Robotic surgery for rectal cancer is gaining popularity, but evidence on long-term oncological outcomes is scarce. We aimed to compare surgical quality and long-term oncological outcomes of robotic and conventional laparoscopic surgery in patients with middle and low rectal cancer. Here we report the short-term outcomes of this trial.This multicentre, randomised, controlled, superiority trial was done at 11 hospitals in eight provinces of China. Eligible patients were aged 18-80 years with middle (5 to 10 cm from the anal verge) or low (≤5 cm from the anal verge) rectal adenocarcinoma, cT1-T3 N0-N1 or ycT1-T3 Nx, and no evidence of distant metastasis. Central randomisation was done by use of an online system and was stratified according to participating centre, sex, BMI, tumour location, and preoperative chemoradiotherapy. Patients were randomly assigned at a 1:1 ratio to receive robotic or conventional laparoscopic surgery. All surgical procedures complied with the principles of total mesorectal excision or partial mesorectal excision (for tumours located higher in the rectum). Lymph nodes at the origin of the inferior mesenteric artery were dissected. In the robotic group, the excision procedures and dissection of lymph nodes were done by use of robotic techniques. Neither investigators nor patients were masked to the treatment allocation but the assessment of pathological outcomes was masked to the treatment allocation. The primary endpoint was 3-year locoregional recurrence rate, but the data for this endpoint are not yet mature. Secondary short-term endpoints are reported in this article, including two key secondary endpoints: circumferential resection margin positivity and 30-day postoperative complications (Clavien-Dindo classification grade II or higher). The outcomes were analysed according in a modified intention-to-treat population (according to the original assigned groups and excluding patients who did not undergo surgery or no longer met inclusion criteria after randomisation). This trial was registered with ClinicalTrials.gov, number NCT02817126. Study recruitment has completed, and the follow-up is ongoing.Between July 17, 2016, and Dec 21, 2020, 1742 patients were assessed for eligibility. 502 patients were excluded, and 1240 patients were enrolled and randomly assigned to receive either robotic surgery (620 patients) or laparoscopic surgery (620 patients). 69 patients were excluded (34 in the robotic surgery group and 35 in the laparoscopic surgery group). 1171 patients were included in the modified intention-to-treat analysis (586 in the robotic group and 585 in the laparoscopic group). Six patients in the robotic surgery group received laparoscopic surgery and seven patients in the laparoscopic surgery group received robotic surgery. 22 (4·0%) of 547 patients in the robotic group had a positive circumferential resection margin as did 39 (7·2%) of 543 patients in the laparoscopic group (difference -3·2 percentage points [95% CI -6·0 to -0·4]; p=0·023). 95 (16·2%) of patients in the robotic group had at least one postoperative complication (Clavien-Dindo grade II or higher) within 30 days after surgery, as did 135 (23·1%) of 585 patients in the laparoscopic group (difference -6·9 percentage points [-11·4 to -2·3]; p=0·003). More patients in the robotic group had a macroscopic complete resection than in the laparoscopic group (559 [95·4%] of 586 patients vs 537 [91·8%] of 585 patients, difference 3·6 percentage points [0·8 to 6·5]). Patients in the robotic group had better postoperative gastrointestinal recovery, shorter postoperative hospital stay (median 7·0 days [IQR 7·0 to 11·0] vs 8·0 days [7·0 to 12·0], difference -1·0 [95% CI -1·0 to 0·0]; p=0·0001), fewer abdominoperineal resections (99 [16·9%] of 586 patients vs 133 [22·7%] of 585 patients, difference -5·8 percentage points [-10·4 to -1·3]), fewer conversions to open surgery (10 [1·7%] of 586 patients vs 23 [3·9%] of 585 patients, difference -2·2 percentage points [-4·3 to -0·4]; p=0·021), less estimated blood loss (median 40·0 mL [IQR 30·0 to 100·0] vs 50·0 mL [40·0 to 100·0], difference -10·0 [-20·0 to -10·0]; p0·0001), and fewer intraoperative complications (32 [5·5%] of 586 patients vs 51 [8·7%] of 585 patients; difference -3·3 percentage points [-6·3 to -0·3]; p=0·030) than patients in the laparoscopic group.Secondary short-term outcomes suggest that for middle and low rectal cancer, robotic surgery resulted in better oncological quality of resection than conventional laparoscopic surgery, with less surgical trauma, and better postoperative recovery.Shenkang Hospital Development Center, Shanghai Municipal Health Commission (Shanghai, China), and Zhongshan Hospital Fudan University (Shanghai, China).

