5. Numerical study of three-dimensional PIC for the surface plasmon excitation based on Drude model

This paper explores the time-domain equations of noble metals, in which Drude model is adopted to describe the dielectric constant, to implement three-dimensional particle-in-cell (PIC) simulations for the surface plasmon excitation with the finite-difference time-domain method (FDTD). A three-dimensional model for an electron bunch movement near the metal film is constructed, and particle-in-cell (PIC) simulations are carried out with various metal films of different thicknesses. The frequency of surface plasmon obtained from PIC simulation is agreed with that from theory. Furthermore, the surface plasmon wave properties of excitation and propagation with the metal film is summarized by PIC results.

8. Association between internal pentachlorophenol exposure characteristics and thyroid hormone indices in a community population in Shanghai, China

ObjectiveTo assess the level of internal exposure to PCP in a community population in Shanghai, to investigate the factors affecting the level of PCP, and to analyze the correlation between the exposure and thyroid hormone levels.MethodsA total of 464 residents of a community in Shanghai were selected as the study subjects. A questionnaire survey was conducted to obtain the demographic information, dietary situation, lifestyle and behavioral habits, and disease history of the individuals, and blood samples were collected. Gas chromatography-electron trap was applied to determine the PCP levels in serum. Multicategorical logistic regression analysis was used to analyze the possible influencing factors of PCP exposure in humans. Thyroid hormone levels were used as the dependent variable and serum PCP as the independent variable. Multiple linear regression analysis was used to assess the association between PCP and thyroid hormones in the community population after controlling the confounding factors such as age, gender, literacy, annual personal income, and chronic diseases.ResultsThe detection rate of serum PCP in 464 subjects was 90.3%, and the median serum PCP level was 0.43 μg·L-1. The differences in PCP levels among different age groups were statistically significant. There were no significant differences in PCP levels among different gender and BMI groups. The study of PCP exposure factors showed that age, frequency of using plastic products, consumption of freshwater fish, type of occupation, annual income, and consumption of tea or coffee were the potential influencing factors for PCP exposure. Among them, age, frequency of using plastic products, consumption of tea or coffee, and consumption of freshwater fish were positively associated with PCP levels, and annual personal income was negatively associated with it. The results of multiple linear regression analysis showed that among men, PCP levels were positively correlated with TSH (b=0.105, 95%CI:0.017‒0.313) and negatively correlated with FT4 (b=-0.026, 95%CI:-0.057‒0.004), and among women, PCP levels were positively correlated with TSH (b=0.092, 95%CI:-0.211‒0.904) and FT3 (b=0.017, 95%CI:-0.058‒0.230) and negatively correlated with FT4 (b=-0.013, 95%CI:-0.011‒0.037).ConclusionSerum PCP detection is common among community residents in Shanghai. Different demographic characteristics or behavioral habits may increase or decrease PCP exposure. PCP exposure then affects human thyroid hormone levels.

9. Field Simulation of Runoff and Sand Production on Degraded and Revegetated Slopes in Alpine Meadows Under Rainfall Conditions

[Objective] The Qinghai-Tibetan Plateau is an important ecological barrier in China, and alpine meadows are an important part of it, and the function of soil and water conservation is an important ecological service function of alpine meadows. Therefore, it is of great significance to carry out the study of soil erosion process in alpine meadows on the Tibetan Plateau to reduce anthropogenic disturbance of soil erosion and to safeguard the ecological security of the southwest region. [Methods] In this study, five different types of slopes, namely, undegraded, mildly degraded, moderately degraded, bare slopes and artificially restored slopes in alpine meadows of the Tibetan Plateau, were selected to analyze the process of flow and sand production on degraded and restored slopes under different rainfall intensities, and to reveal the relationship between the erosion mechanism and water-sand relationship between degraded slopes and restored slopes with artificial vegetation through the simulated rainfall test in the field. [Results] (1) The initial flow production time is relatively short on the undegraded slopes of alpine meadows and the restored slopes with artificial vegetation because of their dense root system, and the water infiltration ability is relatively weak; while on the mildly degraded slopes, the lateral flow and vertical infiltration ability are relatively strong, and the initial flow production time is relatively long. (2) Under regular rainfall conditions (30 and 60 mm/h), the flow rate of mildly degraded slopes is the smallest, and the flow reduction benefit is the highest, reaching 77.13%, the flow rate of artificially restored slopes is close to that of undegraded slopes and is slightly higher than that of mildly degraded slopes; the cumulative flow rate of bare soil slopes is the largest, and it is significantly higher than other slopes; the flow rate of undegraded slopes under extreme rainfall conditions (90 mm/h) is surging, only lower than that of bare slopes and significantly higher than other slopes. Under extreme rainfall conditions (90 mm/h), the flow rate of undegraded slopes increased, which was only lower than that of bare slopes and significantly higher than that of artificially restored slopes. (3) Under regular rainfall conditions (30 and 60 mm/h), the sand reduction benefit of the artificially restored vegetated slopes was the highest, reaching 81.97% and 89.82%, followed by the undegraded slopes, and with the increase of the degradation degree of the vegetation, the sand reduction benefit was gradually reduced; however, under extreme rainfall conditions, the sand reduction benefit of the artificially restored vegetation and the undegraded slopes was almost the same, and the sand production of the moderately degraded slopes was higher than the undegraded slopes, which is significantly higher than the other slopes. However, under extreme rainfall conditions, the sand reduction benefits of artificially restored vegetation and undegraded slopes are almost the same, while the sand production of moderately degraded slopes is 4.59 times of that of undegraded slopes. [Conclusion] The results of these studies can provide important scientific basis for vegetation restoration and ecological security in alpine meadow area.

