This study aimed to study protein expression in diabetic nephropathy (DN) rats and stem cell tissue repair based on nanomembrane concentration technology. Based on the polymer porous self-assembled nanomembrane technology, the content of total protein and albumin in the serum of rats in the control and experimental groups were measured. The obtained images were adopted to analyze the expression of cytoplasmic proteins and membrane proteins, and then the mechanism of stem cell tissue repair function was studied. The results showed that at 56 weeks of age, in contrast to control group, the total protein content in the serum of the experimental group evidently decreased. At 36 weeks of age, the cytoplasmic protein samples of diabetic rats in the experimental group were subjected to three two-dimensional protein electrophoresis. It was found that there were about 701 spots in each gel, and the matching rate was about 87.5%. In contrast to control group, 16 cytoplasmic proteins and 23 membrane proteins of the experimental group changed. The results of the differentially expressed protein analysis indicated that the change trends of protein spots. Based on polymer porous self-assembled nano-film technology, the expression of differential proteins in DN rats was analyzed, which would supplement new research assistance to the mechanism of DN.

To investigate the protective effects of metformin on the diabetic mice with cognitive impairment induced by the combination of streptozotocin (STZ) and isoflurane anesthesia. The isoflurane-anesthetized cognitive impairment model mice were established and then observed via behavioral tests and histopathological examination. Then these model mice were randomly assigned to three groups, which received the PBS, low and high doses of metformin, respectively. The body weight, food and water consumption of model mice were measured every other day. The mechanisms of metformin on ameliorating the cognitive dysfunction were further investigated by histomorphological, biochemical and Western blot analysis. After 14-days treatment of metformin, the diabetic symptoms in STZ-induced diabetic mice were significantly alleviated. Metformin could restore the isoflurane- and STZ-induced hippocampal tissue damage, cognitive and memory impairment in exposed space via improving the oxidative stress, upregulating the contents of glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) in the hippocampus tissues of diabetic mice. Furthermore, chronic treatment of metformin significantly down-regulated the expression of AGEs, RAGE, pNF-κB, iNOS, and COX-2. In conclusion, metformin can improve the isoflurane- and STZ-induced cognitive impairment in diabetic mice via improving oxidative stress and inhibiting the AGEs/RAGE/NF-κB signaling pathway.

Pollen analysis is rarely applied to cave sediments because of the complex range of processes influencing the origin, transport and taphonomy of pollen grains in caves. In order to evaluate the potential of pollen spectra from within caves for vegetation reconstruction, we compared the modern pollen from the interior and exterior of four caves in Guangxi and Guizhou, in southwestern China. The results show that the pollen spectra from within the sack-shaped caves, and from the front and middle of a long, narrow caves, were similar to those outside the caves, and therefore that the pollen spectra from these caves can reliably be used for palaeovegetation reconstruction. Wind is the major transport medium for the pollen entering caves, but animals may be additional transport medium. In addition, cave geometry is likely to be responsible for the observed spatial distribution of pollen concentrations in caves. In long, narrow caves, the entrance is the optimum sampling location for pollen analysis; however, in sack-shaped caves an optimum sampling location cannot be specified due to the gradient in pollen concentration between the entrance and the rear, together with its irregular distribution.

Background The human endometrium in premenopausal women is an active site of physiological angiogenesis, with regenerative cells present, suggesting that the endometrium contains adult angiogenic stem cells. In the context of cardiac repair after ischemic injury, angiogenesis is a crucial process to rescue cardiomyocytes. We therefore investigated whether human endometrium-derived stem cells (hEMSCs) can be used for cardiac repair after ischemic injury and their possible underlying mechanisms. Methods Comparisons were made between hEMSCs successfully isolated from 22 premenopausal women and human bone marrow mesenchymal stem cells (hBMSCs) derived from 25 age-matched patients. Cell proliferation, migration, differentiation, and angiogenesis were evaluated through in vitro experiments, while the ability of hEMSCs to restore cardiac function was examined by in vivo cell transplantation into the infarcted nude rat hearts. Results In vitro data showed that hEMSCs had greater proliferative and migratory capacities, whereas hBMSCs had better adipogenic differentiation ability. Human umbilical cord vein endothelial cells, treated with conditioned medium from hEMSCs, had significantly higher tube formation than that from hBMSCs or control medium, indicating greater angiogenic potentials for hEMSCs. In vivo, hEMSC transplantation preserved cardiac function, decreased infarct size, and improved tissue repair post-injury. Cardiac metabolism, assessed by 18F-FDG uptake, showed that 18F-FDG uptake at the infarction area was significantly higher in both hBMSC and hEMSC groups, compared to the PBS control group, with hEMSCs having the highest uptake, suggesting hEMSC treatment improves cardiomyocyte metabolism and survival after injury. Mechanistic assessment of the angiogenic potential for hEMSCS revealed that angiogenesis-related factors angiopoietin 2, Fms-like tyrosine kinase 1, and FGF9 were significantly upregulated in hEMSC-implanted infarcted hearts, compared to the PBS control group. Conclusion hEMSCs, compared to hBMSCs, have greater capacity to induce angiogenesis, and improved cardiac function after ischemic injury.