Green production of NH 3 , especially Li-mediate electrochemical N 2 reduction reaction (NRR) in non-aqueous solutions, is attracting thronging research interest. Controversies on NRR mechanism greatly impedes its optimization and wide applications. To understand the electrocatalytic process, we treated Au coated carbon fibrous paper (Au/CP) as the model catalyst. In-situ XRD for the first time witnessed the transformation of lithium intermediates during NRR. Au greatly improved electron transfer kinetics to catalyze metallic Li formation, and accordingly highly accelerated NRR spontaneously. The Faradaic efficiency of NRR on Au/CP reached 34.0%, and NH 3 yield was as high as ~50 μg h -1 cm -2 . Our research proved that the key step of Li-mediate non-aqueous NRR is electrocatalytic Li reduction, and offered a novel electrocatalyst design method for Li reduction.

Suppressing cellular signal transducers of transcription 2 (STAT2) is a common strategy that viruses use to establish infections, yet the detailed mechanism remains elusive, owing to a lack of structural information about the viral-cellular complex involved. Here, we report the cryo-EM and crystal structures of human STAT2 (hSTAT2) in complex with the non-structural protein 5 (NS5) of Zika virus (ZIKV) and dengue virus (DENV), revealing two-pronged interactions between NS5 and hSTAT2. First, the NS5 methyltransferase and RNA-dependent RNA polymerase (RdRP) domains form a conserved interdomain cleft harboring the coiled-coil domain of hSTAT2, thus preventing association of hSTAT2 with interferon regulatory factor 9. Second, the NS5 RdRP domain also binds the amino-terminal domain of hSTAT2. Disruption of these ZIKV NS5-hSTAT2 interactions compromised NS5-mediated hSTAT2 degradation and interferon suppression, and viral infection under interferon-competent conditions. Taken together, these results clarify the mechanism underlying the functional antagonism of STAT2 by both ZIKV and DENV.

Lipid metabolic reprogramming plays an essential role in regulating the progression of colorectal cancer (CRC). However, the effect of lysophosphatidic acid (LPA) metabolism on CRC development is incompletely characterized. Here, we compared the mRNA levels of human CRC tissues to those of paracarcinoma tissues and focused on the notably enriched LPA metabolic pathways. We identified and verified that 1-acylglycerol-3-phosphate O-acyltransferase 4 (Agpat4) was aberrantly expressed in CRC tissues and predicted poor survival in CRC patients. Manipulating Agpat4 expression in CRC cells did not affect the growth or migration of CRC cells in vitro, whereas Agpat4 silencing suppressed CRC cell growth in subcutaneous and peritoneal xenograft models. Mechanistically, Agpat4 silencing-induced LPA release from CRC cells and polarized macrophages to an M1-like phenotype through LPA receptors 1 and 3. This M1 activation, characterized by elevated p38/p65 signaling and increased proinflammatory cytokines, promoted the infiltration and activation of CD4+ and CD8+ T cells in the tumor microenvironment. Modulation of the Agpat4/LPA/p38/p65 axis regulated macrophage polarization, T-cell activity and CRC progression. Notably, combined therapy with LPA and regular chemotherapy drugs synergistically suppressed CRC development. Taken together, our results showed that the Agpat4/LPA axis in CRC cells regulated p38/p65 signaling-dependent macrophage polarization, T-cell activation, and CRC progression. The Agpat4/LPA/p38/p65 axis might represent a potential target for therapy in the clinic.