10. Reliability of Girder Bridge System under Lateral Uneven Vehicular Overloads

Truck presence on bridges is random. Trucks may centrically or eccentrically appear on a bridge span in one or more lanes, which will cause extra load effects on girder components and eventually influence structural performance. Especially when trucks are heavily loaded, their eccentric passage is possible to induce damage in components, and, thus, the prescribed load- delivery path will be changed among components within the system. Therefore, component- and system-level structural performance of bridges subjected to aggressive vehicular overloads need to be thoroughly addressed. In this study, a WIM-based 5-axle overloaded truck model is chosen, and it is gradually applied to the finite element models of two girder bridges with different eccentric distance, considering two loading scenarios of single truck and multiple trucks. In detail, trucks are transversely loaded with an incremental eccentric distance, to investigate the impacts of eccentric loadings on structural performance, including failure sequences of components, load distribution factors among components, as well as system redundancy, as structures entering nonlinear stage. Finally, the reliability indices of the two bridges under uneven overloads are assessed at the component level and system level, respectively. The results of this study were beneficial to structural evaluation bridges subjected to overloads to ensure their serviceability and integrity.

Regeneration of adsorption saturated activated carbon by traditional electrochemical technology has the advantages of high regeneration efficiency and no mass loss of activated carbon, but it has defects such as low mineralization efficiency, generation of toxic by-products, and high energy consumption. Electrochemical advanced oxidation(E-AOP) regeneration technology can effectively overcome the problems of traditional electrochemical regeneration. In this paper, the regeneration efficiency and mechanism of activated carbon by traditional electrochemical are briefly described, and the effects of operating parameters (current, electrolyte type and concentration, regeneration location,anode material, etc.) on the desorption-adsorption balance and the mineralization efficiency of pollutants are analyzed. The mechanism and application of three types of E-AOP regeneration technologies(electro-Fenton regeneration, electro-peroxone regeneration, and electro-persulfate regeneration) in the process of activated carbon regeneration are summarized. The electric-Fenton regeneration technology has better regeneration effect, and the problem of metal ion pollution in the system can be effectively overcome by new regeneration. The electro-peroxone regeneration technology does not require the addition of any chemicals and catalysts, which is conducive to the control of reaction conditions and automation of the regeneration process. The electro-persulfate regeneration technology is convenient in operation with low energy consumption and wide applicability. Finally, the problems and development prospects of E-AOP regeneration technology are proposed. This review provides theoretical basis for the development of new efficient and environmentally friendly activated carbon regeneration technologies.

Internal circulation(IC) denitrification reactor was designed on the basis of IC anaerobic reactor,and the engineering application was investigated for the reverse osmosis concentrated water treatment. The results showed that the IC denitrification reactor exhibited a rapid start and stable running. The effluent TN ranged from 1.89 mg/L to 14.0 mg/L with an average of 4.65 mg/L when the influent TN were from 18.7 mg/L to 39.0 mg/L with an average of 26.6 mg/L,which was lower than the required 15 mg/L by Standard B of Table1 in Comprehensive Wastewater Discharge Standards for the Yellow River Basin in Shanxi Province (DB 61/224-2018). The effluent suspended solids mass concentration was lower with an average mass concentration of 18.6 mg/L,and the effluent COD of the subsequent recirculation biofilter was 35.4 mg/L,stably controlled below 50 mg/L,which showed a perfect comprehensive treatment effect. Moreover,the IC denitrification reactor had a small footprint,simple process,and a direct water treatment cost of 1.36 yuan/m3,which had significant economic benefits.

