Background: Early childhood growth and development is critical for long term health. Emerging science spotlights the significance of optimal gut microbiome and bone development during this period. The aim of the Bone And MicroBiOme Onset (BAMBOO) study is to determine age-appropriate trajectories for microbiome maturation and bone development, and to identify the influence of dietary factors in the process. This paper is to describe the rationale and study design, and reports study progress. Methods: BAMBOO is an ongoing prospective observational cohort study conducted in Tianjin, China. Children who meet the following requirements are invited to participate in this study: 1) full-term gestational birth (≥ 37 and ≤ 42 weeks); 2) singleton; and 3) signed informed consent by infant’s parents (or his/her legally accepted representative) and agree to fulfill the requirements of the study protocol. The exclusion criteria include pregnancy complication (such as pre-eclampsia, gestational diabetes), bowel disease, or currently participating or having participated in another clinical trial within 4 weeks prior to the start of this cohort. The study is composed of two groups of children: Group 1 includes children from birth to 12 months of age; group 2 includes children from 6 to 36 months of age. Questionnaires are used at different timepoints to collect information on infant feeding practice, medical history, concomitant medication, adverse events/serious adverse events and development benchmarks. Concurrent anthropometric measurements include length/height, weight, and bone measurements. Children’s dietary intake data are collected using 3-day-food diaries. Biological samples (stool, urine, and breastmilk) are also collected at different timepoints. Discussion: Recruitment of Bamboo started in September 2021 and is still ongoing. Data quality assessment and method validation have been conducted using early available samples. This study will provide unprecedented insights on early life microbiome maturation and bone development in Chinese infants and toddlers, and the impact of diet. The results may contribute to evidence-based policy making and inform nutrition healthcare programs for infants and toddlers aiming to benefit long-term health. Background: To investigate the causal link between early-life exposures and long-term health consequences, we established the Tianjin Birth Cohort (TJBC), a large-scale prospective cohort in northern China. Methods: TJBC aims to enroll 10,000 families with follow-ups from pregnancy until children’s six year-old. Pregnant women and their spouses were recruited through a three-tier antenatal healthcare system at early pregnancy, with follow-ups at mid-pregnancy, late pregnancy, delivery, 42 days after delivery, 6 months after delivery, and each year until 6 years old. Antenatal/neonatal examination, biological samples and questionnaires were collected. Results: From August 2017 to January 2019, a total of 3,924 pregnant women have already been enrolled, and 1,697 women have given birth. We observed the prevalence of gestational diabetes mellitus as 18.1%, anemia as 20.4%, and thyroid hypofunction as 2.0%. In singleton live births, 5.6% were preterm birth (PTB), 3.7% were low birth weight, and 7.3% were macrosomia. Based on current data, we also identified maternal/paternal factors which increased the risk of PTB, including paternal age (OR 1.07; 95% CI, 1.01–1.14 for each year increase), vaginal bleeding during pregnancy (OR 2.82; 95% CI, 1.54–5.17) and maternal early-pregnancy BMI (OR 1.08; 95% CI, 1.01–1.15 for each kg/m2 increase). Conclusion: TJBC has the strength of collecting comprehensive maternal, paternal, and childhood information. With a diverse range of biological samples, we are also engaging with emerging new technologies for multi-omics research. The study would provide new insight into the causal link between macro/micro-environmental exposures of early life and short/long-term health consequences.

A water-soluble and sensitive fluorescent probe N,N′-bis[tris-(2-aminoethyl)amine]-3,4,9,10-perylenetetracarboxylic diimide (PTRIS) was synthesized and, in combination with polydopamine (PDA), utilised in the detection of acetylcholinesterase (AChE) activity. PDA is spontaneously polymerized from dopamine (DA) in aerobic and alkaline solutions. The excellent absorption of PDA results in the aggregation of PTRIS around PDA as well as π–π stacking between them, which consequently quenched the fluorescence of PTRIS due to aggregation induced quenching (AIQ) in 9 min. The hydrolysis product of acetylthiocholine (ATCh) catalyzed by AChE, thiocholine (TCh), was proved to inhibit the polymerization of DA, therefore the free monomeric PTRIS retained its strong fluorescence intensity. The fluorescence was switched off and on depending on the activity of AChE. According to the change of fluorescence intensity at 550 nm, the detection limit of AChE was quantified as 0.02 mU mL−1. It was also proved that this probe possessed excellent selectivity for AChE. Tacrine and the organophosphate pesticide diazinon were further evaluated for inhibitor screening. The half-maximal inhibitory concentration value of tacrine and diazinon was calculated to be 1.4 and 1.6 μM respectively, revealing potential applications for inhibition and pesticide detecting.