Photoelectrochemical water splitting can convert solar energy into clean hydrogen energy for storage. It is desirable to explore non-precious electrocatalysts for practical applications of a photoelectrode in a large scale. Here, we developed a facile spin-coating and in-situ photoelectrochemical reduction method to prepare a dispersed Cu electrocatalyst on a Si photocathode, which improves the performance remarkably. We find that thiourea in the precursor solution for spin-coating plays an important role in obtaining dispersed Cu particles on the surface of a Si photoelectrode. With thiourea in the precursor, the Cu/Si photocathode shows higher performance than the one without thiourea. Moreover, the Cu/Si photocathode also indicates good stability after 16 h illumination.

In light of significant effort conducted to manned deep space exploration, it is of high technological importance and scientific interest to develop the lunar life supporting system for long-term exploration and exploitation. And lunar in situ resource utilization offers great opportunity to provide the material basis of life supporting for lunar habitation and traveling. Based on the analysis of the structure and composition, the Chang’E-5 lunar soil sample was used for lunar-surface solar energy conversion, i.e. the extraterrestrial photosynthesis catalysts. By evaluating the performance of the Chang’E-5 lunar sample as photovoltaic-driven electrocatalyst, photocatalyst and photothermal catalysts, the full water splitting and CO2 conversion are able to be achieved with solar energy, water and lunar soil, with a wide range of product distribution, including O2, H2, CO, CH4 and CH3OH. Thus, we propose a potentially available extraterrestrial photosynthesis pathway on the moon, which could help us to achieve a ‘zero-energy consumption’ environment and life support system on the moon.

ObjectiveMultiple mechanical learning models were used to predict the therapeutic dose of 131I radionuclide in patients with hyperthyroidism, and to compare the calculation results of each prediction model to obtain the optimal model for dose prediction. Meanwhile, the classification model was used to classify the prognosis of the existing clinical hyperthyroidism case data in order to evaluate the administration results and provide reference for the dose given by clinicians.MethodsAccording to the data of hyperthyroidism patients treated with 131I in nuclear medicine department of many hospitals, a prediction model was established based on MATLAB. Firstly, the prediction results of BP neural network, radial basis function (RBF) neural network and support vector machine (SVM) were compared with small sample data, and then the optimal model was selected to predict the drug dose. BP-AdaBoost, SVM and random forest were used to classify the patients after recovery and evaluate whether the dose was accurate.ResultsThe average errors of BP neural network, RBF neural network and SVM models trained with small samples were 6.58%, 17.25% and 14.09% respectively. After comparison, BP neural network was selected to establish the prediction model. The data of 30 cases were randomly selected to verify BP neural network, and average error of the prediction results was 11.99%. Using SVM, BP-AdaBoost and random forest models, 100 groups of case data were selected as the training set and 10 groups as the test set. The classification accuracy were 80%, 90% and 100% respectively. The random forest model with the highest accuracy was selected as the large sample prediction. When 318 groups of cases were trained and 35 groups of cases were used for the test, the classification accuracy was 97.14%.ConclusionThis study compared the prediction effects of various prediction models on 131I therapeutic dose in patients with hyperthyroidism and the accuracy of prognosis classification. BP neural network and random forest achieved the best results respectively. The two models provide reference for clinicians when giving the dose, which has clinical practical significance.

