Abstract Backgrounds Homeobox C6 (HOXC6) is a gene that encodes for a transcription factor involved in various cellular processes, including development and differentiation, and regulates cancer progression. However, the carcinogenesis and effect of HOXC6 in lung adenocarcinoma (LUAD) still need further investigation. Methods The differential HOXC6 expression levels at the mRNA and protein level were explored in multiple public datasets, including The Cancer Genome Atlas (TCGA) and Human Protein Atlas (HPA) dataset. Gene Expression Omnibus (GSE31210), International Cancer Genome Consortium (ICGC) datasets and the LUAD sample from Affiliated Hospital of Guangxi Medical University. We also investigated the relation between HOXC6 expression and clinicopathologic indexes. Furthermore, the correlation of immune infiltration, drug responsiveness and HOXC6 were explored. Results The upregulated HOXC6 expressions at mRNA and protein levels were found in LUAD tissues compared to the normal lung tissues. Besides, the relatively shorter overall survival time, worse T and N stages, and lower immune scores were found in the high-expression HOXC6 subgroup. Notably, T cells regulatory (Tregs), Macrophages M0, and Plasma cells had the higher infiltration levels in the high-HOXC6 expression subgroup, while NK cells activated, Monocytes, Dendritic cells resting, and Mast cells resting had the lower infiltration levels. In drug sensitivity analysis, we revealed that LUAD patients with high-HOXC6 expression may be more susceptible to Camptothecin, Cytarabine, Docetaxel, Elesclomol, Rapamycin, Sorafinib, Temsirolimus, and Vorinostat. Conclusions Taken together, there is a great potential for HOXC6 to become a prognosis biomarker and contribute to develop treatment strategies for LUAD patients. Further mechanism exploration and drug development for HOXC6 are needed.

The yield stress- and zeta potential-pH behaviour of washed spherical α-Al2O3 suspension with and without three typical anionic additives were characterized. The zero zeta potential or isoelectric point (IEP) of washed α-Al2O3 particles locates at pH∼ 9.5. With the increased dosage of the anionic additives, IEP moves to a lower pH range. The maximum yield stress of 50 wt% α-Al2O3 suspension decreases from 78.3 Pa to 50 Pa by about 36 % when citric acid is 0.2 dwb% (dispersant g/100 g powder). According to the simulation of the MM2 model, the type of O-H···O hydrogen bond existed in the CA molecule which results in a stable steric repulsive effect. The pH range where yield stress exists also moves to the lower pH with the addition of anionic additives. However, the addition of Diisobutylene-maleic anhydride sodium salt and Polystyrene sulphonic acid sodium salt increase the maximum yield stress because of the specific attraction force, hydrophobic attraction force and charge attraction force.

A broadband radar cross-section (RCS) reduction employed by the vortex metasurface is proposed. The underlying physics is associated with the Pancharatnam-Berry geometrical phases involved in spin-to-orbital angular momentum conversion. The proposed metasurface generates spin-controlled vortex waves with different topological charges. The vortex singularity implements specular extremely low RCS reduction. The prototypes are fabricated and measured to verify the wideband performance of the RCS reduction. The simulated results illustrate that −10 dB broadband RCS reduction from 9 GHz to 22.5 GHz and the RCS reduction has peak values −37.2 dB, −40.8 dB, and −42.5 dB at 9.8 GHz, 13.1 GHz, and 20.8 GHz, respectively. The measurement results illustrate that −10 dB RCS reduction over 9.1–18 GHz is realized, and the RCS reduction peak −53 dB at 10.1 GHz. The measured electric field intensities and phase distribution of the vortex metasurface also be displayed. Due to the limitation of experimental conditions, no test was done in the range greater than 18 GHz. Far-field results achieve good agreement between simulations and experiments, and the near-field measurements are a subtle difference in the experimental results. This work reveals the potential advantages of vortex waves in the fields of multifunctional electromagnetic stealth, beamforming for communication systems, etc.

