Abstract

Cyclophosphamide (CTX) is an antitumor drug commonly used to treat various cancer types. Unfortunately, its toxic side effects, including gastrointestinal (GI) toxicity, affect treatment compliance and patients' prognosis. Thus, there is a critical need of evaluating strategies that may improve the associated GI toxicity induced by CTX. In this work, we evaluated the capacity of epigallocatechin-3-gallate (EGCG), a major constituent of green tea, to improve the recovery of gut injury induced by CTX in mice. Treatment with CTX for 5 days severely damaged the intestinal structure, increased immune-related cytokines (TNFα, IL-10 and IL-21), reduced the expression levels of tight junction proteins (ZO-1, occludin, claudin-1), induced reactive oxygen species, altered the composition of gut microbiota, and reduced short chain fatty acid levels. EGCG treatment, starting one day after the last CTX dose, significantly improved the intestinal structure, ameliorated gut permeability, and restored ZO-1, occludin and claudin-1 levels. Moreover, EGCG reduced TNFα, IL-10 and IL-21 levels and decreased oxidative stress by regulating the activities of the antioxidant enzymes catalase, superoxide dismutase and glutathione peroxidase. Finally, EGCG treatment restored the composition of gut microbiota and the levels of the short chain fatty acids. In conclusion, these findings indicate that EGCG may function as an effective bioactive compound to minimize CTX-induced GI tract toxicity.

19. Machine learning classifies predictive kinematic features in a mouse model of neurodegeneration.

Abstract

Motor deficits are observed in Alzheimer's disease (AD) prior to the appearance of cognitive symptoms. To investigate the role of amyloid proteins in gait disturbances, we characterized locomotion in APP-overexpressing transgenic J20 mice. We used three-dimensional motion capture to characterize quadrupedal locomotion on a treadmill in J20 and wild-type mice. Sixteen J20 mice and fifteen wild-type mice were studied at two ages (4- and 13-month). A random forest (RF) classification algorithm discriminated between the genotypes within each age group using a leave-one-out cross-validation. The balanced accuracy of the RF classification was 92.3 ± 5.2% and 93.3 ± 4.5% as well as False Negative Rate (FNR) of 0.0 ± 0.0% and 0.0 ± 0.0% for the 4-month and 13-month groups, respectively. Feature ranking algorithms identified kinematic features that when considered simultaneously, achieved high genotype classification accuracy. The identified features demonstrated an age-specific kinematic profile of the impact of APP-overexpression. Trunk tilt and unstable hip movement patterns were important in classifying the 4-month J20 mice, whereas patterns of shoulder and iliac crest movement were critical for classifying 13-month J20 mice. Examining multiple kinematic features of gait simultaneously could also be developed to classify motor disorders in humans.

27. Measuring Road Network Topology Vulnerability by Ricci Curvature

Abstract

Describing the basic properties of road network systems, such as their robustness, vulnerability, and reliability, has been a very important research topic in the field of urban transportation. Current research mainly uses several statistical indicators of complex networks to analyze the road network systems. However, these methods are essentially node-based. These node-based methods are more concerned with the number of connections between nodes, and lack of consideration for interactions. So, this leads to the well-known node paradox problem, and their ability of characterizing the local and intrinsic properties of a network is weak. From the perspective of network intrinsic geometry, this paper proposes a method for measuring road network vulnerability using a discrete Ricci curvature, which can identify the key sections of a road network and indicate its fragile elements. The results show that our method performs better than complex network statistics on measuring the vulnerability of a road network. Additionally, it can characterize the evolution of the road network vulnerability among different periods of time in the same city through our method. Finally, we compare our method with the previous method of centrality and show the different between them. This article provides a new perspective on a geometry to analyze the vulnerability of a road network and describes the inherent nature of the vulnerability of a road system from a new perspective. Abstract

In this study, the effects of adding glutamate (Glu), glutamine (Gln), aspartate (Asp), and asparagine (Asn) on the flavor formation of Huangjiu were investigated, and the effect of Gln concentration on the quality, microbial community structure, and flavor development of Huangjiu was further explored. Varied Gln concentrations influenced yeast growth, sugar utilization, microbial communities, and quality attributes. Additional Gln promoted yeast cell counts and sugar depletion. It increased the complexity of bacterial co-occurrence networks and reduced the impact of stochastic processes on assembly. Correlation analysis linked microorganisms to flavor compounds. Isolation experiments verified the role of Saccharomyces cerevisiae, Aspergillus chevalieri, Bacillus altitudinis, and Lactobacillus coryniformis in flavor production under Gln conditions. This research elucidated the microbiological mechanisms by which amino acid supplementation, especially Gln, enhances Huangjiu quality by modulating microbial metabolic functions and community dynamics during fermentation. Abstract

A Neutron Time-Of-Flight (NTOF) spectrometer based on Heavy Ion Research Facility in Lanzhou (HIRFL) is developed for studies of neutron production of proton induced spallation reactions related to the ADS project. After the presentation of comparisons between calculated spallation neutron production double-differential cross sections and the available experimental one, a detailed description of NTOF spectrometer is given. Test beam results show that the spectrometer works well and data analysis procedures are established. The comparisons of the test beam neutron spectra with those of GEANT4 simulations are presented., Comment: 9 pages, 7 figures
