Early diagnosis of gastrointestinal malignancies in patients with acute coronary syndrome (ACS) is often delayed. The present study aims to demonstrate the value of [68Ga]Ga‐FAPI‐04 PET imaging in ACS with suspected gastrointestinal malignancies. Twelve ACS patients with suspected gastrointestinal malignancies were enrolled, including ST‐elevation myocardial infarction (STEMI) (n = 5), non‐ST‐elevation myocardial infarction (NSTEMI) (n = 5), and unstable angina (UA) (n = 2). All patients underwent coronary angiography (CAG) and [68Ga]Ga‐FAPI‐04 PET/MR or PET/CT within 1 week. All five STEMI and five NSTEMI patients had high [68Ga]Ga‐FAPI‐04 uptake in the injured myocardium compared to remote area (TBR: 2.10 ± 0.72 vs. 0.62 ± 013; p <.001), correlated with peak cTnI level (R =.82, p =.004). No [68Ga]Ga‐FAPI‐04 in the myocardium was found in UA patients. NSTEMI displayed a similar myocardial [68Ga]Ga‐FAPI‐04 intensity as STEMI (p =.42). Compared with STEMI, NSTEMI patients had a significantly delayed door‐to‐balloon time for reperfusion treatment (p =.023). High uptake of [68Ga]Ga‐FAPI‐04 in the gastrointestinal tract was detected in three patients. Because of no myocardial [68Ga]Ga‐FAPI‐04 expression, they discontinued antiplatelet therapy and underwent endoscopy. The rectal, colon, and gastric cancer diagnoses were made by biopsy. The other nine patients showed no accumulation of [68Ga]Ga‐FAPI‐04 beyond the heart, and invasive tumor examinations were delayed. During a median 6‐month follow‐up, no tumor formation was observed. [68Ga]Ga‐FAPI‐04 PET imaging is valuable to assess injured myocardium, detect tumors, and guide invasive examinations in ACS patients with suspected gastrointestinal malignancies. Purpose: We aimed to evaluate whether [68 Ga]Ga-FAPI-04 PET/CT could characterize the early stages of cardiac fibrosis in pressure overload heart failure. Methods: Sprague–Dawley rats underwent abdominal aortic constriction (AAC) (n = 12) and sham surgery (n = 10). All rats were scanned with [68 Ga]Ga-FAPI-04 PET/CT at 2, 4, and 8 weeks after surgery. The expression of fibroblast activation protein (FAP) in the myocardium was detected by immunohistochemistry. [68 Ga]Ga-FAPI-04 PET signal and FAP expression were compared between two groups. Results: Compared with the sham group, the AAC group presented with decreased ejection fraction (EF) and fractional shortening (FS) and increased left ventricular internal dimensions in diastole (LVIDd) and systole (LVIDs) at 4 and 8 weeks (all p < 0.01). The AAC group showed higher [68 Ga]Ga-FAPI-04 accumulation in the heart than the sham group at 2, 4, and 8 weeks, and FAPI increased significantly from 2 to 8 weeks (all p < 0.001). Immunohistochemistry confirmed the higher density of the FAP+ area in the AAC group. The intensity of the [68 Ga]Ga-FAPI-04 correlated with the density of the FAP+ area (p < 0.001). The expression of the [68 Ga]Ga-FAPI-04 at 4 weeks correlated with the deterioration of cardiac function at 8 weeks (EF: R = − 0.87; FS: R = − 0.72; LVIDd: R = 0.77; LVIDs: R = 0.79; all p < 0.001). The AAC group also showed an increased [68 Ga]Ga-FAPI-04 signal in the liver, peaking at 4 weeks and then declining. Cardiac and liver PET signals correlated at 4 weeks in the AAC group (R = 0.69, p = 0.0010), suggesting an early fibrotic link between organs. A combination of the [68 Ga]Ga-FAPI-04 intensity in the heart and liver at 4 weeks better predicted the deterioration of cardiac function at 8 weeks. Conclusions: The activated fibroblasts in the heart and liver after pressure overload can be monitored by [68 Ga]Ga-FAPI-04 PET/CT, which reveals an early fibrotic link in cardio-liver interactions and could better predict nonischemic heart failure prognosis. A serum metabolomic method based on ultra-high-performance liquid chromatography coupled with mass spectrometry was developed to characterize hyperuricemia-related metabolic profiles and delineate the mechanism of Sanmiao wan (SMW), a traditional Chinese medicine (TCM), in treating hyperuricemic rats. With partial least-squares discriminant analysis for classification and selection of biomarkers, 13 potential biomarkers