Abstract The legume albumin-1 gene family, arising after nodulation, encodes linear a- and b-chain peptides for nutrient storage and defense. Intriguingly, in one prominent legume, Clitoria ternatea, the b-chains are replaced by domains producing ultra-stable cyclic peptides called cyclotides. The mechanism of this gene hijacking is until now unknown. Cyclotides require recruitment of ligase-type asparaginyl endopeptidases (AEPs) for maturation (cyclization), necessitating co-evolution of two gene families. Here we compare a chromosome-level C. ternatea genome with grain legumes to reveal an 8 to 40-fold expansion of the albumin-1 gene family, enabling the additional loci to undergo diversification. Iterative rounds of albumin-1 duplication and diversification create four albumin-1 enriched genomic islands encoding cyclotides, where they are physically grouped by similar pI and net charge values. We identify an ancestral hydrolytic AEP that exhibits neofunctionalization and multiple duplication events to yield two ligase-type AEPs. We propose cyclotides arise by convergence in C. ternatea where their presence enhances defense from biotic attack, thus increasing fitness compared to lineages with linear b-chains and ultimately driving the replacement of b-chains with cyclotides.

The temporal contiguity effect (TCE) is the tendency for the recall of one event to cue recall of other events originally experienced nearby in time. Retrieved context theory proposes that the TCE results from fundamental properties of episodic memory: binding of events to a drifting context representation during encoding and the reinstatement of those associations during recall. If these processes are automatic, the TCE should not be dependent on any encoding strategy and should, in fact, be present regardless of encoding intentionality. Here, we ask whether this theory is compatible with recent findings that the TCE is dramatically reduced under incidental encoding, even though memory accuracy is only modestly reduced. We begin by attempting to replicate this finding in a new large-scale study with over 5,000 participants in which we manipulated encoding intentionality between participants in both delayed free recall and continual distractor free recall. A small, but reliable, TCE was observed in all conditions, although the effect was dramatically reduced in incidental encoding. In a simulation study, we demonstrated that retrieved context theory can simultaneously account for both overall recall and the strength of the TCE in incidental encoding conditions. Additional analyses revealed that the incidental TCE is not an artifact of theoretically uninteresting factors, such as recency, and is consistent with being generated by the core contextual dynamics of retrieved context theory.

In this study, the plant biomass from the phytoremediation was recovered, prepared, and investigated catalytic ability for the α-pinene isomerization. The results show that the Fe_loaded AAL biochar can catalyze the isomerization of a-pinene, with the α-pinene conversion of 90.5 % and the selectivities for monocyclic terpenes (limonene, terpinolene and γ-terpinene) of 57.1 %, bicyclic terpene (camphene) of 24.6 %. Iron in the plant biomass from phytoremediation is considered a decisive factor that heightened the conversion of α-pinene and the yield of isomers. This research has initially opened up a new application for the plant biomass absorbing heavy metal from the phytoremediation stage to resolve contaminants efficiently. [ABSTRACT FROM AUTHOR]

Long non-coding RNAs (lncRNAs) are non-coding RNA molecules longer than 200 nucleotides that regulate gene expression at the transcriptional, post-transcriptional, and translational levels. Abnormal expression of lncRNAs has been identified in many human diseases. Future improvements in diagnostic, prognostic, and therapeutic techniques will be facilitated by a deeper understanding of disease etiology. Cardiovascular diseases (CVDs) are the main cause of death globally. Cardiac development involves lncRNAs, and their abnormalities are linked to many CVDs. This review examines the relationship and function of lncRNA in a variety of CVDs, including atherosclerosis, myocardial infarction, myocardial hypertrophy, and heart failure. Therein, the potential utilization of lncRNAs in clinical diagnostic, prognostic, and therapeutic applications will also be discussed.

Pore structure is a key determinant of soil functioning, and both root growth and activity of soil fauna are modified by and interact with pore structure in multiple ways. Cover cropping is a rapidly growing popular strategy for improving agricultural sustainability, including improvements in pore structure. However, since cover crop species encompass a variety of contrasting root architectures, they can have disparate effects on formation of soil pores and their characteristics, thus on the pore structure formation. Moreover, utilization of the existing pore systems and its modification by new root growth, in conjunction with soil fauna activity, can also vary by cover crop species, affecting the dynamics of biopores (creation and demolition). The objectives of this study were (i) to quantify the influence of 5 cover crop species on formation and size distribution of soil macropores (>36 μm Ø); (ii) to explore the changes in the originally developed pore architecture after an additional season of cover crop growth; and (iii) to assess the relative contributions of plant roots and soil fauna to fate and modifications of biopores. Intact soil cores were taken from 5 to 10 cm depth after one season of cover crop growth, followed by X-ray computed micro-tomography (CT) characterization, and then, the cores were reburied for a second root growing period of cover crops to explore subsequent changes in pore characteristics with the second CT scanning.Our data suggest that interactions of soil fauna and roots with pore structure changed over time. While in the first season, large biopores were created at the expense of small pores, in the second year these biopores were reused or destroyed by the creation of new ones through earthworm activities and large root growth. In addition, the creation of large biopores (>0.5 mm) increased total macroporosity. During the second root growing period, these large sized macropores, however, are reduced in size again through the action of soil fauna smaller than earthworms, suggesting a highly dynamic equilibrium. Different effects of cover crops on pore structure mainly arise from their differences in root volume, mean diameter as well as their reuse of existing macropores.

