Abstract The mechanisms underlying myopia pathogenesis are not well understood. Using publicly-available human and animal datasets, we expound on the roles of known, implicated proteins, and new myopia-related signaling pathways were hypothesized. Proteins identified from human serum or ocular fluids, and from ocular tissues in myopic animal models, were uploaded and analyzed with the QIAGEN Ingenuity Pathway Analysis (IPA) software (March 2023). With each IPA database update, more potentially-relevant proteins and signaling pathways previously unavailable during data acquisition are added, allowing extraction of novel conclusions from existing data. Canonical pathway analysis was used to analyze these data and calculate an IPA activation z-score—which indicates not only whether an association is significant, but also whether the pathway is likely activated or inhibited. Cellular immune response and cytokine signaling were frequently found to be affected in both human and animal myopia studies. Analysis of two publicly-available proteomic datasets highlighted a potential role of the innate immune system and inflammation in myopia development, detailing specific signaling pathways involved such as Granzyme A (GzmA) and S100 family signaling in the retina, and activation of myofibroblast trans-differentiation in the sclera. This perspective in myopia research may facilitate development of more effective and targeted therapeutic agents.

The proton exchange membrane electrolysis cell (PEMEC) has attracted considerable attention for large-scale and efficient hydrogen production because of its high current density, high hydrogen purity and fast dynamic response. Flow field distributions and water and thermal management characteristics of a PEMEC are vital for electrolytic cell structure and the determination of operating condition. A three-dimensional, non-isothermal, electrochemical model of a PEMEC was established in this manuscript. The flow field distribution and water and thermal management of the PEMEC are discussed. The corresponding results showed that the pressure of the flow channel decreased diagonally from the inlet to the outlet, and the pressure and velocity distribution exhibited a downward opening shape of a parabola. At the same inlet flow rate, when the voltage was 1.6 V, the oxygen generation rate was 15.74 mol/(cm2·s), and when the voltage was 2.2 V, the oxygen generation rate was 332.05 mol/(cm2·s); due to the change in the oxygen production rate, the pressure difference at 2.2 V was 2.5 times than that at 1.6 V. When the stoichiometric number was less than two, the average temperature of the catalyst layer (CL) decreased rapidly with the increase in the water flow rate. When the voltage decreased to 2.1 V, the current density came to the highest value when the stoichiometric number was 0.7, then the current density decreased with an increase in the stoichiometric number. When stoichiometric numbers were higher than five, the surface temperature and current density remained basically stable with the increase in the water flow rate, and the water and thermal management and electrolysis characteristics performed better. The research results could optimize the water supply of electrolysis cells. According to the velocity distribution law of the flow field, the water and thermal management performance of the PEMEC could be estimated, further promoting safety and reliability.

Abstract Background Inhibition of hepatic lipogenesis is widely regarded as an effective treatment for metabolic-associated fatty liver disease (MAFLD), although numerous related drugs have failed to reach clinical application. The goal of this study is to identify a novel small compound that can effectively treat MAFLD. Methods Primary hepatocytes were first exposed to palmitic acid and oleic acid, then treated with compounds prior to high through screening for cellular lipid content. The efficacy of these compounds was measured by Nile Red staining and triglyceride analysis. The potential cellular toxicity caused by these compounds was evaluated by CCK8 assay. qPCR and Western blot were used to determine expression of RNAs and proteins, respectively. The compound was intraperitoneally injected into diet-induced obese (DIO) mice to examine its efficacy in vivo. Results We identified the dimethyl 1-methyl-2-thioxoindoline-3,3-dicarboxylate (TOIDC) as a powerful chemical to reduce cellular lipid with minimal cellular toxicity. When injected intraperitoneally, TOIDC effectively ameliorates MAFLD in DIO mice. Mechanically, TOIDC suppresses de novo lipogenesis through inhibiting sterol regulatory element-binding protein 1 (SREBP1). Conclusions Our findings indicate that TOIDC could be a promising lead compound to develop new drugs to treat MAFLD. Graphical Abstract

