Abstract

Diabetic wounds are prone to develop chronic wounds due to bacterial infection and persistent inflammatory response. However, traditional dressings are monofunctional, lack bioactive substances, have limited bacterial inhibition as well as difficulties in adhesion and retention. These limit the therapeutic efficacy of traditional dressings on diabetic wounds. Therefore, finding and developing efficient and safe wound dressings is currently an urgent clinical need. In this study, an antimicrobial gel loaded with silver nanoparticles (AgNPs) (referred to as AgNPs@QAC-CBM) was prepared by crosslinking quaternary ammonium chitosan (QAC) with carbomer (CBM) as a gel matrix. AgNPs@QAC-CBM exhibited a reticulated structure, strong adhesion, good stability, and remarkable bactericidal properties, killing 99.9% of Escherichia coli, Staphylococcus aureus, Candida albicans, and Pseudomonas aeruginosa within 1 min. Furthermore, AgNPs@QAC-CBM improved the wound microenvironment and accelerated wound healing in diabetic mice by promoting tissue production and collagen deposition, inducing M2 macrophages, reducing pro-inflammatory factor secretion and increasing anti-inflammatory factor levels. Moreover, AgNPs@QAC-CBM was proven to be safe for use through skin irritation and cytotoxicity tests, as they did not cause any irritation or toxicity. 29. Two ‘faces’ of ENSO-induced surface waves during the tropical cyclone season

Abstract

The response of the wave climate to tropical cyclones (TCs) is investigated using the significant wave height (SWH) observed from satellites and the WAVEWATCH III (WW3) model. Tropical cyclone wind generates local waves (wind seas) under the TC and longer-period waves (swells) that propagate long distances. The genesis location, intensity, and frequency of tropical cyclones over the global ocean are strongly affected by the phases of the El Niño Southern Oscillation (ENSO). It is shown that the interannual variation of global ocean surface waves in the subtropics during summer is dominated by ENSO-related TC activity. In particular, in the subtropical western North Pacific, the wind power is stronger in the TC season before an El Niño and weaker before a La Niña. These ENSO-related TC variations are shown, through composite and empirical orthogonal function analyses, as well as modeling, to dominate the spatial distribution and temporal variation of the SWH over the western North Pacific. The model confirms that longer-period waves (swells) are driven into northern South China Sea, toward Japan in mid latitudes and toward the central Pacific along the equator due to ENSO-related TC activities. The wind power and SWH over the subtropical western North Pacific lead and regress well with the ENSO index, suggesting that they may potentially serve as useful ENSO predictors.

Abstract

In a preceding paper [J. Chem. Phys. 131, 154103 (2009)], we introduced a new, hybrid explicit/implicit method to treat electrostatic interactions in computer simulations, and tested its performance for liquid water. In this paper, we report further tests of this method, termed the image-charge solvation model (ICSM), in simulations of ions solvated in water. We find that our model can faithfully reproduce known solvation properties of sodium and chloride ions. The charging free energy of a single sodium ion is in excellent agreement with the estimates by other electrostatics methods, while offering much lower finite-size errors. Similarly, the potentials of mean force computed for Na-Cl, Na-Na, and Cl-Cl pairs closely reproduce those reported previously. Collectively, our results demonstrate the superior accuracy of the proposed ICSM method for simulations of mixed media. [ABSTRACT FROM AUTHOR]

Most patients with NSCLC and somatic mutations have shown a dramatic initial clinical response to reversible EGFR inhibitors. The clinical efficacy of reversible inhibitors is, however, ultimately limited due to the emergence of drug resistance, which is usually conferred by the EGFR T790M mutation. Importantly, irreversible, synthetic small molecule inhibitors are currently evaluated and some of them have been shown to overcome the acquired resistance that is oftentimes observed in these patients. Thus far, irreversible EGFR inhibitors as a drug class have not received regulatory approval due in part to their poor effectiveness at clinically achievable concentrations. Therefore, there is an urgent need to discover and develop novel, potent irreversible inhibitors against the EGFR T790M mutation.Material and methods: In the following study, we report a novel “hybrid strategy” to identify irreversible EGFR inhibitors with active scaffolds starting with the identification and extraction of a common chemical reactive feature and a pharmacophore feature.
