Abstract Liver transplantation (LT) rejection remains the most pervasive problem associated with this procedure, while the mechanism involved is still complicated and undefined. One promising solution may involve the use of myeloid-derived suppressor cells (MDSC). However, the immunological mechanisms underlying the effects of MDSC after LT remain unclear. This study is meant to clarify the role MDSCs play after liver transplantation. In this study, we collected liver tissue and peripheral blood mononuclear cells (PBMC) from LT patients showing varying degrees of rejection, as well as liver and spleen tissue samples from mice LT models. These samples were then analyzed using flow cytometry, immunohistochemistry and multiple immunofluorescence. M-MDSCs and CD8 + T-cells extracted from C57/BL6 mice were enriched and cocultured for in vitro experiments. Results, as obtained in both LT patients and LT mice model, revealed that the proportion and frequency of M-MDSC and PD-1 + T-cells increased significantly under conditions associated with a high degree of LT rejection. Within the LT rejection group, our immunofluorescence results showed that a close spatial contiguity was present between PD-1 + T-cells and M-MDSCs in these liver tissue samples and the proportion of CD84/PD-L1 double-positive M-MDSC was greater than that of G-MDSC. There was a positive correlation between the activity of CD84 and immunosuppressive function of M-MDSCs including PD-L1 expression and reactive oxygen species (ROS) production, as demonstrated in our in vitro model. M-MDSCs treated with CD84 protein were able to induce co-cultured CD8 + T-cells to express high levels of exhaustion markers. We found that CD84 regulated M-MDSC function via expression of PD-L1 through activation of the Akt/Stat3 pathway. These results suggest that the capacity for CD84 to regulate M-MDSC induction of CD8 + T-cell exhaustion may play a key role in LT rejection. Such findings provide important, new insights into the mechanisms of tolerance induction in LT.

The aim of this study is to enhance the carbonation resistance of fully recycled aggregate concrete through diverse measures in an effort to enhance solid waste disposal, reduce the consumption of natural aggregates, and broaden the utilization of recycled aggregate concrete. Six sets of fully recycled aggregate concrete specimens were prepared and subjected to rapid carbonation tests. Carbonation depth and compressive strength measurements were taken at different ages (3, 7, 14, and 28 days). Subsequent calculations and analyses were conducted on both parameters for each set of specimens. Results indicate that the incorporation of microspheres and high-toughness polypropylene fibers (HTPP) substantially improves the carbonation resistance of fully recycled aggregate concrete, leading to a 48% reduction in carbonation depth by the 28th day. Furthermore, a relative compressive strength model for fully recycled aggregate concrete post-carbonation was established based on the strength data of each specimen group. This model accurately depicts the growth pattern of compressive strength after carbonation. Additionally, a carbonation depth prediction model was developed through fitting analysis of carbonation depth data, effectively foreseeing the depth of carbonation in fully recycled aggregate concrete. Based on the carbonation depth, the carbonation life of fully recycled aggregate concrete was predicted. The carbonation life of recycled aggregate concrete with added microspheres and HTPP fibers can be increased by up to 278%. Finally, scanning electron microscopy (SEM) was employed to examine the microstructure of fully recycled aggregate concrete, revealing the mechanisms by which various methods enhance its carbonation resistance. The carbonation resistance improvement technology of fully recycled aggregate concrete is selected through this study characteristics such as simplicity, convenience, and cost-effectiveness, which are crucial for the widespread application of recycled aggregate concrete in building structures.

Zero-dimensional heterometallic halide hybrids have emerged as multifunctional emitters, due to their multiple emission features. Herein, high pressure is employed to regulate the multiple emissions of (C9NH20)9[Pb3Br11](MnBr4)2. Pressure-triggered rigidness of [Pb3Br11]5- alters the emission equilibrium of STEs, generating a new blue-shifted STE emission. Meanwhile, [MnBr4]2- contraction promotes the confinement effect to increase the 4D-6A1 transition efficiency. Notably, [Pb3Br11]5- could act as ‘antenna' to enhance the Mn2+ emission with blocked back-energy transfer. It indicates that enough incident energy could excite the STE emission and 4D-6A1 transitions simultaneously, which clarifies the conventional understanding of the emission origin at ambient conditions.

Abstract The aging process of the kidneys is accompanied with several structural diseases. Abnormal fiber formation disrupts the balance of kidney structure and function, causing to end-stage renal disease and subsequent renal failure. Despite this, the precise mechanism underlying renal damage in aging remains elusive. In this study, ABI3BP gene knockout mice were used to investigate the role of ABI3BP in renal aging induced by irradiation. The results revealed a significant increase in ABI3BP expression in HK2 cells and kidney tissue of aging mice, with ABI3BP gene knockout demonstrating a mitigating effect on radiation-induced cell aging. Furthermore, the study observed a marked decrease in Klotho levels and an increase in ferroptosis in renal tissue and HK2 cells following irradiation. Notably, ABI3BP gene knockout not only elevated Klotho expression but also reduced ferroptosis levels. A significant negative correlation between ABI3BP and Klotho was established. Further experiments demonstrated that Klotho knockdown alleviated the aging inhibition caused by ABI3BP downregulation. This study identifies the upregulation of ABI3BP in aged renal tubular epithelial cells, indicating a role in promoting ferroptosis and inducing renal aging by inhibiting Klotho expression.

Abstract Rhenium (Re) and uranium (U) are essential proxies in reconstructing past oceanic oxygenation evolution. However, their removal in continental shelf sediments, hotspots of early diagenesis, were previously treated as quantitatively unimportant sinks in the ocean. Here we examine the sedimentary reductive removal of Re and U and their coupling with organic carbon decomposition, utilizing the 224Ra/228Th disequilibria within the East China Sea shelf. We identified positive correlations between their removal fluxes and the rates of sediment oxygen consumption or organic carbon decomposition. These correlations enable an evaluation of global shelf reductive sinks that are comparable to (for Re) or higher than (~4-fold for U) previously established suboxic/anoxic sinks. These findings suggest potential imbalances in the modern budgets of Re and U, or perhaps a substantial underestimation of their sources. Our study thus highlights shelf sedimentary reductive removal as critical yet overlooked sinks for Re and U in the modern ocean.

Abstract Background Doxorubicin (Dox) is associated with various liver injuries, limiting its clinical utility. This study investigates whether NSUN2 participates in Dox-induced liver injury and the associated molecular mechanism. Methods In vivo and in vitro liver cell injury models were constructed based on Dox therapy. The protein levels of NSUN2 and oxidative stress indicators Nrf2, HO-1, and NQO1 were evaluated by Western blot. The RNA binding potential was detected by RNA methylation immunoprecipitation (RIP). Additionally, the effect of NSUN2 on Nrf2 mRNA synthesis and localization was evaluated using an RNA fluorescence probe. Results NSUN2 was downregulated, and liver tissue suffered significant pathological damage in the Dox group. The levels of ALT and AST significantly increased. NSUN2 interference exacerbated Dox-induced liver cell damage, which was reversed by NSUN2 overexpression. RIP demonstrated that NSUN2 recognized and bound to Nrf2 mRNA. Western blot analysis showed the protein level of Nrf2 in the NSUN2-WT group was significantly higher than that of the control group, whereas there was no significant change in Nrf2 level in the mutant NSUN2 group. Luciferase analysis demonstrated that NSUN2 could recognize and activate the Nrf2 5′UTR region of LO2 cells. In addition, RIP analysis revealed that ALYREF could recognize and bind to Nrf2 mRNA and that ALYREF controls the regulatory effect of NSUN2 on Nrf2. Conclusion NSUN2 regulates Dox-induced liver cell damage by increasing Nrf2 mRNA m5C methylation to inhibit inhibiting antioxidant stress. The regulatory effect of NSUN2 on Nrf2 depends on ALYREF.