The venom of marine cone snails is mainly composed of peptide toxins called conopeptides, among which conotoxins represent those that are disulfide-rich. Publications on conopeptides frequently state that conopeptides attract considerable interest for their potent and selective activity, but there has been no analysis yet that formally quantifies the popularity of the field. We fill this gap here by providing a bibliometric analysis of the literature on cone snail toxins from 2000 to 2022. Our analysis of 3028 research articles and 393 reviews revealed that research in the conopeptide field is indeed prolific, with an average of 130 research articles per year. The data show that the research is typically carried out collaboratively and worldwide, and that discoveries are truly a community-based effort. An analysis of the keywords provided with each article revealed research trends, their evolution over the studied period, and important milestones. The most employed keywords are related to pharmacology and medicinal chemistry. In 2004, the trend in keywords changed, with the pivotal event of that year being the approval by the FDA of the first peptide toxin drug, ziconotide, a conopeptide, for the treatment of intractable pain. The corresponding research article is among the top ten most cited articles in the conopeptide literature. From the time of that article, medicinal chemistry aiming at engineering conopeptides to treat neuropathic pain ramped up, as seen by an increased focus on topological modifications (e.g., cyclization), electrophysiology, and structural biology.

MicroRNAs (miRNAs) are a group of endogenous non-coding RNAs that regulate gene expression. Alteration in miRNA expression results in changes in the profile of genes involving a range of biological processes, contributing to numerous human disorders. With high stability in human fluids, miRNAs in the circulation are considered as promising biomarkers for diagnosis, as well as prognosis of disease. In addition, the translation of miRNA-based therapy from a research setting to clinical application has huge potential. The aim of the current review is to: (i) discuss how miRNAs traffic intracellularly and extracellularly; (ii) emphasize the role of circulating miRNAs as attractive potential biomarkers for diagnosis and prognosis; (iii) describe how circulating microRNA can be measured, emphasizing technical problems that may influence their relative levels; (iv) highlight some of the circulating miRNA panels available for clinical use; (v) discuss how miRNAs could be utilized as novel therapeutics, and finally (v) update those miRNA-based therapeutics clinical trials that could potentially lead to a breakthrough in the treatment of different human pathologies.

Hydro power can provide a source of dispatchable low-carbon electricity and a storage solution in a climate-dependent energy mix with high shares of wind and solar production. Therefore, understanding the effect climate has on hydro power generation is critical to ensure a stable energy supply, particularly at a continental scale. Here, we introduce a framework using climate data to model hydro power generation at the country level based on a machine learning method, the random forest model, to produce a publicly accessible hydro power dataset from 1979 to present for twelve European countries. In addition to producing a consistent European hydro power generation dataset covering the past 40 years, the specific novelty of this approach is to focus on the lagged effect of climate variability on hydro power. Specifically, multiple lagged values of temperature and precipitation are used. Overall, the model shows promising results, with the correlation values ranging between 0.85 and 0.98 for run-of-river and between 0.73 and 0.90 for reservoir-based generation. Compared to the more standard optimal lag approach the normalised mean absolute error reduces by an average of 10.23% and 5.99%, respectively. The model was also implemented over six Italian bidding zones to also test its skill at the sub-country scale. The model performance is only slightly degraded at the bidding zone level, but this also depends on the actual installed capacity, with higher capacities displaying higher performance. The framework and results presented could provide a useful reference for applications such as pan-European (continental) hydro power planning and for system adequacy and extreme events assessments.

Goniothalamus elegans Ast is a plant commonly used in traditional medicine for the treatment of heart disease and diarrhea. Although there has been one published study on its chemical composition, limited research has been conducted on its bioactivity. In this study, the aerial part of G. elegans was extracted using the soaking method and further subjected to column chromatography for isolation. The isolated compounds were then evaluated for cytotoxicity using SRB staining. From the aerial part of G. elegans, four compounds were successfully isolated and identified as (23R)-21,23-epoxy-5a-cycloart-24-en-3ß-ol (1), stigmasterol (2), aristolactam BII (3), and piperolactam C (4). Among these compounds, aristolactam BII and piperolactam C exhibited moderate cytotoxicity against SW-489, AGS, LU-1, and HepG2 cell lines, with IC50 values ranging from 55.55 to 96.21 µg/mL. This study reveals that the phytochemical composition of G. elegans and demonstrates its potential cytotoxic activity, indicating promising avenues for further exploration in future research endeavors.