Objective To observe the effect of thunder-fire moxibustion on joint function recovery after surgery for fractures around the knee. Methods Totally 50 patients were divided into 2 groups (control group and observation group) by random number method. The control group was treated with routine treatment and nursing interventions after surgery, and the observation group was treated with thunder-fire moxibustion on the basis of routine treatment. The clinical effect of the two groups was compared. Results A total of 48 patients were followed up with complete results, including 24 cases in the control group and 24 cases in the observation group. The upper and lower diameters of the affected limb in the observation group were smaller than those in the control group after 3 and 7 days of treatment, with a significant difference(P<0. 05). The Visual Analogue Scale (VAS) score of the observation group was significantly lower than that of the control group after 3, 5 and 7 days of treatment, with a significant difference(P<0. 05). After 1, 3 and 5 weeks of treatment, the Hospital for Special Surgery (HSS) knee scores in two groups were higher than those before treatment (P<0. 05), and the HSS knee score in the observation group was higher than that in the control group after 3 and 5 weeks of treatment (P<0. 05). The range motion of the knee in the observation group was better than that of the control group after 7 days of treatment, with a significant difference(P<0. 05). Conclusion Postoperative nursing combined with thunder-fire moxibustion can effectively reduce the swelling and pain of the affected limb, improve the range of motion and function of the knee joint, and improve the clinical treatment effect. (目的 观察雷火灸联合护理干预促进膝关节周围骨折术后关节功能恢复的效果。方法 采用随机数字表法将50例患者分为对照组和观察组。对照组给予常规治疗和护理, 观察组在对照组基础上增加雷火灸治疗。比较两组干预效果。结果 随访共得到48例患者完整结果, 对照组及观察组各24例。治疗3 d、7 d后, 观察组患者患侧上、下周径均小于对照组, 差异有统计学意义(P<0. 05)。治疗3 d、7 d后, 观察组患者患侧VAS评分低于对照组, 差异有统计学意义(P<0. 05)。治疗1周、3周、5周后, 观察组患者患侧膝HSS评分均较治疗前提高(P<0. 05); 治疗3周、 5周后, 观察组患者患侧膝HSS评分高于对照组(P<0. 05)。治疗7 d后, 观察组患者患侧膝关节活动度大于对照组, 差异有统计学意义(P<0. 05)。结论 膝关节周围骨折术后护理配合实施雷火灸治疗, 能有效减轻患肢肿胀、疼痛, 促进康复, 改善膝关节活动度及功能, 有利于提高临床治疗效果。)

To improve the construction of a food safety standards system, the current status of analysis standards for radioactive substances in food in China was studied. Examining the content, application, and revision status of the current standards for the analysis of radioactive substances in food, shortcomings, and proposed suggestions were analyzed. Analysis standards are not suitable for practical needs in terms of timeliness and suitability, and it is necessary to improve the construction of relevant analysis standards to provide strong technical support for risk monitoring, assessment, and emergency disposal. It is recommended to establish a multiparty standard management mechanism, accelerate the preparation and revision of limit standards and response action level standards, improve testing methods and standards that are both urgent and routine, and continuously maintain the suitability and effectiveness of analytical standards.

Objective: To analyze and evaluate the uncertainty of the determination of folic acid content in milk powder by microplate method. Methods: The content of folic acid in infant formula milk powder was determined by microplate method according to GB 5009.211—2022 "National Standard for Food Safety-Determination of Folic Acid in Foods". Various experimental factors affecting the test results were analyzed and calculated according to the uncertainty evaluation methods and guidelines such as JJF 1059.1—2012, CNAS-CL01-G003:2011 and CNAS-GL05:2011, and the synthetic standard uncertainty and expanded uncertainty were obtained. Results: The expanded uncertainty U95=22.08 μg/100 g(k=2), and the folate content of the sample was (282.33±22.08) μg/100 g. The uncertainty mainly comes from the process of microplate inoculation, the fitting of standard curve and the repeatability of measurement. Conclusion: The accuracy of pipettes and microbial factors have a great impact on the experimental results, so it is necessary to ensure the quality of the instrument and strictly follow the standardized procedures to minimize the uncertainty.

The reconstruction and analysis of building models are crucial for the construction of smart cities. A refined building model can provide a reliable data support for data analysis and intelligent management of smart cities. The colors, textures, and geometric forms of building elements, such as building outlines, doors, windows, roof skylights, roof ridges, and advertisements, are diverse; therefore, it is challenging to accurately identify the various details of buildings. This article proposes the Multi-Task Learning AINet method that considers features such as color, texture, direction, and roll angle for building element recognition. The AINet is used as the basis function; the semantic projection map of color and texture, and direction and roll angle is used for multi-task learning, and the complex building facade is divided into similar semantic patches. Thereafter, the multi-semantic features are combined using hierarchical clustering with a region adjacency graph and the nearest neighbor graph to achieve an accurate recognition of building elements. The experimental results show that the proposed method has a higher accuracy for building detailed edges and can accurately extract detailed elements.

