3. WTAP‐induced N6‐methyladenosine of PD‐L1 blocked T‐cell‐mediated antitumor activity under hypoxia in colorectal cancer.

Abstract

N6‐Methyladenosine (m6A) is a important process regulating gene expression post‐transcriptionally. Programmed death ligand 1 (PD‐L1) is a major immune inhibitive checkpoint that facilitates immune evasion and is expressed in tumor cells. In this research we discovered that Wilms' tumor 1‐associated protein (WTAP) degradation caused by ubiquitin‐mediated cleavage in cancer cells (colorectal cancer, CRC) under hypoxia was inhibited by Pumilio homolog 1 (PUM1) directly bound to WTAP. WTAP enhanced PD‐L1 expression in a way that was m6A‐dependent. m6A "reader," Insulin‐like growth factor 2 mRNA‐binding protein 2 (IGF2BP2) identified methylated PD‐L1 transcripts and subsequently fixed its mRNA. Additionally, we found that T‐cell proliferation and its cancer cell‐killing effects were prevented by overexpression of WTAP in vitro and in vivo. Overexpression prevented T cells from proliferating and killing CRC by maintaining the expression of PD‐L1. Further evidence supporting the WTAP–PD‐L1 regulatory axis was found in human CRC and organoid tissues. Tumors with high WTAP levels appeared more responsive to anti‐PD1 immunotherapy, when analyzing samples from patients undergoing treatment. Overall, our findings demonstrated a novel PD‐L1 regulatory mechanism by WTAP‐induced mRNA epigenetic regulation and the possible application of targeting WTAP as immunotherapy for tumor hypoxia. 14. Characterization of the microbial community response to replant diseases in peach orchards

Abstract

This study attempted to monitor the development of microbial communities and reveal the correlation between the soil microbial community and soil nutrient factors over different years following the replanting of peach trees. The replanted soil (RS) and nonreplanted soil (NRS) were collected from peach orchards with different growth years (1, 3, 5, 7, 9, 11, and 13 years) in the same region. The soil bacterial and fungal community diversities were analyzed by high-throughput sequencing technology. Redundancy analysis (RDA) was used to show the correlation between the soil microbial community and environmental variables. The alpha diversities of the bacterial and fungal communities indicated that RS contained a higher abundance of bacterial and fungal operational taxonomic units (OTUs) than NRS. NMDS and ANOSIM analyses showed that the soil bacterial and fungal communities were significantly (P<0.01) affected by planting years, and that the main changes occurred in the first and ninth planting years. The presence of the bacterial orders Sphingobacteriales, Burkholderiales and Actinomycetales changed significantly after replanting. Some bacteria associated with bioremediation, such as Burkholderiales and Intrasporangiaceae, and some harmful pathogens, such as Penicilliumand Ophiostomatales, significantly increased after replanting (LDA score>3.0). In addition, the soil nutrient contents were lower in RS than in NRS in the early stage (1–5 years), and the RDA showed that bacterial and fungal phyla are closely associated with environmental variables, including the potential of hydrogen (pH), ammonium nitrogen (AN), available phosphorus (AP) and available potassium (AK). These results lead to a deeper understanding of the microbial responses to replanting in peach orchards.

28. Chemical composition and insecticidal properties of the essential oil of Bidens frondosa L (Asteraceae) against booklice (Liposcelis bostrychophila)

Abstract

Purpose: To investigate contact and fumigant toxicity of the essential oil of Bidens frondosa and its isolated constituents against booklice (Liposcelis bostrychophila).Methods: The essential oil of B. frondosa was obtained by hydro-distillation and analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) with HP-5MS column. The active constituents were purified from the oil by bioactivity-guided fractionation. Contact (impregnated filter paper method) and fumigant toxicity (sealed space) of the oil and its isolates were determined.Results: Thirty-two compounds, representing 98.88 % of the total oil, were determined and the major constituents of the essential oil were caryophyllene oxide (20.50 %), borneol (17.66 %), 4-terpineol (17.26 %), and β-cedrene (6.94 %). The essential oil displayed fumigant toxicity against booklice, with a median lethal concentration (LC50) of 507.35 μg/L while the isolated constituents, borneol and 4-terpineol, had LC50 values of 2.20 mg/L and 335.24 μg/L against booklice, respectively. The essential oil also exhibited contact toxicity against L. bostrychophila with an LC50 value of 210.73 μg/cm2. Borneol, caryophyllene oxide, β-cedrene, and 4-terpineol showed acute toxicity against booklice with LC50 of 98.04, 84.62, 458.79 and 211.35 μg/cm2, respectively.Conclusion: The results suggest that the essential oil and its isolates possess potential for cultivation into natural insecticides or fumigants, for control of insects in stored grains.Keywords: Bidens frondosa, Liposcelis bostrychophila, Contact toxicity, Essential oil, Boolice, Stored grains, Natural insecticides, Fumigants

Abstract

Purpose: To evaluate the larvicidal activity of the essential oil of Dipsacus asper Wallich ex Candolle (Dipsacaceae) roots against the larvae of Aedes aegypti L. and Culex pipiens pallens Coquillett.Methods: Essential oil was extracted from D. asper roots by hydrodistillation and analyzed for its composition by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). The oil was evaluated for larvicidal activity, using World Health Organization (WHO) procedures, against the fourth larvae of A. aegypti and C. pipiens pallens within 24 h, and activity was recorded for various concentrations of the oil ranging from 12.5 – 200.0 μg/mL.Results: A total of 34 components of the essential oil of D. asper were identified. The major compounds of the essential oil were caryophyllene oxide (13.29 %), caryophyllene (9.14 %), cubebene (7.87 %), β-gurjunene (6.43 %), carvone (5.38 %), 1,8-cineole (5.29 %), and calamenene (5.05 %). The oil exhibited larvicidal activity against A. aegypti and C. pipiens pallens at median lethal concentrations (LC50) of 56.29 μg/mL and 47.49 μg/mL, respectively.Conclusion: The essential oil of D. asper roots has potentials for use in the control of A. aegypti and C. pipiens pallens and may be useful in the search for new, safer and more effective natural larvicides.Keywords: Dipsacus asper, Aedes aegypti, Culex pipiens pallens, Essential oil, Larvicidal activity, Caryophyllene, Cubebene, β-Gurjunene, Carvone
