Objective To investigate the effect of pathogenic bacterium-Porphyromonas gingivalis (P.g) on the proliferation and inflammatory factor expression of human colorectal cancer Caco-2 cells, to determine whether the Janus kinase 2-signal transducers and activators of transcription 3 (JAK2-STAT3) pathway is involved in the regulation of Caco-2 cell proliferation by P.g and to provide an experimental basis for further exploring the relationship between P.g and colorectal cancer. Methods Caco-2 cells were cultured in vitro, and P.g at different multiplicities of infection (MOIs) (0, 1, 10, 25) was selected to stimulate for 12, 24 and 48 h. The effect of P.g on the proliferation of Caco-2 cells was detected by CCK8. The stimulation time was set as 12, 24 and 48 h. MOI=0 was the control group, and MOI=1, 10 and 25 comprised the experimental group. qRT-PCR and Western blot were used to detect the changes in interleukin-6 (IL-6), interleukin-10(IL-10), JAK2 and STAT3 gene and protein (phosphorylated protein) levels in each group. Results After P.g infection of Caco-2 cells, P.g had a sustained stimulatory effect on the cells for 12, 24 and 48 h at MOI=1 and MOI=10 compared with the control group. Hyperglycemia drives dysfunction of the intestinal barrier. 5-Hydroxytryptaine 4 receptor (5-HT4R) agonists have been considered therapeutics for constipation in clnic. However, the roles of 5-HT4R activation in mucosa should be fully realized. Here, we investigate the effects of 5-HT4R activation on diabetes-induced disruption of the tight junction (TJ) barrier in the colon. Not surprisingly, the TJ barrier in diabetic mice with or without 5-HT4R is tremendously destroyed, as indicated by increased serum fluorescein isothiocyanate (FITC)-dextran and decreased transepithelial electrical resistance (TER). Simultaneously, decreased expressions of TJ proteins are shown in both wild-type (WT) and 5-HT4R knockout (KO) mice with diabetes. Notably, chronic treatment with intraperitoneal injection of a 5-HT4R agonist in WT mice with diabetes repairs the TJ barrier and promotes TJ protein expressions, including occludin, claudin-1 and ZO-1, in the colon, whereas a 5-HT4R agonist does not improve TJ barrier function or TJ protein expressions in 5-HT4R KO mice with diabetes. Furthermore, stimulation of 5-HT4R inhibits diabetes-induced upregulation of myosin light chain kinase (MLCK), Rho-associated coiled coil protein kinase 1 (ROCK1), and phosphorylated myosin light chain (p-MLC), which are key molecules that regulate TJ integrity, in the colonic mucosa of WT mice. However, such action induced by a 5-HT4R agonist is not observed in 5-HT4R KO mice with diabetes. These findings indicate that 5-HT4R activation may restore TJ integrity by inhibiting the expressions of MLCK, ROCK1 and p-MLC, improving epithelial barrier function in diabetes.

To provide regulatory elements for the regulation of metabolic fluxes in Saccharomyces cerevisiae cells and to control the efficient and precise expression of key genes at the transcriptional level in the cells, cloning and characterization studies of partial promoter elements of S. cerevisiae were carried out. The pRS41H-Promoter-EGFP expression vector was constructed using OE-PCR and enzymatic ligation using EGFP (enhanced green fluorescent protein) as the reporter gene. The relative strength and characterization of the S. cerevisiae promoters were characterized by measuring the fluorescence value of EGFP. The characterization results show that six new S. cerevisiae promoters are screened and cloned, namely PCBFI, PPAFI, PPOLI, PPOLII, PPOLIII and PRTFI, and their relative strengths are PPOLI>PPAFI>PPOLIII>PPOLII>PRTFI>PCBFI. Finally, the expression of promoter PPOLI in S. cerevisiae cells was investigated. It was found that PPOLI can continue to function in S. cerevisiae cells as they grow. This study provide a reference for the application of promoters in the regulation of metabolic flow homeostasis in chassis cells.

In this paper, the synthesis of ginsenoside Rh2 was catalyzed by using heterologous expression of Bacillus subtilis SL-44 glycosyltransferase. The synthesis parameters of ginsenoside Rh2 were optimized by the selection of strains and chemical supplies, the establishment of kinetic equations for the respiration rate of UGT enzyme, the effect of storage temperature on the model, and the glycosylation reaction of ginsenoside PPD with UGT. The effect of Rh2 saturation on the thermal denaturation temperature of the protein was analyzed along with the kinetic properties of the enzyme GE02773 (GE03484) while varying the saturation of Rh2. The results showed that the conversion of ginsenoside Rh2 reached 84% at a temperature of 35℃, pH 8, 5% DMSO, 0.4 of M-UDPG, and 1M-PPD in reaction with GE02773. In this paper, we successfully achieved the efficient synthesis of ginsenoside Rh2, which provides new ways and ideas for the application and synthesis of ginsenoside Rh2, with important practical significance and scientific value.