Forest can provide multiple benefits, such as supplying a variety of ecosystem services. However, the planet is currently at risk of deforestation as a result of increased urbanization and growing food demand. Countries around the world have introduced a wide range of policies to mitigate this issue. In China, the Linyi City government has launched the cross-county forest credit market transactions (CFCMT) policy, which showcased significant innovations in compliance credits and transaction entities to enhance motivation for afforestation. This study creates a no-policy counterfactual scenario to analyze the woodland growth and ecosystem service trade-offs between 2013 and 2021. The results demonstrate that the woodland grew faster after this policy, with a growth rate 1.78 times higher than that from 2013 to 2017. The areas of woodland growth with the policy are mainly located on the edges of cropland and woodland. The increased woodland primarily originates from cropland conversion, and compared to the no-policy scenario, these growing woodlands have gentler slopes and lower altitudes. After the policy’s enforcement, there were more increases in carbon storage, soil retention, and habitat quality. And ecosystem services showed a more optimizing trend in the direction of synergy at grid and town scales. We also find that larger spatial scales exhibit synergies and less accurately represent trade-off/synergistic change trends. The ecosystem services bundle consisting of carbon stocks, soil conservation and habitat quality showed a significant increase. This study reveals woodland growth and trade-offs among ecosystem services after the implementation of CFCMT policy, offering valuable insights for future afforestation policy optimization and promotion.

Nonlinear effects in optical fiber frequency transfer have a significant impact on the precision of frequency transfer. We investigate the main nonlinear effects, including the Brillouin scattering and the Raman scattering, in optical fiber frequency transfer through theoretical and simulation calculations in detail. The calculation results show that the threshold powers of the Brillouin scattering and the Raman scattering decrease with the increase in the fiber length; however, the fiber length has little to no impact on the threshold powers when the fiber length is greater than 10 km. The threshold powers, including the Brillouin scattering and the Raman scattering, increase as the attenuation coefficient increases. Conversely, when it comes to the gain coefficients, the outcomes exhibit a reverse trend. When the linewidth Δvlaser of the laser source is from 1 Hz to 1 MHz, the linewidth Δvlaser does not affect the threshold powers of the Brillouin scattering. This study seeks to offer design guidance aimed at mitigating nonlinear effects in optical fiber frequency transfer. The calculated results hold considerable potential in guiding various applications reliant on Brillouin and Raman scattering properties, such as laser technology and optical fiber sensing.

Since multiple studies have reported that small nucleolar RNAs (snoRNAs) can be serve as prognostic biomarkers for cancers, however, the prognostic values of snoRNAs in lung adenocarcinoma (LUAD) remain unclear. Therefore, the main work of this study is to identify the prognostic snoRNAs of LUAD and conduct a comprehensive analysis. The Cancer Genome Atlas LUAD cohort whole-genome RNA-sequencing dataset is included in this study, prognostic analysis and multiple bioinformatics approaches are used for comprehensive analysis and identification of prognostic snoRNAs. There were seven LUAD prognostic snoRNAs were screened in current study. We also constructed a novel expression signature containing five LUAD prognostic snoRNAs (snoU109, SNORA5A, SNORA70, SNORD104 and U3). Survival analysis of this expression signature reveals that LUAD patients with high risk score was significantly related to an unfavourable overall survival (adjusted P = 0.01, adjusted hazard ratio = 1.476, 95% confidence interval = 1.096-1.987). Functional analysis indicated that LUAD patients with different risk score phenotypes had significant differences in cell cycle, apoptosis, integrin, transforming growth factor beta, ErbB, nuclear factor kappa B, mitogen-activated protein kinase, phosphatidylinositol-3-kinase and toll like receptor signaling pathway. Immune microenvironment analysis also indicated that there were significant differences in immune microenvironment scores among LUAD patients with different risk score. In conclusion, this study identified an novel expression signature containing five LUAD prognostic snoRNAs, which may be serve as an independent prognostic indicator for LUAD patients.

