Abstract The study aimed to compare the effectiveness between Hiporfin-photodynamic therapy (PDT) and loop electrosurgical excision procedure (LEEP) in treating cervical high-grade squamous intraepithelial lesion (HSIL). We conducted a retrospective analysis of 104 women diagnosed with cervical HSIL at our hospital from April 2019 to July 2023. Abstract Lung adenocarcinoma (LUAD) is the most common and aggressive subtype of lung cancer, and coronavirus disease 2019 (COVID-19) has become a serious public health threat worldwide. Patients with LUAD and COVID-19 have a poor prognosis. Therefore, finding medications that can be used to treat COVID-19/LUAD patients is essential. Bioinformatics analysis was used to identify 20 possible metformin target genes for the treatment of COVID-19/LUAD. PTEN and mTOR may serve as hub target genes of metformin. Metformin may be able to cure COVID-19/LUAD comorbidity through energy metabolism, oxidoreductase NADH activity, FoxO signalling pathway, AMPK signalling system, and mTOR signalling pathway, among other pathways, according to the results of bioinformatic research. Metformin has ability to inhibit the proliferation of A549 cells, according to the results of colony formation and proliferation assays. In A549 cells, metformin increased glucose uptake and lactate generation, while decreasing ATP synthesis and the NAD+/NADH ratio. In summary, PTEN and mTOR may be potential targets of metformin for the treatment of COVID-19/LUAD. The mechanism by which metformin inhibits lung adenocarcinoma cell proliferation may be related to glucose metabolism regulated by PI3K/AKT signalling and mTOR signalling pathways. Our study provides a new theoretical basis for the treatment of COVID-19/LUAD.

The rapid progress in point-of-care testing (POCT) has become a promising decentralized patient-centered approach for the control of infectious diseases, especially in resource-limited settings. POCT devices should be inexpensive, rapid, simple operation and preferably require no power supply. Here, we developed a simple bacterial sensing platform that can be operated by a smartphone for bacteria identification and antimicrobial susceptibility testing (AST) based on using a polydiacetylene (PDA) arrayed membrane chip. Each PDA array produced a unique color ‘fingerprint’ pattern for each bacteria based on different modes of action of toxins from bacteria on biomimetic lipid bilayers within PDA-lipid assemblies. We show that the PDA-based device can detect viable cells of bacteria as low as 104 ​CFU/mL within 1.5 ​h compared with several days of conventional bacterial identification, with the aid of a smartphone app. The device can also be used for an antimicrobial susceptibility test (AST) for at least two broad-spectrum antimicrobials within 4 ​h and provide identification of antimicrobial susceptibility and resistance, enabling the selection of appropriate therapies. This PDA-based sensing platform provides an alternative way for bacterial detection and could be used as a portable and inexpensive POCT device for the rapid detection of bacterial infection in limited-resource settings.

This article proposes a switched-coupled-inductor impedance source network integrating capacitor-winding-diode technology. Next, voltage-boost switched-coupled-inductor dc–dc converters based on the proposed impedance source network are deduced out. The proposed dc–dc converters can obtain higher voltage gain and own common ground. Also, input currents of the proposed converters are continuous, which is beneficial of input power. Compared to conventional impedance source dc–dc converters, the proposed converters can have the higher boost ability and largely reduce the voltage stresses across switches in the same voltage gain. Theoretical analysis including operation principles, voltage gain derivations, stress analysis, parameter design, efficiency analysis, and feature comparisons of the proposed topologies are given. The specific experimental results are shown to verify the aforementioned analysis.

Carbon dots doped with boron and nitrogen atoms (B, N-CDs) were designed and prepared by hydrothermal strategy using citric acid, urea and borax as raw materials. The structure and optical properties of B, N-CDs were characterized by various characterization technologies. The results showed that the B, N-CDs exhibited good mono-dispersion, excellent fluorescence stability, and high fluorescence intensity (quantum yield up to 32 %). A dual-functional Fe3+ and pH detection platform based on B, N-CDs was constructed. The Fe3+ sensing method based on B, N-CDs fluorescence quenching showed an excellent linear relationship between 0.044–70 μM (R2 = 0.994), with a detection limit as low as 0.044 μM. Then, this detection method was successfully applied to the high-accuracy detection of Fe3+ in water samples and iron supplement tablets. The study of the quenching mechanism showed that the fluorescence of B, N-CDs was quenched by Fe3+ in two ways: static quenching and dynamic quenching. Moreover, B, N-CDs can also be used as a pH fluorescence sensor in the ranges of pH 2.0–4.8 and 5.2–7.6. The developed sensing system based on B, N-CDs has high selectivity and sensitivity for the detection of Fe3+, and a wide detection range for pH.

