12. Soil Quality Evaluation and Analysis of Driving Factors of Pinus tabuliformis in Loess Hilly Areas.

Abstract

The selection of suitable tree species and the reasonable allocation of planting areas are important measures for improving soil quality. To evaluate the soil quality (SQ) and its driving factors of Pinus tabuliformis forests in loess hilly areas where forestry ecological projects, such as returning farmland to forest (grass), have been implemented, this study selected P. tabuliformis forests with different restoration years (1a, 6a, 11a, 18a, and 22a) in Wuqi County and used grassland before afforestation (PRG) and abandoned grassland (AG) with 22 years as controls. In this study, soil physicochemical indices, soil fauna indices, and herbaceous plant indices obtained via principal component analysis were used to establish a soil quality evaluation model via the fuzzy comprehensive evaluation method to comprehensively evaluate SQ. Structural equation modeling (SEM) was used to identify the key factors affecting the SQ of P. tabuliformis forests. The goal was to create a model that could effectively evaluate the SQ while considering all relevant factors. The findings of the study showed that: (1) by performing a principal component analysis on the 27 indicator factors, the first six principal components had eigenvalues > 1, and the cumulative contribution rate was 90.028%, effectively encompassing the information of the original variables. (2) The highest soil quality index (SQI) was 0.592 (p < 0.05) in the restored 6a P. tabuliformis forest, whereas the lowest SQI was 0.323 in the restored 1a P. tabuliformis forest. As the number of years of restoration increased, the SQ of the P. tabuliformis plantation forest progressively approached that of the long-term abandoned grassland, with only a 1.8% difference after 22 years of restoration. The SQI of the P. tabuliformis woodland in restored 6a was 83% higher than that of 1a, and following 6a of restoration, the SQI showed a decreasing trend with increasing restoration years. Nevertheless, the SQI increased by >52% compared with the early stage of restoration (1a) and by 31% compared with the grassland before afforestation (PRG). (3) SEM revealed that the SQ of P. tabuliformis forest land was mainly driven by soil physical and herbaceous plant indicators, and soil fauna indicators and restoration years had a negative effect on the evolution of SQ in P. tabuliformis forests. The driving factors of P. tabuliformis forests of different restoration years were different, and with the increase in restoration years, the effects of soil fauna and herbaceous plant indicators on the SQ of P. tabuliformis plantation forests showed an overall upward trend. Abstract

Objectives: To determine the imaging details and diagnostic information of the treatment response to neoadjuvant chemoradiotherapy (nCRT) of rectal adenocarcinoma at 9.4T magnetic resonance imaging (MRI) by ex vivo. Methods: Fifteen cases with locally advanced rectal cancer (LARC) followed by radical surgery after nCRT between September 2022 and February 2023 were recruited. Resected specimens were fixed in a perfluoropolyether‐filled test tube and scanned with a 3.0T and 9.4T MRI system ex vivo. The residual tumor depth and MRI‐based tumor regression grade (TRG) were subjectively assessed and then compared with the pathological findings. Results: The ex vivo 9.4T T2WI without fat suppression clearly differentiated tumor tissue, fibrosis and normal rectal wall, which clearly corresponded to the pathologic tissues of the rectal specimens. The TRG could be accurately assessed on ex vivo 9.4T images in 13/15 specimens (86.7%), while in 11/15 specimens (73.3%) on ex vivo 3.0T images. Conclusion: Ex vivo 9.4T MR imaging clearly displayed the components of rectal wall and proved excellent diagnostic performance for evaluating the treatment response to nCRT, which allow radiologists to understand and then assess more accurately the TRG of LARC after nCRT. Abstract

