarkably, the μFFE was first designed for the sensing of monosaccharide via the supermolecule interaction of synthesized ο-BBV, fluorescent dye, and monosaccharide. Under the optimized conditions, the minimum concentration of monosaccharide that could be detected was 1 × 10(-11) M. Finally, the developed device was used for the detection of 0.3 mM glucose spiked in human urine. All of the results demonstrated the feasibility of monosaccharide detection via the μFFE.

An immobilized pH gradient (IPG) has strong power against instability (e.g., drifting and plateau) existing in classic isoelectric focusing (IEF). However, the relevant mechanism against the instability of pH gradient is still unclear. In this work, the theories of diffusional current and water products in IEF were developed based on the Svensson–Tiselius's differential equation and concept of moving reaction boundary (MRB). Two novel methods of pH gradient mobilization in IPG–IEF and non-IPG–IEF (opposite to IPG–IEF) were developed to unveil stability mechanism of IPG–IEF. The theoretical and experimental results indicated that (i) the drifting of pH gradient in non-IPG–IEF could be effectively controlled by IPG technique due to the existence of equal-fluxes of hydroxyl and hydrogen ions in the IPG–IEF system, (ii) there existed high diffusional current in non-IPG–IEF because of the existence of free carrier ampholyte (CA), but weak current in the IPG–IEF due to the immobilization of CA species in gel matrix, and (iii) the high diffusional current resulted in a great amount of water formation in neutral zone of pH gradient that led to distinct plateau in non-IPG–IEF, conversely the weak diffusional current caused little of water formation and weak plateau of pH gradient in IPG–IEF. These studies have considerable significance to the understanding of mechanism and development of protein IEF separation technique.

In this paper, a mathematical model and computer simulator were developed for offline sample pretreatment of heavy metal ion based on moving precipitate boundary (MPB) electrophoresis. The simulation indicates that (i) the program can easily accomplish numerical computations, such as the velocities of MPB and elution boundary (EB), and enrichment factor (EF) etc; (ii) the simulator can vividly imitate the dynamics of MPB, EB, precipitate zone, and precipitate-elution; and (iii) the software may simply optimized experimental conditions via the influence factors (e.g., voltage, hydroxyl, hydrogen and metal ions) on the EF. As a proof of concept, copper ion and its precipitate with definite blue color were, respectively chosen as mode heavy metal ion and alkaline precipitate for the relevant experiments of MPB-based sample preconcentration of heavy metal ion in large tube. All of the experimental results manifest the validity of developed mathematical model and the relevant simulation results. The model and simulator advanced herein are of clear significance to the optimization of experimental conditions and understanding of offline MPB- based sample condensation of heavy metal ion.

Moving reaction boundary titration (MRBT) has a potential application to immunoassay and protein content analysis with high selectivity. However, air bubbles often impair the accuracy of MRBT, and the leakage of electrolyte greatly decreases the safety and convenience of electrophoretic titration. Addressing these two issues a reliable MRBT device with modified electrolyte chamber of protein titration was designed. Multiphysics computer simulation was conducted for optimization according to two-phase flow. The single chamber was made of two perpendicular cylinders with different diameters. After placing electrophoretic tube, the resident air in the junction next to the gel could be eliminated by a simple fast electrolyte flow. Removing the electrophoretic tube automatically prevented electrolyte leakage at the junction due to the gravity-induced negative pressure within the chamber. Moreover, the numerical simulation and experiments showed that the improved MRBT device has following advantages: (i) easy and rapid setup of electrophoretic tube within 20 s; (ii) simple and quick bubble dissipates from the chamber of titration within 2 s; (iii) no electrolyte leakage from the two chambers: and (iv) accurate protein titration and safe instrumental operation. The developed technique and apparatus greatly improves the performance of the previous MRBT device, and providing a new route toward practical application.