Electric vehicles (EVs) are experiencing explosive developments due to their advantages in energy conservation and environmental protection. As a pivotal component of EVs, the safety performance of lithium-ion batteries directly affects driving miles and even safety; hence, a battery thermal management system (BTMS) is especially important. To improve the thermal safety performance of power battery modules, first, a new leak-proof phase change material (PCM)-coupled liquid-cooled composite BTMS for large-scale battery modules is proposed in this research. Second, the numerical simulation analysis method was utilized to analyze the influences of the fluid flow channel shape, working fluid inlet temperature, inlet velocity, and reverse flow conditions on the BTMS. Eventually, the abovementioned performances were compared with the traditional PCM-coupled liquid-cooling strategy. The relative data indicated that the Tmax was reduced by 17.5% and the ΔTmax was decreased by 19.5% compared to the liquid-cooling approach. Further, compared with conventionally designed PCM composite liquid cooling, the ΔTmax was reduced by 34.9%. The corresponding data showed that, when using the e-type flow channel, reverse flow II, the inlet flow velocity was 0.001–0.005 m/s, and the inlet temperature was the ambient temperature of the working condition. The thermal performance of the anti-leakage system with a thermal conduction slot PCM-coupled liquid-cooling composite BTMS reached optimal thermal performance. The outcome proved the superiority of the proposed BTMS regarding temperature control and temperature equalization capabilities. It also further reduced the demand for liquid-cooling components, avoided the problem of the easy leakage of the PCM, and decreased energy consumption.

Abstract Although obstructive sleep apnea (OSA) has been clinically reported to be associated with acute coronary syndrome (ACS), the pathogenesis between the two is unclear. Herein, we analyzed and screened out the prospective molecular marker. To explore the candidate genes, as well as signaling cascades involved in ACS related to OSA, we extracted the integrated differentially expressed genes (DEGs) from the intersection of genes from the Gene Expression Omnibus (GEO) cohorts and text mining, followed by enrichment of the matching cell signal cascade through DAVID analysis. Abstract Background The purpose of this study was to assess the antioxidative activity of selenium-enriched Chrysomyia Megacephala (Fabricius) (C. megacephala) larvae powder (SCML) and its impact on the diversity and structure of intestinal microflora in a mouse model of D-galactose (D-gal)-induced oxidative damage. Methods Sixty male ICR mice were equally randomized to a normal control (NC) group, a model group, a positive group, a low-dose SCML (L-SCML) group, a mid-dose SCML (M-SCML) group, and a high-dose SCML (H-SCML) group. Animals in NC and model groups received water, animals in the positive group received 40 mg/Kg vitamin E (VE), and those in the three SCML groups received SCML which include 300, 1000 and 3000 μg/Kg selenium (Se) respectively. An oxidative damage model induced by subcutaneous injection of D-gal for 6 weeks via the neck was established. Serum oxidative stress levels and tissue appearance were evaluated. Tissues oxidative stress levels were detected by commercially available kit. Nuclear erythroid 2-related factor (Nrf2) and gut microbiota were determined by western blot and high throughput sequencing 16S rRNA gene respectively. Results An oxidative damage model was established successfully as represented by a significant elevation of malondialdehyde (MDA) and protein carbonylation, and inhibition of the antioxidants including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC) and glutathione (GSH). It was found that oxidative damage and histological alterations were attenuated, the expression of Kelch-like ECH-associated protein (Keap1) was decreased, and the expression of Nrf2 and hemeoxygenase-1 (HO-1) was increased after SCML treatment. In addition, significant changes were observed in the gut microbiota, including Proteobacteria and the ratio of Bacteroidetes to Firmicutes at the phylum level, as well as Helicobacter, Clostridium and Lactobacillus at the genus level. Conclusion SCML exerted an antioxidative effect in vivo, probably by increasing the antioxidant activity and reducing the production of oxidation products via the Nrf2 signaling pathway. SCML could also redress the intestinal flora imbalance induced by oxidative stress. All these findings suggest that SCML could serve as a functional food and natural drug additive to protect the human body against oxidative damage.

