Abstract

Salt stress is one of the major impairments to agricultural soil that significantly reduces growth and productivity in rice (Oryza sativa L.) and other crop plants. The proteomic mechanisms underlying salt stress tolerance in rice have not been well established. Therefore, a comprehensive understanding of molecular mechanisms associated with salt signaling, salt-toxicity detoxification, and other metabolic mechanisms is essential for elucidating salt tolerance mechanisms in rice as well as ensuring global food security. Recent proteome studies have provided a global proteomic signature of rice cultivars. Integrative studies of proteomic, physiological, and molecular responses under salt stress have provided detailed mechanisms associated with salt stress tolerance in rice. This review explores the proteomic mechanisms with finely-tuned salt-responsive networks in this cereal. Several proteomic processes, including salt sensing and signaling, scavenging of reactive oxygen species (ROS) and stress defense, salt compartmentalization and homeostasis, alterations of cell wall components, modulation of the cytoskeleton, regulation of salt-responsive genes, transcription factors, and protein synthesis, protein folding and processing, protein degradation, and strategies of carbohydrate and energy metabolism for organ development, elucidate extensive molecular mechanisms linked to salt stress responses and tolerance in rice. Further, it is updated the prospects of salt stress tolerance in rice using multi-omics and CRISPR/Cas approaches. These finely-tuned molecular insights will be beneficial to rice breeders and farmers for developing high-yielding, salt-tolerant rice cultivars to achieve global food security.

43. Red blood cells increase secretion of matrix metalloproteinases from human lung fibroblasts in vitro

Abstract

Tissue remodeling is an important process in many inflammatory and fibrotic lung disorders. RBC may in these conditions interact with extracellular matrix (ECM). Fibroblasts can produce and secrete matrix components, matrix-degrading enzymes (MMPs) and tissue inhibitors of metalloproteinases (TIMPs). Imbalance in matrix synthesis/degradation may result in rearrangement of tissue architecture and lead to diseases such as emphysema or fibrosis. Neutrophil elastase (NE), a protease released by neutrophils, is known to activate MMP. We hypothesized that RBC can stimulate secretion of MMPs from human lung fibroblasts and that NE can augment this effect. Human fetal lung fibroblasts were cultured in floating collagen gels with or without RBC. After 4 days, the culture medium was analyzed with gelatin zymography, Western blot, and ELISA for MMP-1, -2, -3 and TIMP-1, -2. RBC augmented NE-induced fibroblast-mediated collagen gel contraction compared with NE alone (18.4 [+ or -] 1.6%, 23.7 [+ or -] 1.4% of initial gel area, respectively). A pan-MMP inhibitor (GM-6001) completely abolished the stimulating effect of NE. Gelatin zymography showed that RBC stimulated MMP-2 activity and that NE enhanced conversion to the active form. Addition of GM-6001 completely inhibited MMP-2 activity in controls, whereas it only partially altered RBC-induced MMP activity. Western blot confirmed the presence of MMP-1 and MMP-3 in fibroblasts stimulated with RBC, and ELISA confirmed increased concentrations of pro-MMP-1. We conclude that stimulation of MMP secretion by fibroblasts may explain the ability of RBC to augment fibroblast-mediated collagen gel contraction. Abstract

The effect of Co content on the phase formation, structure, and magnetocaloric effect of rapidly quenched La(Fe(sub 1-x)Co(sub x))(sub 11.4)Si(sub 1.6) were investigated by x-ray diffractometry and magnetic measurements. Increasing Co content drives the order of magnetic transition neat T(sub C) from first order to second order and eliminates the occurrence of the field-induced magnetic transition above T(sub C) which is responsible for the decrease in the magnetocaloric effects.
