4. Coexistence of multiple interfacial states at heterogeneous solid/liquid interface

Abstract

The growing trend towards engineering interfacial complexion (or phase) transitions has been seen in the grain boundary and solid surface systems.Meanwhile, little attention has been paid to the chemically heterogeneous solid/liquid interfaces. In this work, novel in-plane multi-interfacial states coexist within the Cu(111)/Pb(l) interface at a temperature just above the Pb freezing point is uncovered using atomistic simulations.Four monolayer interfacial states, i.e., two CuPb alloy liquids and two pre-freezing Pb solids, are observed coexisting within two interfacial layers sandwiched between the bulk solid Cu and bulk liquid Pb. Through computing the spatial variations of various properties along the direction normal to the in-plane solid-liquid boundary lines for both interfacial layers, a rich and varied picture depicting the inhomogeneity and anisotropy in the mechanical, thermodynamical, and dynamical properties is presented. The bulk values extracted from the in-plane profiles suggest that each interfacial state examined has distinct equilibrium values from each other and significantly deviates from those of the bulk solid and liquid phases, and indicate that the complexion (or phase) diagrams for the Cu(111)/Pb(l) interface bears a resemblance to that of the eutectic binary alloy systems, instead of the monotectic phase diagram for the bulk CuPb alloy. 11. Knockdown of ANP32E inhibits colorectal cancer cell growth and glycolysis by regulating the AKT/mTOR pathway

Abstract

Colorectal cancer (CRC) is the third most common tumor, with an increasing number of deaths worldwide each year. Tremendous advances in the diagnosis and treatment of CRC have significantly improved the outcomes for CRC patients. Additionally, accumulating evidence has hinted the relationship between acidic nuclear phosphoprotein 32 family member E (ANP32E) and cancer progression. But the role of ANP32E in CRC remains unclear. In our study, through TCGA database, it was demonstrated that the expression of ANP32E was enhanced in COAD tissues (n = 286). In addition, the mRNA and protein expression of ANP32E was also confirmed to be upregulated in CRC cell lines. Further investigation uncovered that knockdown of ANP32E suppressed cell proliferation and glycolysis, and facilitated cell apoptosis in CRC. Moreover, inhibition of ANP32E inhibited the AKT/mTOR pathway. Through rescue assays, we discovered that the reduced cell proliferation, glycolysis and the enhanced cell apoptosis mediated by ANP32E repression was reversed by SC79 treatment. In summary, ANP32E aggravated the growth and glycolysis of CRC cells by stimulating the AKT/mTOR pathway. This finding suggested that the ANP32E has the potential to be explored as a novel biomarker for CRC treatment.

37. Can all-atom protein dynamics be reconstructed from the knowledge of C-alpha time evolution?

Abstract

We inquire to what extent protein peptide plane and side chain dynamics can be reconstructed from knowledge of C-alpha dynamics. Due to lack of experimental data we analyze all atom molecular dynamics trajectories from Anton supercomputer, and for clarity we limit our attention to the peptide plane O atoms and side chain C-beta atoms. We try and reconstruct their dynamics using four different approaches. Three of these are the publicly available reconstruction programs Pulchra, Remo Scwrl4. The fourth, Statistical Method, builds entirely on statistical analysis of Protein Data Bank (PDB) structures. All four methods place the O and C-beta atoms accurately along the Anton trajectories. However, the Statistical Method performs best. The results suggest that under physiological conditions, the all atom dynamics is slaved to that of C-alpha atoms. The results can help improve all atom force fields, and advance reconstruction and refinement methods for reduced protein structures. The results provide impetus for development of effective coarse grained force fields in terms of reduced coordinates., Comment: 24 figures