Lysophosphatidic acid (LPA) is an endogenous cell signaling molecule, and dysregulation of LPA signaling pathways is accompanied by several types of cancer. Herein, we developed a chemical proteomic method for the proteome-wide identification of LPA-binding proteins. The method involves the synthesis of a desthiobiotin-conjugated LPA acyl phosphate probe for the covalent labeling, enrichment, and subsequent LC-MS/MS identification of LPA-binding proteins at the proteome-wide level. By conducting labeling reactions at two different probe concentrations (10 and 100 μM) in conjunction with an SILAC (stable isotope labeling by amino acids in cell culture)-based workflow, we characterized the LPA-binding capabilities of these proteins at the entire proteome scale, which led to the identification of 86 candidate LPA-binding proteins in HEK293T cells. Moreover, we validated that two of these proteins, annexin A5 and phosphoglycerate kinase 1, can bind directly with LPA. Together, we developed a novel LPA probe for the identification and characterizations of LPA-binding proteins from the entire human proteome. The method should be adaptable for the identification of other lipid-binding proteins.

An effective passive micromixer based on the principles of asymmetric split and recombine has been designed, developed, and investigated to enhance the mixing performance of the micromixer in a microfluidic system. The effects of geometrical parameters and the number of mixing units on mixing performance were studied at different Reynolds (Re) number (ranging from 1 to 80) via CFD software ANSYS CFX. The results revealed that the preferable number of mixing units is 6, and the optimal direction of reducer is opposite to the main channel flow; the D-shaped mixing channel forces the mixed fluid to produce the extended vortex and Dean vortices simultaneously which caused a strong collision at the confluence of fluids, thus leading to the superposition and enhancement of the vortex system; when Re number is 80 and the width ratio of mixing channel W2/W3 = 1, the mixing efficiency of micromixer can exceed 95%.

Brazing is an effective technique for joining tungsten and steel. However, the high residual stresses are produced due to the different coefficients of thermal expansions between tungsten and steel. Compared with the direct brazing with BNi-2 foil filler, BNi-2/Cu/ BNi-2 multiple interlayer was used as filler to minimize the residual stresses between tungsten and 316L steel. The brazing experiments were conducted at 1050 °C for 25 min using Cu foils with different thickness. The results show that tungsten and 316L steel have been successfully joined by brazing. The intermetallic compound of NiW formed at the W/BNi-2 interface, which was detrimental to the strength of the joint. The microhardness of different diffusion zones is higher than that of the substrates owing to the formation of intermetallic compound and solid solution. All specimens of shear testing fractured at the W/BNi-2 interface close to W substrate, and the average strength of joints was 197, 275 and 268 MPa with multiple interlayer thickness of 0.2 , 0.1 and 0.05 mm copper foil, respectively, while the average strength of joints was 143 MPa with BNi-2 foil filler. The significant increase in the joint shear strength can be ascribed to the Cu foil in the multiple interlayer because of with excellent plasticity and toughness. DNA methylation and trimethylated histone H4 Lysine 20 (H4K20me3) constitute two important heterochromatin-enriched marks that frequently cooperate in silencing repetitive elements of the mammalian genome. However, it remains elusive how these two chromatin modifications crosstalk. Here, we report that DNA methyltransferase 1 (DNMT1) specifically ‘recognizes’ H4K20me3 via its first bromo-adjacent-homology domain (DNMT1BAH1). Engagement of DNMT1BAH1-H4K20me3 ensures heterochromatin targeting of DNMT1 and DNA methylation at LINE-1 retrotransposons, and cooperates with the previously reported readout of histone H3 tail modifications (i.e., H3K9me3 and H3 ubiquitylation) by the RFTS domain to allosterically regulate DNMT1’s activity. Interplay between RFTS and BAH1 domains of DNMT1 profoundly impacts DNA methylation at both global and focal levels and genomic resistance to radiation-induced damage. Together, our study establishes a direct link between H4K20me3 and DNA methylation, providing a mechanism in which multivalent recognition of repressive histone modifications by DNMT1 ensures appropriate DNA methylation patterning and genomic stability., How histone modifications crosstalk with DNA methylation to regulate epigenomic patterning and genome stability in mammals remains elusive. Here, the authors show that DNA methyltransferase DNMT1 is a reader for histone H4K20 trimethylation via its BAH1 domain, which leads to optimal maintenance of DNA methylation at repetitive LINE-1 elements.