Background: Icaritin is a natural product with a wide range of anti-tumor effects. However, its anti-tumor mechanism has not been thoroughly studied. This study examined the inhibitory effect of icaritin on nasopharyngeal cancer and its underlying mechanism using network pharmacology along with in vivo and in vitro experiments.Methods: MTT and clone formation assays were used to detect the effects of icaritin on the viability and proliferation of nasopharyngeal carcinoma cells, followed by the construction of a HONE1 xenograft tumor model to evaluate the anti-tumor efficacy of icaritin in vivo. A public database was used to predict prospective targets, built a protein-protein interaction (PPI) network, and analyze gene enrichment and biological processes. Based on network pharmacological data, cell cycle-related proteins were identified using western blotting. Besides, cell cycle distribution, apoptosis, and intracellular reactive oxygen species (ROS) generation were identified using flow cytometry. In addition, SA-β-Gal staining was performed to detect cellular senescence, and western blotting was performed to detect the expression of P53, P21, and other proteins to verify key signaling pathways.Results: Icaritin effectively inhibited the viability and proliferation of nasopharyngeal carcinoma cell lines and showed good anti-tumor activity against HONE1 nasopharyngeal carcinoma cells in vivo. Key protein targets, including AKT1, HSP90AA1, CDK4, CCND1, and EGFR, were screened using PPI network topology analysis. GO and KEGG analysis revealed that the cell cycle, p53 signaling, and cell senescence pathways may be the main regulatory pathways. Flow cytometry and western blot experiments showed that icaritin caused S-phase arrest and promoted an increase in ROS. SA-β-Gal staining showed that icaritin significantly induced cellular senescence, and western blotting showed that the expression of senescence-related proteins p53 and P21 increased significantly. Moreover, inhibition of ROS levels by N-Acetylcysteine (NAC) enhanced cell viability, reversed cellular senescence and reduced cellular senescence-associated protein expression.Conclusion: The results of network pharmacological analysis and in vivo and in vitro experiments showed that icaritin effectively inhibited the growth of nasopharyngeal carcinoma cells, promoted ROS production, induced cellular senescence, and inhibited tumor cells, which are related to the regulation of P53/P21 signal pathway.

Abstract

Vib1, a member of the Ndt80/PhoG-like transcription factor family, has been shown to be essential for cellulase production of Trichoderma reesei. Here, we combined transcriptomic and genetic analyses to gain mechanistic insights into the roles of Vib1 during cellulose degradation. Our transcriptome analysis showed that the vib1 deletion caused 586 genes with decreased expression and 431 genes with increased expression on cellulose. The downregulated genes were enriched for Gene Ontology terms associated with carbohydrate metabolism, transmembrane transport, oxidoreductase activity, and transcription factor activity. Of the 258 genes induced by cellulose, 229 showed no or decreased expression in Δvib1 on cellulose, including almost all (hemi)cellulase genes, crucial sugar transporter genes (IDs:69957, 3405), and the genes encoding main transcriptional activators Xyr1 and Ace3. Additionally, Vib1 also regulated the expression of genes involved in secondary metabolism. Further comparison of the transcriptomes of Δvib1 and Δxyr1 in cellulose revealed that the genes regulated by Vib1 had much overlap with Xyr1 targets especially for the gene set induced by cellulose, presumably whose expression requires the cooperativity between Vib1 and Xyr1. Genetic evidence indicated that Vib1 regulates cellulase gene expression partially via Xyr1. Our results will provide new clues for strain improvement.

50. Experimental study on substrate with hierarchical nested channels for thermal interface resistance control.

Abstract

Thermal interface resistance is an important parameter for the thermal management of electronics packaging. In order to reduce thermal interface resistance, hierarchical nested channels were designed and fabricated on the surfaces of aluminum and copper sheets. Experiments were conducted to measure the thermal interface resistance of the sheets. A sequence of experimental results revealed that thermal interface resistance of the sheets with hierarchical nested channels is 2-3 times larger than that of the original flat sheets without the channels, which is contrary to the experimental objective. A comprehensive experimental analysis was presented to explain the phenomenon. It is found that the roughness of contact interface plays a key role. In the reported references, the thermal interface material composed of metal particles is easy to form particle stacking in the smooth contacting surface, therefore, the hierarchical nested channels can effectively reduce the particle stacking and the corresponding thermal interface resistance. However, in the present experiment samples, the surface roughness is larger than 10 μm, the phenomenon of particle stacking is difficult to occur, the hierarchical nested channels cannot perform well for decreasing particle stacking and thermal resistance. Otherwise, the hierarchical nested channels increases the bulk thermal resistance because of the replacement of the high thermal conductivity metal material with thermal greases that have relatively low thermal conductivity.