An innovative strategy for circularly polarized luminescence (CPL) of carbon dots (CDs) has been developed: The achiral CDs displayed contrary supramolecular chirality and opposite CPL in two different bi-solvent systems, which are due to the formation of self-assembled helical nanostructures with two diverse assembly modes (P and M) in aggregate state via intermolecular π-π interactions and differential hydrogen bonding (H-bonding) without the need of any additional element of chirality.

Chiral materials related to circularly polarized luminescence (CPL) make up a rapidly developing new field that has broad application prospects in optoelectronic devices, selective recognition, biomedicine, and other fields. Biofunctional chiral materials are also attracting increasing attention because of their unique biocompatibility, chiral recognition, and coding. However, there has been little discussion on biomolecule-based CPL till now. In this Review, the latest progress in CPL materials related with biomolecules are reviewed, including the chiral construction from molecular level to giant microstructure, as well as their emerging applications. In addition, we discuss the challenges and prospects of bio-based CPL materials, hoping this work will provide new perspectives and insights for more related research.

Serotonin is one of the important neurotransmitters in human nervous system and associated with central nervous system diseases. Herein, we have prepared a novel electrochemical aptasensor for rapid and sensitive detection of serotonin by using the pre-designed and prepared DNA aptamers. In the absence of serotonin, the electron transfer rate on the aptasensor was faster than that in the presence of serotonin due to the hairpin structure of the aptamer was loose and MB could be closer to the electrode surface. While in the presence of serotonin, the hairpin structure of the aptamer was extended and MB was far away from the electrode surface. The effect of MB labeled sites on analytical performances of the proposed aptasensors was discussed by comparing sensitivity of the aptasensors that MB labeled in the intermediate of the aptamer with that MB labeled at the 3′ end of the aptamer. It was found that sensitivity of the intermediate-labeled aptasensor was much higher than the terminal-labeled aptasensor due to the specific conformational changes before and after aptamer binding to serotonin. The developed aptasensors exhibits a rapid electrochemical response and high sensitivity for the determination of serotonin. Under the optimal experimental conditions, the linear range for serotonin concentrations by the intermediate-labeled aptasensor was 1 pM–10 nM with a detection limit of 0.017 fM (S/N = 3). Moreover, the proposed aptasensor is reusable and shows good reproducibility and selectivity for the detection of serotonin in 100-fold diluted rat cerebrospinal fluid, suggesting a good application prospect in the detection of serotonin in real samples.

Dihydronicotinamide adenine dinucleotide (NADH) is an important enzyme in all living cells, which is found to be abnormally expressed in cancer cells. Since it is redox-active, an electrochemical detection method would be suitable for monitoring its concentration in biological fluids. Here we present a strategy for specific determination of NADH in real human serum by using RhIr@MoS2 nanohybrids based microsensor. To implement the protocol, RhIr nanocrysrals are in-situ grown onto MoS2 interlayers forming a nanohybrid structure (RhIr@MoS2). After being locally deposited on an electrochemical microsensor, it could be used for the analysis of NADH. The developed RhIr@MoS2 nanohybrids based microsensor possesses the ability for analyzing NADH at the applied potential of 0.07 V (much lower than most reported values). The detection limit is evaluated as low as 1 nmol/L even in bovine serum albumin (BSA) media. In addition, the sampling analysis of human serum from cancer patients and health controls shows that the microsensor displays good diagnostic sensitivity and specificity, illustrating that this developed detection technique is a relatively accurate method for measuring NADH in biological fluids. The proposed electrochemical microsensor assay also owns the benefits of convenience, disposable and easy processing, which make it a great possibility for future point-of-care cancer diagnosis.