The Arithmetic Optimization Algorithm (AOA) is a meta-heuristic algorithm inspired by mathematical operators, which may stagnate in the face of complex optimization issues. Therefore, the convergence and accuracy are reduced. In this paper, an AOA variant called ASFAOA is proposed by integrating a double-opposite learning mechanism, an adaptive spiral search strategy, an offset distribution estimation strategy, and a modified cosine acceleration function formula into the original AOA, aiming to improve the local exploitation and global exploration capability of the original AOA. In the proposed ASFAOA, a dual-opposite learning strategy is utilized to enhance population diversity by searching the problem space a lot better. The spiral search strategy of the tuna swarm optimization is introduced into the addition and subtraction strategy of AOA to enhance the AOA’s ability to jump out of the local optimum. An offset distribution estimation strategy is employed to effectively utilize the dominant population information for guiding the correct individual evolution. In addition, an adaptive cosine acceleration function is proposed to perform a better balance between the exploitation and exploration capabilities of the AOA. To demonstrate the superiority of the proposed ASFAOA, two experiments are conducted using existing state-of-the-art algorithms. First, The CEC 2017 benchmark function was applied with the aim of evaluating the performance of ASFAOA on the test function through mean analysis, convergence analysis, stability analysis, Wilcoxon signed rank test, and Friedman’s test. The proposed ASFAOA is then utilized to solve the wireless sensor coverage problem and its performance is illustrated by two sets of coverage problems with different dimensions. The results and discussion show that ASFAOA outperforms the original AOA and other comparison algorithms. Therefore, ASFAOA is considered as a useful technique for practical optimization problems.

Harmonizing human activities with the natural environment has received much attention as a development goal and is a longstanding pursuit of human society. In this study, we investigated the sustainability of green development from the perspective of the coordination between economic and environmental subsystems at the place-based scale, with 290 cities in China as research objects. An interdisciplinary method combining an improved data envelopment analysis model and Lotka-Volterra model was conducted to examine the two subsystems’ development efficiency and their correlation (i.e., mutualism, commensalism, amensalism, sacrifice, competition, independence). The results indicate that one-third of the cities’ economic and environmental subsystems are in a mutualistic state, with a relatively stable social-ecological system. However, the two subsystems are in a state of competition in one-fourth of the cities, with rather intense vulnerability, where the harmonious coexistence of humans and nature is facing more significant challenges. In addition, vulnerability of the social-ecological system in economically developed cities shows substantial polarization. The two subsystems in most of these cities are in mutualism or amensalism relations, whereas the social-ecological system in economically underdeveloped cities shows even greater vulnerability.

We characterize the frequency response of channel-interleaved photonic analog-to-digital converters (CI-PADCs) theoretically and experimentally. The CI-PADC is composed of a photonic frontend for photonic sampling and an electronic backend for quantization. The photonic frontend includes a photonic sampling pulse generator for directly high-speed sampling and an optical time-division demultiplexer (OTDM) for channel demultiplexing. It is found that the frequency response of the CI-PADC is influenced by both the photonic sampling pulses and the OTDM, of which the combined impact can be characterized through demultiplexed pulse trains. First, the frequency response can be divided into multiple frequency intervals and the range of the frequency interval equals the repetition rate of demultiplexed pulse trains. Second, the analog bandwidth of the CI-PADC is determined by the optical spectral bandwidth of demultiplexed pulse trains which is broadened in the OTDM. Further, the effect of the OTDM is essential for enlarging the analog bandwidth of the CI-PADC employing the photonic sampling pulses with limited optical spectral bandwidth.

In order to reverse the global trend of biodiversity loss, the concept of ecosystem services has been widely applied to make policymakers and the general public realize that conserving biodiversity possesses both intrinsic and utilitarian values. However, to achieve this goal, it is necessary to first have a clear understanding of the relationships between biodiversity and ecosystem services (BES). To advance our understanding of this issue, we first reviewed the major progress in current BES studies, with an emphasis on three biodiversity dimensions (i.e., taxonomic diversity, functional diversity, and ecosystem diversity). Based on the findings, we then propose three research topics as future directions: (1) More direct and explicit studies on the effects of different dimensions of biodiversity on various ecosystem service types; (2) developing a biodiversity-based understanding of the formation of ecosystem services; (3) creation of science-based ecosystem management plans and policies that can maximize synergies between biodiversity conservation and ecosystem service enhancement. By conducting such research, we will be able to not only further understand the complex relationships between biodiversity and ecosystem services but also better promote the concept of ecosystem services for more successful biodiversity conservation in the future.