in mouse serum were identified in the screen, primarily involved in purine metabolism, arginine and proline metabolism, citrate cycle, phenylalanine metabolism, tryptophan metabolism and glycerophospholipid metabolism. Taking these potential biomarkers as screening indexes, SMW could reverse the pathological process of hyperuricemia through partially regulating the perturbed metabolic pathway except for glycerophospholipid metabolism. Our results showed that a metabolomic approach offers a useful tool to identify hyperuricemia-related biomarkers and provides a new methodological cue for systematically dissecting the underlying efficacies and mechanisms of TCM in treating hyperuricemia. Introduction: Calcific aortic valve disease(CAVD) is a complex pathological process for which no effective pharmacological therapy currently exist. Transformation of valvular interstitial cells (VICs) to osteoblasts is believed to be one of the most important causes of valve calcification. Recently, emerging evidence suggests that pro-osteogenic MicroRNAs (miRs) play essential roles in the calcification of the aortic valve. The purpose of this study is to determine whether miR-22 is critically involved in the osteogenic differentiation of VICs, and if so, to determine the molecular mechanisms involved. Methods and Results: A total of 33 CAVD patients were enrolled in the study. The severity of CAVD was determined by standard echocardiographic methods. To identify the aberrant expression of miRNAs in calcified aortic valve, real-time PCR was performed to detect the expression profiles of osteogenic miRNAs in CAVD patients. Subsequently, we identified miR-22 as one of the most significantly up-regulated miRNAs in calcified aortic valves. Fluorescence in situ hybridization assay showed that miR-22 was expressed throughout the regions of the calcified valves and predominantly localized in VICs, as indicated by the co-expression of vimentin. Elevated miR-22 levels were positively correlated with the expression of OPN (rs=0.820, P<0.01) and Runx2 (rs=0.563, P<0.01) as well as VIC osteogenic differentiation. Furthermore, we identified calcium binding protein 39 (CAB39) as a novel downstream target of miR-22 in VICs, as determined by dual-luciferase reporter assay, real-time PCR, and western blot assays. Furthermore, we found that the CAB39 expression was negatively correlated with the calcification severity in clinical CAVD samples, as determined by immunohistochemical staining analysis. Adenovirus-mediated both gain- and loss-of-function analyses demonstrated that miR-22 is critically involved in the osteogenic differentiation of VICs, specifically through regulating the CAB39-AMPK-mTOR signaling pathway. Conclusions: MicroRNA-22 serves as a potential inducer of CAVD through inhibiting the CAB39/AMPK/mTOR signaling pathway. Purpose: Although the adverse effects of atrial fibrillation (AF) on cancers have been well reported, the relationship between the AF and the adverse outcomes in prostate cancer (PC) remains inconclusive. This study aimed to explore the prevalence of AF and evaluate the relationship between AF and clinical outcomes in PC patients., Methods: Patients diagnosed with PC between 2008 and 2017 were identified from the National Inpatient Sample database. The trends in AF prevalence were compared among PC patients and their subgroups. Multivariable regression models were used to assess the associations between AF and in-hospital mortality, length of hospital stay, total cost, and other clinical outcomes., Results: 256,239 PC hospitalizations were identified; 41,356 (83.8%) had no AF and 214,883 (16.2%) had AF. AF prevalence increased from 14.0% in 2008 to 20.1% in 2017 ( P  < .001). In-hospital mortality in PC inpatients with AF increased from 5.1% in 2008 to 8.1% in 2017 ( P  < .001). AF was associated with adverse clinical outcomes, such as in-hospital mortality, congestive heart failure, pulmonary circulation disorders, renal failure, fluid and electrolyte disorders, cardiogenic shock, higher total cost, and longer length of hospital stay., Conclusions: The prevalence of AF among inpatients with PC increased from 2008 