High-entropy materials (HEMs) including high-entropy alloys (HEAs) and high-entropy ceramics (HECs) at nanoscale have promising prospects in many fields, yet a robust synthesis strategy is lacking. Herein, we present a simple and general approach, laser scanning ablation (LSA), to synthesize a vast library of HEA and HEC nanoparticles (NPs) including alloys, sulfides, oxides, borides, nitrides, phosphides. The LSA method takes only 5 nanoseconds per pulse to ablate the corresponding NPs precursors at atmospheric temperature and pressure in alkanes. The ultra-rapid process ensures up to 9 dissimilar metallic elements combined uniformly regardless of their thermodynamic solubility. As laser pulse precisely confines energy to desired microregions, the LSA method enables HEM NPs loading on various substrates, even thermally-sensitive ones such as metals and glass. Applied as electrocatalysts for overall water splitting, HEM NPs achieved an overpotential of 185 mV @ 10mA cm-2, which was among the best activities. The LSA technique discloses a large collection of new nanostructured HEMs with unique properties and attractive functions. We believe this general strategy will provide a versatile and flexible material platform for a wild range of fields such as biology, catalysis, electronics and magnetics.

In mammals, repressive histone modifications such as trimethylation of histone H3 Lys9 (H3K9me3), frequently coexist with DNA methylation, producing a more stable and silenced chromatin state. However, it remains elusive how these epigenetic modifications crosstalk. Here, through structural and biochemical characterizations, we identified the replication foci targeting sequence (RFTS) domain of maintenance DNA methyltransferase DNMT1, a module known to bind the ubiquitylated H3 (H3Ub), as a specific reader for H3K9me3/H3Ub, with the recognition mode distinct from the typical trimethyl-lysine reader. Disruption of the interaction between RFTS and the H3K9me3Ub affects the localization of DNMT1 in stem cells and profoundly impairs the global DNA methylation and genomic stability. Together, this study reveals a previously unappreciated pathway through which H3K9me3 directly reinforces DNMT1-mediated maintenance DNA methylation.

DNA methylation plays a critical role in regulating gene expression, genomic stability, and cell fate commitment. Mammalian DNA methylation, which mostly occurs in the context of CpG dinucleotide, is installed by two de novo DNA methyltransferases, DNMT3A and DNMT3B. Oligomerization of DNMT3A and DNMT3B permits both enzymes to comethylate two CpG sites located on the same DNA substrates. However, how DNMT3A- and DNMT3B-mediated co-methylation contributes to the DNA methylation patterns remain unclear. Here we generated covalent enzyme–substrate complexes of DNMT3A and DNMT3B, and performed bisulfite sequencing-based single-turnover methylation analysis on both complexes. Our results showed that both DNMT3A- and DNMT3B-mediated co-methylation preferentially gives rise to a methylation spacing of 14 base pairs, consistent with the previous structural observation for DNMT3A in complex with regulatory protein DNMT3L and CpG DNA. This study provides a novel method for mechanistic investigation of DNMT3A- and DNMT3B-mediated DNA co-methylation.

Designing efficient oxygen evolution reaction (OER) electrocatalysts is essential for numerous sustainable energy conversion technologies. An obstacle that impedes the development of OER electrocatalysts is the insufficient emphasis on the spin attribution of electrons. Recently, the different spin configuration of reactants and products in the OER has been recognized as the factor that slows down the reaction kinetics. In this work, Mn substitution was introduced to LaCoO3, which brought about lattice expansion and reduced crystalline field splitting energy. This led to the increase in the effective magnetic moment, which triggers the transfer of Co3+ from low to higher spin states. Thus, the hybridization of Co eg and O 2p states across the Fermi level was strengthened. Specifically, with 25 at. % Mn substitution, LaCoO3 transits from a semiconductor to a half-metal, which benefits the spin-oriented electronic transport and resultantly promotes the OER. This method paves the way for the construction of spin pathways in catalysts.

