18. TSPY potentiates cell proliferation and tumorigenesis by promoting cell cycle progression in HeLa and NIH3T3 cells

Abstract

Background: TSPY is a repeated gene mapped to the critical region harboring the gonadoblastoma locus on the Y chromosome (GBY), the only oncogenic locus on this male-specific chromosome. Elevated levels of TSPY have been observed in gonadoblastoma specimens and a variety of other tumor tissues, including testicular germ cell tumors, prostate cancer, melanoma, and liver cancer. TSPY contains a SET/NAP domain that is present in a family of cyclin B and/or histone binding proteins represented by the oncoprotein SET and the nucleosome assembly protein 1 (NAP1), involved in cell cycle regulation and replication. Methods: To determine a possible cellular function for TSPY, we manipulated the TSPY expression in HeLa and NIH3T3 cells using the Tet-off system. Cell proliferation, colony formation assays and tumor growth in nude mice were utilized to determine the TSPY effects on cell growth and tumorigenesis. Cell cycle analysis and cell synchronization techniques were used to determine cell cycle profiles. Microarray and RT-PCR were used to investigate gene expression in TSPY expressing cells. Results: Our findings suggest that TSPY expression increases cell proliferation in vitro and tumorigenesis in vivo. Ectopic expression of TSPY results in a smaller population of the host cells in the G(2)/M phase of the cell cycle. Using cell synchronization techniques, we show that TSPY is capable of mediating a rapid transition of the cells through the G(2)/M phase. Microarray analysis demonstrates that numerous genes involved in the cell cycle and apoptosis are affected by TSPY expression in the HeLa cells. Conclusion: These data, taken together, have provided important insights on the probable functions of TSPY in cell cycle progression, cell proliferation, and tumorigenesis.

34. Inhibition of shoot‐expressed NRT1.1 improves reutilization of apoplastic iron under iron‐deficient conditions.

Abstract

SUMMARY: Iron deficiency is a major constraint for plant growth in calcareous soils. The interplay between NO3− and Fe nutrition affects plant performance under Fe‐deficient conditions. However, how NO3− negatively regulates Fe nutrition at the molecular level in plants remains elusive. Here, we showed that the key nitrate transporter NRT1.1 in Arabidopsis plants, especially in the shoots, was markedly downregulated at post‐translational levels by Fe deficiency. However, loss of NRT1.1 function alleviated Fe deficiency chlorosis, suggesting that downregulation of NRT1.1 by Fe deficiency favors plant tolerance to Fe deficiency. Further analysis showed that although disruption of NRT1.1 did not alter Fe levels in both the shoots and roots, it improved the reutilization of apoplastic Fe in shoots but not in roots. In addition, disruption of NRT1.1 prevented Fe deficiency‐induced apoplastic alkalization in shoots by inhibiting apoplastic H+ depletion via NO3− uptake. In vitro analysis showed that reduced pH facilitates release of cell wall‐bound Fe. Thus, foliar spray with an acidic buffer promoted the reutilization of Fe in the leaf apoplast to enhance plant tolerance to Fe deficiency, while the opposite was true for the foliar spray with a neutral buffer. Thus, downregulation of the shoot‐part function of NRT1.1 prevents apoplastic alkalization to ensure the reutilization of apoplastic Fe under Fe‐deficient conditions. Our findings may provide a basis for elucidating the link between N and Fe nutrition in plants and insight to scrutinize the relevance of shoot‐expressed NRT1.1 to the plant response to stress. 39. Application of modified sepiolite as reusable adsorbent for Pd(II) sorption from acidic solutions

Abstract

The adsorption characteristics and mechanisms of modified sepiolite as an adsorbent to recover Pd(II) from acidic solutions were studied. The Pd(II) adsorption properties were analysed through isotherm, kinetic and thermodynamic models. In addition, SEM−EDS, TEM and XPS were applied to investigating the Pd(II) adsorption mechanisms onto modified sepiolite. The equilibrium data were well fitted to Langmuir isotherm model with maximum Pd(II) adsorption capacity of 322.58 mg/g at 30 degrees C. The kinetic data could be satisfactorily simulated by the pseudo-second order model, indicating that the rate-controlling step was chemical adsorption. 99% of Pd(II) could be recovered using 1 g/l modified sepiolite when initial concentration of Pd(II) was 100 mg/l. The results of reusability studies indicated the modified sepiolite had an acceptable stability and reusability. This study indicated that the modified sepiolite might be an efficient and cost-effective material for Pd(II) recovery. Abstract

The prevalence of fatty liver disease (FLD), a chronic disorder that includes alcoholic liver disease (ALD) and non-alcoholic fatty liver disease (NAFLD) ranges from 1% to 50% and varies with age, gender and occupation in different districts of China. Some studies have shown that FLD especially NAFLD, has a relationship with cardiac disease, metabolic syndrome, dyslipidemia and obesity. To determine the prevalence of FLD and related diseases in the Dehui City of Jilin Province in the Northeastern region of China, we surveyed the population of Dehui City of Jilin Province in China. The objective of this study was to evaluate the prevalence of FLD in relation to different factors, such as age, location, occupations and educational levels, and analyze the risk factors of FLD. The study sample was separated into three groups according to alcohol consumption: non-alcoholic fatty liver disease (NAFLD), alcoholic liver disease (ALD) and suspected alcoholic liver disease (SALD) groups. All subjects (18- 78 years) completed a standard questionnaire, physical examination and hepatic ultrasound examination. Multivariate logistic regression was used to screen risk factors of FLD. The study indicated that the prevalence of FLD was 23.1%, strati ed by NAFLD 14.5%, SALD 5.5% and ALD 3.0%. 50. Lactational Breast Abscesses Caused by Methicillin-Resistant or Methicillin-Sensitive Staphylococcus aureusInfection and Therapeutic Effect of Ultrasound-Guided Aspiration

Abstract

Background and Objective:We investigated whether (1) there are any differences in lactational breast abscesses between patients from whom methicillin-resistant Staphylococcus aureus(MRSA) and those from whom methicillin-sensitive S. aureus(MSSA) were isolated from pus samples and (2) there are differences in the effects of ultrasound-guided aspiration.Materials and Methods:The clinical data of 171 patients with lactational breast abscesses treated by ultrasound-guided aspiration in Beijing from January to July 2018 were retrospectively analyzed. The patients were divided into MSSA infection (N= 132) and MRSA infection (N= 39) groups according to their bacterial culture results. Abscess cavity location, abscess cavity number, maximum abscess cavity size, antibiotic utilization rate, and cure rate were compared between the groups. Cure rate refers to the proportion of the total number of cases remaining after the elimination of failed cases. The number of ultrasound-guided aspiration procedures performed for healing between the two groups was also compared.Results:There were no significant differences in abscess cavity location, abscess cavity amount, and abscess cavity size between both groups. The antibiotic utilization rate of the two groups were 58.3% (MSSA, 77/132) and 69.2% (MRSA, 27/39), respectively, and there were no significant differences between both groups. The cure rates of ultrasound-guided aspiration of the two groups were 97.7% (MSSA, 129/132) and 92.3% (MRSA, 36/39), and there were no significant differences between both groups. There were also no significant differences in the median number of aspiration performed for cure between the MRSA infection group (median = 3, range = 1–10) and the MSSA infection group (median = 3, range = 1–14).Conclusion:Lactational breast abscesses are the same irrespective of the type of S. aureusinfection. Treatment by ultrasound-guided aspiration for patients with MRSA infection can achieve the same effect as that for those with MSSA infection.