In this work, charge-to-mass ratio (C/M) and band broadening analyses were combined to provide better guidance for the design of free-flow zone electrophoresis carrier buffer (CB). First, the C/M analyses of hemoglobin and C-phycocyanin (C-PC) under different pH were performed by CLC Protein Workbench software. Second, band dispersion due to the initial bandwidth, diffusion, and hydrodynamic broadening were discussed, respectively. Based on the analyses of the C/M and band broadening, a better guidance for preparation of free-flow zone electrophoresis CB was obtained. Series of experiments were performed to validate the proposed method. The experimental data showed high accordance with our prediction allowing the CB to be prepared easily with our proposed method. To further evaluate this method, C-PC was purified from crude extracts of Spirulina platensis with the selected separation condition. Results showed that C-PC was well separated from other phycobiliproteins that have similar physicochemical properties, and analytical grade product with purity up to 4.5 (A620/A280) was obtained.

In this work, a simple and novel sheath-flow sample injection method (SFSIM) is introduced to reduce the band broadening of free-flow zone electrophoresis separation in newly developed self-balance free-flow electrophoresis instrument. A needle injector was placed in the center of the separation inlet, into which the BGE and sample solution were pumped simultaneously. BGE formed sheath flow outside the sample stream, resulting in less band broadening related to hydrodynamics and electrodynamics. Hemoglobin and C-phycocyanin were successfully separated by the proposed method in contrast to the poor separation of free-flow electrophoresis with the traditional injection method without sheath flow. About 3.75 times resolution enhancement could be achieved by sheath-flow sample injection method.

Herein, a simple novel free-flow electrophoresis (FFE) method was developed via introduction of organic solvent into the electrolyte system, increasing the solute solubility and throughput of the sample. As a proof of concept, phenazine-1-carboxylic acid (PCA) from Pseudomonas sp. M18 was selected as a model solute for the demonstration on feasibility of novel FFE method on account of its faint solubility in aqueous circumstance. In the developed method, the organic solvent was added into not only the sample buffer to improve the solubility of the solute, but also the background buffer to construct a uniform aqueous-organic circumstance. These factors of organic solvent percentage and types as well as pH value of background buffer were investigated for the purification of PCA in the FFE device via CE. The experiments revealed that the percentage and the types of organic solvent exerted major influence on the purification of PCA. Under the optimized conditions (30 mM phosphate buffer in 60:40 (v/v) water-methanol at an apparent pH 7.0, 3.26 mL/min background flux, 10-min residence time of injected sample, and 400 V), PCA could be continuously purified from its impurities. The flux of sample injection was 10.05 μL/min, and the recovery was up to 93.7%. An 11.9-fold improvement of throughput was found with a carrier buffer containing 40% (v/v) methanol, compared with the pure aqueous phase. The developed procedure is of evident significance for the purification of weak polarity solute via FFE.

Interval free-flow zone electrophoresis (FFZE) has been used to suppress sample band broadening greatly hindering the development of free-flow electrophoresis (FFE). However, there has been still no quantitative study on the resolution increase of interval FFZE. Herein, we tried to make a comparison between bandwidths in interval FFZE and continuous one. A commercial dye with methyl green and crystal violet was well chosen to show the bandwidth. The comparative experiments were conducted under the same sample loading of the model dye (viz. 3.49, 1.75, 1.17, and 0.88 mg/h), the same running time (viz. 5, 10, 15, and 20 min), and the same flux ratio between sample and background buffer (= 10.64 × 10⁻³). Under the given conditions, the experiments demonstrated that (i) the band broadening was evidently caused by hydrodynamic factor in continuous mode, and (ii) the interval mode could clearly eliminate the hydrodynamic broadening existing in continuous mode, greatly increasing the resolution of dye separation. Finally, the interval FFZE was successfully used for the complete separation of two-model antibiotics (herein pyoluteorin and phenazine-1-carboxylic acid coexisting in fermentation broth of a new strain Pseudomonas aeruginosa M18), demonstrating the feasibility of interval FFZE mode for separation of biomolecules.

