Basic medical laboratory courses (BMLCs) play an important role in medical educational courses helping the student acquire three important skills of surgical operating, collaborative learning, and problem solving. The outcome-based student assessment (OBSA) is a learning evaluation method that establishes specific evaluation points based on performance of students in three aspects: surgical operating, collaborative learning, and problem solving in the BMLC curriculum practices. The purpose of the present randomized controlled trial study is to explore the efficiency of OBSA program in BMLCs. The 233 students attending BMLCs were randomly divided into 2 groups, 118 in the OBSA group and 115 in the control group. We conducted multiple-choice examination questions (MCQs) test and two questionnaires with the method of two-sample t test for statistics. The results of MCQs in total eight BMLC blocks showed that the academic performance of the OBSA group was significantly better than that of the control group (P < 0.05). In addition, the average scores of direct observation of procedural skills (DOPS) and mini-experimental evaluation exercise in OBSA group were significantly higher than those in control group (P < 0.05). The majority of the medical students preferred the OBSA and considered OBSA could effectively improve their surgical operating skills (83.9%), collaborative learning skills (92.1%), and problem-solving skills (91.1%). From the above, OBSA is an effective evaluation method for the implementation of the BMLC curriculum

The pathogenesis of diabetes is accompanied by increased levels of inflammatory factors, also known as "metabolic inflammation", which runs through the whole process of the occurrence and development of the disease. Mitochondria, as the key site of glucose and lipid metabolism, is often accompanied by mitochondrial function damage in type 2 diabetes mellitus (T2DM). Damaged mitochondria release pro‐inflammatory factors through damage‐related molecular patterns that activate inflammation pathways and reactions to oxidative stress, further aggravate metabolic disorders, and form a vicious circle. Currently, the pathogenesis of diabetes is still unclear, and clinical treatment focuses primarily on symptomatic intervention of the internal environment of disorders of glucose and lipid metabolism with limited clinical efficacy. The proinflammatory effect of mitochondrial damage‐associated molecular pattern (mtDAMP) in T2DM provides a new research direction for exploring the pathogenesis and intervention targets of T2DM. Therefore, this review covers the most recent findings on the molecular mechanism and related signalling cascades of inflammation caused by mtDAMP in T2DM and discusses its pathogenic role of it in the pathological process of T2DM to search potential intervention targets. To investigate the antidiabetic effects and mechanisms of quinoa on type 2 diabetes mellitus (T2DM) mice model. In this context, we induced the T2DM mice model with a high‐fat diet (HFD) combined with streptozotocin (STZ), followed by treatment with a quinoa diet. To explore the impact of quinoa on the intestinal flora, we predicted and validated its potential mechanism of hypoglycemic effect through network pharmacology, molecular docking, western blot, and immunohistochemistry (IHC). We found that quinoa could significantly improve abnormal glucolipid metabolism in T2DM mice. Further analysis showed that quinoa contributed to the improvement of gut microbiota composition positively. Moreover, it could downregulate the expression of TAS1R3 and TRPM5 in the colon. A total of 72 active components were identified by network pharmacology. Among them, TAS1R3 and TRPM5 were successfully docked with the core components of quinoa. These findings confirm that quinoa may exert hypoglycemic effects through gut microbiota and the TAS1R3/TRPM5 taste signaling pathway. [ABSTRACT FROM AUTHOR]

Currently, a series of in vitro methods have been developed on the basis of the adverse outcome pathway (AOP). KeratinoSensT assay which has been developed to address the second key event (KE) of skin sensitization AOP, is the first cell-based in vitro assay t maximize the simulation of human skin sensitization. It is one of the most important in vitro alternative methods for solving the end poi skin allergy that is harmful to human health after exposure to cosmetics. In this review, the mechanism and method of KeratinoSensT assay were briefly introduced. The development of its use in evaluating the skin sensitization of cosmetic raw materials and chemicals we summarized. The limitations in the detection of cosmetic allergens were considered and prospected. The assay is applicable to the detectio of allergens present in cosmetics, including perfumes, plant extracts, metal nano materials, etc., indicative of a high degree of agreemei with in vivo data. In addition, different combination strategies can be used to combine this assay with other test methods to reliabl determine the safety and skin sensitization potential of the substance. The future development direction of KeratinoSens™ assay was als introduced. KeratinoSens™ assay under animal product-free conditions has been adapted, in which human equivalents are used instea of a large number of animal-derived components. In short, this assay provides reference for the selection of appropriate in vitro detectio methods for evaluating the skin sensitization for the raw materials and finished products of cosmetics. Introduction: The functions and mechanisms of prion proteins (PrPC) are currently unknown, but most experts believe that deformed or pathogenic prion proteins (PrPSc) originate from PrPC, and that there may be plural main sites for the conversion of normal PrPC into PrPSc. In order to better understand the mechanism of PrPC transformation to PrPSc, the most important step is to determine the replacement or substitution site.

The streamer discharge occurring on the surface of a conductor in rain is one of the critical problems associated with the design of high voltage direct current (HVDC) transmission lines. In this paper, the streamer discharges on the conductor surface in the presence of raindrops at atmospheric pressure are studied by use of two-dimensional particle-in-cell simulations with Monte Carlo collisions included. The influences of the drop angles, volumes, and spatial distributions of raindrops on the development of streamer discharges have been demonstrated. The formation of streamer discharges and the involved electric fields, plasma densities, propagation velocities, and discharge currents are presented. It is found that the discharge intensity decreases significantly with an increasing of the drop angle. The dividing point at the 95° angle is suggested. With an increase of the raindrop volume and the distance of raindrops, strong discharges with the advanced discharge time, faster propagation velocity, larger electric field, and higher plasma density are initiated. Moreover, the electron energy probability functions of different discharges have been studied, which are nonequilibrium and reveal kinetic behavior of the discharges. The simulation results, which are qualitatively in agreement with the previous experimental observations, bring new insight into the discharge dynamics and provide useful references for the protection of HVDC transmission lines in rainy days. Introduction: Peroxiredoxin 6 (Prdx6) is a bifunctional protein and a unique 1-Cys Prdx of the peroxiredoxin family. The expression and regulation of Prdx6 are implicated in numerous physiological and pathological processes.
