4. LBL: Logarithmic Barrier Loss Function for One-class Classification

Abstract

One-class classification (OCC) aims to train a classifier only with the target class data and attracts great attention for its strong applicability in real-world application. Despite a lot of advances have been made in OCC, it still lacks the effective OCC loss functions for deep learning. In this paper, a novel logarithmic barrier function based OCC loss (LBL) that assigns large gradients to the margin samples and thus derives more compact hypersphere, is first proposed by approximating the OCC objective smoothly. But the optimization of LBL may be instability especially when samples lie on the boundary leading to the infinity loss. To address this issue, then, a unilateral relaxation Sigmoid function is introduced into LBL and a novel OCC loss named LBLSig is proposed. The LBLSig can be seen as the fusion of the mean square error (MSE) and the cross entropy (CE) and the optimization of LBLSig is smoother owing to the unilateral relaxation Sigmoid function. The effectiveness of the proposed LBL and LBLSig is experimentally demonstrated in comparisons with several state-of-the-art OCC algorithms on different network structures. 29. MRE11A:a novel negative regulator of human DNA mismatch repair

Abstract

DNA mismatch repair (MMR) is a highly conserved pathway that corrects DNA replication errors. Although well characterized, MMR factors remain to be identified. As a 3’-5’ exonuclease and endonuclease, meiotic recombination 11 homolog A (MRE11A) is implicated in multiple DNA repair pathways. However, the role of MRE11A in MMR is unclear. Here, we show that MRE11A deficiency increased the sensitivity of HeLa cells to N-methyl-N’ nitro-N nitrosoguanidine (MNNG) treatment, implying a potential role of MRE11 in MMR. Moreover, we found MRE11A was largely recruited to chromatin and negatively regulated the DNA damage signals within the first cell cycle after MNNG treatment. We also showed that knockdown of MRE11A increased, while overexpressing MRE11A decreased, MMR activity in HeLa cells, suggesting that MRE11A negatively regulates MMR activity. Furthermore, we show that the recruitment of MRE11A to chromatin requires MLH1 and that MRE11A competes with PMS2 for binding to MLH1. This decreases PMS2 levels in whole cell and on chromatin, and consequently comprises MMR activity. Collectively, our findings reveal that MRE11A is a negative regulator of human MMR.

Abstract

We focus on the problems of the accurate time delay estimation, the design of training pilots, and hybrid matrix optimization within the large-scale antenna array Terahertz (THz) broadband communication system. In contrast to the existing researches based on narrow-band arrays, we hereby shed light on the time delay estimation of broadband arrays. In THz broadband communication systems, the data symbol duration is relatively short when comparing with the dimension of the antenna array. In large-scale antenna systems, signals received in each antenna are no longer different phase-shifted copies of the same symbol, but completely different symbols in which occasion traditional narrow-band structure is no longer suitable. Based on the above conclusion, firstly, we put forward a system model based on large-scale antenna arrays and Time delay line (TDL) structure. Secondly, we deduce the Cramer-Rao lower bound (CRLB) of the time delay estimation, and present a time delay estimation algorithm that could reach the CRLB. Thirdly, by minimizing the CRLB, we address the design of the training pilot and optimized TDL structure under the condition of constant envelope training pilot and modulus TDL structure. Finally, we disclose the numerical simulation results. According to the simulation results, the aforementioned method is workable in reaching the CRLB, the TDL structure can significantly surpass that of the traditional model, and the optimal pilot design method outperforms the pseudo-random pilot structure.

Abstract

DNA mismatch repair (MMR) is a highly conserved pathway that corrects both base–base mispairs and insertion-deletion loops (IDLs) generated during DNA replication. Defects in MMR have been linked to carcinogenesis and drug resistance. However, the regulation of MMR is poorly understood. Interestingly, CNOT6 is one of four deadenylase subunits in the conserved CCR4-NOT complex and it targets poly(A) tails of mRNAs for degradation. CNOT6 is overexpressed in acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML) and androgen-independent prostate cancer cells, which suggests that an altered expression of CNOT6 may play a role in tumorigenesis. Here, we report that a depletion of CNOT6 sensitizes human U2OS cells to N-methyl-N′nitro-N-nitrosoguanidine (MNNG) and leads to enhanced apoptosis. We also demonstrate that the depletion of CNOT6 upregulates MMR and decreases the mutation frequency in MMR-proficient cells. Furthermore, the depletion of CNOT6 increases the stability of mRNA transcripts from MMR genes, leading to the increased expression of MMR proteins. Our work provides insight into a novel CNOT6-dependent mechanism for regulating MMR.

Abstract

Titanium dioxide (TiO2), a conventional n-type semiconductor, was widely used in photocatalysis, electrocatalysis and photoelectrocatalysis due to its good UV absorption and stable physical and chemical properties. However, its wide band gap and low oxygen reaction (OER) activity limited its application in photoelectrochemical (PEC) water splitting. In this work, we successfully constructed type-II Sb2(S,Se)3@TiO2core–shell heterojunctions. Antimony sulfide selenide (Sb2(S,Se)3) was a quasi-one-dimensional light-absorbing material with an adjustable band gap (1.1–1.8 eV), which broadened the TiO2light absorption range and effectively promoted the photogenerated carrier separation, transportation and utilization. Of particular note, novel Sb2(S,Se)3nanospheres (NSPs) (ca.69 nm) were in situgrown inside the tubes attributed to the unique space-confinement effect of TiO2nanotubes (NTs). The IPCE value for Sb2(S,Se)3@TiO2at 734 nm was 10.808% compared to 0.030% for TiO2. The separation efficiency and injection efficiency increased from 2.48% and 31.62% to 4.90% and 36.48%, respectively. The onset potential was moved negatively by 60 mV, and the maximum photocurrent density of 1.53 mA cm−2at 1.23 V vs.RHE was 13.9 times higher than that of TiO2(0.11 mA cm−2). This work provided a new idea for the application of TiO2in the field of PEC water splitting.