In this paper, a moving neutralization boundary (MNB) electrophoresis is developed as a novel model of visual offline sample stacking for the trace analysis of heavy metal ions (HMIs). In the stacking system, the cathodic-direction motion MNB is designed with 1.95–2.8 mM HCl + 98 mM KCl in phase alfa and 4.0 mM NaOH + 96 mM KCl in phase beta. If a little of HMI is present in phase alfa, the metal ion electrically migrates towards the MNB and react with hydroxyl ion, producing precipitation and moving precipitation boundary (MPB). The alkaline precipitation is neutralized by hydrogen ion, leading to a moving eluting boundary (MEB), release of HMI from its precipitation, circle of HMI from the MEB to the MPB, and highly efficient visual stacking. As a proof of concept, a set of metal ions (Cu(II), Co(II), Mn(II), Pb(II) and Cr(III)) were chosen as the model HMIs and capillary electrophoresis (CE) was selected as an analytical tool for the experiments demonstrating the feasibility of MNB-based stacking. As shown in this paper, (i) the visual stacking model was manifested by the experiments; (ii) there was a controllable stacking of HMI in the MNB system; (iii) the offline stacking could achieve higher than 123 fold preconcentration; and (iv) the five HMIs were simultaneously stacked via the developed stacking technique for the trace analyses with the limits of detection (LOD): 3.67 × 10 −3 (Cu(II)), 1.67 × 10 −3 (Co(II), 4.17 × 10 −3 (Mn(II)), 4.6 × 10 −4 (Pb(II)) and 8.40 × 10 −4 mM (Cr(III)). Even the off-line stacking was demonstrated for the use of CE-based HMI analysis, it has potential applications in atomic absorption spectroscopy (AAS), inductively coupled plasma-mass spectrometry (ICP-MS) and ion chromatography (IC) etc.

In China, some forensic cases are caused by barbiturates. Thus, the determination of trace level barbiturates in body fluid is important for the poisoning investigation. In this study, an online large-volume sample stacking (LVSS) with polarity switching in capillary electrophoresis (CE) was applied for the sensitive determination of barbiturates. This technique involves injecting a large volume of sample into a capillary and removing the sample matrix plug out of the capillary by reversing the polarity. Quantitation limit obtained was 0.048, 0.057, 0.039, and 0.015 μg/mL for secobarbital, amobarbital, barbital, and phenobarbital (signal-to-noise ratio = 9). An increasing number of small biosamples (e.g. proteins and enzymes) need micropreparation in lab. However, neither large-scale free-flow electrophoresis (LS-FFE) nor chip FFE (C-FFE) could fit the growing demands. Herein, a simple quasi-chip FFE (QC-FFE) was constructed. In contrast to C-FFE, the features of QC-FFE are as follows: (i) its separation chamber is reassemblable and rewashable avoiding discard of C-FFE due to blockage of solute precipitation in chamber; (ii) its chamber size is 45 mm × 30 mm × (80-500) μm (108-654 μL volume) having function of micropreparation; (iii) there are up to 16 outlets in QC-FFE bestowing fine fraction for micropurification. The QC-FFE was used for the micropurification of model enzyme of self-digestible trypsin in crude pancreatin. Under the given conditions, the purification factor of enzyme was 11.7, the specific activity reached 6236 U/mg, the run time for 19 μL sample purification was 45 s and the throughput of trypsin was 3.34 mg/h, and the yield of pure trypsin was 55.2%. All of the results show the feasibility of enzyme micropreparation via QC-FFE. The developed device and procedure have potential use to other micropurification of protein or peptide sample.

In this paper, we describe the synthesis of a novel copper ion hapten using the copper mercaptide of penicillenic acid (CMPA) derived from penicillin. Results from tests with immune rabbits indicate that: (i) A new antigen synthesized with CMPA has good stability and is safe for immunizing animals with no toxic phenomena being found in animal experiments; (ii) the immunogenic antigen (CMPA-BSA) can stimulate the immune system to produce specific antibodies with high titrations, up to 150000; and (iii) antibodies in antisera showed higher affinity to OVA-GSH-CuCl than OVA-GSH, which indicates that the antibodies have specific affinity towards copper ions. These results confirm that the novel hapten and relevant antigen for copper ion have been successfully synthesized, giving progress towards an immunoassay for copper ions in environmental and food samples.