Objectives Gigantopithecus blacki, the largest hominoid known, is one of the representative Pleistocene mammals in southern China and northern Southeast Asia. Here we investigate the feeding ecology of G. blacki in its core habitat (Guangxi, Southern China) during the early Early Pleistocene, which was the early period in its evolution. Materials and methods The stable isotopic (C, O) analysis of tooth enamel of the fauna associated with G. blacki (n = 58), including the largest number of G. blacki teeth (n = 12) to date from the Liucheng Gigantopithecus Cave (~2 Ma), Guangxi, China, is undertaken. Results The δ13 C values of Liucheng fauna range from -12.9 to -19.0‰ with an average of -16.1 ± 1.3‰ (n = 58) and the δ18 O values range from -4.3 to -9.6‰ with an average of -6.9 ± 1.2‰ (n = 58). The δ13 C values of G. blacki range from -15.9‰ to -17.0‰ with an average of -16.5 ± 0.4‰ (n = 12), and the δ18 O values vary from -5.9‰ to -7.5‰ with an average of -6.6 ± 0.5‰ (n = 12). Conclusions The isotopic data show Guangxi was characterized by closed C3 forest and humid climate in the early Early Pleistocene. Niche partitioning is found among G. blacki, Sinomastodon, Ailuropoda and Stegodon, the typical megafauna in South China in the early Early Pleistocene. This could be one of the important factors for them to co-exist until the Middle Pleistocene. Smallest isotopic variations of G. blacki are found compared with those of contemporary animals, indicating a conservative foraging ecology i.e., limited foraging area and/or narrow dietary flexibility. Furthermore, the more confined foraging ecology of G. blacki is also seen in comparison with fossil and extant large-bodied primates. However, the unique dietary pattern of G. blacki does not seem to have hindered its survival. The environment in Guangxi during the early Early Pleistocene offered the suitable conditions for G. blacki to become one of the typical species in the faunal assemblages.

Fruit quality in most fleshy fruit crops is fundamentally linked to ripening-associated traits, including changes in colour. In many climacteric fruits, including tomato (Solanum lycopersicum), the phytohormone ethylene plays a key role in regulating ripening. Previous map-based cloning of YELLOW FRUITED-TOMATO 1 (YFT1) revealed that it encodes the EIN2 protein, a core component in ethylene signal transduction. A YFT1 allele with a genetic lesion was found to be down-regulated in the yft1 tomato mutant that has a yellow fruit phenotype and perturbed ethylene signalling. Based on bioinformatic analysis, yeast one hybrid assays and electrophoretic mobility shift assays, we report that transcription factor WRKY32 regulates tomato fruit colour formation. WRKY32 binds to W-box and W-box-like motifs in the regulatory region of the YFT1 promoter and induces its expression. In tomato fruits of WRKY32-RNAi generated lines, ethylene signalling was reduced, leading to a suppression in ethylene emission, a delay in chromoplast development, decreased carotenoid accumulation, and a yellow fruit phenotype. These results provide new insights into the regulatory networks that govern tomato fruit colour formation via ethylene signal transduction.

Parkinson's disease (PD) is a disorder characterized by a progressive loss of the dopaminergic neurons in the substantia nigra and a depletion of the neurotransmitter dopamine in the striatum. Our published results indicate that fasciculation and elongation protein zeta-1 (FEZ1) plays a role in the astrocyte-mediated protection of dopamine neurons and regulation of the neuronal microenvironment during the progression of PD. In this study, we examined the effects of engrafted type-2 astrocytes (T2As) with high expression of FEZ1 on the improvement of the symptoms and functional reconstruction of PD rats. T2As were stereotactically transplanted into the striatum of rats with PD induced by 6-hydroxydopamine (6-OHDA). An examination of apomorphine (APO)-induced rotations was performed to evaluate dopamine neuron damage and motor functions. Remarkably, the grafted cells survived in the lesion environment for six weeks or longer after implantation. In addition, the transplantation of T2As decrease the average velocity and the duration time of the APO-induced rotations, and increase the actuation time, as measured in the rotation behavioural tests. In the substantia nigra, the transplantation of T2As reduced the PD-induced GFAP, TH and FEZ1 downregulation. The grafted cells exclusively migrated to other regions near the injection site in the striatum and differentiated into GFAP+ astrocytes or TH+ neurons. Furthermore, by detecting monoamine neurotransmitters through high-performance liquid chromatography, we found that the nigrostriatal pathway had been repaired to some extent. Taken together, these results suggest that engrafted T2As with high expression of FEZ1 improved the symptoms and functional reconstruction of PD rats, providing a theoretical basis for FEZ1 as a potential target and engraftment of T2As as a therapeutic strategy in the treatment of PD.