In this paper, the HSV color space model is used to collect the color characteristics of modern interior design and classify them into three types: hue (H), saturation (S), and luminance (V). At the same time, it combines with the theory of color psychology to construct a PAD emotion model for interior design. Then, it selects experimental samples and data analysis tools to carry out an example analysis of interior design from the emotional perspective. The data show that the selection of people warm and bright color interior design in the interior design of the background color across the amplitude of the largest and more concentrated in the 47.5°, the main body color across the amplitude of the smallest and most stable, the accent color value of the whole relatively high, the highest 123.6°, the background color saturation is the highest of 34.6°, and the brightness of more than 63.9°, the overall presentation of the warmth of comfort indoor environment. Error-values were 0.004, 0.0698, and 0.038 in order, with the errors all below 10%. This study integrates psychology and color into interior emotional design to better create visually appropriate, comfortable, and culturally rich interior environments for people, and also to promote the development of the interior design field.

The endocrown, a modified overlay or crown, is considered to have the advantages of adhesive retention and mechanical retention. Marginal adaptation and mechanical strength are common criteria for evaluating endocrowns. This review studied these aspects of endocrowns to guide further clinical application. Results from previous academic studies indicate that endocrowns made of different materials, such as glass-based ceramics, zirconia-based ceramics and resin-based ceramics, exhibit clinically acceptable marginal adaptation. Zirconia is a common dental material, but when used in endocrowns, it tends to present a risk of irreparable tooth root fractures. Compared with products manufactured with zirconia, endocrowns manufactured with resin-based ceramics and glass-based ceramics, such as lithium disilicate ceramics, do not tend to cause irreparable results, such as tooth root fracture, but their mechanical strengths are not as good as those of zirconia. At the same time, the tooth prepration design such as the types of endocrowns, the pulp chamber extension depth and angles, the endocrown thickness can influence the mechanical strength of endocrowns as well. Compared with traditional zirconia, self-glazed zirconia, a new type of material used to restore defective teeth, has the similar elasticity modulus（210 GPa）and better aesthetic advantages, and is suitable for short crown patients with insufficient occlusal space. Further study is warranted to improve the performance of endocrowns made from ceramic materials to prevent root fracture.

The stitched foam-core sandwich composites with different stitching methods and stitch density were manufactured using vacuum assisted resin infusion (VARI) molding process. The effect of stitching parameters on their flatwise tensile, three-point flexural, core shear and climbing drum peel properties was investigated. The results show that although stitching makes flatwise tensile strength and core shear strength of foam-core sandwich composites to decrease obviously, it can improve flexural properties as well as substantially enhance climbing drum peel properties. Modified lock stitch method is better than tufting stitch method. Properly increasing stitch space has certain positive effect for mechanical properties, but it is not conducive to the enhancement of climbing drum peel properties. For modified lock stitched foam-core sandwich composites with the stitch density of 30 mm×10 mm, flatwise tensile strength and core shear strength are reduced by 14.75% and 24.79%, respectively, whereas flexural strength and average peel strength are respectively increased by 7.96% and 80.78% compared to those of unstitched foam-core sandwich composites.

In recent years, due to precise control of the amorphous mineral precursor in the demineralization of dentine collagen fibers in orderly deposition, forming apatite crystals similar to the natural mineralized dentin, the bottom-up remineralization approach which does not depend on the existence of seed crystallites, dentin biomimetic mineralization techniques gradually become a hotspot in the research field of restoration of demineralized dentin caused by dental caries. This paper reviews the changing concepts and practices of the remineralization of demineralized dentin, emphasizing biomimetic remineralization studies. The results of the literature review show that the traditional dentin remineralization method is usually a disordered mixture of demineralized dentin and minerals, so mineralized dentin is not comparable to natural mineralized dentin in terms of the morphological characteristics and mechanical properties. With its gradual increase in recent years, dentine biomimetic mineralization technology perfectly resembles the minerals in the dentin overlapping sequence arranged with the dentine collagen fiber structure characteristics, leading to greatly improved microstructural, physical and chemical properties. As a result, dentine biomimetic mineralization technology is expected to achieve new breakthroughs in the fields of resin-dentin bonding mixing layers and the decay of dentin. At present, the technical obstacles that need to be overcome in the clinical application of the biomimetic remineralization of dentin are how to continuously supplement all the active ingredients needed for mineralization in the process of remineralization and how to keep the mechanical properties of the parent material unchanged while slowly releasing all ingredients. Researchers have successively proposed three-step transportation of the biomimetic remineralization of raw materials, as well as the preparation of mineralization precursors stabilized by polymers in advance and the reuse of mesoporous silicon nanomaterials for the transportation of the mineralized ingredient system. The concept described above provides the preliminary in vitro experimental basis for the transformation of the biomimetic remineralization strategy of dentin in clinical applications.