IntroductionFusarium wilt (FW) caused by Fusarium oxysporum f. sp. cucumerinum (Foc) is a destructive soil-borne disease in cucumber (Cucumis sativus. L). However, there remains limited knowledge on the molecular mechanisms underlying FW resistance-mediated defense responses in cucumber.MethodsIn this study, metabolome and transcriptome profiling were carried out for two FW resistant (NR) and susceptible (NS), near isogenic lines (NILs) before and after Foc inoculation. NILs have shown consistent and stable resistance in multiple resistance tests conducted in the greenhouse and in the laboratory. A widely targeted metabolomic analysis identified differentially accumulated metabolites (DAMs) with significantly greater NR accumulation in response to Foc infection, including many phenolic acid and flavonoid compounds from the flavonoid biosynthesis pathway.ResultsTranscriptome analysis identified differentially expressed genes (DEGs) between the NILs upon Foc inoculation including genes for secondary metabolite biosynthesis and transcription factor genes regulating the flavonoid biosynthesis pathway. Joint analysis of the metabolomic and transcriptomic data identified DAMs and DEGs closely associated with the biosynthesis of phenolic acid and flavonoid DAMs. The association of these compounds with NR-conferred FW resistance was exemplified by in vivo assays. These assays found two phenolic acid compounds, bis (2-ethylhexyl) phthalate and diisooctyl phthalate, as well as the flavonoid compound gallocatechin 3-O-gallate to have significant inhibitory effects on Foc growth. The antifungal effects of these three compounds represent a novel finding.DiscussionTherefore, phenolic acids and flavonoids play important roles in NR mediated FW resistance breeding in cucumber.

Abstract Background No uniform consensus has been achieved regarding the ambulation protocol after transfemoral cerebral angiography (TFA). Until now, in most hospitals patients are prescribed 8-12 h strict immobilization along with bed rest in the supine position after TFA in China, which causes great discomfort to patients. Objective To evaluate the effect of an evidence-based early ambulation protocol on the prevention of vascular complications and general discomfort in patients following transfemoral cerebral angiography (TFA). Methods A prospective quasi-experimental study was conducted on 214 patients undergoing TFA with manual compression. Patients in the experimental group were placed supine position for 2 h with a sandbag placed on the wound dressing, followed by a semi-seated position for another 2 h. After this period, patients took 2 h bed rest (move freely) with the sandbag removed, and were allowed to get out of bed 6 h after TFA. Patients in the control group were restricted to an 8 h bed rest in a supine position with the affected leg straight and immobilized. The vascular complications (bleeding, hematoma, ecchymosis) and levels of comfort (low back pain, leg pain, and blood pressure) were evaluated after the procedure. Numeric Rating Scale (NRS) pain scores, systolic blood pressure (SBP); diastolic blood pressure (DBP) were measured hourly for 8 h after TFA. Results There was no significant difference in the two groups with regard to vascular complications including bleeding events (P = 0.621), bleeding volume (P = 0.321), and area of hematoma (P = 0.156). The area of ecchymosis in the experimental group was significantly smaller than the control group (P = 0.031). The MCrAlY(M=Ni, Co or NiCo)has become a widely used bond-coat material of thermal barrier coatings (TBCs) due to its advantages of high-temperature oxidation resistance and high thermal expansion coefficient, with the continuous increase in turbine inlet temperature of aero engines and gas turbines. However, the stress distribution at the interface between the bond and top coating in thermal barrier coatings becomes more complicated under a high-temperature service environment. The failure of the bond-coat leads to the spalling of the top coating, which limits the development of the thermal protection coating field. The development process of the bond-coat was briefly described.The failure behavior of the bond-coat interface due to high-temperature phase transition, increased thermal and growth stresses, and S-element diffusion was focused on, and the failure mechanism of the bond-coat interface was analyzed. The research on improving the bond-coat interface failure at home and abroad was summarized. The synergistic strengthening of MCrAlY by rare earth and nanoparticles is proposed, which provides a research direction for the future optimization design of thermal bond-coat systems.

The microscopic pore-fracture structure and wettability have a significant influence on the two-phase seepage of shale gas and water. Due to the limitation of experimental conditions, the seepage patterns of gas and water in shale pores and slits under different wetting conditions have not been clarified yet. In this study, the three-dimensional digital rock models of shale inorganic pores, organic pores, and microfractures are established by focused ion beam-scanning electron microscopy scanning, and gas-driven water seepage simulation in shale microscopic pore-fracture structure under different wetting conditions is carried out based on volume of fluid method. The simulation results show that the gas–water relative permeability curves of microfractures are up-concave, and the gas–water relative permeability curves of inorganic and organic pores are up-convex; the gas–water two-phase percolation in microfractures is least affected by the change of wettability, the gas–water two-phase percolation in inorganic pores is most affected by the change of wettability, and the organic pores are in between; the gas–water two-phase percolation zone of microfractures is the largest, and the isotonic saturation is the highest; under the water-wet conditions, the critical gas saturation of microfractures, inorganic pores, and organic pores are 0.13, 0.315, and 0.34, respectively, and the critical gas saturation of organic pores under non-water-wet conditions is 0.525, indicating that under water-bearing conditions, the shale gas flow capacity in water-wet microfractures is the strongest, followed by water-wet inorganic pores, water-wet organic pores, and hydrophobic organic pores, respectively.

Abstract Background In this study, we investigated the effects of alpine meadow in different phenological periods on ruminal fermentation, serum biochemical indices, and gastrointestinal tract microbes in grazing yak on the Qinghai-Tibetan Plateau. A total of eighteen female freely grazing yaks with an average age of 3 years old and a body weight of 130 ± 19 kg were selected. According to the plant phenological periods, yaks were randomly allocated to one of three treatments: (1) regreen periods group (RP, n = 6); (2) grassy periods group (GP, n = 6); and (3) hay periods group (HP, n = 6). At the end of the experiment, the blood, rumen fluids, and rectal contents were collected to perform further analysis. Results The concentrations of total volatile fatty acid (TVFA), acetate, glucose (GLU), triglyceride (TG), cholesterol (CHO), high density lipoprotein (HDL), and low density lipoprotein (LDL) were higher in the GP group than in the HP group (P

The skin is subjected to various external factors that contribute to aging including oxidative stress from hydrogen peroxide (H2O2). This study investigated the distribution of aquaporin-8 (AQP8), a protein that transports H2O2 across biological membranes, in skin cells, and its effects in mitigating H2O2-induced oxidative damage. Human dermal fibroblasts were treated with increasing concentrations of H2O2 to evaluate oxidative damage. Cell viability, reactive oxygen species (ROS) generation, and the expression of specific genes associated with skin aging (IL-10, FPR2, COL1A1, KRT19, and Aggrecan) were evaluated and AQP8 expression was assessed via quantitative polymerase chain reaction and western blotting. Small-interfering RNA was used to silence the AQP8 gene and evaluate its significance. The results show that H2O2 treatment reduces cell viability and increases ROS generation, leading to oxidative damage that affects the expression of target molecules. Interestingly, H2O2-treated cells exhibit high levels of AQP8 expression and gene silencing of AQP8 reverses high levels of ROS and low levels of COL1A1, KRT19, and Aggrecan expression in stressed cells, indicating that AQP8 plays a vital role in preventing oxidative damage and consequent aging. In conclusion, AQP8 is upregulated in human dermal fibroblasts during H2O2-induced oxidative stress and may help prevent oxidative damage and aging. These findings suggest that AQP8 could be a potential therapeutic target for skin aging. Further research is necessary to explore the feasibility of using AQP8 as a preventive or therapeutic strategy for maintaining skin health.