Magnetic viscosity (MV) effect is a disturbance to the transient electromagnetic method (TEM). This effect will cause slow decay at the intermediate-late time of the TEM responses, leading to erroneous interpretation. Although there are extensive researches on the MV distortion to TEM data, they are focused on conventional loop source configuration, while little has been studied for grounded-wire source configuration that has become the essential role for relatively deep-buried targets. Hence, we present the study of MV effect on the grounded-wire source TEM data. We first derived the formulas for calculating the secondary field due to MV effect, based on the Chikazumi model. The dependence of the MV effect on model parameters, such as resistivity, susceptibility, offset and geometry of the superparamagnetic layer, is then examined to illustrate the intrinsic physical mechanism of MV effect. We find that the MV effect on the grounded-wire source TEM data can be suppressed by adjusting the parameters of the survey design. Thereafter, we propose the method to quantify the MV effect when estimates of resistivity and susceptibility are available. Finally, the method for choosing the optimal offset to suppress the MV effect is proposed, which can be used as a guide for the installation of fieldwork of grounded-wire TEM survey.

Optical reference cavities are a key component of ultra-stable lasers. Based on the theory of elastic deformation, the displacement difference between the top and bottom mirrors of an optical vertical cavity supported on the midplane was analyzed in the present research. The displacements and tilt angles of the two mirrors of the optical reference cavity were deduced based on Poisson's ratio. In order to verify the validity of the theoretical analysis, the vibration sensitivity induced by the elastic deformation of vertical optical reference cavities was simulated by finite element analysis. In order to compensate for the displacement difference between the top and bottom mirrors under an external force, the zero vibration sensitivity of the optical cavities was achieved by changing the thicknesses of Ultra-Low Expansion glass rings and the mirrors. The results and methods presented in the present paper may facilitate the design of vertical reference cavities, especially transportable optical reference cavities.

Since the beginning of the 21st century, the net primary productivity (NPP) of grassland in the Three-Rivers Headwaters (TRH) region has changed significantly. In this study, NPP was assessed by the Carnegie-Ames-Stanford approach (CASA) model in TRH from 2000 to 2019. The abrupt changes of NPP and meteorological factors were analyzed by cumulative departure, MK test, and Pettitt test. The contributions of meteorological and human factors to changes in grassland NPP were quantitatively assessed using the scenario simulation method. The obtained results showed that: (1) From 2000 to 2019, the NPP of grasslands increased from 135.72 to 141.16 gC/m−2a−1. However, the overall growth trend was not significant, and the proportion of significant growth was only 31.45%; (2) An abrupt increase in meteorological factors occurred around 2005, while an abrupt increase in NPP occurred around 2008, which showed that 2008 was the year when human factors, such as ecological projects and policies, began to show a significant impact on the growth of NPP; and (3) The contribution of human factors to the abrupt increase in NPP was significantly greater than the contribution of meteorological factors. The contribution of human factors exceeded 70% in 93.68% of the studied area, reaching 98% in general, while the contribution of meteorological factors was less than 2%. Among them, the human contributions to the Yangtze River Source, the Yellow River Source, and the Lancang River Source all exceeded 95%. The negative effects of meteorological factors on the growth of NPP in the abovementioned three regions were as high as 47.35%, 48.66%, and 36.91%, respectively. Human factors have contributed greatly to the increase in NPP in most of the source areas of the Yellow River, the eastern part of Tanggulashan County, the southeastern part of Zhiduo County, and the western part of Zaduo County at the source of the Yangtze River.

In this study, a new closed-form expression is developed to determine the equivalent electromagnetic parameters (permittivity and permeability) of biaxial anisotropic honeycomb radar-absorbing materials (RAMs). Based on the equivalent medium theory, the nonmagnetic substrate and absorbing coating of the honeycomb structure are equivalent to one kind of material, and then, the effective electromagnetic properties of irregular hexagonal (compressed or stretched) honeycombs are obtained by using the layered anisotropic model. In order to validate the effectiveness and correctness of this new closed-form expression, experimental tests and numerical simulation are adopted. The results show good agreement among the experimental tests, numerical simulation, and theoretical analysis within the specified frequency range (2–18 GHz), which confirms the validly of the proposed new method. Compared with the previous layered anisotropic model, the modified new model is more accurate for irregular hexagonal honeycomb RAMs. Furthermore, it can predict the effective permittivity (permeability) precisely without experimental measurements or numerical simulation, and it is of great significance for the design of honeycomb RAMs.