The multilayer soil moisture Internet of things sensor is designed to monitor the moisture of multiple soil profiles in real time. Its sensitivity and accuracy are of great concern to improve the performance of sensors. This article introduces the system composition of the end-cloud integrated multilayer soil moisture Internet of things sensor and then focuses on the design of key technologies, such as the moisture detection circuit, the time division multiplexing detection technology, and the deredundancy circuit in the analog–digital integrated design. The performance of the soil moisture detection circuit is directly related to the measurement accuracy of the sensor. A detection method is proposed using a high-frequency double-resonance circuit, which can detect small changes in moisture by changing the circuit detuning voltage. The maximum root mean square error of the calibration is less than 1.35% for five typical soils from different places. Compared with that of an independent detection method, the output consistency of the time division multiplexing detection is significantly improved by using the time division multiplexing detection method, which has a root mean square error of only 0.12%. In order to reduce errors caused by inconsistency in each burial, the gravimetric analysis is used in the sensitivity monitoring test, which shows that small changes in soil moisture can be detected by the circuit.

With the continuous reduction of renewable resources and the increasing awareness of environmental protection, the phenomenon of replacing oil with electricity has become more and more common. Electric vehicles are an obvious example. In the past few years, electric vehicles have developed rapidly and become more and more popular in people’s lives. However, imperfect technology has caused frequent safety accidents. The main reason for this is the charging safety risk. In this paper, there are many complex factors affecting the charging safety of electric vehicles in terms of the safety of electric vehicle charging and the energy and data exchange direction between charging piles and electric vehicles. Based on the analysis of the factors that affect the charging safety of electric vehicles, this paper combines big data technology to study the charging safety of electric vehicles.

The purpose of this paper was to investigate the surface modification of zeolite with polyethylene glycol (PEG) and the modified zeolite as a reinforcing filler partly replacing carbon black (CB) into rubber to prepare the rubber composites. The surface characteristics of zeolite modified by PEG and the unmodified zeolite were characterized by infrared (IR) spectroscopy and scanning electron microscopy (SEM). Moreover, the evaluations of the zeolite/rubber composite were carried out on characteristics such as tensile property, abrasion loss, and hysteresis heat generation with different amounts of zeolite and PEG. The results showed that the tensile strength and elongation at break of the composites were improved, and the abrasion and lag heat generation were reduced when 5–8 phr zeolite modified by 3 wt% PEG was filled into rubber. These results indicated that the modified zeolite has a good compatibility and dispersion in rubber.

Airport terminal is a typical large space building with large high interspaces and multitudinous functional zones. The task of heating, ventilation and air conditioning (HVAC) system is to ensure the thermal environment of these zones with different functions. A typical terminal in cold region of China is investigated in present paper, where indoor environment and performance of air conditioning system in summer are emphasized. The results indicate that the indoor temperature and humidity parameters at a height of 1.5 m in each zone of the terminal can be effectively controlled. The indoor temperature at a height of 1.5 varies in the range of 24~27°C, and the temperature difference is 0.5~4.9°C in the vertical direction. The temperature at the top of the terminal is greatly affected by the outdoor temperature where is higher and fluctuates obviously, while the temperature at the height of the occupant area is lower and more stable under the control of the air-conditioning system. The indoor carbon dioxide concentration is also at a reasonable level, all below 900 ppm, indicating that the equivalent outdoor air volume per capita is greater than 35 m3/h. Even in the case of high load caused by flight delays, the thermal environment in the terminal can be basically controlled at a comfortable level. As for the air handling unit, air handling process is tested to investigate the cooling capacity. The results show that the actual water supply of the AHU is relatively higher than the set value, but the air volume is almost equal to the set value which leads to an unsatisfactory energy efficiency. The tested energy efficiency ratio (EER) of AHU is only 5~7, almost approaching to that of the cooling plant. Reducing air flow rate could greatly improve the EER of the current air-conditioning system.