A label-free fluorescent nanoprobe for iodide ion (I−) detection was developed based on the direct fluorescence quenching of spermine-functionalized carbon dots (SC-dots), whether in complex biological fluids or living cells. The positively charged SC-dots were fabricated via one-step microwave synthesis and exhibited excellent optical properties. Due to the strong quenching ability of I−, SC-dots were utilized for I− detection with high sensitivity and excellent selectivity, which offered a relatively low detection limit of 0.18 μM. This strategy was also successfully applied for I− detections in human serum and HeLa cells. The detection process is facile, highly sensitive and selective, providing a new insight into the potential applications of SC-dots for anion nanoprobe designs in clinical diagnosis and other biologically related areas.

Signal amplification strategies increase the complexities of biosensors while improving the response signals. Herein, a novel electrochemical biosensor was developed based on a DNA hydrogel for sensitive analysis using microRNA-21 (miRNA-21) as a detection model. Poly C sequences combined with C-Ag(I)-C hydrogel formed a DNA hydrogel by the unique interaction between the cytosines and silver ions. Thus, with a three-way conjunction structure of DNA, this C-Ag(I)-C hydrogel was constructed as a novel biosensor for the detection of miRNAs. With the assistance of this hydrogel, numerous silver ions gathered around DNA strands, which would amplify the signal. Under these conditions, the silver ions produced distinct square wave voltammetry oxidation peak currents. This electrochemical biosensor we designed exhibited a great linear relationship for the logarithm of the concentration of miRNA-21 from 1 fM to 100 pM with a detection limit of 0.117 fM. Furthermore, our sensors were able to differentiate miRNA-21 from its homologous family with satisfactory responsiveness in the dilute bovine serum system.

Limited healthcare capacity highlights the needs of integrated and simple sensing systems for personalized health monitoring. However, only a limited set of sensors can be employed for point-of-care applications, emphasizing the lack of a generalizable engineering strategy for sensor construction. Here, we report a de novo rational engineering strategy for the construction of an integrated electrochemical liquid biopsy (ELB) platform capable of direct profiling cancer exosomes from blood. Using a bottom-up approach for sensor design, a series of critical sensing functions is considered and encoded into the material interface by programming the electrode material with different chemical and structure features. We present that the rationally engineered electrochemical liquid biopsy platform is able to achieve one-step sensor fabrication, target isolation, non-fouling and high-sensitivity sensing, direct signal transduction and multiplexed detection. Integrating the multiplexed sensing with principal component analysis, we demonstrate the capability of the programmed sensing system on differentiating cancerous groups from healthy controls by analyzing clinical samples from lung cancer patients.

Theranostic sonosensitizers with combined sonodynamic and near infrared (NIR) imaging modes are required for imaging guided sonodynamic therapy (SDT). It is challenging, however, to realize a single material that is simultaneously endowed with both NIR emitting and sonodynamic activities. Herein, we report the design of a class of NIR-emitting sonosensitizers from a NIR phosphorescent carbon dot (CD) material with a narrow bandgap (1.62 eV) and long-lived excited triplet states (11.4 μs), two of which can enhance SDT as thermodynamically and dynamically favorable factors under low-intensity ultrasound irradiation, respectively. The NIR-phosphorescent CDs are identified as bipolar quantum dots containing both p- and n-type surface functionalization regions that can drive spatial separation of e−–h+ pairs and fast transfer to reaction sites. Importantly, the cancer-specific targeting and high-level intratumor enrichment of the theranostic CDs are achieved by cancer cell membrane encapsulation for precision SDT with complete eradication of solid tumors by single injection and single irradiation. These results will open up a promising approach to engineer phosphorescent materials with long-lived triplet excited states for sonodynamic precision tumor therapy.

Right (R)- and left (L)-handed helical G-quartet nanostructures were synthesized for the first time simultaneously via the self-assembly of 5'-guanosine monophosphate (GMP), the helical handedness of which is well regulated by metal ions. These g-nanostructures were further applied as circularly polarized luminescence (CPL) templates to realize full-color R-/L-CPL and Förster resonance energy transfer CPL. The glum value reached 10-2, indicating their excellent template function for CPL materials design and application.

