3. Dichloromethane Extract of Fermentation Broth by Co-Culture of Morchella esculenta and Coprinus comatus Induces Apoptosis in U251 Cells via Mitochondrial Intrinsic Pathway

Abstract

The current study was designed to investigate the inhibitory effects of dichloromethane extract of fermentation broth by co-culture of Morchella esculenta and Coprinus comatus (DCMM) on human glioma U251 cells in vitro and its possible underlying mechanisms. The proliferation of U251 cells was inhibited by DCMM with different concentrations by the CCK-8 assay. Besides, flow cytometry assay was used to evaluate the DCMM promoted U251 cell apoptosis rate in a dose-dependent manner. DCMM with different concentrations (10 μg·mL-1, 20 μg·mL-1, and 40 μg·mL-1) significantly enhanced the expression of caspases-3 activity after 24 h. In addition, DCMM with different concentrations significantly increased caspase-3 and Bax, and decreased Bcl-2 expressions at both mRNA and protein levels. DCMM can remarkably inhibit the proliferation and promote cell apoptosis of human glioma U251 cells. The possible underlying mechanisms could be related to induction of apoptosis of human glioma U251 cells by mitochondrial intrinsic pathway.

Abstract

Undoped and Eu3+-doped Sr2Mg1-xZnxSi2O7 (0≤x≤1) powder crystal samples were obtained via a conventional solid-state reaction method. The XRD result and FT-IR spectra demonstrated the complete solid-solution as well as lattice distortion between Sr2MgSi2O7 and Sr2ZnSi2O7. The UV emission spectra showed the main emission around 616nm, which implied the non-inversion symmetry around Eu3+ in Sr2Mg1-xZnxSi2O7:Eu3+ (0≤x≤1). The UV excitation spectra exhibited the Eu3+-O2- CT transitions centered at 250nm and 258nm in Sr2MgSi2O7:Eu3+ and Sr2ZnSi2O7:Eu3+, respectively. The emission of Sr2(Mg, Zn)Si2O7:Eu3+ under 147nm light excitation is very weak, which suggests that energy transfer from the host to the activator is not so efficient in Sr2Mg1-xZnxSi2O7:Eu3+ (0≤x≤1) system.

5. Biosorption of Heavy Metal Ions from Aqueous Solution on Chinese Fir Bark Modified by Sodium Hypochlorite

Abstract

The oxidation conditions and adsorption ability of Cu (II), Cd (II), and Pb (II) from an aqueous solution of sodium hypochlorite-oxidized fir bark powder were investigated. The optimum amount of NaClO was 9.6%, the pH was 9, and the oxidation time was 120 min. Batch adsorption experiments were carried out with various times, pH values, and initial metal ion concentrations. The adsorption isotherms and kinetics of adsorption were also studied. The maximum adsorption capacity of oxidized fir bark was 0.983, 1.223, and 0.966 mmol/g for Cu (II), Cd (II), and Pb (II), respectively, within 30 min at pH 5, higher adsorption capacity than that of unoxidized fir bark. The isothermal adsorption of heavy metal ions in aqueous solution was a good fit to the Langmuir equation, with a coefficient of determination, R2, above 0.99. The adsorption of fir bark on metal ions can be appropriately approximated by a pseudo-second order adsorption kinetics model. Results indicate that the fir bark powder modified with sodium hypochlorite can adsorb large amounts of metal ions and could be a good adsorbent for the removal of Cu(II), Cd(II), and Pb(II) from aqueous solution.