It is a great challenge for sedimentologists to perform the facies analysis of shales due to the lack of macro-structures and high heterogeneity in shale, and thus, conventional research methods are poorly applied in the field of shale sedimentology. To establish a typical depositional model for marine shales, a new method adopted from marine carbonate rocks was employed to perform the microfacies analysis of marine shales of the Wufeng and Longmaxi Formation in Sichuan Basin. This method emphasizes the fact that many skeletal constituents in marine shales have specific bathymetric distributions, allowing for a better understanding of the depositional process of shales. With this approach, a total of nine microfacies were identified and two depositional models for marine shales were proposed. The depositional models show that sea levels were high during the Middle to Late Wufeng period, which was followed by a rapid regression that led to a significant sea-level drop by over 50 m at the topmost of the Wufeng Formation, and consequently, widespread fine-grained sandy shales with large amounts of benthic bioclasts were deposited in the study region. The eustatic sea-level changes recovered by using the method of microfacies analysis are in line with the global trend, indicating that the method has promising applications in the field of marine shale sedimentology.

[Objective] Green ammonia, produced through carbon-free processes, has emerged as a high-quality carrier of hydrogen energy and stands as an alternative energy source vital for achieving carbon neutrality. Therefore, the planning and construction of green ammonia terminals hold crucial significance in facilitating the rapid development of the hydrogen energy industry chain and promoting the green transformation of the societal energy structure. [Methods] In this paper, a concise overview of ammonia's properties and advantages was provided that ammonia, as an efficient medium for energy and hydrogen storage, was able to reduce significantly the costs associated with hydrogen storage, transportation, and consumption, and enhance overall energy security due to its particular characteristics of high energy density, easy storage and transportation, zero carbon emissions at terminals, high safety and long-term storage. Various ammonia hydrogen projects and their trading details in different countries were investigated to analyze the global growth in green ammonia production capacity and demand, and to present a review of the current status of ammonia energy trade and terminal construction. In contrast to established technologies regarding LNG terminals, key processes and technologies involved in green ammonia terminals were elaborated on in this paper, including the technological system for green ammonia storage, liquid ammonia evaporator processing, and ammonia unloading and filling. [Results] This paper emphasized safety concerns associated with green ammonia terminals, specifically addressing the toxicity and potential for combustion-induced explosibility that may arise from liquid ammonia leakage. Additionally, it provided relevant preventive and protective measures. [Conclusion] The research and development of technologies for green ammonia terminals are of great significance for enhancing the utilization of green hydrogen energy in both the chemical fertilizer and energy sectors. The evolving demands of the new era call for a continued expansion of green ammonia terminals in terms of quantity and scale in the future. Furthermore, it is suggested that the renovation of bulk storage and transportation facilities, such as LNG terminals, can be an effective approach to cost reduction.

Background Elderly hemodialysis (HD) patients have a high risk of death. The effect of different types of HD membranes on survival is still controversial. The purpose of this study was to determine the relationship between the use of low-flux or high-flux membranes and all-cause and cardiovascular mortality in elderly hemodialysis patients.Methods This was a retrospective clinical study involving maintenance hemodialysis patients which were categorized into low-flux and high-flux groups according to the dialyzer they were using. Propensity score matching was used to balance the baseline data of the two groups. Survival rates were compared between the two groups, and the risk factors for death were analyzed by multivariate Cox regression.Results Kaplan–Meier survival analysis revealed no significant difference in all-cause mortality between the low-flux group and the high-flux group (log-rank test, p = 0.559). Cardiovascular mortality was significantly greater in the low-flux group than in the high-flux group (log-rank test, p = 0.049). After adjustment through three different multivariate models, we detected no significant difference in all-cause mortality. Patients in the high-flux group had a lower risk of cardiovascular death than did those in the low-flux group (HR = 0.79, 95% CI, 0.54–1.16, p = 0.222; HR = 0.58, 95% CI, 0.37–0.91, p = 0.019).Conclusions High-flux hemodialysis was associated with a lower relative risk of cardiovascular mortality in elderly MHD patients. High-flux hemodialysis did not improve all-cause mortality rate. Differences in urea distribution volume, blood flow, and systemic differences in solute clearance by dialyzers were not further analyzed, which are the limitations of this study.

Mutton is one of the most popular meats among the general public due to its high nutritional value. This study evaluated the differences in meat quality among Chaka (CK), Black Tibetan (BT) and Oula (OL) sheep and investigated the metabolic mechanisms affecting meat quality using targeted and untargeted metabolomics and 16S rRNA. The results showed that the meat quality of CK sheep was superior to that of BT and OL sheep in terms of meat color, muscle fiber characteristics and nutritional quality. Pseudobutyrivibrio, Alloprevotella, Methanobrevibacter, unidentified_Christensenellaceae, and unidentified_Bacteroidales were key microbes involved in regulating meat color, muscle fiber characteristics, amino acid and fatty acid content. Protein digestion/absorption, pentose phosphate metabolism, carbon metabolism, and glyoxylate and dicarboxylate metabolism were important metabolic pathways involved in meat quality regulation. Our study is important for the development of sheep breeding strategy and sheep meat industry in Qinghai-Tibetan Plateau.

BackgroundC-reactive protein-to-albumin ratio (CRP/ALB) has been proven to represent a biomarker for predicting prognosis in many groups of patients with severe diseases. However, few studies have investigated the association between CRP/ALB and mortality in Japan older people with dysphagia patients.ObjectiveThis retrospective cohort study aimed to assess the prognostic value of C-reactive protein/albumin ratio (CAR) in older Japanese patients with dysphagia.MethodsWe analyzed data from 253 patients diagnosed with dysphagia at a single center between January 2014 and January 2017. Cox regression analysis was used to compare the mortality rates across the CAR tertiles. Subgroup analyses were conducted, and Kaplan–Meier curves were used to determine the median survival times.ResultsThe study included 154 female and 99 male patients, with a median age of 83 years. After adjusting for all covariates, the multivariable Cox regression analysis revealed a significant association between increasing CAR (HR = 1.19, 95% CI: 1.03–1.37, P = 0.022) and the risk of mortality. Compared to the reference group T1 (< 0.149), the adjusted hazard ratios for T2 (0.149–0.815) and T3 (> 0.815) were 1.75 (95% CI: 1.07–2.87, P = 0.027) and 2.15 (95% CI: 1.34–3.46, P = 0.002), respectively. Kaplan–Meier curves indicated median survival times of 864, 371, and 223 days for T1, T2, and T3, respectively.ConclusionThe C-reactive protein/albumin ratio was positively related to mortality in Japan older people with dysphagia patients. There was no interaction for the subgroup analysis. The result was stable.

In the deep Longmaxi Formation shale gas reservoirs of the southern Sichuan Basin, strong overpressure is universally developed to varying degrees. However, there is currently a lack of in-depth research on the formation mechanisms, evolutionary patterns, and the controlling effects on reservoir pore characteristics of strong overpressure. This limitation significantly restricts the evaluation of deep shale gas reservoirs. This study selected typical overpressured shale gas wells in Yongchuan, Luzhou, and Dazu areas as research subjects. Through comprehensive methods such as log analysis, fluid inclusion analysis, and numerical simulation, the dominant mechanisms of strong overpressure formation were determined, and the pressure evolution from early burial to late strong uplift was characterized. Additionally, the impact of varying degrees of overpressure on reservoir pore characteristics was studied using techniques such as scanning electron microscopy, gas adsorption-mercury intrusion, and helium porosity testing. The research findings indicate that hydrocarbon generation expansion is the primary mechanism for strong overpressure formation. The pressure evolution in the early burial phase is controlled by the processes of kerogen oil generation and residual oil cracking into gas. The reservoir experienced three stages: normal pressure (Ordovician to Early Triassic), overpressure (Early Triassic to Early Jurassic), and strong overpressure (Early Jurassic to Late Cretaceous), with pressure coefficients of approximately 1.08, 1.56, and 2.09, respectively. During the late strong uplift phase, the adjustment of early overpressure occurred due to temperature decrease and gas escape, leading to a decrease in formation pressure from 140.55 MPa to 81.63 MPa, while still maintaining a state of strong overpressure. Different degrees of strong overpressure exert a significant control on the physical properties of shale reservoirs and the composition of organic matter pores. Variations exist in the organic matter pore morphology, structure, and connectivity within the deep Wufeng-Longmaxi shale. Higher overpressure favors the preservation of organic large pores and reservoir porosity. Under conditions of strong overpressure development, deep siliceous shales and organically rich clay shales exhibit favorable reservoir properties. By determining the dominant mechanisms of strong overpressure in the Wufeng-Longmaxi Formation and studying pore characteristics, this research not only deepens the understanding of the geological features of deep shale gas reservoirs but also provides a new perspective for understanding the overpressure mechanisms and reservoir properties of deep shale gas reservoirs. Moreover, it is of significant importance for guiding the exploration and development of deep Longmaxi shale and provides valuable references for further research in related fields.