Vincristine is extensively used chemotherapeutic medicine to treat leukemia. However, it remains a critical clinical problem with regard to its toxicity and drug-resistance. AMP-activated protein kinase (AMPK) is an energy sensor that is pivotal in maintaining cell metabolic homeostasis. It is reported that AMPK is involved in vincristine-induced apoptosis. However, whether AMPK is involved in chemotherapy-resistance is largely unclear. It is well-documented that metformin, a widely used medicine to treat type II diabetes, possesses anti-cancer activities, yet whether metformin affects leukemia cell viability via vincristine is unknown. In this study, we showed that both AMPKα1 mRNA and phosphorylated AMPK protein levels were significantly decreased in clinical leukemia samples. We further demonstrated that metformin sensitized leukemia cells to vincristine-induced apoptosis in an AMPK-dependent manner. In addition, knockdown of AMPKα1 significantly reduced the effects of metformin on vincristine-induced apoptosis. Taken together, these results indicate that AMPK activation is critical in metformin effects on vincristine-induced apoptosis and suggest a putative strategy of a combination therapy using metformin and vincristine in treatment of leukemia.

DNA methyltransferases (DNMTs) are enzymes responsible for establishing and maintaining DNA methylation in cells. DNMT inhibition is actively pursued in cancer treatment, dominantly through the formation of irreversible covalent complexes between small molecular compounds and DNMTs that suffers from low efficacy and high cytotoxicity, as well as no selectivity towards different DNMTs. Herein, we discover aptamers against the maintenance DNA methyltransferase, DNMT1, by coupling Asymmetrical Flow Field-Flow Fractionation (AF4) with Systematic Evolution of Ligands by EXponential enrichment (SELEX). One of the identified aptamers, Apt. #9, contains a stem-loop structure, and can displace the hemi-methylated DNA duplex, the native substrate of DNMT1, off the protein on sub-micromolar scale, leading for effective enzymatic inhibition. Apt. #9 shows no inhibition nor binding activity towards two de novo DNMTs, DNMT3A and DNMT3B. Intriguingly, it can enter cancer cells with over-expression of DNMT1, colocalize with DNMT1 inside the nuclei, and inhibit the activity of DNMT1 in cells. This study opens the possibility of exploring the aptameric DNMT inhibitors being a new cancer therapeutic approach, by modulating DNMT activity selectively through reversible interaction. The aptamers could also be valuable tools for study of the functions of DNMTs and the related epigenetic mechanisms.

DNA methylation, one of the major epigenetic mechanisms, plays critical roles in regulating gene expression, genomic stability and cell lineage commitment. Establishment and maintenance of DNA methylation in mammals is achieved by two groups of DNA methyltransferases: DNMT3A and DNMT3B, which are responsible for installing DNA methylation patterns during gametogenesis and early embryogenesis, and DNMT1, which is essential for propagating DNA methylation patterns during replication. Both groups of DNMTs are multi-modular proteins, containing a large N-terminal regulatory region in addition to the C-terminal methyltransferase domain. Recent structure-function investigations of the individual domains or large fragments of DNMT1 and DNMT3A have revealed the molecular basis for their substrate recognition and specificity, intramolecular domain-domain interactions, as well as their crosstalk with other epigenetic mechanisms. These studies highlight a multifaceted regulation for both DNMT1 and DNMT3A/3B, which is essential for the precise establishment and maintenance of lineage-specific DNA methylation patterns in cells. This review summarizes current understanding of the structure and mechanism of DNMT1- and DNMT3A-mediated DNA methylation, with emphasis on the functional cooperation between the methyltransferase and regulatory domains.