Diabetic peripheral neuropathy (DPN) is one of the most important complications in diabetes mellitus (DM), which has been reported to be modulated by long non-coding RNAs (lncRNAs). The purpose of the current study is to explore the regulatory mechanism of lncRNA HCG18 on DPN in vitro. The expression of lncRNA HCG18, miR-146a, TRAF6, CD11c, and iNOS was detected by qRT-PCR. Through Enzyme-linked immunosorbent assay, the levels of inflammatory factors (TNF-α, IL-1β, and IL-6) were determined. M1 macrophage polarization was measured by flow cytometry analysis. The interactions between miR-146a and HCG18/TRAF6 were predicted by Starbase/Targetscan software and verified by the dual luciferase reporter assay. Western blot assay was performed to determine the protein expression of TRAF6. LncRNA HCG18 was highly expressed in DPN model and HG-induced macrophages. The levels of inflammatory factors (TNF-α, IL-1β, and IL-6) were elevated in DPN model. The expression of M1 markers (CD11c and iNOS) was visibly up-regulated in DPN model and was positively correlated with HCG18 expression. LncRNA HCG18 facilitated M1 macrophage polarization. In addition, miR-146a was identified as a target of lncRNA HCG18. Overexpression of miR-146a reversed the promoting effect of HCG18 on M1 macrophage polarization. Simultaneously, TRAF6 was a target gene of miR-146a TRAF6 expression was positively modulated by HCG18 and was negatively modulated by miR-146a. Down-regulation of TRAF6 reversed the promoting effect of HCG18 on M1 macrophage polarization. LncRNA HCG18 promotes M1 macrophage polarization via regulating the miR-146a/TRAF6 axis, facilitating the progression of DPN. This study provides a possible therapeutic strategy for DPN.

Objectives We aimed to investigate molar enamel development in fossil orangutans from Guangxi and shed light on the evolution of Asian great apes. Materials and methods We collected 32 fossil orangutan molars, most of which were from Guangxi apothecaries and the Guangxi Daxin Heidong cave, and prepared histological sections of each molar. We then characterized aspects of dental development, including long period line periodicity, number of Retzius lines and lateral enamel formation time, cuspal enamel thickness, and enamel formation time. Results The long period line periodicity in fossil orangutans ranged from 9 to 10 days (mean, 9.09 days). The molar lateral enamel formation time ranged from 1.48 to 3.17 years (540-1,152 days). Cuspal enamel thickness in fossil orangutan molars ranged from 949 to 2,535 μm, and cuspal enamel formation time ranged from 0.64 to 1.87 years. Molar enamel formation time of fossil orangutans ranged from 2.47 to 4.67 years. Discussion Long-period line periodicity of fossil orangutans from Guangxi was within the variation range of extant orangutans, and the average long period line periodicity (9.09 days) of fossil orangutans from Guangxi in this study was lower than the values for extant orangutans (9.5 days) and fossil orangutans (10.9 days) from Sumatra and Vietnam. Orangutan enamel thickness may have gradually decreased from the Middle Pleistocene to Holocene. Crown formation time of fossil orangutans was slightly longer than that of extant orangutans, and the M1 emergence age of fossil orangutans from Guangxi was about 4-6 years. These findings might indicate the regional difference or evolutionary changes in orangutans since Pleistocene. Dental development of the Guangxi fossil orangutans were more similar to that of Asian Miocene apes, suggesting the closer evolutionary relationship of orangutans to Miocene Asian fossil apes.

Dental remains provide crucial evidence for interpreting a populations’ affinity as well as human evolution. Recent studies of fossil human teeth have enriched our current view of morphological variation during the Pleistocene in East Asia. In the present paper, we describe a newly discovered late Pleistocene human tooth, a lower third premolar from the Weijiadong cave in Bijie in south-western China, which is dated around 18-20Ka BP using AMS Carbon 14 dating. The tooth is identified as belonging to an early modern human based on its morphology and size. The Tomes’ root is present in this lower third premolar, one of the non-metrical dental traits developed genetically. Moreover, we examine most of the fossil lower third premolars collected from China, as well as material from several Neolithic and historical human assemblages. We find that the Tomes’ root is very common in Chinese human fossils and occurs in high frequency in Neolithic and historic human remains from China. We suggest the high frequency of Tomes’ root may serve as a non-metric dental trait to support inferences regarding regional continuity of human evolution in East Asia.

1 Objectives The present study investigated the distribution of perikymata on anterior teeth of Miocene Lufengpithecus lufengensis so as to broaden the comparative data of developmental variation within and among hominoids. We also compared perikymata-spacing pattern of Lufengpithecus lufengensis with hominins and extant African great apes to understand the implication of dental development. 2 Materials and methods A total of 30 anterior teeth (including 6 I1, 10 I2, and 14 C) of Lufengpithecus lufengensis were examined using a scanning electron microscope and Keyence VHX-600EOS digital microscope to document the number and distribution of perikymata on their labial surfaces. The labial crown height of each tooth was divided into 10 equal deciles and the total perikymata number in each decile was recorded. The mean number of perikymata per millimeter was then calculated for each decile. SPSS statistical software was used to perform analyses of these data, including t-tests for sexual dimorphism and plots showing the perikymata distribution in Lufengpithecus lufengensis. 3 Results Perikymata counts of Lufengpithecus lufengensis in the first three deciles are fewer than the remaining deciles, and with perikymata becoming increasingly more closely packed as growth progresses from cusp to cervix, but decrease in density in the cervical decile. Besides, total labial perikymata counts of canines tend to display very significant sexual dimorphism. 4 Discussion Lufengpithecus lufengensis anterior teeth are more similar in their distribution of labial perikymata to those of Australopithecus than those of other hominins and extant African great apes from previous studies.

