Abstract

Fanconi anemia (FA) is an inheritable disorder that presents with bone marrow failure, developmental anomalies, and an increased susceptibility to cancer. The etiology of this condition stems from a genetic mutation that disrupts the proper repair of interstrand DNA cross-links (ICLs). The resultant dysregulation of the DNA damage response mechanism can induce genomic instability, thereby elevating the mutation rates and the likelihood of developing cancer. The FA pathway assumes a pivotal role in safeguarding genome stability through its involvement in the repair of DNA cross-links and the maintenance of overall genomic integrity. A mutation in the germ line of any of the genes responsible for encoding the FA protein results in the development of FA. The prevalence of aberrant FA gene expression in somatic cancer, coupled with the identification of a connection between FA pathway activation and resistance to chemotherapy, has solidified the correlation between the FA pathway and cancer. Consequently, targeted therapies that exploit FA pathway gene abnormalities are being progressively developed and implemented. This review critically examines the involvement of the FA protein in the repair of ICLs, the regulation of the FA signaling network, and its implications in cancer pathogenesis and prognosis. Additionally, it explores the potential utility of small-molecule inhibitors that target the FA pathway.

2. Study on the Path of Coupled and Coordinated Development of New Urbanization and Ecological Environment in Ya’an City

Abstract

Rapid urbanisation in Ya’an City, Sichuan Province, has caused environmental pollution and energy depletion in all districts, and the situation of coupled coordinated development is critical. In this study, with the help of PSR model, indicators are established and analysed from two aspects, namely, new urbanization and ecological environment, and a coupled coordinated development evaluation model is constructed. Finally, based on the calculation results, it is concluded that there are large gaps in the coupled and coordinated development of the eight districts and counties, and the problem of unbalanced and insufficient development is prominent. This study puts forward relevant path analyses and suggestions.

10. Adapting Transformer Language Models for Predictive Typing in Brain-Computer Interfaces

Abstract

Brain-computer interfaces (BCI) are an important mode of alternative and augmentative communication for many people. Unlike keyboards, many BCI systems do not display even the 26 letters of English at one time, let alone all the symbols in more complex systems. Using language models to make character-level predictions, therefore, can greatly speed up BCI typing (Ghosh and Kristensson, 2017). While most existing BCI systems employ character n-gram models or no LM at all, this paper adapts several wordpiece-level Transformer LMs to make character predictions and evaluates them on typing tasks. GPT-2 fares best on clean text, but different LMs react differently to noisy histories. We further analyze the effect of character positions in a word and context lengths.

Abstract

Perception of threats is essential for survival. Previous findings suggest that parallel pathways independently relay innate threat signals from different sensory modalities to multiple brain areas, such as the midbrain and hypothalamus, for immediate avoidance. Yet little is known about whether and how multi-sensory innate threat cues are integrated and conveyed from each sensory modality to the amygdala, a critical brain area for threat perception and learning. Here, we report that neurons expressing calcitonin gene-related peptide (CGRP) in the parvocellular subparafascicular nucleus in the thalamus and external lateral parabrachial nucleus in the brainstem respond to multi-sensory threat cues from various sensory modalities and relay negative valence to the lateral and central amygdala, respectively. Both CGRP populations and their amygdala projections are required for multi-sensory threat perception and aversive memory formation. The identification of unified innate threat pathways may provide insights into developing therapeutic candidates for innate fear-related disorders.

Abstract

Bony fusion caused by pathological new bone formation manifests the clinical feature of ankylosing spondylitis (AS). However, the underlying mechanism remains elusive. Here we discovered spontaneous kyphosis, arthritis and bony fusion in mature CD4-Cre;Ptpn11f/f mice, which present the pathophysiological features of AS. A population of CD4-Cre-expressing proliferating chondrocytes was SHP2 deficient, which could differentiate into pre-hypertrophic and hypertrophic chondrocytes. Functionally, SHP2 deficiency in chondrocytes impeded the fusion of epiphyseal plate and promoted chondrogenesis in joint cavity and enthesis. Mechanistically, aberrant chondrocytes promoted ectopic new bone formation through BMP6/pSmad1/5 signaling. It is worth emphasizing that such pathological thickness of growth plates was evident in adolescent humans with enthesitis-related arthritis, which could progress to AS in adulthood. Targeting dysfunctional chondrogenesis with Smo inhibitor sonidegib significantly alleviated the AS-like bone disease in mice. These findings suggest that blockade of chondrogenesis by sonidegib would be a drug repurposing strategy for AS treatment.

Abstract

Opioid-induced respiratory depression (OIRD) causes death following an opioid overdose, yet the neurobiological mechanisms of this process are not well understood. Here, we show that neurons within the lateral parabrachial nucleus that express the µ-opioid receptor (PBL Oprm1 neurons) are involved in OIRD pathogenesis. PBL Oprm1 neuronal activity is tightly correlated with respiratory rate, and this correlation is abolished following morphine injection. Chemogenetic inactivation of PBL Oprm1 neurons mimics OIRD in mice, whereas their chemogenetic activation following morphine injection rescues respiratory rhythms to baseline levels. We identified several excitatory G protein-coupled receptors expressed by PBL Oprm1 neurons and show that agonists for these receptors restore breathing rates in mice experiencing OIRD. Thus, PBL Oprm1 neurons are critical for OIRD pathogenesis, providing a promising therapeutic target for treating OIRD in patients.

46. On the Idiosyncrasies of the Mandarin Chinese Classifier System

Abstract

While idiosyncrasies of the Chinese classifier system have been a richly studied topic among linguists (Adams and Conklin, 1973; Erbaugh, 1986; Lakoff, 1986), not much work has been done to quantify them with statistical methods. In this paper, we introduce an information-theoretic approach to measuring idiosyncrasy; we examine how much the uncertainty in Mandarin Chinese classifiers can be reduced by knowing semantic information about the nouns that the classifiers modify. Using the empirical distribution of classifiers from the parsed Chinese Gigaword corpus (Graff et al., 2005), we compute the mutual information (in bits) between the distribution over classifiers and distributions over other linguistic quantities. We investigate whether semantic classes of nouns and adjectives differ in how much they reduce uncertainty in classifier choice, and find that it is not fully idiosyncratic; while there are no obvious trends for the majority of semantic classes, shape nouns reduce uncertainty in classifier choice the most.
