In response to an investigation on the paths of changes in the crystallization and radial differences during the forming process of nascent fibers, in this study, we conducted numerical simulation and analyzed the changes in crystallization mechanical parameters and tensile properties through a fluid dynamics two-phase model. The model was based on the melt-spinning method focusing on melt spinning, the environment of POLYFLOW, and the method of joint simulation, coupled with Nakamura crystallization kinetics, including the development of process collaborative parameters, stretch-induced crystallization, viscoelasticity, filament cooling, gravity term, inertia, and air resistance. Finally, for nylon 6 BHS and CN9987 resin spinning, the model successfully predicted the distribution changes in temperature, velocity, strain rate tensor, birefringence, and stress tensor along the axial and radial fibers and obtained the variation pattern of fibers' crystallinity along the entire spinning process under different stretching rates. Furthermore, we also explored the effects of spinning conditions, including inlet flow rate, winding speeds, and the extrusion temperature, on the fibers' crystallization process and obtained the influence rules of different spinning conditions on fiber crystallization. Knowing the paths of changes in mechanical performance can provide important guidance and optimization strategies for the future industrial preparation of high-performance fibers. Background: Organ weight change is widely accepted as a measure of toxicologic pathology. We and other groups have shown that excessive alcohol exposure leads to decreased spleen weight in rodents. This study explores the mechanisms underlying alcohol‐induced splenic injury through a network meta‐analysis. Methods: QIAGEN Ingenuity Pathway Analysis (IPA) and Mammalian Phenotype (MP) Ontology were used to identify alcohol‐related molecules associated with the small spleen phenotype. Search Tool for the Retrieval of Interacting Genes/Proteins (STRING) and IPA bioinformatics tools were then used to analyze the biologic processes and enriched signaling pathways engaging these molecules. In addition, the "downstream effects analysis" algorithm was used to quantify alcohol's effects. Results: IPA identified 623 molecules affected by alcohol and a Venn diagram revealed that 26 of these molecules overlapped with those associated with the MP Ontology of small spleen. The 26 molecules are TGFB1, CASP8, MTOR, ESR1, CXCR4, CAMK4, NFKBIA, DRD2, BCL2, FAS, PEBP1, TRAF2, ATM, IGHM, EDNRB, MDM2, GLRA1, PRF1, TLR7, IFNG, ALOX5, FOXO1, IL15, APOE, IKBKG, and RORA. Some of the 26 molecules were also associated with the MP Ontology of abnormal white pulp and red pulp morphology of the spleen, abnormal splenic cell ratio, decreased splenocyte number, abnormal spleen physiology, increased splenocyte apoptosis, and reduced splenocyte proliferation. STRING and IPA "Core Analysis" showed that these molecules were mainly involved in pathways related to cell apoptosis, proliferation, migration, and immune responses. IPA's "Molecular Activity Predictor" tool showed that concurrent effects of activation and inhibition of these molecules led to decreased spleen size by modulating apoptosis, proliferation, and migration of splenocytes. Conclusions: Our network meta‐analysis revealed that excessive alcohol exposure can damage the spleen through a variety of molecular mechanisms, thereby affecting immune function and human health. We found that alcohol‐mediated splenic atrophy is largely mediated by increased apoptosis signaling, migration of cells, and inhibition of splenocyte proliferation. [ABSTRACT FROM AUTHOR]

Objective To investigate the effect of miR-145 on the migration and invasion of non-small cell lung cancer (NSCLC) cells and possible mechanism. Methods The expression of miR-145 and activating enhancer binding protein 4 (AP4) mRNA in NSCLC tissues were detected by qRT-PCR. After the transfection with miR-145 mimic or AP4 siRNA, the migration and invasion of A549 cells were examined by wound healing and Transwell assay, respectively; The mRNA and protein expressions of AP4 were detected by qRT-PCR and Western blot, respectively. The relationship between AP4 mRNA and miR-145 was detected by double luciferase reporter gene method. Results Compared with adjacent normal tissues, the level of miR-145 was significantly decreased in NSCLC tissues (P < 0.05), however, the mRNA and protein levels of AP4 were significantly increased (P < 0.05). Compared with control group, the level of miR-145 was significantly increased (P < 0.05), the mRNA and protein levels of AP4 were significantly decreased (P < 0.05), and cells migration and invasion were significantly decreased (P < 0.05) in A549 cells after the transfection with miR-145 mimic. Moreover, AP4 was a direct target of miR-145. After the transfection with AP4 siRNA, the migration and invasion of A549 cells were significantly decreased (P < 0.05). Conclusion miR-145 upregulation could efficiently attenuate the migration and invasion of A549 cells via inhibiting target gene AP4 expression.

With the upsurge of magnetic levitation construction and research in China, in order to improve the running speed of mid-low-speed maglev train, the 13th Five-Year Science and Technology Major Proposal put forward a medium speed maglev solution with long stator synchronous traction. According to this new plan, the magnetic levitation subsystem, including the control system, needs re-co-ordination design, and the car network system must be changed accordingly. In this paper, based on the original scheme, a new magnetic levitation vehicle network system is designed to meet the needs of localization. In this paper, colored Petri nets are used to model and analyze the original scheme and the improved scheme. It proves theoretically that the improvement scheme is superior to the original scheme in failure rate and can meet the actual project requirements.