Myopia is a globally emerging concern accompanied by multiple medical and socio-economic burdens with no well-established causal treatment to control thus far. The study of the genomics and transcriptomics of myopia treatment is crucial to delineate disease pathways and provide valuable insights for the design of precise and effective therapeutics. A strong understanding of altered biochemical pathways and underlying pathogenesis leading to myopia may facilitate early diagnosis and treatment of myopia, ultimately leading to the development of more effective preventive and therapeutic measures. In this review, we summarize current data about the genomics and transcriptomics of myopia in human and animal models. We also discuss the potential applicability of these findings to precision medicine for myopia treatment.

Lung cancer is a fatal tumor threatening human health. It is of great significance to explore a diagnostic method with wide application range, high specificity, and high sensitivity for the detection of lung cancer. In this study, data fusion and wavelet transform were used in combination with Fourier transform infrared (FTIR) spectroscopy and Raman spectroscopy to study the serum samples of patients with lung cancer and healthy people. The Raman spectra of serum samples can provide more biological information than the FTIR spectra of serum samples. After selecting the optimal wavelet parameters for wavelet threshold denoising (WTD) of spectral data, the partial least squares–discriminant analysis (PLS-DA) model showed 93.41% accuracy, 96.08% specificity, and 90% sensitivity for the fusion data processed by WTD in the prediction set. The results showed that the combination of FTIR spectroscopy and Raman spectroscopy based on data fusion and wavelet transform can effectively diagnose patients with lung cancer, and it is expected to be applied to clinical screening and diagnosis in the future.

Segmented filamentous bacteria (SFB) are commensal organisms that grow by anchoring a specialized holdfast structure to the intestinal walls of a variety of animals. Interaction of SFB with Peyer’s patches in mice promotes the post-natal maturation of the immune system. We previously reported that the colonization of SFB in humans mainly occurs by 36 months of age, and is difficult to be detected afterward. In this study, we measured the level of SFB in intestinal fluids of human children. SFB were found via qPCR to represent a small fraction of the whole SFB-positive microbiota (105 SFB in 1011 total bacteria). Bacteria with filamentous segmented morphology were observed in intestinal fluids via fluorescent in situ hybridization, and from gut biopsies via scanning electron microscopy. SFB-specific DNA and peptide fragments were also identified via multiple displacement amplification PCR and mass spectrometry. There was an overall positive correlation between the presence of SFB and the titer of total secretory immunoglobulin A (sIgA), which is more apparent in intestinal fluids of the age group of 8–36 months. Afterward there was a decline of SFB in numbers correlated with a reduction of total sIgA. RT-qPCR analysis of the terminal ileal biopsies revealed that the expression of Th17 pathway genes were induced in SFB-positive samples, while the markers of T and B cell receptor signaling pathways were also upregulated. Collectively, these data suggest that SFB is a rare member of microbiota, and may play an important role in the development of human gut immunity.

To explore the topological properties of paddy arthropod food webs is of significance for understanding natural equilibrium of rice pests. In present study, we used Pajek software to analyze the topological properties of four full arthropod food webs in South China. The results showed that predators were significantly abundant than preys, and the proportion of predators to preys (3.07) was significantly higher than previously reported by Cohen in 1977 (1.33). In the food webs, the number of top species was the largest, accounted for about 50% of the total. The number of intermediate-intermediate links was far greater than the other three links. The average degree of paddy arthropod food webs is 6.0, 6.04, 5.74 and 7.75, respectively. Average degree and link density did not change significantly with the change of the number of species, but the connectance reduced significantly. In the paddy ecosystems, the increase of species diversity does not lead to an increase proportionally to the links among species. The link density and connectance of food webs of early season rice field were less than that from late season rice field. Cycles of all food webs cycles were 0. The maximum chain length of the basal species was 3, and the largest chain length of the top species was typically 2 or 3. Neutral insects were found to play a very important role in the paddy ecosystem. Nilaparvata lugens and Sogatella furcifera were found to be the dominant species of rice pests. Pardosa pseudoannulata, Tetragnatha maxillosa, Pirata subparaticus, Arctosa stigmosa and Clubiona corrugate were identified as the important predatory species that may effectively control the pest population. The keystone species calculated from keystone index and network analysis are analogous, indicating either keystone index or network analysis can be used in the analysis of keystone species.