High efficient and durable catalysts are always needed to lower the kinetic barriers as well as prolong the service life associated with oxygen evolution reaction (OER). Herein, a sequential synthetic strategy is considered to prepare a hierarchical nanostructure, in which each component can be configured to achieve their full potential so that endows the resulting nanocatalyst a good overall performance. In order to realize this, well-organized cobalt oxide (Co3O4) nanopillars are firstly grown onto ultrathin 1T-molybdenum sulfide (1T-MoS2) to obtain high surface area electrocatalyst, providing electron transfer pathways and structural stability. After that, zeolitic imidazolate framework-67 (ZIF-67) derived carbonization film is further in situ deposited on the surface of nanopillars to generate plentiful active sites, thereby accelerating OER kinetics. Based on the combination of different components, the electron transfer capability, catalytic activity and durability are optimized and fully implemented. The obtained nanocatalyst (defined as 1T-MoS2/Co3O4/CN) exhibits the superior OER catalytic ability with the overpotential of 202 mV and Tafel slope of 57 mV·dec-1 at 10mA·cm-2 in 0.1 M KOH, and good durability with a minor chronoamperometric decay of 9.15 % after 60000 s of polarization.

The formation of chirality of G-quartet materials has been of concern for a long time, however, the helix-handedness of G-quartet materials is still ambiguous, as well as the novel circularly polarized luminescence (CPL) properties. Here, we demonstrated that the handedness of G-quartet materials highly depends on their formation kinetics. By controlling the temperature or the initial concentration of reactants, we found that right-handed helical G-quartet nanostructures were synthesized in the slow process, while left-handed structures were synthesized in the fast process via orderly stacking. The phenomenon can be explained by the theory of kinetic trapping, in which a slow process leads to the thermodynamic equilibrium, while a fast process results in the kinetic trap state. Furthermore, the first kinetic trapping-controlled reversal CPL system was designed in G-quartet materials via chirality transfer, which has potential applications in CPL materials design and application.

Autophagy is a conserved cellular process playing a role in maintenance of cellular homeostasis and response to changing nutrient conditions via degradation and recirculation of cellular redundant components. Autophagy-related proteins (Atg) play important function in autophagy pathway. Aedes albopictus mosquito is an effective vector transmitting multiple viruses which cause serious human diseases. Moreover, Aedes albopictus mosquito is becoming a serious threat to human health due to its widening distribution in recent years and thus worth of more research attention. It was reported that autophagy might play a role in viral infection in Aedes mosquito. To better understand the interaction between autophagy and arbovirus infection in mosquito system, it is necessary to identify autophagy pathway in the system. However, autophagy in Aedes albopictus mosquito is still poorly understood so far. We recently identified AaAtg8, the first Atg protein reported in Aedes albopictus mosquito. This work further identified twelve atg genes in Aedes albopictus mosquito. Sequence and phylogenetic analysis of the twelve atg genes were performed. Expression profiles of all the twelve Aaatg genes in different developmental stages and genders of Aedes albopictus mosquito were conducted. Effects of chemicals inhibiting or inducing autophagy on the levels of eight identified AaAtg proteins were examined. The function of two identified AaAtg proteins AaAtg6 and AaAtg16 and their response to arbovirus SINV infection were studied preliminarily. Taken together, this work systematically identified Aedes albopictus atg genes and provided basic information which might help to elucidate the autophagy pathway and the role of autophagy in arbovirus infection in Aedes mosquito system.