A large-scale free-flow electrophoresis (LS-FFE) is often too large for cell separation of lab scale, whereas micro-FFE (μFFE) has great difficulty in cell isolation due to easy blockage by cell accumulation in μFFE. In this study, a mid-scale FFE (MS-FFE) is developed for cell and protein separations. The volume of the separation chamber (70×40×0.1-0.8 mm) is from 280 μL to 2.24 mL, much lower than that in an LS-FFE but higher than that in a μFFE. Gravity is used for uniform flow of the background buffer only via a single pump with 16 channels and the sample is injected via an adjuster originally used for clinical intravenous injection. The experiments reveal that the hydrodynamic and electrohydrodynamic flows are much stable, and the Joule heat can be effectively dispersed without obvious positive or negative deviation as shown by the omega plots. By the device, Escherichia coli and Staphylococcus aureus, which easily accumulate to block μFFE and are separated with difficulty due to their same negative charges carried, can be well isolated under the conditions of 4.5 mM pH 8.5 Tris-boric buffer (4.5 mM Tris, 4.5 mM boric acid) with 0.10 mM ethylene diamine tetraacetic acid and 5% m/v sucrose, 200 μL/min, 800 V, and sample injection via inlet 4. The mid-scale FFE device could also be used for the separation of three model proteins of horse heart cytochrome c, myoglobin and bovine serum albumin. The device has clear significance for mid-scale separation of cells and proteins.

To demonstrate the theoretic method on the stacking of zwitterion with moving reaction boundary (MRB) in the accompanying paper, the relevant experiments were performed. The experimental results quantitatively show that (1) MRB velocity, including the comparisons between MRB and zwitterionic velocities, possesses key importance to the design of MRB stacking; (2) a much long front alkaline plug without sample should be injected before the sample injection for a complete stacking of zwitterion if sample buffer is prepared with strong base, conversely no such plug is needed if using a weak base as the sample buffer with proper concentration and pH value; (3) the presence of salt in MRB system holds dramatic effect on the MRB stacking if sample solution is a strong base, but has no effect if a weak alkali is used as sample solution; (4) all of the experiments of this paper, including the previous work, quantitatively manifest the theory and predictions shown in the accompanying paper. In addition, the so-called derivative MRB-induced re-stacking and transient FASI-induced re-stacking were also observed during the experiments, and the relevant mechanisms were briefly demonstrated with the results. The theory and its calculation procedures developed in the accompanying paper can be well used for the predictions to the MRB stacking of zwitterion in CE.

A sensitive analytical method based on capillary electrophoresis with field-amplified sample stacking, Soxhlet’s extraction and ultrasonic extraction for studying the degradation of trace amounts of pyoluteorin (Plt, a broad-spectrum fungicide produced by fluorescent Pseudomonas) in soil samples was first developed. The limits of detection and the limits of quantification for Plt of the developed method were 0.107 and 0.36 μg mL−1, respectively. The recoveries of Plt at three spiking concentrations of 500, 200, and 50 μg kg−1 were 94.18, 79, and 78.25%, respectively. In this communication, we successfully performed the investigation on the dynamic residue of Plt during its decomposition in agriculture soil using the method developed. The calculated t 1/2 values for Plt residue in near-surface and rhizosphere soil were 42.26 and 32.84 h, respectively. The results show that Plt has good environmental compatibility.