Many researchers and educational institutions are committed to exploring the modes and strategies of industry-education integration, which promotes the close connection between education and industrial needs by jointly carrying out teaching, research, and practice activities. This paper proposes a multi-objective optimization strategy based on genetic algorithms, which aims to enhance and optimize the talent cultivation model through adjustments to the resource matching scheme and teaching task allocation scheme for industry-education integration. The mechanical specialty of a higher vocational college puts forward 10 kinds of industry-teaching integration teaching resource allocation schemes based on teaching tasks, combined with enterprise demand and students’ ability, and substitutes them into the constructed multi-objective integration model, and solves them by genetic algorithm to arrive at the optimal resource allocation scheme G, which has an adaptability value of 0.571, and the matching degree of teaching task 6 under the industry-teaching integration teaching resource allocation scheme G is the highest, which means that the mechanical specialty needs to strengthen the professional knowledge teaching about task 6. Teaching of specialized knowledge about task 6. Additionally, the satisfaction distribution graph from the questionnaire data indicates that students feel more content with the construction and development of the mechanical specialty during the optimized talent cultivation mode of industry-teaching integration. To explore consumer feedback on route planning of urban tourist attractions in order to better propose optimization measures. This paper establishes the consumer satisfaction evaluation index system, determines the index weights based on the G1 method, CRITIC method and game theory method, and carries out evaluation calculation through the consumer satisfaction evaluation cloud model. This paper analyzes the satisfaction of consumers of different genders, age groups and incomes on each evaluation index and explores the credibility of this paper’s method by comparing and analyzing it with the satisfaction of online reviews. In terms of travel shopping satisfaction, the cloud droplets have the highest degree of aggregation, between 0.75-0.88 and a lower degree of aggregation, between 0.49-0.75, indicating that consumers are very satisfied. At the same time, the consumer satisfaction analyzed in this paper and the network review data satisfaction as a whole converge; the network review satisfaction and this paper’s analysis of the satisfaction in tourism services are around 84%-86%. The consumer satisfaction evaluation method constructed in this paper has a high degree of feasibility and accuracy.

This paper explores the interaction mechanism between urbanization and the ecological environment based on the unique correlation between urbanization and the ecological environment. BISE and Savizky-Golay algorithms are combined to establish the time series data, and for the characteristic curves of the EVI time response of the land cover, the spectral angle mapping method is combined with the minimum distance method to form the spectral angle-minimum distance classification method. The application of time series data involves calculating the time and spatial change of urban land cover and analyzing the evolution of urban ecological environment elements. Based on the temporal and spatial consistency test and land cover analysis of urban ecological environment construction, as well as the development of urbanization construction, relevant development paths are proposed. The comprehensive score of new urbanization in the four districts and one city of Hengyang City is concentrated towards 1.6, indicating that the level of economic development and the construction of new urbanization have reached coupling at different levels, respectively.

CHIPIC has been developed for more than one decade, starting from simulating vacuum devices. During the last decade of development, the functionality of CHIPIC is extended from 2-dimension to 3-dimension, from text interface to graphical interface, from serial computation to parallel computation, from simple boundary condition to multiple boundary conditions, and from electron-only PIC simulations of vacuum devices to complex plasma particle-in-cell/Monte Carlo collision (PIC/MCC) simulations of various devices.

ObjectiveTo establish an High Performance Liquid Chromatography-Inductively Coupled Plasma-Mass Spectrometry (HPLC-ICP-MS) method for determination of six arsenic species in human urine,including arseniccholine (AsC), arsenobetaine (AsB), arsenite (As3+), dimethylarsinic acid (DMA5+), monomethylarsonic acid (MMA5+), and arsenate (As5+).MethodsThe pH value of mobile phase and the content of anhydrous ethanol were optimized. Ammonium carbonate (50 mmol·L-1, containing 2% anhydrous ethanol, pH-8.5) mobile phase was selected. Cl- interference was eliminated by He mode. The arsenic species in 10-fold diluted human urine samples were separated by an Hamilton PRP X-100 anionic column. A method for the determination of six arsenic species was established.ResultsSix arsenic species could be separated in 13 minutes. The linear correlation coefficients were above 0.999. The limits of detection were 0.10‒0.20 μg·L-1, and the limits of quantification were 0.30‒0.50 μg·L-1. Precision experiments showed that RSD ranged from 5.96% to 9.07% when adding concentration 0.20 μg·L-1; from 2.48% to 6.38% when adding concentration 2.00 μg·L-1; and from 1.41% to 2.57% when adding concentration 5.00 μg·L-1. Accuracy test showed that the recoveries were 80%‒125%.ConclusionThe established HPLC-ICP-MS method for determination of six arsenic species in human urine is rapid, accurate and sensitive. It can be applied to the determination of arsenic species in human urine.