Abstract Objectives This study aimed to identify the related risk factors and potential predictors of SARS-CoV-2 RNA negative conversion by describing the dynamics of viral shedding in infected children admitted to two hospitals from Shanghai during the Omicron variant outbreak. Methods This retrospective cohort included laboratory-confirmed cases of SARS-CoV-2 infection from Shanghai between March 28 and May 31, 2022. Clinical characteristics, personal vaccination, and household vaccination rates were collected through electronic health records and telephone interviews. Results A total of 603 paediatric patients confirmed to have COVID-19 were included in this study. Both univariate and multivariate analyses were performed to filter independent factors for the duration to viral RNA negative conversion. Data on the redetection of SARS-CoV-2 in the patients after they showed negative results on the RT‒PCR test (intermittent negative status) were also analysed. The median duration of virus shedding was 12 (interquartile range, IQR: 10–14) days. The severity of clinical outcome, personal vaccination-2doses, household vaccination rates, and abnormal defecation were factors indecently affecting negative conversion of SARS-CoV-2 RNA, suggesting that patients who had abnormal defecation or with more severe conditions would have delayed virological clearance, while patients who previously had 2 doses of vaccination or had higher household vaccination rates would have accelerated virological clearance. Loss of appetite (odds ratio (OR): 5.343; 95% CI: 3.307–8.632) and abnormal defecation (OR: 2.840; 95% CI: 1.736–4.645) were significantly associated with intermittent negative status. Conclusion These findings could provide clues for the early identification of paediatric patients with prolonged viral shedding and could enrich the evidence for the development of prevention and control strategies, especially vaccination policies for children and adolescents.

Nitrogen (N) doped CDs (N-CDs) were prepared using citric acid and hexamethylenetetramine by a one-step hydrothermal method, characterized by transmission electron microscopy (TEM), Powder X-ray Diffractometer (PXRD), Nuclear Magnetic Resonance (13C NMR), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared (FT-IR), and ultraviolet–visible (UV–vis) absorption spectra. The doping mechanism and drug-loading characteristics of N-CDs were also studied. The results showed that N-CDs had both down- and up-conversion fluorescence properties and the average particle size was about 4.28 nm. N-CDs could be used as a drug carrier for doxorubicin (DOX) (N-CDs-DOX) via the formation of the NC bond, and the drug-loading rate of N-CDs-DOX was 35.88%. The N-CDs-DOX had pH-targeted properties and low cytotoxicity. In vitro release of N-CDs-DOX belonged to the Weibull model and Fick diffusion. The results showed that N-CDs could deliver DOX successfully, and collocate with DOX in the SH-SY5Y cells through two-photon imaging. The N-CDs-DOX system had sustained-release and pH-targeted properties. N-CDs were expected to deliver a type of drugs containing CO group and be used in cancer treatment.

Heteroatom-doping carbon dots (CDs) have excellent fluorescence properties. In the paper, nitrogen (N) and phosphorus (P) co-doped CDs (N, P-CDs) were prepared through the hydrothermal method and characterized by TEM, PXRD, FT-IR, 13C NMR, XPS, UV–vis, and fluorescence spectra. The optical properties and drug (doxorubicin) - delivery performance of N, P-CDs were also studied. The results showed that the average size of N, P-CDs was 3.64 nm and the quantum yield was 30 %. The N, P-CDs had down- and up-conversion fluorescence properties, and the luminescence mechanism of the N, P-CDs was explained. The drug loading capacity of N, P-CDs was 39.11 %, and the drug delivery system (N, P-CDs-DOX) had low cytotoxicity, sustained release, and pH-targeted properties. The in vitro release of N, P-CDs-DOX belonged to the Weibull model and Fick diffusion, exhibiting the same release model and mechanism with free DOX. The N, P-CDs could deliver DOX successfully, which was demonstrated by the co-collocation of N, P-CDs and DOX in SH-SY5Y cells through single/two-photon imaging. N, P-CDs could potentially be used in drug-targeted delivery and cell imaging.

