The Wnt/β-catenin pathway contains multiple high-confidence risk genes that are linked to neurodevelopmental disorders, including autism spectrum disorder. However, its ubiquitous roles across brain cell types and developmental stages have made it challenging to define its impact on neural circuit development and behavior. Here, we show that TCF7L2, which is a key transcriptional effector of the Wnt/β-catenin pathway, plays a cell-autonomous role in postnatal astrocyte maturation and impacts adult social behavior. TCF7L2 was the dominant Wnt effector that was expressed in both mouse and human astrocytes, with a peak during astrocyte maturation. The conditional knockout of Tcf7l2 in postnatal astrocytes led to an enlargement of astrocytes with defective tiling and gap junction coupling. These mice also exhibited an increase in the number of cortical excitatory and inhibitory synapses and a marked increase in social interaction by adulthood. These data reveal an astrocytic role for developmental Wnt/β-catenin signaling in restricting excitatory synapse numbers and regulating adult social behavior.

The Atlantic sturgeon (Acipenser oxyrinchus) faces unique conservation challenges due to its extended maturation period and irregular spawning patterns, which restrict its reproductive capacity. Therefore, in order to overcome these difficulties we have made a first attempt to develop effective cryopreservation procedure for Atlantic sturgeon through evaluation of the effects of the glucose concentration in extender and final sperm concentration in straw on sperm motility parameters. A remarkably high (81 ± 8%) postthaw sperm motility was obtained. The other sperm velocity and trajectory parameters were differentially affected both by concentrations of glucose or sperm and cryopreservation process. Our results undoubtedly show that the post-thaw sperm motility of Atlantic sturgeon is highly influenced by the final glucose and sperm concentration. The recommended cryopreservation procedure includes 0.05 M glucose and 7.5% methanol at 1.0×109 spermatozoa ml−1. We have confirmed that our elaborated protocol can be used for cryopreservation of Atlantic sturgeon semen which can be subsequently used for long-term conservation of this endangered species, offering a valuable tool in efforts to safeguard its genetic diversity and contribute to its survival.

Polybrominated diphenyl ethers (PBDEs) have been used for many years as flame retardants. Due to their physicochemical and toxicological properties, they are considered to be persistent organic pollutants (POPs). BDE-209 is the main component of deca-BDE, the one PBDE commercial mixture currently approved for use in the European Union. The aim of this study was to analyse BDE-209 in surface soil samples from Warsaw and surrounding areas (Poland) as an indicator of environmental pollution with PBDEs, and to characterise the associated health risk. A total of 40 samples were analysed using gas chromatography with electron capture detection (GC-µECD). Concentrations of BDE-209 in soil ranged from 0.4 ng g−1 d.w. (limit of quantification) to 158 ng g−1 d.w. Overall, 52.5% of results were above the method’s limit of quantification. The highest levels were found at several locations with heavy traffic and in the vicinity of a CHP plant in the city. The lowest concentrations were observed in most of the samples collected from low industrialized or green areas (−1 d.w.). Exposure to BDE-209 was estimated for one of the most sensitive populations, i.e., young children. The following exposure routes were selected: oral and dermal. No risk was found to young children’s health.

The results of comparative studies on the fabrication and characterization of GaN/Ag substrates using pulsed laser deposition (PLD) and magnetron sputtering (MS) and their evaluation as potential substrates for surface-enhanced Raman spectroscopy (SERS) are reported. Ag layers of comparable thicknesses were deposited using PLD and MS on nanostructured GaN platforms. All fabricated SERS substrates were examined regarding their optical properties using UV–vis spectroscopy and regarding their morphology using scanning electron microscopy. SERS properties of the fabricated GaN/Ag substrates were evaluated by measuring SERS spectra of 4-mercaptobenzoic acid molecules adsorbed on them. For all PLD-made GaN/Ag substrates, the estimated enhancement factors were higher than for MS-made substrates with a comparable thickness of the Ag layer. In the best case, the PLD-made GaN/Ag substrate exhibited an approximately 4.4 times higher enhancement factor than the best MS-made substrate.

