Abstract

Massive stars end their life as core-collapse supernovae, amongst which some extremes are Type Ic broad-lined supernovae associated with long-duration gamma-ray bursts (LGRBs) having powerful relativistic jets. Their less-extreme brethren make unsuccessful jets that are choked inside the stars, appearing as X-ray flashes or low-luminosity GRBs. On the other hand, there exists a population of extragalactic fast X-ray transients (EFXTs) with timescales ranging from seconds to thousands of seconds, whose origins remain obscure. Known sources that contribute to the observed EFXT population include the softer analogs of LGRBs, shock breakouts of supernovae, or unsuccessful jets. Here, we report the discovery of the bright X-ray transient EP240414a detected by the Einstein Probe (EP), which is associated with the Type Ic supernova SN 2024gsa at a redshift of 0.401. The X-ray emission evolution is characterised by a very soft energy spectrum peaking at < 1.3 keV, which makes it distinct from known LGRBs, X-ray flashes, or low-luminosity GRBs. Follow-up observations at optical and radio bands revealed the existence of a weak relativistic jet that interacts with an extended shell surrounding the progenitor star. Located on the outskirts of a massive galaxy, this event reveals a new population of explosions of Wolf-Rayet stars characterised by a less powerful engine that drives a successful but weak jet, possibly owing to a progenitor star with a smaller core angular momentum than in traditional LGRB progenitors., Comment: 43 pages, 9 figures, 4 tables, submitted. Abstract

Long gamma-ray bursts (GRBs) are believed to originate from core collapse of massive stars. High-redshift GRBs can probe the star formation and reionization history of the early universe, but their detection remains rare. Here we report the detection of a GRB triggered in the 0.5--4 keV band by the Wide-field X-ray Telescope (WXT) on board the Einstein Probe (EP) mission, designated as EP240315a, whose bright peak was also detected by the Swift Burst Alert Telescope and Konus-Wind through off-line analyses. At a redshift of $z=4.859$, EP240315a showed a much longer and more complicated light curve in the soft X-ray band than in gamma-rays. Benefiting from a large field-of-view ($\sim$3600 deg$^2$) and a high sensitivity, EP-WXT captured the earlier engine activation and extended late engine activity through a continuous detection. With a peak X-ray flux at the faint end of previously known high-$z$ GRBs, the detection of EP240315a demonstrates the great potential for EP to study the early universe via GRBs., Comment: 41 pages, 8 figures, 7 tables

Abstract

The Lobster Eye Imager for Astronomy (LEIA), a pathfinder of the Wide-field X-ray Telescope of the Einstein Probe (EP) mission, was successfully launched onboard the SATech-01 satellite of the Chinese Academy of Sciences on 27 July 2022. In this paper, we introduce the design and on-ground test results of the LEIA instrument. Using state-of-the-art Micro-Pore Optics (MPO), a wide field-of-view (FoV) of 346 square degrees (18.6 degrees * 18.6 degrees) of the X-ray imager is realized. An optical assembly composed of 36 MPO chips is used to focus incident X-ray photons, and four large-format complementary metal-oxide semiconductor (CMOS) sensors, each of 6 cm * 6 cm, are used as the focal plane detectors. The instrument has an angular resolution of 4 - 8 arcmin (in FWHM) for the central focal spot of the point spread function, and an effective area of 2 - 3 cm2 at 1 keV in essentially all the directions within the field of view. The detection passband is 0.5 - 4 keV in the soft X-rays and the sensitivity is 2 - 3 * 10-11 erg s-1 cm-2 (about 1 mini-Crab) at 1,000 second observation. The total weight of LEIA is 56 kg and the power is 85 W. The satellite, with a design lifetime of 2 years, operates in a Sun-synchronous orbit of 500 km with an orbital period of 95 minutes. LEIA is paving the way for future missions by verifying in flight the technologies of both novel focusing imaging optics and CMOS sensors for X-ray observation, and by optimizing the working setups of the instrumental parameters. 24. MiR-146a relieves kidney injury in mice with systemic lupus erythematosus through regulating NF-κB pathway.