Lead halide perovskites (LHPs) have been investigated for photoelectrochemical hydrogen generation from water splitting. However, the harsh requirements in preparing the environment, i.e. isolating water and oxygen, hinder the wide applications of lead halide perovskites. Herein, an all-inorganic perovskite, i.e. a CsPbBr3-based photocathode, has been prepared to generate hydrogen. It is notable that as a valuable trial for a potential large-scale production, the whole preparation process was completed in an open-air environment. The LHP photocathode achieved the highest photocurrent of about 1.2 mA cm-2 at 0 VRHE. And the photocurrent remains around 94% after continuous illumination for 1 h with the Faradaic efficiency of 90%, illustrating a good photoelectrochemical stability. The all-inorganic LHP photocathodes are facile to prepare with a relatively good performance, and can be improved via band engineering and structure optimization, of which large-scale applications can be expected.

DNA methylation by de novo DNA methyltransferases 3A (DNMT3A) and 3B (DNMT3B) at cytosines is essential for genome regulation and development. Dysregulation of this process is implicated in various diseases, notably cancer. However, the mechanisms underlying DNMT3 substrate recognition and enzymatic specificity remain elusive. Here we report a 2.65-angstrom crystal structure of the DNMT3A-DNMT3L-DNA complex in which two DNMT3A monomers simultaneously attack two cytosine-phosphate-guanine (CpG) dinucleotides, with the target sites separated by 14 base pairs within the same DNA duplex. The DNMT3A-DNA interaction involves a target recognition domain, a catalytic loop, and DNMT3A homodimeric interface. Arg836 of the target recognition domain makes crucial contacts with CpG, ensuring DNMT3A enzymatic preference towards CpG sites in cells. Haematological cancer-associated somatic mutations of the substrate-binding residues decrease DNMT3A activity, induce CpG hypomethylation, and promote transformation of haematopoietic cells. Together, our study reveals the mechanistic basis for DNMT3A-mediated DNA methylation and establishes its aetiological link to human disease.

In the present study, diffusion bonding of two dissimilar materials TC6 and copper alloy was investigated in vacuum chamber by directly bonding and using Ni foil as interlayer. Interface quality of the joints was evaluated by mechanical property and microstructure. The maximum shear strength of directly bonding was found to be 64 MPa for the speciemen bonded at 850 °C, 5 MPa for 30 min; and the maximum shear strength with Ni foil interlayer was 113 MPa under the same bonding parameters. The bonding interfaces and fracture surfaces were analyzed by energy disperse spectrometer, scanning electron microscopy and X-ray diffraction. The results show that the diffusion region of directly bonding specimen generated several IMCs (Ti2Cu and Ti5CuSn3, etc.). Fracture morphology showed that brittle fracture present at the Ti5CuSn3 IMCs, which was the weak point of the joint. While the diffusion zone of the specimen with Ni foil interlayer consists of various phase including Ti2Ni, TiNi, TiNi3 at TC6 side, and Cu-Ni solid solution at ZQSn11-4-3 side, and fracture surface of joint present a mixture of brittle and ductile characteristics, and fracture initiated at the TiNi3/Ni interface.

The Yersinia outer protein J (YopJ) family of bacterial effectors depends on a novel acetyltransferase domain to acetylate signalling proteins from plant and animal hosts. However, the underlying mechanism is unclear. Here, we report the crystal structures of PopP2, a YopJ effector produced by the plant pathogen Ralstonia solanacearum, in complex with inositol hexaphosphate (InsP6), acetyl-coenzyme A (AcCoA) and/or substrate Resistance to Ralstonia solanacearum 1 (RRS1-R)WRKY. PopP2 recognizes the WRKYGQK motif of RRS1-RWRKY to position a targeted lysine in the active site for acetylation. Importantly, the PopP2-RRS1-RWRKY association is allosterically regulated by InsP6 binding, suggesting a previously unidentified role of the eukaryote-specific cofactor in substrate interaction. Furthermore, we provide evidence for the reaction intermediate of PopP2-mediated acetylation, an acetyl-cysteine covalent adduct, lending direct support to the 'ping-pong'-like catalytic mechanism proposed for YopJ effectors. Our study provides critical mechanistic insights into the virulence activity of YopJ class of acetyltransferases.