Abstract Therapeutic angiogenesis represents a promising avenue to revascularize the ischemic heart. Its limited success is partly due to our poor understanding of the cardiac stroma, specifically mural cells, and their response to ischemic injury. Here, we combine single-cell and positional transcriptomics to assess the behavior of mural cells within the healing heart. In response to myocardial infarction, mural cells adopt an altered state closely associated with the infarct and retain a distinct lineage from fibroblasts. This response is concurrent with vascular rarefaction and reduced vascular coverage by mural cells. Positional transcriptomics reveals that the infarcted heart is governed by regional-dependent and temporally regulated programs. While the remote zone acts as an important source of pro-angiogenic signals, the infarct zone is accentuated by chronic activation of anti-angiogenic, pro-fibrotic, and inflammatory cues. Together, our work unveils the spatiotemporal programs underlying cardiac repair and establishes an association between vascular deterioration and mural cell dysfunction.

Abstract Adult tissue-resident macrophages (RMs) are either maintained by blood monocytes or through self-renewal. While the presence of a nurturing niche is likely crucial to support the survival and function of self-renewing RMs, evidence regarding its nature is limited. Here, we identify fibro-adipogenic progenitors (FAPs) as the main source of colony-stimulating factor 1 (CSF1) in resting skeletal muscle. Using parabiosis in combination with FAP-deficient transgenic mice (Pdgfrα CreERT2 × DTA) or mice lacking FAP-derived CSF1 (Pdgfrα CreERT2 × Csf1 flox/null ), we show that local CSF1 from FAPs is required for the survival of both TIM4- monocyte-derived and TIM4+ self-renewing RMs in adult skeletal muscle. The spatial distribution and number of TIM4+ RMs coincide with those of dipeptidyl peptidase IV (DPPIV)+ FAPs, suggesting their role as CSF1-producing niche cells for self-renewing RMs. This finding identifies opportunities to precisely manipulate the function of self-renewing RMs in situ to further unravel their role in health and disease.

Oil is a global, non-renewable energy source, which plays a pivotal role in the development of the global economy and the strategic reserve system. With the expansion of crude oil futures trading scale, crude oil is no longer a pure energy commodity, but is increasingly controlled by the financial market. Based on Bayesian network, this paper studies and analyzes the financial risks of non renewable energy such as oil. Finally, the prediction models of BP neural network and Bayesian network are given. Neural network is a mathematical model based on neurons, a nonlinear adaptive dynamic system formed by the connection between neurons, and based on these two models, discusses the financial risk early warning method of petroleum in non-renewable energy. The test results of this paper show that the Bayesian network and BP neural network are used for early warning analysis based on the comprehensive oil financial risk data from 1987 to 2015. The prediction results of neural network show that there is 65% correlation between the two. From the fitting results of the prediction results and real data, the fitting effect of Bayesian network is better than that of BP neural network, and the fitting success rate is as high as 80%. The Bayesian network prediction model is analyzed by using the autoregressive movement of the model itself and the time series measurement results of the data itself. The results show that the prediction model is highly reliable. Therefore, it is very necessary to adopt Bayesian network prediction model in the risk early warning of petroleum finance.

The current limitations of air-cooled proton exchange membrane fuel cells (AC-PEMFCs) in water and heat management remain a major obstacle to their commercialization. A 90 cm2 full-size AC-PEMFC multi-physical field-coupled numerical model was constructed; isothermal and non-isothermal calculations were performed to explore the effects of univariate and multivariate variables on cell performance, respectively. The isothermal results indicate that lower temperature is beneficial to increase the humidity of MEA, and distribution uniformity at lower stoichiometric ratios and lower temperatures is better. The correlation between current density distribution and temperature, water content, and concentration distribution shows that the performance of AC-PEMFCs is influenced by multiple factors. Notably, under high current operation, the large heat generation may lead to high local temperature and performance decline, especially in the under-channel region with drier MEA. The higher stoichiometric ratio can enhance heat dissipation, improve the uniformity of current density, and increase power density. Optimal fuel cell performance is achieved with a stoichiometric ratio of 300, balancing the mixed influence of multiple factors.

Respiratory system diseases caused by respiratory viruses are common and exert tremendous pressure on global healthcare system. In our previous studies, we found that Long non-coding RNA NRAV (Lnc NRAV) and its target molecule Rab5c plays a significant role in respiratory virus infection. However, the mechanism by which Rab5c affects virus replication remains unclear. Rab5c, a protein mainly localized on the cell membranes and in early endosomes and phagosomes, participates in endocytosis mediated by clathrin and regulates the fusion of early endosome, maturation of early phagosomes, and autophagy. Therefore, we inferred that Rab5c impacts virus replication, which might be related to endocytosis or autophagy. We selected RSV (respiratory syncytial virus) as a representative enveloped virus and ADV (Adenovirus) as a representative non-enveloped virus to explore the possible mechanism of RSV and ADV replication promoted by Rab5c in A549 cells and in Rab5c-overexpressing mice. Here, we confirmed that the activated Rab5c promotes RSV and ADV replication and the inactivated Rab5c inhibits their replication. However, Rab5c promoting RSV and ADV replication is not mediated by endocytosis rather by autophagy in respiratory epithelial cells. Our study showed that Rab5c upregulates LC3-Ⅱ (microtubule-associated protein 1 light chain 3 beta) protein expression levels by interacting with Beclin1, a key autophagy molecule, which can induce autophagy and promote replication of ADV and RSV. This study enriches the understanding of the interaction between respiratory viruses and Rab5c, providing new insights for virus prevention and treatment.

Intervertebral disc (IVD) degeneration (IDD) is a worldwide spinal degenerative disease. Low back pain (LBP) is frequently caused by a variety of conditions brought on by IDD, including IVD herniation and spinal stenosis, etc. These conditions bring substantial physical and psychological pressure and economic burden to patients. IDD is closely tied with the structural or functional changes of the IVD tissue and can be caused by various complex factors like senescence, genetics, and trauma. The IVD dysfunction and structural changes can result from extracellular matrix (ECM) degradation, differentiation, inflammation, oxidative stress, mechanical stress, and senescence of IVD cells. At present, the treatment of IDD is basically to alleviate the symptoms, but not from the pathophysiological changes of IVD. Interestingly, the p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway is involved in many processes of IDD, including inflammation, ECM degradation, apoptosis, senescence, proliferation, oxidative stress, and autophagy. These activities in degenerated IVD tissue are closely relevant to the development trend of IDD. Hence, the p38 MAPK signaling pathway may be a fitting curative target for IDD. In order to better understand the pathophysiological alterations of the intervertebral disc tissue during IDD and offer potential paths for targeted treatments for intervertebral disc degeneration, this article reviews the purpose of the p38 MAPK signaling pathway in IDD.