Brownian thermal noise (TN) of ultra-stable cavities (USCs) imposes a fundamental limitation on the frequency stability of ultra-narrow linewidth lasers. This work investigates the TN in cylindrical USCs with the four support pads in detail through theoretical estimation and simulation. To evaluate the performance of state-of-the-art ultra-narrow linewidth lasers, we derive an expression of the TN for a cylindrical spacer according to the fluctuation–dissipation theorem, which takes into account the front face area of the spacer. This estimation is more suitable for the TN of the cylindrical USC than the previous one. Meanwhile, we perform detailed studies of the influence of the four support pads on the TN in cylindrical USCs for the first time by numerical simulations. For a 400 mm long cylindrical USC with an ultra-low expansion spacer and fused silica substrates, the displacement noise contributed from the four support pads is roughly four times that of the substrates and the GaAs/AlGaAs crystalline coating. The results show that the four support pads are the primary TN contributors under some materials and geometries of USCs.

We extend the array theory to design 2D curved coding metasurfaces in order to achieve arbitrary wavefront control. The extended array theory establishes the relationship between wavefront and curved-surface, which provides a simple and effective method to obtain the layout of desired metasurfaces. According the theory, we present 2D curved metasurfaces with 3-bit coding elements. Both simulations and measurements demonstrate that the proposed designs can manipulate wavefront effectively. For normal incidence, the aperture efficiency of normal reflection is more than 90%. For general oblique incidence, the proposed designs almost meet the expectances, but the aperture efficiency decreases with the increase of desired scattering elevation angle. The curved coding metasurfaces are expected to be applied to many devices such as conformal antenna array.

Conventional flammable liquid electrolytes are not suitable for next generation lithium metal batteries with high energy density, stable performance, and excellent safety. Although solid polymer electrolytes have been studied for decades, it is still quite challenging for them to be pushed forward for practical application. Developing polymer/inorganic hybrid electrolytes that take advantage of both polymers and inorganic materials is considered to be an important route to make breakthrough. Ionic liquids are green materials in electrochemical process with unique advantages. This review summarizes recent research advances in developing ionic liquid containing, polymer/inorganic hybrid electrolytes, including their preparation routes, ion conduction, electrochemical properties, and mechanical and thermal properties. Abstract Background Fibroblast growth factor 9 (FGF9) is a heparin-binding growth factor, secreted by both mesothelial and epithelial cells, which participates in hair follicle regeneration, wound healing, and bone development. A suitable source of recombinant human FGF9 (rhFGF9) is needed for research into potential clinical applications. We present that expression of oleosin-rhFGF9 fusion protein in safflower (Carthamus tinctorius L.) seeds stimulates hair growth and wound healing. Results The oleosin-rhFGF9 expressed in safflower seeds, in which it localizes to the surface of oil bodies. The expression of oleosin-rhFGF9 was confirmed by polyacrylamide gel electrophoresis and western blotting. According to BCA and Enzyme-linked immunosorbent assay (ELISA) assay, the results show that the expression level of oleosin-rhFGF9 was 0.14% of oil body protein. The oil body bound oleosin-rhFGF9 showed mitogenic activity towards NIH3T3 cells in a methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay. The efficacy of oil body bound oleosin-rhFGF9 in promoting hair growth and wound healing was investigated in C57BL/6 mice. In a hair regeneration experiment, 50 μg/μl oil body bound oleosin-rhFGF9 was applied to the dorsal skin of mice in the resting phase of the hair growth cycle. After 15 days, thicker hair and increased number of new hairs were seen compared with controls. Furthermore, the number of new hairs was greater compared with rhFGF9-treated mice. The hair follicles of mice treated with oil body bound oleosin-rhFGF9 expressed β-catenin more abundantly. In a wound healing experiment, dorsal skin wounds were topically treated with 50 μg/μl oil body bound oleosin-rhFGF9. Wound healing was quicker compared with mice treated with rhFGF9 and controls, especially in the earlier stages of healing. Conclusions The oil body bound oleosin-rhFGF9 promotes both hair growth and wound healing. It appears to promote hair growth, at least in part, by up-regulating β-catenin expression. The potential of oil body bound oleosin-rhFGF9 as an external drug can treat the alopecia and wounds or use in further clinical application.