Abstract

OBJECTIVE: To explore the effect of micro ribonucleic acid (miRNA)-146a on kidney injury in mice with systemic lupus erythematosus (SLE), and to investigate its possible mechanism. MATERIALS AND METHODS: A total of 45 female MRL/lpr mice were randomly divided into control group, miR-146a mimic group and miR-146a inhibitor group. Urine protein level was measured every 2 weeks. Meanwhile, the levels of serum anti-dsdeoxyribonucleic acid (anti-dsDNA), anti-ssDNA, antinuclear antibody (ANA) and anti-chromatin were measured using enzyme-linked immunosorbent assay (ELISA). At 2 weeks after drug treatment, the effects of miR-146a mimic and inhibitor on kidney tissues of MRL/lpr mice were detected and analyzed by gene chip and gene set enrichment analysis, respectively. The mice were executed at the age of 24 weeks, and the blood samples were collected. Subsequently, the level of blood urea nitrogen (BUN) was measured using the BUN analyzer. After that, kidney tissues were taken, and the effect of drug treatment on the morphology of kidney tissues was detected via hematoxylin-eosin (HE) staining. Moreover, the effects of drug treatment on the mRNA levels of inflammatory factors and the nuclear factor-κB (NF-κB) signaling pathway in kidney tissues were detected via quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting, respectively. RESULTS: MiR-146a mimic significantly reduced urine protein in a time-dependent manner, which also significantly reduced BUN level at 24 weeks. The results of HE staining showed that both glomerular injury and renal vascular injury in miR-146a mimic group were significantly alleviated. In miR-146a mimic group, serum autoantibodies of anti-dsDNA, anti-ssDNA, anti-chromatin and ANA decreased significantly. However, the survival time of mice was significantly prolonged. High-throughput gene expression chip technique elucidated that in miR-146a mimic group, the expression of positive regulatory gene of NF-κB showed a decreasing trend. However, the expression of negative regulatory gene of NF-κB showed an increasing trend. MiR-146a mimic remarkably down-regulated the expression levels of RELA, IRAK1, interleukin-1B (IL1B) and IL-10 in kidney tissues. Furthermore, the results of Western blotting showed that miR-146a mimic inhibited both the classical and non-classical NF-κB signaling pathways. 25. LINC00052 inhibits tumor growth, invasion and metastasis by repressing STAT3 in cervical carcinoma.

Abstract

OBJECTIVE: The vital role of long noncoding RNAs (lncRNAs) in tumor progression has been identified in numerous studies. In this research, the biological function of lncRNA LINC00052 during the development of cervical cancer was mainly explored. PATIENTS AND METHODS: LINC00052 expression was detected by quantitative Real-time polymerase chain reaction (qRT-PCR) in cervical cancer tissue samples and cell lines. Moreover, the correlation between LINC00052 expression level and disease-free survival rate of cervical cancer patients was analyzed. In vitro functions of LINC00052 in cervical cancer cells were evaluated by proliferation assay, wound healing assay and transwell assay. In addition, qRT-PCR and Western blot were utilized to explore the underlying mechanism of LINC00052 in mediating the progression of cervical cancer. RESULTS: LINC00052 expression level was lower in cervical cancer samples than that in adjacent tissues, which was correlated with disease-free survival time. Moreover, cell proliferation, migration and invasion were inhibited through overexpression of LINC00052 in vitro. The mRNA and protein expression of signal transducers and activators of transcription 3 (STAT3) was downregulated after overexpressing LINC00052 in cervical cancer cells. The STAT3 expression level was negatively correlated with the expression of LINC00052 in cervical cancer tissues. CONCLUSIONS: LINC00052 could repress metastasis and invasion of cervical cancer cell via suppressing STAT3. LINC00052 might be a novel tumor suppressor in cervical cancer. [ABSTRACT FROM AUTHOR]