BackgroundWe previously found that the respiratory epithelial cells could eliminate the invaded group A streptococcus (GAS) through autophagy induced by binding a fibronectin (Fn) binding protein (FnBp) expressed on the surface of GAS to plasma protein Fn and its receptor integrin α5β1 of epithelial cells. Is autophagy initiated by FnBp+ bacteria via FnBp-Fn-Integrin α5β1 axis a common event in respiratory epithelial cells?MethodsWe chose Staphylococcus aureus (S. aureus/S. a) and Listeria monocytogenes (L. monocytogenes/L. m) as representatives of extracellular and intracellular FnBp+ bacteria, respectively. The FnBp of them was purified and the protein function was confirmed by western blot, viable bacteria count, confocal and pull-down. The key molecule downstream of the action axis was detected by IP, mass spectrometry and bio-informatics analysis.ResultsWe found that different FnBp from both S. aureus and L. monocytogenes could initiate autophagy through FnBp-Fn-integrin α5β1 axis and this could be considered a universal event, by which host tries to remove invading bacteria from epithelial cells. Importantly, we firstly reported that S100A8, as a key molecule downstream of integrin β1 chain, is highly expressed upon activation of integrin α5β1, which in turn up-regulates autophagy.ConclusionsVarious FnBp from FnBp+ bacteria have the ability to initiate autophagy via FnBp-Fn-Integrin α5β1 axis to promote the removal of invading bacteria from epithelial cells in the presence of fewer invaders. S100A8 is a key molecule downstream of Integrin α5β1 in this autophagy pathway.

Objective(s): Liver fibrosis is a common liver disease caused by chronic liver damage. However, there are currently no approved drugs available to treat it. Therefore, the therapeutic effect of indirubin on liver fibrosis was evaluated. This study investigated the protective effect and related molecular mechanism of indirubin against CCl4-induced liver fibrosis in mice.Materials and Methods: We first detected the effect of indirubin on liver fibrosis in mice (n=8 per group, 32 mice total) by ELISA, HE, and Masson staining. Subsequently, the proliferation of activated HSCs was detected by MTT and EdU. Finally, the changes of related proteins and signaling pathways in mice treated with indirubin were investigated by qRT-PCR and Western blot. One-way ANOVA or two-tailed student’s t-test was used for comparison between groups.Results: Firstly, we found that indirubin (25 mg/kg) therapy could attenuate liver injury and significantly down-regulate α-SMA (P=0.0038) and collagen 1 (P=0.0057) in the liver using CCl4-induced liver fibrosis in mice. Secondly, we showed that indirubin (25 μM) could significantly inhibit hepatic stellate cell (HSC) trans-differentiation into myofibroblasts and proliferation (P=0.0063) in HSC-T6 cells treated by TGF-β. Finally, we showed that indirubin could greatly reduce the protein levels of p-Smad2/3, p38, p-ERK, and p-JNK in vivo and in vitro.Conclusion: Our results suggested that indirubin alleviated liver fibrosis and HSC activation mainly through TGF-β-mediated signaling pathways in vivo and in vitro. In conclusion, our data showed that indirubin could be a promising clinical therapeutic drug for the prevention and treatment of liver fibrosis.

Abstract Cellular senescence serves as a fundamental and underlying activity that drives the aging process, and it is intricately associated with numerous age-related diseases, including Alzheimer's disease (AD), a neurodegenerative aging-related disorder characterized by progressive cognitive impairment. Although increasing evidence suggests that senescent microglia play a role in the pathogenesis of AD, their exact role remains unclear. In this study, we quantified the levels of lactic acid in senescent microglia, and hippocampus tissues of naturally aged mice and AD mice models (FAD4T and APP/PS1). We found lactic acid levels were significantly elevated in these cells and tissues compared to their corresponding counterparts, which increased the level of pan histone lysine lactylation (Kla). We aslo identified all histone Kla sites in senescent microglia, and found that both the H3K18 lactylation (H3K18la) and Pan-Kla were significantly up-regulated in senescent microglia and hippocampus tissues of naturally aged mice and AD modeling mice. We demonstrated that enhanced H3K18la directly stimulates the NFκB signaling pathway by increasing binding to the promoter of Rela (p65) and NFκB1(p50), thereby upregulating senescence-associated secretory phenotype (SASP) components IL-6 and IL-8. Our study provides novel insights into the physiological function of Kla and the epigenetic regulatory mechanism that regulates brain aging and AD. Specifically, we have identified the H3K18la/NFκB axis as a critical player in this process by modulating IL-6 and IL-8. Targeting this axis may be a potential therapeutic strategy for delaying aging and AD by blunting SASP.

Regenerative fuel cells and the phenomenon of cell reversal (CR) necessitate creating robust catalyst layers for consistent performance in fuel cells. This research used in situ Raman spectroscopy to observe molecular alterations on carbon nanotube-supported platinum catalysts (Pt/CNT) during ethanol oxidation. Following a CR event simulation, the ethanol oxidation efficiency on Pt/CNT was amplified 2.8 times after high-potential scanning but reverted to its initial efficiency after 100 cycles. The adsorbed *CO2− species on Pt/CNT was pivotal for initiating ethanol oxidation, with the rate assessed through Raman analysis. In addition to water electrolysis, the carbon substrate was degraded. This study sheds light on the mechanisms behind catalyst degradation, steering the creation of more advanced catalysts.

Abstract Background Ankylosing spondylitis (AS) is one of the most common immune-mediated arthritic diseases worldwide. Despite considerable efforts to elucidate its pathogenesis, the molecular mechanisms underlying AS are still not fully understood. Methods To identify candidate genes involved in AS progression, the researchers downloaded the microarray dataset GSE25101 from the Gene Expression Omnibus (GEO) database. They identified differentially expressed genes (DEGs) and functionally enriched them for analysis. They also constructed a protein–protein interaction network (PPI) using STRING and performed cytoHubba modular analysis, immune cell and immune function analysis, functional analysis and drug prediction.The results showed that DEGs were mainly associated with histone modifications, chromatin organisation, transcriptional coregulator activity, transcriptional co-activator activity, histone acetyltransferase complexes and protein acetyltransferase complexes. Results The researchers analysed the differences in expression between the CONTROL and TREAT groups in terms of immunity to determine their effect on TNF-α secretion. By obtaining hub genes, they predicted two therapeutic agents, AY 11–7082 and myricetin. Conclusion The DEGs, hub genes and predicted drugs identified in this study contribute to our understanding of the molecular mechanisms underlying the onset and progression of AS. They also provide candidate targets for the diagnosis and treatment of AS.

ABSTRACTMitochondria are good targets for viruses to manipulate their hosts. However, it remains obscure whether respiratory syncytial virus (RSV) target mitochondria to suppress the type I interferon (IFN) responses. Here, we show that nonstructural protein 1 (NS1) protein of RSV interacts with Tu translation elongation factor mitochondrial (TUFM), which can lead to its localization in mitochondria and finally induce TUFM-dependent mitophagy and inhibition of IFNβ. Mechanically, NS1-mediated TUFM-dependent mitophagy does not depend on the PINK1-PARKIN pathway and classic mitophagy receptors. Importantly, NS1 may act as a new receptor protein to bridge mitochondria and autophagosomes by interacting with TUFM and LC3B. The LIR motif of NS1 protein is essential for its interaction with LC3B and is of great importance for its mitophagy induction and IFNβ suppression. Finally, NS1-induced TUFM-dependent mitophagy was essential for its attenuated IFNβ response using autophagy-deficient cells and mice. Our study provides a novel mitophagy receptor molecular and a new antiviral option by suppressing antiviral innate immune via targeting TUFM-dependent mitophagy.IMPORTANCEIt is a worthy concern for us to understand virus-host interactions which affect progression and prognosis of disease. We demonstrated that the non-structural protein 1 of respiratory syncytial virus (RSV NS1) may act as a novel mitophagy receptor to induce mitophagy by binding LC3B and mitochondrial protein TUFM, and finally dampen interferon (IFN) responses induced by RIG1 and RSV infection. TUFM is beneficial for RSV replication in vivo and vitro. It is new and interesting that RSV NS1 may function as a mitophagy receptor to interact with LC3B. The LIR motif of NS1 protein is essential for its interaction with LC3B. We further confirm that RSV NS1 inhibited IFNβ response and promoted RSV replication in autophagy-dependent mechanisms in vivo and vitro. Our study contributes to understanding virus-host interaction, enriching our insights into RSV pathogenic mechanism and exploiting new antiviral treatments targeting TUFM.