The yft1 tomato mutant has a yellow-fruited phenotype controlled by a recessive gene of YFT1 allele, which has been shown by map-based cloning to be a homolog of ETHYLENE INSENSITIVE 2 (EIN2). Genetic lesion of YFT1 allele in yft1 is attributed to a 573 bp DNA fragment (IF573) insertion at 1,200 bp downstream of the transcription start site. Transcriptomic analysis revealed that YFT1 lesion resulted in 5,053 differentially expressed genes (DEGs) in yft1 pericarp compared with the M82 wild type cultivar. These were annotated as being involved in ethylene synthesis, chromoplast development, and carotenoid synthesis. The YFT1 lesion caused a reduction in its own transcript levels in yft1 and impaired ethylene emission and signal transduction, delayed chromoplast development and decreased carotenoid accumulation. The molecular mechanism underlying the downregulated YFT1 allele in yft1 was examined at both RNA and DNA levels. The IF573 event appeared to introduce two negative regulatory sequences located at -272 to -173 bp and -172 to -73 bp in the YFT1 allele promoter, causing alterative splicing due to introduction of aberrant splicing sites, and breaking upstream open reading frames (uORF) structure in the 5'-UTR. Those results a new provided insight into molecular regulation of color formation in tomato fruit.

Background Long non-coding RNAs (lncRNAs) are involved in multiple biological processes in both mammals and plants. There is growing evidence that they are associated with development; but their expression and regulation during fruit ripening in the model plant tomato (Solanum lycopersicum) has yet to be described. Results Following integration of 134 RNA-seq data sets, we identified 79,322 putative lncRNAs, consisting of 70,635 lincRNAs, 8085 antisense non-coding RNAs (ancRNAs) and 602 sense lncRNAs (slncRNAs). lncRNAs had specific features that are distinct from mRNAs, including tissue-specificity, and shorter and fewer exons. Notably, more than 5000 of the novel lincRNAs were found to be expressed across the mature green (MG), breaker (BR) and breaker plus 7 days (BR + 7) developmental stages. The differently expressed lincRNAs had different DNA methylation profiles from the mRNAs. Conclusions Integrating transcriptome datasets and genome-wide screening enabled the identification of a comprehensive set of tomato lncRNAs. Here, we found that the lncRNAs DNA methylation profiles were different from those of mRNAs. This will help future investigation of lncRNA function, especially for the dissection of the molecular mechanisms involved in the regulation of fruit development. Lithospermum extract from Lithospermum is a kind of naphthoquinone, which has good anti-ultraviolet and anti-bacterial function. In this paper, the effects of different treatment temperature, time and ratio of liquid to liquid on the UV resistance of Lithospermum erythrorhizon extract were studied. The optimum extraction conditions were as follows: extraction temperature 60 ℃, extraction time 2 h, ratio of liquid to liquid of Lithospermum and ethanol 1:11. In this paper, the anti-UV finishing of cotton fabric was carried out, and the anti-ultraviolet and whiteness of the fabric were taken as the main indexes. The optimum process of the anti-UV finishing was as follows: the impregnation temperature was 70 ℃, the immersion time was 2h, 1:40. Compared with the uncoated cotton fabric, the fabric UPF value of the fabric was improved from 12.31 to 83.25, and the anti-ultraviolet performance was excellent, and it had certain bacteriostatic effect on Bacillus subtilis and Escherichia coli.

Twenty-eight (28) isolated orangutan fossil teeth are reported in this paper, which are newly discovered from Sanhe Cave in Chongzuo Ecological Park of Guangxi in southern China. The geological age is Early Pleistocene, approximately 1.2 Ma, based on the mammal fauna and paleomagnetic stratigraphy. The tooth crown areas are within the range of extant orangutans and subfossil orangutans from Indonesia. However, by comparing the distribution of the crown areas, the sample from Sanhe has crown areas that fall above the mean value of extant orangutans and Holocene orangutans. Morphological differences between the Sanhe, subfossil and extant orangutans are difficult to discern, although the occlusal wrinkles of the postcanine teeth of the fossil orangutans from Sanhe are less numerous but coarser than that of extant orangutans. The fossil orangutans from Sanhe are smaller than those from other caves from Chongzuo Ecological Park, and are slightly larger than samples from Mulanshan Cave. We conclude that the fossil orangutan teeth from the Early Pleistocene of Sanhe Cave are provisionally classified as Pongo pygmaeus weidenreichi, and better evidence is needed to interpret the evolution and taxonomy of the fossil orangutans in southern China.