To this end, the noise of this frequency is reproduced on the bench, the laser Doppler vibration measurement technology is used to obtain the operational deflection shape(ODS) at this frequency. Finite element analysis(FEA) model is established and modified according to the experimental modal. The complex eigenvalue analysis is adopted to retrieve the complex eigenvalues and mode shapes.The correlation analysis is used to confirm that the complex eigenvalue results are valid for the actual noise ODS results.Afterwards, analysis of the components contribution and the components modal contribution of this unstable mode are used to provide corresponding structural optimization plan, and a bench test has been carried out to verify the effectiveness of this method.

The vibration acceleration monitoring of the cart with the frame damping system found that due to the frame damping system, nylon components are used for isolation between the frame connection parts and between the wheel and the seat pocket so that the nylon can effectively absorb the impact energy.

Under the “dual carbon” goal, China’s energy structure is accelerating transformation and upgrading, and new energy represented by solar energy, wind energy, geothermal energy is gradually replacing traditional coal, oil, gas and other fossil energy. Promoting the optimal combination of coal and new energy is a new requirement for achieving the “double carbon” goal, and also an important path for the green and low-carbon transformation of coal mines in the future. Based on the analysis of the energy distribution characteristics of coal-endowed areas in China, it is found that the fossil energy distribution areas dominated by coal are often also areas rich in solar energy, wind energy, thermal energy and other new energy, which are suitable for large-scale development of new energy in in coal-endowed areas. The research results are as follows: ① Put forward the concept of full life cycle energy development in coal-endowed areas, divide the energy development process in coal-endowed areas into three stages: coal, coal and new energy optimization combination, and new energy, and build a general framework for new energy development in coal-endowed areas; ② In terms of solar energy development, the theoretical reserves and development potential of solar energy in the mining area are evaluated based on the overlapping relationship between the coal occurrence and the solar energy resource rich area, and a hierarchical comprehensive utilization scheme of solar energy coupled with photochemical utilization, such as distributed/centralized photovoltaic power generation, solar thermal utilization and solar hydrogen production, is proposed to achieve diversified and efficient conversion and utilization of solar energy and collaborative mining with coal resources; ③ In terms of geothermal energy development, the potential of geothermal energy development in coal-endowed areas is evaluated based on the overlapping relationship between the coal bearing area and the geothermal energy resource rich area. In combination with the formation mechanism, distribution law and mining situation of the mine geothermal, a collaborative development system of deep mineral resources and geothermal resources is established to form a comprehensive application scheme for the graded mining of mine geothermal energy and the cascade utilization of thermal users, including the hot water type, rock temperature type and mixed type of the mine, realizing the strategic transfer of “harm” to “benefit” of mine geothermal; ④ In terms of wind energy development, the potential of wind energy development in the mining area is evaluated based on the overlapping relationship between the coal occurrence and the wind energy resource rich area, and the application approaches of wind power generation, wind water lifting, and wind heating technologies in the coal bearing area are proposed, and the wind energy utilization framework for power supply, water cycle power, heating self-sufficiency, and grid connected power generation of underground and underground facilities is constructed; ⑤ Considering that the roof is generally managed by the caving method in the current underground coal mining, and the caving space cannot be effectively used for underground energy storage, a technical path of energy storage in the goaf of the mine is proposed, which is “advanced planning of energy storage reservoir → construction of energy storage reservoir using functional backfilling→ Energy storage and release operation control”; ⑥ In view of the complex and changeable characteristics of multi energy complementary integrated energy system (MCIES) in coal-endowed areas, a heterogeneous energy flow coupling model of energy production and supply units in coal-endowed areas is established, the mathematical expressions of different energy hubs in MCIES are unified, and the energy management and optimization logic method and operation mode of MCIES in coal-endowed areas are proposed. This paper aims to explore the combination path of coal and new energy, demonstrate the feasibility of large-scale development of new energy in coal-endowed areas, endow coal bearing areas with the function of energy supply in the whole life cycle, and form a new mode of energy industry development of coal enterprises integrating “mine-wind-light-heat-storage”.