Summary: Metformin prevents progression of non-alcoholic fatty liver disease (NAFLD). However, the potential mechanism is not entirely understood. Ferroptosis, a recently recognized nonapoptotic form of regulated cell death, has been reported to be involved in the pathogenesis of NAFLD. Here, we investigated the effects of metformin on ferroptosis and its potential mechanism in NAFLD. We found that metformin prevented the progression of NAFLD, and alleviated hepatic iron overload (HIO), ferroptosis and upregulated ferroportin (FPN) expression in vivo and in vitro. Mechanically, metformin reduced the lysosomal degradation pathway of FPN through activation AMPK, thus upregulated the expression of FPN protein, alleviated HIO and ferroptosis, and prevented progression of NAFLD. These findings discover a mechanism of metformin, suggesting that targeting FPN may have the therapeutic potential for treating NAFLD and related disorders.

Summary: Efficient skeletal muscle regeneration necessitates fine-tuned coordination among multiple cell types through an intricate network of intercellular communication. We present a protocol for generation of a time-resolved cellular interactome during tissue remodeling. We describe steps for isolating distinct cell populations from skeletal muscle of adult mice after acute damage and extracting RNA from purified cells prior to the generation of RNA sequencing data. Purpose The aim of this study was to further assess whether the platelet-to-lymphocyte ratio (PLR) is independently associated with all-cause mortality and cardiovascular mortality in maintenance hemodialysis (MHD) patients.Methods From January 1, 2014, to December 31, 2014, patients undergoing regular hemodialysis in the Blood Purification Center of the General Hospital of Northern Theatre Command were retrospectively selected. A total of 303 MHD patients were enrolled in accordance with the inclusion and exclusion criteria. For each patient, the endpoint of follow-up was either death or December 31, 2021. The primary endpoints were all-cause and cardiovascular death. A receiver operating characteristic (ROC) curve was drawn to detect the predictive ability of PLR, and the optimal critical value of PLR was determined to be 107.57. Kaplan–Meier curves and Cox proportional analysis were used to assess the prognostic value of PLR. We used the same method to evaluate the correlation between the neutrophil-to-lymphocyte ratio (NLR) and the prognosis of MHD patients.Results At the end of follow-up, 128 MHD patients had progressed to all-cause death, and 73 MHD patients had progressed to cardiovascular death. In multivariate Cox regression, both the high PLR group and the high NLR group were independently associated with all-cause mortality (HR 2.608, 95% CI 1.579–4.306, p

The clinical management of impacted incisors affected by root dilaceration is challenging. This case report presents the treatment of an adolescent female patient who presented with a deeply impacted, rotated and dilacerated maxillary central incisor and also reviews the current literature on the definition, prevalence, aetiology, diagnosis, and treatment of the anomaly. The impacted and dilacerated incisor was successfully orthodontically treated following a two-stage surgical exposure and after being rotated 100 degrees in the sagittal plane (due to the deep impaction and severe displacement). A Ballista spring and piggyback appliance technique were used to elevate the tooth. The dilacerated incisor was successfully preserved and relocated in the dental arch. The pre-operative and post-operative CBCT scans showed no significant root resorption of the repositioned tooth.

Abstract Background The concept of acute kidney injury (AKI) substages has been recommended to better phenotype AKI and identify high-risk patient groups and therefore improve the diagnostic accuracy of AKI. However, there remains a gap between the recommendation and the clinical application. The study aimed to explore the incidence of AKI substages based on a sensitive AKI biomarker of urinary cystatin C (uCysC), and to determine whether AKI substages were relevant with respect to outcome in critically ill children. Results The multicenter cohort study enrolled 793 children in pediatric intensive care unit (PICU) of four tertiary hospitals in China. Children were classified as non-AKI, sub-AKI and AKI substages A and B according to uCysC level at PICU admission. Sub-AKI was defined by admission uCysC level ≥ 1.26 mg/g uCr in children not meeting the KDIGO criteria of AKI. In children who fulfilled KDIGO criteria, those with uCysC

Abstract Doxorubicin (DOX) is a commonly used antitumor drug, but its application has been limited because of its strong cardiac damage. This study aims to explore the role of NSUN2 in DOX-induced heart injury. C57BL/6J mice were intraperitoneally injected with 20 mg/Kg DOX to induce heart injury. After 3 days, the cardiac function, cardiac histopathology, myocardial apoptosis, and the expression level of NSUN2 were detected. In vitro, H9C2 cells were transfected with NSUN2 siRNA or overexpressed lentivirus and then treated with 500 ng/ml DOX. After 24 h, the changes in reactive oxygen species (ROS), apoptosis, and NSUN2 expression were detected. After DOX treatment, both in vitro and in vivo experiments showed that the cardiac function decreased, the number of apoptotic cells increased, and the expression level of NSUN2 increased. Interfering the expression of NSUN2 by siRNA promoted DOX-induced heart injury, while overexpression of NSUN2 could inhibit DOX-induced heart injury. Further study showed that NSUN2 promoted antioxidative stress by upregulating the Nrf2 protein level. In addition, NSUN2 overexpression could increase the half-life of Nrf2 mRNA. m5C RNA methylation immunoprecipitation (MeRIP) also showed that the level of Nrf2 m5C mRNA was significantly increased in NSUN2 overexpressed group when compared to the GFP group. NSUN2 enhances the expression of Nrf2 by promoting Nrf2 mRNA m5C modification and enhances its antioxidative stress effect to alleviate DOX-induced myocardial injury.

Abstract Background Traditional Chinese Medicine (TCM) nurse‐led clinics (NLC) is an innovative working model in China, representing the specialization and extension of nurses’ role. However, as a pioneer in TCM nursing, this new model of working is facing both opportunities and challenges because it is known little about the operational status of NLCs. Aims To explore the experiences of nurses who work in NLC in TCM hospitals. Materials & Methods A focused ethnographic study was conducted in three TCM hospitals affiliated with Guangzhou University of Chinese Medicine. We interviewed eleven nurses in those hospitals and observed seven of them working with patients. We used snowball sampling for data collection including interview, non‐participant observation and documents from medical records. All the data were processed as following steps: (a) coding for descriptive labels; (b) sorting for patterns; (c) identifying outliers or negative cases; (d) generalizing with construction and theories and (e) noting reflective remarks. Results Nurse‐led clinics help nurses develop their skills and knowledge that are highly recognized by public since they meet the growing needs of patients and also relieve the workload of physicians in the hospitals. However, lack of specialization is still a major challenge in NLCs due to insufficiency of full‐time staff with specialized education, nurse‐led practice without standardized guidelines, restrictions on prescription right of nurses, and also inadequate support from hospitals. Discussions As a revolutionary innovation of working model for nurses in TCM hospitals, NLCs could improve quality of care and lead to a comprehensive promotion of nursing career. However, there are several challenges on providing high quality care for patients whilst improving educational development of nurses. This study suggests that nurses, hospital administration and the government should cooperate with each other to develop standard nursing programs for NLCs. Conclusions It is imperative to identify nursing roles, collect available resources, and develop supportive policies and training programs to enhance the quality of NLCs.