In this paper, an improved coupled-inductor impedance source network (CL-ISN) with more freedom in winding match is proposed. Through introducing three-winding coupled-inductor cells, the proposed CL-ISN can produce the same voltage gain by using different turn ratios among three windings, which will largely increase the freedom in winding match. Also, the proposed CL-ISN can produce the higher boost ability, realize the continuous input current and suppress the start-up inrush current. The topology derivations, math calculations and parameter design of proposed CL-ISN are given out. The proposed CL-ISN is applied for DC-AC converter, and a full comparison between proposed CL-ISN and other impedance source networks are made based on DC-AC converter. Finally, the experiment prototype is built to verify the validity of the theoretical analysis.

A novel fault tolerant machine with integral slot non-overlapping concentrated winding (ISNCW) is proposed in this paper. With the distribution of phase windings in axial, the proposed machine realized excellent isolation capability between different phases. The performance comparisons between the proposed machine and the traditional integer slot overlapping winding (ISOW) machine are made by the analytical method and finite element analysis (FEA). Compared to the ISOW machine, the proposed machine has better performance in low speed high torque applications, and much better fault tolerant capability and flux weakening capability. Especially, the proposed machine appears excellent performance in limiting the short circuit current and preventing the irreversible demagnetization of permanent magnet under short-circuit fault. The phase independent structure makes the maintenance and replacement of the fault module convenient and saves the maintenance cost further.

A 9-week growth trial was carried out to assess the influence of replacing poultry by-product meal protein with conventional cottonseed meal protein (CCMP) or low gossypol cottonseed meal protein (LGCMP) on growth, feed utilization, gut micromorphology, and immunity of hybrid grouper (Epinephelus fuscoguttatus♀× Epinephelus lanceolatus♂) juveniles fed low-fish meal (18.53%, dry matter) diets. Eleven experimental diets were prepared. The control diet (PBMP) contained 46.15% poultry by-product meal protein. Both conventional cottonseed meal protein (CCMP) and low-gossypol cottonseed meal protein (LGCMP) were used in replacement ratios of 20, 40, 60, 80, and 100% of poultry by-product meal protein (PBMP) from the control diet, forming ten experimental diets (CCMP20, CCMP40, CCMP60, CCMP80, CCMP100, LGCMP20, LGCMP40, LGCMP60, LGCMP80, and LGCMP100). Results demonstrated that weight-gain percentage (WG%) was not different between different sources of cottonseed meal (CCMP and LGCMP). However, values of WG% significantly differed among different replacement levels, with CCMP80 and LGCMP40 having significantly higher values compared to other treatments. Fish fed CCMP80 and LGCMP40 exhibited higher protein efficiency ratios (PERs) than fish fed other experimental diets. The regression analysis from a second-order or third-order polynomial model based on WG% showed that the optimal PBMP replacement levels by CCMP and LGCMP are 74% and 33%, respectively. The whole-body lipid contents remarkably decreased as dietary CCMP or LGCMP inclusion levels increased. The relative mRNA expression of insulin-like growth factor-1(IGF-1) in liver was higher in fish fed CCMP80 and LGCMP40 diets compared to fish fed other diets. Generally, in low-FM diets of hybrid grouper, CCMP and LGCMP could replace 74% and 33% of PBMP, respectively.

Signaling lymphocytic activation molecule (SLAM) and SLAM-associated protein (SAP) play important role in inflammatory and autoimmune diseases. Our study is aimed at detecting the expression of SLAM and SAP in patients with Graves’ disease (GD) and analyzing the effect of SLAM/SAP on circulating blood CD4+CXCR5+Foxp3+ follicular regulatory T (Tfr) cells. The level of SAP in CD4+CXCR5+ T cells and the level of SLAM on CD19+ B cells were significantly increased in the patients with GD, but no significant difference in the level of SLAM on CD4+CXCR5+ T cells was observed between the patients with GD and the healthy controls. A decrease in the percentage of Foxp3+ cells in CD4+CXCR5+ T cells was observed following anti-SLAM treatment, but the percentages of IFN-γ+ cells, IL-4+ cells, and IL-17+ cells showed no obvious differences. The proportion of circulating Tfr cells was decreased in the patients with GD, and the proportion of circulating Tfr cells had a negative correlation with the level of SAP in CD4+CXCR5+ T cells and the levels of autoantibodies in the serum of the patients with GD. Our results suggested that the SLAM/SAP signaling pathway is involved in the decrease of circulating Tfr cells in Graves’ disease.

