Background/objectives To evaluate the role of the potential inflammatory effects of diet using the Energy adjusted Dietary Inflammatory Index (E-DII) for oral cancer. Subjects/methods A case-control study including 295 oral cancer cases and 425 controls from September 2010 to June 2018 was performed in Fujian Province, China. The E-DII was calculated based on the food frequency questionnaire (FFQ) and adjusted by total energy intake. The association between E-DII and the risk of oral cancer was estimated with unconditional logistic regression model. Results Compared with E-DII score in the lowest quartile, those with E-DII score in the fourth quartile were at the higher risk of oral cancer (OR = 2.57; 95% CI: 1.54, 4.29, P.sub.trend = 0.013). When analyses were carried out using E-DII as a continuous variable, one-unit increase in E-DII increased the odds of having oral cancer by 3% (95% CI: 1.00, 1.06). Moreover, there was a significant interaction between the E-DII and oral hygiene for oral cancer (P.sub.interaction < 0.001, in those without and with poor hygiene, the OR (95% CI) were 1.96 (0.96, 4.00) and 4.23 (1.83, 9.81), respectively). Conclusions The present study suggests that the higher E-DII score, indicated a pro-inflammatory diet, may be a risk factor for oral cancer in southeast of China. Objective: To assess the prognostic role of pretreatment lymphocyte percentage (LY%) for patients with oral squamous cell carcinoma (OSCC). Methods: A large‐scale prospective cohort study between July 2002 and March 2021 was conducted. Propensity score‐matched (PSM) analysis and inverse probability of treatment weighting (IPTW) analysis were performed to adjust for potential confounders. Using random survival forest (RSF), the relative importance of pretreatment LY% in prognosis prediction was also assessed. Results: A total of 743 patients were enrolled and followed up (median: 2.75 years, interquartile range: 1.25–4.42 years). A high pretreatment LY% was significantly associated with better disease‐specific survival of patients with OSCC (Hazard ratio [HR] = 0.60, 95% confidence interval [CI]: 0.42, 0.84). The same tendency was observed in PSM (HR = 0.57, 95% CI: 0.38, 0.85) and IPTW analysis (HR = 0.57, 95% CI: 0.40, 0.82). RSF showed that LY% ranked the fifth among importance ranking of all prognostic factors. Conclusion: Pretreatment LY% showed a moderate predictive ability, suggesting it might be a valuable tool to predict prognosis for patients with OSCC. [ABSTRACT FROM AUTHOR]

Objective To explore the effects of oral hygiene, vegetables and fruits intake on gingival cancer. Methods A case-control study including 131 patients pathologically confirmed as gingival cancer from October 2010 to November 2016 and 669 health controls was performed by questionnaire survey. Unconditional Logistic regression was used to assess the effect of oral hygiene, vegetables and fruits intake on gingival cancer. Results Teeth brushing < 2 times/day, lacking of teeth, wearing dentures, irregular dental visits and recurrent dental ulceration were risk factors of gingival cancer. We developed a composite oral hygiene score to measure the combined effects of various oral hygiene. The risk of gingival cancer among patients scored 3-5 was 6.991 times(95%CI: 4.267, 11.453) as those scored 0-2. Eating green leafy vegetables < 2 times/day, non-green leafy vegetables < 2 times/day, fruit < 1 time/day were risk factors of gingival cancer. The people scored 3-5 had higher risk of gingival cancer in all subgroups. Furthermore, the people with lower frequency consumption of green leafy vegetables, non-leafy vegetables and fruits had the higher risk of gingival cancer. Multiplicative interaction of green leafy vegetables, non-leafy vegetables, fruits with oral hygiene was observed. Conclusion The people with higher oral hygiene index have higher risk of gingival cancer. Eating green leafy vegetables < 2 times/day, non-green leafy vegetables < 2 times/day, fruit < 1 time/day could increase the risk of gingival cancer.

Background: To evaluate whether exosomal circRNAs could serve as diagnostic biomarkers for the accurate preoperative prediction of lymph node metastasis (LNM) risk in oral squamous cell carcinoma (OSCC) patients. Methods: A combinative strategy of exosomal circRNAs microarray and qRT‐PCR verification was employed to dig LNM‐related circRNA signatures. Then, a dynamic nomogram was developed based on candidate circRNAs and preoperative clinical features and the calibration, discrimination, and clinical use of the nomogram were evaluated. Results: According to the microarray, three circRNAs derived from the tumor were associated with preoperative LNM risk, including hsa_circRNA_047733, hsa_circRNA_024144 and hsa_circRNA_403472. The hsa_circRNA_047733 was further verified to be significantly downregulated in patients with LNM (+) as compared with those with LNM (−) (p = 0.007). Patients with the higher expression of hsa_circRNA_047733 showed a lower risk of LNM (multivariate‐adjusted OR = 0.22, 95%CI: 0.06–0.83). The bioinformatics prediction showed that hsa_circRNA_047733 might sponge miR‐4464/miR‐4748 to regulate RPS21 expression. A dynamic nomogram integrating exosomal hsa_circRNA_047733 with five clinicopathological characteristics (tumor site, leukocyte level, maximum tumor diameter, and LNM reported by MRI and preoperative biopsy differentiation) was developed. The model displayed an excellent discrimination ability (AUC = 0.868, 95%CI: 0.781–0.955) and great calibration. The decision curve revealed a higher net benefit superior to the baseline model at an 80% threshold probability. Conclusion: The data provide preliminary evidence that exosomal hsa_circRNA_047733 might be a novel biomarker for the LNM of OSCC. The hsa_circRNA_047733‐based dynamic nomogram could serve as a convenient preoperative assessment tool to predict the risk of LNM for OSCC patients. [ABSTRACT FROM AUTHOR]

Objective: To explore the prognostic value of systemic inflammatory biomarkers (albumin/globulin ratio [AGR], neutrophil/lymphocyte ratio [NLR], and platelet/lymphocyte ratio [PLR]) in patients with oral squamous cell carcinoma (OSCC), and further develop a novel prognostic score (AGR‐NLR). Methods: A large‐scale prospective study enrolling 792 eligible patients from December 2002 to June 2018 was carried out at the First Affiliated Hospital of Fujian Medical University. Three multivariate Cox regression models were performed to assess the association of overall survival (OS) with systemic inflammatory biomarkers, quantified by Akaike information criterion (AIC). Then, a novel AGR‐NLR score was established and incorporated into a prognostic nomogram. Results: In the univariate analysis, the increased AGR was associated with a reduced risk of death. Conversely, the higher NLR and PLR, the worse the OS. In the multivariate Cox regression models, AGR and NLR were stably independent prognostic indicators in all models, with Model 2 showing a lowest AIC (AGR: HR = 0.56, 95%CI: 0.41–0.78; NLR: HR = 1.80, 95%CI: 1.07–3.04). Then, a novel AGR‐NLR score was established, which showed a more excellent performance than either AGR or NLR alone (area under curve [AUC]: 0.589, 0.559, and 0.556, respectively). The C‐index of the nomogram based on AGR‐NLR was superior to that of traditional TNM staging system (C‐index: 0.658 versus. 0.596, p <.001). Similar results were also showed by decision curve analysis, indicating the nomogram had more positive net benefit compared to TNM staging system. Conclusion: The novel AGR‐NLR score is strongly associated with outcome in patients with OSCC and could be serve as a useful tool to accurately predict the OS of OSCC patients. [ABSTRACT FROM AUTHOR]