In this regard, this paper introduces the Game Idea and unfolds the analysis from the perspective of security resource point site selection and security resource path planning, hoping to improve the emergency efficiency. From scratch, the event diffusion influence factor is introduced to construct the security resource site selection strategy model to realise the site selection decision on regional road network security resource points. Further on, the security resource scheduling path planning platform with u-Dijkstra algorithm as the core under the site selection strategy is designed to realise the path planning for security resource scheduling, providing more efficient decision suggestions that are more applicable to colleges and universities. Finally, combing the traffic network of a campus, we conduct a simulation analysis while designing a small-range security resource path planning. Subsequently, the optimal decision and the optimal route of the safety resource planning are worked out, which demonstrates the effectiveness of the mathematical model in a simulative manner.

The existing research results showed that circular RNA zinc finger RNA binding protein (circRNA-ZFR) was abnormally expressed in many malignant tumors, and its expression level was closely related to tumor clinical stage, cell metastasis and patient prognosis. circRNA-ZFR could participate in the regulation of tumor occurrence and development through endogenous competitive adsorption of miRNA, regulation of cell cycle and activation of multiple signal pathways. Further study on the expression and mechanisms of circRNA-ZFR in malignant tumors may support early diagnosis, targeted therapy of related tumors and patients' prognosis.

40. METTL3/LINC00662/miR-186-5p feedback loop regulates docetaxel resistance in triple negative breast cancer

Abstract Insight into the mechanism of docetaxel resistance in breast cancer may help to improve prognosis. We aimed to investigate the role of N6-methyladenosine (m6A) and the METTL3/LINC00662/miR-186-5p pathway in regulating docetaxel resistance in triple negative breast cancer (TNBC). We have recruited 193 pathologically diagnosed TNBC patients from 2016 to 2017 in our hospital. Quantitative real-time PCR was used to evaluate the expression of LINC00662 and miR-186-5p both in vivo and in vitro. CCK8 tests were used to assess cell viability. ELISA was used for protein expression evaluation. Dual luciferase reporter gene assay and RNA pull-down were used to evaluate the interaction between LINC00662 and miR-186-5p. m6A levels were enhanced in breast cancer tissues and cells. LINC00662, miR-186-5p and METTL3 were differentially expressed in vivo, and METTL3 expression was associated with LINC00662 and miR-186-5p expression. LINC00662 and miR-186-5p were differentially expressed in vitro; LINC00662 promoted cell viability and decreased the apoptosis rate, whereas miR-186-5p inhibited cell viability and increased the apoptosis rate. Furthermore, we found that METTL3 regulated m6A levels in docetaxel-resistant breast cancer cells by regulating the expression of LINC00662. Moreover, LINC00662 and miR-186-5p regulated the cell viability rate of docetaxel-resistant breast cancer cells. Further experiments showed that LINC00662 directly interacted with miR-186-5p to exert biological functions; besides miR-186-5p could regulate the expression of METTL3. METTL3 promotes m6A levels and docetaxel resistance in breast cancer by regulating the expression of LINC00662 and miR-186-5p; more experiments are needed to clarify the role of m6A regulation in drug resistance.

Aiming at the problems of low segmentation accuracy and high loss of deep learning image semantic segmentation methods, image semantic segmentation method with fusion of transposed convolution and deep residual is proposed. Firstly, in order to solve the problems of decreasing segmentation accuracy and slow convergence speed caused by increasing of the depth of neural network, a deep residual learning module is designed to improve the training efficiency and convergence speed of the network. After that, in order to make the feature map fusion more accurate in upsampling and feature extraction process, two upsampling methods of UpSampling2D and transposed convolution in the deep residual U-net model are merged to form a new upsampling module. Finally, to solve the over-fitting of the weights between training set and validation set in the process of network training, Dropout is introduced in the skip connection layer of the improved network, which enhances learning ability of the model. The performance of algorithm is proven on the CamVid datasets. The semantic segmentation accuracy of the algorithm reaches 89.93% and the loss is reduced to 0.23. Compared with U-net model, the verification set accuracy is improved by 13.13 percentage points, and the loss is reduced by 1.20, which is better than the current image semantic segmentation methods. The proposed model of image semantic segmentation combines the advantages of U-net, which makes the image semantic segmentation more accurate, with better effect, and effectively improves the robustness of algorithm.

Objective: To improve the accuracy of the results of cadmium (Cd) content in kelp (Laminaria japonica) by graphite furnace atomic absorption spectrometry (GF-AAS). Methods: The concentration of Cd element in kelp sample was measured by using GF-AAS after microwave digestion according to the standard of GB 5009.15—2014. Combining with the experimental process and mathematical model, the possible sources of measurement uncertainty were firstly identified, then the components of each standard uncertainties were calculated, and finally each parameter of uncertainties was synthesized, and the whole uncertainty was evaluated. Results: when the value of Cd concentration in kelp was 0.202 mg/kg, the expanded uncertainty was 0.008 mg/kg (k=2). Conclusion: The uncertainty of the method is mainly derived from the determination of Cd concentration in the sample solution, which is basically introduced by standard working solution preparation. Moreover, the uncertainty caused by the sample recovery rate is also large. So the quality control of the parameter of uncertainties should be strengthened in the future.