Theoretical background: The global financial crisis (GFC) has shown the importance of the funding model for the bank’s stability. In this context, deposits were of particular importance as they proved to be a stable source of funding during market turmoil. As a result, many banks have changed the funding model, paying greater attention to financing obtained on the deposits market. Purpose of the article: In this paper, we analyze the impact of funding models on the EU banks’ risk after GFC, i.e. in 2011–2018. We put particular emphasis on the funding structure measured by the deposits to total assets ratio and changes that take place according to the type of institution (i.e. listing status, specialization, and funding model). Research methods: In our research, we use panel data models together with a set of tests that allow us to deduce about properties of proposed models and allow us to analyze the significance of the impact of the bank-specific, macroeconomic, and dummy variables on the bank’s risk. We apply “within”, “fixed time effects” estimator from plm R package. Main findings: We confirm the stabilizing function of deposits, but also the non-linear nature of the impact of the funding structure on the bank’s stability, depending on the bank’s specialization. This means that the stabilizing role of deposits for the bank’s stability is just as important in the post-crisis period as it was during the outbreak of GFC in 2008, although the excessive growth of deposits in some types of banks may, however, lead to an increase in the risk level.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disease caused by a mutation in the HTT gene. To generate human-induced pluripotent stem cells (hiPSCs), we used dermal fibroblasts from 1 healthy adult control (K-Pic2), 1 HD manifest patient (M-T2), 1 healthy juvenile control (jK-N1), and 1 juvenile HD patient (jHD-V1). HD stage of patients was assessed by neurological tests and donors were without comorbidities and were non-smokers. Characterization showed that the obtained hiPSCs have the same number of CAG repeats as the parental fibroblast lines, express pluripotency markers and have the ability to differentiate into all 3 germ layers.

ABSTRACT: The tissue-specific profile of alternatively spliced genes (ASGs) and their involvement in reproduction processes characteristic of turkey testis, epididymis, and ductus deferens were investigated for the first time in birds. Deep sequencing of male turkey reproductive tissue RNA samples (n = 6) was performed using Illumina RNA-Seq with 2 independent methods, rMATs and SUPPA2, for differential alternative splicing (DAS) event prediction. The expression of selected ASGs was validated using quantitative real-time reverse transcriptase-polymerase chain reaction. The testis was found to be the site of the highest number of posttranscriptional splicing events within the reproductive tract, and skipping exons were the most frequently occurring class of alternative splicing (AS) among the reproductive tract. Statistical analysis revealed 86, 229, and 6 DAS events in the testis/epididymis, testis/ductus deferens, and epididymis/ductus deferens comparison, respectively. Alternative splicing was found to be a mechanism of gene expression regulation within the turkey reproduction tract. In testis, modification was observed for spermatogenesis specific genes; the changes in 5’ UTR could act as regulator of MEIG1 expression (a player during spermatocytes meiosis), and modification of 3’ UTR led to diversification of CREM mRNA (modulator of gene expression related to the structuring of mature spermatozoa). Sperm tail formation can be regulated by changes in the 5’ UTR of testicular SLC9A3R1 and gene silencing by producing dysfunctional variants of ODF2 in the testis and ATP1B3 in the epididymis. Predicted differentially ASGs in the turkey reproductive tract seem to be involved in the regulation of spermatogenesis, including acrosome formation and sperm tail formation and binding of sperm to the zona pellucida. Several ASGs were classified as cilia by actin and microtubule cytoskeleton organization. Such genes may play a role in the organization of sperm flagellum and post-testicular motility development. To our knowledge, this is the first functional investigation of alternatively spliced genes associated with tissue-specific processes in the turkey reproductive tract.

