Ulva green tides have adversely affected coastal ecosystems. In June 2023 in Changhua County, Taiwan, an unprecedented floating tubular Ulva bloom spanning about 30 km of coastline caused about 30 % reduction in oyster yield as reported by local farmers. Understanding its taxonomic and ecological basis is crucial for preemptive and remedial measures. Based on molecular (rbcL and tufA) and morphological evidence, Ulvameridionalis was the cause of this green tide. Haplotype network analysis (based on ITS) suggests that this green tide originated from northern China via the China Coastal Current. Historical survey data indicate that U. meridionalis arrived in Taoyuan Algal Reef (about 150 km north of the bloom area) as far back as 2018. Our ecophysiological experiments revealed that U. meridionalis exhibited a rapid daily growth rate with biomass increment up to 13–21 % when subjected to local nutrient-rich waters under lower salinity and spring-like conditions. Although historical ecological and poultry/livestock data analyses showed no noticeable change in sea surface temperature and rainfall over the past decade, a gradual rise in agricultural nitrogen and phosphorus output was observed. This taxonomic and ecological background lays the groundwork for long-term ecological monitoring. The polyextremophilic red algal class Cyanidiophyceae inhabits a wide range of extreme habitats (e.g. acidic geothermal areas and dim-light caves), and shows various cell morphologies (e.g., subspherical and club-shape) and trophic strategies (e.g., photoautotrophy, heterotrophy and mixotrophy). Historically, inconsistent observations of morphology and physiology have hindered taxonomic advancements in Cyanidiophyceae despite indications of species diversity from molecular studies. Here we revisited this taxonomic conundrum and explored their cryptic speciation based on multi-locus and morpho-physiological evidence examined from expanded taxa sampling, including 20 cultures and one environmental sample. Our multi-locus phylogeny revealed two well-supported monophyletic lineages: the C-lineage that is generically rich (including Cavernulicola, Cyanidiococcus, Cyanidiofrigus, Cyanidioschyzon, Cyanidium, Gronococcus, and Sciadococcus) and the G-lineage that currently only contains the genus Galdieria. Species delimitation analyses revealed at least 21 species in Cyanidiophyceae. Although our morpho-physiological observations failed to differentiate closely related species apart within a genus, we propose Cyanidiofrigus pintoensis gen. et sp. nov. as this species is in a highly divergent lineage with much lower thermal tolerance than other recognized species. Samples of the freshwater red algae, Bangiadulcis atropurpurea, collected from the mountain waterfalls and its close species, Bangia fuscopurpurea, collected from coasts, were phylogenetically analyzed. The sequences of the rbcL gene and RuBisCO spacer region of the freshwater Bangiadulcis atropurpurea species were identical or similar to those of B. atropurpurea from Japan, North America and Europe. This result indicated that the freshwater Bangiadulcis species from Taiwan shared a common ancestor with the three above-mentioned populations and formed a distinct clade from the marine Bangia species in the phylogenetic trees. It is suggested that all the previous records on marine Bangia species should be revised and the name B. fuscopurpurea be used for the marine species in Taiwan. In this study, the freshwater alga B. atropurpurea presents a new record in the algal flora of Taiwan. This finding is important for the protection of the biodiversity of red algal flora, and provides useful information on the ecological conservation of the species in Taiwan.