to 2017. AF was associated with poor prognosis and higher health resource utilization. Thoracic aortic dissection (TAD) is a highly lethal vascular disease characterized by medial degeneration. Heat shock protein 90 (HSP90) had been proved as a potential target for a variety of diseases. The aim of this study was to identify the effect of HSP90 inhibitor on TAD progress, and to explore the potential utility of HSP90 inhibitors as therapeutic avenue for TAD. In clinical samples, the elevated HSP90 expression was detected in aortic walls from TAD patients (n=20) by real-time PCR and western blot, and was positively correlated with osteopontin (OPN), a synthetic phenotypic marker of smooth muscle cells (SMCs), while negatively correlated with SM22, a contractile phenotypic marker. In a β-aminopropionitrile fumarate-induced AD mice model, 17-DMAG, a HSP90-inhibitor, effectively reduced the incidence and mortality of TAD. Histological examination confirmed that 17-DMAG significantly alleviated the loss of elastic fibers integrity. Meanwhile, the phenotypic switch of SMCs was significantly suppressed by 17-DMAG, demonstrated by the change of phenotypic markers expression. On the cellular level, 17-DMAG suppressed phenotypic switch of SMCs induced by PDGF-bb, and significantly depressed the excessive proliferation and migration of SMCs. Flow cytometry analysis showed that 17-DMAG induced cell cycle arrest in G1 phase. In summary, 17-DMAG could effectively alleviate the TAD progress by suppressing SMCs phenotypic switch, and inhibition of HSP90 might be a potential avenue for TAD therapy., Competing Interests: None.

Endocytosis is a cellular process mostly responsible for membrane receptor internalization. Cell membrane receptors bind to their ligands and form a complex which can be internalized. We previously proposed that F-BAR protein initiates membrane curvature and mediates endocytosis via its binding partners. However, F-BAR protein partners involved in membrane receptor endocytosis and the regulatory mechanism remain unknown. In this study, we established database mining strategies to explore mechanisms underlying receptor-related endocytosis. We identified 34 endocytic membrane receptors and 10 regulating proteins in clathrin-dependent endocytosis (CDE), a major process of membrane receptor internalization. We found that F-BAR protein FCHSD2 (Carom) may facilitate endocytosis via 9 endocytic partners. Carom is highly expressed, along with highly expressed endocytic membrane receptors and partners, in endothelial cells and macrophages. We established 3 models of Carom-receptor complexes and their intracellular trafficking based on protein interaction and subcellular localization. We conclude that Carom may mediate receptor endocytosis and transport endocytic receptors to the cytoplasm for receptor signaling and lysosome/proteasome degradation, or to the nucleus for RNA processing, gene transcription and DNA repair.

Carom is a novel protein that regulates membrane curvature and transmits pathophysiological signaling. The tissue expression of Carom is unclear and its functional role and signaling are unknown. We employed a group of combined database mining strategies and established a working model of Carom signaling. We identified 26 Carom partners and established their expression profiles in human and mouse tissues. We classified three tiers of tissues for Carom/partner expression and found lymph node was the tier 1 tissue expressing Carom and most of its partners. Using GEO database, we discovered that four conditions (hypoxia, endometriosis, PPARgamma deletion and iPSC reprogramming) altered Carom/partner expression in endothelial cells. We identified 26 Carom partner signalings by Ingenuity pathway analysis. Ten of the 26 pathways and three genes (ITSN1, UBC and HSPA5) were reported to be regulated in the above four conditions. Paired induction of Carom/ITSN1 elevation was associated with pathological angiogenesis. Whereas, paired reduction of Carom/HSPA5 or UBC was associated with iPSC generation. These results provide an insight on identifying Carom complex model and predicting its functional implications.