Abstract Background Epidermal growth factor (EGF) can promote cell proliferation as well as migration, which is feasible in tissue wound healing. Oil bodies have been exploited as an important platform to produce exogenous proteins. The exogenous proteins were expressed in oil bodies from plant seeds. The process can reduce purification steps, thereby significantly reducing the purification cost. Mostly, the diameter of oil body particle ranges between 1.0 and 1.5 µm in the safflower seeds, however, it reduces to 700–1000 nm in the transgenic safflower seeds. The significant reduction of particle size in transgenic seeds is extremely beneficial to skin absorption. Results The diameter of oil body in the transgenic safflower seeds was recorded in the range of 700–1000 nm. The smaller particle size improved their skin absorption. The expression level of oleosin-hEGF-hEGF in T3 transgenic seeds was highest at 69.32 mg/g of seeds. The oil body expressing oleosin-hEGF-hEGF had significant proliferative activity on NIH/3T3 cells and improved skin regeneration thereby accelerating wound healing in rats. The wound coverage rate exceeded 98% after treatment for 14 days with oil body expressing oleosin-hEGF-hEGF, while the saline without EGF group and wild type oil body group both showed less than 80%. The neonatal fibroblast and collagen were found to be increased in the safflower oil body expressing oleosin-hEGF-hEGF treatment group. TGF-β1, bFGF and VEGF were noted as important growth factors in the repair of cutaneous wounds. Their expression level increased after 4 and 7 day treatment, but decreased after 14 days. Therefore, it can promote skin regeneration to accelerate wounds healing. Conclusions The expression of oleosin-hEGF-hEGF in T3 transgenic seeds was 80.43 ng/μL oil body. It had significant proliferative activity on NIH/3T3 cells and improved skin regeneration to accelerate wound healing in rats. The expression process of TGF-β1, bFGF and VEGF increased at first and then gradually declined.

Abstract In this paper, we fabricated a series of FeCoBSi multistoried patterned magnetic films with different thickness by traditional UV lithography method and DC sputtering deposition. Broad resonance band phenomenon was observed during high frequency property characterization, with full width half maximum (FWHM) of 4 GHz when the film thickness is 45 nm. The broad resonance band effect was contributed to the existence of multiple resonance peaks due to different stripe width of the combined stripe pattern, which induced distinguish shape anisotropic field in each stripe. Each resonance peak was independent due to the gap between the stripes, leading to a controllable method to tune the microwave properties of such structure. With thickness varied, the resonance band could be altered according to the mathematic prediction. This work presents an effective method for tuning the microwave resonance characterization in magnetization dynamic.

Ultra-stable optical cavities (USOCs) as fragile precision instruments have many important applications in space. In order to protect them from being damaged during a rocket launch, we analyzed a USOC by means of finite element methodology. The shock acceleration limits that the USOC can withstand in different directions and under various conditions are given. To increase the shock acceleration limit, the midplane thickness and the fixed hole diameter should be selected to be as high as possible. It is worth noting that the launch direction of the USOC should be selected as the horizontal direction, for which the shock acceleration limit that the USOC can withstand is approximately two times that of the vertical direction. In this paper, results provide guidance for the design of USOCs for space applications, especially the design to prevent the damage caused by a shock. The method could then be applied to other space optical cavities, providing a tool to improve the effect of shock at high accelerations.

Thermal noise in optical cavities sets a fundamental limit to the frequency instability of ultra-stable lasers. Numata et al. derived three equations based on strain energy and the fluctuation–dissipation theorem to estimate the thermal noise contributions of the spacer, substrates, and coating. These equations work well for cylindrical cavities. Extending from that, an expression for the thermal noise for a cubic spacer based on the fluctuation–dissipation theorem is derived, and the thermal noise in cubic optical cavities is investigated in detail by theoretical analysis and finite element simulation. The result shows that the thermal noise of the analytic estimate fits well with that of finite element analysis. Meanwhile, the influence of the compressive force Fp on the thermal noise in cubic optical cavities is analyzed for the first time. For a 50 mm long ultra-low expansion cubic cavity with fused silica substrates and GaAs/AlGaAs crystalline coating, the displacement noise contributed from every Fp of 100 N is about three times more than that of the substrate and coating.

