Two lactic acid bacteria (LAB) strains of Lactobacillus plantarum 21802 and Lactobacillus brevis 6239 were selected and combined for fermentation of grape juice. The changes of sugars, organic acids, volatiles, phenolic compounds as well as antioxidant capacity of the grape juice were characterized. Results showed that fermentation with LAB led to a slight decrease in sugar content and increased in organic acid content. The sugars were mainly consumed at the initial stage of fermentation, while the malic acid was consumed completely during the fermentation. The contents of total phenolics and flavonoids in grape juice decreased gradually during the fermentation process. As compared with the initial contents in unfermented grape juice, the contents of proanthocyanin B2, rutin, isoquercitrin, rhamnose-3-O-glucoside in the final fermented grape juice were increased by 26.83%, 249.24%, 138.48%, 139.60%, respectively, while the contents of gallic acid, proanthocyanin B1, catechin and epicatechin were decreased by 24.49%, 14.85%, 71.68%, 42.52%, respectively. The mice fed with the fermented grape juice showed higher activity of superoxide dismutase and lower level of malondialdehyde both in serum and liver tissue than those fed with the unfermented grape juice. Fermentation with LAB also enhanced the flavor profile by increasing the total number and content of volatile compounds, especially those of monoterpene alcohols, which are the main and characteristic aroma volatile compounds in Muscat aroma grapes. This research confirmed that fermentation with LAB might be a desirable option for improving the flavor and nutritional attributes of grape juice by altering the ratio of sugar to organic acid, improving the phenolic compounds, enhancing the antioxidant capacity, intensifying of the characteristic aroma and pleasant flavor of the grape juice.

This study is aimed at determining the relationship of flavonoid structures to their chemical and intracellular antioxidant activities. The antioxidant activities of 60 flavonoids were investigated by three different antioxidant assays, including 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, oxygen radical absorption capacity (ORAC), and cellular antioxidant activity (CAA) assays. The result showed 6 flavonoids as good cellular antioxidants evaluated for the first time. The cellular antioxidant activities of compounds 7-methoxy-quercetin, 3-O-methylquercetin, 8-hydroxy-kaempferol, quercetin-3-O-α-arabinofuranose, kaempferol-7-O-glucopyranoside, and luteolin6-C-glucoside were linked with the upregulation of antioxidant enzyme activities (superoxide dismutase, catalase, and glutathione peroxidase). A structure-activity relationship suggested that 2,3-double bond, 4-keto groups, 3′,4′-catechol structure, and 3-hydroxyl in the flavonoid skeleton played important roles in the antioxidant behavior. Furthermore, the cell proliferative assay revealed a low cytotoxicity for 3-O-methylquercetin. The present results provide valuable information for the dietary application of flavonoids with different structures for high antioxidant.

The physicochemical properties, organic acid, and phenolic profiles of hawthorn wines prepared by different pretreatments were analyzed. It was found that fermentation together with smashed seeds was favorable for ethanol production, while thermal maceration, addition of pectinase and amyloglucosidase significantly increased the levels of sugar-free extract, residual sugar, total phenolics, flavonoids, and most individual phenolic comounds, showing beneficial effects on improving nutritive value of hawthorn wine. Addition of pectinase and amyloglucosidase caused an increase of methanol content by 34.88% and 26.97%, respectively, while thermal maceration decrease the methanol content by 36.20%. Addition of amyloglucosidase yielded the highest contents of procyanidins as well as flavanols, while thermal maceration and addition of pectinase were favorable for improving phenolic acid level. All the pretreatments significantly increased the contents of succinic acid and malic acid in the final wines, among which thermal maceration and addition of pectinase yielded the highest content of succinic acid and malic acid respectively.

A comparison of sugar and organic acid profiles among different fruit juices (including apple, pear, peach, grape, sweet cherry, strawberry, and blueberry with various varieties) was performed to assess the possibility for authentication coupled with chemometrics. It was found that the distribution of each sugar and organic acid in juices showed some specific characteristics related to fruit species, despite the fact that great differences in the content existed among different varieties. Sucrose was the most abundant sugar in peach juice, accounting for 58.26–77.11% of the total sugar content. However, in grape, blueberry, and sweet cherry juice, glucose and fructose were the predominant sugars. Pear juice contained the highest level of sorbitol, which contributed to 15.02–43.07% of the total sugar content. Tartaric acid was detected only in grape juice among the seven species of fruit juice, with a proportion of 57.95–89.68% in the total acid content. Malic acid was the predominant organic acid in apple and sweet cherry juice, accounting for 69.92–88.30% and 97.51–98.73% of the total acid content of each species. Citric acid was the predominant organic acid in strawberry and blueberry juice, which contributed to 62.39–83.73% and 73.36–89.56% of the total acid content of each species. With the aid of principal component analysis and linear discriminant analysis (LDA), the juice samples could be successfully classified according to fruit species by using the sugar and/or organic acid composition as analytical data. Combination of sugar and organic acid composition gave the best differentiation of these seven species of juices, with a 100% correct classification rate for both the original and the cross-validation method in LDA. Adding malic/citric into the dataset of the organic acid content may also improve the differentiation effect. Furthermore, the adulteration of sweet cherry juice, blueberry juice, raspberry juice, and grape juice with apple juice, pear juice, or peach juice could also be distinguished from their corresponding pure juices based on sugar and organic acid composition by LDA.