Abstract Background Gene duplication has been a fundamental process in the evolution of eukaryotic genomes. After duplication one copy (or both) can undergo divergence in sequence, expression pattern, and function. Two divergent copies of the ribosomal protein S13 gene (rps13) of chloroplast origin are found in the nucleus of the rosids Arabidopsis, Gossypium, and Glycine. One encodes chloroplast-imported RPS13 (nucp rps13), while the other encodes mitochondria-imported RPS13 (numit rps13). The rps13 gene has been lost from mitochondrial DNA (mt rps13) of many rosids. Results We studied sequence evolution of numit rps13 in comparison with nucp rps13 in seven rosid genera. Ka/Ks analysis and likelihood ratio tests showed considerably higher Ka values and Ka/Ks ratios in numit rps13 than in nucp rps13, indicating increased amino acid sequence divergence in numit rps13. Two positively selected codons were detected in numit RPS13 in regions that are inferred to interact with the 16S rRNA. Several amino acids in numit RPS13 have changed from the one present in nucp RPS13 to the one present in mt RPS13, showing that numit rps13 is becoming more like mt rps13. Comparison of expression patterns and levels of numit rps13 and nucp rps13 in Arabidopsis using microarray data indicated divergence in gene expression. We discovered that in addition to numit rps13, Malus (apple) contains a transcribed mt rps13 gene. To determine if partitioning of expression takes place between numit rps13 and mt rps13, expression of both copies and RNA editing of mt rps13 were examined by RT-PCR, qRT-PCR, and sequencing from 14 different organ types plus seedlings subjected to five different abiotic stresses. Co-expression of numit rps13 and mt rps13 was observed in all the organs and various stress treatments. We determined that purifying selection is acting on both numit rps13 and mt rps13 in Malus. Conclusion Our data provide evidence that numit rps13 genes in rosids have experienced adaptive sequence evolution and convergent evolution with mt rps13. Co-expression of numit rps13 and mt rps13 and purifying selection on both genes in Malus suggest that both are functional. The three organellar rps13 genes in rosids provide a distinctive case of gene duplication involving the co-evolution of the nuclear and cytoplasmic genomes.

Calcareous green alga in the genus Halimeda are important contributors to the marine carbonate budget. Dongsha Island is located in the northernmost South China Sea and is a seagrass-dominated ecosystem with intermixed Halimeda macroloba patches, making it an excellent system to better examine the extent of carbonate contribution by H. macroloba in such an ecosystem. To this end, we examined the standing stock and actual CaCO3 contribution of H. macroloba in the seagrass-dominated ecosystem (herein Dongsha Island) and compared them with those in Halimeda-dominated ecosystems. The density, growth rate, calcification rate and CaCO3 content of H. macroloba at four life stages were investigated. The mean density of H. macroloba was around 8.82 ± 1.57 thalli m−2 and the estimated standing stock was 61,740 to 72,730 thalli. Thalli produced 1 to 2 new segments day−1, giving a growth rate of 0.003 ± 0.001 g dry weight thallus−1 day−1. Calculated algal biomass and annual areal production were 0.03 g m−2 and 9.66 g m−2 year−1. In each square metre of this area, H. macroloba produced 8.82 to 17.64 new segments day−1, accumulating 0.002 ± 0.001 g CaCO3 thallus−1 day−1 or around 6.44 g CaCO3 m−2 year−1. Mean CaCO3 content was 0.32 ± 0.05 g thallus−1. As expected, the growth rate and CaCO3 production of H. macroloba in Dongsha Island were lower than in other studies from Halimeda tropical ecosystems. Overall, this work provides the baseline of carbonate production of H. macroloba in Dongsha Island and relevant systems where the ecosystem is dominated by seagrasses. [ABSTRACT FROM AUTHOR]

The evolutionary and population demographic history of marine red algae in East Asia is poorly understood. Here, we reconstructed the phylogeographies of two upper intertidal species endemic to East Asia, Gelidiophycus divaricatus and G. freshwateri. Phylogenetic and phylogeographic inferences of 393 mitochondrial cox1, 128 plastid rbcL, and 342 nuclear ITS2 sequences were complemented with ecological niche models. Gelidiophycus divaricatus, a southern species adapted to warm water, is characterized by a high genetic diversity and a strong geographical population structure, characteristic of stable population sizes and sudden reduction to recent expansion. In contrast, G. freshwateri, a northern species adapted to cold temperate conditions, is genetically relatively homogeneous with a shallow population structure resulting from steady population growth and recent equilibrium. The overlap zone of the two species roughly matches summer and winter isotherms, indicating that surface seawater temperature is a key feature influencing species range. Unidirectional genetic introgression was detected at two sites on Jeju Island where G. divaricatus was rare while G. freshwateri was common, suggesting the occurrence of asymmetric natural hybrids, a rarely reported event for rhodophytes. Our results illustrate that Quaternary climate oscillations have left strong imprints on the current day genetic structure and highlight the importance of seawater temperature and sea level change in driving speciation in upper intertidal seaweed species.