Furan and its derivatives are found in various heat-treated foods. Furan is classified as a possible human carcinogen. The European Union authorities recommend collecting data on the occurrence of these compounds, estimating consumer exposure, and taking measures to protect human health based on a scientific risk assessment. The aim of this study was to estimate the exposure of infants and toddlers to furan and its methyl derivatives—2-methylfuran, 3-methylfuran, and ∑2,5-dimethylfuran/2-ethylfuran—present in home-prepared foods and to characterize the associated health risks. The compounds of interest were determined using the HS-GC/MS. The risk was characterized by the calculation of the margin of exposure (MoE). Huntington’s disease (HD) is a progressive neurodegenerative disorder with autosomal-dominant heritability that affect the central nervous system and peripheral tissues. The human-induced pluripotent stem cells (hiPSC) lines were generated from dermal fibroblasts of patients without comorbidities, non-smokers, at the pre-manifest (IIMCBi004-A), early-manifest (IIMCBi005-A), and manifest (IIMCBi006-A) HD stage assessed by neurological tests, as well as from a healthy donor (IIMCBi003-A). Characterization showed that the obtained hiPSC lines contained different CAG repeats consistent with the number of CAG repeats in original fibroblasts. Moreover, hiPSCs expressed pluripotency markers and were able to differentiate into three-germ layers in vitro.

Recently, the photovoltaic technology has become very popular as a means to produce renewable energy. One of the problems that are still unsolved in this area of the industry is that photovoltaic panels are subject to a significant loss of efficiency due to the accumulation of dust and dirt. In addition, during the winter season, the accumulation of snow and ice also reduces or stops the energy production. The current methods of dealing with this problem are inefficient and pollute the environment. One way with high potential to prevent the build-up of dirt and ice is to use transparent coatings with self-cleaning and icephobic properties. In this work, the chemical modification of an epoxy–silicone hybrid resin using dually functionalized polysiloxanes was carried out. The icephobic properties (ice adhesion and freezing delay time of water droplets), hydrophobic properties (water contact angle, contact angle hysteresis, and roll-off angle), average surface roughness, and optical properties were characterized. It can be concluded that the performed chemical modification resulted in a significant improvement of the icephobic properties of the investigated coatings: ice adhesion decreased by 69%, and the freezing delay time increased by 17 times compared to those of the unmodified sample. The polysiloxanes also caused a significant reduction in the contact angle hysteresis and roll-off angle. The chemical modifications did not negatively affect the optical properties of the coatings, which is a key requirement for photovoltaic applications.

Abstract Background Mammalian/mechanistic target of rapamycin (mTOR) complexes are essential for cell proliferation, growth, differentiation, and survival. mTORC1 hyperactivation occurs in the tuberous sclerosis complex (TSC). mTORC1 localizes to the surface of lysosomes, where Rheb activates it. However, mTOR was also found on the endoplasmic reticulum (ER) and Golgi apparatus (GA). Recent studies showed that the same inputs regulate ER-to-GA cargo transport and mTORC1 (e.g., the level of amino acids or energy status of the cell). Nonetheless, it remains unknown whether mTOR contributes to the regulation of cargo passage through the secretory pathway. Methods The retention using selective hooks (RUSH) approach was used to image movement of model cargo (VSVg) between the ER and GA in various cell lines in which mTOR complexes were inhibited. We also investigated VSVg trafficking in TSC patient fibroblasts. Results We found that mTOR inhibition led to the overall enhancement of VSVg transport through the secretory pathway in PC12 cells and primary human fibroblasts. Also, in TSC1-deficient cells, VSVg transport was enhanced. Conclusions Altogether, these data indicate the involvement of mTOR in the regulation of ER-to-GA cargo transport and suggest that impairments in exocytosis may be an additional cellular process that is disturbed in TSC.