A polarization-insensitive recirculating delayed self-heterodyne method (PI-RDSHM) is proposed and demonstrated for the precise measurement of sub-kilohertz laser linewidths. By a unique combination of Faraday rotator mirrors (FRMs) in an interferometer, the polarization-induced fading is effectively reduced without any active polarization control. This passive polarization-insensitive operation is theoretically analyzed and experimentally verified. Benefited from the recirculating mechanism, a series of stable beat spectra with different delay times can be measured simultaneously without changing the length of delay fiber. Based on Voigt profile fitting of high-order beat spectra, the average Lorentzian linewidth of the laser is obtained. The PI-RDSHM has advantages of polarization insensitivity, high resolution, and less statistical error, providing an effective tool for accurate measurement of sub-kilohertz laser linewidth.

The National Time Service Center of China is developing a compact, highly stable, 698 nm external-cavity diode laser (ECDL) for dedicated use in a space strontium optical clock. This article presents the optical design, structural design, and preliminary performance of this ECDL. The ECDL uses a narrow-bandwidth interference filter for spectral selection and a cat’s-eye reflector for light feedback. To ensure long-term stable laser operation suitable for space applications, the connections among all the components are rigid and the design avoids any spring-loaded adjustment. The frequency of the first lateral rocking eigenmode is 2316 Hz. The ECDL operates near 698.45 nm, and it has a current-controlled tuning range over 40 GHz and a PZT-controlled tuning range of 3 GHz. The linewidth measured by the heterodyne beating between the ECDL and an ultra-stable laser with 1 Hz linewidth is about 180 kHz. At present, the ECDL has been applied to the principle prototype of the space ultra-stable laser system.

In this research, monthly hydrological and daily meteorological data were collected across China for the period 1956–2012. Modified Mann–Kendall tests, double mass curve analysis, and correlation statistics were performed to identify the long-term trends and interrelation of the hydrometeorological variables and to examine the influencing factors of streamflow and sediment. The results are as follows: (1) In the last 60 years, the streamflow in northern China has shown different decreasing trends. For the southern rivers, the streamflow presented severe fluctuations, but the declining trend was insignificant. For the streamflow in western China, an increasing trend was shown. (2) In the northern rivers, the streamflow was jointly controlled by the East Asian monsoon and westerlies. In the southern rivers, the runoff was mainly influenced by the Tibet–Qinghai monsoon, the South Asian monsoon, and westerlies. (3) Sediment loads in the LCRB (Lancang River Basin) and YZRB (Yarlung Zangbo River Basin) did not present significant change trends, although other rivers showed different degrees of gradual reduction, particularly in the 2000s. (4) Underlying surface and precipitation changes jointly influenced the streamflow in eastern rivers. The water consumption for industrial and residential purposes, soil and water conservation engineering, hydraulic engineering, and underlying surface changes induced by other factors were the main causes of streamflow and sediment reduction.

RsmI and RsmH are conserved S-Adenosylmethionine (AdoMet)-dependent methyltransferases (MTases) that are responsible for the 2'-O-methylation and N4-methylation of C1402 in bacterial 16S rRNA, respectively. Methylation of m4Cm1402 plays a role in fine-tuning the shape and functions of the P-site to increase the decoding fidelity, and was recently found to contribute to the virulence of Staphylococcus aureus in host animals. Here we report the 2.20-Å crystal structure of homodimeric RsmI from Escherichia coli in complex with the cofactor AdoMet. RsmI consists of an N-terminal putative RNA-binding domain (NTD) and a C-terminal catalytic domain (CTD) with a Rossmann-like fold, and belongs to the class III MTase family. AdoMet is specifically bound into a negatively charged deep pocket formed by both domains by making extensive contacts. Structure-based mutagenesis and isothermal titration calorimetry (ITC) assays revealed Asp100 and Ala124 are vital for AdoMet-binding. Although the overall fold of RsmI shows remarkable similarities to the characterized MTases involved in vitamin B12 biosynthesis, it exhibits a distinct charge distribution especially around the AdoMet-binding pocket because of different substrate specificity. The docking model of RsmI-AdoMet-RNA ternary complex suggested a possible base-flipping mechanism of the substrate RNA that has been observed in several known RNA MTases. Our structural and biochemical studies provide novel insights into the catalytic mechanism of C1402 methylation in 16S rRNA.