High-resolution, multi-channel remote sensing images have complex spatial layout and channel structure. Traditional Convolutional Neural Networks (CNNs) methods do not make full use of the features of remote sensing images, ignoring contextual relevance and globality, resulting in insufficient features discrimination and limiting the accuracy of scene classification. To address these issues, we propose a discriminative network HCA-TSA based on head-tail global joint dual attention mechanism. Considering the characteristics between the spatial dimension and the channel dimension, the channel attention module Head-Channel Attention (HCA) and the spatial attention module Tail-Spatial Attention (TSA) are proposed. This network uses the context information discrimination ability of the Gated Recurrent Unit (GRU) module and the structure of the global layout to learn and output the importance weights of different channels and spatial features, pay more attention to the salient features in the remote sensing image, and ignore the insignificant features, thus improve the discriminative power of feature representation. The proposed network can be connected to any benchmark CNNs, and the whole network structure can be trained end-to-end. Through a comprehensive comparative experiment on three public datasets with large differences, AID, UC-Merced and HSRS-SC, the highest classification accuracies in the experimental results are 97.68%, 99.41% and 98.38%, respectively. The results show that the proposed method can effectively discriminate scenarios and obtain competitive classification results, which verifies the effectiveness of the proposed method.

By designing structures with different gradients, the strength of lattice structure can be improved, and its mechanical properties such as impact resistance, energy absorption and vibration reduction can be improved. The Voronoi method was used to generate random points in the Eulerian space domain to form the basic unit cell of lattice structure. The distribution forms of different gradient unit cell structure were set, and finally the lattice structures with different gradient were generated. In this experiment, “inward” gradient and “outward” gradient were designed, and five different lattice structures with gradient grades of 0.5, 0.6, 0.7, 0.8 and 0.9 were designed for “inward” and “outward” structures, respectively. And gradient level of the uniform lattice structure was 1.0. The finite element analysis software Abaqus was used to simulate the mechanical properties of lattice structures with different gradients. The lattice structures were fabricated by 3D printing technology, and the mechanical compression experiment was carried out to verify the finite element analysis results. The results of finite element analysis and experiments show that the mechanical properties of lattice structures with different gradients generated by Voronoi method are better than that of the uniform lattice structure; in the “inward” gradient lattice structures, the mechanical properties of the lattice structure with gradient grade 0.6 are the best, with good shock resistance and vibration reduction; in the “outward” gradient lattice structures, the mechanical properties of the lattice structure with gradient grade 0.8 is the best. The overall performance of “outward” gradient lattice structure is better than that of “inward” gradient lattice structure. The gradient lattice structures based on Voronoi method have good mechanical properties such as high strength, vibration resistance and energy absorption. The mechanical properties of lattice structures are closely related to the gradient grade.

Hyperspectral images possess the characteristics of high dimensionality, which causes “dimensional disaster” and low classification accuracy. In order to solve the problems, based on traditional k-means algorithm, considering the importance of different bands for classification, and also combining both intra-class and inter-class information, a Kmeans-CM (K-means with correlation coefficient and maximize inter-class distance) algorithm with spectral angle mapper for hyperspectral image classification is proposed. First, we define weights of bands by introducing coefficient of variation and spectral angle mapping, which measures the importance of bands for classification, so as to make up for the deficiency of the traditional K-means algorithm to treat each band equally. Second, we introduce correlation coefficient to reset the intra-class distances in order to intensify correlation between pixels in the same category. Then, in order to reduce effect of local optimum of clustering effect, we introduce inter-class information for clustering by maximizing the distance between class centers and global center. Finally, the K-means clustering objective function is redefined according to the band weights and intra-class, inter-class information, and also solving optimally. The overall classification accuracy of the method reached 84.47%, 90.08% and 80.45% on classical hyperspectral data sets Pavia University, Salinas and Botswana respectively. And comparing with the traditional K-means algorithm, the CV-K-means algorithm and the CK-means algorithm, the experimental results show that the proposed algorithm can effectively improve classification accuracy of hyperspectral images, thus proving that the algorithm has good classification performance.

The efficient catalysis of ethanol oxidation (EOR) is of paramount importance for the progress of energy conversion and storage technologies. We investigate the improved catalytic performance of commercial J-M Pt/C and Pd/C catalysts for ethanol oxidation by utilizing high potential scanning. By employing in situ Raman spectroscopy, we analyze the reaction intermediates during electrochemical reactions. Our findings highlight that the Pd/C catalyst exhibits remarkable enhancements in catalytic activity and stability, which can be attributed to the activation at high potentials on Pd/C electrode. Interestingly, different results are observed between Pt/C and Pd/C catalysts under the same experimental conditions. This study provides valuable insights into the design and development of efficient catalysts for ethanol oxidation.

Molecular dynamics simulations, the perturbation and scattering methods are used to study the transmission and reflection of an incident pulse due to dust impurities in a chain of dust particles. It is found that the transmission and reflection depend strongly on the number of impurities. How the momentum of both the transmitted and reflected wave depends on that of the incident wave is given analytically and numerically for only one impurity and numerically for multiple impurities. It is worth noting that the amplitude of the transmitted and reflected waves remains a constant when the impurity length exceeds the width of the solitary wave. The present results offer a means of detecting impurities in a dusty plasma crystal by measuring the transmitted-to-incident and reflected-to-incident amplitude ratios because they contain information about the impurities in a dusty plasma crystal.

Introduction and objectives: To investigate the efficacy of combined immunosuppressive regimens of cyclosporine (CsA), tacrolimus (TAC), or cyclophosphamide (CTX) combined with steroids in the treatment of idiopathic membranous nephropathy (IMN). Materials and methods: A total of 150 biopsy-proven IMN patients were divided into three groups: CTX, TAC, and CsA groups (50 cases each). Patients received a selected regimen for 48 weeks. The efficacy (remission rate, 24 h urinary protein, and serum albumin and creatinine) and safety (adverse events) profiles of administered regimens were evaluated at 12, 24 and 48 weeks. Results: At 12 weeks, the response rates for CsA, TAC, and CTX groups were 14%, 50%, and 22%, respectively. This increased to 74%, 84%, and 82%, respectively at 48 weeks. During follow-up, 24 h urinary protein significantly reduced from baseline in all regimens (P 0.05). Safety was comparable in all groups, with lower respiratory tract infection being the most frequent adverse event. Conclusions: The combined regimens (i.e., TAC, CsA, and CTX) are effective in the treatment of patients with IMN at 48 weeks, while TAC and CTX might be more beneficial in terms of shortened time to remission and increased complete response rate. Resumen: Introducción y objetivos: Investigar la eficacia de los regímenes inmunosupresores combinados de ciclosporina (CsA), tacrolimús (TAC) o ciclofosfamida (CTX) combinados con esteroides en el tratamiento de la nefropatía membranosa idiopática (NMI). Materiales y métodos: Un total de 150 pacientes con NMI comprobada por biopsia se dividieron en 3 grupos: grupos CTX, TAC y CsA (50 casos cada uno). Los pacientes recibieron un régimen seleccionado durante 48 semanas. Se evaluaron los perfiles de eficacia (tasa de remisión, proteína en orina de 24 h y albúmina y creatinina séricas) y seguridad (eventos adversos) de los regímenes administrados a las 12, 24 y 48 semanas. Resultados: A las 12 semanas, las tasas de respuesta para los grupos CsA, TAC y CTX fueron del 14, el 50 y el 22%, respectivamente. Esto aumentó al 74, el 84 y el 82%, respectivamente, a las 48 semanas. Durante el seguimiento, la proteína urinaria de 24 h se redujo significativamente desde el inicio en todos los regímenes (p 0.05). La seguridad fue comparable en todos los grupos, siendo la infección del tracto respiratorio inferior el evento adverso más frecuente. Conclusiones: Los regímenes combinados (es decir, TAC, CsA y CTX) son eficaces en el tratamiento de pacientes con NMI a las 48 semanas, mientras que TAC y CTX podrían ser más beneficiosos en términos de reducción del tiempo de remisión y aumento de la tasa de respuesta completa.

