Abstract Background The paper mulberry (Broussonetia papyrifera L.) is a valuable source of woody forage that can be used for ruminants, such as goat and lambs. However, there is limited knowledge about how paper mulberry silage affects carcass characteristics, meat physicochemical attributes, amino acid and unsaturated fatty acids in Hu lambs compared to alfalfa silage. The objective of this experiment was to investigate the impact of substituting alfalfa silage with paper mulberry silage on the slaughter performance, meat quality, free amino acid and fatty acid composition in muscles of Hu lambs. Results Thirty 60-day-old male Hu lambs with 17.13 ± 0.26 kg body weight were randomly allocated into paper mulberry silage (30% of total mixed ration) and alfalfa silage (30% of total mixed ration) treatment, and the feeding trial lasted 70 days. The results indicated no significant differences in all measurements (P > 0.05). However, compared to the group fed with alfalfa silage, the group fed with paper mulberry silage exhibited a reduction in meat drip loss (P 0.05). Lung cancer is the most common cancer worldwide. Epigenetic modifications like DNA methylation play fundamental roles in the dynamic process of lung cancer. The objective of this study was to use methyl-CpG binding domain protein-enriched genome sequencing (MBD-Seq) to identify novel and high-confidence DNA methylation in lung tumor. We first compared the whole-genome DNA methylation of three lung cancer cell lines, including A549, H1299, and SK-MES-1, against BEAS-2B, a lung/bronchial normal epithelial cell line. We then used pyrosequencing and OneStep qMethyl kit methods to verify the results in the cell line specimens. MBD-Seq identified 279, 8046, and 22 887 differentially methylated regions (DMRs), respectively, with 120 common DMRs among three comparison groups. Three DMRs were consistent with the MBD-Seq results by both pyrosequencing and OneStep qMethyl validations. Furthermore, OneStep qMethyl kit was also performed for functional validation of these three potential DMRs in sputum DNA from clinical participants. We successfully identified one new DMR adjacent to ATG16L2. The novel DMR might have an important function in lung carcinogenesis. Further validation of the finding in clinical specimens of lung cancer patients and functional analysis of this novel DMR in the development of lung cancer through transcriptional silencing of ATG16L2 are warranted. L'objectif de ce travail était d'avoir recours au séquençage de la fraction du génome qui est enrichie en séquences méthylées en contexte CpG (MBD-Seq) pour identifier des sites méthylés inédits et avec grande confiance au sein de tumeurs des poumons. Les auteurs ont d'abord comparé la méthylation pangénomique de l'ADN chez trois lignées de cellules cancéreuses (A549, H1299 et SK-MES-1) avec celle décelée chez BEAS-2B, une lignée de cellules épithéliales normales du poumon ou des bronches. Les auteurs ont ensuite pratiqué un pyroséquen-çage et une analyse à l'aide d'une trousse [beaucoup moins que] OneStep qMethyl [beaucoup plus grand que] pour vérifier les résultats chez ces lignées cellulaires. A simplified column-buckling model is developed to understand the buckling mechanism of thin-walled strips restrained by uniform lateral pressure in the milling process. The strip is simplified as two rigid columns connected by a rotation spring, resting on a smooth surface, restrained by a uniform pressure and loaded by an axial force. Two loading cases are considered, i.e., the dead load and the follower load. Analytical solutions for the post-buckling responses of the two cases are derived based on the energy method. The minimum buckling force, Maxwell force and stability conditions for the two cases are established. It is demonstrated that the application of higher uniform pressure increases the minimum buckling force for the column and thus makes the column less likely to buckle. For the same pressure level, the dead load is found to be more effective than the follower load in suppressing the buckling of the system. The effect of initial geometric imperfection is also investigated, and the imperfection amplitude and critical restraining pressure that prevent buckling are found to be linearly related. The analytical results are validated by finite element simulations. This analytical model reveals the buckling mechanism of strips under lateral pressure restraint, which cannot be explained by the conventional bifurcation buckling theory, and provides a theoretical foundation for buckling-prevention strategies during the milling process of thin-walled strips, plates and shells commonly encountered in aerospace or automotive industries.