The importance of a species is correlated with its topological properties in a food web. Studies of keystone species provide the valuable theory and evidence for conservation ecology, biodiversity, habitat management, as well as the dynamics and stability of the ecosystem. Comparing with biological experiments, network methods based on topological structure possess particular advantage in the identification of keystone species. In present study, we quantified the relative importance of species in Carpinteria Salt Marsh food web by analyzing five centrality indices. The results showed that there were large differences in rankings species in terms of different centrality indices. Moreover, the correlation analysis of those centralities was studied in order to enhance the identifying ability of keystone species. The results showed that the combination of degree centrality and closeness centrality could better identify keystone species, and the keystone species in the CSM food web were identified as, Stictodora hancocki, small cyathocotylid, Pygidiopsoides spindalis, Phocitremoides ovale and Parorchis acanthus.

In this study, we used Pajek to analyze the effect of parasitism on food webs collected from Carpinteria Salt Marsh (CSM). Results showed that median and mean of generality and vulnerability for predator-prey and parasite-host sub-webs were greater than the reported previously. Inclusion of parasites significantly increased the mean generality and vulnerability of the full food web. Effectiveness of cascade model was tested using CSM and arthropod food webs. The results demonstrated that fitting goodness on the predator-prey sub-web without parasites was lower than that on the full CSM food web. Also, cascade model performed worse in fitting arthropod food webs.

An efficient and general copper-catalyzed Goldberg reaction at 90–110 °C between aryl bromides and amides providing the desired products in good to excellent yields has been developed using N,N-dimethylglycine as the ligand. The reaction is tolerant toward a wide range of amides and a variety of functional group substituted aryl bromides. In addition, hindered, unreactive aromatic and aliphatic secondary acyclic amides, known to be poor nucleophiles, are efficiently coupled with aryl iodides through this simple and cheap copper/N,N-dimethylglycine catalytic system.

Hexadepsipeptide 2, the precursor of Hirsutellide A (1), was synthesized in an overall yield of 45% from N-Boc-Me-Gly via three coupling reactions using dicyclohexylcarbodiimide (DCC), O-(7-azabenzotriazol-1-yl)-N,N,N’,N’-tetramethyl- uronium hexafluorophosphate (HATU) and bis(2-oxo-3-oxazolidinyl)phosphinic chloride (BOP-Cl), respectively.

Significance: A data-based calibration method with enhanced depolarization contrast in polarization-sensitive optical coherence tomography (PS-OCT) was developed and demonstrated effective for detecting melanin content in the eye. Aim: We aim to mitigate the dependence between the measured depolarization metric and the intensity signal-to-noise ratio (SNR) for improved visualization of depolarizing tissues, especially in low SNR regions, and to demonstrate the enhanced depolarization contrast to evaluate melanin presence. Approach: A function for calibrating the depolarization metric was experimentally derived from the young albino guinea pig, assuming depolarization free in the retina. A longitudinal study of guinea pigs (9 weeks) was conducted to assess the accumulation of melanin during early eye growth. Furthermore, the melanin content of the sub-macular choroid was compared in eyes with light and dark irides involving 14 human subjects in early middle adulthood. Results: We observed an increase in the improved depolarization contrast, which indicates potential melanin accumulation in the early eye development with age in the pigmented guinea pig eyes. We found a significant difference in melanin content between human eyes with light and dark colors. Conclusions: Our proposed calibration method enhanced the visualization of depolarizing structures in PS-OCT, which can be generalized to all kinds of polarization-sensitive imaging and can potentially monitor melanin in healthy and pathological eyes.