Compared to medium-high temperature petroleum reservoirs (30°C-73°C), little is known about microbial regulation by nutrients in low-temperature reservoirs. In this study, we report the performance (oil emulsification and biogas production) and community structure of indigenous microorganisms from a low-temperature (22.6°C) petroleum reservoir during nutrient stimulation. Culture-dependent approaches indicated that the number of hydrocarbon-oxidizing bacteria (HOB), nitrate-reducing bacteria (NRB) and methane-producing bacteria (MPB) increased by between 10- and 1000-fold, while sulfate-reducing bacteria (SRB) were observed at low levels during stimulation. Phylogenetic analysis of the 16S rRNA gene indicated that Pseudomonas, Ochrobactrum, Acinetobacter, Halomonas and Marinobacter, which have the capability to produce surfactants, were selectively enriched. Methanoculleus, Methanosaeta, Methanocorpusculum and Methanocalculus showed the largest increase in relative abundance among archaea. Micro-emulsion formed with an average oil droplet diameter of 14.3 μm (ranging between 4.1 μm and 84.2 μm) during stimulation. Gas chromatographic analysis of gas production (186 mL gas/200 mL medium) showed the levels of CO2 and CH4 increased 8.97% and 6.21%, respectively. Similar to medium-high temperature reservoirs, HOB, NRB, SRB and MPB were ubiquitous in the low-temperature reservoir, and oil emulsification and gas production were the main phenomena observed during stimulation. Oil emulsification required a longer duration of time to occur in the low-temperature reservoir.

The present paper reports the recently discovered fossil teeth of Early Pleistocene Gigantopithecus blacki and associated mammalian fauna from Baeryan Cave, Bijie County in Guizhou Province, and also reviews briefly the known fossil sites of Gigantopithecus in south China of Pleistocene. In Early Pleistocene, Gigantopithecus had a wider distribution, but withdrew southward in Middle Pleistocene to a limited area mainly in South China, and it disappeared in the Late Pleistocene according to the present fossil records. Diet and habitat analysis from carbon isotope evidence is used to investigate the reasons for the extinction of Gigantopithecus, which fed on a pure C3 diet and lived in a forest habitat. It was clearly different from early hominins in South and East Africa, such as Australopithecus africanus, Paranthropus robustus and Paranthropus boisei, which had C4 diets. The extinction of Gigantopithecus was also related to the great changes of climate and environment of the Pleistocene, especially the last one million years, during which Homo became more and more prosperous and exerted great pressure on G. blacki.

Rice (Oryza sativa) is one of the most important staple foods for more than half of the global population. Many rice traits are quantitative, complex and controlled by multiple interacting genes. Thus, a full understanding of genetic relationships will be critical to systematically identify genes controlling agronomic traits. We developed a genome-wide rice protein-protein interaction network (RicePPINet, http://netbio.sjtu.edu.cn/riceppinet) using machine learning with structural relationship and functional information. RicePPINet contained 708 819 predicted interactions for 16 895 non-transposable element related proteins. The power of the network for discovering novel protein interactions was demonstrated through comparison with other publicly available protein-protein interaction (PPI) prediction methods, and by experimentally determined PPI data sets. Furthermore, global analysis of domain-mediated interactions revealed RicePPINet accurately reflects PPIs at the domain level. Our studies showed the efficiency of the RicePPINet-based method in prioritizing candidate genes involved in complex agronomic traits, such as disease resistance and drought tolerance, was approximately 2-11 times better than random prediction. RicePPINet provides an expanded landscape of computational interactome for the genetic dissection of agronomically important traits in rice.

Thymosinβ4 (Tβ4) is a peptide that is known to play important roles in protection, regeneration, and remodeling of injured tissues in humans, and that shows great promise in a range of clinical applications. However, current strategies to Tβ4 are insufficient to meet growing demand and have a number of limitations. In this current study we investigated whether expression of recombinant Tβ4 in plants, specifically in tobacco (Nicotiana tabacum) leaves, represents an effective approach. To address this question, a 168 bpTβ4gene optimized for tobacco codon usage bias was constitutively expressed in tobacco as a 4-unit repeat concatemer, fused to a polyhistidine tag. Quantitative polymerase chain reaction and Western blot analyses were used to verify4×Tβ4expression in 14 transgenic tobacco lines and enzyme-linked immunosorbent assay analysis indicated 4×Tβ4 protein concentrations as high as 3 μg/g of fresh weight in the leaves. We observed that direct administration of tobacco-derived Tβ4 was more effective than Tβ4 either obtained commercially or derived from expression inEscherichia coliat promoting splenocyte proliferationin vitroand wound healing in mice through an endothelial migration assay. This study provides new insights into the development of plant-derived therapeutic proteins and their application by direct administration.

The present study investigated the distribution of perikymata on anterior teeth of Miocene Lufengpithecus lufengensis so as to broaden the comparative data of developmental variation within and among hominoids. We also compared perikymata-spacing pattern of Lufengpithecus lufengensis with hominins and extant African great apes to understand the implication of dental development.A total of 30 anterior teeth (including 6 I1, 10 I2, and 14 C) of Lufengpithecus lufengensis were examined using a scanning electron microscope and Keyence VHX-600EOS digital microscope to document the number and distribution of perikymata on their labial surfaces. The labial crown height of each tooth was divided into 10 equal deciles and the total perikymata number in each decile was recorded. The mean number of perikymata per millimeter was then calculated for each decile. SPSS statistical software was used to perform analyses of these data, including t-tests for sexual dimorphism and plots showing the perikymata distribution in Lufengpithecus lufengensis.Perikymata counts of Lufengpithecus lufengensis in the first three deciles are fewer than the remaining deciles, and with perikymata becoming increasingly more closely packed as growth progresses from cusp to cervix, but decrease in density in the cervical decile. Besides, total labial perikymata counts of canines tend to display very significant sexual dimorphism.Lufengpithecus lufengensis anterior teeth are more similar in their distribution of labial perikymata to those of Australopithecus than those of other hominins and extant African great apes from previous studies.