Abstract

Tetrapyrroles, such as chlorophyll and heme, are integral to the metabolism of all living organisms. Glutamyl-tRNA reductase (GluTR) is the first unique enzyme in the tetrapyrrole biosynthetic pathway in plants. We firstly cloned the GluTR complete sequences (1733 bp) from Chinese jujube (Ziziphus jujuba Mill., belonging to the family Rhamnaceae), named as ZjGluTR (GenBank accession no. KF530842). ZjGluTR shares high similarity (90%) to those of Malus domestica and Prunus mume through BLASTX analysis. It contains a 1659-bp ORF and encodes a predicted polypeptide of 552 amino acids, with an estimated molecular mass of 60.0 kDa and a theoretical pI of 8.55. The speculated formula of ZjGluTR is C2615H4318N756 O804S25. ZjGluTR has a chloroplast transit peptide which contains 61 amino acids in the N terminal. The subcellular localization result showed that ZjGluTR protein exists in chloroplasts. ZjGluTR has three typical functional domains, i.e. GluTR N-terminal domain (101-259 aa), NAD(P)-binding Rossmann-fold domain (266-422 aa) and GluTR dimerization domain (425-534 aa). The molecular phylogenetic tree of GluTR indicated that the family Rhamnaceae has a close genetic relationship with the family Rosaceae. Our studies on the GluTR using molecular biology and bioinformatics approaches would play an important role in chlorophyll metabolic research of Chinese jujube. [ABSTRACT FROM AUTHOR]

Abstract

Silicon on insulator (SOI)-based transistors gated by nanogranular SiO2 proton conducting electrolytes were fabricated to mimic synapse behaviors. This SOI-based device has both top proton gate and bottom buried oxide gate. Electrical transfer properties of top proton gate show hysteresis curves different from those of bottom gate, and therefore, excitatory post-synaptic current and paired pulse facilitation (PPF) behavior of biological synapses are mimicked. Moreover, we noticed that PPF index can be effectively tuned by the spike interval applied on the top proton gate. Synaptic behaviors and functions, like short-term memory, and its properties are also experimentally demonstrated in our device. The results show that the initial damage affects the stress, fatigue life and future damage of components.

46. Extraction of rare earth elements from NdFeB scrap by AlF3–NaF melts

Abstract

To conserve both energy and rare earths, a new technology for recycling of NdFeB magnet scrap was proposed in this paper based on the selective fluorination with AlF3–NaF melts. After fluorination, the reaction products were examined by scanning electron microscopy equipped with energy dispersive X-ray spectrometry, X-ray diffraction and inductively coupled plasma. Rare earth elements in the scrap were selectively converted to rare earth fluorides in the yield of ca. 94·2, leaving Fe, B and impurities in the solid residue. The differences in chemical activity between Fe, impurities and rare earth should be mainly responsible for the selective fluorination process. The obtained fluorides can be subsequently processed with molten salt electrolysis for the direct production of Al–Nd based alloy.

Abstract

In Streptococcus mutans, virulence and cariogenicity may be modulated via the two-component regulatory system VicRK. Environmental signals, sensed by VicK, inducing this modulation are still unclear, however, and were investigated in the present study. We found that VicRK displays homology with proteindomains that, in other bacteria, are involved in redox-sensing. After constructing a VicRK-promoter GFP-reporter strain, we showed increased fluorescence intensity under oxidative stress. Potential interference of alternative signals and experimental conditions on GFP expression was excluded by the use of negative and positive control strains. Finally, we constructed a clean vicK knockout mutant, which proved to be more sensitive to H2O2 than the wild-type. In conclusion, this study showed that the VicRK system responds to and protects against oxidative stress. As a result, a link between oxidative/redox stress and the cariogenic nature of S. mutans can be hypothesized. [ABSTRACT FROM AUTHOR]

Abstract

The experimental results of space, azimuthal, and projected angular distributions of target black fragments produced in silicon-emulsion collisions at 4.5A GeV/c (the Dubna energy) are reported. A multi-source ideal gas model is suggested to describe the experimental angular distributions. The Monte Carlo calculated results are in agreement with the experimental data. [ABSTRACT FROM AUTHOR]