Abstract Background Iron deficiency in children can have significant neurological consequences, and iron supplementation is an effective treatment of choice. However, traditional routes of iron supplementation do not allow efficient iron delivery to the brain due to the presence of the blood–brain barrier. So an easily delivered iron formulation with high absorption efficiency potentially could find widespread application in iron deficient infants. Results In this study, we have developed and characterized a nanovesicular formulation of ferric ammonium citrate (ferric ammonium citrate nanoliposomes, FAC-LIP) and have shown that it can increase brain iron levels in rats following nasal administration. FAC was incorporated into liposomes with high efficiency (97%) and the liposomes were small (40 nm) and stable. Following intranasal delivery in rats, FAC-LIP significantly increased the iron content in the olfactory bulb, cerebral cortex, striatum, cerebellum and hippocampus, and was more efficient at doing so than FAC alone. No signs of apoptosis or abnormal cell morphology were observed in the brain following FAC-LIP administration, and there were no significant changes in the levels of SOD and MDA, except in the cerebellum and hippocampus. No obvious morphological changes were observed in lung epithelial cells or tracheal mucosa after nasal delivery, suggesting that the formulation was not overtly toxic. Conclusions In this study, nanoscale FAC-LIP proved an effective system delivering iron to the brain, with high encapsulation efficiency and low toxicity in rats. Our studies provide the foundation for more detailed investigations into the applications of niosomal nasal delivery of liposomal formulations of iron as a simple and safe therapy for iron deficiency anemia. Nanoliposome-encapsulated ferric ammonium citrate (FAC-LIP) was successfully prepared and intranasal administration of FAC-LIP increased both the total iron contents and iron storage protein (FTL) expression in rat olfactory bulb, cerebral cortex, striatum and hippocampus, compared with those of FAC groups. Moreover, there was not overtly toxic affects to brain, lung epithelial cells and tracheal mucosa.

Our previous study found that ferric citrate-loaded liposome (FAC-LIP) exhibited stable physical characteristics and have better efficacy to treat anemia of inflammation (AI) than free ferric citrate (FAC). This study was aimed at oral pharmacokinetics of both FAC-LIP and FAC in Sprague Dawley (SD) rats. Results showed that the main pharmacokinetic parameters of FAC-LIP in vivo were as follows: Cmax of 445.89 ± 23.56 μg/mL, Tmax of 4 h, t1/2 of 13.01 ± 2.04 h, MRT of 20.54 ± 2.06 h and AUC0-24 h of 6446.1 ± 516.58 hμg/mL. The relative bioavailability of FAC-LIP formulation was 215.7%, nearly 2-fold in comparison with free FAC formulation. FAC-LIP showed a sustained-release effect, a wide absorption and a complete elimination. These results support the fact that FAC-LIP has the potential of improving oral bioavailability and reducing iron accumulation in liver, which could influence the available iron level in the blood system. FAC-LIP could be used as an effective iron supplement.

To understand the mechanism of the dehydrogenation of methanol to CO and H2 catalyzed by a ruthenium pincer complex, a density functional theory (DFT) study has been conducted on two different cycles which differ in the substances entering the cycle (methanol (cycle 1) versus methoxymethanol (cycle 2)). Our calculated results show that both cycles consist of three stages: dehydrogenation of alcohol to aldehyde (stage I); hydrogen formation (stage II); and decarbonylation with the regeneration of the catalyst (stage III). The energy barriers of the rate-determining steps for cycles 1 and 2 are 49.6 and 28.5 kcal mol-1, respectively. Thus cycle 2 is more energetically feasible. For stage III of cycle 2, our results did not support the mechanism proposed in the experiment (CO release occurs prior to decarbonylation). Instead, we suggested and examined an alternative pathway, that is, decarbonylation occurs prior to CO release.