Protein-protein interactions (PPIs) are essential to almost all cellular processes. To better understand the relationships of proteins in Arabidopsis (Arabidopsis thaliana), we have developed a genome-wide protein interaction network (AraPPINet) that is inferred from both three-dimensional structures and functional evidence and that encompasses 316,747 high-confidence interactions among 12,574 proteins. AraPPINet exhibited high predictive power for discovering protein interactions at a 50% true positive rate and for discriminating positive interactions from similar protein pairs at a 70% true positive rate. Experimental evaluation of a set of predicted PPIs demonstrated the ability of AraPPINet to identify novel protein interactions involved in a specific process at an approximately 100-fold greater accuracy than random protein-protein pairs in a test case of abscisic acid (ABA) signaling. Genetic analysis of an experimentally validated, predicted interaction between ARR1 and PYL1 uncovered cross talk between ABA and cytokinin signaling in the control of root growth. Therefore, we demonstrate the power of AraPPINet (http://netbio.sjtu.edu.cn/arappinet/) as a resource for discovering gene function in converging signaling pathways and complex traits in plants.

Hepatocellular carcinoma (HCC) is a common clinical primary malignant tumor; however, efficient drugs for the treatment of HCC are still lacking at the present time. To develop a new approach for liver cancer therapy, we designed a chimeric gene (his-HR) encoding a single-chain variable fragment of human HAb25 (hHscFv) fused to a cytotoxic ribonuclease from Rana catesbeiana (RC-RNase) and expressed the corresponding fusion protein in transgenic tobacco (Nicotiana tabacum). Eleven positive transgenic plant lines were identified from 204 regenerated tobacco plants by PCR and Southern blot analysis, and the immunocompetence of the recombinant his-HR protein was confirmed by Western blotting. The expression levels of his-HR protein ranged from 0.75 to 1.99 µg/g in the fresh tobacco leaves. To characterize the bifunction of the expressed his-HR protein in tobacco, binding specificity and cell toxicity to several cell lines were examined by the indirect immunocytochemical streptavidin-biotin complex method and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide assay. Data indicated that the his-HR protein had stronger specific binding affinity to HepG2 (human liver HCC cell line) than to the other tumor cell lines and normal liver cell line, and the capacity to kill the HCC cell lines SMMC7721 and HepG2 with an half maximal inhibiting concentration of 2.0 and 2.4 nM, respectively. The results suggest that recombinant bifunctional his-HR protein derived from transgenic plants may provide a novel strategy to treat HCC in the future.

Flowering time of tomato ( Solanum.chilense ) which is a self-incompatibility wild species flowers was determined using a series of landmark days. We established a correlation between the floral shapes and the days before anthesis. The model of tomato flower development schedule was then used to analyze the size of bud, anther, style, and the pollen scanning electron microscope (SEM) in the days before anthesis. Two new S-RNase genes of the Solanum .chilense which was the stylar SI genes were cloned. The structure of the two S-RNase was also analyzed. Finally, the different topology of the phylogenetic trees produced using available S-RNase sequences suggests that the S-RNase have different evolutionary histories. Enamel stable carbon isotope analyses were conducted on the large fossil ape Gigantopithecus blacki and an associated mammalian megafauna from Longgudong Cave in Jianshi and Juyuandong Cave in Liucheng, South China. The range in delta (13)C values (-18.8aEuro degrees to -14.1aEuro degrees) indicates that G. blacki and other large mammals fed on solely C3 biomass, and lived in forest habitats, and not open country or savannas. These results are consistent with other faunal and floral analyses for that time. The diet and habitat of G. blacki were significantly different from those of early hominins (Australopithecus and Paranthropus) from South and East Africa. Extinction of G. blacki probably was a result of forest habitat fragmentation and deterioration.

Thymosin α1 (Tα1) was widely used for the treatment of hepatitis (B and C) and several cancers. However, current production of Tα1 is difficultly meeting clinical needs. To develop a novel and safety approach for Tα1 production, we synthesized a Tα1 gene (124 bp) based on the plant codon usage bias and constructed a four-copy Tα1 gene concatemer (408 bp) by using isocaudamer technique. This 4 × Tα1 structure was cloned into plant binary expression vector pCAMBIA2300 with twin transfer deoxyribonucleic acids (T-DNAs) and integrated into lettuce genome via Agrobacterium-mediated transformation. Thirteen positive plants were identified by polymerase chain reaction and confirmed by Southern blot analysis, and 11 marker-free lettuce plants were obtained in T2 generation. The content of recombined Tα1 (rTα1) protein reached 1798.317 ± 87.312 ng/g in fresh leaves of transgenic lettuce. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay demonstrated that rTα1 protein stimulated mouse splenic lymphocyte proliferation in vitro. These data suggest that biologically active rTα1 was successfully expressed in marker-free transgenic lettuce, and this method could provide an alternative choice for large-scale production of Tα1 in the future.