Recombinant human fibroblast growth factor 10 (rhFGF-10) is frequently used to treat patients with skin injuries. It can also promote hair growth. However, the effective application of rhFGF-10 is limited because of its poor stability and transdermal absorption. In this study, polymerase chain reaction (PCR) and Southern blotting were used to identify transgenic safflowers carrying a gene encoding an oleosin-rhFGF-10 fusion protein. The size and structural integrity of oleosin-rhFGF-10 in oil bodies extracted from transgenic safflower seeds was characterized by polyacrylamide gel electrophoresis and western blotting. Oil body extracts containing oleosin-rhFGF-10 were topically applied to mouse skin. The absorption of oleosin-rhFGF-10 was studied by immunohistochemistry. Its efficiency in promoting wound healing and hair regeneration were evaluated in full thickness wounds and hair growth assays. We identified a safflower line that carried the transgene and expressed a 45 kDa oleosin-rhFGF-10 protein. Oil body-bound oleosin-rhFGF-10 was absorbed by the skin with higher efficiency and speed compared with prokaryotically-expressed rhFGF-10. Oleosin-rhFGF-10 also enhanced wound closure and promoted hair growth better than rhFGF-10. The application of oleosin-rhFGF-10 in oil bodies promoted its delivery through the skin, providing a basis for improved therapeutic effects in enhancing wound healing and hair growth.

BACKGROUND: Emerging evidence showed the beneficial effect of acidic fibroblast growth factor (aFGF) on heart diseases. The present study investigated whether non-mitogenic aFGF (nm-aFGF) can prevent diabetic cardiomyopathy and the underlying mechanisms, if any. METHODOLOGY/PRINCIPAL FINDINGS: Type 1 diabetes was induced in mice by multiple intraperitoneal injections of low-dose streptozotocin. Hyperglycemic and age-matched control mice were treated with or without nm-aFGF at 10 µg/kg daily for 1 and 6 months. Blood pressure and cardiac function were assessed. Cardiac H9c2 cell, human microvascular endothelial cells, and rat cardiomyocytes were exposed to high glucose (25 mM) for mimicking an in vitro diabetic condition for mechanistic studies. Oxidative stress, DNA damage, cardiac hypertrophy and fibrosis were assessed by real-time qPCR, immunofluorescent staining, Western blotting, and pathological examination. Nm-aFGF significantly prevented diabetes-induced hypertension and cardiac dysfunction at 6 months. Mechanistic studies demonstrated that nm-aFGF showed the similar preventive effect as the native aFGF on high glucose-induced oxidative stress (increase generation of reactive oxygen species) and damage (cellular DNA oxidation), cell hypertrophy, and fibrotic response (increased mRNA expression of fibronectin) in three kinds of cells. These in vitro findings were recaptured by examining the heart of the diabetic mice with and without nm-aFGF. CONCLUSIONS: These results suggest that nm-aFGF can prevent diabetic cardiomyopathy, probably through attenuation of cardiac oxidative stress, hypertrophy, and fibrosis.

To explore the renal protection mechanism of targeted regulating of transforming growth factor beta receptor type 1 expression by let-7a in diabetic nephropathy rats, diabetic nephropathy rats were created through high-sugar and high-fat diet combined with a small dose of streptozotocin. Normally reared rats were taken as control group. Diabetic nephropathy rats were divided into two groups, one group was treated with let-7a analogs group, and the other group was not treated (diabetic nephropathy group). Experimental results showed that diabetic nephropathy rats had symptoms such as increased dietary frequency, weight loss, proteinuria, and decreased creatinine clearance. After let-7a treatment, the proteinuria symptoms of rats in let-7a analogs group were notably improved, and the creatinine clearance rate increased. The expression level of collagen IV in let-7a analogs group was higher than that of the control, but the difference was not significant. The expression level of fibronectin increased significantly in the diabetic nephropathy group (p<0.05), and that of the let-7a analogs group was significantly lower than that of the diabetic nephropathy group (p<0.05). The immunofluorescence expression result was consistent with the expression trend detected by Western blot. Compared with the control, the expressions of transforming growth factor-beta 1, transforming growth factor beta receptor type 1, and mothers against decapentaplegic homolog 2 in the diabetic nephropathy group were evidently increased (p<0.05), and the expression of mothers against decapentaplegic homolog 7 decreased obviously (p<0.05). The protein expression level was consistent with the messenger ribonucleic acid expression result. In summary, by regulating the levels of collagen IV and fibronectin and targeting the transforming growth factor beta receptor type 1 pathway, let-7a inhibited extracellular matrix synthesis to protect kidney function in patients with diabetic nephropathy.