To solve engineering geological problems, including water conservancy, transportation, and mining, it is necessary to obtain information on rock mass structures, such as slopes, foundation pits, and tunnels, in time. The traditional method for obtaining structural information requires manual measurement, which is time consuming and labor intensive. Because geological information is complicated and diverse, it is not practical for general deep learning methods to obtain full-scale structural surface trace images to prepare training samples. Transfer learning can abstract high-level features from low-level features with a small number of training samples, which can automatically express the inherent characteristics of objects. This article proposed a rock mass structural surface trace extraction method based on the transfer learning technique that considers the attention mechanism and shape constraints. For the general test set, the accuracy of rock mass structural surface trace recognition with the proposed method can reach 87.2%. Experimental results showed that the proposed method has advantages in extracting complicated geological structure information and is valuable for providing technical support for the extraction of geological information in the construction of water conservancy, transportation, mining, and related projects.

NKILA is a kind of newly-discovered lncRNA whose expression is aberrant in diverse malignant tumors. The existing researches have confirmed that NKILA participates in the occurrence and development of tumors mainly by regulating the NF-κB signaling pathway, and has significance to the cancer diagnosis, treatment and prognostic evaluation of patients. This article reviews the abnormal expressions and biological effects of NKILA, and the up- and down-stream mechanisms of NKILA regulating malignant biological behavior in different cancers.

Existing video inpainting methods usually produce blurred texture,distorted structure and artifacts,while applying image-based inpainting model directly to the video inpainting will lead to inconsistent time.From the perspective of time,a novel coherent semantic spatial-temporal attention(CSSTA) for video inpainting is proposed,through the attention layer,the model focuses on the information that the target frame is partially blocked and the adjacent frames are visible,so as to obtain the visible content to fill the hole region of the target frame.The CSSTA layer can not only model the semantic correlation between hole features but also remotely correlate the long-range information with the hole regions.In order to complete semantically coherent hole regions,a novel loss function Feature Loss is proposed to replace VGG Loss.The model is built on a two-stage coarse-to-fine encoder-decoder model for collecting and refining information from adjacent frames.Experimental results on the YouTube-VOS and DAVIS datasets show that the method in this paper runs almost in real-time and outperforms the three typical video inpainting methods in terms of inpainting results,peak signal-to-noise ratio (PSNR) and structural similarity (SSIM).

48. Effects of necrotizing enterocolitis on brain injury and its severity in neonatal mice

Objective To investigate the effect of necrotizing enterocolitis (NEC) at different severities on brain injury in neonatal mice. Methods Sixty 5-day old C57BL/6 mice were randomly divided into control group (CON group, n=20) and NEC group (n=40). The CON group was breeding with female mice and breast-fed. The NEC group was fed in an incubator, and induced by formula gavage feeding+hypoxia+oral lipopolysaccharide (LPS) to establish the model of NEC. In 4 d after NEC modeling, all pups were sacrificed by cervical decapitation. Their body weight was measured and general conditions were observed. HE staining was used to observe the pathological changes of intestinal tissue. NEC group was divided into mild NEC subgroup and severe NEC subgroup according to the intestinal injury score of ileocecus. Nissl staining was employed to further observe the pathological changes of the corresponding brain tissue. The correlation between the severity of intestinal injury and the number of nerve cells was analyzed with Pearson analysis. The mRNA levels of the inflammatory factors interleukin-1β (IL-1β), IL-6 and tumor necrosis factor-α (TNF-α) in brain tissues were measured with quantitative real-time PCR (RT-qPCR). Enzyme-linked immunosorbent assay (ELISA) was adopted to detect the protein contents of IL-1β, IL-6 and TNF-α in brain tissues. Immunofluorescence assay was conducted to detect the number of activated microglia and astrocytes in the hippocampus. Results Compared with CON group, the NEC group had decreased activities and lower body weight (3.84±1.25 vs 1.76±0.26 g, P < 0.05), and serious intestinal damage. The number of nerve cells in the hippocampus were significantly reduced in the NEC group than the CON group (P < 0.05), so was in the severe NEC subgroup than that the mild NEC subgroup (P < 0.05). Correlation analysis showed that the pathological changes of brain were negatively correlated with the severity of intestinal injury (r=- 0.943, P < 0.001). When compared with the CON group, the mRNA and protein levels of IL-1β, IL-6 and TNF-α in brain tissues were significantly elevated (P < 0.05), and the numbers of activated microglia and astrocytes were also significantly increased in the NEC group (P < 0.05), especially in the severe NEC subgroup (P < 0.05). Conclusion NEC can lead to acute brain injury in neonatal mice, and the severity of damage is related to the severity of intestinal injury, which may be related to the increase of neuroinflammation.