Fresh peach blossoms of two varieties were dried by using different methods, which include shade drying, freeze drying, microwave drying and hot air drying (at 30, 60, 90, 120 °C). The effects of different drying methods on phenolic contents, antioxidant and tyrosinase inhibitory activity were evaluated. Among the seven drying treatments, microwave drying yielded the highest contents of total phenolics, flavonoids, anthocyanins, proanthocyanidins and individual phenolic compounds in dried peach blossoms, while hot air drying at 30 °C gave the lowest retention of total phenolics, flavonoids, anthocyanins. As compared with shade drying and freeze drying, hot air drying at 60, 90, 120 °C resulted in higher retention of phenolic content. The highest antioxidant activity in dried peach blossoms was obtained by microwave drying, while hot air drying at 30 °C gave the lowest. The effect of different drying methods on antioxidant activity of dried peach blossoms was consistent with that of phenolic content. The highest tyrosinase inhibitory activity was achieved by hot air drying at 120 °C for variety ‘yingchun’ and 90 °C for variety ‘huangjinmeili’, but there was no significant difference among hot air drying at 60, 90, 120 °C for both varieties. The tyrosinase inhibitory activities of peach blossoms by microwave drying and freeze drying were comparable and much lower than that of shade drying. The results indicated that microwave drying was a desirable method for the preservation of phenolic compounds and antioxidant activity in peach blossoms, while shade drying and hot air drying at high temperature were favorable for tyrosinase inhibitory activity.

Supercritical fluid extraction (SFE) was used for the extraction of phenolic compounds from peach blossom (Amygdalus persica). The optimum conditions of SFE were optimized by response surface methodology and confirmed at 64°C, 30 MPa, 143 min, and 35 mL ethanol (100%) as modifier for maximum total phenolic contents (54.10 mg GAE g−1 DW), which was higher than the yield achieved by ultrasonic-assisted extraction (44.04 mg GAE g−1 DW). The antioxidant activities and tyrosinase inhibitory activities of extraction extracted by SFE were 79.55% and 14.42%. Furthermore, chlorogenic acid, quercetin-3-O-galactoside, kaempferol-3-O-galactoside, quercitrin-3-O-rhamnoside, and kaempferol 4′-glucoside were identified in this study.

Peach blossoms were harvested and classified into six developmental stages: (I) bud emerging stage, (II) middle bud stage, (III) large bud stage, (IV) initial-flowering stage, (V) full-flowering stage, and (VI) end-flowering stage. The contents of total phenolics, flavanoids, individual phenolic compounds as well as antioxidant and tyrosinase inhibitory activity of peach blossoms at different developmental stages were investigated. The total phenolic contents varied from 149.80 to 74.80 mg chlorogenic acid equivalents/g dry weight (DW), and the total flavanoid contents ranged from 93.03 to 44.06 mg rutin equivalents/g DW. Both the contents of total phenolics and flavanoids decreased during blossom development. Chlorogenic acid was the predominant component, accounting for 62.08%–71.09% of the total amount of identified phenolic compounds in peach blossom. The antioxidant capacities determined by different assays and tyrosinase inhibitory activity also showed descending patterns during blossom development. Significant correlations were observed between antioxidant capacities with contents of total phenolics and total flavanoids as well as chlorogenic acid, cinnamic acid and kaempferol-3-O-galactoside, while the tyrosinase inhibitory activity had lower correlations with total phenolics and total flavanoids as well as chlorogenic acid, quercetin-3-O-rhamnoside, kaempferol-3-O-galactoside and cinnamic acid. The antioxidant activities of peach blossom seemed to be more dependent on the phenolic compounds than tyrosinase inhibitory activity.

Supercritical fluid extraction (SFE) was utilized for the first time to extract phenolics in date seeds. Orthogonal array design (OA9 (34)) was applied to optimize extraction variables including extraction temperature, extraction pressure, extraction time, and number of extraction. Optimum values of extraction variables were analyzed and number of extractions was found to have a significant effect on total phenolics content (TPC). Optimal SFE conditions for maximum yield of TPC were 50 °C, 350 bar, and two repeated extractions, each for 2 h. Under optimal condition, the TPC was increased to 441.57 mg gallic acid equivalents/100 g fresh weight. Several phenolic compounds were detected in date seeds including chlorogenic acid, rutin, ellagic acid, quercetin, and two unidentified compounds of phenolic acids. In addition, phenolics of date seeds showed a higher antioxidant activity than ascorbic acid at the same concentration in the range of 1.0–8.0 mg/L.