With the rapidly growing number of CT examinations, the consequential radiation risk has aroused more and more attention. The average dose in each organ during CT scans can only be obtained by using Monte Carlo simulation with computational phantoms. Since children tend to have higher radiation sensitivity than adults, the radiation dose of pediatric CT examinations requires special attention and needs to be assessed accurately. So far, studies on organ doses from CT exposures for pediatric patients are still limited. In this work, a 1-year-old computational phantom was constructed. The body contour was obtained from the CT images of a 1-year-old physical phantom and the internal organs were deformed from an existing Chinese reference adult phantom. To ensure the organ locations in the 1-year-old computational phantom were consistent with those of the physical phantom, the organ locations in 1-year-old computational phantom were manually adjusted one by one, and the organ masses were adjusted to the corresponding Chinese reference values. Moreover, a CT scanner model was developed using the Monte Carlo technique and the 1-year-old computational phantom was applied to estimate organ doses derived from simulated CT exposures. As a result, a database including doses to 36 organs and tissues from 47 single axial scans was built. It has been verified by calculation that doses of axial scans are close to those of helical scans; therefore, this database could be applied to helical scans as well. Organ doses were calculated using the database and compared with those obtained from the measurements made in the physical phantom for helical scans. The differences between simulation and measurement were less than 25% for all organs. The result shows that the 1-year-old phantom developed in this work can be used to calculate organ doses in CT exposures, and the dose database provides a method for the estimation of 1-year-old patient doses in a variety of CT examinations.

This Letter presents a facile alternative synthesis of a recoverable Au(III) catalyst supported on Fe3O4@ SiO2 similar to MPS grafted by poly(N-vinyl-2-pyrrolidone) (PVP). The solid magnetic support was prepared by anchoring 3-methacryloxypropyltrimethoxysilane (MPS) onto the Fe3O4@SiO2 surfaces followed by free radical polymerization with N-vinyl-2-pyrrolidone. Au(III) was immobilized onto the magnetic support in aqueous media to afford Au(III)/Fe3O4@SiO2 similar to PVP (catalyst 1). Catalyst 1 was characterized by FT-IR, TEM, VSM, TGA, XRD, and ICP-AES. The amount of Au in catalyst 1 was measured to be 0.64 wt % by ICP-AES. This newly prepared catalyst can catalyze the aromatic bromination reaction with comparable activity as homogeneous AuCl3. Moreover, the supported catalyst is easy to recover and can be used in four cycles without apparent loss of activity. (C) 2012 Elsevier Ltd. All rights reserved.

A practical and simple synthesis of 2,5-disubstituted oxazoles was developed via an iodine-catalyzed tandem oxidative cyclization. A wide range of common commercial aromatic aldehydes can be used as reaction substrates, which displayed excellent functional group compatibility in this reaction.

To understand the situation of radiation exposure to primary operators in the interventional X-ray procedures, a pilot survey was conducted for the 5 most frequently performed procedures in Shanghai in 2011. The distributions of entrance-surface air kerma (ESAK) were measured with thermoluminescent dosimeters (TLD) in 125 primary operators conducting the 5 procedures in 10 hospitals in Shanghai, and the equivalent doses ( H T ) for eye lens and extremity as well as the effective dose ( E ) were estimated. The results showed that the mean ESAKs were usually larger than 10 μGy for unshielded eye lens and extremity in each procedure, and the annual H T for unshielded eye lens exceeded 10 mSv in some procedures, such as the coronary angiography (CA) and angioplasty (PTCA). The mean annual effective doses were estimated to be 0.7–10.4 mSv among the 5 procedures, however, the values varied with different calculation algorithms.