Based on several of the earliest sites (Yuanmou, Renzidong, Longgudong, and Longgupo) in southern China, this article is a general review of some recent discoveries of archaeological evidence. The dominating similarity in mammalian faunas indicates a strong affinity of the environment among the sites. The influence of environmental change including geology and climate has been also considered here. Around 2 Ma in China, the emergence of a group of early hominid sites yielding human fossils, stone artifacts or both indicates that Asia is not a desolate land for early human evolution. The understanding of early human evolution in this region is needed to be better known. In addition to East Africa, Asia is another potential place for exploring early hominid sites. Without further validation, the long dominant theory of “Out of Africa” may be only taken as a tentative hypothesis, especially under the new conditions involving more evidence from other regions. Some wider possibilities are existent beyond East Africa.

Fossil records indicate orangutan-like hominoids have been widely distributed in south China during Pleistocene, although currently only surviving in the tropical forests of Kalimantan and Sumatra in Indonesia. This paper describes the recently discovered hominoid fossil teeth from human site of Mulanshan cave in Chongzuo of Guangxi, whose geological age is the Late Pleistocene, about 11000 yeas age based on associated mammal fauna and U-series dating. Compared with those of modern and subfossil orangutans from Indonesia, other fossil great apes from China, the hominoid teeth from Mulanshan cave are orangutan-like, but show somehow different from Indonesia’s orangutans, the average sizes of cheek teeth larger and occlusal enamel wrinkles less and simpler. They are classified temporarily as the subspecies of Pongo pygmaeus weidenreichi. Concerning the variations of morphological features and dental sizes of orangutan-like teeth from southern China and neighboring northern Vietnam, different subspecies or species or genus possibly, but the key evidence is necessary to be identified.

Different PCR–denaturing gradient gel electrophoresis (DGGE) protocols were employed to investigate bacterial communities in a high temperature and water flooded petroleum reservoir in Dagang oil field, China. Bacterial universal primers sets frequently used in PCR–DGGE were evaluated. Three primers sets P1 (341F-GC and 534R), P2 (341F-GC and 907R) and P3 (1055F and 1406R-GC) showed different DGGE patterns. Good separation and quality of patterns were obtained in DGGE analysis with the set P3. A total of 12 DNA fragments were excised from the DGGE gels and their sequences were determined. Clustering analysis of the DGGE profiles showed that bacteria in this petroleum reservoir belonged to four clusters. These results indicate that the procedure of DGGE analysis with the primer P3 (1055F and 1406R-GC) is suitable for investigating microbial community in petroleum reservoirs.

A field experiment was performed to monitor changes in exogenous bacteria and to investigate the diversity of indigenous bacteria during a field trial of microbial enhanced oil recovery (MEOR). Two wells (26-195 and 27-221) were injected with three exogenous strains and then closed to allow for microbial growth and metabolism. After a waiting period, the pumps were restarted and the samples were collected. The bacterial populations of these samples were analyzed by denaturing gradient gel electrophoresis (DGGE) with PCR-amplified 16S rRNA fragments. DGGE profiles indicated that the exogenous strains were retrieved in the production water samples and indigenous strains could also be detected. After the pumps were restarted, average oil yield increased to 1.58 and 4.52 tons per day in wells 26-195 and 27-221, respectively, compared with almost no oil output before the injection of exogenous bacteria. Exogenous bacteria and indigenous bacteria contributed together to the increased oil output. Sequence analysis of the DGGE bands revealed that Proteobacteria were a major component of the predominant bacteria in both wells. Changes in the bacteria population in the reservoirs during MEOR process were monitored by molecular analysis of the 16S rRNA gene sequence. DGGE analysis was a successful approach to investigate the changes in microorganisms used for enhancing oil recovery. The feasibility of MEOR technology in the petroleum industry was also demonstrated.

Self-incompatibility (SI), a phenomenon that is widespread among flowering plants (angiosperms), promotes outbreeding, resulting in increased genetic diversity and species survival. SI is also important in establishing intra- or interspecies reproductive barriers, such as those that are evident in the tomato clade, Solanum section Lycopersicon, where they limit the use of wild species inbreeding programs to improve cultivated tomato. However, the molecular mechanisms underlying SI are poorly understood in the tomato clade. In this study, an SI (Solanum chilense, LA0130) and a self-compatible (SC, Solanum pimpinellifolium, LA1585) tomato species were chosen to dissect the mechanism of SI formation using a comparative proteomics approach. A total of 635 and 627 protein spots were detected in two-dimensional electrophoresis (2-DE) maps of proteins from the SI and SC species, respectively. In the SC species, 22 differently expressed proteins (DEPs) were detected in SCP versus SCUP (self-pollination versus non-pollination in SC species). Of these, 3 and 18 showed an up-or down-regulated expression in the SCP protein sample, respectively, while only one DEP (MSRA, Solyc03g111720) was exclusively expressed in the SCP sample. In the SI species, 14 DEPs were found between SIP/SIUP, and 5 of these showed higher expression in SIP, whereas two DEPs (MLP-like protein 423-like, gene ID, 460386008 and (ATP synthase subunit alpha, gene ID, Solyc00g042130) were exclusively expressed in SIP or SIUP, respectively. Finally, two S-RNases (gene IDs, 313247946 and 157377662) were exclusively expressed in the SI species. Sequence homology analysis and a gene ontology tool were used to assign the DEPs to the 'metabolism', 'energy', 'cytoskeleton dynamics', 'protein degradation', 'signal transduction', 'defence/stress responses', 'self-incompatibility' and 'unknown' protein categories. We discuss the putative functions of the DEPs in different biological processes and how these might be associated with the regulation of SI formation in the tomato clade.