Ganoderic acids (GAs) were bioactive secondary metabolites produced by a traditional mushroom Ganoderma lucidum. We describe a simple and efficient method for the separation and quantitative determination of four GAs, namely Ganoderic acid T (GA-T), Ganoderic acid Mk (GA-Mk), Ganoderic acid Me (GA-Me) and Ganoderic acid S (GA-S) from dried triterpene-enriched extracts of G. lucidum mycelia powder by capillary zone electrophoresis (CZE). Under the optimum conditions, the four GAs reached the baseline separation in 9 min with Glycyrrhetinic acid (GTA) as internal standard. The four GAs and internal standard (GTA) were detected at a wavelength 245 nm. All calibration curves showed good linearity (r(2)>0.9958) within test ranges. Limit of detection (LOD) and limit of quantification (LOQ) were less than 0.6 and 1.8 microg/mL, respectively. The relative standard deviation (R.S.D.) values of precision and recoveries were less than 5% and recoveries ranged from 91.4% to 103.6%. This was the first report on simultaneous determination of the four GAs and the results provided a firm basis for the trace analysis of GAs in dried fermentation mycelia powder of G. lucidum with high accuracy.

In this paper, the concept of a moving interaction boundary (MIB) is proposed with regard to guest and host molecules. With 2-naphthalene-sulfonate (2-NS) and β-cyclodextrin (CD) as the model guest and host compounds, respectively, the relevant experiments were carried out on the MIB in capillary electrophoresis (CE). The experiments show that (1) there are a MIB and a complex boundary (CB) if proper guest and host molecules are used; (2) the MIB system has the characteristic of selective focusing and probing of the target 2-NS; (3) the system also has the characteristic of selective probing of the target host molecule β-CD without UV-absorbance, making the direct UV determination of β-CD from other CDs possible; (4) interestingly, the focusing of the guest molecule is a kind of leaky-sample stacking rather than a collection of analytes in sample sweeping; (5) the mechanism of MIB-induced separation of target analyte from unwanted ones is similar to but different from that of an affinity chromatography. In addition, the utility of MIB was briefly tested for a real sample of wastewater spiked with 2-NS.

This paper describes a novel free-flow electrophoresis (FFE), which is joined with gratis gravity, gas cushion injector (GCI) and self-balance collector instead of multiple channel pump, for the purpose of preparative purification. The FFE was evaluated by systemic experiments. The results manifest that (i) even though one-channel peristaltic pump is used for the driving of background buffer, there is still stable flow in the FFE chamber; (ii) the stable flow is induced by the gravity-induced pressure due to the difference of buffer surfaces in the GCI and self-balance collector; (iii) the pulse flow of background buffer induced by the peristaltic pump is greatly reduced by the GCI with good compressibility of included air; (iv) the FFE can be well used for zone electrophoretic separation of amino acids; (v) up to 20 inlets simultaneous sample injection and up to five to tenfold condensation of amino acid can be achieved by combining the FFE device with the method of moving reaction boundary. To the best of authors' knowledge, FFE has not been used for such separation and condensation of amino acids. The relevant results achieved in the paper have evident significance for the development of preparative FFE.

This paper introduces a mathematic mode of moving chelation boundary (MCB) for computer simulation of a continuous EDTA-based sample sweeping in capillary electrophoresis (CE). Besides the equations of MCB used herein, the mode also includes electro-neutrality equation, constant current density, jump boundary condition of MCB, Kohlrausch' regulating function expressed in MCB formulation, product of water, ionic apparent mobility, ionic strength and conductivity of electrolyte as well as simple equilibrium reaction, etc. The simulation software is developed based on the mode. With the software the relevant simulation is carried out, and the corresponding experiments on a MCB are performed. The results on the simulation and experiments demonstrate that (1) the software can simulate a dynamic process, characteristic peak shape and relevant electrophoregram of a MCB; (2) the simulator can quantitatively compute velocities of MCB and complex boundary (CB), all of ionic concentrations (especially the concentration of complex) and sweeping efficiency; (3) these simulation results mentioned above are generally in accordance with the experiments. The simulation software holds evident significances for the study on a MCB and conditional optimization in such an EDTA-based sample sweeping of metal ion in CE.