49. Effects of direct current electric field on proliferation and biological characteristics of human adipose derived mesenchymal stem cells

Objective To investigate the effect of direct current (DC) electric field on the proliferation of human adipose-derived stem cells (hASCs) in vitro. Methods The fat tissue obtained in operation was digested with zymogen to prepare hASCs. After the cell culture was passaged to the third generation, the hASCs were inoculated in a 6-well plate. After 24 h, the cells were cultured under a stimulation of 0, 50, 100, 150 and 200 mV/mm DC electric field, at a frequency of 1 h/d for 3 continuous days. Cell morphology and growth were observed under an inverted microscope. Cell counting and CCK8 assay were carried out to measure cell proliferation. Flow cytometry was performed to detect the expression of stem cell phenotype markers, CD105, CD73, CD90, CD14, CD34, CD31 and CD45. The effect of DC electric field stimulation was observed after differentiation induction. Western blotting was used to detect the expression of proteins related to cell proliferation in stimulated and unstimulated cells. Results The proliferation rate and absorbance value were significantly higher in the cells after 50, 100 and 150 mV/mm stimulation than those without (all P < 0.01). When the stimulation intensity of the DC electric field was increased to 200 mV/mm, the proliferation rate was significantly lower (P < 0.001), but the absorbance value was statistically higher than those of the 0 mV/mm stimulation (P < 0.05). The stimulation also resulted in remarkably higher expression of CD105, CD73 and CD90, and lower expression of CD14, CD31, CD34 and CD45. Moreover, the hASCs could differentiate into bone cells and adipocytes with and without DC electric field stimulation, but the stimulated cells had faster differentiation than those without. Compared with the unstimulated cells, the expression of cell proliferation related protein p-ERK1/2 was significantly increased in the stimulated cells (P < 0.01). Conclusion The DC electric field promotes the proliferation of hASCs probably by regulating the phosphorylation of ERK, but has no effect on the biological characteristics of the stem cells.

50. Preparation of gel containing human adipose derived stem cells and its clinical efficacy for adherent scar

Objective To investigate the clinical effectiveness of stromal vascular fraction gel (SVF-gel) containing human adipose derived stem cells (ADSCs) in the treatment of adherent scar. Methods A retrospective analysis was carried out on the patients with adherent scars meeting our inclusion criteria and undergoing the treatment of gel injection during June 2018 and June 2020. The SVF-gel was prepared from the lipoaspirates sucked from the abdomen or the thigh followed by filtration, centrifugation and emulsion, and then injected into the adherent scars (at an interval of 2 mm for every needle puncture, SVF-gel 0.02 mL for every dose). Small amount of granular adipose tissues and SVF-gel were digested, cultured and identified. The scar hardness and thickness were measured for every patient preoperatively and 6 months postoperatively. Patient and observer scar assessment scale (POSAS) was used to evaluate scar condition, and patient satisfaction was investigated. Results There were 8 patients enrolled in this study, with a total of 10 adherent scars and the area of depression of 5.56±4.50 cm2. The injection volume of SVF-gel under the adherent scar was 3.90±3.00 mL. The results of morphological observation showed that short spindle cells grew in both SVF-gel and granular fat after digestion, and the over 95% obtained cells expressed the specific markers of mesenchymal stem cells, CD73, CD90 and CD105. At the same time, the number of primary cells cultured in same volume of SVF-gel was larger than the number from the granular fat tissue. Differentiation study found the growth of adipose-like cells in vitro, which were positive to oil red O staining. The scar hardness was decreased from 48.13±20.59 shore units preoperatively to 30.96±15.66 shore units in 6 months postoperatively(P < 0.05), and the scar skin thickness was increased from 2.00±0.60 mm to 2.72±0.63 mm (P < 0.05). In 6 months after operation, the appearance and activity of scar skin were significantly improved, and no complications were observed. The scores of patient scar assessment scale (PSAS) and observer scar assessment scale (OSAS) were 12.0±2.8 and 13.7±3.9 respectively, and the patient satisfaction score was 4 to 5, with an average of 4.6. Conclusion Our prepared SVF-gel contains more active ADSCs. Its injection can improve the local depression and scar texture in the adherent scar and achieve good satisfaction in the patients.