Tomato style is the pathway for pollen germination and pollen tubes growth from the stigma to the ovules where fertilization occurs. It is essential to supplying the nutrients for pollen tube growth and guidance for the pollen tubes. To our knowledge, style also regulates gametophytic self-incompatibility (SI) in tomato species. This study identified the metabolites and monitored the metabolic changes of self-incompatible and self-compatible tomato with self-pollinated or unpollinated styles by gas chromatography-mass spectrometry (GC-MS). A total of 9 classes of compounds were identified in SI and self-compatibility (SC) self-pollinated and unpollinated styles which included amino acids, sugars, fatty acids/lipids, amines, organic acids, alcohols, nitriles, inorganic acids and other compounds. The contents of d-Mannose-6-phosphate, Cellobiose, Myristic acid, 2,4-Diaminobutyric acid, Inositol and Urea were significantly decreased and the rest did not significantly change in SI styles. But change of metabolites content significantly happened in SC styles. In addition, among the total 9 classes of compounds, the different metabolites accounted for a different proportion in amino acids, sugars, amines, organic acids and alcohols compared SC and SI. The result indicated that the physiological changes of styles existed differences in SC and SI after self pollination.

The isolated yft1 allele controls the formation of fruit color in n3122 via the regulation of response to ethylene, carotenoid accumulation and chromoplast development. Fruit color is one of the most important quality traits of tomato (Solanum lycopersicum) and is closely associated with both nutritional and market value. In this study, we characterized a tomato fruit color mutant n3122, named as yellow-fruited tomato 1 (yft1), which produces yellow colored mature fruit. Fruit color segregation of the progeny from an intra-specific cross (M82 × n3122) and an inter-specific cross (n3122 × LA1585) revealed that a single recessive nuclear gene determined the yellow fruit phenotype. Through map-based cloning, the yft1 locus was assigned to an 88.2 kb region at the top of chromosome 9 that was annotated as containing 12 genes. Sequencing revealed that one gene, Solyc09g007870, which encodes ETHYLENE INSENSITIVE2 (EIN2), contained two mutations in yft1: a 13 bp deletion and a 573 bp insertion at position −318 bp upstream of the translation initiation site. We detected that EIN2 expression was substantially lower in yft1 than in the red-fruited M82 wild type and that, in addition, carotenoid accumulation was decreased, ethylene synthesis and perception were impaired and chromoplast development was delayed. The results implied that the reduced expression of EIN2 in yft1 leads to suppressed ethylene signaling which results in abnormal carotenoid production.

Background Tomato (Solanum lycopersicum) self-compatibility (SC) is defined as self-pollen tubes that can penetrate their own stigma, elongate in the style and fertilize their own ovules. Self-incompatibility (SI) is defined as self-pollen tubes that are prevented from developing in the style. To determine the influence of gene expression on style self-pollination, a transcriptome-wide comparative analysis of SC and SI tomato unpollinated/pollinated styles was performed using RNA-sequencing (RNA-seq) data. Results Transcriptome profiles of 24-h unpollination (UP) and self-pollination (P) styles from SC and SI tomato species were generated using high-throughput next generation sequencing. From the comparison of SC self-pollinated and unpollinated styles, 1341 differentially expressed genes (DEGs) were identified, of which 753 were downregulated and 588 were upregulated. From the comparison of SI self-pollinated and unpollinated styles, 804 DEGs were identified, of which 215 were downregulated and 589 were upregulated. Nine gene ontology (GO) terms were enriched significantly in SC and 78 GO terms were enriched significantly in SI. A total of 105 enriched Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways were identified in SC and 80 enriched KEGG pathways were identified in SI, among which “Cysteine and methionine metabolism pathway” and “Plant hormone signal transduction pathway” were significantly enriched in SI. Conclusions This study is the first global transcriptome-wide comparative analysis of SC and SI tomato unpollinated/pollinated styles. Advanced bioinformatic analysis of DEGs uncovered the pathways of “Cysteine and methionine metabolism” and “Plant hormone signal transduction”, which are likely to play important roles in the control of pollen tubes growth in SI species. The taxonomy, age and environment status of the Yuanmou hominoids had been controversial since their discovery. We launched new excavations and investigations in the Yuanmou Basin and analyzed both old and newly collected data. We conclude that all hominoids from the Yuanmou Basin and the so-called Pliocene hominids can be attributed into a single species, and its valid nomination is Lufengpithecus hudienensis. It is different from Lufengpithecus lufengensis and Lufengpithecus keiyuanensis. Based on the faunal comparison and paleomagnetic correlation, the strata yielding the Yuanmou hominoids are older than those of Lufeng hominoids and younger than those of the Kaiyuan hominoids. The age of the Yuanmou hominoids is Late Miocene. The environments of the Yuanmou hominoids were subtropical, hilly, with some dense forestry and partial transitional bushes and seasonal climate.