The endocytic adaptor protein 2 (AP-2) complex binds dynactin as part of its noncanonical function, which is necessary for dynein-driven autophagosome transport along microtubules in neuronal axons. The absence of this AP-2-dependent transport causes neuronal morphology simplification and neurodegeneration. The mechanisms that lead to formation of the AP-2-dynactin complex have not been studied to date. However, the inhibition of mammalian/mechanistic target of rapamycin complex 1 (mTORC1) enhances the transport of newly formed autophagosomes by influencing the biogenesis and protein interactions of Rab-interacting lysosomal protein (RILP), another dynein cargo adaptor. We tested effects of mTORC1 inhibition on interactions between the AP-2 and dynactin complexes, with a focus on their two essential subunits, AP-2β and p150Glued. We found that the mTORC1 inhibitor rapamycin enhanced p150Glued–AP-2β complex formation in both neurons and non-neuronal cells. Additional analysis revealed that the p150Glued–AP-2β interaction was indirect and required integrity of the dynactin complex. In non-neuronal cells rapamycin-driven enhancement of the p150Glued–AP-2β interaction also required the presence of cytoplasmic linker protein 170 (CLIP-170), the activation of autophagy, and an undisturbed endolysosomal system. The rapamycin-dependent p150Glued–AP-2β interaction occurred on lysosomal-associated membrane protein 1 (Lamp-1)-positive organelles but without the need for autolysosome formation. Rapamycin treatment also increased the acidification and number of acidic organelles and increased speed of the long-distance retrograde movement of Lamp-1-positive organelles. Altogether, our results indicate that autophagy regulates the p150Glued–AP-2β interaction, possibly to coordinate sufficient motor-adaptor complex availability for effective lysosome transport. Inorganic hollow spheres find a growing number of applications in many fields, including catalysis and solar cells. Hence, a simple fabrication method with a low number of simple steps is desired, which would allow for good control over the structural features and physicochemical properties of titania hollow spheres modified with noble metal nanoparticles. A simple method employing sol–gel coating of nanoparticles with titania followed by controlled silver diffusion was developed and applied for the synthesis of Ag-modified hollow TiO2 spheres. The morphology of the synthesized structures and their chemical composition was investigated using SEM and X-ray photoelectron spectroscopy, respectively. The optical properties of the synthesized structures were characterized using UV–vis spectroscopy. Ag–TiO2 hollow nanostructures with different optical properties were prepared simply by a change of the annealing time in the last fabrication step. The synthesized nanostructures exhibit a broadband optical absorption in the UV–vis range.

BackgroundWe have previously shown that transthyretin (TTR) amyloidosis patients have amyloid fibrils of either of two compositions; type A fibrils consisting of large amounts of C-terminal TTR fragments in addition to full-length TTR, or type B fibrils consisting of only full-length TTR. Since type A fibrils are associated with an older age in ATTRVal30Met (p.Val50Met) amyloidosis patients, it has been discussed if the TTR fragments are derived from degradation of the amyloid deposits as the patients are aging. The present study aimed to investigate if the fibril composition type changes over time, especially if type B fibrils can shift to type A fibrils as the disease progresses.Material and methodsAbdominal adipose tissue biopsies from 29 Swedish ATTRVal30Met amyloidosis patients were investigated. The fibril type in the patients´ initial biopsy taken for diagnostic purposes was compared to a biopsy taken several years later (ranging between 2 and 13 years). The fibril composition type was determined by western blot.ResultsAll 29 patients had the same fibril composition type in both the initial and the follow-up biopsy (8 type A and 21 type B). Even patients with a disease duration of more than 12 years and an age over 75 years at the time of the follow-up biopsy had type B fibrils in both biopsies.DiscussionThe result clearly shows that the amyloid fibril composition containing large amounts of C-terminal fragments (fibril type A) is a consequence of other factors than a slow degradation process occurring over time.

Honey is considered to be a health-promoting food product. Therefore, it is assumed that it should be free of contaminants. Although the use of organochlorine pesticides (OCPs) was banned a few decades ago in developed countries, persistent organic pollutants (POPs) are still detected in various environmental and biological matrices, including food. These contaminants exhibit toxic properties and bioaccumulate in some food chains. The validation of a modified QuEChERS extraction method was successfully performed for o,p’-DDT, o,p’-DDE, o,p’-DDD, p,p’-DDT, p,p’-DDE, p,p’-DDD, heptachlor and dieldrin. 2,2′,4,4′,5,5′-hexachlorobiphenyl (PCB 153) was used as an internal standard. The modification involved changing the solvent from acetonitrile to n-hexane after extraction. Quantitation was carried out using gas chromatography with an electron capture detector (µECD). The mean recovery values for o,p’-DDT, o,p’-DDE, o,p’-DDD, p,p’-DDT, p,p’-DDE, p,p’-DDD and dieldrin, spiked at 2.9 ng/g and 20 ng/g, ranged from 64.7% to 129.3%, and, for heptachlor spiked at 5.6 ng/g and 20 ng/g, ranged from 68.0% to 88.3%. The relative standard deviation (RSD) for these concentrations did not exceed 20%, and the within-laboratory reproducibility was below 20%, except o,p’-DDE and p,p’-DDT, which were 25.2% and 20.7%, respectively. This modified QuEChERS extraction method for selected organochlorine compounds was demonstrated as effective for routine testing in honey.