Sideways fall-induced hip fracture is a primary global health concern among the elderly. Existing impact models for predicting peak hip impact force mostly consider the human body-related parameters rather than impact surface parameters. This study proposed improving existing spring-mass-damper models by accounting for the human-structure dynamic interaction during sideways fall for better predicting peak impact force on the hip. Information required to construct the models was extracted from the literature. Different peak hip impact forces were estimated by considering differences in gender, body height, body mass, stiffness, damping coefficients of body tissue over the greater trochanter, and the impact surface stiffness. The predicted peak hip impact forces were compared to measured or simulated results in the literature and found to agree reasonably. Simulation results show that interactions with impact surfaces with lower stiffness can reduce the value of peak impact force applied on the hip by at least 16%.

Abstract Background Explanted livers from patients with inherited metabolic liver diseases possess the potential to be a cell source of good-quality hepatocytes for hepatocyte transplantation (HT). This study evaluated the therapeutic effects of domino HT using hepatocytes isolated from explanted human livers for acute liver failure (ALF). Methods Isolated hepatocytes were evaluated for viability and function and then transplanted into d-galactosamine/lipopolysaccharide-induced ALF mice via splenic injection. The survival rate was analyzed by the Kaplan–Meier method and log-rank test. Liver function was evaluated by serum biochemical parameters, and inflammatory cytokine levels were measured by ELISA. The pathological changes in the liver tissues were assessed by hematoxylin–eosin staining. Hepatocyte apoptosis was investigated by TUNEL, and hepatocyte apoptosis-related proteins were detected by western blot. The localization of human hepatocytes in the injured mouse livers was detected by immunohistochemical analyses. Results Hepatocytes were successfully isolated from explanted livers of 10 pediatric patients with various liver-based metabolic disorders, with an average viability of 85.3% ± 13.0% and average yield of 9.2 × 106 ± 3.4 × 106 cells/g. Isolated hepatocytes had an excellent ability to secret albumin, produce urea, uptake indocyanine green, storage glycogen, and express alpha 1 antitrypsin, albumin, cytokeratin 18, and CYP3A4. Domino HT significantly reduced mortality, decreased serum levels of alanine aminotransferase and aspartate aminotransferase, and improved the pathological damage. Moreover, transplanted hepatocytes inhibited interleukin-6 and tumor necrosis factor-α levels. Domino HT also ameliorates hepatocyte apoptosis, as evidenced by decreased TUNEL positive cells. Positive staining for human albumin suggested the localization of human hepatocytes in ALF mice livers. Conclusion Explanted livers from patients with inheritable metabolic disorders can serve as a viable cell source for cell-based therapies. Domino HT using hepatocytes with certain metabolic defects has the potential to be a novel therapeutic strategy for ALF.

Abstract Background Pulmonary infection is a frequent complication among stroke patients and adversely affects clinical outcomes, increases the length of hospitalization stay and costs, and aggravates the financial burden of the national medical health system. Early identification and management of high-risk patients are necessary and imperative to reduce the incidence of stroke-associated pneumonia (SAP). Aim The evidence-based practice project evaluated the effectiveness of a standard care bundle intervention in preventing the occurrence of SAP. Methods The project was conducted in a neurology department of a teaching hospital. Given the variation in assessment and management standards, evidence-based practice (EBP) methodology was used to establish a process for quality improvement. A thorough literature search was conducted to identify evidence-based interventions to manage and prevent SAP. Thorough critiques of the literature and synthesis of the evidence were completed. A systematic management flow and care bundle interventions were established. The care bundle included interventions, such as the utilization of tools for SAP risk screening; dysphagia screening and rehabilitation; feeding modification, oral care, airway management, position management, and the nursing techniques of traditional Chinese medicine. Results A significant improvement was observed in preventing SAP in patients in the postimplementation group compared with those in the preimplementation group (14.0% vs. 37.2%, p = 0.025). In addition, significantly lower duration of hospitalization, lower rate of aspiration, and improvements in albumin and oral hygiene were found after the implementation of the care bundle. Conclusions Evidence-based care bundles successfully empower nurses to reduce the incidence of SAP. The management flow of SAP prevention could be promoted to other units of the neurology department in the future. The results of the project reflect positively on the capacity to implement EBP in an acute care setting for stroke. The EBP methodology can be utilized to solve other clinical problems.

Abstract Background Substantial interstudy heterogeneity exists in defining acute kidney injury (AKI) and baseline serum creatinine (SCr). This study assessed AKI incidence and its association with pediatric intensive care unit (PICU) mortality under different AKI and baseline SCr definitions to determine the preferable approach for diagnosing pediatric AKI. Methods In this multicenter prospective observational cohort study, AKI was defined and staged according to the Kidney Disease: Improving Global Outcome (KDIGO), modified KDIGO, and pediatric reference change value optimized for AKI (pROCK) definitions. The baseline SCr was calculated based on the Schwartz formula or estimated as the upper normative value (NormsMax), admission SCr (AdmSCr) and modified AdmSCr. The impacts of different AKI definitions and baseline SCr estimation methods on AKI incidence, severity distribution and AKI outcome were evaluated. Results Different AKI definitions and baseline SCr estimates led to differences in AKI incidence, from 6.8 to 25.7%; patients with AKI across all definitions had higher PICU mortality ranged from 19.0 to 35.4%. A higher AKI incidence (25.7%) but lower mortality (19.0%) was observed based on the Schwartz according to the KDIGO definition, which however was overcome by modified KDIGO (AKI incidence: 16.3%, PICU mortality: 26.1%). Furthermore, for the modified KDIGO, the consistencies of AKI stages between different baseline SCr estimation methods were all strong with the concordance rates > 90.0% and weighted kappa values > 0.8, and PICU mortality increased pursuant to staging based on the Schwartz. When the NormsMax was used, the KDIGO and modified KDIGO led to an identical AKI incidence (13.6%), but PICU mortality did not differ among AKI stages. For the pROCK, PICU mortality did not increase pursuant to staging and AKI stage 3 was not associated with mortality after adjustment for confounders. Conclusions The AKI incidence and staging vary depending on the definition and baseline SCr estimation method used. The modified KDIGO definition based on the Schwartz method leads AKI to be highly relevant to PICU mortality, suggesting that it may be the preferable approach for diagnosing AKI in critically ill children and provides promise for improving clinicians’ ability to diagnose pediatric AKI.

Abstract The development of stable barium (Ba) isotope measurements provides a novel tool to investigate the geochemical cycling of Ba in the ocean and its sediments. In sediment pore waters, gradients of dissolved Ba concentrations result from various diagenetic processes. The distribution and fractionation of Ba isotopes in the pore waters are expected to further improve our understanding of these early diagenetic control mechanisms. Here, we present pore water profiles of dissolved stable Ba isotopic signatures (δ138Bapw) from shallow water sediments covering the entire Pearl River Estuary (PRE) in Southern China. We find pronounced depth‐dependent Ba isotope variations generally showing a shift from heavy to light δ138Bapw signatures from the sediment surface down to 15 cm depth. These gradients are well reproduced by a diffusion‐reaction model, which generates an apparent fractionation factor (138ε) of −0.60 ± 0.10‰ pointing to preferential removal of low‐mass Ba isotopes from the pore water during solution‐solid phase interactions. Consequently, the combined diagenetic processes induce the highest δ138Bapw values of +0.5 to +0.7‰ in the pore waters of the topmost sediment layer. Although the detrital fraction dominates the Ba content in the PRE surface sediments, the determined gradients of pore water Ba isotopes, together with concentration variations of Ba and other redox‐sensitive elements such as manganese (Mn), show that non‐detrital excess Ba carriers including Mn oxides and authigenic barite clearly affect the post‐depositional Ba dynamics. Stable Ba isotopes are thus a potentially powerful tracer of Ba geochemistry during early sediment diagenesis in estuarine depositional environments.