β-Lactoglobulin amyloid fibril (BLGF)-capped gold nanoclusters (Au NCs) with red, green and blue emissions were fabricated via pH-dependent reduction strategy. The BLGF-Au NCs exhibited 3.2 times enhancement of fluorescence (λex = 500 nm, λem = 684 nm), a significant 42 nm red shift, a 11.57% quantum yield and a 1.4 μs decay time compared with native β-lactoglobulin (BLG)-stabilized Au NCs. Meanwhile, the multicolor Au NCs were employed for cell imaging via incubation with A549 cells for 14 h. According to the Michaelis-Menten equation, the kinetic parameters of the BLGF-Au NCs showed a lower Km value (66 μmol L-1) for 3,3,5,5-tetramethylbenzidine (TMB) and a higher vmax (3.74 × 10-8 M s-1) for H2O2, which are comparable with other artificial nanoenzymes and natural peroxidases. Based on the highly intrinsic peroxidase-like activity of the BLGF-Au NCs, a colorimetric method was developed for glucose determination with a detection limit of 1.5 μmol L-1 by determining the variation of the absorption at 652 nm, ranging from 5 to 100 μmol L-1. In addition, the glucose assay method also revealed a 101.02 to 104.16% recovery in a real human serum sample.

Since the Material Genome Initiative (MGI) was proposed, high-throughput based technology has been widely employed in various fields of materials science. As a theoretical guide, material informatics has been introduced based on machine learning and data mining and high-throughput computation has been employed for large scale search, narrowing down the scope of the experiment trials. High-throughput materials experiments including synthesis, processing, and characterization technologies have become valuable research tools to pin down the prediction experimentally, enabling the discovery-to-deployment of advances materials more efficiently at a fraction of cost. This review aims to summarize the recent advances of high-throughput materials experiments and introduce briefly the development of materials design based on material genome concept. By selecting representative and classic works in the past years, various high-throughput preparation methods are introduced for different types of material gradient libraries, including metallic, inorganic materials, and polymers. Furthermore, high-throughput characterization approaches are comprehensively discussed, including both their advantages and limitations. Specifically, we focus on high-throughput mass spectrometry to analyze its current status and challenges in the application of catalysts screening.

Enterovirus 71 (EV71) is a small, nonenveloped icosahedral RNA virus and is the predominant causative pathogen of hand-foot-and-mouth disease. Recently, microRNAs (miRNAs) are reported to play important roles in the pathogenesis of EV71 replication. This study investigated the role of miR-545 in the EV71 replication and explored the underlying molecular mechanisms. We showed that miR-545 was upregulated in the EV71-infected human embryonic kidney (HEK) 293 cells and rhabdomyosarcoma (RD) cells. Overexpression of miR-545 promoted the viral replication of EV71 and attenuated the inhibitory effects of EV71 on cell viability in HEK293 and RD cells; while knockdown of miR-545 significantly suppressed the EV71 replication in these two cell lines. Bioinformatics analysis and luciferase reporter assay showed that miR-545 directly targeted the 3'untranslated region of phosphatase and tensin homolog (PTEN) and tumor necrosis factor receptor-associated factor 6 (TRAF6) in HEK293 cells. Furthermore, miR-545 negatively regulated the messenger RNA (mRNA) and protein expression of PTEN and TRAF6. The mRNA and protein expression of PTEN and TRAF6 was also suppressed by EV71 infection, which was attenuated by miR-545 knockdown in HEK293 cells. Overexpression of PTEN and TRAF6 both suppressed the EV71 replication in HKE293 cells, and also attenuated the enhanced effects of miR-545 overexpression on the EV71 replication in HEK293 cells. Collectively, our study for the first time showed that miR-545 had an enhanced effect on the EV71 replication in HEK293 and RD cells. Further mechanistic results indicated that miR-545 promoted EV71 replication at least partly via targeting PTEN and TRAF6.

An inorganic titanium oxide (TiO) gel sensor was demonstrated for convenient detection of ascorbic acid (AA). It is composed of TiO (PI-TiO) functionalized with a perylene diimide derivative containing carboxylic groups as a new soft dopant material. A traditional solvothermal reaction is adopted to prepare the PI-TiO composite, which exhibits a different spectrum according to the reaction time. The final gel possesses a strong chelating affinity with AA molecules, in which phenol hydroxyl groups are shown to compete with those already present in PI. We further utilize the functionalized gel to prepare a series of films with a simple and portable AA response. A visual colour variation can be recognized by the naked eye, together with obvious fluorescence changes for selective and sensitive AA detection. Finally, the as-prepared gel film displays good stability and reproducibility for real sample responses with satisfying results.