W artykule omówiono różne koncepcje na temat charakteru prawnego usiłowania postrzeganego tradycyjnie jako forma stadialna przestępstwa. Stosując metodę analizy przepisów prawnokarnych opartą na systemowym badaniu struktur normatywnych zakodowanych w tych przepisach, niektórzy autorzy dochodzą do odmiennych wniosków i przyjmują, że usiłowanie jest odrębnym typem czynu zabronionego. W artykule podjęto próbę takiego sposobu interpretacji, który uwzględniając dotychczasowy dorobek nauki w zakresie analiz normatywnych przepisów karnych, nie kwestionuje jednocześnie charakteru usiłowania jako nieposiadającej bytu samoistnego postaci stadialnej przestępstwa. Osobnym problemem, związanym ze sposobem odczytania charakteru normatywnego usiłowania, jest kwestia stosowania przepisów o usiłowaniu do przestępstw będących delictum sui generis usiłowania. Analiza normatywna usiłowania ukształtowanego w kodeksie karnym jako forma stadialna przestępstwa prowadzi do wniosku, że u podstaw tej konstrukcji prawnej leży naruszenie tej samej normy sankcjonowanej, którą odczytujemy z przepisu typizującego dokonanie.

The aim of this study was to assess the potential implication of microRNA on tuberous sclerosis (TSC) pathogenesis by performing microRNA profiling on cell lines silencing TSC1 or TSC2 genes using qPCR panels, before and after incubation with rapamycin. Significant differences in expression were observed between samples before and after rapamycin treatment in nineteen miRNAs in TSC1, five miRNAs in TSC2 and seven miRNAs in controls. Of miRNAs dysregulated before rapamycin treatment, three normalized after treatment in the TSC1 group (miR-21-3p, miR-433-3p, let-7g-3p) and one normalized in the TSC2 group (miR-1224-3p). Of the miRNAs dysregulated before rapamycin treatment in the TSC1 and TSC2 groups, two did not normalize after treatment (miR-33a-3p, miR-29a-3p). The results of the possible targets indicated that there are four common genes with seed regions susceptible to regulation by those miRNAs: ZBTB20, PHACTR2, PLXNC1 and ATP1B4. Our data show no changes in mRNA expression of these targets after rapamycin treatment. In conclusion, results of our study indicate the involvement of miRNA dysregulation in the pathogenesis of TSC. Some of the miRNA might be used as markers of treatment efficacy and autonomic miRNA as a target for future therapy.

Introduction: A disturbed calcium-phosphate balance is an important issue for kidney stone formation in nephrolithiasis. Hypercalciuria (HC) has been proposed as an essential etiology of monosymptomatic nocturnal enuresis (MNE). Objectives: We may suspect that patients with MNE may be at risk of stone formation hence the objective of this paper was to assess the risk in MNE children using Bonn Risk Index (BRI). Patients and Methods: The urinary work-up of 204 children (83 with MNE and 121 controls) included urinary calcium (Ca), magnesium (Mg) and sodium (Na) excretion, Ca/creatinine ratio, BRI, ionized calcium (Ca2+), Mg/creatinine and Ca/citrate ratios, urinary citrates and oxalates (Ox). Results: Ca/creatinine and Mg/creat ratios were higher in the MNE group. There were no differences in Mg and Ca amount in urine and Mg/Ca ratio between MNE and the reference group. Both groups differed in Mg and Ca excretion per kg of body mass. MNE children differed from controls regarding BRI, Ox and urinary Ca2+. No differences in urinary citrate excretion nor Ca/citrate ratio between MNE and the controls were found. Correlations between factors important in the crystallization process in MNE children were recorded. Conclusion: MNE patients may be at risk of oxalate nephrolithiasis. Further studies to assess the role of the BRI and Ca/citrate ratio in predicting stone formation in MNE children are needed.
